Abstract:
Brucellosis has become a zoonotic disease that received attention in efforts to prevent and eradicate strategic infectious animal diseases in Indonesia. Brucellosis can be detected early by the rose bengal test (RBT), followed by complement fixation test (CFT) and by enzyme linked immunosorbent assay (ELISA). The aims of this research was to study the indirect enzyme linked immunosorbent assay test (I-ELISA) as an alternative test for detecting brucellosis in dairy cattle. The method was used by conducting tests of RBT, CFT, I-ELISA and commercial I-ELISA to test brucellosis. The test results were calculated sensitivity and specificity, as well as analyzed by calculating the kappa value. The test results were calculated for sensitivity and specificity, as well as analyzed by calculating the Kappa statistical value. The results of the sensitivity and specificity calculation showed that the indirect enzyme linked immunosorbent assay (I-ELISA) test developed a higher sensitivity (100%) compared to RBT test (93.75%) and commercial I-ELISA (93.75%). The developed I-ELISA specificity (74.68%) was still lower than RBT (89.87%), but higher than commercial I-ELISA (70.52%). The calculation of the statistical value of kappa RBT with CFT showed the kappa value 0.7120 which meaned it had a good agreement, commercial I-ELISA with CFT showed kappa value 0.6165 which meaned it had good suitability, whereas I-ELISA developed with CFT showed kappa value 0.4984 which meaned having a moderate agreement.In conclusion, the indirect enzyme linked immunosorbent assay (I-ELISA) which had been developed had low specificity, but the sensitivity was the highest compared to the commercial I-ELISA test and RBT, so this test was appropriate to be used as a screening test, especially in dairy cows movement into brucellosis-free areas or regions.

Abstract:
The use of 1:1:1 (packed red blood cells: fresh frozen plasma: platelets) transfusion ratio has been shown to improve survival in severely injured trauma patients. The aim of this study was to assess the outcomes in patients with traumatic brain injury (TBI) receiving 1:1:1 ratio-based blood product transfusion (RBT). We hypothesized that RBT improves survival in patients with TBI as only major injury. We performed a 3-year retrospective analysis of all patients with TBI as only major injury presenting to our Level I trauma center. Patients receiving blood transfusion were included. Patients were stratified into two groups: those who received RBT and those who did not receive RBT (No-RBT). The outcome measure was inhospital mortality. Multivariate logistic regression analysis was performed. A total of 189 patients were included of which 29 per cent (n = 55) received RBT. The mean age was 48 ± 24 years, median (range) Glasgow Coma Scale score was 12 (3–15), and median head abbreviated injury severity scale was 3 (3–5). The overall mortality rate was 28.5 per cent. Patients in the RBT group had a higher survival rate compared with the patients in the No-RBT group (83.6% vs 66.5%, P = 0.02). In conclusion, the survival benefit of RBT exists even in patients with TBI as major injury. Guidelines for the initial management of TBI patients should focus on the use of RBT. The beneficial effect of platelets in RBT among TBI patients requires further evaluation.

Abstract Forty four pigs with typical characteristics for false positive serological reactions (FPSR) were examined for the presence of Yersinia enterocolitica O:9. The positive reactions were observed in rose bengal test (RBT, N=23 sera), serum agglutination test (SAT, N=16), complement fixation test (CFT, N=9), indirect ELISA (i-ELISA, N=11) in first, and in RBT (N=14), SAT (N=8), CFT (N=7) and i-ELISA (N=18) in second examination, respectively. In bacteriological examination Y. enterocolitica was confirmed in 12 cases. Six of these isolates were identified with PCR as Y. enterocolitica O:9.

ABSTRACT Brucellosis is an infectious disease caused by bacteria of the genus Brucella spp. with diagnosis based on use of serological techniques. The present study aimed to develop and standardize a western blotting (WB) test for detection of antibodies against B. abortus. Samples from two groups of cattle were analyzed: group I: 60 serum samples from true positive and true negative vaccinated animals (30 positive samples from infected animals according to rose bengal test (RBT), 2-mercaptoethanol, serum agglutination test (SAT) and complement fixation test (CFT) and 30 RBT negatives samples); group II: 383 field samples (90 positive and 293 CFT negative sera). The most reactive band in the western blotting, which properly identified and separated infected from non - infected had a molecular weight of ≤ 20kDa. The sensitivity, specificity and accuracy of the WB compared to RBT was 93%, 99%, 98%, respectively and k= 0.938. When compared to CFT, the sensitivity, specificity and accuracy of the WB was 97%, 98% and 97%, respectively and k= 0.929. The WB developed and standardized in the present study is a serological test with potential use as a confirmatory test for the diagnosis of bovine brucellosis.

