Abstract:
The International Gravitational Event Collaboration, IGEC, is a coordinated effort by research groups operating gravitational wave detectors working towards the detection of millisecond bursts of gravitational waves. Here we report on the current IGEC resonant bar observatory, its data analysis procedures, the main properties of the first exchanged data set. Even though the available data set is not complete, in the years 1997 and 1998 up to four detectors were operating simultaneously. Preliminary results are mentioned.

Abstract:
Inhalation of polyaromatic hydrocarbons (PAHs) extracted from diesel exhaust particles (DEP) enhances local (nasal) production of IgE in humans. The aim of the present research is to investigate whether in humans dermal exposure to PAHs which are not extracted from DEPs increases serum IgE and whether host factors modify the immunologic effect. In thirty-two patients with acute psoriatic lesions, a cream containing 3% of coal tar (which holds a variety of PAHs) was applied to the skin for 24 hours. Serum IgE were measured before (IgE0) and four (IgE4) and eight (IgE8) days after application. Replicated means were compared by analysis of variance for repeated measures and by the Newman-Keuls' test. IgE0, IgE4 and IgE8 were 151.19, 159.69 (a 6% excess) and 170.90 kU/L (a 13% excess) respectively; pairwise comparison showed IgE8 was significantly higher than IgE0 (p<0.05). At multiple linear regression analysis, the percentage increase in serum IgE across observation days was the dependent variable against age, sex, cigarettes/day, urinary 1-pyrenol, atopy, skin area treated, and grams of cream. Of the independent variables, only age had a significant (p<0.028) influence: the younger the age, the higher the IgE response to PAHs. We conclude that whatever the source and the route of entry (skin or respiratory tract), PAHs increase total serum IgE, mainly in younger age groups.

Para cada unidade educativa é produzido um relatório que se encontra publicitado na página web da IGEC. Em cada relatório a equipa de avaliação faz uma apreciação do funcionamento da organização educativa avaliada de acordo com três domínios (Resultados; Prestação do Serviço Educativo e Liderança e Gestão). Na última secção de cada relatório, refere-se os principais pontos fortes, bem como às áreas de melhoria. Com a finalidade de se descortinar se a diferenciação pedagógica emerge como uma prática regular do processo de ensino e da aprendizagem analisámos o conteúdo dos 59 relatórios da Área de Inspeção Territorial Norte. Em função dos resultados obtidos concluímos que só 20,3% das escolas avaliadas desenvolvem práticas de diferenciação de forma regular e consistente.

Gravitational events on IGEC data are marked with their detection time td. Although td takes into account phase shifts due to hardware and software, it could be statistically retarded or anticipated by noise with respect to the arrival time ta of a gravitational wave. Then we can estimate a time interval Δ for each event so that we can consider the ta of the event equally likely in Δ. Consequently we can define a n-fold coincidence as a n-tuple of events whose associated time intervals have a non-empty intersection. Using this definition and taking into account some time interval properties we show that a n-fold coincidence set can be computed by an appropriate selection of events from the 2-fold coincidence sets. Furthermore, we stress that a data base management system (DBMS) is a very effective tool to store IGEC data (i.e. events) and to implement n-fold coincidence analysis based on the above considerations. Finally we propose to use a DBMS to build a database of both events and n-fold coincidence analysis data in order to have an efficient and powerful tool for data retrieval and further analysis.

