Minet, Antoine. "Bioaccumulation and effects of mercury in cuttlefish in the context of ocean acidification." Electronic Thesis or Diss., La Rochelle, 2022. http://www.theses.fr/2022LAROS039.

Abstract:
The common cuttlefish, Sepia officinalis, is a coastal cephalopod known for its eco physiological performance supported by a wide repertoire of behaviors that allows it, despite its short life cycle, to play a central role in food webs. However, it is also known to efficiently accumulate metallic elements such as mercury (Hg), a neurotoxic contaminant in its organic form (methylmercury; MeHg). Moreover, in the context of global changes, ocean acidification (OA) induced by the increase of CO2 emissions, can impact metabolism, development and the central nervous system, especially in young life stages. The present work therefore aims to study the effects of OA on Hg bioaccumulation and the effects of these two neurotoxicants (Hg and CO2) on behavior and cognitive performance in juvenile cuttlefish. In the first place, in-situ measurements showed that Hg accumulated in the brain of cephalopods in methylated form (MeHg). Moreover, experimental approaches using isotopic tracers (stable and radioactive) of Hg have shown that the accumulation of total Hg comes mainly from the assimilation of MeHg present in preys, compared to the accumulation of inorganic mercury (iHg) dissolved in seawater. This MeHg is then very strongly retained despite evidence of Hg demethylation processes in the digestive gland. Surprisingly, OA has no effect on the bioaccumulation efficiencies and metabolism of Hg. In terms of effect, behavioral assays showed that Hg and CO2, alone or in combination, did not impact visual acuity and predation performance in juveniles. However, both induced an increase in locomotor activity, compromised defensive behavior through, in particular, an alteration of disruptive coloration and a non-alignment of lateralization. Among the neural processes likely to be impacted by Hg and CO2, the GABAergic system was analyzed in the optic lobes but showed no obvious link with behavioral responses suggesting more complex effect processes. All of this work therefore questions the effects of future ocean conditions on juvenile’s cuttlefish, and more broadly the impacts on cephalopod populations

Daly, J. N. "IgH allelic exclusion at the level of IgH transcription." Thesis, University of Cambridge, 2005. http://ethos.bl.uk/OrderDetails.do?uin=uk.bl.ethos.598257.

Full text
Abstract:
Expression of a productivity rearranged immunoglobulin heavy (μH) chain from a first in-frame VDJH-rearranged allele prevents further recombinant of the second, DJH-rearranged, allele, termed heavy chain allelic exclusion. I investigated at the single cell level the transcriptional status of both IgH alleles in ex vivo cells using RNA fluorescent in situ hybridisation (FISH). Prior to exclusion, in predominantly DJH /DJH-rearranged pro/pre-B1 cells, both IgH alleles are transcribed and detected by RNA-FISH. In contrast, following productive VDJH-recombination and μH expression i.e. in allelically excluded pre-BII cells, mono-allele transcription was detected in the 40% and bi-allelic in 60% of cells. This 40:60 ratio was maintained throughout subsequent B cell developmental stages. A VH-specific transcription assay revealed a VH (J558) transcription co-localising with mono- and bi-allelic μ transcription sites, strongly indicating transcription from VDJH-rearranged alleles in allelically excluded B cells. Combined RNA/DNA FISH showed detection of the VH-DH intergenic regions in cells with mono-allelic μ-transcription. Within these nuclei the μ-transcription site and VH-DH intergenic region were clearly separate and thus do not originate from the same allele, thus suggesting a VDJH/DJH-rearrangement status. In the majority (>80%) of nuclei with two μ-transcription sites no VH-DH-intergenic regions were observed, suggesting VDJH/VDJH-recombination. Semi-quantitative RT-PCR revealed steady-state DJH-Cμ transcripts in splenic B cells, which have been allelically excluded at levels 2-3 fold less than observed in a developmental stage prior to exclusion. Thus the DJH-recombined allele is transcribed after exclusion. Our data show that the process of allelic exclusion results in diminished ability for RNA-FISH to detect the transcription of the DJH- but not the VDJH-rearranged alleles, indicating that the signal for allelic exclusion is received only by the DHJ-recombined alleles. Therefore, in allelically excluded cells, the DJH-but not VDJH-rearranged alleles alter their transcriptional activity and thus act as part of the mechanism for heavy chain allelic exclusion.

