Abstract:

Zusammenfassung Hintergrund Trotz der Verbesserung der Diagnosetechnologien bleibt die Detektion von intraokularen Fremdkörpern (IFK), die sich in der Projektion des Ziliarkörpers befinden, eines der aktuellen Probleme der Augentraumatologie. Ziel Untersuchung der möglichen Visualisierung intraokularer Fremdkörper (IFK) in der Projektion des Ziliarkörpers mithilfe der infrarоten transpalpebralen Transillumination (IRTT). Materialien und Methoden Analyse von 10 Personen (10 Augen) mit einer penetrierenden Augenwunde und nachgewiesenem IFK. In allen Fällen erfolgten Röntgendiagnostik, Ultraschalluntersuchung des Auges und IRTT. Ergebnisse Bei allen Patienten wurden Schatten von den Strukturen des Ziliarkörpers (Pars plana, Pars plicata) auf der Sklera mittels IRTT und Schatten von IFK visualisiert. Schlussfolgerung Die Technik der IRTT bei Patienten mit penetrierenden Augenverletzungen ermöglicht die nicht invasive Visualisierung von IFK in der Projektion des Ziliarkörpers und die Identifizierung ihrer Lokalisation.

A delayed discrete equationΔu(k+n)=−p(k)u(k)with positive coefficientpis considered. Sufficient conditions with respect topare formulated in order to guarantee the existence of positive solutions ifk→∞. As a tool of the proof of corresponding result, the method described in the author's previous papers is used. Except for the fact of the existence of positive solutions, their upper estimation is given. The analysis shows that every positive solution of the indicated family of positive solutions tends to zero (ifk→∞) with the speednot smaller than the speed characterized by the functionk·(n/(n+1))k. A comparison with the known results is given and some open questions are discussed.

Abstract:

Traditional fermentation of dried seeds ofParkia biglobosais used to produce a local product called ‘iru’. It is a condiment consumed mostly in Western Nigeria. In this study, the effect ofBacillussubtilisBC4333starter cultures and different additives, such as local softening agents called ‘kuuru’,Hibiscus sabdariffaseed cotyledon and ash from cashew (Anacardium occidentals) plants as on the microbiological, physicochemical, and enzymatic properties of fermentedParkia biglobosaseeds were investigated. The dried seeds ofParkia biglobosawere processed by boiling the cotyledons withHibiscus sabdariffa,‘kuuru’ andAnacardium occidentals(cashew plant) ash using natural fermentation andBacillus subtilisstrainBC4333as a starter culture. The fermentations were carried out at 35°C for 36 hrs. Commercially fermented Parkiabiglobosaseed (called ‘iru-pete’) was used as control. All the samples were analyzed for microbial, physico-chemical and enzymatic properties using standard biochemical techniques. ‘Iru’ produced using ‘kuuru’ (IFK) had the highest microbial count of 7.88 log CFU/g, while the microbial count of íru’ produced with ash (IFA) and íru’ produced with dried seeds ofHibscus sabdariffa(IFH) were 7.69 log CFU/g and 7.75 log CFU/g respectively. The pH of the fermented products ranged between 7.06 and 7.91, while the unfermented sample (UFS) had pH 6.68. The unfermented sample had the highest total titratable acidity (TTA) of 2.6×10-2N while the ‘iru’ samples produced using ‘kuuru’(IFK) had the least TTA of 1.0×10-2N. Fermentation significantly increased the moisture content of the products. Starter culture fermented ‘iru’ (F14) had the highest protease, amylase and phytase activity of 232.3UPR, 0.41UAM and 71.85UPH respectively. Findings from this study suggest that ‘iru’ produced by addition ofBacillus subtilisstrain BC4333 had the best desirable biochemical qualities when compared to the other test additives and thus recommened for commercial scale.

AbstractWe consider total well-founded orderings on monadic terms satisfying the replacement and full invariance properties. We show that any such ordering on monadic terms in one variable and two unary function symbols must have order typeω,ω2orωω. We show that a familiar construction gives rise to continuum many such orderings of order typeω. We construct a new family of such orderings of order typeω2, and show that there are continuum many of these. We show that there are only four such orderings of order typeωω, the two familiar recursive path orderings and two closely related orderings. We consider also total well-founded orderings onNnwhich are preserved under vector addition. We show that any such ordering must have order typeωkfor some 1 ≤k≤n. We show that ifk<nthere are continuum many such orderings, and ifk=nthere are onlyn!, then! lexicographic orderings.

