Abstract:
The motivation to avoid punishment, or to pursue reward can result in certain behavioral tendencies. It is known that individuals with higher sensitivity towards reward can express traits of extrovertedness, openness to novel situations, and a general appetite for feeling good. Individuals on the other side of the spectrum tend to show a pessimistic attitude towards approaching novel situations. As a consequence, individuals with a higher tendency of approaching novel situations ought to find themselves in risky situations more frequently than individuals who would rather avoid novel situations. Looking at this from a human factors perspective, it becomes apparent that a tendency to approach situations with a lack of conflict monitoring can produce risks, but simeltaneously, disinterest in approaching a risky situation can lead to crisis events. In other words, whilst individuals with a sensitivity towards reward might have a proclivity for producing a crisis, they might also have a higher chance of solving a crisis. In order to solve unexpected crisis events, it is necessary to be able to improvise. In the experiment of this current thesis, individuals were assessed for their sensitivity towards reward and punishment, and following this, the individuals were assessed in improvisational skills. Motivation behavior and improvisation skills were investigated to see if there is a correlation between them. The purpose of this thesis was to present a multidisciplinary literature account for the relevant subjects by tying together aspects of cognition, cognitive neuroscience, human factors, and psychology, and to provide experimental data about the mentioned correlation. The present experiment showed that there is a significant correlation between sensitivity towards reward and improvisation skills. Additionally, it was also found that system-level factors i.e pre-implemented elements by a designer/organization might not facilitate improvisational performance, but this significant result did not appear in the expected manner. Future directions of research on this subject should focus on five mainpoints (1) replication of investigating the relationship between personality styles and improvisation with different methodologies, (2) investigating the mechanisms leading to the emergence of this relationship, (3) approach the subject mainly from a cognitive-neuroscientific standpoint, but complement with a multiple disciplinary approach (4) use the results within the disciplines of human factors, safety, and resilience, and (5) take data into consideration for e.g. future accident preventions policies.

Abstract:
Introduction: HTLV-1 associated Myelopathy / Tropical Spastic Paraparesis (HAM / TSP) is a chronic progressive demyelinating disease that affects the spinal cord and the brain's white matter. The Vestibular Evoked Myogenic Potential (VEMP) is a neural response from three-neuron reflex arc involving the inner ear, the brainstemand vestibular-spinal pathway. This test was evaluated in HTLV-1 infected individuais by cross-sectional study was more change in HAM / TSP compared to examine the asymptomatic individual. It is important to know the VEMP value for the monitoration of asymptomatic patients with symptoms but without the HAM / TSP criteria and the evolution to incipient neurological disorders. Objective: Longitudinal evaluation of theresults of VEMP in subjects with HTLV-1 in different disease progression stages, ranging from asymptomatic to individuais with established HAM / TSP. Methods: Prospective and comparative study, from 2006 to 2009. The participants underwent VEMP with an interval of six months, totaling four serial assessments. Were evaluated adults between 21 and 73 years, of both genders were divided into four groups: control group, consisting of healthy seronegative donors of the FoundationCenter of Hematology in Minas Gerais (Hemominas) and three other groups study comprised of HTLV-1 infected individuais, as follows: asymptomatic infected subjects, possible HAM / TSP and with defined HAM / TSP from the open cohort followed by the Group for interdisciplinary Research in HTLV (GIPH). For the record VEMP, were used tone burst at an intensity of 118 dB and in band-pass filter from 10Hz to 1500Hz, with 200 stimuli presented at a frequency of 1Hz with record time of60 ms. In the analysis, we considered P13 and N23 latency and amplitude. The amplitude variation was controlled by the asymmetry index. Results: 90 subjects, whereas 30 (33,3%) to the control group and 60 (66,7%) of the seropositive group, subdivided into subgroups 18 (30%) asymptomatic patients 25 (41,7%) with probableHAM / TSP and 17 (28,3%) defined HAM / TSP. A higher frequency of VEMP absent in individuais with HAM / TSP and no abnormal test reverted to normal examination or any absent VEMP test reverted to an increase of latency. Thus the altered response in VEMP latency prolongation has changed for the absence of evoked response to the evolution of neuronal damage. In the survival curve, it was observed that during the surveillance was more likely to change in VEMP response in ali groups, this finding being more pronounced in subjects with HAM / TSP, and there isa progression to a worse response. Conclusion: Vestibular Evoked Myogenic Potential (VEMP) was useful to monitoring HTLV-1 individuais and HAM / TSP progression, although is a progressive and slow course neurological disease. Possibly, asymptomatic individuais infected with HTLV-1 with altered VEMP will have more chance to develop neurological disease than those with normal VEMP.
