Abstract:
The aim of the study was to prepare an active recombinant human perforin by comparing 5 candidate segments of human perforin. Full-length perforin, MAC1 (28–349 aa), MAC2 (166–369 aa), C-100, and N-60 of human perforin were selected as candidate active segments and designated, respectively, HP1, HP2, HP3, HP4, and HP5. The target genes were amplified by PCR and the products were individually subcloned into pGEM-T. The genes for HP1, HP2, HP3, and HP5 were subcloned into pET-DsbA, whereas pET-41a (+) was used as the expression vector of HP4. The fusion proteins were expressed in Escherichia coli BL21pLysS(DE3) and purified using nickel nitrilotriacetic acid (NTA) agarose affinity chromatography. The hemolysis microassay was used as an activity assay of fusion protein. From this study, we obtained the recombinant plasmids pGEM-T-HP1, -HP2, -HP3, -HP4 and -HP5, consisting of 1600, 960, 600, 300bp, and 180, respectively. From these recombinant plasmids, expression plasmids were successfully constructed and expressed in E. coli BL21pLysS(DE3). The resultant fusion proteins, affinity purified using Ni–NTA, were ~80, 58, 45, 44, and 30 kDa, respectively. The recombinant proteins were assayed for activity on hemolysis. HP2 and HP5 were the only recombinant proteins that were active in hemolysis, and the hemolytic function was concentration dependent. These results demonstrate that active recombinant forms of perforin can be synthesized in a prokaryote model. The recombinant N-60 and MAC1 (28–349 aa) of human perforin have the function of forming pores. Our study provides the experimental basis for further investigation on the application of perforin.

Abstract:
This study deals with the characterization of the phytochemical profiles and antioxidant activities of cv. Caner pomegranate (Punica granatum) juices obtained from three different juice extraction methods including halved pomegranate (HPJ), arils (AJ), and macerated arils (MAJ) extraction for the first time. It was found that the type of the juice extraction process had substantial effects on the volatiles, anthocyanin compositions, and antioxidant activities of the samples. Results showed that the AJ sample (593 mg L−1) had more anthocyanin compounds followed by HPJ (555 mg L−1) and MAJ (408 mg L−1) samples. GC-MS analysis revealed a total of 34 volatile compounds. The highest number of volatiles was found in the MAJ sample (1872 µg L−1); thus, the aril maceration process played an important role in increasing the volatiles as compared to the HPJ (751.8 µg L−1) and AJ (710.7 µg L−1) samples. Sensory analysis showed that the HPJ sample was the most preferred and its general impression was higher as compared to the AJ and MAJ samples. The findings of this study elucidated that the juice extraction technique had a significant influence on the phytochemical profiles, sensory quality, and antioxidant activity of pomegranate juices.

INTRODUÇÃO: O presente trabalho foi realizado no período de março a maio de 2008, com o objetivo de avaliar a eficácia de diferentes técnicas para diagnóstico de Blastocystis hominis em uma amostra da população atendida pelo Laboratório de Biomedicina da Universidade Feevale, Novo Hamburgo/RS. MÉTODOS: Foram estudadas 100 amostras de fezes de crianças e adultos, que foram coletadas e submetidas às técnicas de sedimentação espontânea (HPJ), sedimentação em formol-éter (Ritchie) e de coloração por Gram e May-Grünwald-Giemsa (MGG). RESULTADOS: A presença de Blastocystis hominis foi observada em 40 amostras, quando utilizadas técnicas de coloração (MGG e Gram), enquanto que as técnicas de sedimentação se mostraram menos eficientes (32 amostras positivas para a técnica de Ritchie e 20 amostras positivas para a técnica de HPJ). CONCLUSÕES: Nossos resultados demonstram que o HPJ foi menos eficiente que outros métodos, indicando a necessidade de se incluir na rotina do laboratório técnicas que permitam a identificação deste parasita.

AbstractThis study aimed to compare three qualitative parasitological methods for the diagnosis ofSyphacia murisinfection in 30 Wistar rats (Rattus norvegicus) infected naturally. Methods of spontaneous sedimentation (Hoffman, Pons and Janer, or HPJ) and spontaneous flotation (Willis) for faecal samples and a method of taping (Graham) were performed and compared. The Graham and Willis methods were more sensitive than the HPJ method (P< 0.05). The Graham method was able to detectS. muriseggs in 100% of the samples. Eggs were detected in 83% and 60% of the samples using the Willis and HPJ methods, respectively. Method choice is important for screening for parasites of rats kept under laboratory conditions, as accurate diagnosis helps prevent future environmental contamination and infection. We concluded that the Graham method was the most efficient of those tested in this study for detection ofS. murisinfection in rats. This method is also rapid, inexpensive and practical, and should be implemented as a necessary measure for infection control.