Abstract:
The Total Antioxidant Index (TAA-I) can be viewed as a wine added value, for which consumers are willing to pay, and as a determinant in developing successful differentiated marketing strategies. This interdisciplinary study aims to harness potentials existing in an evidence-based wine TAA-I and territorial data on wine production. According to the Resource Based Theory (RBT), only above-average performances identify valuable resources. By applying the theory, data on regional areas planted with vines as well as a seminal study ranking thirteen selected Italian wines based on their TAA-I have been analyzed. Wine growing vocation represents a valuable resource only in six out of twenty Italian regions. Eleven vine areas of origin can be assumed as being important, especially in Northern and Central Italy. The TAA-I positions achieved by selected wines are far from being expressed, and thus promoted, by adopting labels based on production traditions. The 1997 sample database adopted by this study needs to be updated with a more recent one and expanded in further researches. Thus, researches of evidencebased marketing, thus hidden and objective quality attributes of wine, are needed and urgent but still in their infancy. Results obtained by this study allow decision makers to set-up a RBT-rooted benchmark system connecting biomedical studies, economic and marketing studies.

Test evaluation in the absence of a gold standard test was conducted for the diagnosis and screening of bovine brucellosis using three commercially available tests including RBPT, CFT, and I-ELISA in National Animal Health Diagnostic and Investigation Center (NAHDIC) Ethiopia. A total of 278 sera samples from five dairy herds were collected and tested. Each serum sample was subjected to the three tests and the results obtained were recorded and the test outcomes were cross-classified to estimate the sensitivity and specificity of the tests using Bayesian model. Prior information generated on the sensitivity and specificity of bovine brucellosis from published data was used in the model. The three test-one population Bayesian model was modified and applied using WinBug software with the assumption that the dairy herds have similar management system and unknown disease status. The Bayesian posterior estimate for sensitivity was 89.6 (95% PI: 79.9–95.8), 96.8 (95% PI: 92.3–99.1), and 94 (95% PI: 87.8–97.5) and for specificity was 84.5 (95% PI: 68–94.98), 96.3 (95% PI: 91.7–98.8), and 88.5 (95% PI: 81–93.8) for RBT, I-ELISA, and CFT, respectively. In this study I-ELISA was found with the best sensitivity and specificity estimates 96.8 (95% PI: 92.3–99.1) and 96.3 (95% PI: 91.7–98.8), compared to both CFT and RBPT.

SummaryReference materials for thromboplastins are available from the World Health Organization (WHO) and the European Commission (EC). The long-term stability of the reference materials is an essential requirement and must be monitored. The relationship between two reference materials for rabbit thromboplastin, i. e. ERM-AD149 (EC) and RBT/90 (WHO), has been monitored in the period 1996–2002. No significant trend with time was detected. In addition,the relationship between ERM-AD149 and the reference material for bovine thromboplastin (i. e. OBT/79) has been determined in 1994 and in 2005 in multicentre studies (n = 11 and n = 9, respectively). No significant changes were observed in the relationships between these reference materials when all results were included (5% significance level).

Background and Aim: Brucellosis is endemic zoonotic and highly contagious bacterial disease. Recently, several brucellosis cases were reported in Lebanon, causing significant economic losses; however, no study was done so far on farms located in the southern part of the country. Thus, the aim of our study was to estimate the prevalence of Brucella abortus in South Lebanon using three different serological tests in the diagnosis of brucellosis in cattle. Materials and Methods: Seventeen farms from 14 locations in Southern Lebanon were selected. Two hundred and three bovine blood samples of different ages, and 121 milk samples collected from older than 2 years cattle were tested using different serological tests: Rose Bengal test (RBT), milk ring test (MRT), indirect enzyme-linked immunosorbent assay (I-ELISA), and confirmed with competitive ELISA (C-ELISA). Results: Results revealed that approximately 15.3% (confidence interval [CI] 95 10.3-20.2%) and 15.7% (CI 95 9.2-22.2%) of samples were positive using RBT and MRT, respectively. This percentage was significantly higher when using I-ELISA (18.3%) (CI 95 12.9-23.5%) and C-ELISA (18.7%) (CI 95 9.8-27.5%). Among used diagnostic tests, our results showed that ELISA was more accurate for the detection of brucellosis, especially since it detects the late stages of the infection, which is characterized by the presence of immunoglobulin G. The seroprevalence of brucellosis was higher among females. All positive tests were of cattle Holstein breed older than 2 years. Tyre and Jezzine cities had a higher significance in bovine brucellosis than Saida. A positive correlation between human and cattle brucellosis was found. Conclusion: Our results showed that bovine brucellosis is prevalent in southern Lebanon. Lack of research, in addition to little feedback of occurring illness or symptoms, creates a gap in helping to control the spread of the disease.