Introdução: A Mielopatia associada ao HTLV-1/ Paraparesia Espástica Tropical (HAM/TSP) é uma doença desmielinizante crônica progressiva que afeta a medula espinal e a substância branca do cérebro. O Potencial Evocado Miogênico Vestibular - Vestibular Evoked Myogenic Potential - (VEMP) é uma resposta neural formada a partir de um arcoreflexo de três neurônios que envolvem a orelha interna, o tronco encefálico e a via vestíbulo-espinhal. Esse teste foi avaliado em indivíduos infectados pelo HTLV-1 a partir de estudo transversal mostrou-se mais alterado na HAM/TSP quando comparado ao exame doindivíduo assintomático. Por isso, é importante conhecer o valor do VEMP no acompanhamento de pacientes assintomáticos e com sintomas, mas sem os critérios definidos para HAM/TSP, quanto a sua evolução a partir de alterações neurológicas incipientes. Objetivo: avaliar, por meio de análise longitudinal do resultado do VEMP,sujeitos com HTLV-1 em diferentes estágios de progressão da doença, variando desde indivíduos assintomáticos até com HAM/TSP estabelecida. Métodos: Realizou-se estudo prospectivo, comparativo, de 2006 a 2009. Os participantes do estudo foram submetidos aoVEMP com intervalo de seis meses, totalizando quatro avaliações seriadas. Foram avaliados indivíduos adultos entre 21 e 73 anos de idade, de ambos os gêneros, subdivididos em quatro grupos: grupo-controle, composto de doadores soronegativos saudáveis da Fundação Centro de Hematologia e Hemoterapia de Minas Gerais (Hemominas); e outros três grupos de estudo compostos de indivíduos infectados pelo HTLV-1, a saber: sujeitos infectados assintomáticos, possível HAM/TSP e com HAM/TSP definida, selecionados da coorte aberta do Grupo Interdisciplinar de Pesquisa em HTLV (GIPH). Para o registro do VEMP, utilizou-se estímulo tone burst rarefeito, com intensidade de 118 dB Na e filtro passa-banda de 10Hz a 1500Hz, sendo apresentados 200 estímulos na freqüência de 1Hz com tempo de registro de 60 ms. Na análise, considerou-se latência e amplitude das ondas P13 e N23. A variação da amplitude foi controlada pelo índice deassimetria. Resultados: 90 sujeitos participaram do estudo, sendo que 30 (33,3%) pertenciam ao grupo controle e 60 (66,7%) ao grupo com sorologia positiva para o HTLV-1, subdividido nos subgrupos 18 (30%) assintomáticos, 25 (41,7%) com provável HAM/TSP e 17 com HAM/TSP definido. Verificou-se maior freqüência de VEMP ausente nos indivíduos com HAM/TSP e, nenhum exame alterado reverteu para exame normal, ou nenhum VEMP ausente reverteu para exame com aumento de latência. Assim, a resposta alterada no VEMP mudou de prolongamento de latência para ausência de resposta evocada com a evolução do dano neuronal. Na curva de sobrevivência, observou-se que ao longo do seguimento houve maior probabilidade de mudança de resposta do VEMP em todos os grupos, sendo mais acentuado este achado nos sujeitos com HAM/TSP, verificando-se progressão para pior resposta. Conclusão: O Potencial Evocado Miogênico Vestibular (VEMP) foi útil para o seguimento de indivíduos infectados pelo HTLV-1 quanto á evolução para HAM/TSP, apesar da evolução neurológica da doença ser progressiva e de curso lento. Possivelmente, indivíduos assintomáticos infectados pelo HTLV-1 com VEMP alterado terão mais chance de evoluir na doença neurológica do que aqueles com VEMP normal.

