Abstract:
A plant auxin hormone indole-3-acetic acid (IAA) can be assimilated by bacteria as an energy and carbon source, although no degradation has been reported for indole-3-propionic acid and indole-3-butyric acid. While significant efforts have been made to decipher the Iac (indole-3-acetic acid catabolism)-mediated IAA degradation pathway, a lot of questions remain regarding the mechanisms of individual reactions, involvement of specific Iac proteins, and the overall reaction scheme. This work was aimed at providing new experimental evidence regarding the biodegradation of IAA and its derivatives. Here, it was shown that Caballeronia glathei strain DSM50014 possesses a full iac gene cluster and is able to use IAA as a sole source of carbon and energy. Next, IacE was shown to be responsible for the conversion of 2-oxoindole-3-acetic acid (Ox-IAA) intermediate into the central intermediate 3-hydroxy-2-oxindole-3-acetic acid (DOAA) without the requirement for IacB. During this reaction, the oxygen atom incorporated into Ox-IAA was derived from water. Finally, IacA and IacE were shown to convert a wide range of indole derivatives, including indole-3-propionic acid and indole-3-butyric acid, into corresponding DOAA homologs. This work provides novel insights into Iac-mediated IAA degradation and demonstrates the versatility and substrate scope of IacA and IacE enzymes.

Abstract:
Abstract BACKGROUND The internal auditory canal (IAC) is an important landmark during surgery for lesions of the cerebellopontine angle. There is significant variability in the position and orientation of the IAC radiographically, and the authors have noted differences in surgical exposure depending on the individual anatomy of the IAC. OBJECTIVE To test the hypothesis that IAC position and orientation affects the surgical exposure of the IAC and facial nerve, especially when performing the translabyrinthine approach. METHODS The authors retrospectively reviewed magnetic resonance imaging studies of 50 randomly selected patients with pathologically confirmed vestibular schwannomas. Measurements, including the anterior (APD) and posterior (PPD) petrous distances, the anterior (APA) and posterior (PPA) petro-auditory angles, and the internal auditory angle (IAA), were obtained to quantify the position and orientation of the IAC within the petrous temporal bone. RESULTS The results quantitatively demonstrate tremendous variability of the position and orientation of the IAC in the petrous temporal bone. The measurement ranges were APD 10.2 to 26.1 mm, PPD 15.1 to 37.2 mm, APA 104 to 157°, PPA 30 to 96°, and IAA –5 to 40°. CONCLUSION IAC variability can have a substantial effect on the surgical exposure of the IAC and facial and vestibulocochlear nerves. Specifically, a horizontally oriented IAC with a small IAA may have significant impact on visualization of the facial nerve within its cisternal segment with the translabyrinthine approach. The retrosigmoid approach is less affected with IAC variability in position and angle.

Abstract:
Integrin adhesion complexes (IACs) form mechanochemical connections between the extracellular matrix and actin cytoskeleton and mediate phenotypic responses via posttranslational modifications. Here, we investigate the modularity and robustness of the IAC network to pharmacological perturbation of the key IAC signaling components focal adhesion kinase (FAK) and Src. FAK inhibition using AZ13256675 blocked FAKY397 phosphorylation but did not alter IAC composition, as reported by mass spectrometry. IAC composition was also insensitive to Src inhibition using AZD0530 alone or in combination with FAK inhibition. In contrast, kinase inhibition substantially reduced phosphorylation within IACs, cell migration and proliferation. Furthermore using fluorescence recovery after photobleaching, we found that FAK inhibition increased the exchange rate of a phosphotyrosine (pY) reporter (dSH2) at IACs. These data demonstrate that kinase-dependent signal propagation through IACs is independent of gross changes in IAC composition. Together, these findings demonstrate a general separation between the composition of IACs and their ability to relay pY-dependent signals.

Abstract:
Background. Endotoxin initiates osteoclastic activity resulting in bone loss. Endotoxin leakage through implant abutment connections negatively influences peri-implant bone levels.Objectives. (i) To determine if endotoxin can traverse different implant-abutment connection (IAC) designs; (ii) to quantify the amount of endotoxins traversing the IAC; (iii) to compare the in vitro comportments of different IACs.Materials and Methods. Twenty-seven IACs were inoculated withE. coliendotoxin. Six of the twenty-seven IACs were external connections from one system (Southern Implants) and the remaining twenty-one IACs were made up of seven internal IAC types from four different implant companies (Straumann, Ankylos, and Neodent, Southern Implants).Results. Of the 27 IACs tested, all 6 external IACs leaked measurable amounts of endotoxin. Of the remaining 21 internal IACs, 9 IACs did not show measurable leakage whilst the remaining 12 IACs leaked varying amounts. The mean log endotoxin level was significantly higher for the external compared to internal types (p=0.015).Conclusion. Within the parameters of this study, we can conclude that endotoxin leakage is dependent on the design of the IAC. Straumann Synocta, Straumann Cross-fit, and Ankylos displayed the best performances of all IACs tested with undetectable leakage after 7 days. Each of these IACs incorporated a morse-like component in their design. Speculation still exists over the impact of IAC endotoxin leakage on peri-implant tissues in vivo; hence, further investigations are required to further explore this.

