Abstract:
Abstract Background: Glycosylation profoundly influences the interactions between cancer cells and microenvironmental stromal cells during the peritoneal disseminated metastasis of ovarian carcinoma (OC), which is the major cause of cancer-related death. Although the characteristic cancer glycoconjugates are widely used as biomarkers for cancer diagnosis, our knowledge about cancer glycome remains quite fragmented due to the technique limitations in analyzing glycan chains with tremendous structural and functional heterogeneity. Method: Given the dysregulated cancer glycome is defined by the altered glycosylation machinery, here we performed a systematic loss-of-function screen on 498 genes involved in glycosylation for key regulators of OC dissemination. We identified neuraminidase 4 (NEU4), an enzyme capable of hydrolyzing terminal sialic acid from glycoconjugates, as a vital peritoneal dissemination-promoting modifier of OC glycome. What’s more, we also identified membrane proteins whose sialylation was able to be modified by NEU4 in OC cells by pull down and mass spectrum to investigate the functional significance of NEU4-mediated desialylation. Results: In human patients with high-grade serous OC (HGSOC), increased NEU4 was detected in the disseminated OC cells when compared with that in the primary tumor cells, which significantly correlated with the worse survival. Among three alternative splice-generated isoforms of human NEU4, we revealed that only the plasma membrane-localized NEU4 isoform 2 (NEU4-iso2) and intracellular isoform 3 promoted the peritoneal dissemination of OC by enhancing the cell motility and epithelial-mesenchymal transition. We also identified NEU4-iso2-regulated cell surface glycoproteome and found that NEU4-iso2 desialylated the epithelial growth factor receptor (EGFR), in particular at N196 residue, for the hyperactivation of EGFR and its downstream tumor-promoting signaling cascades. Conclusion: Our results provide new insights into how the OC glycome is dysregulated by NEU4 during OC progression and reveals a functionally important glycosite on EGFR for its abnormal activation in cancer. Sulfur- and Coking-tolerant Anodes for Solid Oxide Fuel Cells Zhijun Liu,1 Yucun Zhou,1 Weilin Zhang,1 Jie Hou,1 Xueyu Hu,1 Meilin Liu1, * 1 School of Materials Science and Engineering, Georgia Institute of Technology, Atlanta, Georgia 30332-0245, United States Abstract Solid oxide fuel cells (SOFCs) have potential to be one of the most efficient systems for direct conversion of a wide variety of chemical fuels into electricity. 1-3 However, the lack of sulfur- and coking-tolerant anodes still hinders the development of direct hydrocarbon-fueled SOFCs for efficient and cost-effective operation. The conventional Ni-based anodes may suffer from carbon deposition and sulfur poisoning when they are directly exposed to sulfur-containing hydrocarbon fuels, resulting in degradation in performance or even destruction of the anodes. 4-6 In this presentation, we will report a conventional Ni-Zr0.84Y0.16O2-δ (YSZ) anode decorated with Ce and Ba-based catalyst coatings to significantly enhance the sulfur and coking tolerance. Results suggest that the enhanced sulfur tolerance is attributed to the evenly distributed Ce-based catalyst on the inner surface of the porous Ni-YSZ anode while the enhanced coking tolerance is benefited from the Ba-based catalyst layer on the top of the anode. SOFCs with the catalysts-modified Ni-YSZ anodes show minimal degradation when hydrogen with 100 ppm H2S is used as the fuel. More importantly, stable operation is demonstrated for direct utilization of sulfur-containing liquid fuels (e.g., octane and gasoline). Abstract The deadliest anaplastic thyroid cancer (ATC) often transforms from indolent differentiated thyroid cancer (DTC), however the complex intra-tumor transformation process is poorly understood. We investigated an anaplastic transformation model by dissecting both cell lineage and cell fate transitions using single cell transcriptomes and genetic alterations data of patients with different subtypes of thyroid cancer. The resulting model started from stress-responsive DTC cells to inflammatory ATC cells, to mitotic defective ATC cells and extended all the way to mesenchymal ATC cells. In parallel with tumor cell evolution, macrophages shifted from anti-tumor to tumor-promoting states and T cells reprogrammed from cytotoxic to exhausted states. Further, our analysis identified two important milestones: 1) diploid stage, where ATC cells were commonly diploids with non-RAS mutations and inflammatory phenotypes. 2) aneuploid stage, where ATC cells gained aneuploidy with frequent RAS mutations and mesenchymal phenotypes leading to the extreme lethal stage of ATC progression. Abstract:
Alternative Chinese film culture has become increasingly diverse since the 1990s. This diversification is related to the kinds of films being produced and to who has ‘the right’ to make and to view them. A significant number of new films are documentaries, distinct in form from the newsreels (xinwen pian) or special topic films (zhuanti pian) that represent the more traditional official forms of documentary. Some of these new unofficial (non-state funded) documentaries have attempted to create alternative space for the telling of individual stories about sensitive and traumatic historical periods such as the Great Leap Forward, the Great Famine, and the Cultural Revolution. Such stories are in marked contrast to official visual histories of these periods. This paper’s focus is the new alternative space created by two individual filmmakers, Hu Jie and Wang Bing, who have documented unofficial, individual histories. Three of their films are examined in the context of a film industry that offers them severely limited or no access at all to official channels of distribution. The paper begins by examining the manner in which historical narratives have been and are ongoingly integral to the construction of a specific official view of the Chinese nation. Such official narratives are dominant in state-run environments, such as schools and in the media, and they tend to drown out any attempted alternative narratives. Hu Jie and Wang Bing stand out, even within alternative film culture, as filmmakers willing to challenge official doctrine and history. The narrative styles of three of their films are closely analyzed with a view to understanding how and why their work differs from that of official historical narratives. The approaches taken by the two filmmakers are also compared and contrasted. A major aspect of this project has been the translation of Hu Jie’s film In Search of Soul of Lin Zhao into English. The other two films examined have already been translated. This paper argues that by privileging the individual narrative, the two filmmakers present more complex versions of traumatic historical periods that stand in contrast to the often idealized or superficial official histories.

