ABSTRACT Recently a cell culture model supporting the complete life cycle of the hepatitis C virus (HCV) was developed. Searching for host cell determinants involved in the HCV replication cycle, we evaluated the efficiency of virus propagation in different Huh-7-derived cell clones. We found that Huh-7.5 cells and Huh7-Lunet cells, two former replicon cell clones that had been generated by removal of an HCV replicon by inhibitor treatment, supported comparable levels of RNA replication and particle production, whereas virus spread was severely impaired in the latter cells. Analysis of cell surface expression of CD81 and scavenger receptor class B type I (SR-BI), two molecules previously implicated in HCV entry, revealed similar expression levels for SR-BI, while CD81 surface expression was much higher on Huh-7.5 cells than on Huh7-Lunet cells. Ectopic expression of CD81 in Huh7-Lunet cells conferred permissiveness for HCV infection to a level comparable to that for Huh-7.5 cells. Modulation of CD81 cell surface density in Huh-7.5 cells by RNA interference indicated that a certain amount of this molecule (∼7 × 104 molecules per cell) is required for productive infection with a low dose of HCV. Consistent with this, we show that susceptibility to HCV infection depends on a critical quantity of CD81 molecules. While infection is restricted in cells expressing very small amounts of CD81, susceptibility rapidly rises within a narrow range of CD81 levels, reaching a plateau where higher expression does not further increase the efficiency of infection. Together these data indicate that a high density of cell surface-exposed CD81 is a key determinant for productive HCV entry into host cells.

Abstract:
Objective. Hepatocellular carcinoma is a malignant disease occurring in the liver and is one of the main causes of death in cancer patients. Tumor cells are the main components of tumors and have a strong proliferative capacity. They are easily transferred to other parts of the body and can produce harmful substances that destroy the normal organ structure and endanger human life and health. In this study, we investigate the effect of circFOXO3 on the proliferation and invasion of hepatocellular carcinoma cells and its possible mechanism. Methods. Human hepatocellular carcinoma cells BEL-7404, Hep G2, Hep 3B2.1–7, HuH-7, Li-7, and human normal hepatocytes HHL-5 were selected, and the expression level of circFOXO3 in the cell lines was determined by qRT-PCR. The cell line with low circFOXO3 expression level (HuH-7 cells) was used for follow-up experiments. HuH-7 liver cancer cells were divided into the control group (normal cultured), circFOXO3-NC group (transfected with circFOXO3 negative control), circFOXO3 mimic group (transfected with circFOXO3 mimic), PI3K activator group (20 μmol/L PI3K activator 740Y-P), and circFOXO3 mimic + PI3K activator group (transfected with circFOXO3 mimic + treated with PI3K activator 740Y-P). The qRT-PCR method was used to determine the expression level of circFOXO3 in HuH-7 liver cancer cells in each group, WB was used to detect the expression of apoptosis, invasion, and phosphoinositide 3-kinase/protein kinase B (PI3K/Akt) pathway related proteins in HuH-7 liver cancer cells in each group, the CCK-8 method was used to determine the viability of HuH-7 liver cancer cells in each group, flow cytometry was used to determine the apoptotic ability of HuH-7 liver cancer cells in each group, the transwell chamber experiment was used to determine the invasion ability of HuH-7 liver cancer cells in each group, and the scratch test was used to determine the migration ability of HuH-7 liver cancer cells in each group. Results. circFOXO3 showed low expression in liver cancer cells; compared with the control group, the circFOXO3 expression and apoptosis rate of HuH-7 liver cancer cells in the circFOXO3 mimic group were significantly increased ( P < 0.05 ) and the PI3K/Akt pathway-related protein expression, cell viability, invasion, and migration abilities were significantly reduced ( P < 0.05 ); the apoptosis rate of HuH-7 liver cancer cells in the PI3K activator group was significantly reduced ( P < 0.05 ) and the PI3K/Akt pathway related protein expression, cell viability, invasion and migration abilities were significantly increased ( P < 0.05 ). Compared with the circFOXO3 mimic group, the apoptosis rate of HuH-7 liver cancer cells in the circFOXO3 mimic + PI3K activator group was significantly reduced ( P < 0.05 ) and the PI3K/Akt pathway-related protein expression, cell viability, invasion and migration abilities were significantly increased ( P < 0.05 ). Conclusion. Highly expressed circFOXO3 can inhibit the proliferation and invasion of HuH-7 liver cancer cells, which may be achieved by inhibiting the PI3K/Akt pathway.

Abstract:
Inorganic nanoparticles are representing an emerging paradigm in molecular imaging probe design. We have determined lanthanum oxide nanoparticles (La2O3 NPs)-induced toxicity on human livers cells for 48 hrs. Before exposure to La2O3 NPs, the size and shape of NPs were confirmed by transmission electron microscope. It was found at 32 ±1.6 nm with a sheet-like morphological structure. The viability of CHANG and HuH-7 cells was reduced as the concentration of La2O3 NPs increased. HuH-7 cells were more sensitive than CHANG cells to La2O3 NPs. We observed production of intracellular ROS in HuH-7 cells was more than CHANG cells and the LPO level was more in CHANG cells than in HuH-7 cells at 50 μg/ml of La2O3 NPs. Glutathione was decreased and catalase was increased at 50 μg/ml of La2O3 NPs. More apoptotic and necrotic cells were observed at 300 μg/ml in HuH-7 cells FACS. DNA damage was observed by the SGCE test and more DNA damage was found in CHANG cells than HuH-7 cells at 300 μg/ml La2O3 NPs over 48 hrs. Thus, study warrants the application of La2O3 NPs in daily life and provides vital information about the toxicity of La2O3 NPs.

