Abstract:
Recently, wavelength division mulplexing (WDM) technology has been extensively studied, and various WDM-PON architectures have been proposed for next-generation passive optical network (PON). In this paper, we combine the TDM with WDM architecture to achieve high-speed, long-distance transmission. To reduce the cost of ONU, we achive the colorless ONU by placing a Reflective Semiconductor Optical Amplifier (RSOA) in the uplink transmission. We build the whole architecture to simulate the Hybrid WDM/TDM PON System in Optisystem and the experimental results prove that the architecture is feasible and reasonable.

In this paper, the performance and feasibility of a hybrid wavelength division multiplexing/time division multiplexing passive optical network (WDM/TDM PON) system with 128 optical network units (ONUs) is analysed. In this system, triple play services (video, voice and data) are successfully communicated through a distance of up to 28 km. Moreover, we analysed and compared the performance of various modulation formats for different distances in the proposed hybrid WDM/TDM PON. NRZ rectangular emerged as the most appropriate modulation format for triple play transmission in the proposed hybrid PON.

We develop and study the physics of a novel hybrid wavelength-division-multiplexed, optical timedivision-multiplexed (WDM-OTDM) mode-locked semiconductor laser for applications in ultra-high data rate communication links, computer interconnects, and optical sampling applications. The key philosophy behind using a hybrid approach is that state-of-the-art system performance can be achieved without the necessity of operating at the limits of a either a pure WDM or OTDM technology platform.

Abstract:
Recently, metropolitan and access communication networks have markedly developed by utilizing a variety of technologies. Their bearer communication infrastructures will be mostly exploiting the optical transmission medium where wavelength division multiplexing techniques will play an important role. This contribution discusses the symmetric sharing of common optical network resources in wavelength and time domains. Wavelength-Division Multiplexed Passive Optical Networks (WDM-PON) attract considerable attention regarding the next generation of optical metropolitan and access networks. The main purpose of this contribution is presented by the analysis of possible scheduling of wavelengths for our novel hybrid network topologies considered for WDM-PON networks. This contribution briefly deploys adequate Dynamic Wavelength Allocation (DWA) algorithms for selected WDM-PON network designs with the provision of traffic protection when only passive optical components in remote nodes are utilized. The main part of this study is focused on the use of wavelength scheduling methods for selected WDM-PON network designs. For evaluation of offline and online wavelength scheduling for novel hybrid network topologies, a simulation model realized in the Matlab programming environment allows to analyze interactions between various metropolitan and access parts in the Optical Distribution Network (ODN) related to advanced WDM-PON network designs. Finally, wavelength scheduling methods are compared from a viewpoint of utilization in advanced WDM-PON networks designs.

Abstract:
This paper presents on the design of L-Band Multiwavelength laser for Hybrid Time Division Multiplexing/ Wavelength Division Multiplexing (TDM/WDM) Passive Optical Network (PON) application. In this design, an L-band Mulltiwavelength Laser is designed as the downstream signals for TDM/WDM PON. The downstream signals ranging from 1569.865 nm to 1581.973 nm with 100GHz spacing. The multiwavelength laser is designed using OptiSystem software and it is integrated into a TDM/WDM PON that is also designed using OptiSystem simulation software. By adapting multiwavelength fiber laser into a TDM/WDM network, a simple and low-cost downstream signal is proposed. From the simulation design, it is found that the proposed design is suitable to be used in TDM/WDM PON for up to 64 Optical Network Units (ONUs).

Abstract:
A high-capacity, flexible, cost, and energy efficient backhaul network based on the convergence of optical and wireless technologies is proposed this research work. The proposed backhaul network, which is deployed on wavelength-division multiplexing passive optical network (WDM-PON), supports three backhauling solutions including pure WDM-PON, hybrid WDM-PON/free-space optics (FSO), and hybrid WDM-PON/millimeter-wave (MMW) radio frequency (RF). We firstly have developed mathematical models for performance analysis of the downlink for three comparative backhaul solutions. We then theoretically and comprehensively analyzed the system performance while considering various system parameters such as the total distance, wireless link distance, splitting ratio, amplifier gain, and the bit rate as well as key environment parameters including rain attenuation and atmospheric turbulence. We have also compared the performance of the three alternative backhaul solutions to verify the trade-off between the system performance and the system flexibility/mobility. The numerical results verify the system performance of each backhaul solution. It is demonstrated that combination of these approaches can exploit PON, FSO and RF technologies to provide a flexible and gigabit-bandwidth-capable solution for the fifth generation (5G) backhaul networks.

Hybrid optical OFDM/WDM system is proposed to provide longer reach and larger capacity comparing with existing PON systems in order to meet the ever increasing bandwidth demand and data rate of next generation optical access networks. In this paper, two channels using different modulation at 10 Gb/s are used for downstream link. OFDM signals occupy different portions of the available signal spectrum for signal transmissions by using WDM. As a result, the system has good performance based on SNR with 50 km distance and QAM modulation delivers better performance compared with PSK.