Background:Despite the progress in the discovery of antiretroviral compounds for treating HIV-1 infection by targeting HIV integrase (IN), a promising and well-known drug target against HIV-1, there is a growing need to increase the armamentarium against HIV, for avoiding the drug resistance issue.Objective:To develop novel HIV-1 IN inhibitors, a series of 3-hydroxy-pyrane-4-one (HP) and 3- hydroxy-pyridine-4-one (HPO) derivatives have been rationally designed and synthesized.Methods:To provide a significant characterization of the novel compounds, in-depth computational analysis was performed using a novel HIV-1 IN/DNA binary 3D-model for investigating the binding mode of the newly conceived molecules in complex with IN. The 3D-model was generated using the proto-type foamy virus (PFV) DNA as a structural template, positioning the viral polydesoxyribonucleic chain into the HIV-1 IN homology model. Moreover, a series of in vitro tests were performed including HIV-1 activity inhibition, HIV-1 IN activity inhibition, HIV-1 IN strand transfer activity inhibition and cellular toxicity.Results:Bioassay results indicated that most of HP analogues including HPa, HPb, HPc, HPd, HPe and HPg, showed favorable inhibitory activities against HIV-1-IN in the low micromolar range. Particularly halogenated derivatives (HPb and HPd) offered the best biological activities in terms of reduced toxicity and optimum inhibitory activities against HIV-1 IN and HIV-1 in cell culture.Conclusion:Halogenated derivatives, HPb and HPd, displayed the most promising anti-HIV profile, paving the way to the optimization of the presented scaffolds for developing new effective antiviral agents.

A method for the development of press coated tablet of atenolol for pulsatile delivery was investigated for chronotherapy of hypertension. Effect of viscosity of Hydroxypropylcellulose (HPC) on pulsatile release of atenlol was studied by press coating atenolol core tablet using different viscosity grade HPC and varying coat weight. L-HPC, M-HPC and H-HPC viscosity garde with 75, 100 and 150 mg coat weight were press coated over atenolol core tablets to delay release of atenolol. The batches, HP1-HP9, exhibited an increase in lag time in response to increase in viscosity and coat weight. Two of the batches HP5 and HP7 have shown a burst release of atenolol after 6.5 and 6.0 h lag time respectively, which is suitable for pulsatile drug delivery of atenolol for chronotherapy of hypertension.

As parasitoses, entre as quais se destacam as enteroparasitoses, são mais frequentes na população mais carente. Diante de várias metodologias conhecidas para realização do exame parasitológico de fezes para identificação de cistos, protozoários, ovos e larvas de helmintos dos parasitos intestinais, e do surgimento de novas técnicas, o presente trabalho tem como objetivo avaliar o grau de concordância de um novo teste (Parasitokit®) com base em uma metodologia de reconhecida eficiência para o diagnóstico de helmintos e protozoários, em relação a outro método tradicionalmente utilizado em Análises Clínicas (Hoffman, Pons e Janner - HPJ). A técnica do Parasitokit® tem como vantagem o dinamismo na confecção e a organização no que diz respeito ao menor espaço utilizado para a preparação e leitura do exame; o método de HPJ requer maior volume de vidraria na bancada para a confecção e leitura dos exames. Foram avaliadas quatrocentas amostras no período de doze meses e a análise dos dados foi feita por meio de estatística descritiva. O método do Parasitokit® demonstra fácil manuseio, custo baixo e pequena demanda no espaço físico, apresentando um grau de não conformidade insignificante já que ocorreu concordância em 99,5% em relação ao método de HPJ que utiliza o cálice de sedimentação.

We have isolated probes for DNA polymorphisms across the linkage group I gene cluster in Caenorhabditis elegans, using Tc1-linkage selection. The probes detect strain polymorphism between the wild-type strains of var. Bristol and var. Bergerac. As a result of mapping the sites hP4, hP5, hP6, hP7, hP9, and sP1, more than 1000 kilobases (kb) of cloned cosmid DNA has been positioned on the genetic map. We found there is more DNA per map unit in the center of the gene cluster than expected on the basis of the genomic average. Furthermore, the amount is not constant across the entire region but reaches a peak in the hP9 unc-13 interval. To find the coding regions, we examined DNA cross-homology between two species, Caenorhabditis elegans and Caenorhabditis briggsae. Approximately one-third of the DNA in the hP5 hP9 interval was examined for coding regions and 21 sequences were identified within 318 kb of DNA.Key words: Caenorhabditis elegans, physical map, DNA polymorphisms, genetic mapping, Caenorhabditis briggsae.