Abstract:
The family Hepeviridae comprises the species Orthohepevirus A–D (HEV-A to -D). HEV-C genotype 1 (HEV-C1, rat HEV) is able to infect humans. This study investigated whether an optimized HEV-A cell culture system is able to propagate the cell culture-derived rat HEV, and if de novo isolation of the virus from rat liver is possible. We tested the liver carcinoma cell lines PLC/PRF/5, HuH-7, and HuH-7-Lunet BLR for their susceptibility to HEV-C1 strains. Cells were infected with the cell culture-derived HEV-C1 strain R63 and rat liver-derived strain R68. Cells were maintained in MEMM medium, which was refreshed every 3–4 days. The viral load of HEV-C1 was determined by RT-qPCR in the supernatant and expressed as genome copies per mL (c/mL). Rat HEV replication was most efficient in the newly introduced HuH-7-Lunet BLR cell line. Even if the rat HEV isolate had been pre-adapted to PLC/PRF/5 by multiple passages, replication in HuH-7-Lunet BLR was still at least equally effective. Only HuH-7-Lunet BLR cells were susceptible to the isolation of HEV-C1 from the liver homogenate. These results suggest HuH-7-Lunet BLR as the most permissive cell line for rat HEV. Our HEV-C1 cell culture system may be useful for basic research, the animal-free generation of large amounts of the virus as well as for the testing of antiviral compounds and drugs.

Abstract:
Horsetail fern plant is botanically known as Equisetum arvense L., and it is a good source of phenolic flavonoids, phenolic acids, and compounds. Anticancer properties of hexane and chloroform extracts of the horsetail fern plant and their mechanisms involved in the anticancer activity on human hepatocarcinoma (HuH-7) cells were examined. Cytotoxicity was evaluated by using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and NRU (neutral red uptake) assays. Other parameters such as oxidative stress and apoptosis in pretreated hexane and chloroform extracts of the horsetail fern plant were examined in HuH-7 cells. The observation showed that hexane and chloroform extract of the horsetail fern plant exhibited cytotoxicity against HuH-7 cells. The value of IC50-24 h of hexane and chloroform extract of the horsetail fern plant was determined as 199.0 μg/ml and 161.90 0 μg/ml for HuH-7 cells, respectively, and on the basis of IC50 value, three acute concentrations, viz., 75% of IC50, 50% of IC50, and 25% of IC50, were determined for further study. The lower dose of extracts hexane and chloroform extract of the horsetail fern plant did not show significant toxicity. Higher concentrations of extract induced significant antioxidant effects as well as apoptosis effects. However, exposure to hexane and chloroform extract of the horsetail fern plant upregulated the expression of Bax and p53 in HuH-7 cells. These data suggest that hexane and chloroform extract of the horsetail fern plant plays a significant role in the induction of toxicity via the regulation of oxidative stress in HuH-7 cells. This work may be useful for cancer chemotherapy.

Nanotechnology has achieved more commercial attention over recent years, and its application has increased concerns about its discharge in the environment. In this study, we have chosen human hepatic carcinoma (HuH-7) cells because liver tissue has played an important role in human metabolism. Therefore, the objective of this study was to determine DNA damaging and apoptotic potential of cadmium telluride quantum dots (CdTe QDs; average particle size (APS) 10 nm, 1–25 µg/ml) on HuH-7 cells and the basic molecular mechanism of its cellular toxicity. Cytotoxicity of different concentrations of CdTe QDs on HuH-7 cells was determined by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) and lactate dehydrogenase (LDH) tests. Moreover, reactive oxygen species (ROS) generation, mitochondrial membrane potential, DNA damage, and Hoechst 33342 fluorescent staining morphological analysis of necrotic/apoptotic cells were detected; cellular impairment in mitochondria and DNA was confirmed by JC-1 and comet assay, respectively. A dose- and time-dependent cytotoxicity effect of CdTe QDs exposure was observed HuH-7 cells; the significant ( p < 0.05) cytotoxicity was found at 25 μg/ml of CdTe QDs exposure. The percentage of cytotoxicity of CdTe QDs (25 μg/ml) in HuH-7 cells reached 62% in 48 h. CdTe QDs elicited intracellular ROS generation and mitochondrial depolarization, and DNA integrity cells collectively advocated the apoptotic cell death at higher concentration. DNA damage was observed in cells due to CdTe QDs exposure, which was mediated by oxidative stress. This study exploring the effects of CdTe QDs in HuH-7 cells has provided valuable insights into the mechanism of toxicity induced by CdTe QDs.

Five new compounds called Pestalotis A–E (1–5), comprising three monoterpene-lactone compounds (1–3), one tetrahydrobenzofuran derivative (4), and one sesquiterpene (5), were isolated from the EtOAc extract of Pestalotiopsis sp. The structures of the new compounds were elucidated by analysis of their NMR, HRMS, and ECD spectra, and the absolute configurations were established through the comparison of experimental and calculated ECD spectra. All compounds were tested for antitumor activity against SW-480, LoVo, HuH-7, and MCF-7. The results showed that compounds 2 and 4 exhibited potent antitumor activity against SW-480, LoVo, and HuH-7 cell lines. Furthermore, compound 4 was assessed against HuH-7, and the results indicated that the rate of apoptosis was dose-dependent.