Abstract:
Objectives: The aims of this study were to clarify the clinical value of the bony cochlear nerve canal (BCNC) and internal auditory canal (IAC) in children with bilateral sensorineural hearing loss (b-SNHL) and to reveal the correlation between these parameters and outcomes after cochlear implantation (CI). Methods: Ninety-four ears with b-SNHL that received CI and 100 ears with normal hearing were enrolled. Parameters of IAC and pre- and post-CI categories of auditory performance scores were analyzed. Results: The width of the BCNC and the width, height, and length of the IAC were shorter in the b-SNHL group. BCNC and IAC width were associated with b-SNHL. The calculated cutoff values for BCNC and IAC width were 2.055 mm in the BCNC and 4.245 mm in the IAC, setting the sensitivity to 90%. Patients with narrow BCNCs and IACs had significantly worse post-CI auditory performance. Conclusions: BCNC and IAC widths were narrower in children with b-SNHL than in normal-hearing children. Narrow BCNC and IAC width had a negative impact on post-CI outcomes. The proposed cutoff values for BCNC and IAC width were meaningful when predicting the auditory outcome after CI, especially considering both.

Abstract:
Abstract Two different immunoaffinity columns (IACs) were prepared for detection of staphylococcal enterotoxins (SETs) from dairy products. First, a specific IAC for staphylococcal enterotoxin A (SEA), IAC-1, was prepared by coupling monoclonal antibody (mAb) directed against SEA; second, a polyspecific IAC for SEA, staphylococcal enterotoxin B (SEB), staphylococcal enterotoxin C (SECs), and staphylococcal enterotoxin D (SED), IAC-2, was prepared by coupling a mixture of mAbs against SEA, SECs, and SED, and rabbit IgG against SEB. These columns were applied for detection of SETs in dairy products, after extraction, immunoaffinity chromatography, and enzyme immunosorbent assay (EIA). Overall recoveries from dairy products spiked with 1 ng SEA/25 g averaged 81.2% (range, 76–85%) on IAC-1. The repeated use of IAC-1 was then determined with good efficiency of 91.5%, in more than 10 runs. On the other hand, a recovery yield of 77%of SETs (SEA, SEB, SEC, and SED) from dairy products spiked with 2.5 ng of each enterotoxin per 25 g, was obtained with IAC-2. IAC-2 was also successfully subjected to the chromatography of naturally contaminated foods implicated in staphylococcal food poisoning outbreaks. This new extraction–concentration–immunoaffinity–chromatography method (ECIC) is very useful for improving staphylococcal enterotoxin detection and eliminating matrix effect in EIA of dairy products.

Abstract:
ABSTRACT PCR-based detection assays are prone to inhibition by substances present in environmental samples, thereby potentially leading to inaccurate target quantification or false-negative results. Internal amplification controls (IACs) have been developed to help alleviate this problem but are generally applied in a single concentration, thereby yielding less-than-optimal results across the wide range of microbial gene target concentrations possible in environmental samples (J. Hoorfar, B. Malorny, A. Abdulmawjood, N. Cook, M. Wagner, and P. Fach, J. Clin. Microbiol. 42:1863-1868, 2004). Increasing the number of IACs for each quantitative PCR (qPCR) sample individually, however, typically reduces sensitivity and, more importantly, the reliability of quantification. Fortunately, current advances in high-throughput qPCR platforms offer the possibility of multiple reactions for a single sample simultaneously, thereby allowing the implementation of more than one IAC concentration per sample. Here, we describe the development of a novel IAC approach that is specifically designed for the state-of-the-art Biotrove OpenArray platform. Different IAC targets were applied at a range of concentrations, yielding a calibration IAC curve for each individual DNA sample. The developed IACs were optimized, tested, and validated by using more than 5,000 unique qPCR amplifications, allowing accurate quantification of microorganisms when applied to soil DNA extracts containing various levels of PCR-inhibiting compounds. To our knowledge, this is the first study using a suite of IACs at different target concentrations to monitor PCR inhibition across a wide target range, thereby allowing reliable and accurate quantification of microorganisms in PCR-inhibiting DNA extracts. The developed IAC is ideally suited for high-throughput screenings of, for example, ecological and agricultural samples on next-generation qPCR platforms.