An advanced electro-optic hybrid rotary joint (EOHRJ) has been developed in Phase II of an AF SBIR effort with Physical Optics Corporation (POC) to replace cable wrap structure for multi-channel rotation-to-fixed (RTF) signal transmission. The EOHRJ meets AFFTC and other range special needs with a generic, high performance, rotary joint solution. At the moment, we have successfully installed and tested the EOHRJ on our KTM tracker system with the following capabilities: 1) able to accommodate hundreds of transmission channels, including electrical power, control, feedback, and low-speed signals; 2) able to accommodate multiple channel, high data rate (over gigabits per second), and bi-directional signal transmission; 3) able to be reliable for harsh environmental operation, adaptive to stringent sized requirement, and accommodating existing electrical and mechanical interfaces. The completed EOHRJ contains three uniquely integrated functional rings. The first and the outmost one is power ring, which provides RTF transmission channels for over 50 high voltage and high current channels. The second and the middle one is low speed electrical signal ring, which provides RTF transmission for over hundred control, feedback, and low speed data signals. The third and the inmost one is optical fiber slip ring, which, incorporating with current advanced signal multiplexing technologies (either time division or wavelength division multiplexing ) is able to provide multiple channel, high data rate, and bi-directional signal transmission. At the moment, the prototype module of the tree-layer EOHRJ has been successfully assembled in Air Force’s tracker system, and is providing a satisfactory performance. This paper presents our joint work on this project.

Abstract:
The ever-increasing demand for telecommunications needs has highlighted the intrinsic limitations of electronics. Photonics has proven to be a suitable solution to its limitations. STMicroelectronics has developed a silicon photonic platform called PIC25G that allows single-channel transmission at 25 Gb/s. The data rate increase with wavelength division multiplexing (WDM) encounter some constraints. The proposed solution is based on a hybrid approach integrating silicon photonics and glass ions exchanged photonics developed at the IMEP-LaHC laboratory. The solution consists of a glass interposer on which a silicon photonics chip is assembled. First, the studies focused on the components of the silicon chip especially on the optimization of grating couplers and (de)-multiplexers based on cascaded Mach Zehnder interferometers. Then, the optical and electrical passive components of the glass interposer were studied and realized. The feasibility of an optical coupling between the silicon chip and the glass interposer has been demonstrated. Finally, the test structures needed to validate the proposed solution were studied. These test structures allowed to transmit radiofrequency signals up to 40 GHz between the silicon chip and the glass interposer. A new approach to realize the optical waveguides of the interposer has been suggested and carried out in order to address the fabrication issues. Ultimately, this approach will provide an electro-optical module for high-speed applications

Abstract:
Essentially all biological processes depend on protein-protein interactions (PPIs). Timing of such interactions is crucial for regulatory function. Although circadian (~24 hrs) clocks constitute fundamental cellular timing mechanisms regulating important physiological processes PPI dynamics on this timescale are largely unknown. To elucidate so far unknown regulatory mechanisms within the circadian clockwork, I have systematically mapped PPIs among 46 circadian components using high-throughput yeast-two-hybrid (Y2H) interaction experiments. I have identified 109 so far uncharacterized interactions and successfully validated a sub-fraction via co-immunoprecipitation experiments in human cells. Among the novel PPIs, I have identified modulators of CLOCK/BMAL1 function and further characterized the role of protein phosphatase 1 (PP1) in the dynamic regulation of BMAL1 abundance. Furthermore, to generate a more comprehensive circadian PPI network, the experimental network was enriched and extended with additional interactions and interaction partners from literature, some of which turned out to be essential for normal circadian dynamics. The integration of circadian mRNA expression profiles allowed us to determine the interaction dynamics within our network. Systematic genetic perturbation studies (RNAi and overexpression in oscillating human cells) revealed a crucial role of dynamic regulation (via rhythmic PPIs) for the molecular clockwork. Furthermore, dynamic modular organization as a pervasive circadian network feature likely contributes to time-of-day dependent control of many cellular processes. Global analysis of the proteome regarding circadian regulation of biological processes via rhythmic PPIs revealed time-of-day dependent organization of the human interactome. Circadian PPIs dynamically connect many important cellular processes like signal transduction and cell cycle, which contribute to temporal organization of cellular physiology.