Abstract:
INTRODUCTION: Inflammatory manifestations are characteristic of the Human T-lymphotropic virus type 1 (HTLV-1)/ associated myelopathy/ tropical spastic paraparesis (HAM/TSP), which can affect the vestibulospinal tract and could be the explanation for the higher frequency of otoneurological manifestations described in HAM/TSP. However, this finding has not been systematically studied, so far. OBJETIVE: To compare the frequency of otoneurological manifestations in individuals infected with HTLV-1 that are asymptomatic and with HAM/TSP. METHODOLOGY: The participants of this study are included in the cohort of the Interdisciplinary Group of HTLV Researches GIPH, started at 1997 and with 637 ex-blood donors infected by HTLV-1 and 232 blood donors (controls) enrolled in the cohort at the moment of this analysis. The present study is a sectional evaluation of 120 individuals of the cohort, 60 of whom were infected with HTLV-1 (30 asymptomatic and 30 with HAM/TSP, according to neurological evaluation) and 60 not-infected, who were matched for gender and age with the HTLV-1-infected group. All participants underwent an interview about general health status and otoneurological manifestations. The participants who complained of dizziness, tinnitus or hearing loss were evaluated by the otolaryngologist and submitted to specific tests to define the diagnosis. The vestibular-evoked myogenic potential (VEMP) was performed to test the function of the vestibulospinal tract in those that complained of dizziness. The frequency of otoneurologic manifestations was then compared. RESULT: The complaint of hearing loss (p=0.506) and tinnitus (p=0.498) were similar. Dizziness predominated in the HAM/TSP group (p=0,002). Instability was the most frequent type of dizziness among the individuals infected by HTLV-1 (p=0,006). VEMP was altered in 43% of the asymptomatic carries and in 85% of the individuals with HAM/TSP. CONCLUSION: Dizziness as the sensation of instability was found to be a clinical manifestation of HAM/TSP. Vestibulospinal tract injury was demonstrated through VEMP to occur more frequently in HAM/TSP. Possibly, the complaint of dizziness in asymptomatic carriers may be an early indicator of progression to HAM/TSP.
INTRODUÇÃO: Alterações inflamatórias são características da infecção pelo Vírus Linfotrópico Humano de Células T tipo 1 (HTLV-1), em especial dos portadores da mielopatia associada ao HTLV-1/paraparesia espástica tropical (HAM/TSP), o que pode comprometer o trato vestibuloespinhal e poderia ser a justificativa para a maior frequência de manifestações otoneurológicas descritas na HAM/TSP. Contudo, esse achado não foi, até o presente momento, estudado de forma sistematizada. OBJETIVO: Comparar a frequência de manifestações otoneurológicas em indivíduos infectados HTLV-1 assintomáticos e com HAM/TSP. METODOLOGIA: Os participantes desse estudo estão incluídos na coorte do Grupo Interdisciplinar de Pesquisas em HTLV-1 - GIPH, iniciada em 1997 e com 637 ex-doadores de sangue infectados pelo HTLV-1 e 232 doadores de sangue (controles) cadastrados na coorte no momento dessa análise. O presente estudo trata-se de uma avaliação transversal de 120 indivíduos da coorte, sendo 60 infectados pelo HTLV-1 (30 assintomáticos e 30 com HAM/TSP, segundo avaliação neurológica) e 60 não infectados que foram pareados por gênero e idade com o grupo infectado pelo HTLV-1. Todos os participantes foram submetidos à entrevista sobre saúde geral e queixa otoneurológica. Todos os participantes que apresentaram queixas de tontura, zumbido ou perda auditiva foram avaliados pelo otorrinolaringologista e realizados exames específicos para definição do diagnóstico. O potencial evocado miogênico vestibular (VEMP) foi utilizado para testar a função do trato vestibuloespinhal naqueles com queixa de tontura. A frequência de alterações otoneurológicas foi comparada entre os grupos. RESULTADOS: Perda auditiva (p=0,506) e zumbido (p=0,498) foram semelhantes entre os grupos. A queixa de tontura predominou naqueles com HAM/TSP (p=0,002). Instabilidade foi o tipo mais frequente de tontura entre os indivíduos infectados pelo HTLV-1 (p=0,006). Dentre esses indivíduos com tontura do tipo instabilidade, o VEMP foi alterado em 43% dos portadores assintomáticos e em 85% dos indivíduos com HAM/TSP. CONCLUSÃO: Tontura se apresentando como uma sensação de instabilidade pode ser uma manifestação clínica da HAM/TSP. Lesão do trato vestibuloespinhal foi demonstrada através da alteração do VEMP ocorrer de modo mais frequente na HAM/TSP. Possivelmente, a queixa de tontura em portadores assintomáticos pode ser um indicador de início de evolução para HAM/TSP.

