Cultured skin fibroblasts were examined from two probands with type II (lethal) osteogenesis imperfecta. One proband had a single base mutation which converted the glycine codon at position alpha 1-244 in the alpha 1(I) chain of procollagen I into a cysteine codon whereas the other had a similar mutation that converted the glycine codon at position alpha 2-787 of the alpha 2(I) chain into a cysteine codon. Both mutations produced post-translational overmodification of procollagen I. The Cys alpha 1-244 mutation, however, had a minimal effect on the thermal stability or secretion of the protein whereas the Cys alpha 2-787 mutation markedly decreased the thermal stability and, apparently as a result, essentially none of the mutated protein was secreted. The results provide clear exceptions to two previous generalizations about the position-specificity of glycine substitutions in procollagen I.

Abstract:
The PB2 subunit of the influenza RNA-dependent RNA polymerase (RdRp) has been identified as a promising target for the treatment of influenza. To expand the chemical space of the known influenza polymerase PB2 inhibitor–pimodivir (formerly VX-787) and improve its pharmacokinetic profile, two pimodivir analogs containing 2,3-dihydro-imidazopyridine fragment (comp. I and comp. II) were designed, synthesized, and evaluated for anti-influenza virus activity. In the cytopathic effect (CPE) inhibition assay, comp. I and comp. II showed IC50 values of 0.07 and 0.09 μM for A/Puerto Rico/8/34 (H1N1) and 0.04 and 0.07 μM for A/Hong Kong/8/68 (H3N2), respectively. Protein-binding affinity assay results showed a concentration-dependent association and dissociation pattern, with KD values of 1.398 and 1.670 μM, respectively. In vitro metabolic stability assays showed that comp. I and comp. II exhibited good stability to liver microsomes and considerably less sensitivity to aldehyde oxidase compared to pimodivir. The binding modes of comp. I and comp. II were similar to those of VX-787; however, comp. I and comp. II had lower structural adaptability to PB2 than VX-787. Our results provide helpful information regarding the structure–activity relationship for the design of novel PB2 inhibitors and a reference for the development of drugs containing 2,3-dihydro-imidazopyridine fragments.

Jednym z wielkich nauczycieli duchowych tego okresu był Józef Hazzāyā, który dokonał syntezy nauki poprzedników. Fundament jego twórczości stanowi nauka o kontemplacji Boga, która była obca dla tradycji Antiocheńskiej i dlatego została potępiona jako heretycka przez patriarchę Tymoteusza I na synodzie w latach 786-787. W ar­tykule dokonano analizy nauki Józefa o kontemplacji Boga w celu krytycznego spojrzenia na podstawy jego potępienia. Autorka przeprowadziła analizę termi­nów, które Józef stosuje na określenie stanu kontemplacji Boga, a także przedsta­wiła specyfikę jego nauczaniu w tej kwestii. W ostatnim punkcie artykułu został przedstawiony krótki zarys chrystologii Kościoła Wschodu oraz nauka dotycząca doświadczenia w sferze kontemplacji Boga.

Abstract:
Abstract Background Colistin is a last-resort antibiotic for multidrug resistant gram-negative infections. Recently, a new allele of the mobile colistin resistance (mcr) gene family designated mcr-9, has been reported. However, its clinical and phenotypic significance remains unclear. Methods The Centers for Diseases Control and Prevention-funded Georgia Emerging Infections Program (EIP) performs population- and laboratory- based surveillance for CRE isolated from sterile sites or urine in metropolitan Atlanta, GA including standardized chart abstraction. We queried genomes of carbapenem-resistant Enterobacterales (CRE) for mcr-9 from a convenience sample of Georgia EIP clinical isolates between 2012-2017. Isolates underwent phenotypic characterization by broth microdilution and population analysis profiling. Nine available E. cloacae (two mcr-9 positive, seven mcr-9 negative) genomes from the National Institutes of Health were included in downstream genomic analysis. Fastq files underwent de novo assembly, annotation and AMR and virulence gene prediction, pan-genome association analysis, pairwise comparisons of average nucleotide identity and phylogenetic tree construction based on core genes. We compared characteristics and outcomes of mcr-9 positive and negative CRE cases. Results Among 449 sequenced CRE genomes, thirteen (2.9%) were found to harbor mcr-9, all of which were E. cloacae. Fourteen mcr-9 negative E. cloacae (n=14) were included as a comparative group. E. cloacae was most commonly isolated from the urine (22/24, 86%), and none were community associated. The median colistin MIC, rates of heteroresistance and inducible resistance were similar between mcr-9 positive and negative isolates (Table 1). 90-day mortality was high in both mcr-9 positive (31%) and negative (7% cases (p=0.28, Table 1). Phylogenetic analysis revealed no geo-temporal clustering (Figure 1). Plasmid-associated genes were significantly associated with the presence of mcr-9 (p< 0.001). Pairwise comparisons of average nucleotide identity on the assembled genomes were performed with the Mashmap method using fastANI v1.32. ABSTRACT The knowledge of root canal morphology and the existing anatomical variations is essential for successful endodontic therapy. This report presents an extraordinary case of unusual tooth morphology involving the mandibular first molar with a single root and a single canal in a patient. Endodontic treatment of the affected tooth 36 was performed. Cone beam computed tomography (CBCT) images confirmed our diagnostic and therapeutic measures after endodontic management of the teeth. This morphologic variation has been reported once in the literature. The availability of three-dimensional images further provided the opportunity for the precise description of the anatomy of mandibular first molar with single roots and single canals. Syftet med denna kvalitativa studie var att få kännedom i hur lärare i år 1-3 hanterar det ökade ansvaret med att erbjuda sina elever daglig fysisk aktivitet. Ambitionen var även att få insikt i lärarnas kunskaper kring fysiska aktiviteters effekter samt få insyn i deras kunskaper kring Läroplanens (Lpo 94) innehåll rörande daglig fysisk aktivitet. Utifrån semistrukturerade intervjuer kunde slutsatsen dras att alla tio medverkande lärare ansåg att det var betydelsefullt att arbeta med fysiska aktiviteter. Samtliga arbetade i mer eller mindre stor omfattning med fysiska aktiviteter i klassrummet, både som pausmoment och integrerat i undervisningen. Lärarna menade att elevernas behov lämpligen måste styra aktiviteternas omfattning och form. Slutligen var det dock en fråga om prioritet och en avvägning av vad eleverna klarade av som avgjorde mängden. Den ambition som fanns resulterade följaktligen inte alltid i att de fysiska aktiviteterna genomfördes i praktiken.