Abstract:

This study investigates the meaning of the Qur'ānic near-synonyms الإفك al-'ifk and الكذب al-kadhib (lie) and their English translation. It aims to identify the contextual meaning of these Qur'ānic near-synonyms based on the exegeses of the Holy Qur'ān. Then, it explains the nuances that exist between these two lexical items in terms of denotative meaning. This study also examines how the semantic differences between these Qur'ānic near-synonyms are reflected in the English translation of the Holy Qur'ān by Ali (2006). This study adopts the RC-S approach by Murphy (2003) as a theoretical framework for data analysis. It also employs the qualitative approach to collect and analyze the data of the study. The exegeses of the Holy Qur'ān are consulted to identify the semantic differences between the Qur'ānic near-synonyms. The findings reveal that there exist some differences in meaning between these Qur'ānic near-synonyms in terms of denotative meaning and that the semantic differences between these Qur'ānic near-synonyms are not reflected in Ali's English translation of the Holy Qur'ān. The study provides recommendations for readers and translators especially the translators of the Holy Qur'ān.

Abstract:

This article discusses the motives for wearing a face veil in early Islam. With the lens of prophetic historical reflection, it focuses on two narratives of the hadith on veil: the Arab women who sold in the market of Bani Qaynuqa‘ and ‘Aishah on hadith al-ifk. This article argues that the motives of covering their faces has close related to the situation and conditions occurred at the time. Historically, the two stories explained that wearing a face veil as a protection of dignity and self-esteem from an uncomfortable milieu. Sociologically, the face veil was cultural habits in guarding women from any social disorders and negative treatment. While psychologically, wearing a face veil symbolized the purity of an honor and the high value of personality embedded in the dignity of women so as not to invite a slander.

<p><em>This paper seeks to parse the solution to the problem of hoax from the perspective of Quranic interpretation holistically and comprehensively. This study is a qualitative research by reading the relationship (munasabah) between verses about hoax in the Holy Quran that are represented by the terms ifk, fa>siq, muna>fiq, murjifu>n, and tabayyun. After reading the verses related to the term, it was concluded that hoax news can be minimized by critical thinking, having emotional maturity, performing tabayyun, and broadening insight. In addition, the Holy Quran also teaches good communication ethics, namely qaulan sadi>dan (correct speech), qaulan bali>gan (good words that leave an imprint on the soul), qaulan maisu>ran (proper speech), qaulan layyinan (soft words), qaulan kari>man (noble words), and qaulan ma'ru> fan (good words).</em></p><p> </p>

Uzvišeni Allah, dž. š., šaljući Poslanike na zemlju, odgajao ih je, kao i njihove sljedbenike kroz upečatljive događaje i iskušenja iza kojih je davao odgojne mjere i koristi koje se nikada neće zaboraviti. Nije poslao ni jednog od poslanika, a da ga nisu optužili da je luđak, ludi pjesnik, čarobnjak, prorok i sl., pa su tako i nevjernici, politeisti, idolopoklonici Kurejša, na koncu svih poslaničkih misija, govorili za Muhammeda, a. s., čekajući priliku da mu na bilo koji način mogu naštetiti, napakostiti fizički ili moralno: njegovom ugledu i moralu ili njegovoj porodici. Samo jedan u nizu od tih napada jeste događaj ,,ifka-a“ (hadisetul-ifk) u kojem su htjeli da Allahovog Poslanika, a. s., degradiraju moralno i psihički sa potvorom i objedom na njegovu ženu, majku muslimana, Aišu, r. a., kćerku Ebu-Bekra es-Siddika Istinoljubivog. Nakon mjesec dana strahovitih muka i neprijatnosti, koje su se odrazile ne samo na Allahova Poslanika i Aišu, r. a., njenu porodicu, nego i na čitavo novoformirano muslimansko društvo i ashabe, Uzvišeni Allah će poslati Objavu kojom dokazuje nevinost Aiše, r. a., i kojom daje odgojne mjere i smjernice kojima će dalje kročiti muslimansko društvu nakon ovih strašnih događaja, ali će ukazati i na koristi svih ovih događanja kroz obznanjivanje i otkrivanje nečega što muslimani nisu znali, a to je da među njima ima prikrivenih licemjera koji su u njihovoj blizini i među njima. Dakle, ima onih koji nisu iskreni vjernici. Ovaj događaj će raslojiti muslimansko društvo na dvije kategorije: na iskrene vjernike koji su se držali istine i na one koji nisu, nego su se pretvarali da su vjernici i svoj prikriveni munafikluk i licemjerstvo koristili za napade na vjeru i vjernike.