Abstract:
Objective Evaluate the feasibility of an adequate exposure with anatomical preservation of labyrinth structures through retrosigmoid transmeatal approach (RSA) in surgeries for resection of acoustic neuromas/vestibular schwannomas (VS). Method Thirty patients underwent surgical resection and were preoperatively evaluated with fine slice high definition CT scans and 3D-MRI volumetric reconstructions. Extension of internal auditory canal (IAC) opening during surgery was measured using 3 mm right-angle calibrated hook and neuronavigation parameters. Postoperatively, the extension of IAC opening and integrity of the labyrinth were confirmed through preoperatively images procedures. Results The preoperative length of IACs varied between 7.8 and 12.0 mm (mean 9.3 mm, SD 0.98, 95%CI 8.9 to 9.6, and median 9.0 mm). Postoperative images demonstrated adequate opening of the IAC and semicircular channels integrity. Conclusion A complete drilling of the posterior wall of IAC through the RSA is feasible and allows direct visualization of the IAC-fundus without damaging the semicircular canals.

Abstract:
BackgroundNew device technology has changed the techniques used for revascularization of emergent large vessel occlusion in acute stroke. We report technical results using stent retrievers (SRs) for thrombectomy alone versus SRs used in conjunction with a new group of devices, intracranial aspiration catheters (IACs). Our aim is to demonstrate differences in procedural time and thrombectomy attempts between these two groups.MethodsA retrospective evaluation was performed of a prospectively maintained database of 97 patients treated at a single institution for anterior circulation stroke with SRs. Patients were divided into two groups, a combination group defined as the SR/IAC group and the SR alone group defined as the SR group.ResultsPatients in the SR/IAC group had a mean age of 66 years vs 59 years in the SR group (p=0.008). Mean presenting National Institutes of Health Stroke Scale (NIHSS) scores in the SR/IAC and control groups were 18.7 and 18.2, respectively (p=0.50). Recanalization rates (Thrombolysis In Cerebral Infarction (TICI) 2b or 3) in the SR/IAC and SR groups were 85% (58/68) and 90% (26/29), respectively (p=0.41). Mean time from groin arteriotomy to recanalization was 50±3.6 min (range 19–136) in the SR/IAC group (n=59) and 61±6.6 min (range 28–140) in the SR group (n=27) (p=0.049). The total number of thrombectomy attempts in the SR/IAC and SR groups were 1.9±0.1 (range 1–4) and 2.5±0.6 (range 1–6), respectively (p=0.009). Post-procedural subarachnoid hemorrhage was seen in 15% (10/68) and 10% (3/29) of cases in the SR/IAC and SR groups, respectively (p=0.41).ConclusionWhen using SRs for intracranial stroke thrombectomy, the concurrent use of IACs is associated with a decrease in procedural time and thrombectomy attempts compared with use of SRs alone.

Abstract:
Peanut (Arachis hypogaea L.) is one of the most consumed oil in the world and Brazil has the potential to emerge as a major producer and exporter. Peanut different cultivars have been developed to improve the productivity, nutritional quality and shelf life of grain and its derivatives. The literature indicates difference in the chemical and nutritional composition of cultivars obtained by genetic improvement. This project aimed to identify peanut cultivars in production and its nutritional composition with a view to food, nutritional profile and government programs to boost consumption. The selected cultivars were Granoleico (Argentine origin), OL3 IAC, IAC 886, IAC 505 and IAC Red Tatu (developed in Brazil). Samples of five cultivars were collected in cooperatives of the main producing regions of São Paulo, in the cities of Tupa and Marilia, representatives of Alta Paulista region with light soils and Jaboticabal and Sertaozinho, corresponding to the region of Upper Mogiana with red soil. These five cultivars produced in the State of São Paulo, reaching 83% of cultivation area, which is responsible for 92% of production in Brazil. The samples were analyzed for chemical composition, the presence of anti-nutritional factors (phytic acid, tannins, saponins and activity of urease), minerals (iron, zinc, phosphorus, calcium, manganese and magnesium), fatty acid profile, total tocopherols and β-carotene. Statistical analysis was performed by analysis of variance - ANOVA and application of the Tukey test for comparison of means. The observed results showed variations of 18% carbohydrate, 9.7% protein, 28% fiber and 8.2% lipids, with IAC 505 having the highest percentage of lipids and proteins. Mineral varied to 45% Fe, 40% Ca, 12% Mg, 39% for Mn, 41% to 30% for P and Zn. Differences were observed in anti-nutritional content, confirming if cultivars with saponins content (Red Tatu), phytic acid (IAC OL3), condensed tannins (IAC 886) and urease activity (IAC 886), higher than the others. The levels of total tocopherols and β-carotene were higher in cultivars Tatu Vermelho and Granoleico respectively. The quality viewpoint of the fatty acid profile, the IAC 505 is highlighted with the composition 42% monounsaturated fatty acids, 1.4% from polyunsaturated, saturated and 6% oleic reason / linoleic 26. Were elaborated comparison tables between the results obtained and the dietary intake recommendations to check the supply of nutrients. With the results, it can be concluded that the chemical and nutritional composition of developed cultivars may be affected by genetic improvement, but further studies should be conducted. There are varieties with higher nutritional quality to other supplying organic needs and are more interesting for consumption according to the nutritional purpose. These should be included in the incentive programs for agricultural production and encouraged consumption due to beneficial impact on the nutritional profile of populations.