Le présent travail souhaite contribuer à la recherche sur des créations documentaires indépendantes dans la Chine contemporaine qui se confrontent à l’histoire nationale. Dans un contexte intellectuel et artistique d’exhumation et transmission de faits encore enfouis, refoulés ou privés de représentation publique, nous observons trois cas exemplaires à cet égard : HU Jie, le Folk Memory Project du Caochangdi Workstation, et MAO Chenyu. Nous tentons dans notre étude de caractériser les propositions et gestes principaux de leur recherche d’écriture documentaire. Pour HU Jie, le geste principal est aussi le geste séminal du documentaire classique : établir les faits. À partir d’une réflexion sur les matériaux collectés et sur les images d’archive, HU Jie s’interroge sur la mise en forme de son sujet et sur les principes d’intelligibilité du réel que son écriture documentaire met en œuvre. Projet collectif à l’initiative du cinéaste WU Wenguang et de la chorégraphe WEN Hui, l’entreprise du Folk Memory Project consiste à s’engager des amateur-filmeurs ou de simples citoyens qui retournent dans leurs propres villages, filment et recueillent les témoignages de leurs proches ayant connu la Grande famine (1958-1961) en Chine. Le projet conçoit ainsi la pratique documentaire comme geste à la fois de création, d’interrogation, de transmission et d’introspection. MAO Chenyu, quant à lui, centre son travail sur le monde rural ainsi que la vie paysanne en Chine. En cultivant des formes fragmentaires, en recherche perpétuelle d’une articulation possible du cinéma avec d’autre pratiques actuelles et à venir, l’écriture documentaire de MAO Chenyu propose une démarche qui s’apparente à une ethnologie historique. This dissertation wishes to contribute to the studies about the independent documentary film creations in contemporary China which confront national history. In an intellectual and artistic context, where the exhumation and transmission of the buried facts are repressed or bereft of public representation, we would like to observe three exemplary cases in this regard : HU Jie, Folk Memory Project, and MAO Chenyu. Our study will try to characterise the different propositions and the principal gestures in their experimentations of documentary writing style.For HU Jie, the principal gesture is also the seminal gesture of classical documentary filmmaking : to establish the facts. Based on collected testimonials and archival footage, HU Jie explores in his practice how the subjects of his films and the historical facts would come to shape intelligibly. Folk Memory Project is an ongoing participatory documentary experience that engages amateur filmmakers or citizens to go back to their families’ rural villages, and film elders’ memories about difficult times. The project sees its practice as creation, investigation, transmission, but also intimate introspection. In the case of MAO Chenyu, who centers his work in the rural region and China’s peasantry, with his innovative approach that explores perpetually about the possible articulation between cinema and other practices, his work provides an ethnographic thinking to history writing.These three practices all participate to the critical approach of history writing in sounds and images, which reclaims history's collective and plurality becoming

This chapter critically reflects upon the Deleuzian notion of cinematic ethics, and that it provides a reason to believe in this world. Through close analysis of the Chinese documentary Wo sui si qu/Though I am Gone (Hu Jie, 2008) – a film in which Deleuzian time-images are used to reconsider history much as Deleuze considered typical of modern political or minor cinema – the sticking point of Deleuze’s cinematic ethics is revealed: namely, in a world of cinemas broadly conceived it is not this world which a cinematic ethics asks us to rejuvenate our belief in, but this world’s history (or rather, in the complexities of a world history of which we can only glimpse a fraction through cinema).