Zusammenfassung Hintergrund In Ländern mit Direktzugang zur Physiotherapie wählen Physiotherapeuten autonom das bevorzugte Heilmittel zur Behandlung ihrer Patienten. Anders gestaltet sich die Situation in Deutschland: Hier verordnen die Ärzte das zu verwendende Heilmittel. Ziel Im Rahmen des Modellvorhabens Physiotherapie gemäß § 63 Abs. 3b SGB V wird (unter anderem) evaluiert, welche Heilmittel bei Behandlungsautonomie in welcher Frequenz und Dauer angewendet werden und welche Auswirkungen diese Wahl auf die Behandlungsergebnisse der Patienten hat. Methode Eine randomisierte kontrollierte Studie vergleicht die herkömmliche Behandlung nach ärztlicher Verordnung mit der Behandlung im neuen Versorgungsmodell hinsichtlich Wahl der Heilmittel, Behandlungsdauer und -frequenz sowie den Behandlungsergebnissen Patientenzufriedenheit (Fragebogen ZUF-8), Schmerz (visuelle Analogskala, VAS), gesundheitsbezogene Lebensqualität (Fragebogen EuroQol) und Funktion (Back Performance Scale [BPS], Roland-Morris Disability Questionnaire [RMDQ], Lower Extremity Functional Scale [LEFS]). Ergebnisse Am Modellvorhaben nahmen 630 Patienten teil, davon 296 in der Kontroll- (KG) und 334 in der Modellgruppe (MG). In der MG wendeten die Physiotherapeuten häufiger (in 33 % der Fälle) Allgemeine Krankengymnastik (AKG) und/oder Manuelle Therapie (MT) zusammen mit einem ergänzenden Heilmittel (z. B. Wärmetherapie-Fango) an (vs. 10 % in der KG; x2 = 51,525, p < 0,001) sowie häufiger eine Kombination aus AKG mit MT (in 32 % der Fälle vs. 3 % in der KG, x2 = 100,455, p < 0,001). Die Behandlungsdauer war in der MG mit 9,7 Wochen (Standardabweichung [SD] = 13,2) kürzer als in der KG mit 11,8 Wochen (SD = 5,4; T = 2,542, p < 0,05). Weitgehend unabhängig von ihrer Zugehörigkeit zur KG oder MG, berichteten die Patienten eine hohe Zufriedenheit und signifikante Verbesserungen hinsichtlich Schmerz, Lebensqualität und Funktion (Back Performance Scale [BPS], Roland-Morris Disability Questionnaire [RMDQ], Lower Extremity Functional Scale [LEFS]). Schlussfolgerung Bei Behandlungsfreiheit entschieden sich Physiotherapeuten vermehrt für eine Kombination von aktiven und passiven Behandlungstechniken, ohne die Behandlungsdauer auszudehnen. Dieses Vorgehen ist mit international anerkannten Behandlungsleitlinien konform.

ABSTRACT A primer extension inhibition (toeprint) assay was developed using ribosomes and ribosomal subunits from Streptomyces lividans. This assay allowed the study of ribosome binding to streptomycete leaderless and leadered mRNA. Purified 30S subunits were unable to form a ternary complex on aph leaderless mRNA, whereas 70S ribosomes could form ternary complexes on this mRNA. 30S subunits formed ternary complexes on leadered aph and malE mRNA. The translation initiation factors (IF1, IF2, and IF3) from S. lividans were isolated and included in toeprint and filter binding assays with leadered and leaderless mRNA. Generally, the IFs reduced the toeprint signal on leadered mRNA; however, incubation of IF1 and IF2 with 30S subunits that had been washed under high-salt conditions promoted the formation of a ternary complex on aph leaderless mRNA. Our data suggest that, as reported for Escherichia coli, initiation complexes with leaderless mRNAs might use a novel pathway involving 70S ribosomes or 30S subunits bound by IF1 and IF2 but not IF3. Some mRNA-ribosome-initiator tRNA reactions that yielded weak or no toeprint signals still formed complexes in filter binding assays, suggesting the occurrence of interactions that are not stable in the toeprint assay.

