Abstract:
Baroreceptors play a pivotal role in the regulation of blood pressure through moment to moment sensing of arterial blood pressure and providing information to the central nervous system to make autonomic adjustments to maintain appropriate tissue perfusion. A recent publication by Zeng and colleagues (Zeng WZ, Marshall KL, Min S, Daou I, Chapleau MW, Abboud FM, Liberles SD, Science 362: 464–467, 2018) suggests the mechanosensitive ion channels Piezo1 and Piezo2 represent the cellular mechanism by which baroreceptor nerve endings sense changes in arterial blood pressure. However, before Piezo1 and Piezo2 are accepted as the sensor of baroreceptors, the question must be asked of what criteria are necessary to establish this and how well the report of Zeng and colleagues (Zeng WZ, Marshall KL, Min S, Daou I, Chapleau MW, Abboud FM, Liberles SD, Science 362: 464–467, 2018) satisfies these criteria. We briefly review baroreceptor function, outline criteria that a putative neuronal sensor of blood pressure must satisfy, and discuss whether the recent findings of Zeng and colleagues suitably meet these criteria. Despite the provocative hypothesis, there are significant concerns regarding the evidence supporting a role of Piezo1/Piezo2 in arterial baroreceptor function.

AbstractStudy ObjectivesTo report functional recovery, symptomatic remission, and sustained symptomatic remission rates after treatment with aripiprazole once-monthly 400mg (AOM 400) administered every 4weeks for up to 52weeks as maintenance treatment in a mixed cohort of AOM 400 naïve (de novo) and experienced adults (rollover) with bipolar I disorder (BP-I).MethodThis open-label study (NCT01710709) enrolled de novo patients with a diagnosis of BP-I and ≥1 previous manic or mixed episode and rollover patients who completed a randomized, double-blind, placebo-controlled study assessing the efficacy and safety of AOM 400 (NCT01567527). Efficacy was assessed by mean changes from baseline in Young-Mania Rating Scale (YMRS), Montgomery-Asberg Depressive Rating Scale (MADRS), and Clinical Global Impression- Bipolar Version-Severity of Illness (CGI-BP-S) scores. Sustained functional recovery was defined as a total score of ≤11 on the Functioning Assessment Short Test (FAST) for ≥8 consecutive weeks. Remission was defined as YMRS and MADRS total scores ≤12, and sustained remission was defined as meeting criteria for remission for 8 consecutive weeks. The study included a screening phase (6weeks) for de novo patients, an oral aripiprazole conversion phase (4–6weeks), an oral stabilization phase (4–12weeks), and an AOM 400 maintenance phase (up to 52weeks). Rollover patients entered directly into the AOM 400 maintenance phase.ResultsA total of 464 subjects entered the maintenance phase and 63% (291/464) completed the trial. Of patients entering the maintenance phase, 379 (82%) were de novo and 85 (18%) were rollover. The most frequent reasons for discontinuation were withdrawal of consent (11%) and adverse events (AEs) (10%). Weight increase (1.5%, 7/464) and BP-I (0.9%, 4/464) were the most common reasons for discontinuation due to AEs. Improvements in mean YMRS, MADRS, CGI-BP-S, and FAST scores achieved in previous phases were maintained over 52weeks. Treatment-emergent AEs experienced by >10% of the patients were akathisia (14.7%), weight increased (13.4%), nasopharyngitis (12.1%), and insomnia (11.0%). A high proportion of de novo patients met the criteria for symptomatic remission (87.2%, 328/376) and sustained remission (77%, 292/379) by last visit. Rollover patients’ remission rate remained stable (98.8%, 84/85) by last visit. Of the rollover patients, 35/85 (43%) and 35/116 (36%) of de novo subjects met the criteria for sustained functional recovery after study completion.ConclusionsPatients treated with AOM 400 maintained symptomatic and functional stability for up to 52weeks. Importantly, more than one-third of patients achieved sustained functional recovery using a strict criterion. Overall, AOM 400 was safe and well tolerated in patients with BP-I. Background Abecarnil, a novel anxiolytic beta-carboline, was investigated in five four-week double-blind, European multicentre studies. Overall, 451 patients with generalised anxiety disorder were randomised to abecarnil, 461 to placebo and 464 to active controls.Method Data includes inferential statistics based on individual studies and descriptive analysis of 323 patients in open-label abecarnil longterm continuation up to 52 weeks.Results Abecarnil was safe, the most frequent adverse event being drowsiness. Onset of effect was at week I. At week 4 the Hamilton Anxiety Scale score had improved by 12–13 points on average. Due to notably large and variable placebo effects abecarnil was not consistently superior to placebo. No rebound or withdrawal symptoms were observed after fast-tapered discontinuation. Safety, extent of efficacy and incidence of rebound or withdrawal did not change during longterm treatment.Conclusions Abecarnil is safe and effective. Further research into its therapeutic potential seems warranted.