Abstract:
The main objectives of this thesis were to analyse the relative roles of food availability and predation on the distribution, population dynamics and habitat selection of small mammals in Mediterranean areas of the Iberian Peninsula.
Main results and conclusions
Composition and abundance of small mammals communities can be completely ascertained by using two indirect methods of small mammals sampling, genet scats and barn owl pellets. Both methods reported all the small mammals species known to be present in the study area. Genet scats were used for the first time to study the composition of small mammal communitites with a biogeographical application.
Small mammals species richness was negatively related to a gradient of productivity, which decreased with elevation. The increase of species density along elevation was observed at two spatial scales (plot and elevation belt). Small mammals mean abundance decreased with elevation, and this pattern was may be related to the decreased availability of resources and harsh environmental conditions with elevation.
The interaction between negative feedback (density-dependence) and natural environmental disturbances (seasonal and interannual changes in cumulative rainfall) seem to be relevant forces driving the dynamics of wood mouse populations in Mediterranean forests. The role of intraspecific competition and climate were equally responsible of the strong population oscillations observed during three consecutive years.
All small mammal species studied showed significant associations with gradients of short vegetation cover, either shrubs or herbs, in natural and man-disturbed habitats. Overall, species richness and abundance also showed positive associations with gradients of short vegetation cover. These patterns highlighted the dependence of small mammals on vegetation which provides food and shelter.
Early successional stages supported larger small mammal abundances than unburnt forests after controlling for structural differences between plots. I hypothesised that early successional stages would be favourable habitats to small mammals due to a combination of reduced predation risk (high vegetation cover) and reduced predation pressure (less predators), and unburnt areas would be unfavourable habitats due to a combination of increased predation risk (low vegetation cover) and increased predation pressure (more predators). Fire-related fragmentation could have promoted the isolation of forest predators (owls and carnivores) in unburned forest patches, a fact that could have produced a higher predation pressure for small mammals. Behavioural responses of small mammals to predation were also surmised, with positive associations to vegetation cover in unburnt forests and negative associations in early burned habitats. These changes in small mammal-microhabitat relationships suggest differences in perceived predation risk that may have promoted the observed changes in microhabitat use. Foraging activity of mice shifted spatially and temporally in response to the presence of genets at relevant spatial scales, and decissions about how often and how long to forage were affected by the foraging activity of genets. Summarising, our findings demonstrate that predators have a prominent role in determining the patterns of distribution of mice along the post-fire succession, and that this role is mediated, at least in part, by indirect, behavioural responses of foraging mice to increased predator pressure along such succession.
Effects of grazing on small mammals were mainly due to decreased availability of food and refuges for surviving harsh climatic conditions during winter, but not to food quality or increased predation risk. Taken together, our results indicate that small mammal communities in Mediterranean montane grasslands are bottom-up controlled by interspecific competition for food with cattle and by the limiting role of the availability of winter refuges, whereas top-down control due to generalists predators does not appear to be at work in these systems.