Gydymo išangės fistulės kamščiu sėkmingumas – nuo 15,6 % iki 88 %. Iš 551 IFK gydytų ligonių 300 (54,4 %) pasveiko. Kriptoglanduliarinės kilmės (K) fistulės buvo sėkmingai išgydytos 55,1 % atvejų, o įvairios kilmės (I) – 54,9 %. Neaptikome patikimo ryšio tarp sėkmingo gydymo rezultatų ir paciento lyties, amžiaus, fistulės ilgio ar kilmės. Išvados: Išangės fistulės kamščio metodas yra patikimas gydyti sudėtingas fistula-in-ano. Ateities tyrimai turėtų padėti aiškiau apibrėžti gydymo išangės fistulės kamščiu metodo vietą išangės fistulių chirurgijoje. Methods: A total of 31 studies, published since 2006 to 2010 were extracted and 19 of them were finally included in this systematic review. 8 were retrospective case series, 9 prospective studies, 1 prospective non-randomized controlled trial and 1 randomized controlled trial. In these 19 studies, a total of 551 patients were included, who were treated with anal fistula plug. Results: The follow-up period ranged from 3–24 months. The AFP procedure had a success rate ranging from 15.6–88 %. From 551 patients treated with AFP, 300 (54.4 %) were cured. The success rate in patients with cryptoglandular origin (K) was 55.1 % and the success rate in patients with fistulas of other origin was 54.9 %. We did not notice difference in success rate between age, gender, fistula anatomy and etiology. Conclusion: The anal fistula plug method is a reliable method in treat of complex fistula-in-ano. Future studies should help to clarify the place of anal fistula plug in anal fistula surgery. Key words: Fistula-in-ano, anal plug, recurrence, advancement flap.

The development of technology has been shifted human’s way from the manual era to the digital era. Humans have been able to communicate without border and time. However, information in the modern era is not always true and many of them are false or hoaxes. Al Qur’an as a guide for humans has actually explained the hoax. Some words in the Al Qur'an illustrated the context of hoaxes such as ifk, kaẑaba, fitnah, khud'a, qaul al-zūr, buhtan, iftara, tahrif, and garur. In this paper, it will discuss the text of the hoax in the Al Qur’an in its text and textual verses when the verse is revealed. Furthermore, the context of the hoax in the Al Qur’an is explained in the context of the current issues with a semantic approach. This paper concludes that the meaning of the hoax in the Al Qur'an is in accordance with the recent context but only the media and problems are more complex. Furthermore, the Qur'an has provided solution to the issues of hoax such as checking the correctness of the data before sharing (Q. S Al-Hujurat: 6, Q. S Isra: 36), increasing the culture of literacy (Q. S Al-Qalam: 1 -2), awareness of prejudice (Q. S Al-Hujurat: 12), and so on.

Unicellular tubes are components of internal organs and capillaries. It is unclear how they meet the architectural challenge to extend a centered intracellular lumen of uniform diameter. In an RNAi-based Caenorhabditis elegans screen, we identified three intermediate filaments (IFs)—IFA-4, IFB-1, and IFC-2—as interactors of the lumenal membrane-actin linker ERM-1 in excretory-canal tubulogenesis. We find that IFs, generally thought to affect morphogenesis indirectly by maintaining tissue integrity, directly promote lumenogenesis in this capillary-like single-cell tube. We show that ERM-1, ACT-5/actin, and TBB-2/tubulin recruit membrane-forming endosomal and flux-promoting canalicular vesicles to the lumen, whereas IFs, themselves recruited to the lumen by ERM-1 and TBB-2, restrain lateral vesicle access. IFs thereby prevent cystogenesis, equilibrate the lumen diameter, and promote lumen forward extension. Genetic and imaging analyses suggest that IFB-1/IFA-4 and IFB-1/IFC-2 polymers form a perilumenal triple IF lattice, sandwiched between actin and helical tubulin. Our findings characterize a novel mechanism of capillary-like lumenogenesis, where a tensile trilayered cytoskeletal endotube transforms concentric into directional growth.