This research proposes and implements a web-based New Student Admission Information System (PPDB) using the Simple Additive Weight (SAW) method as a decision support system for school selection. The case study was conducted at the Lidah Kulon I/464 Surabaya Elementary School. The background of this research is driven by the significant impact of information technology advancements in the education sector. Lidah Kulon I/464 Surabaya Elementary School, as an educational institution committed to achieving excellence, discipline, faith, science and technology (IPTEK), as well as environmental awareness, strives to reduce paper usage in the new student registration process by transitioning to a more efficient administrative information system. The results of the tests state that the system has succeeded in various aspects, including the use of the Certainty Factor Method, diagnosis menu, information menu, login menu, and others. The conclusion of this research is that the system has been implemented successfully, supporting the efficiency of the PPDB process, and providing easy access to users. System development recommendations for the future include flexibility without hard coding, more comprehensive user management, the development of a more advanced school selection system, more accurate entrance fee calculations, and considerations for seasonal factors and school promotions. In suitable pH value of Walpole buffer solution and in the presence of surfactant Triton X-10 and alcohol, PtCl62- reacts with I- to form PtI62-. By means of the electrostatic and hydrophobic forces, proteins with positive charge, such as human serum albumin (HSA), bovine serum albumin (BSA), human immunoglobulin (IgG) and ovalbumin (OVA), associates with PtI62- to form stable [protein-(PtI6)n]m association particles which cause substantial enhancement of Rayleigh scattering and interface fluorescence. They produce three Rayleigh scattering peaks at 330 nm, 420 nm and 464 nm and a resonance scattering peak at 580 nm, among which the strongest peak is at 464 nm. The four kinds of protein association particle system all exhibit a strong fluorescence peak at 466 nm, while the HSA fluorescence peak at 360 nm was quenched with the formation of the association particle. The cause of the Rayleigh scattering peaks of the association particle system was considered in detail, and the relationship between the Rayleigh scattering and fluorescence of the association particle (the means diameter is about 470 nm) and the fluorescence quenching of HSA was interpreted. Under the optimal experimental conditions, there is a good linear relationship between the scattering intensity (I464 nm) and protein concentration in the range of 0.05~25μg/mL HSA, 0.05~25μg/mL BSA, 0.3~30μg/mL IgG and 0.1~16μg/mL OVA respectively, with a detection limit of 20 ng/mL HSA, 26 ng/mL BSA, 40 ng/mL IgG and 70 ng/mL OVA. This assay has been applied to the determination of the albumin of human serum samples with satisfactory results.