Ziel: Das Ziel dieser Studie war die Untersuchung der akuten Hornhaut-Toxizität von Diquas®, einer auf dem Markt erhältlichen 3%igen Diquafosol-Lösung für das Auge mit C12-Benzalkoniumchlorid (BAC) als Konservierungsmittel. Methoden: Bei lebenden Kaninchen maßen wir die Veränderung des transepithelialen elektrischen Widerstands (TEER) der Hornhaut nach 60-sekündiger Exposition gegenüber Diquas® (Diquafosol 3%, mit 0,0075% C12-BAC als Konservierungsmittel) sowie 0,0075%iger C12-BAC-Lösung und 0,0075%igem C12/C14/C16-BAC-Gemisch. Außerdem wurde die Schädigung der Hornhaut mittels Rasterelektronenmikroskopie (REM) untersucht. HBSS (Hank's Balanced Salt Solution) diente als Kontrolle. Ergebnisse: Diquas® und 0,0075%ige C12-BAC-Lösung bewirkten im Vergleich zur HBSS-Kontrolle keine signifikante Veränderung des Hornhaut-TEER. Eine signifikante Abnahme des TEER war nach Exposition der Hornhaut gegenüber dem 0,0075%igen C12/C14/C16-BAC-Gemisch festzustellen (p < 0,01). Die REM ergab, dass die oberflächlichen Zellen der mit dem 0,0075%igen BAC-Gemisch behandelten Netzhäute geschädigt waren und degenerierte Mikrovilli aufwiesen. Nach Exposition gegenüber Diquas® oder 0,0075%iger C12-BAC-Lösung sahen die oberflächlichen Hornhautzellen in der REM hingegen normal aus und hatten normale Mikrovilli. Schlussfolgerung: Die akute Hornhaut-Toxizität von Diquas® ist geringer als die des 0,0075%igen BAC-Gemischs. Zusammenfassung Einleitung Das thermische Trauma im Kindes- und Jugendalter ist weiterhin von hoher sozioökonomischer Relevanz, da es seit Jahren zu keiner Reduktion der jährlich stationär behandelten Patienten/innen gekommen ist. Die Therapie des pädiatrischen Verbrennungspatienten stellt besondere Anforderungen an das medizinische Personal, die verwendeten Verbandsmaterialien und die angewandten Operationstechniken. Ziel dieser Studie war es einen aktuellen Überblick über das stationäre Management und die Therapie des thermischen Traumas bei pädiatrischen Patienten/innen an einer spezialisierten Klinik zu geben. Patienten/Material und Methode Kinder und Jugendliche, die zwischen 2012 und 2016 an der Kinderstation der chirurgischen Fächer der Medizinischen Universität Wien von der klinischen Abteilung für Plastische und Rekonstruktive Chirurgie bei thermischen Trauma behandelt wurden, wurden identifiziert und ihr Therapieverlauf retrospektiv analysiert. Ergebnisse 115 Patienten/innen wurden auf Grund eines akuten thermischen Traumas stationär behandelt. Das mediane Alter lag bei 2 Jahren (0–18). Die Verbrühung stellte mit 74 % den häufigsten Verletzungsmechanismus dar. Im Median zeigten sich 5 % (1–40) der Körperoberfläche betroffen. Konservatives Wundmanagement bei 2a°igem Trauma (62 %) wurde in 98 % mit MepilexAg durchgeführt und dauerte im Median 11 Tage (4–34) bis zur Abheilung. In 38 % (n = 44) lag eine 2b°ige oder 3°ige Verbrennung vor. Die Verwendung von Suprathel nach tangentialer Nekrosektomie bzw. Bürstendebridement bei oberflächlichen 2b°igen Verletzungen zeigte zufriedenstellende Heilungsverläufe. Schlussfolgerung Durch den Einsatzvon modernen Wundverbänden in der konservativen Therapie 2a°iger Wunden kann eine schnelle und unkomplizierte Abheilung mit atraumatischen Verbandswechseln erfolgen. Bei oberflächlichen 2b°igen Wunden bietet Suprathel eine gute Alternative zur klassischen Hauttransplantation, welche jedoch bei tieferen 2b°igen Verbrennungen immer noch als Therapiestandard gelten sollte. Narbenprophylaxe und regelmäßige Nachkontrollen sind wichtig um sekundäre Funktionseinschränkungen durch Narbenstränge zu vermeiden. Um zukünftig die Inzidenz des thermischen Traumas im Kindesalter zu senken ist die prophylaktische Aufklärung und Sensibilisierung der Eltern von großer Wichtigkeit.

Zusammenfassung Gegenstand und Ziel: Untersuchungen zur Beeinflussung des Blutspiegels von Glukose und anorganischem Phosphat bei gesunden, laktierenden Kühen nach 5- und 20%iger Glukoseinfusion. Material und Methoden: Für den Versuch standen 10 gesunde, laktierende und nichtgravide, 3- bis 6-jährige Kühe der Rasse Deutsch Holstein mit einer Tagesmilchleistung von 20,3±2,7 Litern zur Verfügung. Den Tieren wurde vor, während sowie 0 und 60 Minuten nach der Infusion von 0,9%iger NaCl-Lösung, 5- oder 20%iger Glukoselösung Blut zur Bestimmung des Gehalts an Glukose und anorganischem Phosphat entnommen. Ergebnisse: Es konnte ein signifikanter Einfluss der Glukose infusion auf den Blutglukosespiegel der Kühe nachgewiesen werden. Nach Infusion von 1000 ml einer 20%igen Glukoselösung (200 g Glukose) stieg die Glukosekonzentration um durchschnittlich 13,26 mmol/l, nach Verabreichung von 1000 ml einer 5%igen Glukoselösung (50 g Glukose) um durchschnittlich 3,31 mmol/l. Kein signifikanter Zusammenhang ergab sich zwischen einer 0,9%igen NaCl-Infusion, 5- oder 20%igen Glukoseinfusion und der Blutkonzentration an anorganischem Phosphat. Schlussfolgerung: Eine Infusion von 1000 ml 5- oder 20%iger Glukoselösung bedingt keinen nachteiligen Abfall des Blutphosphatspiegels und eignet sich zur Verbesserung der Blutglukose versorgung der Kuh.

ZusammenfassungDie Wurzel von Liebstöckel (Levisticum officinale Koch), der v. a. als Gewürzpflanze bekannt ist, wird seit Jahrhunderten zur Herstellung von Arzneimitteln eingesetzt. Neben der traditionellen Verwendung zur Durchspülungstherapie bei Nierenleiden findet in der anthroposophischen Medizin ein 5 %iger Auszug der getrockneten Wurzel in nativem Olivenöl Anwendung bei Mittelohrentzündung. Sicherheit und Wirkungen dieses Auszugs waren Ziele einer präklinischen und klinischen Untersuchung.Methoden: Präklinisch wurden die Effekte des 5 %igen Auszugs sowie des Auszugsöls auf die 5-Lipoxygenase und die Cyclooxygenase-2 und die von ihnen katalysierte Synthese von antiinflammatorischen Mediatoren, Leukotrien B4 und Prostaglandin E2 untersucht. Klinisch lieferte eine nichtinterventionelle Studie mit 85 Patienten Daten aus der ärztlichen Routineanwendung des Auszugs von Levistici radix bei Otitis media.Ergebnisse: Der 5 %ige Auszug von Levistici radix in nativem Olivenöl reduzierte deutlich die Leukotrien-B4- und weniger deutlich die Prostaglandin-E2-Synthese. Die Wirkung des parallel getesteten Olivenöls fiel wesentlich geringer aus. Die klinische Anwendung des Auszugs in Form von Ohrentropfen ergab eine als gut bis sehr gut beurteilte Wirkung und Verträglichkeit.Fazit: Die präklinische Untersuchung liefert eine rationale Basis für die lokale klinische Anwendung des 5 %igen Auszugs von Levistici radix in nativem Olivenöl bei Otitis media, die in der Praxis bestätigt wird.