Abstract:
Dengue virus (DENV) is a flavivirus associated with clinical manifestations ranging in severity from self-limiting dengue fever, to the potentially life threatening condition, severe dengue. There are currently no approved antiviral therapies for the treatment of DENV. Here, we evaluated the antiviral potential of four broad-spectrum antivirals, UV-4B, interferon-alpha (IFN), sofosbuvir (SOF), and favipiravir (FAV) against DENV serotype 2 as mono- and combination therapy in cell lines that are physiologically relevant to human infection. Cell lines derived from human liver (HUH-7), neurons (SK-N-MC), and skin (HFF-1) were infected with DENV and treated with UV-4B, IFN, SOF, or FAV. Viral supernatant was sampled daily and infectious viral burden was quantified by plaque assay on Vero cells. Drug effect on cell proliferation in uninfected and infected cells was also assessed. UV-4B inhibited DENV in HUH-7, SK-N-MC, and HFF-1 cells yielding EC50 values of 23.75, 49.44, and 37.38 µM, respectively. Clinically achievable IFN concentrations substantially reduced viral burden in HUH-7 (EC50 = 102.7 IU/mL), SK-N-MC (EC50 = 86.59 IU/mL), and HFF-1 (EC50 = 163.1 IU/mL) cells. SOF potently inhibited DENV in HUH-7 cells but failed to produce the same effect in SK-N-MC and HFF-1 cells. Finally, FAV provided minimal suppression in HUH-7 and SK-N-MC cells, but was ineffective in HFF-1 cells. The two most potent anti-DENV agents, UV-4B and IFN, were also assessed in combination. UV-4B + IFN treatment enhanced antiviral activity in HUH-7, SK-N-MC, and HFF-1 cells relative to monotherapy. Our results demonstrate the antiviral potential of UV-4B and IFN against DENV in multiple physiologically relevant cell types.

Abstract:
Hepcidin is the main regulator of systemic iron homeostasis, acting by inhibiting macrophages iron release and intestinal iron absorption. During hypoxia hepcidin expression is inhibited to allow iron requirement to bone marrow to sustain erythropoietic expansion, but mechanisms of hypoxia-induced hepcidin down-regulation are partially unknown. Our aim was to improve our knowledge on hypoxia modulation of hepatic hepcidin, investigating the role of erythroid and iron-related pathways using in vitro and in vivo studies. We firstly studied the effect of hypoxia on healthy volunteers living at high altitude for 24h and 72h, confirming that hypoxia is a strong inhibitor of hepcidin production and that this inhibition followed EPO induction. Indeed s-hepcidin was strictly down-regulated at 72h of hypoxia while EPO levels were already markedly increased at 24h slightly decreasing thereafter. We then analyzed the effect of sera of these subjects on hepcidin expression in HuH-7 cells using both luciferase assay and RT-PCR. HAMP wild-type promoter activity decreased after exposure to hypoxic sera, suggesting that circulating factors present in sera may modulate hepcidin expression. However, analysis of the transcripts gave less homogenous results. In fact only a subgroup of hypoxic sera significantly inhibited hepcidin. These sera also induced a significant ID1 down-regulation indicating that inhibitory factor(s) seems to act through the SMADs pathway. Our results also suggest that a cellular model cannot reproduce what really happens in vivo that might depend to complex events involving both circulating and tissue factors. Therefore, we took advantages from a mouse model. For hypoxia challenge, C57BL/6 mice were housed in a hypoxic chamber at 10% O2 for 6 and 15h. mRNA levels of erythroid genes were assessed in liver, spleen and bone marrow by RT-PCR. Hamp1 levels were slightly increased after 6h of hypoxia and strongly down-regulated after 15h. After 6h we already observed a marked activation of transcription of all the erythroid genes analyzed (Pdgf-b, Fam132b, Gdf-15 and Twsg-1) in bone marrow (but not in spleen) that return to baseline levels after 15h. To better analyze the role of PDGF-BB, normoxic and hypoxic mice were treated with PDGF-BB neutralizing antibody or PDGF-BB receptor inhibitor immediately prior to hypoxic challenge. Inactivation of PDGF-BB or its receptor suppressed the hypoxia-induced hepcidin inhibition confirming the importance of PDGF-BB in hepcidin regulation. However, no changes in CREB/H protein were observed suggesting that this molecule is not involved in hepcidin regulation in this setting. In conclusion, our studies demonstrated that hypoxia-induced hepcidin regulation is a quick event secondary to erythropoietic activation. Indeed hepcidin decreased after up-regulation of transcription of several erythroid genes that return to baseline when hepcidin reached the maximum inhibition. Our studies indicate that PDGF-BB is a good candidate as hepcidin erythroid regulators but also suggest that hypoxia induced-hepcidin suppression is the result of cooperation of multiple signals that need to be dissected.