Celem prowadzonych badań była ocena skuteczności chwastobójczej wybranych herbicydów stosowanych w jęczmieniu jarym, odmiana Poldek. Doświadczenie przeprowadzono w latach 2012–2014. W doświadczeniu stosowano następujące kombinacje herbicydowe: kontrola (bez odchwaszczania), Pike 20 WG (metsulfuron metylu), Aurora 40 WG (karfentrazon etylu), Tayson 464 SL + Pike 20 WG (2,4-D + dikamba + metsulfuron metylu), Pike 20 WG + Aurora 40 WG (metsulfuron metylu + karfentrazon etylu), Galmet 20 SG + Galaper 200 EC (metsulfuron metylu + fluroksypyr), Granstar Ultra SX 50 WG (tifensulfuron metylu + tribenuron metylu). Ocenę skuteczności zwalczania najliczniej występujących gatunków chwastów przeprowadzono 30 dni po zastosowaniu preparatów metodą szacunkową. Dodatkowo dwa tygodnie przed zbiorem jęczmienia oceniono wtórne zachwaszczenie łanu metodą botaniczno-wagową, określając skład florystyczny, liczebność poszczególnych gatunków oraz powietrznie suchą masę chwastów. Gatunkami dominującymi w uprawie jęczmienia były: Galinsoga parviflora, Polygonum lapathifolium subsp. lapathifolium, Galium aparine, Matricaria maritima subsp. inodora, Chenopodium album i Stellaria media. Najbardziej skuteczne w zwalczaniu dominujących w jęczmieniu gatunków chwastów były kombinacje herbicydowe Tayson 464 SL + Pike 20 WG oraz Pike 20 WG + Aurora 40 WG, Galmet 20 SG + Galaper 200 EC. Po aplikacji tych kombinacji herbicydowych uzyskano również najwyższy plon ziarna jęczmienia.

Com vistas ao reconhecimento da obra poética, elabora-se uma proposta de leitura dos versos 464 a 466 do Canto IV das Geórgicas de Virgílio, que tratam do mito de Orfeu. Em particular, o interesse por poesia, haja vista a expressividade verbal e o artesanato do poeta, motivou o estudo da imagem poética, a partir das relações entre som e sentido. Sob essa proposta, abre-se uma discussão acerca dos possíveis efeitos estéticos advindos da leitura atenta dos três versos latinos, incluindo sua escansão e contextualização de referências míticas, com ênfase na inteligência sensorial do texto poético. O objetivo do artigo, em síntese, consiste na descrição de leitura detalhada de um excerto, a partir do qual se estabelecem comentários sobre expressividade poética e imagem verbal. Nesse ensejo, comparam-se os recursos métrico-prosódicos da poesia latina com a poesia em língua portuguesa, mais acessível ao falante nativo. Assim, acrescentam-se, na proposta de leitura, a estrofe 19 do Canto I dos Lusíadas de Camões e o poema “Rondó dos cavalinhos”, de Manuel Bandeira, perpassando o antigo e o moderno.

In an open-label, long-term safety study (NCT01710709), AOM 400 was safe and effective as long-term maintenance treatment (2).Objective:To evaluate the effect of long-term AOM 400 maintenance treatment on manic and depressive symptoms in BP-I patients from the open-label, long term safety study.Methods:Manic and depressive symptoms were assessed post-hoc by analysis of change from study entry in the Young-Mania Rating Scale (YMRS) and Montgomery-Åsberg Depression Rating Scale (MADRS) total scores and line item scores. The mean changes from baseline at last visit in YMRS and MADRS total scores, and single items were calculated using descriptive statistics, using last observation carried forward for total scores and observed cases for single items.Results:A total of 464 patients entered the maintenance phase: 379 were de novo and 85 were rollover patients who completed the double-blind, placebo-controlled withdrawal study. Overall, 63% (291/464) completed 52 weeks of open-label treatment. Mean YMRS and MADRS total scores were minimally changed from baseline (YMRS: 2.31, endpoint change -0.30; MADRS: 3.23, endpoint change +1.24) across the study in the total population.Conclusion:Patients entering an open-label safety study with stable manic and depressive symptoms maintained stability in both types of symptoms, as shown by minimal mean changes from baseline in YMRS and MADRS scores, suggesting that treatment with AOM 400 is effective in preventing re-emergence of both manic and depressive symptoms.Funding Acknowledgements:The study was supported by Otsuka Pharmaceutical Development & Commercialization, Inc.