Abstract:
Luminogens are molecules that emit light upon exposure to high-energy light, and fluorophores are one class of luminogens. Applications of fluorophores range from microviscosity sensors to light emitting diodes (LEDs), as well as biosensors, just to name a few. Many of these applications require the fluorophore to be in the aggregate or solid state. Some fluorophores become highly emissive in the aggregate state; these fluorophores are aggregation-induced emission (AIE) luminogens. Currently, very few quantum mechanical mechanisms have been proposed to describe the unique AIE behavior of luminogens. Boron difluorohydrazone (BODIHY) dyes are a new type of AIE fluorophore. The bright emission is from the S>1 excited state (“anomalous” emission) contrary to Kasha’s Rule. Thus, the mechanism Suppression of Kasha’s Rule (SOKR) was proposed to be responsible for the family of BODIHY dyes. We hypothesize that the SOKR mechanism can explain AIE as well as the anomalous emission of other fluorophores. New BODIHY derivatives (para-CO2H BODIHY, aluminum difluorohydrazone (ALDIHY), and paranitro ALDIHY) were predicted to be bright anomalous fluorophores through density functional theory (DFT) and time-dependent DFT (TDDFT) investigations. In addition, a series of anomalous fluorophores were investigated to determine if their photophysical properties could be explained by the SOKR mechanism (azulene, 1,6-diphenyl-1,3,5hexatriene, and zinc tetraphenylporphyrin). Finally, several triazolopyridinium and triazoloquinolinium dyes were computationally investigated by DFT and TDDFT calculations, and an accurate computational model for the large Stokes shifts of these dyes was developed. In conclusion, a better understanding of the photophysical properties through DFT and TDDFT modeling and spectroscopic investigation of hydrazone-based fluorophores has been achieved. In addition, the metal active sites and cofactors of metalloproteins were probed by optical spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, and DFT modeling. In conjunction, these techniques can be used to elucidate the electronic structure responsible for the unique function of these metalloproteins. Specifically, a novel ironsulfur cluster of a metalloprotein that may be involved in endospore formation of Clostridium difficile, CotA, was characterized by magnetic circular dichroism (MCD) spectroscopy. We propose that CotA contains a high-spin [4Fe-4S] cluster and a Rieske [2Fe-2S] cluster. It appears that the multimerization of the protein is related to the cluster conversion at the interface of monomeric subunits where two [2Fe-2S] clusters combine to form the [4Fe-4S] cluster. In addition, a putative cobalamin acquisition protein from Phaeodactylum tricornutum, CBA1, was not expressed at sufficient concentrations in Escherichia coli for spectroscopic investigation. Finally, a new technique was developed using cobalt-59 NMR spectroscopy to better understand the nucleophilic character of cobalt tetrapyrroles, such as cobalamin (vitamin B12), as biological cofactors as well as synthetic catalysts. New insight into the electronic structure provides valuable information related to the mechanism of these metalloproteins.

This essay is a study about school success for immigrant pupils whose ages are between 16 and 19 years when they arrive to Sweden.

The purpose of this study is to examine pupils difficulties in the school and the faktors which affect their success in the school and even possibilities which they can be offered by the school.

This research carried out at two upper secondary schools in the municipality of “Södertälje”. The method which I have used is a qualitative research method which depends on interviews.

The results of the research shows pupils difficulties and the different faktors which influence their success in the school.

My conclusions are that these pupils have many difficulties and to reach a successful schooling, the shcool have to give these pupils a variety of possibilities which builds with pupils self possibilities a foundation for a good and successful schooling.

The research also shows that the both schools are not giving enough possibilities to these pupils. The education system in these schools doesn’t fits for multi – lingual pupils. The pupils in these schools are trying to adapt themselves to the education system.