Introduction : L'hémophilie B (HB) est une maladie hémorragique héréditaire due à un défaut en facteur IX (FIX) de la coagulation. Le traitement substitutif est efficace mais pose deux problèmes : la fréquence des injections intraveineuses de concentrés de FIX et leurs coûts. La production d'un FIX recombinant à activité améliorée et à demi-vie prolongée est un enjeu important du traitement. Matériel et méthodes : Afin d'améliorer l'activité du FIX, l'étude s'est portée sur un résidu impliqué dans l'interaction du FIX activé avec son cofacteur, le facteur VIII activé (FVIIIa). Quatre FIX mutés au niveau de l'acide aminé E410 ont été développés. Afin de prolonger la demi-vie, un ADN complémentaire de FIX chimérique a été généré de façon à produire la protéine correspondante par la lignée cellulaire humaine Huh-7. Résultats : L'activité coagulante in vitro des FIX-E410 était 3 à 5 fois plus élevée que celle du FIX wild-type (FIX-WT). De plus, le FIX-E410H induisait une génération de thrombine 5,2 fois plus élevée comparée à celle du FIX-WT. Chez la souris HB, l'activité coagulante et la capacité de génération de thrombine du FIX-E410H in vivo étaient significativement plus élevées que celles du FIX-WT. La protéine chimérique a présenté une activité molaire spécifique 10 fois augmentée in vitro et une demi-vie jusqu'à 2,8 fois plus allongée, par rapport à celles du FIX-WT. Conclusion : Nous avons développé et caractérisé quatre molécules de FIX ayant une activité améliorée in vitro et in vivo, ainsi qu'un FIX modifié à activité augmentée et à demi-vie prolongée in vivo. Ces nouvelles molécules pourraient ouvrir de nouvelles perspectives thérapeutiques de traitement de l'HB
Introduction: Hemophilia B (HB) is an inherited X-linked recessive bleeding disorder, due to a defect in human factor IX (FIX). Replacement therapy, in severe HB is very effective but is limited by FIX concentrates injections frequency and cost issues. Production of a recombinant FIX with enhanced clotting activity and prolonged half-life is one of the current challenges for HB treatment. Materials and Methods: To improve activity, we focused on an important residue known to be involved in the interaction of activated FIX with its cofactor, activated factor VIII (FVIIIa), and four mutated FIX-E410 were developed. To prolong stability, a new chimeric FIX cDNA was constructed too. Recombinant FIX molecules were produced by the human hepatoma cell line Huh-7. Results: The in-vitro clotting activity of FIX-E410 was 3 to 5-fold higher than wild-type FIX (FIX-WT) and this improvement was confirmed using thrombin generation assay. FIX-E410H induced 5.2-fold higher thrombin generation than FIX-WT. In HB mice, we observed significantly higher in-vivo clotting activity and thrombin generating capacity with FIX-E410H compared to FIX-WT, mainly explained by 2.5-fold enhanced affinity of the mutant for FVIIIa. Chimeric FIX showed a 10-fold increase in the in-vitro molar specific activity and a significantly increased half-life in mice (up to 2.8-fold), compared to FIX-WT. Conclusion: We have engineered and characterized four improved FIX proteins with enhanced in- vitro and in-vivo activity, and a new chimeric FIX with in-vivo increased activity and prolonged half- life. These results suggest that these new molecules could optimize protein replacement therapy forHB treatment

Várias doenças relacionadas à desregulação do sistema imunológico têm sido, há muito tempo, tratadas com a medicina fitoterápica, considerando que vários efeitos terapêuticos podem ser induzidos pela modulação de citocinas. Entretanto, não são encontrados relatos científicos do uso de plantas medicinais na dengue. A identificação de compostos com propriedades imunomoduladoras é extremamente importante para o tratamento da dengue, uma vez quea gravidade da doença está relacionada a uma produção exacerbada da resposta imunológica, principalmente, de citocinas que podem levar à distúrbios hemodinâmicos e de coagulação. A família Marcgraviaceae, pertencente à flora brasileira, tem sido avaliada quanto às suas atividades biodinâmicas, devido ao seu quase completo ineditismo químico e farmacológico. A atividade anti-inflamatória dessa família tem sido descrita por alguns autores. Assim sendo, o objetivo principal do trabalho foi caracterizar os potenciais antivirais e imunomoduladores de uma espécie de Marcgraviaceae. Foi estabelecido e utilizado um modelo in vitrocultivando uma linhagem de hepatócitos \2013 as células HuH-7- que foram infectadas com DENV-2 As células HuH-7 infectadas foram incubadas, por 48 horas, com uma fração butanólica e 4 subfrações (26-30, 40-43, 89-98 e 99-134) derivadas de um extrato bruto etanólico de folhas de uma espécie de Marcgraviaceae. As amostras passaram por uma triagem, a qual identificou a atividade antiviral das mesmas, através da detecção do antígeno viral (DENV Ag) nas células HuH-7, pela citometria de fluxo e a atividade imunomoduladora, através da dosagem de MIF, por ELISA, no sobrenadante das culturas infectadas e tratadas. Os resultados da triagem indicaram que a fração butanólica e sua subfração 89-98 apresentaram ambasas atividades, sendo selecionadas para os demais estudos de caracterização dos mecanismos de ação. Após a seleção, a atividade antiviral destas frações/subfrações foram avaliadas quanto à produção de óxido nítrico (NO), através dareação de Griess, e detecção da proteína viral NS1, através de ELISA. A ação imunomoduladora foi avaliada através da dosagem de IL-8 também por ELISA. Nossosresultados demonstraram que a fração butanólica e sua subfração 89-98, foram capazes de reduzir a taxa de produção de NS1, além de estimular a produção de NOpelas células HuH-7 Além disso, a subfração 89-98 também reduziu a produção de IL-8. Em conclusão, acreditamos que estas frações/subfrações sejam boascandidatas para o desenvolvimento de fitoterápicos a serem usados no tratamento da dengue. Considerando que o organismo é capaz de controlar ainfecção por DENV em poucos dias e que as reações imunológicas possuem um papel importante na patogênese da dengue, o estudo de fatores imunomoduladores é uma abordagem de investigação inovadora e relevante
Various diseases related to immunodesregulation have been long treated by herbal medicine, considering that several therapeutical effects are observed in the modulation of cytokines. However, there are few reports regarding to the use of medicinal plants in dengue. The identification of compounds with immunomodulatory properties is extremely important for the treatment of dengue, since the disease severity is related to an exacerbated production of the immunological response, mainly cytokines that may lead to hemodynamic and coagulation disorders. Marcgraviaceae, a family of the Brazilian flora, has been evaluated for its biodynamic activities, due to its almost complete chemical and pharmacological uniqueness. The anti-inflammatory activity of this family has been described by some authors. Therefore, the aim of this work was to characterize the antiviral and immunomodulator potentials of a Marcgraviaceae species. It was established and used an in vitro model cultivating an hepatocyte cell line – HuH-7 cells – that were infected with DENV-2. Infected, HuH-7 cells were incubated with a buthanolic fraction and four subfractions ((26-30, 40-43, 89-98 e 99-134) derived from a crude ethanol leave extract from a Marcgraviaceae species, for 48 hours. The samples went through a screening that revealed their antiviral activity, by detection of viral antigen (Ag DENV) in HuH-7 cells, using flow cytometry.The immunomodulator activity was measured by ELISA detecting MIF levels in the supernatant of infected and treated cultures. The results of screening indicated that the buthanolic fraction and its subfraction 89-98 presented both activities, being selected for the other studies to characterize the mechanisms of action. After selection, the antiviral activity of these fractions/subfractions were evaluated for the production of nitric oxide (NO) by Griess reaction, and viral protein NS1 detection, by ELISA. The immunomodulator action was evaluated by IL-8 measurement by ELISA as well. Our results demonstrated that the buthanolic fraction and its subfraction 89-98 were able to reduce NS1 production ratio, besides stimulating NO production by HuH-7 cells. Furthermore, the 89-98 subfraction also reduced IL-8 production. In conclusion, we believe that these are good candidates for the development of herbal medicines for the treatment of dengue. Considering that the host is able to control infection by DENV in a few days and that immune responses develop an important role in the pathogenesis of dengue, the study of immunomodulatory factors is a relevant approach in research.