Abstract:
This chapter examines Ezra Pound's and J. H. Prynne's use of Chinese poetry to understand the problem of motivation—and the incentive structures that govern modern life. It considers how difficult poetry illuminates the difficulty of motivating social change. Here, Prynne's work, “Jie ban mi Shi Hu,” exemplifies a problem for readers of his work, and for literary and social theory: How and why should we read texts that make extravagant, even impossible demands? The question asks us to justify the value of particular texts and the values of the world that receive them. To put this point a different way, when we ask for reasons to accept a poem's invitation to do work, we should also ask what kind of world would have to exist to make the invitation seem reasonable, and whether we would want to live in that world. Similar questions are relevant to debates about utopianism.

This chapter describes a shift in Jia Zhangke’s cinematic language that reveals that his approach to filmmaking has evolved in unexpected ways following the release of his 2013 film A Touch of Sin. It explains how, in a highly stylized crime drama, Jia openly experiments with genre elements that borrow extensively from Hong Kong triad films while also paying homage to several classics within the wuxia tradition. The film, thus, marks a shift towards violent, formally ambitious crime stories continued with Jia’s two subsequent films, Mountains May Depart (2015) and Ash is Purest White (2018). The chapter, furthermore, offers an alternative reading of Jia’s films from a genre-oriented perspective, prioritizing his interest in the conventions of the crime genre over the meditative, naturalistic cinematic language that characterized his early work by tracing the genealogy of references and intertextual elements that inform Jia’s trilogy by identifying several sources of inspiration, including John Woo and King Hu, by bringing to light cinema as a site of the ongoing processes of conflict and reconciliation between a culturally independent Hong Kong and the consolidated nationalisms of China, and by exploring how Jia utilizes multi-episodic storytelling strategies to combine crime film conventions with social commentary.

Abstract The reliability of surface mount leadless solder joints, e.g., LCCC (leadless ceramic chip carrier), LCR (leadless chip resistor), and LCC (leadless chip capacitor), has been a long-term research topic due to the concern of thermal fatigue failure resulting from significant stresses and strains induced in the joint. Numerous studies of failure analysis of leadless solder joints exist in the literature, and a number of life prediction models based on different failure characterizing parameters, e.g., total strain, plastic strain, strain energy density, etc., have been developed for particular loading conditions, such as isothermal or thermal cyclic fatigue tests. ABSTRACT Exploring ways to enhance the liquid-phase seepage ability and increase the flow-back rate of external fluids in the production process is an important way to solve the problem of water blocking in tight reservoirs. Based on the analysis of the characteristics of water phase retention damage caused by foreign fluid invasion during the development of tight sandstone gas reservoirs, a set of ether nano-displacement agent system was optimized. The mechanism of wetting modification is revealed from the perspective of the physical composition of nano-displacement agent, and the mechanism of action between nano-displacement agent and rock and different clay minerals is clarified. Laboratory experiments show that :(1) 0.5 wt% CNDAD1# nano-displacement agent fluid can change the wettability of sandstone to gas-phase wetting, "air-salt water-rock" three-phase static contact angle increased from 38.5° to 126°. (2) SEM and EDX results prove that the nano-displacement agent has good macroscopic static and dynamic adsorption performance on the sandstone surface. This paper provides a reference method for exploring the use of environmentally friendly chemical agents to remove water blocking damage in tight sandstone gas reservoirs. INTRODUCTION Tight sandstone gas reservoirs refer to sandstone gas reservoirs with overburden matrix permeability less than or equal to 0.1 mD. Single wells generally have no natural productivity or natural productivity is lower than the lower limit of industrial gas flow. Industrial natural gas production needs to be obtained through fracturing, horizontal wells, multi-branch wells, etc. (Wei G, Zhang F, Li J, et al., 2016; Jia C, Zheng M and Zhang Y, 2012; Zou N, Zhu R, Wu S, et al., 2012). According to the data, China contains a wealth of tight sandstone gas reservoirs, tight sandstone gas resources account for about 27.5 % of China ‘s natural gas resources, mainly distributed in different basins in China, such as Sichuan, Tarim Basin and other regions, and most of the reserves have not been effectively developed (Wang J, Hu Y, Liu Y, et al., 2019; Fuhai J, Tang D, Xu H, et al., 2012; Bybee K, 2007). Therefore, the development of tight sandstone gas reservoirs will become one of the key sources of future energy security in China, and it is also the focus of unconventional oil and gas development. However, most of the tight sandstone gas reservoirs have low natural productivity of single wells. The combination of horizontal well drilling and fracturing is one of the main ways to realize the commercial exploitation of tight sandstone gas reservoirs. However, during the fracturing process, the fracturing fluid flowback is not timely or the edge and bottom water invades the reservoir during the production process. The reservoir will produce a large amount of external water phase retention or edge and bottom water invasion water, forming a ‘ liquid phase trap ‘ and further tight sandstone gas well productivity (Dutta R, Lee C H, Odumabo S, et al., 2012; You Q, Wang H, Zhang Y, et al., 2018; Zhang L, Zhou F, Mou J Y, et al., 2018).