Perkembangan wilayah pesisir Teluk Kendari cukup pesat dengan berbagai macam aktivitas baik berupa jasa kelautan seperti pelabuhan untuk pelayaran dan perikanan maupun kegiatan-kegiatan di sekitar pantai seperti pemukiman, industri, usaha dan pertambakan. Adanya logam berat di perairan, berbahaya baik secara langsung terhadap kehidupan kerang darah (Anadara. granosa) dan kerang bakau (Polymesoda bengalensis), maupun efeknya secara tidak langsung terhadap kesehatan manusia. Penelitian ini bertujuan untuk menganalisis kandungan logam berat Pb dan Zn pada air, sedimen, dan jaringan kerang dan mengkaji faktor bioakumulasi logam berat Pb dan Zn pada dua jenis kerang. Sampel air, sedimen dan kerang diambil pada kedua lokasi pengambilan kerang, tiap lokasi 3 titik pengambilan dengan 3 kali ulangan analisis. Hasil Penelitian menunjukkan bahwa logam Pb dan Zn di air pada tiap lokasi telah melampau baku mutu, kadar tertinggi masing-masing 0,018 mg/L dan 0,793 mg/L, sedangkan pada sedimen tertinggi masing-masing 0,823 mg/Kg dan 6,919 mg/Kg, dan pada jaringan kerang menunjukan hasil bahwa kerang ukuran besar mengandung logam Pb dan Zn lebih tinggi, masing-masing 1,750 dan 9,863 mg/Kg. (sedimen dan kerang belum melampau baku mutu). Nilai BCFo-w logam Pb tertinggi 119,20 pada kerang bakau dan Zn tertinggi 35,99 pada kerang darah. Berdasarkan kategori nilai IFK (BCF) untuk logam Pb dan Zn termasuk dalam kategori rendah hingga sedang.

Based on the activity plan report of ORI Difteri 2018, it is known that the logistics needs of some vaccines do not comply with the allocation received by Jember regency of the province. This condition This was a descriptive study using questionaire and observations data as well as the documentation study of the annual Report of Health Pharmacy Installation (IFK) year 2019. Respondent to this study were immunization officers at primary health care of Jember District. The result of cold chain management evaluation is known that the most are good enaugh. In some primary health care are still hampered insufficient means so that the maintenance of vaccine is still not optimalBased on the result that cold chain management evaluation, the knowledge officer on cold chain management is still lacking good. In the some primary health care, it is known that the equipment canstraints are unavailable. While in the conformity of temperature only one primary helath care is not recommended. And stirage activities are mostly in accordance with SOP.

Abstract:

This study aimed to examine and explain the passages or propositions for the inquiry of veil use, some opinions of leading scholars, and to correct or refute some weak and controversial statements and interpretations related to veil and its law. This research employed qualitative-descriptive approach with content analysis technique and library research. The result shows that the veil is prescribed in Islam. This is based on the word of Allah in the Quran surah Al-Ahzab verse 59 which is interpreted by commentators among the companions and tabi'in as a command to cover the face. In the long story of ‘ifk, it is also mentioned that Aisyah covered her face. The scholars of schools (Hanafiyyah, Malikiyyah, Syafiiyyah, and Hanabilah), even though they disagree whether covering the face is obligatory or sunnah, they agree that veil is something recommended, specifically at the time when it is feared of triggering tribulation in society due to showing off the faces of women. Some commentators also hold that they must cover their faces as they emphasize in their commentaries. Some interpretations or statements related to veil as well as its law are weak. For example, the interpretation that the use of veil that prevailed in Arab societies was a tradition for certain societies is such a fallacious statement. Considering the existing history, the previous Arab communities which were commonly called the jahiliyyah community were ignorant about covering, let alone the veil.

Abstract:

Milk from different cattle breeds can present different casein and fat contents, which are reflected in different cheese yields (CY). However, CY is also related to some breed-related molecular characteristics. The aim of the present work was to quantify the effect of these characteristics by comparing a series of Parmigiano Reggiano (PR) cheese-making trials made with milks from Italian Brown (IB) and Italian Friesian (IF) cattle herds. Twelve trials were carried out in a cheese factory in one year (one trial per month), each one consisting of four vats processed in parallel: three vats contained milk from three different IF cattle herds (IF1, IF2 and IF3) and one contained milk from a single IB cattle herd. A 24-h CY prediction formula was developed with data from IF1, IF2 and IF3 trials (calibration) and successively validated by applying it to 12 PR trials made with IF milk in six different cheese factories (external validation). The predicted values of 24-h CY were no different to the actual ones in both calibration and external validation. Finally, the formula was tested on trials made with IB milk. In this case, the predicted values were lower than the actual ones. The quantity of IF milk casein necessary to give the same CY of IB milk was 0.20 g/100 g.