ABSTRACT Brucella ceti causes disease in Odontoceti. The absence of control serum collections and the diversity of cetaceans have hampered the standardization of serological tests for the diagnosis of cetacean brucellosis. Without a “gold” standard for sensitivity and specificity determination, an alternative approach was followed. We designed an indirect enzyme-linked immunosorbent assay (iELISA) that recognizes immunoglobulins G (IgGs) from 17 odontocete species as a single group. For the standardization, we used Brucella melitensis and Brucella abortus lipopolysaccharides, serum samples from seven resident odontocetes with no history of infectious disease displaying negative rose bengal test (RBT) reactions, and serum samples from seven dolphins infected with B. ceti. We compared the performance of the iELISA with those of the protein G ELISA (gELISA), the competitive ELISA (cELISA), and the immunofluorescence (IF) and dot blot (DB) tests, using 179 odontocete serum samples and RBT as the reference. The diagnostic potential based on sensitivity and specificity of the iELISA was superior to that of gELISA and cELISA. The correlation and agreement between the iELISA and the gELISA were relatively good (R i/g 2 = 0.65 and κi/g = 0.66, respectively), while the correlation and agreement of these two ELISAs with cELISA were low (R i/c 2 = 0.46, R g/c 2 = 0.37 and κi/c = 0.62, κg/c = 0.42). In spite of using the same anti-odontocete IgG antibody, the iELISA was more specific than were the IF and DB tests. An association between high antibody titers and the presence of neurological symptoms in dolphins was observed. The prediction is that iELISA based on broadly cross-reacting anti-dolphin IgG antibody would be a reliable test for the diagnosis of brucellosis in odontocetes, including families not covered in this study.