Abstract:
As anêmonas do mar utilizam rico complexo proteico para capturar suas presas e se defender de predadores. A peçonha destes animais contem neurotoxinas (35 kDa), com ação em canais iônicos específicos, e hemolisinas (1820 kDa), que atuam formando poros em membranas. No entanto, pouco se conhece sobre a atividade biológica de substâncias de baixo peso molecular isoladas da peçonha destes animais. Foi demonstrado, pelo nosso grupo, que a Bunodosina 391 (BDS 391), um composto de baixo peso molecular isolado da peçonha da anêmona do mar Bunodosoma cangicum, induz potente efeito antinociceptivo periférico, em modelos experimentais de dor aguda e persistente. Ensaios farmacológicos mostraram que a atividade analgésica do BDS 391 é mediada pela ativação de receptores serotoninérgicos 5-HT3. O BDS 391 é um composto bromado formado por um núcleo molecular semelhante a serotonina (5-HT), conectado, por meio de uma ligação peptídica, a uma histidina. A semelhança estrutural com a 5-HT e o envolvimento de receptores 5-HT3 no efeito antinociceptivo deste composto, torna de especial interesse a ampliação da caracterização dos mecanismos moleculares envolvidos na ação do BDS 391. Há evidências crescentes da co-expressão de receptores 5-HT e TRPV1 e a interação entre esses dois receptores na modulação da excitabilidade de neurônios sensitivos envolvidos na nocicepção. O receptor TRPV1 é considerado um receptor polimodal, uma vez que pode ser ativado por diversos estímulos. Baseado em estudos farmacológicos, mostrando que o BDS 391 apresenta potente efeito antinociceptivo em modelo de hipernocicepção mecânica, mediado pela ativação de receptores 5-HT3 periféricos o presente projeto de pesquisa teve como objtivo ampliar a caracterização dos mecanismos moleculares envolvidos na ação do BDS 391 investigando a interação deste composto com os receptores 5-HT3, por meio de ensaios de ligação (binding). Uma vez que nossos resultados demonstraram que o composto BDS 391 não é capaz de interagir diretamente com os receptores 5-HT3, sugerindo que o envolvimento destes receptores no efeito analgésico induzido pelo BDS 391 pode decorrer de uma acão indireta do BDS 391, avaliamos outros possíveis alvos farmacológicos para este composto. Por tanto investigamos, por meiode ensaios eletrofisiológicos e ensaios de imageamento de influxo de Ca2+ em cultura de neurônios do DRG de ratos, a possível ação do BDS 391 sobre correntes iônicas Ca2+ em canais para cálcio dependentes de voltagens (Cavs). Outro alvo avaliado foram os canais TRPV1. Foram realizados emsaios in vivo, utilizando modelo de hiperalgesia térmica induzida pela capsaicina, e ensaios in vitro (imageamento do influxo de Ca2+). Nossos resultados evidenciaram que o BDS 391 não interage com os Cavs, porem bloqueia o influxo de Ca2+ induzido pela capsaicina, além de inibira hiperalgesia térmica induzida pela capsaicina. Em conjunto, estes dados sugerem que os canais TRPV1 são alvos importantes da ação do BDS 391.
Sea anemones employ a wide range of bioactive compounds to capture their prey or fend off possible predators. Sea anemone venom contains neurotoxins (3-5 kDa), with action on ion channels, and also hemolysin (18-20 kDa), which acts by forming pores in membranes. However, little is known about the biological activity of low molecular weight substances isolated from the venom of these animals. One of these substances is Bunodosine 391 (BDS 391), a low molecular weight (391 Da) and non-peptidic compound purified from the venom of the Brazilian Bunodosoma cangicum sea anemone. We have demonstrated that BDS 391, administered by intraplantar (i.pl.) route into a rat hind paw, induces potent peripheral antinociceptive effect in models of acute and chronic pain. Pharmacological studies have shown that the antinociceptive effect of the BDS 391 is mediated by activation of 5-HT3 receptors. Studies on the structure of BDS 391 have demonstrated that this compound is made up of a bromoindole group connected to histidine. Data from the literature have shown that the peripheral release of 5-HT interferes with thermal hyperalgesia, via modulation of TRPV1 receptors. The aim of the present work is to further characterize the mechanisms involved in the antinociceptive effect of BDS 391 (a) evaluating the ability of this substance to directly activate 5-HT3 receptors, (b) characterizing the effect of BDS 391 on capsaicin-evoked thermal hyperalgesia and (c) investigating the involvement of TRPV1 ion channels in this effect. Pharmacological studies have shown that BDS 391 induces antinociception in the capsaicin-evoked thermal hyperalgesia. Ondansentron (5-HT3 receptor antagonist) inhibits the effect of BDS 391, indicating the involvement of 5-HT3 in the antinociceptive effect of this substance. However, binding studies have shown that BDS 391 does not directly interact with the 5-HT3 receptor. The possible effect of BDS 391 on TRPV1 ion channels was investigated in vitro, through Ca2+ imaging studies. Cultured DRG neurons were used for Ca2+ imaging. BDS 391 inhibited capsaicin-evoked Ca2+ influx in DRG neurons, indicating a modulatory action on the activity of TRPV1. Our results suggest that the antinociceptive effect of BDS 391 could involve the inhibition of TRPV1 channels and, indirectly, the modulation of 5-HT3 receptors.