Abstract : Innovation habitats are systematically organized enterprises to promote innovation and provide the interaction and integration of different actors. They are contemporary phenomena of high relevance for the development of a region or a country because they operate with knowledge assets. The implementation of an innovation habitat directly implies the socioeconomic and cultural development of the surrounding community. These provide differentiated conditions for developing innovative processes and products to foster economic, social, local and regional dynamics as it can bring together people, technologies, financial resources, and knowledge. The issue regarding this research analyzes the communities surrounding the innovation habitats about their needs, potentialities, and expectations. The theoretical background for this thesis is built on the intersection between concepts of innovation habitats, technology parks, changes, social innovation, triple and quadruple propeller. The aim is to develop a methodology for reading and analysis, identifying needs, potentialities, and expectations of communities surrounding the innovation habitats. The study is characterized as descriptive and exploratory. It develops from a systematic review to obtain basic concepts and a benchmarking to identify surrounding reading practices. With such elements, it is proposed a methodology of environment interpretation developed in three phases: elaboration, prototyping, and application. The elaboration was constituted by an online instrument that was prototyped in the Orion Park?s surrounding, in the city of Lages. The methodology application for its configuration was developed in the Sapiens Park?s surrounding, in Florianópolis. The actors involved in the process were eight managers of the technological parks visited, ten managers of internal organizations of the Sapiens Parque, 201 managers of external companies and 622 families located around the Sapiens Parque. The result obtained with the study relates to an adaptive methodological proposal for different types of innovation habitats, which makes it possible to read and analyze the enterprises? surroundings, generating information that characterizes thelocal population profile regarding their needs, potentialities, and expectations. With the application of InHab-Read - IHR - Environment Reading Methodology for innovation habitats, it is possible to propose common policies and actions between the surrounding population and habits internal organizations, enhancing the interaction among all the actors, and broadening dimensions of technological and economic innovation processes for social change.

Abstract:
This study investigates how non-ing initial adverbials and adverbial ing-clauses regardless of position are translated from English to Swedish in a medical text. It discusses what different kinds of positions and forms they take in the translation. The primary source is a medical text that is translated wherein the non-ing initial adverbials and adverbial ing-clauses are identified and categorized, and solutions are discussed. The results show that most non-ing initial adverbials keep their form, but are changed in some cases because of stylistic preferences. The most common form of the non-ing initial adverbials is prepositional phrases that mostly also are translated as prepositional phrases. The adverbial ing-clauses, regardless of position, change their form in all cases in the translation and the most common correspondence is subordination. Both non-ing initial adverbials and adverbial ingclauses keep their position in most cases, but are sometimes moved due to stylistic preferences.

Hoechtl, Nina. "If only for the length of a lucha : queer/ing, mask/ing, gender/ing and gesture in lucha libre." Thesis, Goldsmiths College (University of London), 2012. http://research.gold.ac.uk/8056/.

Full text
Abstract:
This PhD uses a queer reading strategy to explore the performative sites of lucha libre (wrestling in Mexico). My research inhabits the space behind the scene, the space between the ring and the audience, and the space of being part of the audience itself. My reading of the luchas takes place through the camera, the interview, printed works, theoretical investigation, and through the work of artists who draw on lucha libre – including myself. As lucha libre itself cannot be narrowed down to one specific medium, my subject matter allows me to utilize an interdisciplinary perspective to examine its various encounters, spaces, subjectivities and gestures. As well as attending live events in the arenas, watching lucha libre on television, exploring its circulation in artistic and filmic productions and its appropriation in advertisements and political discourse, I have carried out an intervention in a regular lucha libre programme by inventing a character, promoting, constructing and staging a match in an arena in the north of Mexico City. My methodology therefore makes use of a whole range of strategies including those borrowed from the discipline of anthropology and from practices of documentary making. Through my writing and my practice, I attempt to query and complicate these disciplinary conventions and my own use of them. I place particular emphasis in this PhD upon the possibility and use of a queer reading strategy in relation to lucha libre. Drawing on the works of Gloria Anzaldúa, Judith Butler, Judith ʻJackʼ Halberstam, José Javier Maristany, and José Esteban Muñoz, the thesis argues that a queer reading strategy has the potential to complicate ways of seeing gender and sexuality as well as race, ethnicity, class, time and space in this context. I identify the rich queer legacies within lucha libre, film and popular culture, and focus on the multiple and often conflicting readings made possible by adopting queer theory and reading practices. In doing so, the thesis interrogates the different ways in which popular cultures can go beyond accepted notions of what it means to be Mexican, a woman, macho, gay and so on. Throughout this work, I pay close attention to forms of audience participation, their verbal and gestural acts and how key these are in to the event of the lucha. These verbal and gestural acts, I argue, produce a unique complicity in the arena manifesting a transient trace of queer histories, and suggesting potential utopias.