Abstract Integrative genetic elements (IGEs) are mobile multigene DNA units that integrate into and excise from host bacterial genomes. Each IGE usually targets a specific site within a conserved host gene, integrating in a manner that preserves target gene function. However, a small number of bacterial genes are known to be inactivated upon IGE integration and reactivated upon excision, regulating phenotypes of virulence, mutation rate, and terminal differentiation in multicellular bacteria. The list of regulated gene integrity (RGI) cases has been slow-growing because IGEs have been challenging to precisely and comprehensively locate in genomes. We present software (TIGER) that maps IGEs with unprecedented precision and without attB site bias. TIGER uses a comparative genomic, ping-pong BLAST approach, based on the principle that the IGE integration module (i.e. its int-attP region) is cohesive. The resultant IGEs from 2168 genomes, along with integrase phylogenetic analysis and gene inactivation tests, revealed 19 new cases of genes whose integrity is regulated by IGEs (including dut, eccCa1, gntT, hrpB, merA, ompN, prkA, tqsA, traG, yifB, yfaT and ynfE), as well as recovering previously known cases (in sigK, spsM, comK, mlrA and hlb genes). It also recovered known clades of site-promiscuous integrases and identified possible new ones.

Indirect genetic effects (IGEs) describe how an individual's behaviour—which is influenced by his or her genotype—can affect the behaviours of interacting individuals. IGE research has focused on dyads. However, insights from social networks research, and other studies of group behaviour, suggest that dyadic interactions are affected by the behaviour of other individuals in the group. To extend IGE inferences to groups of three or more, IGEs must be considered from a group perspective. Here, I introduce the ‘focal interaction’ approach to study IGEs in groups. I illustrate the utility of this approach by studying aggression among natural genotypes of Drosophila melanogaster. I chose two natural genotypes as ‘focal interactants’: the behavioural interaction between them was the ‘focal interaction’. One male from each focal interactant genotype was present in every group, and I varied the genotype of the third male—the ‘treatment male’. Genetic variation in the treatment male's aggressive behaviour influenced the focal interaction, demonstrating that IGEs in groups are not a straightforward extension of IGEs measured in dyads. Further, the focal interaction influenced male mating success, illustrating the role of IGEs in behavioural evolution. These results represent the first manipulative evidence for IGEs at the group level.

However, the genetic basis for this variation is poorly understood and a mixed ESS for fighting can be interpreted in more than one way. Specifically, we do not know whether variation in aggression within a population arises from among-individual differences in fixed strategy (determined by an individual’s genotype—direct genetic effects [DGEs]), or from within-individual variation in strategy across contests. Furthermore, as suggested by developments of the original Hawk-Dove model, within-individual variation in strategy may be dependent on the phenotype and thus genotype of the opponent (indirect genetic effects—IGEs). Here we test for the effect of DGEs and IGEs during fights in the beadlet sea anemone Actinia equina. By exploiting the unusual reproductive system of sea anemones, combined with new molecular data, we investigate the role of both additive (DGE + IGE) and non-additive (DGE × IGE) genetic effects on fighting parameters, the latter of which have been hypothesized but never tested for explicitly. We find evidence for heritable variation in fighting ability and that fight duration increases with relatedness. Fighting success is influenced additively by DGEs and IGEs but we found no evidence for non-additive IGEs. These results indicate that variation in fighting behavior is driven by additive indirect genetic effects (DGE + IGE), and support a core assumption of contest theory that strategies are fixed by DGEs.

Motivated by the presence of deep connections among dynamical equations, experimental data, physical systems, and statistical modeling, we report on a series of findings uncovered by the authors and collaborators during the last decade within the framework of the so-called Information Geometric Approach to Chaos (IGAC). The IGAC is a theoretical modeling scheme that combines methods of information geometry with inductive inference techniques to furnish probabilistic descriptions of complex systems in presence of limited information. In addition to relying on curvature and Jacobi field computations, a suitable indicator of complexity within the IGAC framework is given by the so-called information geometric entropy (IGE). The IGE is an information geometric measure of complexity of geodesic paths on curved statistical manifolds underlying the entropic dynamics of systems specified in terms of probability distributions. In this manuscript, we discuss several illustrative examples wherein our modeling scheme is employed to infer macroscopic predictions when only partial knowledge of the microscopic nature of a given system is available. Finally, we include comments on the strengths and weaknesses of the current version of our proposed theoretical scheme in our concluding remarks.