Ericssons Business Communication Suite (BCS) är en Unified Communication-lösning, och är avsedd för större företag och teleoperatörer. BCS erbjuder tjänster som snabbmeddelanden, videosamtal, lägga till kontakter i grupper samt grupphantering mm. Syftet med detta projekt var tvåfaldigt; att utveckla och integrera ett program som kan kommunicera med Ericssons BCS, samt att dokumentera denna integration genom de erhållna kunskaperna från utvecklingen. Applikationen utvecklades för Windows Sidebar widget miljön och BCS miljön simulerades med hjälp av flera program och applikationer som tillhandahölls av Ericsson. Det första resultatet är en steg-för-steg integrationsguide, som innehåller installationsintruktioner för BCS-miljön samt riktlinjer för denna integration. Det andra resultatet är en widget som kan söka efter personer på sökmotorn www.eniro.se och utnyttja tjänster som tillhandahålls från BCS, som kontakt-och grupphantering samt videosamtal. Detta projekt visar att en tredjepartsutvecklad applikation kan integreras med BCS, och att tredjepartsutvecklare samt anställda på Ericsson kan använda steg-för-steg integrationsguiden för att sätta upp test- och utvecklingsmiljö för BCS.
Ericsson Business Communication Suite (BCS) is a Unified Communication solution destined for larger companies and telecommunication operators. BCS offers services such as instant messaging, video calls, contact group management and much more. The purpose of this project was twofold; developing and integrating an application that could communicate with Ericsson's Business Communication Suite, but also to document this integration through the knowledge acquired from the development. The application was developed for the Windows Sidebar widget engine and a BCS environment was simulated with the help of several programs and applications provided by Ericsson. The results of the project are a step-by-step integration guide, which contains environment setup as well as integration guidelines, and also a widget application that can search for people on the search engine www.eniro.se and utilize services provided by BCS such as contact and group management and video calls. This project demonstrates that a third party developed application can be integrated with Ericsson BCS, and that third party developers and people at Ericsson can use the step-by-step integration guide to easily setup the necessary BCS test environments.

Abstract:
Substation primary plant and instrument transformers generally remain in service for many years and are only renewed when physically or mechanically life-expired. However, substation protection and control systems, also known as secondary systems, are changed more frequently, especially electronic or computer based systems. One way to overcome the lifecycle-coordination problem is to develop a new architecture for substation secondary systems by deploying new technology such as standard interface modules, the bay process bus and IEC 61850 communication protocol. IEC 61850-9-2 'process bus' has proposed an Ethernet based communication network to replace traditional hardwiring and cabling, for communication of analogue and binary signals with Ethernet messages. The application of processbus in substations is able to reduce the life-time cost of the secondary systems and improve flexibility and functionality. Sufficient confidence must be demonstrated in the application of this new technology. This thesis describes the design and performance evaluation of a prototype protection scheme suitable for application on a corner of a mesh transmission substation of the type used in the UK on the transmission network.

CubeSats are becoming a game changer in the space industry. Appearing first for univer-sity mission, its popularity is increasing for commercial use and for deep space missionssuch as the on HERA mission that will orbit in 2026 around an asteroid as part of aplanetary defence mission. Standardisation and industrial collaboration is key to a fastdevelopment, assuring the product quality and lower development expenditures.In this study the focus is set elaborating a low cost demonstrator platform to be usedfor developing and testing onboard software on physical hardware: a Hardware-Softwaretesting facility. The purpose of such a platform is to create an interactive and accessibleenvironment for developing on board software. The application chosen to be elaboratedon this platform is a module the subsystem of attitude and orbit control of the satelliteorbiting around asteroid.In order to create this platform the simulation of the asteroid environment of theCubeSat has been made using open source software libraries. During this task the per-formance of open source libraries has been compared to commercial alternatives. In thedevelopment of simulation different orbit perturbations have been studied by modellingthe asteroid as a cube or spheroid and additionally the effect of a third perturbing bodyand radiation pressure.As part of this project two microcontroller have been set up communicating using acommunication bus and communication protocols used for space applications to simulatehow the attitude and orbit control is commanded inside the CubeSat.