Abstract:
This thesis marks a point where the priorities determining my occupation with architecture might shift. In part it is a reflection of an approach to architecture, which generates an awareness that can be extended to an environment of practice. This awareness should provide direction towards the making of architecture, regardless to the number of constraints or limitations which reality might impose. This project is the documenting vehicle for the examination of a design process. The proposal is the evidence of this design process. Its documentation should prove and expose qualities which derived from my approach to architecture.

Abstract:
The main purpose of the essay is to serve as a documentation of my research practice on the movement of the tailbone and its connections to my dancing body. The essay is being written from my personal perspective which springs out of my interest in the importance of using the tailbone whilst dancing. In the first part of the text I have included personal information to the reader about where my interest in the movement of the tailbone arose. One of the methods during the research was to write a process diary as a way to combine a physical practice and writing practice. This process diary is now a big part of this essay. In the essay you as a redear can follow how the research has transformed and changed throughout working time on the project. The second part of the essay reflects on the process of researching the tailbone-ing movement practice and what the practical presentation should look like. In the last pages of the essay you find a choreographic score which is both a documentation of the practice as well as a score to perform it. Working on the essay opens up new possibilties for further research on the importance of the tailbone and the pelvic floor in the dancing body.

This master work includes both a performing and a written part.

Martin, Susanne. "Dancing age(ing) : rethinking age(ing) in and through improvisation practice and performance." Thesis, Middlesex University, 2016. http://eprints.mdx.ac.uk/18984/.

Full text
Abstract:
Set in the context of contemporary dance this thesis investigates how improvisation practice and performance making participate in a critical rethinking of age(ing). Advancing the notion of an age critical dance practice, the research draws on the theoretical frameworks of age studies – a multidisciplinary field of critical inquiry informed by, largely speaking, feminist and poststructuralist theories. The age critical dance practice developed in this thesis, in turn, enters into conversation with the discourses established in age studies and dance studies as a way to incorporate age critique into dance. The thesis is a Practice as Research project consisting of a written thesis, two solo performances (The Fountain of Youth, premiered 2013, and The Fountain of Age, premiered 2015), and employs immersive dance based research methods such as the development of a Solo Partnering practice (as documented on DVD). The research also remodels the method of qualitative interviewing into a performative method that allows the participating expert practitioners to tap into their unique improvisation and performance expertise when addressing their particular understanding of age(ing). Through the development and analysis of improvised practice and performance making, alongside in-depth performative interviews, the findings of this research point to ways in which improvisation and performance embody age critical potential. The long-term, open-ended and agentic artistic processes that improvisation experts develop all share a range of characteristics that serve to challenge the established youth-orientation in dance and constitute an implicitly critical position to dominant understandings of age(ing) in dance. Consequently, the thesis argues that improvisation practices ‘do’ age(ing) in ways less prone to dualistic stereotyping and reiterations of (self‐) discriminatory age(ing)-as‐decline narratives that dominate our culture as a whole. The research also suggests strategies in performance making that enable representations of age(ing) in ways that collide with, resist, or complicate normative expectations on age(ing). The dance works presented in this thesis allow the dancer to articulate shifting perspectives and experiences, creating ambiguous meanings and disjunctive narratives of age(ing), and thereby making explicit a critical position towards the grand narratives of age(ing). In conclusion, this research argues that specific approaches to a long-term, open-ended dance practice, alongside critical images and new imaginations of age(ing) in performance, allow dance to evolve as an age critical arts practice.

Abstract:
The history of jews in Wriezen begins in 1677, shortly after Kurfürst Friedrich Wilhelm allowed jews to settle in Brandenburg again through his edict of 1671. However, during the whole of the 18th century Prussian policy toward jews was extremely restrictive . The results of this policy are clearly visible in the life of the jews of Wriezen: they always remained a small congregation since this was the will of the king for small towns. Life was dominated by the struggle for the right of residence. Status as "Schutzjude" (i.e. "Protected Jew") was restricted to few individuals, leading to the separation of families as younger siblings were forced to leave. State regulated economic policy strongly restricted the freedom of trade, the main source of income for the jews, leading to the impoverishment of most jewish families in Wriezen. In the 18th century, there was no organized congregational activity. This only developed in the course of the 19th century. The jews of Wriezen built their first synagogue in 1820, replacing it with a new and larger one in 1886. The emancipation edict of 1812 improved the general situation of individuals and the new freedom of movement led to an influx of jews to Wriezen. But full legal equality with other citizens was not achieved until the German Empire was founded in 1871. In the first half of the 19th century, the economic situation of the jews of Wriezen was still quite modest, only improving toward the end of the century. There were numerous fluctuations in the membership of the jewish community in the last decades of the 19th and the beginning of the 20th century. But the number of jewish inhabitants of Wriezen remained relatively constant at 100 -120 individuals. There is hardly any written information on the sudden and complete end of jewish life in Wriezen after 1938. At least 59 jewish citizens of Wriezen were deported and murdered between 1940 and 1945. The victims are commemorated in the public database of the Yad-VaShem Memorial in Jerusalem. There has been a jewish cemetery in Wriezen since 1730, in which 131 gravestones still survive today. The oldest dates back to 1773, the last is from 1940.