Abstract:
O presente trabalho teve por objetivo verificar o efeito de diferentes níveis de IGF-I no colostro de vacas que receberam rbST durante o período pré-parto, no desenvolvimento do trato intestinal e na expressão do gene do IGF-I e de seu receptor no fígado e intestino de bezerros. Quarenta e duas vacas da raça Holandesa, gestantes e multíparas foram distribuídas ao acaso em dois grupos de 21 animais, o grupo rbST que recebeu hormônio de crescimento (500 mg rbST) e o grupo controle que recebeu injeção de vitamina E. As aplicações tiveram início aos 35 dias pré-parto, em intervalos de 14 dias até a data de parição. Os bezerros recém-nascidos foram distribuídos aleatoriamente, de acordo com as seguintes idades em que foram abatidos: após o nascimento e sem a ingestão de colostro, dois e sete dias de vida com ingestão de colostro das respectivas mães. Após o abate, amostras do fígado e segmentos do intestino delgado (jejuno e íleo) foram coletadas para quantificação de DNA, RNA, proteína total e o RNAm do IGF-I e do receptor tipo I. O delineamento experimental utilizado foi o inteiramente casualizado, numa estrutura fatorial 2x3, correspondendo aos grupos das mães (rbST ou controle) e as idades dos bezerros (após o nascimento, dois e sete dias de vida). No fígado, as concentrações de RNA e proteína (mg/g tecido) foram maiores no segundo dia de vida e a relação proteína/RNA aumentou no sétimo dia de vida (P<0,05). O jejuno apresentou interações entre os tratamentos nas concentrações de DNA, proteína e relações proteína/RNA e RNA/DNA (P<0,05). Os bezerros que consumiram colostro proveniente de vacas que receberam rbST apresentaram, no jejuno, maiores concentrações de DNA no segundo dia de vida e diminuição a níveis intermediários entre nascimento e dois dias, comparados aos sete dias de vida. Este efeito também foi observado na relação proteína/RNA. No grupo controle, também se verificou aumento de DNA aos dois dias, mas não houve diferenças aos sete dias, e a relação proteína/RNA foi semelhante entre as idades. A concentração de proteína no jejuno do grupo rbST aumentou no segundo dia de vida e diminuiu no sétimo, enquanto que no grupo controle, este aumento foi verificado apenas no sétimo dia de vida. A relação RNA/DNA diminuiu apenas no grupo controle (P<0,05). A expressão do gene do IGF-I foi maior ao nascimento e aos sete dias de idade no fígado dos bezerros do grupo rbST (P<0,05). As concentrações do gene do receptor tipo I diminuíram com a idade dos bezerros (P<0,05). Ao nascimento, o intestino dos bezerros apresentou condição de resposta celular à presença do IGF-I proveniente do colostro. Os resultados sugerem que as células do jejuno de bezerros do grupo rbST apresentaram um diferente estágio de maturação.
The objective of this study was to verify the effect of different levels of IGF-I in colostrum of cows treated with rbST during dry period, in the development of the intestinal tract and IGF-I and receptor gene expression in the liver and intestine of calves. Forty-two Holstein cows, in gestation and multiparous, were randomly assigned in two groups of 21 animals. Group rbST received injection of growth hormone (rbST), and group control received vitamin E injection. Both treatments started 35 days pre-partum, and were administered every 14 days until parturition. Newborn calves were randomly assigned to the following ages of slaughter: just after birth and without colostrum ingestion; two and seven days of life with colostrum ingestion from their respective mothers. After slaughter, samples from liver and small intestine (jejunum and ileum) were collected for quantification of DNA, RNA, total protein and mRNA of IGF-I and receptor type I. A completely randomized design was used with 2X3 factorial arrangements of treatments, which the factors were the mother’s group (control and rbST) and age (just after birth, two and seven days of life). In the liver, RNA and protein concentrations (mg/g tissue) were higher in the second day of age and protein/RNA ratio increased in the seventh day (P<0.05). The jejunum showed interaction among the treatments in the concentrations of DNA and protein, and protein/RNA RNA/DNA ratios (P<0.05). Calves fed colostrum from mothers that received rbST presented, in jejunum, higher DNA concentration in the second day of life, and decrease to intermediate levels between birth and two days, compared to seven days of age. This effect was also observed in protein/RNA ratio. In the control group, it was also verified DNA increase at two days, but there was not difference in the seventh day, and protein/RNA ratio was similar among ages. Protein concentration in jejunum of rbST group increased in the second day and decreased in the seventh, but in the control group, this increase was verified only in the seventh day of age. RNA/DNA ratio decreased in the control group (P<0.05). Expression of IGF-I was higher at birth and seven days old in the liver of the calves from rbST group (P<0.05). The concentrations of receptor type I mRNA decreased with calves’ age (P<0.05). At birth, the small intestine of calves showed a condition of cellular response to the presence of IGF-I in colostrum. These results suggest that cells in jejunum of calves from rbST group presented a different phase of maturation.

Abstract:
Despite efforts to eliminate the HIV-1 virus from infected individuals, the virus persists in a latent chromosomally integrated pool of provirus that evades current drug therapy. Upon cessation of highly active anti-retroviral therapy (HAART), the latent virus begins to replicate resulting in an increased viral titre in the patient's serum, leading to Acquired Immune Deficiency Syndrome (AIDS). A great amount of effort has gone into determining the mechanisms involved in the establishment of viral latency, in order to find targets that can be used in combination with existing therapies to eradicate the virus. In this thesis, I characterize a protein complex that is required for viral replication of integrated virus and therefore has potential as a therapeutic target. The data presented in this thesis identify USF1, USF2, and TFII-I as the proteins that comprise the Ras-response element Binding Factor-2 (RBF-2), which binds constitutively to the Ras-response Factor Binding Elements I and III (RBEI and RBEIII) within the Long Terminal Repeat (LTR) promoter. RBEIII is highly conserved in sequence and position relative to the transcriptional start site. In addition, both RBEIII and RBEI are situated near nucleosomes in the LTR enhancer region. Furthermore, I provide evidence that RBF-2 proteins are modified by phosphorylation in response to T-cell activation. Finally, I demonstrate that mutations in RBEIII that prevent the binding of these factors, as measured by electrophoretic mobility shifting assays, also prevent HIV-1 activation, as observed from stably integrated reporter virus.