Abstract:
Through calculations as well as a literature study the purpose of this work was to apply modern energy efficiency measures on an old existing building and on the same time take economical aspects and thermal inside climate into account. The work also carries out a case study on the existing building Lådberga 1:78. The calculations that has been carried out acknowledges a change of the building’s thermal transmittance for each measure that has been simulated as well as a change of the buildings active heating. In order to carry out the comparison between the buildings active heating, before and after applying the modern building measures, the buildings current energy consumption was calculated.  To examine the proposed actions from an economical point of view, the payback method was used. When applying actions that include additional interior insulation, diffusion calculations was used. The energy efficiency action that was calculated to be most financially viable for the building was exchange of the heat system. This action included a change from an existing pellet boiler to a more efficient geothermal heat pump as well as the exchange of four radiators.  The second-best alternative was to add insulation to the attic. Also, one simple yet effective action that could be done in the building to improve the thermal inside climate was to mount sealing stripes around the existing windows. The energy efficiency actions that was found to be least financially viable was exchange of windows, doors as well as adding insulation to the walls. Calculations was carried out through derivatives that has been acknowledged for during the current study. The results that has been reported has been chosen regarding the general understanding of the study as well as the relevance it has on the final results in order to clearly answer the questions at issue. Tillgång till energi är i dagens samhälle en viktig del i vardagen. Energi nyttjas bland annat för att värma upp tappvarmvatten och bostäder samt för att förse byggnader med den energi som behövs för att dessa ska uppnå deras planerade funktioner. Krav på tillåten energianvändning finns för nybyggnationer och vid ändring av befintliga byggnader, dock gäller inte dessa krav för byggnader som väsentligt inte ska ändras. I och med detta har äldre byggnader i många fall en högre energianvändning än yngre byggnationer vilken kan minskas genom byggnadstekniska och installationstekniska åtgärder. Då byggnader energieffektiviseras sänks Sveriges totala energianvändning, kostnaderna för byggnadens aktiva uppvärmning minskar och den termiska komforten i en äldre byggnad kan förbättras. Genom beräkningar samt litteraturstudier är syftet med arbetet att i teorin energieffektivisera en äldre befintlig byggnad med hänsyn till ekonomi och avgränsad termisk komfort. Arbetet bygger på en empirisk studie vilken innefattar en fallstudie som har utförts på villan Lådberga 1:78 samt litteraturinsamling. Den datainsamling som har nyttjats för arbetet är fältmätningar, observationer samt dokumentanalyser. De energieffektiviseringsåtgärder som har undersökts i arbetet har valts ut med hjälp av arbetets litteraturstudie och för att undersöka hur olika energieffektiviseringsåtgärder påverkar den aktiva uppvärmningen i Lådberga 1:78 har beräkningar utförts. De beräkningar som har utförts uppvisar förändringar av byggnadens värmegenomgångskoefficienter vid respektive åtgärd samt en förändring av byggnadens aktiva uppvärmning. För att möjliggöra en jämförelse mellan byggnadens aktiva uppvärmning innan utförda åtgärder samt efter utförda åtgärder har även byggnadens energianvändning innan energieffektivisering beräknats. För att undersöka åtgärderna ur ett ekonomiskt perspektiv med hänsyn till återbetalningstid har Payback-metoden nyttjats. Vid åtgärder vilka innefattar tilläggsisolering har diffusionsberäkningar utförts. De åtgärder som har undersökts i arbetet är byggnadstekniska åtgärder i klimatskalet samt byte av värmesystem. Den åtgärd som har beräknats vara den mest lönsamma energieffektiviseringsåtgärden i byggnaden är byte av värmesystem. Åtgärden inkluderar byte från en pelletspanna till en bergvärmepump inklusive byte av fyra radiatorer. Denna åtgärd medför ett inbetalningsöverskott på cirka 18 100 kr/år och återbetalningstiden har beräknats till 10 år. Installationens livslängd har uppskattats vara mellan 20 till 40 år beroende på installationsdel. Den näst mest effektiva energieffektiviseringsåtgärden är tilläggsisolering av byggnadens vindsbjälklag. Denna åtgärd medför ett inbetalningsöverskott på cirka 2700 kr/år och har återbetalningstiden 14 år. Åtgärdens livslängd har uppskattats till minst byggnadens livslängd. Nackdelarna med åtgärderna är att båda leder till en minskad mängd värme till byggnadens kallvind vilket kan leda till fuktproblem, installation av en bergvärmepump påverkar även byggnadens självdragsventilation. De åtgärder som inte är lönsamma vid energieffektivisering av Lådberga 1:78 är byte av fönster, byte av dörrar samt tilläggsisolering av byggnadens träregelytterväggar då dessa åtgärder har återbetalningstider mellan 61 år till 92 år. Ytterligare en åtgärd som inte bör genomföras är tilläggsisolering av byggnadens betongytterväggar inifrån, detta på grund av att den relativa luftfuktigheten mellan isoleringen och betongen blir 100 %. De byggnadsdelar i byggnadens klimatskal vilka inte har undersökts via beräkningar är byggnadens krypgrund samt byggnadens betongplatta på mark. Byggnadens aktiva uppvärmning innan energieffektivisering är enligt beräkningar . För att förbättra den termiska komforten i byggnaden finns det möjlighet att utföra mindre förändringar av byggnadsdelarna i byggnadens klimatskal. Exempel på simpla åtgärder vilka kan utföras i Lådberga 1:78 är montering av tätningslister samt isolerglas på byggnadens fönster samt noggrann drevning runt av byggnadens fönster och dörrar. La recerca de la relació entre els enfocaments artístic i tècnic en les especialitats com la veu i del violí, conduirà a la seva aplicació pràctica i innovadora a l'aula de violí. El treball de camp es concreta en: la cerca de tots aquells recursos en la tècnica i la interpretació del cant a través d'entrevistes a cantants; en l'assistència en classes de cant; en l'escolta atenta de cantants; en la lectura de literatura específica; i finalment, en l'aplicació de tot això en el treball amb els alumnes de violí, registrant els resultats. L'objectiu principal d'aquesta tesi és investigar i compartir aquest estudi i recerca amb altres músics i persones interessades en el tema, així com despertar una altra consciència per a tots aquells instrumentistes de corda que desitgin incorporar aquest treball en la seva vocació de músic i el professor. És a dir, obrir una nova línia de treball que faciliti i engrandeixi al violinista com a tal, que pugui fer desaparèixer aquestes barreres o impediments que tècnicament de vegades no aconseguim superar, si no des d'una altra visió totalment diferent i innovadora.
El presente trabajo podría clasificarse dentro del ámbito de la psicopedagogía y la interpretación, donde el enfoque interdisciplinario pretende mejorar las deficiencias comunes en la ejecución del violín. La investigación de la relación entre los enfoques artístico y técnico en las especialidades como la voz y del violín, conducirá a su aplicación práctica e innovadora en el aula de violín. El trabajo de campo se concreta en: la búsqueda de todos aquellos recursos en la técnica y la interpretación del canto a través de entrevistas a cantantes; en la asistencia en clases de canto; en la escucha atenta de cantantes; en la lectura de literatura específica; y finalmente, en la aplicación de todo ello en el trabajo con los alumnos de violín, registrando los resultados. El objetivo principal de esta tesis es investigar y compartir este estudio y investigación con otros músicos y personas interesadas en el tema, así como despertar otra conciencia para todos aquellos instrumentistas de cuerda que deseen incorporar este trabajo en su vocación de músico y el profesor. Es decir, abrir una nueva línea de trabajo que facilite y engrandezca al violinista como tal, que pueda hacer desaparecer esas barreras o impedimentos que técnicamente a veces no logramos superar, si no desde otra visión totalmente diferente e innovadora.