Abstract:
This thesis is performed at International Automotive Components (IAC) Group located in Färgelanda, Sweden. The purpose of the work is to propose a layout of a workstation in a new production line that will be launched in autumn 2017. A layout means physical location of the components in the station. The parts produced by this production line are details of a roof shelf which is part of the interior of a Swedish leading truck manufacturer. During the project, the production line is currently being assembled and some test series is run.The work station that this thesis deals with is a welding operation of the lids before transported to a final assembly. Because the lids are manufactured in different models, the weld needs replacement of tools in order to process the current part. Different assembly tables also needs to be replaced depending on the model of the lid witch is to be manufactured. The work station needs to effectively have a flow of material, while the operators must have an ergonomically designed workplace, and that requires a carefully thought out layout. The process of preparation of this layout was done with methods of product development where concepts are generated and evaluated. The concept development process has been in consultation with the company supervisor and production. A total of six concepts were generated and they were graded and weighted in a matrix, which later led to the choosing of a basic model. After selecting the basic model were more concepts generated in a more detailed cell design. The last generated concepts were presented to the staff involved that eventually opted for a final concept when it met the requirements and preferences that were set. Part of the thesis was also to simulate the work of the operators, ensure the balancing and suggest possible improvements. The results in the report describes the final concept together with recommendations and guidelines of lean production. The actual implementation of the layout will be done by the company themselves.

Three field trials were carried out on Paranapanema Valley during the 1998/99 growing season, focusing soybean IAC 18 and IAC 22 cultivar management. The sowing dates were established in the first week of the following months: October, November and December; the sowing densities were: 8, 15, 22, 29 and 36 plants/m, in rows spaced 0,50m. The trials were placed at Pedrinhas Paulista (SP) on 22º48´55´´ south latitude, 50º46´36´´ west longitude, 331 m altitude coordinates, with high fertility soil; Tarumã (SP) on 22º47´17´´ south latitude, 50º32´44´´ west longitude, 400 m altitude coordinates, with medium fertility soil, and Assis (SP) on 22º37´09´´ south latitude, 50º22´27´´ west longitude, 560 m altitude coordinates, with low fertility soil. The studies aimed to determine soybean features as: grain production, leaf, pod and stream dry weight, plant and first pod high, leaf area index (LAI) and crop growth rate (CGR) as affected by plant density, cultivars and sowing time. The experimental design used was the completely randomized blocks with four replications and the treatments were arranged in a factorial of 3 sowing dates x two cultivars x 5 sowing densities. The results were compared through F test and densities, and planting dates were compared by regression analysis. The variance analysis showed significant interactions of all parameters analyzed for the three locations tests. Higher grain yelds were obtained when soybean was sowed in October. The soybean cultivars may be cultivated under different plant densities on different sowing dates…(Complete abstract, click electronic access below)

Abstract: The global demand for excellence of floriculture products requires the use of appropriate technologies to ensure a good quality product. One important factor is the acquisition of selected plant propagation material, free of contamination, guaranteed by specialized laboratories. Aiming for the assessment of clones of the main brazilian variety of anthurium, this work was conducted to evaluate some preharvest and postharvest quality indices of plant growth and flowers, respectively. Anthurium andraeanum Lind. ('IAC Eidibel') was grown in closed hydroponic system with coconut fiber as growing media, with plants and flowers evaluated qualitative and quantitatively during the experimental period. As a result, the anthurium 'IAC Eidibel' presented: full adaptation and development in closed hydroponic system with coconut fiber; early production and plant height, constant and homogeneous; average production of one leaf/plant per month; constant increment of dry matter in leaves bigger than the increase in leaf area; greater accumulation of dry matter of spathe at the point of maturation with 1/3 of their flowers still immature; uniform color and shape of the spathes; an average stem strainghtness index of 15%; flowers at the commercial stage in the second year of cultivation and; prunning control of leaves after the sixth month of cultivation.