Abstract:
For 270 years there was Jewish life in the Brandenburg town of Schwedt on the Oder. The reader of this volume participates in the changing history of the Jewish people and their community. The first Jew to settle in Schwedt in the 17th century was Bendix Levi. After his house in Oderberg burned down, Kurfürst Friedrich Wilhelm granted him permission to take up residence in Schwedt. The Prussian Kings wanted only few Jews to live in small Brandenburg towns, so during the 18th century the Jewish community in Schwedt consisted of only three families whose heads were Bendix Levi, Simon Salomon and Wulff Salomon. Their life was dominated by the fight for residency and trade permits. Solidarity amongst each other was not always self evident, since the residency permit of one was often in real or imagined conflict with a permit for others. The extensive sources in archives in Berlin, Brandenburg and Jerusalem show intensely the consequences of the restrictive Prussian “Judenpolitik” on the life of Jews in Schwedt and they also give us insight into Jewish traditions and customs. We bear witness to a Jewish oath in the townhouse of Schwedt and take part in the ceremonial signing of a marriage treaty. The “Emanzipationsedikt” of 1812 finally made Jews into Prussian citizens and simplified their life by giving them freedom of settlement and trade. This resulted in considerable growth of the Jewish community in Schwedt which was also joined by the Jewish residents of the nearby town of Vierraden. The first of these new settlers in Schwedt was David Loewenheim from Tütz / Westprussia. No less than seven families from his hometown followed. There was also immigration from other towns, increasing the number of Jewish residents in Schwedt to 200 by the late 19th century. After about 1880 the community shrank again because many moved away, particularly to Berlin. Since 1840 Schwedt was the seat of a Rabbi. Nathan Hirsch Kuttner stayed in Schwedt until his retirement in 1895. Although this long sojourn might indicate an agreeable relationship between him and his community, this was certainly not the case in his first years. Kuttner had to fight for recognition by his community and even had to obtain a court order to collect his salary. The building of a new Synagogue in 1862 gave a new sense of cooperation to the community and calmed the struggles with the Rabbi. Under the impression of increasing anti-Semitism from the late 19th into the 20th century, the community joined organizations which attempted to resist anti-Semitic attacks. For example, in 1894 it joined the “Deutsch- Israelitische Gemeindebund” and in 1901 the “Verein zur Abwehr des Antisemitismus”. In 1922 the “Reuchlin Loge”, was formed within the “Independent Order of Bnai Brith”. Soon followed the fast and complete anihilation of the Jewish community in Schwedt by the national socialist mass murder. The Central Database of Shoah Victims' Names at Yad Vashem / Jerusalem preserves the memory of 71 murdered Jews of Schwedt who are known by name. Descendants of survivors are scattered to all parts of the world. Intensive genealogical research in cooperation with Peter Lowe from Hertford (UK), descendant of the Loewenheim family, and Yehuda Meinhardt (Israel), made it possible to reconstruct a number of family trees. The volume ends with a description of the Jewish cemetery and a record of the grave inscriptions. An entire documentation with photographs can be found in the Internet (www.uni-potsdam.de/juedische-friedhoefe).