Abstract:
Notre étude sur le virus de l'hépatite C (VHC) de génotype 2a a initialement été entreprise pour pallier le problème lié à la production virale du VHC. Ayant obtenu des titres comparables à ceux publiés, nous avons cherché à développer une approche expérimentale nous permettant de les améliorer. Pour cela, nous avons effectué des infections successives de cellules Huh-7 naïves. Nous avons montré, dans un premier temps, qu'il était possible d'obtenir des cellules Huh-7 chroniquement infectées pendant plusieurs mois. La localisation subcellulaire des protéines structurales, telles que la protéine de la capside et les glycoprotéines E1 et E2, ou de la protéine non structurale NS3 a alors été réexaminée dans le contexte d'un cycle infectieux du VHC. L'analyse de la localisation subcellulaire des protéines structurales du VHC, en immunofluorescence en microscopie confocale, a confirmé que les glycoprotéines E1 et E2 sont maintenues dans le réticulum endoplasmique des cellules infectées. Cependant, et contrairement à d'autres études, ces glycoprotéines ne s'accumulent pas dans les autres compartiments intracellulaires ou à la surface cellulaire. L'association entre la protéine de la capside et les gouttelettes lipidiques a également été confirmée. Cependant, contrairement à certaines études précédentes, la protéine C n'a pas été trouvée dans le noyau de la cellule ou en association avec des mitochondries. De façon surprenante, nous n'avons pas observé de colocalisation entre les hétérodimères E1E2 et la protéine C en cellules infectées. Par ailleurs, nous avons observé une colocalisation partielle entre la protéine de la capside et la protéine NS3, et lorsque les gouttelettes lipidiques sont révélées la protéine NS3 se retrouve autour de ces gouttelettes. Ainsi, la localisation subcellulaire des protéines structurales du VHC a pu être étudiée pour la première fois dans le contexte d'un cycle infectieux. Dans un second temps, nous avons cherché à définir si des changements dans le génome viral du clone JFH-1 pouvaient expliquer l'augmentation des titres infectieux que nous avions obtenu au cours des infections successives des cellules Huh-7. Le séquençage de l'ARN du VHC nous a permis de déterminer qu'une mutation majeure, N534K, était apparue dans la séquence codant la glycoprotéine E2 du virus. De manière intéressante, cette mutation empêche la glycosylation d'un des sites de N-glycosylation présent sur la glycoprotéine E2. En outre, lorsque cette mutation est directement introduite dans la séquence du JFH-1, elle facilite l'infection des cellules Huh-7 naïves. Dans une deuxième approche, et fort de nos résultats obtenus dans une étude portant sur des virus chimériques 1a-2a du VHC, nous avons mis en évidence que la sécrétion des particules virales du VHC de génotype 2a pouvait être améliorée par la substitution de deux acides aminés (F172C et P173S) localisés dans la séquence codant la protéine de la capside du VHC. L'insertion de ces variations (F172C, P173S et N534K) dans le génome du VHC de génotype 2a permet de produire des titres infectieux très conséquents. Ainsi, la substitution de certains acides aminés dans la séquence codant les protéines structurales permet de produire des titres infectieux du VHC importants et indépendemment de la lignée cellulaire Huh-7 utilisée. Lors de l'étude des mutants hautement productifs du clone JFH-1, nous avons observé un phénomène de cytotoxicité important sur la lignée cellulaire Huh-7. Bien que ce phénomène ne soit pas encore expliqué, nous avons été en mesure de sélectionner plusieurs clones cellulaires résistants à cet effet cytopathique. L'analyse de ces clones a montré qu'ils étaient résistants à l'infection par le VHC. Cette résistance est le résultat de la perte d'expression d'un récépteur majeur pour l'infection du VHC, la molécule de surface CD81. L'étude de ces clones cellulaires nous a permis de confirmer le rôle primordial que joue CD81 dans l'infection par le VHC.

Inom butiken ställs det allt större krav på köpmannens/butikschefens ledaregenskaper för att hon/han ska kunna motivera och därigenom behålla sin personal och kunderna i slutändan. Inom en franchiseverksamhet som 7-Eleven så driver den egna företagaren sitt företag med uppbackning ifrån franchisegivaren Reitan Servicehandel. Att lära sig motivera personalen i sin verksamhet är flerdimensionellt och inte alltid det lättaste att lyckas med.