Initiation of translation involves the assembly of a ribosome complex with initiator tRNA bound to the peptidyl site and paired to the start codon of the mRNA. In bacteria, this process is kinetically controlled by three initiation factors—IF1, IF2, and IF3. Here, we show that deletion of helix H69 (∆H69) of 23S rRNA allows rapid 50S docking without concomitant IF3 release and virtually eliminates the dependence of subunit joining on start codon identity. Despite this, overall accuracy of start codon selection, based on rates of formation of elongation-competent 70S ribosomes, is largely uncompromised in the absence of H69. Thus, the fidelity function of IF3 stems primarily from its interplay with initiator tRNA rather than its anti-subunit association activity. While retaining fidelity, ∆H69 ribosomes exhibit much slower rates of overall initiation, due to the delay in IF3 release and impedance of an IF3-independent step, presumably initiator tRNA positioning. These findings clarify the roles of H69 and IF3 in the mechanism of translation initiation and explain the dominant lethal phenotype of the ∆H69 mutation.

SUMMARY Valuable information on translation initiation is available from biochemical data and recently solved structures. We present a detailed description of current knowledge about the structure, function, and interactions of the individual components involved in bacterial translation initiation. The first section describes the ribosomal features relevant to the initiation process. Subsequent sections describe the structure, function, and interactions of the mRNA, the initiator tRNA, and the initiation factors IF1, IF2, and IF3. Finally, we provide an overview of mechanisms of regulation of the translation initiation event. Translation occurs on ribonucleoprotein complexes called ribosomes. The ribosome is composed of a large subunit and a small subunit that hold the activities of peptidyltransfer and decode the triplet code of the mRNA, respectively. Translation initiation is promoted by IF1, IF2, and IF3, which mediate base pairing of the initiator tRNA anticodon to the mRNA initiation codon located in the ribosomal P-site. The mechanism of translation initiation differs for canonical and leaderless mRNAs, since the latter is dependent on the relative level of the initiation factors. Regulation of translation occurs primarily in the initiation phase. Secondary structures at the mRNA ribosomal binding site (RBS) inhibit translation initiation. The accessibility of the RBS is regulated by temperature and binding of small metabolites, proteins, or antisense RNAs. The future challenge is to obtain atomic-resolution structures of complete initiation complexes in order to understand the mechanism of translation initiation in molecular detail.

Abstract:

Abstract Initiation factor IF3 is an essential protein that enhances the fidelity and speed of bacterial mRNA translation initiation. Here, we describe the dynamic interplay between IF3 domains and their alternative binding sites using pre-steady state kinetics combined with molecular modelling of available structures of initiation complexes. Our results show that IF3 accommodates its domains at velocities ranging over two orders of magnitude, responding to the binding of each 30S ligand. IF1 and IF2 promote IF3 compaction and the movement of the C-terminal domain (IF3C) towards the P site. Concomitantly, the N-terminal domain (IF3N) creates a pocket ready to accept the initiator tRNA. Selection of the initiator tRNA is accompanied by a transient accommodation of IF3N towards the 30S platform. Decoding of the mRNA start codon displaces IF3C away from the P site and rate limits translation initiation. 70S initiation complex formation brings IF3 domains in close proximity to each other prior to dissociation and recycling of the factor for a new round of translation initiation. Altogether, our results describe the kinetic spectrum of IF3 movements and highlight functional transitions of the factor that ensure accurate mRNA translation initiation.

Abstract:

