Full textBiomaterials for tissue engineering applications have to comply with several requirements, such as safety, biocompatibility and appropriate mechanical features. The development process of these biomaterials encompasses several scientific approaches, ranging from in-silico to in-vivo. The in-silico optimization of biomaterials features is attracting even larger attention. Indeed, the improvement of this approach will allow to reduce additional costs int the biomaterials development process, due to unnecessary experimental characterizations. According to this point-of-view, in this thesis is presented a molecular dynamics approach for biomaterial characterization. More in details, self-assembling peptides (SAPs) hydrogels scaffolds have been investigated at the nano-scale and micro-scale, to elucidate their intrinsic structure-property-function relationships. The atomistic and coarse-grained molecular dynamics (CG-MD) have been used for the elucidation of self-assembling pathways of peptide-based scaffolds. Due to the lack of crucial structural information in CG-MD simulations, the innovative software suite, dubbed Morphoscanner, has been employed for the elucidation of conformational aggregation patterns of SAPs. Then, the mechanical properties and failure mechanisms of SAPs nanostructures have been investigated through the steered MD simulations. These evidences led the development of a CG-MD approach aiming to elucidate the complex interplay between cell membranes and SAPs nanofibrils. In particular, MARTINI CG-MD simulations have been used for understanding the effects of SAPS nanofibril on dynamics of lipid domains in neural membranes. Such achievements open up new dimensions in the field of biomateriomics, allowing to understand and eventually orchestrate the complex phenomena which affect the mechanical properties and biocompatibility of SAPs biomaterials for tissue engineering applications.

Full textTissue engineering aims to replace lost, damaged or failing tissue and organs, starting with cultured proliferating cells and ending with tissue-like structures. The extracellular matrix (ECM) plays a pivotal role in determining cell behavior during tissue remodelling processes leading to complete differentiated structures. Therefore it is desirable to control cell differentiation during tissue regeneration by mimicking the ECM using engineered biomaterials. To modify the biophysical and mechanical properties of these biomaterials can be used as a strategy to elicit specific cellular responses. Moreover, an injectable hydrogel biomaterial, having such capabilities, should have the advantage to be easily engrafted in vivo in order to carry stem cell and/or bioactive molecules to promote tissue renewal. Our research is thus focused on the use of an injectable hydrogel made from polyethylene glycol (PEG) conjugated to Fibrinogen. The PEG-Fibrinogen (PF) is used as scaffold for in vitro muscle reconstruction, seeded and cultured with mesoangioblasts (vessel associated progenitor cells) and freshly isolated muscle satellite cells (SCs). The PF, which can be controlled in terms of its matrix modulus, is tested in vivo as mesoangioblast carrier for muscle regeneration. The PF scaffold used for in vitro 3-D cultures promoted good survival of miogenic precursors and accelerated skeletal muscle differentiation (contractile myotubes) within 24 hours (normally, 2-D cultures take three days to exhibit myotube formation). The confining geometry of the microchannels created with microablation in the PF scaffolds promoted the development of oriented mature muscle fibers. Sub-cutaneous PF/miogenic precursors implants in Rag2 Chain -/- mice were able to form a “muscle organoid”. Finally, in vivo experiments using PF as a cell carrier showed increased transplanted cell survival, ameliorated in situ cellular retention and an overall improvement in cell engraftment. Injectable hydrogel biomaterials can be readily applied for skeletal muscle cell delivery by a simple one-step injection procedure and could have a noteworthy impact in the treatment of muscular dystrophy.