När flera yrkanden står emot varandra vid ett klyvningsförfarande, och där inget alternativ anses vara bättre än det andra, har lantmätaren till uppgift att avgöra vilket yrkande som medför minsta olägenhet utan att oskälig kostnad uppkommer, FBL 11 kap. 7 §. I arbetet var detta vår frågeställning. Syftet med examensarbetet var att undersöka hur denna frågeställning löses i praktiken samt utreda klassificeringar av typsituationer för att underlätta tillämpningen av FBL 11:7. För att få en djupare förståelse om klyvningsinstitutets innebörd och användning krävdes omfattande litteraturstudier. Detta var det första steget i vårt arbete där vi studerade propositioner, förarbeten och kommentarer till fastighetsbildningslagen. Vi läste även rättsfall, rapporter och utförde intervjuer på lantmäterimyndigheten i Stockholm. Nästa steg i processen blev att undersöka klyvningsärenden. Lantmäterimyndigheten i Stockholm tillhandahöll arbetsrum, datorutrustning och inloggningsuppgifter så att vi kunde använda oss av sökmotorn Trossen , som är ett digitalt databasarkiv. Genom programvaran kunde vi söka upp klyvningsärenden och sedan granska dess akter i fastighetsregistret. Syftet med granskningen var att gallra ut alla överenskommelser och endast använda de ärenden som hade något samband med vår frågeställning om tvistande lottilldelningar. Sökningen begränsades till fem år tillbaka i tiden och med hälften av Sveriges län. Hela sökningen har statistiskt redovisats i undersökningen. Vi kunde sedan klassificera typsituationer utifrån de rättsfall och ärenden som vi erhöll genom undersökningen. Klassificeringarna blev följande: - Dispositiva metoder - Medling - Tvingande metoder (minsta olägenhet utan oskälig kostnad) - Separering - Hävd och avsikt - Sämjedelad fastighet - Hus på ofri grund - Lottning Lagrummet bör utformas i en riktning som ger lantmätaren ökade möjligheter att besluta i situationer där parterna är oense om uppdelningen. Med klarare riktlinjer för hur de personliga omständigheterna ska användas, undviks slumpen genom lottning.

Sammanfattning   I den här rapporten har vi undersökt sex lärares uppmuntrande av resonemang under matematiken med extra fokus på algebra. Lärarna undervisar årskurserna 5-7. Syftet var att se vilka resonemang som lärarna uppmuntrar till i matematiken och hur de introducerar algebra för eleverna. Vi har i rapporten utgått från två typer av resonemang nämligen det imiterande och det kreativa. Med det imiterande resonemanget menas att eleverna plagierar en metod som visats för dem av läraren, läroboken eller liknande. Med ett kreativt resonemang menas att eleverna får reflektera, diskutera och få en djupare förståelse av metoderna. För att undersöka detta har vi gjort kvalitativa intervjuer med samtliga sex lärare och dessutom observerat tre av dessa lärares matematiklektioner. Resultatet visade att fyra av de sex lärarna inleder med en genomgång följt av att eleverna får räkna i matematikboken eller annan form av enskilt arbete. Detta medför att eleverna ofta får träna imiterande resonemang. Även när läraren står och hjälper eleven vid dennes bänk syntes det imiterande resonemanget dominera. En orsak som framkom var tidspressen över att hinna hjälpa samtliga elever i klassen. Ett fåtal inslag av det kreativa resonemanget syntes dock bland annat då en av lärarna använde sig mycket av problemlösning under sina lektioner vilket gynnar kreativiteten. Vad gällde algebran så blev vi presenterade för några olika sätt att introducera och det är svårt att säga vilket sätt som är att föredra. Dock är lärarna överens om att algebra är något som eleverna uppfattar som svårt i början och därför anser vi att introduktionen är av största vikt för att fånga elevernas intresse.

Abstract:

Vi vill med vår uppsats ta reda på om vad mobbning egentligen är, hur den uppkommer, vad som kännetecknar mobbare, mobboffer och medlöpare samt hur man kan förebygga och åtgärda mobbning.

Vårt syfte med uppsatsen är också att ta reda på hur elever uppfattar mobbning. Anledningen till att vi valt att skriva om mobbning är vi vill få ökad kunskap om detta inför vår kommande yrkesroll och vi anser att elever har en viktig roll i arbetet mot mobbning.

I uppsatsen tar vi del av hur litteraturen beskriver mobbning och vi gör en undersökning genom kvalitativa intervjuer med elever i årskurs 3 och 7. Det vi får ta del av kopplar vi till vårt resultat.

Undersökningens resultat påvisar att elever definierar mobbning som att det är när flera elever retas eller slår en elev och att detta sker ofta. Eleverna tycker också att mobbning förebyggs genom ökad kunskap och information om mobbning, ett bra rastvaktssystem och mobbningsgrupper i alla åldersgrupper. De anser också att mobbning kan åtgärdas genom samtal med elever, lärare, rektor och föräldrar.

Syftet med studien är att undersöka hur programmering för matematisk problemlösning kan integreras i matematikundervisning i årskurs 7-9. Skolverket har nyligen ändrat kursplanen för matematik så att programmering för matematisk problemlösning inkluderas i det centrala innehållet. För att undersöka syftet genomfördes studien i två delar. Lärare med programmeringskompetens intervjuades om hur de anser att programmering för matematisk problemlösning kan integreras i matematikundervisning. Två programmeringsverktyg undersöktes för att få svar på vilka möjligheter till arbete med matematisk problemlösning programmeringsverktyg erbjuder. Resultaten analyserades utifrån Bernsteins teori om klassifikation och inramning och visar att det finns många olika sätt att integrera programmering i matematikundervisning. Stora möjligheter finns för integrering av programmering för matematisk problemlösning med det centrala innehållet i kursplanen för matematik. Olika verktyg, uppgifter och arbetssätt erbjuder olika pedagogiska möjligheter.

