Hygroscopic insulator field effect transistors (HIFETs) are organic transistors with promising characteristics for biosensing applications. However, their fundamental sensing mechanisms are not yet fully understood. This thesis explores HIFET sensors through detailed electrical and optical characterisation, providing vital insights into the distinct mechanisms by which HIFETs detect biologically relevant chemicals. Hydrogen peroxide, a by-product of enzymatic reactions, oxidises the organic semiconductor, modulating the output current. Ionic solutions, such as KCl, NaCl and HCl, modulate the c