Cette étude concerne les propriétés structurales, optiques et électriques des couches minces d'oxynitrure de silicium ( SiOxNy) élaborées par pulvérisarion cathodique radiofréquence d'une cible de silicium dans un plasma argon-oxygène-azote. La zone d'instabilité de ce procédé réactif a été précisée par spectroscopie d'émission optique et par le suivi de la pression totale et du potentiel d'autopolarisation. Les conditions adéquates d'élaboration qui ont été définies, ont permis de déposer des films dont la composition varie presque linéairement, entre celles du nitrure et de l'oxyde de silicium. Ces couches sont formées par un mélange de nanophases de type SiO2 et Si3N4 incorporées dans une phase de SiOxNy amorphe. Des liaisons pendantes ont aussi été détectées sur les atomes de silicium. Fluorescence polarization assay (FPA), a serological assay, was validated as an alternative test for the rapid and cost-effective diagnosis of bovine brucellosis, with the aim of improving the control of brucellosis in South Africa. The FPA is anticipated to distinguish between vaccinated and infected cattle, circumventing the challenge associated with the tests that are currently used. Positive cattle serum samples (n =420) confirmed by Complement Fixation Test were tested in conjunction with serum samples (n = 446) from non-infected cattle initially tested on Rose Bengal Test, CFT and compared with FPA. The optimum cut-off value that offers the highest diagnostic sensitivity (Dsn) and diagnostic specificity (Dsp) was determined as 87 mP with the use of ROC analysis. The Dsn and Dsp of FPA using this cut-off value was calculated at 99.09% - 100% and 68.09%- 76.61% respectively with a 95% confidence interval (cl). The area under curve (AUC) was calculated at 0.9842 with a 95% standard error (S.E) of 0.005532 with positive and negative likelihood ratio (+LR) (-LR) at 3.643 and 1.002, respectively. The FPA was found to be as effective as CFT and should be considered because of its accuracy and other advantages such as speed, high throughput and the objectivity of the interpretation of results that can be obtained electronically by the (PHERAstar) machine. The test should be included in routine serological diagnosis for brucellosis.

Abstract:
HIVs are classified into types 1 or 2, groups, subtypes, and specific isolates. They infect CD4+ T-lymphocytes and monocyte/ macrophages. Infection and destruction of CD4+ T-lymphocytes occurs at the pace of billions per day. This massive HIV and CD4+ T-lymphocyte turnover, combined with mutation rate, results in extensive HIV sequence polymorphism. After an average of 11 years, CD4 decline from unbalanced turnover, leads to AIDS and inexorably death. In this thesis I first determined that HIV-1 in Vancouver are of envelope and LTR group M, subtype B. I then extensively investigated the LTR. The LTR contains sequences responsible for RNA pol II recognition and transcription of the HIV-1 genes. I first derived 476 LTR sequences from patients at all stages of HIV-1 infection. I found a surprising number of point and length polymorphisms, predicted to impair binding to targets for dogmatically entrenched transcription factors deemed essential to HIV-1. Some are novel, others have previously been described. I further investigated the function for some LTR variants. I found a wide range of transcription activities, and correlate some to specific sequence polymorphisms. I found no obvious correlation with disease stage for any polymorphism. A -121 position polymorphism, that I termed the Most Frequent Naturally occurring Length Polymorphism (MFNLP), was further investigated. MFNLPs occurred in 38% of patients I sampled. I found no phenotype for MFNLP bearing LTRs. Importantly, I first propose and then prove that MFNLPs all harbor binding sites for the recently characterized transcription factor RBF-2, known to bind the HIV-1 LTR. Thus the MFNLP duplicates an RBF-2 site. Although none of the postulated transcription factor binding sites of the HIV-1 LTR escapes mutation in vivo, only an RBF-2 site is invariably compensated by co-occurrence of it's duplication. Because of MFNLPs, I found an RBF-2 target to be the most conserved site of naturally occurring HIV-1 LTRs. Using MFNLP deleted isogenic mutants, I found a repressive phenotype for the MFNLPs, but only in cells with detectable levels of RBF-2. I further describe the biochemical purification of RBF-2. Analysis of oligo-affinity purified RBF-2 suggests it is a novel complex. All LTRs I have examined, that are derived from patients that progress towards AIDS, have the ability to bind RBF-2. Assuming the current view that viral turnover is itself pathogenic, then by extension, I conclude that the RBF-2 cis-acting element, encompassed in MFNLPs, is required for HIV-1 pathogenesis in vivo. The elements which constitute Group 1 of the Periodic Table are known as the alkali metals. They are lithium Li, sodium Na, potassium K, rubidium Rb, cesium Cs, and francium Fr. (Sometimes the NH4+ ion is included among these since it resembles K+ or Rb+ in many of its reactions.) All six of the elements have atoms characterized by an outer electron structure of ns1 with n representing the principal quantum number. The elements exhibit marked resemblances to each other with Li deviating the most. This deviation is assignable to the small size of Li which causes the positive charge of Li+ to be concentrated, that is, the charge density is high. All of the elements exhibit oxidation numbers of 0 and I, with exceptions being rare, such that their chemistries are dominated by the oxidation state I. The six metals are exceptionally reactive, being strong reductants, reacting with HOH at all pH values to give H2 and M+, and having hydroxides MOH which are strong and soluble. Ionic sizes in pm for the members of the group are as follows: Li (76), Na (102), K (139), Rb (152), Cs (167), and Fr (180). The E° values for the M+/M couples are as follows: Li (−3.04 v), Na (−2.71 v), K (−2.93 v), Rb (−2.92 v), Cs (−2.92 v), and Fr (about −3.03 v). a. E–pH diagram. The E–pH diagram for 10−1.0 M Li is presented in Figure 5.1. The figure legend provides an equation for the line that separates Li+ and Li. The horizontal line appears at an E value of −3.10 v. Considerably above the Li+/Li line, the HOH ≡ H+/H2 line appears, which indicates that Li metal is unstable in HOH, reacting with it to produce H2 and Li+. Note further that Li+ dominates the diagram reflecting that the aqueous chemistry of Li is largely that of the ion Li+.