AbstractThis chapter focuses on the carbon footprint of travelling to academic conferences. The cases I present are the last seven General Conferences of the European Consortium for Political Research (ECPR), which are the biggest European conferences in political science, with up to 2000 participants. My estimations show that the travel-induced carbon footprint of a single conference can amount to more than 2000 tons of greenhouse gases—as much as approximately 270 UK citizens emit in a whole year. The average participant produces between 500 and 1500 kg of CO2-eq per conference round-trip. However, by applying three measures (more centrally located conference venues, the promotion of more land-bound travel and the introduction of online participation for attendees from distant locations), the carbon footprint could be reduced by 78–97 per cent. In 2020, the COVID-19 pandemic caused a general shift towards online conferences—the ECPR switched to a virtual event as well. Estimating the carbon footprint of this online-only conference in a more detailed manner shows that the travel-induced carbon emissions—if the event had taken place in physical attendance as originally intended—would have been between 250 and 530 times higher than those from the online conference.

78 unrelated X chromosomes from Southern Chinese (56 normal and 22 haemophiliac) were studied. DNA was restricted by Bel I, Bgl I or Taq I and hybridized to 3' factor VIII:C cDNA probe (5 kb, Chiron) or St 14.1 probe(3 kb, Oberle &Mandel) by standard techniques. The intragenic Bel I polymorphic site was positive in 82%, while Bgl I polymorphic site was positive in all. Thus, 29.5%(2 x×0.82 × 0.18) of Chinese females carried the Bel I polymorphism. Asto the Taq I polymorphism in the closely linked DXS52 DNA segment, the incidences for the various alleles were :System I - allele (3) 10.2%, (4) 2.6%, (5) 2.6%,(6) 17.9%, (7) 21.8% and (8) 44.9% System II - α a allele 56%, 6 allele 44%. Approximately 80% of females were heterozygous for two different alleles. Hence the Bel I and Taq I polymorphisms can be used to track the defective factor VIII gene for carrier detection and prenatal diagnosis. Furthermore, their frequencies in the Chinese are different from those previously reported in other ethnic groups.

Gastrointestinal haemorrhage causes a hypercoagulable state and early blood transfusion increases both rebleeding and transfusion requirements [1]. The role of x- and B-adreno-receptors in the mechanism of this hypercoagulable state and the effects of infusions have been studied in NZW rabbits.Haemorrhage was simulated by aspirating 20% of the blood volume from the median ear artery. Coagulation was measured before haemorrhage and at 30-minute intervals using the Biobridge Impedance Clotting Time (ICT). Three different groups of rabbits were used: (a) normal controls, (b) B-blocked with propranolol 1mg/kg, and (c) x- and B-blocked with phentolamine 3mg/kg and propranolol 1mg/kg. Equal volumes of (i) Haemaccel, (ii) fresh blood or (iii) stored blood were then given 30 minutes after haemorrhage.Mean ± sem ICT was 6.0±0.15 minutes before haemorrhage, with no difference between the groups, suggesting that x- and B-blocking drugs have no effect on baseline coagulation. Haemorrhage produced a hypercoagulable state with the mean ICT shortened from 6.1±0.2 to 2,2±0.2 minutes (p<0.01). This response was significantly reduced to 5.2±0.4 and 5.1±0.2 minutes by both B-blockade and combined x- and B-blockade respectively.Haemaccel had no significant effect on the hypercoagulable state, with the ICT going from 2.6±0.2 to 2.8±0.3 minutes. However, fresh and stored blood both reversed hypercoagulability with ICT increasing from 2.6±0.3 to 4.6+0.2 and 5.1±0.1 minutes respectively.In this model the hypercoagulable response to haemorrhage appears to be mainly B- mediated and is partially reversed by both fresh and stored blood, but not by Haemaccel.1. Industry demands new engineering and material solutions. One of these solutions are fibre reinforced polymers. They are light and strong for application as a material for I-beams. An I-beam is the best section for a homogeneous material because of the highest resistance moment. The popularity of composite materials introduces wide use in most branches of engineering and mostly as fibre reinforced polymers (FRP). This paper presents numerical and analytical studies on the mechanical behaviour of the I-beams made of fibre reinforced polymers reinforced by glass fibres (GFRP) and carbon fibres (CFRP) comparing to the structural steel S235JR. Five I-beams with different composite structures and one steel I-beam were numerically tested. Four-point test according to ASTM C 78 – 02 was conducted. Numerical simulation made in SOLIDWORKS software in the Static simulation mode was used. The obtained results were analysed and an attempt to determine the optimal parameters for combination of different composite materials was conducted. As a result of numerical analysis values of deflection and normal stress were obtained for polymer I-beam reinforced by glass and carbon fibres comparing to the steel I-beam.