Abstract:
The conserved N-linked glycosylation site on the Fc domain of IgG1 antibodies is essential for maintaining a functionally active conformation of the antibody. Different glycoforms of the Fc exhibit widely different effector functions. Similarly, therapeutic antibodies, with engineered glycosylation, exhibit altered binding to cellular Fc receptors (FcRs). Here, X-ray crystallographic structures were obtained for biosynthetic intermediate glycoforms of human IgG1 Fc bearing: unprocessed oligomannose-type, intermediate hybrid-type, and mature complex-type glycans. The fully processed Fc protein crystallised in an “open” conformation with glycans forming canonical stabilising interactions on the protein surface. Analysis of the biosynthetic intermediates revealed that these stabilising hydrophobic protein-glycan interactions are formed only after processing by Golgi -mannosidase II. Mutagenesis of hydrophobic residues on Fc disrupted crucial protein-glycan interactions resulting in the selective destabilization of the 3-arm of the glycan chain with the 6-arm closely matching that seen for the native structure. However, carbohydrate analysis of released glycans shows increased processing on both arms indicating a more accessible and flexible glycan in the mutant structure suggesting that the crystallographic structure of these antibody glycans represents a minor low-energy conformation. The importance of Fc glycosylation is highlighted by endoglycosidases which eliminate Fc effector function. The crystallographic structure of enzymatically deglycosylated IgG Fc revealed a significant collapse of the of Cγ2 domains resulting in a ‘closed’ quaternary conformation, incompatible with Fc receptor binding. This provides a structural explanation for immune deactivating properties of endoglycosidases including those under preclinical development for the treatment of antibody-mediated immune pathology. One such bacterial endoglycosidase, Endo S, was studied further and revealed a specificity for complex-type glycans of the type found on IgG but no hydrolytic activity towards an engineered IgG Fc with oligomannose-type glycans. Introduction of both the engineered monoclonal IgG and endoglycosidase in serum led to a dramatic increase in FcR binding as the competitive binding of serum IgG for FcRs was selectively eliminated. This approach is a general technique for boosting the effector signal of therapeutic antibodies.

Abstract:
In order to explain the role of proper names in narrative fiction the literary text is seen as a system which, however, is connected to the “real world' in many ways. As a work of art the literary text is characterized by the predominance of the aesthetic function. From applying R. JAKOBSON’S and J. MURAKOVSKY’S concept of the aesthetic sphere as the sphere where the linguistic sign primarily reveals its inherent aesthetic and semantic values follows that the communicative functions of the proper name in literary texts are reverse to their order in everyday communication: It is not the identifying function of the proper name which is dominant in literary texts, but rather all the other semantic and phonic values which in real-world communication usually are not realized.

By considering an approach which relates postcolonial studies and translation theories, this study questions the use of linguistic loans and foreignisms in Oswald de Andrade s poetic productions as translation acts and anti/postcolonial critique. The postcolonial categories presented by researchers as Edward Said, Homi Bhabha and Gayatri Spivak namely, the rupture of frontiers, difference as an enunciative category and temporality in the in-between space and questions on translation metaphors and untranslatability, in dialogue with the rich critique produced on Oswald de Andrade, stimulate the careful analysis of anthropophagy in the literary texts taken as our corpus: O Manifesto Antropófago (2011) and Memórias Sentimentais de João Miramar (2004). The result of the present research stresses the need for considering, within our current postcolonial debates, the anticipation of this anthropophagic reasoning, thus revealing a better understanding of important postcolonial aspects in Brazil

Tuberculosis continues to be one of the deadliest diseases in the world. The emergence of drug-resistant strains of Mycobacterium tuberculosis, the unbearable side effects of the available drugs and the frequent patient non-compliance in completing the therapy have increased the need for development of new effective agents. We have previously demonstrated a potent in vitro inhibitory activity for two pentacyano(isoniazid)ferrate(II) compounds, namely IQG-607 and IQG-639 against the M. tuberculosis enoyl-ACP reductase enzyme. Importantly, IQG- 607 and IQG-639 were active against cultures of M. tuberculosis H37Rv and two isoniazid-resistant clinical isolates in vitro. In the present study, the activity of these compounds was evaluated by using an in vivo murine model of tuberculosis. Swiss mice were infected with M. tuberculosis H37Rv strain, and IQG-607 and IQG-639 (250 mg/kg) were administered during 28 or 56 days. As well, a dose-response study was performed with IQG-607 (at 5, 10, 25, 50, 100, 200 and 250 mg/kg). The activity of test compounds was compared with that of the positive control drug isoniazid at 25 mg/kg. After 28 or 56 days of treatment, either IQG-607 or isoniazid significantly reduced M. tuberculosis-induced splenomegaly, and also significantly diminished the colony-forming units in both spleens and lungs. IQG-607 or isoniazid ameliorated the lung macroscopic aspect, reducing the lung lesions to a similar extent. However, IQG-639 was not capable of significantly modifying any evaluated parameter. IQG-607 did not display a classical dose-dependent profile in our murine model of tuberculosis, and 10 mg/kg was the lowest dose able to show significant activity, which was similar to the inhibition observed for higher doses. In addition, experiments using early and late controls of infection revealed a bactericidal activity for IQG-607 in our model. The promising activity of IQG-607 in M. tuberculosis-infected mice suggests that this compound might represent a good candidate for clinical development as a new antimycobacterial agent.