När en konkursförvaltare förvaltar ett konkursbo, måste han välja vilka frågor i konkursen som är värda att driva och vilka frågor som riskerar att kosta mer än den potentiella nyttan av en framgångsrikt driven fråga. Alla beslut som förvaltaren ställs inför, ska fattas med beaktande av den allmänt formulerade bestämmelsen i 7 kap. 8 § konkurslagen (1987:672) om att tillvarata borgenärernas gemensamma rätt och bästa. Utifrån den bestämmelsen finns det olika åsikter inom doktrinen avseende vilken metod förvaltaren ska tillämpa för att uppnå bestämmelsens ändamål. Förespråkare för metoden tillgångsmaximering menar att vissa frågor som förvaltaren ställs inför, ska beslutas av konkursborgenärerna och inte av konkursförvaltaren. Dessa frågor betecknas medlemsfrågor och avser i regel åtgärder som inte tillför konkursboet ökade till-gångar, men som likväl kan gynna borgenärerna. Förespråkare för metoden utdelningsmaxime-ring menar att det ingår i förvaltarens åligganden att ta beslut även i dessa medlemsfrågor. Rättsutvecklingen inom konkursrätten har medfört dels att konkursboet numer ses som en juri-disk person och inte enbart en förmögenhetsmassa, dels att konkursförvaltarens koppling till borgenärerna har försvagats och numer är indirekt. Rättsutvecklingen talar därför för att förval-taren ska agera som konkursboets företrädare och tillvarata borgenärernas intressen indirekt, genom att tillgångsmaximera konkursboet. Metoden gynnar främst resursstarka borgenärer med kunskap inom konkursrätt. Det faktum att förvaltaren har fått en mer självständig roll och med ett vidgat ansvarsområde utöver de strikt borgenärsorienterade frågorna, talar däremot för att metoden utdelningsmaximering bör tillämpas. Ytterligare stöd för utdelningsmaximering är att kraven för vem som får vara konkursförvaltare har skärpts, vilket innebär att förvaltaren kan anses lämplig att fatta adekvata beslut även i medlemsfrågor, samt att metoden ger förvaltaren större möjligheter att påverka borgenärernas utdelning positivt. Metoden gynnar främst resurs-svaga borgenärer utan kunskap inom konkursrätt och förefaller vara marginellt mer lämpad utifrån ändamålet i 7 kap. 8 § konkurslagen. Att fastställa den ena eller den andra metoden som gällande rätt, vore emellertid att begränsa förvaltaren i sitt uppdrag, eftersom borgenärers behov kan variera. Det finns således ett värde i att kunna välja metod. För att avgöra vad som är en borgenärers rätt och bästa, måste först borgenärens behov fastställas. En resurssvag borgenär kan ha en avsevärd nytta av att låta för-valtaren avgöra medlemsfrågorna, medan en resursstark borgenär kan gynnas av ett större in-flytande i konkursen. Skulle borgenärernas behov krocka, torde förvaltaren vara väl lämpad att fatta beslut även i frågan om vilken metod som ska tillämpas.

Hepatocellular carcinoma is a type of primary liver cancer that accounts for 75% of all liver cancers globally. HCC is said to be the 6th most prevalent cancer and the 3rd major cause of cancer related death accounting for approximately 0.32 million annual deaths across the globe. The increasing incidence of HCC with a very high mortality rate is a major problem encountered by many regions of the world including both developing and developed countries and is a source of increasing economic burden. Therefore, effective treatment strategies for HCC are the need of the hour. Oxidative stress can be referred to as the lack of proportion between generation of reactive oxygen species and their removal by antioxidant defense mechanisms. This imbalance can have deleterious effects and contributes to the pathophysiology of several disorders including inflammation, aging cataract, diabetes and cancer. However, this controversial role of oxidative stress leading to apoptosis can be acquired as a treatment for HCC. The current study aimed to investigate the effect of induced oxidative stress via hydrogen peroxide (H2O2) on the proliferation of HCC Huh-7 cells in-vitro. The toxicity generated by H2O2 produce anti- proliferative effect on HCC Huh-7 cells and up regulates the oxidative stress markers. Proteomic studies were conducted in order to identify differential protein expression as potential therapeutic targets for the treatment of HCC.

Abstract:
Solid tumors in the liver such as hepatocellular carcinoma, often are not amenable to chemotherapy or surgical therapies. Local “ablation” methods including thermal and chemical ablation are therefore options for treatment. Hyperthermic ablation methods (RF, microwave, HIFU and laser) can potentially accomplish treatment in a single session but are considerably more costly than chemicals. It was therefore of interest to investigate if a known protein denaturant such as urea would destroy liver tumors. Further, we wished to assess whether this destruction occurs by protein denaturation mechanisms similar to hyperthermic destruction. Specifically, Lepock showed that hyperthermic cell destruction occurs for many cells when overall protein denaturation is greater than 5% (i.e. survival drops from 95% to 5% beyond this range). In this study we report on the cytotoxicity of urea on human hepatoma HuH-7. We then quantify the amount of cellular protein denaturation associated with cytotoxicity and show that protein denaturation in excess of 5% of total protein must occur in order for significant cell killing.