As the use of renewable energy and the implementation of smart grids becomemore prevalent in Europe, there will be a need to ensure that the quality ofpower supply is not compromised during the integration of distributedgeneration to the main grid. Europe's electricity networks should be flexible,accessible, reliable and economic. In the UK, National Grid has standardisedits substation protection and control equipment commissioning andreplacement policies, yet issues affecting system long life availability remain,one reason being long outage periods during substation secondary equipmentinstallation, commissioning and maintenance. The present use of directhardwired point to point connections between the primary power system plantequipment and substation secondary system protection and control devicesdoes not allow for easy upgrading or replacement of these substationsecondary devices without an outage of the primary plant or substation. Outage and the consequent availability problems associated with secondaryequipment can be addressed by the open utility communication architecturestandard IEC 61850. A well-designed simple, highly reliable, secure, flexibleand long-life communication IEC 61850-based architecture can help mitigatethe impact of using protection and control IEDs (Intelligent Electronic Devices).Faulty IEDs can be replaced with little or no interruption to the overall operationof the substation. Interoperability is a key feature of the adoption of IEC 61850in substations. IEC 61850-compliant protection and control devices cancommunicate with one another, even if they made from different manufacturers. This thesis has proposed a simple, long life IEC 61850 based communicationarchitecture which is expected to be flexible and robust enough to cope withboth growth and outages. Reliability analyses have been carried out on varioushypothetical applications of the proposed process bus architecture to NationalGrid substation bays. A detailed description of how to determine the optimalprocess bus architecture using the life cycle cost evaluation technique has beenprovided. The design and implementation of a test bed used for evaluating theperformance characteristics of merging units has been presented. The resultsof the tests have been fed back to National Grid and the manufacturers, whomay then use the data to assist with the drafting of a Merging Unit Test BedSpecification, and also to help the manufacturers to make refinements to themerging units in order to make interoperability more readily achievable.

Abstract:
About 5% of HTLV-1 carriers may develop an inflammatory disease of the central nervous system known as HAM/TSP (HTLV-1-associated myelopathy/tropical spastic paraparesis). The HTLV-1 proviral load is an important risk marker for diseases associated with the virus, because symptomatic patients have an average proviral load levels significantly higher than asymptomatic carriers. However, there are asymptomatic carriers (AC) with high levels of proviral load, whereas there are patients with HAM/TSP (HAM) with low levels of proviral load. Therefore, other factors are important in the outcome of the diseases associated with HTLV-1. Here, it were evaluated the levels of tax and sHBZ mRNA expression besides 22 cellular genes involved in innate and adaptive immune responses (TLR4, IL-12B, CD40, CD40L, CD80, CD86, CD28, perforin, granzyme, FasL, Fas , IFN-, TNF-, IL-8, IL-10, RANTES, CCR1, MIP-1, MCP-1, CCR2B, LFA-1 and MMP-9) in individuals AC and HAM classified in low (L) or high (H) proviral load level, using real time RT-PCR. The expression of tax and sHBZ mRNA were analyzed in 21 ACL, 16 ACH, 7 HAML and 19 HAMH, and the cellular genes mRNA expression were analyzed in 14 ACL, 9 ACH, 7 HAML and 9 HAMH, beyond 14 seronegative blood donors (SN). There was no significant difference in sHBZ mRNA expression normalized by the proviral load between the AC and HAM groups. In contrast, tax mRNA load normalized by the proviral load was significantly higher in HAM group than in AC group and in the ACH vs. ACL subgroups, but there was no significant difference between HAMH and HAML subgroups. There was significant positive correlation of tax and sHBZ mRNA expression with the proviral load in AC and HAM groups, but it was stronger in the AC group. The expression level of tax and sHBZ mRNA were also positively and significantly correlated in the AC and HAM groups, however it was stronger in HAM group. The data showed that the AC individuals are a more heterogeneous group than HAM in relation to tax and sHBZ mRNAs expression, and should be carefully evaluated when compared with HAM/TSP patients. In conclusion, it is important evaluate the levels of tax and sHBZ mRNA, and not only the HTLV-1 load proviral, to better estimate the risk of development of HAM/TSP. Regarding the 23 cellular genes analyzed, the ACL subgroup showed the largest number of genes with differential mRNA expression of at least two times comparing to the SN group, with decreased levels of TLR4 (-2.51x) and increased levels of IFN- (2.06x), IL-8 (3.6x) and MIP-1 (2.63x). The ACH and HAML subgroups presented only one gene with reduced mRNA expression in relation to SN group (-2.29x of of MIP-1 and -3.37x of MMP9, respectively). The HAMH subgroup not presented mRNA expression variation of at least twice in relation to SN group for none of the genes analyzed. There was a significant difference in Foxp3 mRNA expression between AC and HAM groups, but not between AC and SN or between HAM and SN. Moreover, Foxp3 mRNA expression was correlated with the proviral load, tax and sHBZ mRNA load only in AC group. Together, what differentiated the ACL subgroup from ACH subgroup was the lowest tax mRNA expression, the lower Foxp3 mRNA expression and the increased IFN- mRNA expression. Comparing HAML vs. HAMH subgroups, HAML showed similar level of tax mRNA load, but increased level of IL-10 and reduced level of Foxp3 mRNA expression. Together, the data of gene expression pointed to the importance of the innate immune response in the protection of HTLV-1 and suggests possible genetic component in the susceptibility to HAM/TSP.