Red blood cells (RBCs) or erythrocytes are biconcave disks with diameter of 8μm and thickness of 2 μm. RBC can be considered as a nucleus free deformable capsule enclosed by a flexible membrane and consisting of an incompressible viscous fluid [8, 9]. The deformation of RBCs is thought to play a major role in both RBC dynamics and functionality. Previous studies have indicated that two important characteristics related to deformation of the RBC [1]: i) the interior of the RBC is a liquid state that surrounded by an elastic membrane, ii) the biconcave shape of the RBC enables it to deform into a wide varieties of shapes without inducing any stresses in the cell membrane.

Abstract:
This paper develops a cost model for onshore wind farms in the U.S.. This model is then used to analyze the influence of different designs and economic parameters on the cost of a wind farm. A response surface based cost model is developed using Extended Radial Basis Functions (E-RBF). The E-RBF approach, a combination of radial and non-radial basis functions, can provide the designer with significant flexibility and freedom in the metamodeling process. The E-RBF based cost model is composed of three parts that can estimate (i) the installation cost, (ii) the annual Operation and Maintenance (O&M) cost, and (iii) the total annual cost of a wind farm. The input parameters for the E-RBF based cost model include the rotor diameter of a wind turbine, the number of wind turbines in a wind farm, the construction labor cost, the management labor cost and the technician labor cost. The accuracy of the model is favorably explored through comparison with pertinent real world data. It is found that the cost of a wind farm is appreciably sensitive to the rotor diameter and the number of wind turbines for a given desirable total power output.

Abstract:
This paper explores the effectiveness of the recently developed surrogate modeling method, the Adaptive Hybrid Functions (AHF), through its application to complex engineered systems design. The AHF is a hybrid surrogate modeling method that seeks to exploit the advantages of each component surrogate. In this paper, the AHF integrates three component surrogate models: (i) the Radial Basis Functions (RBF), (ii) the Extended Radial Basis Functions (E-RBF), and (iii) the Kriging model, by characterizing and evaluating the local measure of accuracy of each model. The AHF is applied to model complex engineering systems and an economic system, namely: (i) wind farm design; (ii) product family design (for universal electric motors); (iii) three-pane window design; and (iv) onshore wind farm cost estimation. We use three differing sampling techniques to investigate their influence on the quality of the resulting surrogates. These sampling techniques are (i) Latin Hypercube Sampling (LHS), (ii) Sobol’s quasirandom sequence, and (iii) Hammersley Sequence Sampling (HSS). Cross-validation is used to evaluate the accuracy of the resulting surrogate models. As expected, the accuracy of the surrogate model was found to improve with increase in the sample size. We also observed that, the Sobol’s and the LHS sampling techniques performed better in the case of high-dimensional problems, whereas the HSS sampling technique performed better in the case of low-dimensional problems. Overall, the AHF method was observed to provide acceptable-to-high accuracy in representing complex design systems.