It has been suggested that a vWF High Molecular Weight Multi-mers (HMWM) decrease or a PAA were involved in the pathogenesis of HUS. We have studied 8 children (6 girls,_2 boys; 7 months-8_1/2 years old) with HUS : plasma creatinine /μmol/l; mean(range)/=306 (105-524), hemoglobin (g/100ml)-7(6.3-7.8), schistocytes (%)=8(1-18), platelets (x103/mm3)-57(10-115). The vWF was studied quantitatively (antigen ; vWF RAg assay) and qualitatively (multimeric pattern : immunoblotting and autoradiography). PAA studied by incubating the patient's platelet poor plasma (RPR) with washed normal platelets (aggregometer, % light transmission) and confirmed by Thromboxane B2 (TXB2) assay and [14C] Serotonine release study. The PAA was characterized by studying the in vitro effect of several platelet aggregation inhibitors, Immunoglobulins (Igs) and Fresh Frozen Plasma (FFP) on the platelet aggregation.An increase of vWF RAg (%) was observed in 6 cases : mean:330, and possibly related with renal failure. A vWF HMWM decrease was found in 3 patients : 2/3 with associated infection(E.Coli, Pneumococcus), 1/3 with severe hemolysis. Two of these 3 patients had a favourable renal outcome and 1 a severe course (chronic hemodialysis, Arterial Thrombotic MicroAngiopathy at renal histology).An important PAA was evidenced only in 1 patient : post bone-marrow graft HUS during neuroblastoma(NB),arterial hypertension and chronic renal failure. This PAA was Ca++, TXB2 and cAMP dependent; it was moderately inhibited in vitro by Igs and FFP, but persisted after 5 days of Igs infusion (0.3g/Kg/day). Treatment with aspirin and dipyridamole (10mg/Kg/day each) suppressed the patient platelet auto-aggregation although the PAA persisted (follow up:10months). The PAA did not seem to be related with the NB (absence of GD2 ganglioside, specific marker of NB); it could be related with anti platelet antibodies. The coexistence of the two abnormalities could not be demonstrated in our patients.In conclusion, a vWF HMWM decrease was found in 3 out of 8 children patients with HUS. Its presence was not correlated with the severity of the disease. We could demonstrate the presence of PAA during childhood HUS in only 1 post bone-marrow graft case. The PAA characterization is useful for therapeutic decisions and contributes to a better pathogenetic understanding.

Abstract:
This study experimentally measured the efficiency of new generation bicycle hub gears. Since efficiency of bicycle drivetrains can be very close to 100% and vary by small amounts between gears bias errors and measurement accuracy must be identified and controlled. For this study an ergometer frame was altered to support research test equipment. A 1 HP motor and gearbox were used to drive the crankshaft. The hubs were attached directly to the steel flywheel using shortened bicycle spokes, eliminating extra chains or drive components. This setup minimizes measurement uncertainty in the drivetrain. Force-transducers were used to measure the motor and flywheel torque, and two magnetic reed switches were used to measure the speed of the motor and flywheel. Efficiency for each individual gear in each hub was calculated for 14 different power speed combinations. The efficiency of each gear was plotted against flywheel Torque, and an exponential model was fit to the data. This model accounts for known variations in efficiency with power and speed, and provides insight into the torque-speed-efficiency relationship. Four internally-geared hubs were measured and compared with a single speed direct drive train and a belt drive. The internal planetary gear hubs measured were the Shimano Alfine 11, Rohloff 500/14 Speedhub, SRAM Dual-Drive, and the Sturmey Archer X-RK8(W). In addition a single-speed direct chain drive, a single-speed belt drive, and a 7-speed derailleur system were measured. The efficiency of the Shimano Alfine ranged from 90.4% to 96.6%. The efficiency of the Rohloff speed hub ranged from 95.8% to 99.5%. The efficiency of the Sturmey Archer hub ranged from 84.6% to 99.8%. The efficiency of the single speed chain drive was found to be 99.71% and the belt drive 98.0%. The efficiency of the 7-speed derailleur ranged from 97.7% to 99.4%. These values found for efficiency are comparable to other studies.

In 2020, the University of Dundee initiated the development of an Open Research strategy. As part of this initiative, in February 2021 the University’s Library and Learning Centre together with Open Research Champions from the Schools of Health Sciences and Dentistry, formed an Open Research Working group. To build on the University’s open research policy and infrastructure, the purpose of the group was to facilitate ongoing research and development of best practice approaches for our interdisciplinary environment to make outputs, data and other products of our research publicly available, building on University of Dundee’s Open Research policy and infrastructure. Through informal consultations with academic staff and students, the Open Research Working Group found that: → access and reach of research findings can be amplified through effective knowledge mobilisation, and stakeholder and patient and public involvement; and → there was a need for guidance and resources on how-to implement knowledge mobilisation activities with and for stakeholders throughout the entire research process – from proposal development to project completion. In June 2021, the Open Research working group, in partnership with Simon Fraser University’s Knowledge Mobilization Hub began the development of an Integrated Knowledge Translation (IKT) Toolkit, with funding support from the University of Dundee’s Doctoral Academy and Organisational Professional Development. IKT is an approach to knowledge translation that emphasises working in an engaged and collaborative partnership with stakeholders throughout the research cycle in order to have positive impact. The aim was to co-produce evidence-informed, best practice learning materials on how-to: → maintain ongoing relationships between researchers, community stakeholders and decision-makers in research development and implementation; and → facilitate an integrated, participatory way of knowledge production whereby researchers, practitioners and other knowledge users can collaborate to co-generate new and accessible knowledge that can be utilised in contexts ranging from supporting community development to policy guidance for practice. The IKT Toolkit was informed by a focused evidence review and synthesis of published peer-reviewed and grey literature and consists of 8 knowledge briefs and a slide deck co-produced for use in any discipline or sector. Each knowledge brief provides practical guidance and resources to support an IKT process in each of eight key research stages: (i) Partnership Building; (ii) Generating Priorities and Ideas; (iii) Proposal development; (iv) Study Design; (v) Data Collection; (vi) Data Analysis; (vii) Reporting and (viii) Dissemination.