Background An estimated 292 million people are living with chronic hepatitis B virus (HBV) infection globally, including 223,000 people in Australia. HBV diagnosis and linkage of people living with HBV to clinical care is suboptimal in Australia, with 27% of people living with HBV undiagnosed and 77% not receiving regular HBV clinical care. This systematic review aimed to characterize population-level interventions implemented to enhance all components of HBV care cascade and analyse the effectiveness of interventions. Review questions Question 1: What population-level interventions, programs or policy approaches have been shown to be effective in reducing the incidence of hepatitis B; and that may not yet be fully rolled out or evaluated in Australia demonstrate early effectiveness, or promise, in reducing the incidence of hepatitis B? Question 2: What population-level interventions and/or programs are effective at reducing disease burden for people in the community with hepatitis B? Methods Four bibliographic databases and 21 grey literature sources were searched. Studies were eligible for inclusion if the study population included people with or at risk of chronic HBV, and the study conducted a population-level interventions to decrease HBV incidence or disease burden or to enhance any components of HBV care cascade (i.e., diagnosis, linkage to care, treatment initiation, adherence to clinical care), or HBV vaccination coverage. Studies published in the past 10 years (since January 2012), with or without comparison groups were eligible for inclusion. Studies conducting an HBV screening intervention were eligible if they reported proportion of people participating in screening, proportion of newly diagnosed HBV (participant was unaware of their HBV status), proportion of people received HBV vaccination following screening, or proportion of participants diagnosed with chronic HBV infection who were linked to HBV clinical care. Studies were excluded if study population was less than 20 participants, intervention included a pharmaceutical intervention or a hospital-based intervention, or study was implemented in limited clinical services. The records were initially screened by title and abstract. The full texts of potentially eligible records were reviewed, and eligible studies were selected for inclusion. For each study included in analysis, the study outcome and corresponding 95% confidence intervals (95%CIs) were calculated. For studies including a comparison group, odds ratio (OR) and corresponding 95%CIs were calculated. Random effect meta-analysis models were used to calculate the pooled study outcome estimates. Stratified analyses were conducted by study setting, study population, and intervention-specific characteristics. Key findings A total of 61 studies were included in the analysis. A large majority of studies (study n=48, 79%) included single-arm studies with no concurrent control, with seven (12%) randomised controlled trials, and six (10%) non-randomised controlled studies. A total of 109 interventions were evaluated in 61 included studies. On-site or outreach HBV screening and linkage to HBV clinical care coordination were the most frequent interventions, conducted in 27 and 26 studies, respectively. Question 1 We found no studies reporting HBV incidence as the study outcome. One study conducted in remote area demonstrated that an intervention including education of pregnant women and training village health volunteers enhanced coverage of HBV birth dose vaccination (93% post-intervention, vs. 81% pre-intervention), but no data of HBV incidence among infants were reported. Question 2 Study outcomes most relevant to the HBV burden for people in the community with HBV included, HBV diagnosis, linkage to HBV care, and HBV vaccination coverage. Among randomised controlled trials aimed at enhancing HBV screening, a meta-analysis was conducted including three studies which implemented an intervention including community face-to-face education focused on HBV and/or liver cancer among migrants from high HBV prevalence areas. This analysis demonstrated a significantly higher HBV testing uptake in intervention groups with the likelihood of HBV testing 3.6 times higher among those participating in education programs compared to the control groups (OR: 3.62, 95% CI 2.72, 4.88). In another analysis, including 25 studies evaluating an intervention to enhance HBV screening, a pooled estimate of 66% of participants received HBV testing following the study intervention (95%CI: 58-75%), with high heterogeneity across studies (range: 17-98%; I-square: 99.9%). A stratified analysis by HBV screening strategy demonstrated that in the studies providing participants with on-site HBV testing, the proportion receiving HBV testing (80%, 95%CI: 72-87%) was significantly higher compared to the studies referring participants to an external site for HBV testing (54%, 95%CI: 37-71%). In the studies implementing an intervention to enhance linkage of people diagnosed with HBV infection to clinical care, the interventions included different components and varied across studies. The most common component was post-test counselling followed by assistance with scheduling clinical appointments, conducted in 52% and 38% of the studies, respectively. In meta-analysis, a pooled estimate of 73% of people with HBV infection were linked to HBV clinical care (95%CI: 64-81%), with high heterogeneity across studies (range: 28-100%; I-square: 99.2%). A stratified analysis by study population demonstrated that in the studies among general population in high prevalence countries, 94% of people (95%CI: 88-100%) who received the study intervention were linked to care, significantly higher than 72% (95%CI: 61-83%) in studies among migrants from high prevalence area living in a country with low prevalence. In 19 studies, HBV vaccination uptake was assessed after an intervention, among which one study assessed birth dose vaccination among infants, one study assessed vaccination in elementary school children and 17 studies assessed vaccination in adults. Among studies assessing adult vaccination, a pooled estimate of 38% (95%CI: 21-56%) of people initiated vaccination, with high heterogeneity across studies (range: 0.5-93%; I square: 99.9%). A stratified analysis by HBV vaccination strategy demonstrated that in the studies providing on-site vaccination, the uptake was 78% (95%CI: 62-94%), significantly higher compared to 27% (95%CI: 13-42%) in studies referring participants to an external site for vaccination. Conclusion This systematic review identified a wide variety of interventions, mostly multi-component interventions, to enhance HBV screening, linkage to HBV clinical care, and HBV vaccination coverage. High heterogeneity was observed in effectiveness of interventions in all three domains of screening, linkage to care, and vaccination. Strategies identified to boost the effectiveness of interventions included providing on-site HBV testing and vaccination (versus referral for testing and vaccination) and including community education focussed on HBV or liver cancer in an HBV screening program. Further studies are needed to evaluate the effectiveness of more novel interventions (e.g., point of care testing) and interventions specifically including Indigenous populations, people who inject drugs, men who have sex with men, and people incarcerated.