The fact that the intergenerational income elasticity (IGE)—the workhorse measure of economic mobility—is defined in terms of the geometric mean of children’s income generates serious methodological problems. This has led to a call to replace it with the IGE of the expectation, which requires developing the methodological knowledge necessary to estimate the latter with short-run measures of income. This article contributes to this aim. The author advances a “bracketing strategy” for the set estimation of the IGE of the expectation that is equivalent to that used to set estimate (rather than point estimate) the conventional IGE with estimates obtained with the ordinary least squares and instrumental variable (IV) estimators. The proposed bracketing strategy couples estimates generated with the Poisson pseudo–maximum likelihood estimator and a generalized method of moments IV estimator of the Poisson or exponential regression model. The author develops a generalized error-in-variables model for the IV estimation of the IGE of the expectation and compares it with the corresponding model underlying the IV estimation of the conventional IGE. By considering both bracketing strategies from the perspective of the partial-identification approach to inference, the author specifies how to construct confidence intervals for the IGEs, in particular when the upper bound is estimated more than once with different sets of instruments. Finally, using data from the Panel Study of Income Dynamics, the author shows that the bracketing strategies work as expected and assesses the information they generate and how this information varies across instruments and short-run measures of parental income. Three computer programs made available as companions to the article make the set estimation of IGEs, and statistical inference, very simple endeavors.

ABSTRACT The existence of specific immunoglobulin E (IgE) allows us to determine the allergens that cause the allergic disease. For the purposes of allergen avoidance and immunotherapy, the measurement of specific IgE is widely applied in clinical laboratories. However, if IgE from the serum of an allergic patient exhibits reactivity to multiple allergens, it would cause a problem. The present study analyzes whether the serum IgE with multiple reactivity is due to the presence of unique IgE against the common epitope shared by different allergens or the presence of multiple IgEs against different epitopes on different allergens. The quantitative-competitive inhibition tests and the immunoblotting were applied to analyze the immunosimilarity among examined allergens. The result shows that the competitive inhibition of IgE binding between shrimp and crab allergens is higher than those between either shrimp and cockroach or between crab and cockroach. Furthermore, the results of immunoblotting are consistent with those of quantitative-competitive inhibition tests. These results allow us to detect the cross-reactivity for atopic IgE against multiple allergens.

Background. To reveal the regularities of sensitization formation in children with atopic dermatitis (AD) in first four years of life. Methods. Continuous crosssectional study of 69 children with an established diagnosis of AD from age 0 to 4 years was conducted. Severity was assessed according to the SCORAD scale. Clinical examination included: general and biochemical blood tests, determination of total IgE and allergenspecific IgE (IgEs) in serum against the most common epidermal and food allergens with ImmunoCAP, Phadia IDM (Sweden) technology. Results. Diagnostically significant levels of IgEs to these allergens had 68,11% of patients. Sensitization to cow’s milk was estimated in 38,98% (n=23), to chicken egg and its components in 67,92% (n=36), to chicken protein in 67,44% (n=29), to yolk in 54,28% (n=19), to cat epithelium in 47,5% (n=19) of children. Conclusion. Sensitization to food allergens marks the early development of sensitization to exogenous allergens, predominantly to the cat epithelium. Sensitization to house dust mites develops less frequently, but can also occur in children under one year of age. Sensitization to cat epithelium can accompany food sensitization as well as to be an independent cause of AD.

e20002 Background: Allergy has been studied as a risk factor for several malignancies, including childhood leukemia; yet, the tentative etiological nature of this association needs to be further explored. Published studies suffer inappropriate study design and accuracy of exposure variables. In response to the latter need, this study aims to use country specific biological markers, namely levels of the most prevalent allergen-specific immunoglobulin E (IgE) antibodies in Greece as an alternative exposure measurement to history of allergy and compare their concordance with allergic history. Methods: Allergen-specific-IgEs against 24 most prevalent inhalant and food allergens were determined for 199 incident childhood acute lymphoblastic leukemia (ALL), newly diagnosed cases across Greece and registered in the Nationwide Registry for Childhood Hematological Malignancies (NARECHEM) and 113 hospital controls. K statistic was used to check the concordance between serum IgE specific allergens and allergic history overall, as well as among cases and controls. Results: Concordance between self-reported food allergy and food IgE levels in the same individual among both cases and controls was 87% and 83% for respiratory allergens. Among cases, concordance between self reported food allergies and food IgEs was 92% and 80% for controls (p-value 0.003) and the respective κ statistics were 0.28 for cases and 0.10 for controls. Concordance between self reported respiratory allergies and respiratory IgEs was 84% for cases and 81% for controls (p-value 0.57); κ statistics 0.09 for cases and 0.07 for controls. Conclusions: Much of the discordance among cases and controls (self-report false positives) might probably be a reflection of non allergic food hypersensitivity, an allergy that was surpassed or extended allergen avoidance. Other discordance (self-report false negatives) seems to be the result of food sensitization, either hypoclinical or not acknowledged as a type of allergy by mothers of the children. Nevertheless, these measurements jointly analyzed are valuable in exploring the stated hypothesis, especially in well designed prospective studies.