In an open-label, long-term safety study (NCT01710709), AOM 400 was safe and effective as long-term maintenance treatment (2).Objective:To evaluate the effect of long-term AOM 400 maintenance treatment on manic and depressive symptoms in BP-I patients from the open-label, long term safety study.Methods:Manic and depressive symptoms were assessed post-hoc by analysis of change from study entry in the Young-Mania Rating Scale (YMRS) and Montgomery-Åsberg Depression Rating Scale (MADRS) total scores and line item scores. The mean changes from baseline at last visit in YMRS and MADRS total scores, and single items were calculated using descriptive statistics, using last observation carried forward for total scores and observed cases for single items.Results:A total of 464 patients entered the maintenance phase: 379 were de novo and 85 were rollover patients who completed the double-blind, placebo-controlled withdrawal study. Overall, 63% (291/464) completed 52 weeks of open-label treatment. Mean YMRS and MADRS total scores were minimally changed from baseline (YMRS: 2.31, endpoint change -0.30; MADRS: 3.23, endpoint change +1.24) across the study in the total population.Conclusion:Patients entering an open-label safety study with stable manic and depressive symptoms maintained stability in both types of symptoms, as shown by minimal mean changes from baseline in YMRS and MADRS scores, suggesting that treatment with AOM 400 is effective in preventing re-emergence of both manic and depressive symptoms.Funding Acknowledgements:The study was supported by Otsuka Pharmaceutical Development & Commercialization, Inc.

In the present case study, I will discuss the so-called ‘dossier of Thermouthas’. It currently consists of six private letters, but I will argue in favor of the addition of three more letters to this collection: viz. P.Mich. viii 464 (tm 17238), P.Col. viii 215 (tm 17627) and P.Corn. 49 (tm 25711). My hypothesis is based on both internal arguments (linguistic and content related elements) and external ones (e.g. museum archaeology, onomastics and prosopography). This paper illustrates how idiolectic features and the close linguistic investigation of the letters’ phraseology can help the scholar reunite papyri in their original context. It refines the date of P.Corn. 49 and shows that some of the documents in this collection were preserved as an (archaeological) archive.

Investigating moderating effects of culture between body dissatisfaction (BD) and quality of life (QoL) is paramount, as BD affects psychosocial functioning. Participants include 866 females (18–25) years old from Australia ( n = 464) M (20.88) SD (3.38) and Malaysia ( n = 402) M (20.63) SD (2.05). Higher levels of BD predicted lower levels of QoL across all four domains. BD had the strongest effect on psychological QoL for both cultures. Culture moderated the relationship between BD and: (i) physical QoL and (ii) environmental QoL. The adverse impact of BD on all domains of QoL, highlights the importance of BD as a public health problem.

However, anatomical recurrence patterns may differ depending on the therapeutic approaches. The aim of this post-hoc retrospective analysis was to investigate if the relapse pattern as assessed by 68Ga-PSMA-11 PET/CT was different depending on the type of local pelvic therapy (RP, dRT, salvage RT (SRT), pelvic lymph node dissection (PLND), pelvic lymph node RT (PLNRT)) in patients with non-metastatic castrate sensitive (nmCS) recurrent disease after primary definitive therapy. Methods: Patients who underwent a 68Ga-PSMA-11 PET/CT for nmCS PCa recurrent disease after primary definitive therapy were screened from a database of 4 prospective studies (NCT02940262, NCT03515577, NCT04050215, NCT03582774). Patients who underwent primary staging (n = 95), without definitive therapy (n = 68), with known metastatic disease (M1) (n = 68) or with castrate resistant (CR) disease (n = 291) were excluded. We examined the relationship between recurrence patterns as assessed by 68Ga-PSMA-11 PET/CT (PROMISE criteria) and prior local treatments: i) RP, ii) dRT, iii) RP + SRT. Results: 787 patients were included in the analysis. Positive scan rates were 60%, 94% and 75% in RP, dRT and RP + SRT populations, respectively. Median pre-scan PSA levels were 0.50 (0.02-72.5) ng/ml, 4.4 (0.1-202) ng/ml, and 1.07 (0.04-33) ng/ml for patients who underwent RP (n = 464), dRT (n = 109) and post-RP SRT (n = 214). Median time to first recurrence was 27.7 after RP and 54.6 months after dRT (p = < 0.0001). Patients who underwent RP had lower local recurrence (LR) pattern (T+) rates by PSMA PET than those with dRT (99/464; 21% vs 69/109; 63%; p = < 0.0001). Nodal metastasis (N1) positivity rate was similar between RP and dRT (179/464; 39% vs 43/109; 39%; p = 0.87). Extrapelvic metastasis (M1) positivity rate was lower for RP than dRT (93/464; 20% vs 51/109; 47%; p = < 0.0001). Median time from post-RP SRT to second recurrence was 22.3 months. In patients who had a second recurrence after RP and SRT the positivity rate of LR (T+), N1 disease and M1 disease by PSMA PET/CT was 12% (24/214), 46% (99/214) and 44% (95/214). Conclusions: In this cohort of patients with nmCS PCa recurrent disease after primary definitive therapy, the patterns of failure differ based on prior local treatments.