Full textOsteoarthritis (OA) is the most common rheumatic disease in the world and represents the first cause of disability in the world. OA results from the loss of balance between degradation and repair inside cartilage, in favor of degradation, with increased activity of catabolic enzymes such as matrix metalloproteinases and decreased production of ECM proteins. Chondrocytes are responsible for the repair and biosynthesis of elements of the extracellular matrix. Experimental findings support the hypothesis that diabetes is an independent risk factors for OA. However, correct molecular mechanisms underlying the diabetes-associated OA phenotype is still largely unknown. Firstly chondrocytes cell growth, ROS levels and apoptosis were analyzed using different glucose concentration. Results shown that chondrocytes prefer 2.5 mM of glucose which was used as normal glucose concentration and 25 mM of glucose was used as high glucose concentration. ROS levels and cell death increase in chondrocytes growth in high glucose environment. Also cytoskeletal network is more disorganized in C28/I2 cells growth at high glucose concentration, this correlates with different RalA-GTP levels which is involved in the regulation of cytoskeletal organization. Experiments were performed even using medium supplemented by ITS (Insulin-transferrin-selenium) to promote chondrocyte differentiation and IL-1β was used to simulate osteoarthritic cartilage environment. Ral A-GTP levels are lower in cells grown in 25 mM of glucose and stimulated with IL1β. Levels of p-ERK1/2 decrease in cells grown at high glucose concentration and in cells stimulated with IL1β. Furthermore, NF-κB, iNOS and LC3II levels were evaluated. Results demonstrate that high glucose media block autophagic process in chondrocytes. Effect of glucose concentration on human primary chondrocytes cells was evaluated after only 24 h to understand which signaling pathways is activated by high glucose environment. Phosphorylation of ERK1/2, p38, Akt and p65 is altered in chondrocytes growth at high glucose concentration and this correlates with an increase secretion of MMP-13. To better analyze the role of ROS levels in the chondrocytes I worked for 6 months in the Dr. Loeser Lab at the School of Medicine in the University of North Carolina at Chapel Hill; one of the best lab in the cartilage biology field. I worked on PRX6 which is involved in the recovery from H2O2. Oxidation state of PRX6 was evaluated in chondrocytes treated with different stimuli, like H2O2, Fn-f, menadione (men) and DMNQ. After this experiment, we wanted to see if PRX6 could impact the MAPK signaling pathways in cells treated with IGF-1, menadione, combination of menadione and IGF-1 and with Fn-f. Localization of PRX6 was analyzed using nuclear and cytoplasm extraction. Then I worked in collaboration with Prof. Laura Cipolla and Prof. Maddalena Collini to develop and characterized a new gelatin-based hydrogel using Diethylsquarate as crosslinker.

Full textHyadd4-G is a derivatized hyaluronic acid having 1÷3% of the carboxyl groups grafted with a hexadecylic amide side-chains. The introduction of a hexadecyl side chain onto hyaluronic acid (HA) chains yields a polysaccharide with new properties, capable of forming physical hydrogels, stable at very low polymer concentrations, whereas native HA forms viscous solutions at ten times higher concentrations. We addressed the structural and dynamic behavior of Hyadd4-G hydrogels at different time and length scales. Dynamic structural factor, f(Q,t), obtained by DLS, was used to extract the microscopic characteristic times and pore size of the network. Fluorescence recovery after photobleaching methodology, FRAP, was used to measure diffusion coefficients of fluorescent probes with different size. The hydrodynamic coil dimensions of dextrans evidencing a sieving effect of the hydrogels by FRAP experiments can be compared with the mesh size, δ, of Hyadd4-G hydrogels studied by DLS. Rheology measurements in the linear viscoelastic regime show that Hyadd4-G is a soft viscoelastic gel. Rheological measurements showed an unusual self-healing mechanism (structure recovery) after destruction of the gel network in a shear flow and the injectability of the hydrogel is supported by a pronounced shear-thinning behavior under nonlinear deformations. Neutron scattering techniques can provide information about the microstructure and the diffusional properties of water within the pores of the network. Characterization of Hyadd4-G showed that the driving force for its gel-like behavior is the occurrence of hydrophobic interactions involving hydrophobic side-chains stabilizing a 3D-network with nano-sized architecture, despite the low degree of substitution. This picture provides the necessary background to assess Hyadd4-G as one of the potential new hydrogel systems suitable for treatment of osteoarticular diseases. Xanthan and konjac glucomannan pair provides one of the most studied synergistic hydrogels. The junction zones stabilizing the 3D structure of this gel are present as macromolecular complexes in solution formed by the partially depolymerised polysaccharidic chains. The non-covalent interactions stabilizing the structure of the polysaccharidic complex cause the melting of the ordered structure of the complex in the solution and of the hydrogels. Introduction of chemical cross-links in the 3D structure of the synergistic hydrogel removes this behaviour, adding new features to the swelling and to the viscoelastic properties of the cured hydrogel. The use of epichlorohydrin as low molecular weight cross-linker does not impact on the viability of NIH 3T3 fibroblasts.

Full textScope of this work is to examine design issues concerning the reforming of the hydrocarbons in compact systems, which can be used for hydrogen generation onboard vehicles. Several models have been developed in order to build a comprehensive fluid dynamics tool to simulate physical and chemical phenomena that affect reforming process. Models have been extensively validated on results from scientific literature and "Direct" project, a research program funded by the European Union which concern Diesel fuel reforming. The models were then used successfully in the design of two reactors, the first called "Prometer", the other "Syngas UTV", which are capable of operating Catalytic Partial Oxidation (CPOx) of liquid fuels and gaseous fuels respectively. The two multifuel systems have been tested with Diesel fuel and methane. In particular, the choice of run "Prometer" reactor with Diesel fuel is due to the fact that gasoil CPOx is particularly difficult due to soot formation, which often involves fast and irreversible shutdown of the reactor. For this reason, the development of stable and long-life CPOx reactors for Diesel fuel is still an objective for scientific community. The experimental result were successful, even for Diesel reforming, since no soot formation was observed during experiments. To this goal, fuel vaporization and mixing were found to have a key role.