Here, we analyzed differential expression of miRNA and potential target genes of ileal and jejunal lymphatic tissues fromEnterococcus faeciumNCIMB 10415-fed piglets versus untreated controls by using next-generation sequencing. We identified miR-423-5p as being greatly affected by the treatment group (2.32-fold;P= 0.014). Validation by reverse transcription-quantitative PCR (RT-qPCR) confirmed a significant upregulation of miR-423-5p (2.11-fold;P= 0.03) and, additionally, downregulation of the important immune-relevant immunoglobulin lambda light C region (IGLC) (0.61-fold;P= 0.03) and immunoglobulin kappa constant (IGKC) (0.69-fold;P= 0.04) target genes. Expression analysis of miR-423-5p and IGLC at different age points shows a clear anticorrelated relationship. Luciferase reporter assays with a HeLa cell line verified IGLC as a target of miR-423-5p. The results provided evidence for an effect of feeding ofE. faeciumon the expression of miR-423-5p and on the regulation of the IGLC gene through miR-423-5p. This might be a possible mode of action ofE. faeciumon immune cell regulation in the small intestine.

Several animal food allergens assemble into amyloids under gastric-like environments. These aggregated structures provide Gad m 1 with an enhanced immunoglobulin E (IgE) interaction due to the fibrillation of the epitope regions. However, whether these properties are unique to Gad m 1 or shared by other food allergens has not yet been addressed. Using Bos d 5, Bos d 12 and Gal d 2 as allergen models and Gad m 1 as the control, aggregation reactions and the sera of milk, egg and fish allergic patients have been analyzed, assessing the IgE interactions of their amyloids. We found that amyloids formed by Bos d 12 and Gal d 2 full-length and truncated chains are recognized by the IgEs of milk and egg allergic patient sera. As with Gad m 1, in most cases amyloid recognition is higher than that of the native structure. Bos d 5 was not recognized under any fold by the IgE of the sera studied. These results suggest that the formation of IgE-binding amyloids could be a common feature to animal food allergens.

Ascaris lumbricoides is the most common soil-transmitted helminth and infects 447 million people in impoverished areas worldwide. It causes serious morbidity including wheezing and influences various aspects of human immunity, such as type 2 innate lymphoid cells, regulatory T cell function, and acquired immunity. Thus, it is crucial to elucidate its influence on human immunity. We aimed to classify wheezing children based on their Ascaris infection intensity and other risk factors using hierarchical cluster analysis to determine the mechanisms of and the degree to which Ascaris contributes to childhood wheezing in rural Bangladesh. We analyzed relevant data collected in 2001. The participants included 219 5-year-old wheezing children who were randomly selected from 1705 children living in the Matlab Health and Demographic Surveillance area of the International Centre for Diarrhoeal Disease Research, Bangladesh. Hierarchical cluster analysis was conducted using variables of history of pneumonia, total and specific immunoglobulin E levels, Ascaris infection intensity, and parental asthma. Three distinct wheezing groups were identified. Children in Cluster 1 (n=50) had the highest titers of the total, anti-Ascaris, anti-Dermatophagoides pteronyssinus, and anticockroach IgEs and experienced the fewest episodes of pneumonia. Cluster 2 (n=114), the largest group, experienced few episodes of pneumonia and had the lowest titers of the total, anti-Ascaris, anti-Dp, and anticockroach IgEs. Cluster 3 (n=32) consisted of participants with the most episodes of pneumonia and lower titers of the total and specific IgEs. The extremely high prevalence of Ascaris infection found in Clusters 1-3 was 78%, 77%, and 72%, respectively. Childhood wheezing in rural Bangladesh could be divided into three groups, with 26% of wheezing attributable to anti-Ascaris IgE and 16% to history of pneumonia during early childhood, and 58% might have been due to Ascaris infection without elevated anti-Ascaris IgE.

Personalized therapy for female infertility means determining of immune mechanisms, including allergic sensitization towards sperm antigens. In order to determine antisperm serum IgE antibodies in women with infertility suffering from pelvic inflammatory diseases, the modified Anti-Spermatozoa Antibody protocol was used (ASA Serum ELISA, Demeditec Diagnostisc, Germany). The modification of the protocol is designed to detect only serum antisperm IgE antibodies carrying Fab fragments to sperm antigens. Allergen-specific IgEs to a common panel of 176 respiratory and food allergenic molecules were determined using the MeDALL scientific chip developed as a part of the European project. Forty patients suffering from infertility of inflammatory etiology and 16 practically healthy women of reproductive age were examined. Specific IgE antibodies towards sperm antigens were detected in blood serum in 7/40 (17.5%) patients with infertility. The maximum level of sIgE was 4 times higher than the maximal value of fertile women. No correlation with total IgE was detected. Women with sIgE-ASA complained of burning and itching immediately after coition. Systemic and long-term allergic reactions were not noted. Women with positive sIgE-ASA values were 2 times more likely to suffer from chronic recurrent vaginal dysbiosis. The presence of specific anti-sperm IgE antibodies is likely to have pathogenetic significance in female infertility, and they should be taken into consideration for creating personalized therapy approaches.