Abstract:
Protein methylation on arginine and lysine residues is a largely unexplored posttranslational modification which regulates diverse cellular processes. The development of efficient proteome-wide approaches for detecting protein methylation is limited and technically challenging. We developed a novel workload reduced yeast-two hybrid (Y2H) approach to detect protein-protein interactions utilizing second generation sequencing. The novel Y2H-seq approach was systematically evaluated against our state of the art Y2H-matrix screening approach and used to screen 8 protein arginine methyltransferases, 17 protein lysine methyltransferases and 10 demethylases against a set of 14,268 proteins. Comparison of the two approaches revealed a higher sensitivity of the new Y2H-seq approach. The increased sampling rate of the Y2H-seq approach is advantageous when assaying transient interactions between substrates and methyltransferases. Overall 523 interactions between 22 bait proteins and 324 prey proteins were identified including 11 proteins known to be methylated. Network analysis revealed enrichment of transcription regulator activity, DNA- and RNA-binding function of proteins interacting with protein methyltransferases. The dataset represents the first proteome-wide interaction network of enzymes involved in methylation and provides a comprehensively annotated resource of potential new methylation substrates. An in vitro methylation assay coupled to mass spectrometry revealed amino acid methylation of candidate proteins. Seven of nine proteins tested were methylated including SPIN2B, DNAJA3, QKI, SAMD3, OFCC1, SYNCRIP and WDR42A indicating that the interaction network is likely to contain many putative methyltransferase substrate pairs. The presented protein-protein interaction network demonstrates that protein methylation is involved in diverse cellular processes and can inform hypothesis driven investigation into molecular mechanisms regulated through methylation.

Abstract:
With the development of fiber optical data transmission technologies and infrastructure on a global scale, the use of fiber optical sensors to perform sensing functions is becoming increasingly important mainly due to their advantages, such as small size, light weight, multiplexing capabilities and others. Therefore, in order to ensure more efficient use of optical sensor technologies, it is necessary to research and search for new solutions for the integration and use of fiber optical sensors in existing and future fiber optical communication system architecture. The main goal of the Doctoral Thesis is to study and evaluate the interaction of FBG fiber optical sensor networks and WDM-PON metro-access transmission systems implemented in a single system with theoretical and experimental research. In order to evaluate their performance in a unified system, a detailed study of the interaction between the FBG optical sensor network and the fiber optic data transmission system has been performed by implementing mathematical modeling and experimental development of such systems.

Abstract:
Arabidopsis mutants that we have isolated, encode for fertilization-independent endosperm (fie), fertilization-independent seed2 (fis2) and medea (mea) genes, act in the female gametophyte and allow endosperm to develop without fertilization when mutated. We cloned the FIE and MEA genes and showed that they encode WD and SET domain polycomb (Pc G) proteins, respectively. Homologous proteins of FIE and MEA in other organisms are known to regulate gene transcription by modulating chromatin structure. Based on our results, we proposed a model whereby both FIE and MEA interact to suppress transcription of regulatory genes. These genes are transcribed only at proper developmental stages, as in the central cell of the female gametophyte after fertilization, thus activating endosperm development. To test our model, the following questions were addressed: What is the Composition and Function of the Polycomb Complex? Molecular, biochemical, genetic and genomic approaches were offered to identify members of the complex, analyze their interactions, and understand their function. What is the Temporal and Spatial Pattern of Polycomb Proteins Accumulation? The use of transgenic plants expressing tagged FIE and MEA polypeptides as well as specific antibodies were proposed to localize the endogenous polycomb complex. How is Polycomb Protein Activity Controlled? To understand the molecular mechanism controlling the accumulation of FIE protein, transgenic plants as well as molecular approaches were proposed to determine whether FIE is regulated at the translational or posttranslational levels. The objectives of our research program have been accomplished and the results obtained exceeded our expectation. Our results reveal that fie and mea mutations cause parent-of-origin effects on seed development by distinct mechanisms (Publication 1). Moreover our data show that FIE has additional functions besides controlling the development of the female gametophyte. Using transgenic lines in which FIE was not expressed or the protein level was reduced during different developmental stages enabled us for the first time to explore FIE function during sporophyte development (Publication 2 and 3). Our results are consistent with the hypothesis that FIE, a single copy gene in the Arabidopsis genome, represses multiple developmental pathways (i.e., endosperm, embryogenesis, shot formation and flowering). Furthermore, we identified FIE target genes, including key transcription factors known to promote flowering (AG and LFY) as well as shoot and leaf formation (KNAT1) (Publication 2 and 3), thus demonstrating that in plants, as in mammals and insects, PcG proteins control expression of homeobox genes. Using the Yeast two hybrid system and pull-down assays we demonstrated that FIE protein interact with MEA via the N-terminal region (Publication 1). Moreover, CURLY LEAF protein, an additional member of the SET domain family interacts with FIE as well. The overlapping expression patterns of FIE, with ether MEA or CLF and their common mutant phenotypes, demonstrate the versatility of FIE function. FIE association with different SET domain polycomb proteins, results in differential regulation of gene expression throughout the plant life cycle (Publication 3). In vitro interaction assays we have recently performed demonstrated that FIE interacts with the cell cycle regulatory component Retinobalsoma protein (pRb) (Publication 4). These results illuminate the potential mechanism by which FIE may restrain embryo sac central cell division, at least partly, through interaction with, and suppression of pRb-regulated genes. The results of this program generated new information about the initiation of reproductive development and expanded our understanding of how PcG proteins regulate developmental programs along the plant life cycle. The tools and information obtained in this program will lead to novel strategies which will allow to mange crop plants and to increase crop production.