464 Background: The rest-activity rhythm is a biomarker of the circadian timing system (CTS), whose disruption accelerates cancer progression in experimental models. Prior studies in cancer pts identified a reliable measure of CTS function in cancer pts based on non invasive wrist actigraphy monitoring. Methods: The prognostic value of I<O, the ratio between activity counts in-bed vs out-of-bed, was explored for PFS and OS in mCRC. The rest-activity rhythm was recorded for >48 h in 436 mCRC pts from 3 international cohorts. Data were stratified per cohort, and analyzed with Kruskall-Wallis ANOVA, log rank tests, and multivariate Cox analyses. Results: Median age was 59.2 y. Pts were mainly men (62.6%); had good performance status (PS = 0, 58.2%; 1, 33%); > 2 metastatic sites (51.6%); metastases in liver (79%) or lung (38.8%); were chemotherapy-naïve (54.6%). After baseline actigraphy recording, pts received a median of 8 courses of oxaliplatin- or irinotecan-based chemotherapy, including chronomodulated schedules (chrono; 81.4%). Despite a strong correlation with PS (p< 0.0001), I<O independently influenced both PFS and OS. Thus, both median PFS and OS nearly doubled in the pts with I<O above 97.5% (upper two quartiles) as compared to those with I<O below 93.2% (lowest quartile). Median PFS ranged from 4.8 to 10.6 months (log rank, p<0.001) and median OS from 10.1 to 20.6 months (p<0.001). In multivariate analyses, I<O ranked as the second best independent predictor of PFS, after metastases surgery, with a hazard ratio (HR) of 0.967 [95% C.L., 0.95-0.98] (p<0.001). I<O ranked as first independent predictor of OS, with a HR of 0.95 [0.94-0.97] (p<0.001). Forrest plots revealed consistent HR of I<O for PFS and OS in each cohort. Conclusions: The circadian biomarker indicator I<O is a robust, consistent and independent quantitative predictor of both PFS and OS in mCRC pts. Rest-activity rhythm monitoring deserves further testing for helping accurate determination of mCRC prognosis. The biological significance of I<O implies that specific treatments of circadian disruption could enhance survival in cancer pts.

The finding shows that accrual accounting implementation is affected by accounting training and organization culture, but it is not affected by the usage of SAIBA. Tujuan : Mengetahui hubungan manajemen disaster dengan kesiapsiagaan mahasiswa/i Prodi DIII Keperawatan Tegal dalam tanggap bencana. Metode : Jenis penelitian ini adalah deskriptif korelasi dengan pendekatan cross sectional. Pengambilan sampel menggunakan teknik total sampling dengan responden sebanyak 464 orang. Hasil : Penelitian menunjukkan bahwa pemahaman manajemen disaster mahasiswa/i, Prodi DIII Keperawatan Tegal tergolong baik (94,8%), tetapi kesiapsiagaan yang dimiliki mayoritas sedang (57,3%). Hasil uji statistik Spearman Rank (Rho) diperoleh p (value) = 0,312 (p0,05), sehingga tidak ada hubungan antara manajemen disaster dengan kesiapsiagaan mahasiswa/i dalam tanggap bencana Prodi DIII Keperawatan Tegal Poltekkes Kemenkes Semarang. Kesimpulan dan saran : Intitusi Kesehatan sebaiknya memberikan kesempatan kepada mahasiswa untuk terjun langsung di tengah-tengah masyarakat dalam tim gawat darurat bencana serta peneliti berharap agar komitmen pengembangan ilmu keperawatan dibidang manajemen bencana dapat ditingkatkan melalui beberapa alternatif kegiatan seperti edukasi, simulasi, membentuk organisasi khusus penanggulangan bencana dan bekerjasama antar institusi atau organisasi kebencanaan tingkat daerah atau nasional.