Interleukin-13 (IL-13) is a recently discovered immunoregulatory cytokine. The cellular sources of IL-13 and the regulation of its expression are largely unknown. Here we show that human basophils produce IL-13 in response to IgE-receptor (IgER) crosslinking, IL-3, IL-3 plus C5a, but not C5a alone. Human basophils express IL-13 in a restricted manner since, apart from IL-4, no other cytokines encoded on the cytokine gene cluster (IL-3, IL-5, and granulocyte macrophage-colony-stimulating factor [GM-CSF]), are induced. Highest levels of IL-13 are formed after IgE-independent activation leading to a prolonged secretion of IL-13. The response to IgER-cross-linking is more transient preferentially inducing IL-4, IL-3 is a unique cytokine regulating IL-13 production by human basophils: Among a large number of cytokines tested, only IL-3 is capable of directly inducing IL-13 expression. Furthermore, although some IL-13 is produced in response to C5a in the presence of IL-5, GM-CSF, IGF-1 or IL-1 beta, IL-3 is by far the most effective. IL-13 production was blocked by actinomycin D and cycloheximide and conditions leading to IL-13 release also lead to the induction of IL-13 mRNA. This study supports an important immunoregulatory role of human blood basophils, owing to their capacity to simultaneously express IL-13 and IL-4 in a restricted manner.

While immunoglobulin (Ig) E is a prominent biomarker for early-onset, its levels are often elevated in non-allergic late-onset asthma. However, the pattern of IgE expression in the latter is mostly polyclonal, with specific IgEs low or below detection level albeit with an increased total IgE. In late-onset severe asthma patients, specific IgE to Staphylococcal enterotoxins (se-IgE) can frequently be detected in serum, and has been associated with asthma, with severe asthma defined by hospitalisations, oral steroid use and decrease in lung function. Recently, se-IgE was demonstrated to even predict the development into severe asthma with exacerbations over the next decade. Staphylococcus aureus manipulates the airway mucosal immunology at various levels via its proteins, including superantigens, serine-protease-like proteins (Spls), or protein A (SpA) and possibly others. Release of IL-33 from respiratory epithelium and activation of innate lymphoid cells (ILCs) via its receptor ST2, type 2 cytokine release from those ILCs and T helper (Th) 2 cells, mast cell degranulation, massive local B-cell activation and IgE formation, and finally eosinophil attraction with consequent release of extracellular traps, adding to the epithelial damage and contributing to disease persistence via formation of Charcot–Leyden crystals are the most prominent hallmarks of the manipulation of the mucosal immunity by S. aureus. In summary, S. aureus claims a prominent role in the orchestration of severe airway inflammation and in current and future disease severity. In this review, we discuss current knowledge in this field and outline the needs for future research to fully understand the impact of S. aureus and its proteins on asthma.

Auf der Grundlage von spezifischen Laborparametern (Hämatokrit, aktueller Basenüberschuss, Natrium-, Chloridkonzentration) erfolgte in Abhängigkeit von der Herzfrequenz (HF) die Berechnung von Infusionsrichtmengen in Form von 0,9%iger Natriumchlorid- und 8,4%iger Natriumbikarbonatlösung, die im Rahmen einer Notfallbehandlung durch den praktischen Tierarzt bei diesen Patienten erforderlich sind. Pferde mit Krampfkolik und Obstipation der LVLL wiesen unabhängig von der HF physiologische Werte auf und bedürfen in der Regel keiner intravenösen Applikation von Flüssigkeit. Pferde mit Dislocatio coli zeigten ab einer HF von >60/min ein Flüssigkeitsdefizit von drei bis sieben Litern und ein Basendefizit von 250 mmol. Die Elektrolyte Natrium und Chlorid waren unverändert. Pferde mit Torsio coli totalis benötigen mindestens sieben Liter Flüssigkeit, bei einer HF von >80/min 20-27 Liter und 500-1800 ml 8,4%ige Bikarbonatlösung zum Ausgleich von Dehydratation, metabolischer Azidose und zur Verhinderung eines akuten Nierenversagens. Bei Pferden mit Dünndarmstrangulation ist eine Infusion von mindestens fünf Litern erforderlich, bei einer HF von >80/min die Verabreichung von sieben bis 20 Litern isotone Natriumchloridlösung und 400-1000 mmol Bikarbonat. Pferde mit akuter Kolitis benötigen unabhängig von der HF mindestens sechs bis 17 Liter 0,9%ige Natriumchloridlösung und bei einer HF von >70/min bis zu 1100 mmol Bikarbonat.

The present paper describes the design of a HtTA (heterotetravalent allergen) as a multi-component experimental system that enables an integrative approach to study mast cell degranulation. The HtTA design allows presentation of two distinct haptens, each with a valency of 2, thereby better reflecting the complexity of natural allergens by displaying epitope heterogeneity and IgE antibody variability. Using the HtTA design, synthetic allergens HtTA-1 and HtTA-2 were synthesized to model a combination of epitope/IgE affinities. HtTA-1 presented DNP (2,4-dinitrophenyl) and dansyl haptens (Kd=22 and 54 nM for IgEDNP and IgEdansyl respectively) and HtTA-2 presented dansyl and the weak-affinity DNP-Pro (DNP-proline) haptens (Kd=550 nM for IgEDNP). Both HtTAs effectively induced degranulation when mast cells were primed with both IgEDNP and IgEdansyl antibodies. Interestingly tetravalent DNP-Pro or bivalent dansyl were insufficient in stimulating a degranulation response, illustrating the significance of valency, affinity and synergy in allergen–IgE interactions. Importantly, maximum degranulation with both HtTA-1 and HtTA-2 was observed when only 50% of the mast cell-bound IgEs were hapten-specific (25% IgEdansyl and 25% IgEDNP). Taken together, results of the present study establish the HtTA system as a physiologically relevant experimental model and demonstrates its utility in elucidating critical mechanisms of mast cell degranulation.