Abstract:
The main goal of this research plan was to elucidate regulatory mechanisms controlling the development, function of the bovine corpus luteum (CL). The CL contains two different sterodigenic cell types and therefore it was necessary to obtain pure cell population. A system was developed in which granulosa and theca interna cells, isolated from a preovulatory follicle, acquired characteristics typical of large (LL) and small (SL) luteal cells, respectively, as judged by several biochemical and morphological criteria. Experiments were conducted to determine the effects of granulosa cells removal on subsequent CL function, the results obtained support the concept that granulosa cells make a substaintial contribution to the output of progesterone by the cyclic CL but may have a limited role in determining the functional lifespan of the CL. This experimental model was also used to better understand the contribution of follicular granulosa cells to subsequent luteal SCC mRNA expression. The mitochondrial cytochrome side-chain cleavage enzyme (SCC), which converts cholesterol to pregnenolone, is the first and rate-limiting enzyme of the steroidogenic pathway. Experiments were conducted to characterize the gene expression of P450scc in bovine CL. Levels of P450scc mRNA were higher during mid-luteal phase than in either the early or late luteal phases. PGF 2a injection decreased luteal P450scc mRNA in a time-dependent manner; levels were significantly reduced by 2h after treatment. CLs obtained from heifers on day 8 of the estrous cycle which had granulosa cells removed had a 45% reduction in the levels of mRNA for SCC enzymes as well as a 78% reduction in the numbers of LL cells. To characterize SCC expression in each steroidogenic cell type we utilized pure cell populations. Upon luteinization, LL expressed 2-3 fold higher amounts of both SCC enzymes mRNAs than SL. Moreover, eight days after stimulant removal, LL retained their P4 production capacity, expressed P450scc mRNA and contained this protein. In our attempts to establish the in vitro luteinization model, we had to select the prevulatory and pre-gonadotropin surge follicles. The ratio of estradiol:P4 which is often used was unreliable since P4 levels are high in atretic follicles and also in preovulatory post-gonadotropin follicles. We have therefore examined whether oxytocin (OT) levels in follicular fluids could enhance our ability to correctly and easily define follicular status. Based on E2 and OT concentrations in follicular fluids we could more accurately identify follicles that are preovulatory and post gonadotropin surge. Next we studied OT biosynthesis in granulosa cells, cells which were incubated with forskolin contained stores of the precursor indicating that forskolin (which mimics gonadotropin action) is an effective stimulator of OT biosynthesis and release. While studying in vitro luteinization, we noticed that IGF-I induced effects were not identical to those induced by insulin despite the fact that megadoses of insulin were used. This was the first indication that the cells may secrete IGF binding protein(s) which regonize IGFs and not insulin. In a detailed study involving several techniques, we characterized the species of IGF binding proteins secreted by luteal cells. The effects of exogenous polyunsaturated fatty acids and arachidonic acid on the production of P4 and prostanoids by dispersed bovine luteal cells was examined. The addition of eicosapentaenoic acid and arachidonic acid resulted in a dose-dependent reduction in basal and LH-stimulated biosynthesis of P4 and PGI2 and an increase in production of PGF 2a and 5-HETE production. Indomethacin, an inhibitor of arachidonic acid metabolism via the production of 5-HETE was unaffected. Results of these experiments suggest that the inhibitory effect of arachidonic acid on the biosynthesis of luteal P4 is due to either a direct action of arachidonic acid, or its conversion to 5-HETE via the lipoxgenase pathway of metabolism. The detailed and important information gained by the two labs elucidated the mode of action of factors crucially important to the function of the bovine CL. The data indicate that follicular granulosa cells make a major contribution to numbers of large luteal cells, OT and basal P4 production, as well as the content of cytochrome P450 scc. Granulosa-derived large luteal cells have distinct features: when luteinized, the cell no longer possesses LH receptors, its cAMP response is diminished yet P4 synthesis is sustained. This may imply that maintenance of P4 (even in the absence of a Luteotropic signal) during critical periods such as pregnancy recognition, is dependent on the proper luteinization and function of the large luteal cell.