Abstract: Most of the Connemara granites, including the Galway Granite batholith, have a clearly defined circular or ellipsoidal outcrop shape and are I-Type granites often with K-feldspar phenocrysts, whereas the Oughterard Granite (OG) forms numerous small intrusions scattered east-west over 40km with two larger linked bodies near Oughterard and is S-Type and aphyric. The only comprehensive mapping, mineralogical and chemical study of the OG was published over 50 years ago before the important 1974 recognition of the distinction of I- and S-Type granites was first made. New mapping of the southern Oughterard area with small OG intrusions is presented and many scattered published chemical and Rb, Sr, S and Pb isotopic studies integrated to confirm overall their individual conclusions that the peraluminous OG is of S-Type, being formed from melted Dalradian rocks as are the fluid-carried associated uneconomic mineral deposits. The OG is confined to the high sillimanite zone, mostly south of, and mainly later than, the 465-464Ma D4 Connemara Antiform, where a wide (>15km N-S) >70km E-W band of 469Ma gabbros and 467Ma quartz diorite gneisses intruded into already hot Dalradian rocks undergoing regional metamorphism. This generated the OG magmas by partial melting of pelites and semi-pelites. The upward intrusion of the OG magma was significantly delayed, as has been shown by precise dating in other granites elsewhere, to ∼464-461Ma during the late stages and mostly after D4 but before the country rocks had completely cooled. The OG intrusions as a whole probably form the second largest area of S-Type granite in Ireland, after the Leinster Granite.

The effects of the N-methyl-D-aspartate (NMDA) receptor antagonist MK-801 and the dihydropyridine calcium antagonist nimodipine on NMDA-induced phenomena were investigated using an in vivo fluorometric technique with indo-1. Indo-1, a fluorescent cytosolic free calcium ([Ca2+]i) indicator, was loaded into the cat cortex approximately 500 μm in depth by super-fusion with the membrane-permeant indo-1 acetoxymethyl ester (indo-1-AM). Changes in [Ca2+]i signals (400 and 506 nm) and reduced nicotinamide adenine dinucleotide (NADH) fluorescence (464 nm) were simultaneously measured directly from the cortex during ultraviolet excitation (340 nm). Superfusion of 100 μM NMDA over the exposed cortex induced an elevation of the [Ca2+]i signal ratio (400/506 nm), biphasic changes in NAD/NADH redox state (initial oxidation followed by progressive reduction), and characteristic changes in the EEG (abrupt depression in amplitude followed by an excitatory pattern of 18–22 Hz poly spikes or sharp waves). These changes were completely blocked by treatment with MK-801 and reduced by nimodipine. The mechanism underlying the protective effects of systemically administered MK-801 on the NMDA-induced neuronal injury was verified in vivo.

The compound crystallized in methyl alcohol system. The crystal belongs to the Monoclinic system, space groupP21/C, witha= 10.228(2) Å,b= 8.9260(18) Å,c= 12.064(2) Å,α= 90°,β= 91.85(3)°,γ= 90°,M= 219. 23,V= 1100.8(4) Å3,Z= 4,Dc = 1.323 g/cm3,μ=0.096 mm-1,F(000) = 464,R= 0.0594 andwR= 0.1611. A total of 2136 unique reflections were collected, of which 2020 withI>2(I) were observed. The quantum chemistry calculation study on the title compound has been performed by quantum chemical method in SVWN5/6-31G**level. Additionally, IR,1HNMR spectra, thermal analysis and the fluorescent properties are discussed .