Cerca de 5% das pessoas portadoras do HTLV-1 poderão desenvolver uma doença inflamatória degenerativa do sistema nervoso central conhecida como HAM/TSP (mielopatia associada ao TLV-1/paraparesia espástica tropical). A carga proviral do HTLV-1 é um importante marcador de risco para doenças associadas ao vírus, visto que portadores sintomáticos têm em média níveis de carga proviral significativamente mais elevados que portadores assintomáticos. No entanto, há portadores assintomáticos (AC) com altos níveis de carga proviral, do mesmo modo que pacientes com HAM/TSP (HAM) com baixos níveis de carga proviral. Portanto, outros fatores são importantes no desfecho das doenças associadas ao HTLV-1. Foram avaliados os níveis de expressão do mRNA dos genes virais tax e HBZ e de 23 genes celulares envolvidos na resposta imune inata e adaptativa (TLR4, IL-12B, CD40, CD40L, CD80, CD86, CD28, perforina, granzima, FasL, Fas, IFN-, TNF-, IL-8, IL-10, RANTES, CCR1, MIP-1, MCP-1, CCR2B, LFA-1, MMP-9 e Foxp3) em indivíduos AC e HAM classificados de acordo com o nível da carga proviral em baixa (L) ou alta (H), usando ensaios de RT-PCR em tempo real. Na avaliação da expressão dos mRNA de tax e de sHBZ foram analisadas amostras de 21 ACL, 16 ACH, 7 HAML e 19 HAMH, e na avaliação da expressão dos mRNA dos genes celulares foram analisadas amostras de14 ACL, 9 ACH, 7 HAML e 9 HAMH, além de 14 doadores de sangue soronegativos (SN). Não houve diferença significativa na expressão do mRNA de sHBZ entre os subgrupos AC e HAM quando normalizado pela carga proviral. Em contraste, o mRNA de tax normalizado pela carga proviral foi significativamente maior no grupo HAM do que no grupo AC e em ACH x ACL, mas não foi observada diferença significativa entre os subgrupos HAML e HAMH. Houve correlação positiva e significativa das expressões dos mRNA de ambos os genes virais com a carga proviral nos grupos AC e HAM, mas esta foi mais forte no grupo AC. O nível de expressão dos mRNA de tax e de sHBZ também foram correlacionados positivamente e significativamente nos grupos AC e HAM, porém esta foi mais forte no grupo HAM. Os dados mostraram que os indivíduos AC constituem um grupo mais heterogêneo do que o grupo HAM em relação à expressão dos mRNAs de tax e de sHBZ, e devem ser cuidadosamente avaliados quando comparados com pacientes com HAM/TSP. Em conclusão, é importante avaliar os níveis dos mRNA de tax e de sHBZ, e não apenas da carga proviral, para melhor estimar o risco de desenvolvimento de HAM/TSP. Em relação aos 23 genes celulares analisados, o grupo ACL foi o que mostrou maior número de genes com expressão diferenciada do mRNA de no mínimo duas vezes em relação ao grupo SN, com diminuição do nível de TLR4 (-2.51x) e aumento do nível de IFN- (2.06x), IL-8 (3.6x) e CCL3 (2.63x). Os subgrupos ACH e HAML apresentaram apenas um gene com expressão diminuída do mRNA em relação ao grupo SN (-2,29x de MIP-1 e 3,37x de MMP-9, respectivamente). O grupo HAMH não apresentou variação de pelo menos duas vezes em relação ao grupo SN para nenhum dos genes estudados. Houve diferença significativa na expressão do mRNA de Foxp3 entre AC e HAM, mas não entre AC e SN ou entre HAM e SN. Além do mais, a expressão do mRNA de Foxp3 foi correlacionada com a carga proviral e a expressão dos mRNA de tax e de sHBZ apenas no grupo AC. Em conjunto, o que diferenciou o subgrupo ACL do subgrupo ACH foi a menor expressão do mRNA de tax, a menor expressão do mRNA de Foxp3 e a maior expressão do mRNA de IFN-. Comparando HAML vs. HAMH, HAML mostrou nível similar do mRNA de tax, mas nível aumentado de IL-10 e diminuído de Foxp3. Em conjunto, os dados de expressão dos genes analisados envolvidos na resposta imune apontam para a importância da resposta imune inata na proteção da infecção pelo HTLV-1 e sugere possíveis componentes genéticos na susceptibilidade à HAM/TSP.