Research objectives were: 1) To study the effect of diet energy density, level of exercise, thermal conditions and reproductive state on cardiovascular function as it relates to oxygen (O2) mobilization. 2) To validate the use of heart rate (HR) to predict energy expenditure (EE) of ruminants, by measuring and calculating the energy balance components at different productive and reproductive states. 3) To validate the use of HR to identify changes in the metabolizable energy (ME) and ME intake (MEI) of grazing ruminants. Background: The development of an effective method for the measurement of EE is essential for understanding the management of both grazing and confined feedlot animals. The use of HR as a method of estimating EE in free-ranging large ruminants has been limited by the availability of suitable field monitoring equipment and by the absence of empirical understanding of the relationship between cardiac function and metabolic rate. Recent developments in microelectronics provide a good opportunity to use small HR devices to monitor free-range animals. The estimation of O2 uptake (VO2) of animals from their HR has to be based upon a consistent relationship between HR and VO2. The question as to whether, or to what extent, feeding level, environmental conditions and reproductive state affect such a relationship is still unanswered. Studies on the basic physiology of O2 mobilization (in USA) and field and feedlot-based investigations (in Israel) covered a , variety of conditions in order to investigate the possibilities of using HR to estimate EE. In USA the physiological studies conducted using animals with implanted flow probes, show that: I) although stroke volume decreases during intense exercise, VO2 per one heart beat per kgBW0.75 (O2 Pulse, O2P) actually increases and measurement of EE by HR and constant O2P may underestimate VO2unless the slope of the regression relating to heart rate and VO2 is also determined, 2) alterations in VO2 associated with the level of feeding and the effects of feeding itself have no effect on O2P, 3) both pregnancy and lactation may increase blood volume, especially lactation; but they have no effect on O2P, 4) ambient temperature in the range of 15 to 25°C in the resting animal has no effect on O2P, and 5) severe heat stress, induced by exercise, elevates body temperature to a sufficient extent that 14% of cardiac output may be required to dissipate the heat generated by exercise rather than for O2 transport. However, this is an unusual situation and its affect on EE estimation in a freely grazing animal, especially when heart rate is monitored over several days, is minor. In Israel three experiments were carried out in the hot summer to define changes in O2P attributable to changes in the time of day or In the heat load. The animals used were lambs and young calves in the growing phase and highly yielding dairy cows. In the growing animals the time of day, or the heat load, affected HR and VO2, but had no effect on O2P. On the other hand, the O2P measured in lactating cows was affected by the heat load; this is similar to the finding in the USA study of sheep. Energy balance trials were conducted to compare MEI recovery by the retained energy (RE) and by EE as measured by HR and O2P. The trial hypothesis was that if HR reliably estimated EE, the MEI proportion to (EE+RE) would not be significantly different from 1.0. Beef cows along a year of their reproductive cycle and growing lambs were used. The MEI recoveries of both trials were not significantly different from 1.0, 1.062+0.026 and 0.957+0.024 respectively. The cows' reproductive state did not affect the O2P, which is similar to the finding in the USA study. Pasture ME content and animal variables such as HR, VO2, O2P and EE of cows on grazing and in confinement were measured throughout three years under twenty-nine combinations of herbage quality and cows' reproductive state. In twelve grazing states, individual faecal output (FO) was measured and MEI was calculated. Regression analyses of the EE and RE dependent on MEI were highly significant (P<0.001). The predicted values of EE at zero intake (78 kcal/kgBW0.75), were similar to those estimated by NRC (1984). The EE at maintenance condition of the grazing cows (EE=MEI, 125 kcal/kgBW0.75) which are in the range of 96.1 to 125.5 as presented by NRC (1996 pp 6-7) for beef cows. Average daily HR and EE were significantly increased by lactation, P<0.001 and P<0.02 respectively. Grazing ME significantly increased HR and EE, P<0.001 and P<0.00l respectively. In contradiction to the finding in confined ewes and cows, the O2P of the grazing cows was significantly affected by the combined treatments (P<0.00l ); this effect was significantly related to the diet ME (P<0.00l ) and consequently to the MEI (P<0.03). Grazing significantly increased O2P compared to confinement. So, when EE of grazing animals during a certain season of the year is estimated using the HR method, the O2P must be re measured whenever grazing ME changes. A high correlation (R2>0.96) of group average EE and of HR dependency on MEI was also found in confined cows, which were fed six different diets and in growing lambs on three diets. In conclusion, the studies conducted in USA and in Israel investigated in depth the physiological mechanisms of cardiovascular and O2 mobilization, and went on to investigate a wide variety of ruminant species, ages, reproductive states, diets ME, time of intake and time of day, and compared these variables under grazing and confinement conditions. From these combined studies we can conclude that EE can be determined from HR measurements during several days, multiplied by O2P measured over a short period of time (10-15 min). The study showed that RE could be determined during the growing phase without slaughtering. In the near future the development microelectronic devices will enable wide use of the HR method to determine EE and energy balance. It will open new scopes of physiological and agricultural research with minimizes strain on animals. The method also has a high potential as a tool for herd management.