Before insects can be used widely as an alternative source of dietary protein, their allerginicity should be investigated. Therefore, the aim of our study was to assess the potential adverse reactions of the immune system of dogs against Tenebrio molitor proteins. Dogs sensitised to storage mites T. putrescentiae and A. siro were included. Clinically healthy and clinically allergic dogs were compared. Proteins were extracted from mealworm larvae and their digestibility determined by in vitro incubation with digestive proteases. Mealworm protein extracts and digests were analysed by SDS–PAGE. Canine sera tested for the presence of mite-specific IgEs were used for subsequent Western blotting. LC-MS/MS analysis was used to identify mealworm proteins and their allergenic potential was predicted with the AllermatchTM tool. The binding of canine sera IgEs to mealworm proteins was confirmed; however, the differences between the two groups of dogs were not significant. Moreover, no clear correlation was found between sensitisation to storage mites and clinical status of the dogs. Altogether, 17 different proteins were identified, including tropomyosin, α-amylase, and Tm-E1a cuticular protein that are known cross-reacting IgE-binding allergens. Our results suggest that dogs allergic to mites may clinically express also the cross-reactivity with mealworm proteins.

To study the sensitization to inhalation allergens Dermatophagoides pteronyssinus (Der. p.) and Dermatophagoides farinaе (Der. f.) in Moscow and the Moscow region in 2010–2019 and to identify the structure, features and dynamic changes in sensitization to Der. p. and Der. f. based on the analysis of the allergosensitivity profile of patients with IgE-mediated allergic diseases (IgEA).Materials and methods. Blood serum samples of Moscow and the Moscow region residents with IgEA (n = 2849) were examined by RIDA AllergyScreen method in 2010–2019.Results. According to the results of the study spanning the last ten years, about 18–21% of patients with IgEA in Moscow and the Moscow region have sensitization to Der. p. and/or Der. f. There was a statistically significant decrease in sensitization to Der. p. from 2017 to 2019. House dust mites of the type Der. f. more often cause a higher level of sensitization compared to Der. p.Discussion. The data obtained indicate the change in the sensitization of patients to Pyroglyphidae, namely Der. p. against the background of changes in the housing acarocomplex of Moscow and the Moscow region. Features of protein molecules of these species of dust mites may contribute to their varying degrees of the impact on immunological reactivity in the population studied.Conclusions. The decrease in the prevalence of Der. p. is most likely accounted for the changes in the ecosystem of homes of residents of Moscow and the Moscow region.

ZusammenfassungDie Autoren berichten über Methodik, Ergebnisse und Komplikationen bei der Verödungsbehandlung in den vergangenen 12 Jahren bei über 8000 Behandlungen an über 2700 Patienten. Methoden: Neben der klinischen Untersuchung war die Dopplersonographie kombiniert mit der (Farb-)Duplexsonographie fester Bestandteil der Diagnostik. Von wenigen Ausnahmen abgesehen, wurde die Stammvarikose der Vena saphena magna und parva immer einer möglichst begrenzten varizenchirurgischen Intervention zugeführt. Eine postoperativ verbleibende Varikose wurde mit der Sklerotherapie behandelt. Dabei wurden groß- und kleinkalibrige Seitenäste mit mehreren besonders kurzen Kanülen am stehenden Patienten punktiert, der Patient anschließend auf der Kippliege in eine liegende Position gebracht und Aethoxysklerol® (2, 3 und 4% je nach Kaliber der Varize) injiziert (Technik der »offenen Nadel«). Retikuläre Varizen wurden mit 1%igem, Besenreiser mit 0,5%igem Aethoxysklerol® gemischt mit 10%iger NaCl (1: 2) am liegenden Patienten mit vielfacher direkter Injektion behandelt (Technik der »geschlossenen Nadel«). Bei allen Patienten wurde ein Kompressionsverband bis zum Oberschenkel angelegt und eine sofortige Mobilisierung durchgeführt. Als Nachbehandlung trugen die Patienten für 6 Wochen einen Schenkelkompressionsstrumpf der Klasse 2. Ergebnisse: Mit dieser Methode hatten wir nach 6 Wochen gute Ergebnisse in 82%, mäßige in 14% und schlechte (Varize nicht verödet) in 3%. Fast alle nicht obliterierten Varizen konnten erfolgreich nachverödet werden. Die Verödung von Astvarizen war erfolgreicher als die von Besenreisern und retikulären Varizen. Als schwerste Komplikation ereignete sich eine tödliche Lungenembolie. Weitere schwere Komplikationen waren zwei ausgedehnte tiefe Beinvenenthrombosen, zwei (größere) Hautnekrosen und zwei aszendierende Magnaphlebitiden (je 0,02% der Behandlungen). Schlussfolgerung: Verglichen mit den Literaturdaten ist die vorgestellte Methode als besonders risikoarm einzustufen.