One of the bottlenecks in relation to the production of forest seedlings is irrigation, especially in less-tech nurseries. The small volumes of substrate, where the seedlings develop, lead to low water storage. This fact can lead to water deficit and significant losses in the production of seedlings, generating even mortality. The objective of this study was to determine the best irrigation frequencies for Eucalyptus grandis seedling production in less-tech nurseries. The experiment was conducted between 2013/08/04 and 2013/23/07 (106 days) and conducted in Chapadão do Sul county, Brazil. The experiment was set up in a randomized complete block design, with five replications. Four irrigation frequencies were tested: IF1 (one daily irrigation - 11:00 a.m.), IF2 (two daily irrigations - 11:00 a.m. and 7:00 p.m.), IF3 (three daily irrigations - 07:00 a.m., 11:00 a.m. and 7:00 p.m.) and IF4 (four daily irrigations - 07:00 a.m., 11:00 a.m., 3:00 p.m. and 7:00 p.m.). The sample units were composed of eight seedlings in 50 cm3 conical tubes filled with soil and vermiculite in a volume ratio of 1:1. The irrigation depth was estimated by reference evapotranspiration (Penman-Monteith) multiplied by a crop coefficient equal to two. Plant height, number of definitive leaves, shoot dry mass (root and total), seed quality index, survival and efficiency of water use by eucalyptus seedlings were evaluated. The average daily irrigation depth in the experimental period was 5.2 mm. Based on the results, it is recommended for eucalyptus seedling producers, in less-tech nursery, irrigation management twice per days. (11:00 a.m. and 7:00 p.m.).

Abstract:

Synthetic oligodeoxynucleotides (ODN) that contain unmethylated CpG motifs (CpG ODN) induce macrophages to secrete IL-12, which induces interferon (IFN)-γ secretion by natural killer (NK) cells. Since these cytokines can induce T helper 1 (Th1) differentiation, we examined the effects of coadministered CpG ODN on the differentiation of Th responses to hen egg lysozyme (HEL). In both BALB/c (Th2-biased) and B10.D2 (Th1-biased) mice, immunization with HEL in incomplete Freund's adjuvant (IFA) resulted in Th2-dominated immune responses characterized by HEL-specific secretion of IL-5 but not IFN-γ. In contrast, immunization with IFA-HEL plus CpG ODN switched the immune response to a Th1-dominated cytokine pattern, with high levels of HEL-specific IFN-γ secretion and decreased HEL-specific IL-5 production. IFA-HEL plus CpG ODN also induced anti-HEL IgG2a (a Th1-associated isotype), which was not induced by IFA-HEL alone. Control non–CpG ODN did not induce IFN-γ or IgG2a, excepting lesser increases in B10.D2 (Th1-biased) mice. Thus, CpG ODN provide a signal to switch on Th1-dominated responses to coadministered antigen and are potential adjuvants for human vaccines to elicit protective Th1 immunity.

Abstract:

ABSTRACT Laboratory diagnosis of Bartonella henselae infections can be accomplished by serology or PCR assay on biopsy samples. The purpose of our work was to assess immunofluorescence detection (IFD) in lymph node smears using a specific monoclonal antibody directed against B. henselae and a commercial serology assay (IFA) compared with PCR detection. Among 200 lymph nodes examined from immunocompetent patients, 54 were positive for B. henselae by PCR, of which 43 were also positive by IFD. Among the 146 PCR-negative lymph nodes, 11 were positive by IFD. Based on PCR results, the specificity of this new technique was 92.5%, the sensitivity was 79.6%, and the positive predictive value was 79.6%. At a cutoff titer of 64, the sensitivity of the IFA was 86.8% and the specificity was 74.1%. Diagnosis of cat scratch disease (CSD) may be improved, with a specificity of 100%, when the two tests (IFD and IFA) were negative; the sensitivity was 97.4% if one of the two tests was positive. Since PCR-based detection with biopsy samples is available only in reference laboratories, we suggest using IFD coupled with the commercial serology test for the diagnosis of CSD.

Abstract:

A Bacillus stearothermophilus in vitro translational system has been developed to study the expression of the three cistrons (infC, rpmI, and rplT) constituting the infC operon of this bacterium. When directed by homologous in vitro transcribed infC tricistronic mRNA, this system, which consists of partially purified and purified components of the B. stearothermophilus translational apparatus, synthesizes with high efficiency and specificity the three gene products (IF3, L35, and L20) in a ratio similar to that found in vivo (i.e., about 1:6:6). The three cistrons are translationally coupled and expressed in a specific temporal order: a low level of IF3 synthesis stimulates the expression of L35 which, in turn, greatly stimulates the synthesis of L20 and IF3. Protein L20 and an excess of IF3 were found to act as translational feedback inhibitors of the entire operon. The synthesis of IF3 displayed a strong dependence on IF2. This dependence as well as the repressibility by excess IF3 were found to be due to the presence of the rare AUU initiation triplet at the beginning of infC.Key words: translational coupling, IF3, IF2, L35, L20.