At the aim of alleviating the computational burden of complicated engineering optimization problems, metamodels have been widely employed to approximate the expensive blackbox functions. Among the popular metamodeling methods RBF metamodel well balances the global approximation accuracy, computational cost and implementation difficulty. However, the approximation accuracy of RBF metamodel is heavily influenced by the width factors of kernel functions, which are hard to determine and actually depend on the numerical behavior of expensive functions and distribution of samples. The main contribution of this paper is to propose an optimized RBF (ORBF) metamodel for the purpose of improving the global approximation capability with an affordable extra computational cost. Several numerical problems are used to compare the global approximation performance of the proposed ORBF metamodeling methods to determine the promising optimization approach. And the proposed ORBF is also adopted in adaptive metamodel-based optimization method. Two numerical benchmark examples and an I-beam optimization design are used to validate the adaptive metamodel-based optimization method using ORBF metamodel. It is demonstrated that ORBF metamodeling is beneficial to improving the optimization efficiency and global convergence capability for expensive engineering optimization problems.

Abstract:
The probability that the safety I&C system fails to actuate or advertently actuates RT or ESF functions, in part, essentially determines whether a nuclear power plant could operate safely and efficiently. Since more conservative assumptions and simplifications are introduced during the analysis, this paper achieves solid results by performing the modeling and calculation based on a relatively simple approach, the reliability block diagram (RBD) method. A typical safety I&C platform structure is involved in the model presented in this paper. From the perspective of conservation and simplicity, some assumptions are adopted in this paper. A group of formulas is derived in this paper based on Boolean algebra, probability theory, basic reliability concepts and equations, to facilitate the calculations of probabilities that the safety I&C system fails to actuate or advertently actuates RT or ESF functions. All the inputs of the analysis and calculation in this paper, which includes the I&C platform structure, the constitution of the hardware modules, and reliability data, are referenced to the nuclear power plant universal database where applicable. Although the conclusion drawn in the paper doesn’t apply to the I&C platform assessment for a specific plant, the method of modeling and process of analysis provides an illustration of an alternative quantitative reliability assessment approach for a typical safety I&C system installed in the nuclear power plant.

Abstract:
Purpose: Bodybuilding becomes more visible and acceptable within mainstream society thanks to social media, which is promoting, and developing grooving interest in bodies, fit-ness and active lifestyle. However, this is concerning knowing that according to the latest world anti-doping agency report bodybuilding is one of two sports with the highest number of Anti-Doping Rule Violations (ADRVs) committed by athletes. This study aimed to evaluate doping attitudes and correlates of doping attitudes in top level body builders. Methods: Study included 26 competitive bodybuilders form Croatia. Variables were collect-ed by a previously validated Questionnaire of Substance Use (QSU). Statistical procedures included means and standard deviations (for parametric variables), frequencies and percent-ages (for ordinal and nominal variables). Spearman’s correlations were calculated to deter-mine associations between studied variables. Results: The most positive attitudes are found towards injectable anabolic steroids (mean ± standard deviation; 4.00 ± 1.52), followed by fat burners (3.73 ± 1.46), growth hormone (3.69 ± 1.64), and estrogen blockers (3.60 ± 1.22), oral anabolic steroids (3.58 ± 1.27). Sig-nificant correlation was identified between: (i) result achieved in bodybuilding (RBB) and alcohol consumption, (R= -0.57 p < 0.05) (ii) RBB and subjective knowledge on nutrition (R=0.66, p < 0.05), (iii) RBB and subjective knowledge on doping (R=0.72, p < 0.05). Conclusion: The lack of correlation between self-perceived competence and objectively eval-uated knowledge on nutrition is alarming due to the possible “anchoring effect”, accordingly even though objective knowledge is not correlated with attitudes towards doping substances, it is important to properly educate athletes who are in the misconception of their true knowl-edge.