Background The present report was a follow-up investigation at 2.5- and 7-year intervals of a previous study of 20 children with moderate-to-severe immunoglobulin E (IgE) mediated hen's egg (HE) allergy who received oral immunotherapy (OIT) with raw HE. The study design of the previous study divided the 20 subjects into two groups of 10 each: (1) group 1, the OIT group (OIT-G), and, (2) group 2, an age-matched control group (C-G). In that study, 8 of 10 of the children in the OIT-G were successfully desensitized, one child was partially desensitized, and desensitization failed in one child. The aims of the present study were to evaluate the long-term effectiveness and safety profile of OIT with raw HE, and to assess the course and prognostic value of skin-prick tests (SPT) and serum-specific HE-IgEs in this study population. Methods Of the 20 children who were recalled, 2 dropped out, which left 18 to be evaluated. Information on their HE intake was recorded, and SPTs with HE allergen extracts and with raw and hard-boiled HE were performed. Ovomucoid- and ovalbumin-specific IgE levels were also measured. Results At the first (2.5-year) and second (7-year) follow-ups, 87.5% of the children in the OIT-G who tolerated raw HE were still tolerant, whereas the children in the C-G were significantly less tolerant. Overall, cutaneous sensitivity to HE significantly decreased after the 6-month desensitization period and at both follow-ups with regard to the OIT-G but not with regard to the C-G. A significant reduction in serum ovomucoid- and ovalbumin-specific IgE levels was seen in both the OIT-G and the C-G. Conclusion Clinical raw HE tolerance induced by OTT persists over time. Negativization of SPTs could be considered a more reliable prognostic indicator of clinical tolerance to raw HE than the reduction in specific-HE IgE levels. Raw-HE OIT would seem to be a promising method to treat HE allergy.

IgE-mediated mast cell degranulation and excessive Th2 cells activation are major features of various allergic diseases.Sohamhyoong-tanghas been reported to have anti-inflammatory and antibacterial effects. In this study, we investigated the inhibitory effect ofSohamhyoong-tangextract (SHHTE) on allergic symptoms and inflammatory responses in ovalbumin- (OVA-) sensitized BALB/c mice. The mice were sensitized with OVA and alum at 2-week intervals and then orally given SHHTE for 13 days followed by intradermal OVA injection. Administration of SHHTE significantly reduced edema formation and inflammatory-cell infiltration in ear tissues. Total and OVA-specific IgEs as well as proinflammatory cytokine TNF-αand Th2-associated cytokine IL-4 levels were lower in the SHHTE-treated group than in the vehicle. SHHTE treatment significantly suppressed both mRNA and protein levels of IL-4 and IL-5 in OVA-stimulated splenocytes. SHHTE decreased Th1 (IFN-γ) and Th17 (IL-17a) cytokine mRNA expression but increased Treg cytokines (IL-10 and TGF-β1). Moreover, SHHTE significantly inhibited degranulation of RBL-2H3 cell line in a dose-dependent manner. Thus, SHHTE efficiently inhibited the allergic symptoms in an OVA-sensitized mouse model and its action may correlate with the suppression of IgE production by increasing IL-10 and TGF-β1, which can limit the function of other T helper cells and prevent the release of inflammatory mediators from mast cells. These results suggest that SHHTE could be a therapeutic agent for treating various allergic diseases.

Abstract:
This study aimed to determine the incidence and the prognosis of incidental carcinoma of the gallbladder (IGBC) after cholecystectomy through a meta-analysis. This meta-analysis included 51 studies and 436,636 patients with cholecystectomy. The incidence rate of IGBC after cholecystectomy was 0.6% (95% confidence interval (CI) 0.5–0.8%). The incidence rate of recent studies was not significantly different from those of past studies. The mean age and female ratio of the IGBC subgroup were not significantly different from those of the overall patient group. The estimated rates of IGBC were 13.0%, 34.1%, 39.7%, 22.7%, and 12.5% in the pTis, pT1, pT2, pT3, and pT4 stages, respectively. Patients with IGBC had a favorable overall survival rate compared to patients with non-IGBC (hazard ratio (HR) 0.574, 95% CI 0.445–0.739). However, there was no significant difference of disease-free survival between the IGBC and non-IGBC subgroups (HR 0.931, 95% CI 0.618–1.402). IGBC was found in 0.6% of patients with cholecystectomy. The prognosis of patients with IGBC was favorable compared to those with non-IGBC. In the pathologic examination after cholecystectomy for benign diseases, a sufficient examination for histology should be guaranteed to detect IGBC.

Integrated gasification combined cycle (IGCC) is an efficient, low-pollution clean coal power generation technology and efficient use of IGCC technology is helpful to realize clean conversion and comprehensive utilization of coal resources in China. This paper analyzed the factors that influence the power supply efficiency of IGCC power plants, determined assessment model for power supply efficiency of IGCC power plants. Then based on IGCC power plant construction specific data including power supply efficiency, combining the general technical and economic evaluation of the project, the technical and economic evaluation step of IGCC plant was determined. The economic evaluation of IGCC power plant was carried out as well as sensitivity analysis in terms of static total investment, utilization hours and standard coal tax price. The results showed that introduced IGCC of 40 megawatt had higher tariff and utilization hours had maximum impact on tariff.