Abstract:
IEC 61850 standards are the global standard for communication in substations. It is gaining popularity in power substation automation and will dominate the future substation automation and protection system design. The standards provide new approaches for protection, control and metering function via communication. The secondary circuits in substation are simplified significantly and the massive hardwired cables are replaced by the high-speed process bus which transmit analogue and binary signals with Ethernet messages. However, the conformity of a device with the standards does not necessarily guarantee the interoperability with devices from different manufacturers. The use of devices compatible with IEC 61850 standards presents a challenge to many system integrators, especially due to lack of familiarity with features such as Generic Object Oriented Substation Event (GOOSE), reporting, Sampled Values and the need for system redundancy. To facilitate a smooth implementation, all the features and the data exchanges between devices need to be tested to ensure the system operates correctly. This project was carried out to study the protection performance of secondary schemes with IEC 61850 process bus architecture in substation. The tests were performed including current differential protection and distance protection on a transmission substation of the type used in the UK on the transmission network. The protection schemes were tested under IEC 61850 environment with multi-vendor IEDs like Alstom MiCOM IEDs, NARI IEDs with the OMICRON test set. More tests were carried out to verify the interoperability and the performance of time-critical messages were evaluated under different network architectures. The impact of the background traffic on these two messages was investigated and the response of the IEDs when the Sampled Values packets were lost or overwritten was recorded and provided to utility as a reference. This project also presented a technique to assess the performance of Merging Units from different manufacturers, when operating with Intelligent Electronic Devices (IEDs) performing a distance protection function. The performance of the process bus with parallel redundancy protocol is evaluated using a closed loop approach involving a Real Time Digital Simulator. The results indicate that protection using process bus communication is feasible, and meets grid code requirements when implemented with commercially available products. It was found that process bus operation is robust, even with network traffic conditions far beyond what would be experienced in an active substation.

Abstract:
This thesis assesses the novels of B.S. Johnson and, building on earlier socio-cultural readings, for the first time identifies affect, mood and space as key drivers of Johnson’s work. It suggests fresh interactions with the texts via the artistic practice and philosophical thought of the late twentieth to early twenty-first centuries and reveals what more we can gain from reading Johnson now. Rather than fix this author in a canonized literary past it presents Johnson as a writer with inter-disciplinary appeal and influence. My reading champions the continued significance of Johnson’s work and endeavours to resist teleology; it dips in and out of the seven novels, at times circling back to key passages and episodes that can be assessed in multiple ways. The thesis thus follows Johnson’s practice and is palimpsestic, it is formed from multiple layers. Working with affective energies the chapters unfold to build upon each other but also stand to be read individually or even, in true Johnsonian style, at random. The reading moves in a range of directions exploring different nodes of an organic whole that constitutes the body of Johnson’s literary output. Any thesis must build momentum and therefore this thesis culminates in an extended conclusion which for the first time places Johnson at the vanguard of a spatial turn in the humanities. My final analysis suggests that Johnson’s practice advances the novel towards a model of “creative research” or project work – a reflexive adventure peculiar to the making process. This model is concerned with possibilities and processes rather than final resolution and happily accommodates Johnson’s vision of a chaotic, unknowable world.

This research explores the complexities of young peoples’ personal understanding and experiences of violence and safety in public spaces. The research itself is constructed through establishing the interrelationships between the theories, practices and policies of safety and young people. Working through these links has facilitated an original framework for understanding by accessing data using young people’s own experiences and views. There is a significant body of published research exploring young people as offenders but a real absence, especially in the UK literature, of young people as potential victims of violent crime. In particular children’s own conceptualisations of risk, safety and victimisation are little understood. This research explores young people’s thoughts on exactly this. The empirical research draws upon qualitative data derived from semi-structured interviews with 21 young people aged from 10 to 18 years old taken from a socioeconomically mixed area of London. The findings show that irrespective of age, the young people have constructed a very real understanding of safety and risk. Children, even at a young age have developed a myriad of personal safety strategies that involve awareness of teenagers, locations and individuals who they perceive as guardians. However, these strategies emerge without meaningful reference to police or government policy and are largely embedded in a world far away from those in reach of official community safety agents. This research suggests that there needs to be a move away from portraying young people as ‘folk devils’ who sit at the heart of many ‘moral panics’ towards involving them as significant actors and contributors to social policy making by giving them a voice on the political stage.