Abstract:
St. John's Wort herb extracts often contain undesirable or volitional polysaccharides. As polysaccharides exhibit structure-dependent biological functions in the present study water-soluble polysaccharides were extracted from herb material, fractionated by anion exchange chromatography into four main polysaccharide fractions (denominated as Hp1, Hp2, Hp3 and Hp4) and characterized by HPAEC-PAD, CE, IR and GC-MS. Biological activity on human skin keratinocytes and fibroblasts was assessed by investigation of their effect on proliferation, metabolism, cytotoxicity, apoptosis and differentiation. The underlying mechanisms were investigated in gene expression studies. Polysaccharide fraction Hp1 was mainly composed ofβ-D-glucose. Hp2, Hp3 and Hp4 contained pectic structures and arabinogalactan proteins varying in composition and quantity. Polysaccharides of Hp1 induced the keratinocyte differentiation by inhibiting the gene expression of the epidermal growth factor and insulin receptor. While the collagen secretion of fibroblasts was stimulated by each polysaccharide fraction only Hp1 stimulated the synthesis. The fibroblast proliferation was reduced by Hp1 and increased by Hp4. This effect was related to the influence on genes that referred to oxidative stress, metabolism, transcription processes and extracellular proteins. In conclusion polysaccharides have been shown as biologically active ingredients of aqueous St. John's Wort extracts with a relation between their structural characteristics and function.

Abstract:
From each of a group of 217 adult males selected through enzyme-immunoassay or skin-test (Group A), six stool samples were examined by both the Lutz/Hoffman, Pons & Janer (Lutz/HPJ) and Kato/Katz methods. In addition, one oogram of the rectal mucosa was performed. By these methods, schistosomiasis was detected in 44.7%, 47.5% and 40.1% of the individuals respectively. To evaluate the methods in the assessment of cure, the last 40 patients from group A, treated with a single oral dose of oxamniquine at 15 mg/kg were followed up for six months (Group B). The criteria for parasitological cure included three stool examinations by Kato/Katz and Lutz/HPJ methods, one, three and six months post-treatment and a rectal biopsy between the fourth and sixth months post-treatment. The examinations were negative in 87.5%, 90% and 95% of the patients, respectively. The efficacy of oxamniquine was 82.5% when the three methods were considered together and there was no statistically significant difference between the sensitivity of the individual methods.

Abstract:
The Hypothalamic-Pituitary-Adrenal (HPA) and –Gonadal (HPG) axes have been considered mutually inhibitory; however, emerging evidence supports the proposition that this might not necessarily be the case. This idea is termed “coupling,” in which the HPA-HPG axis are mutually activated or deactivated. Coupling is examined across three data sets with different time-courses of stress exposure, and results demonstrate HPA-HPG co-activation occurs. Furthermore, stress exposure influences this relationship. The discussion shows how it is physiologically possible to have positive coupling or co-activation between these axes according to complex regulatory feedback systems and overlapping neural structures. Findings are interpreted developmentally, because adolescence may be a critical time for this co-activation to occur. Finally, the discussion emphasizes an individual difference perspective because each individual differs in the duration and type of stress they experience, and these exerted individualized effects on HPA-HPG coupling.

Abstract:
China's population is aging rapidly, while the traditional long-term care (LTC) system that heavily relies on families is eroding. In response, China has embarked on a journey of policy experimentation for long-term care insurance (LTCI) since 2016, launching LTCI pilots in 15 pioneer cities. These pilots have a great diversity in participation, eligibility, and provision. This paper estimates the prevalence of LTC needs and analyzes the impact of the LTCI pilots on access. Although substantial progress has been achieved, the overall coverage of LTCI is still relatively small, and a large proportion of vulnerable people needing LTC seem to be left behind because of the strict eligibility criteria. This analysis suggests that future policy experimentation on LTCI reform in China needs to address the following pressing policy issues: expanding the coverage of LTCI; narrowing rural-urban disparities in access; improving access for vulnerable subpopulations; and reducing the heavy reliance on institutional care.

Abstract:
The participation of children and early and late adolescents in resistance training activities, including training for sport, appears to continually be on the rise. The involvement of youth in the sport of weightlifting and the use of weightlifting methods as part of training for sport performance also appears to be growing. The increased acceptance of these activities is evidenced by the number of both lay and scientific articles, specialized magazines and publications devoted to the different types of resistance training. It is also indicated by simple observation of fitness centres and sports training facilities. The use of resistance training appears to be growing in popularity despite poorly supported position statements, anecdotal reports and speculation concerning the potential for injury. Children and adolescents participating in resistance training programs can make noticeable increases in strength and power as well as physiological parameters associated with both health and sport performance. Although the number of children and adolescents (aged 8–18 years) involved in some form of resistance training is increasing and evidenced-based reviews that support these activities have been published, controversy still exists. There are still those who insist that training with free weights, particularly the sport of weightlifting, should not take place until a person is fully developed, even though there is not much objective evidence to support such a position. More recently published scientific data support the notion that properly supervised resistance training programs can improve sport performance, reduce injury potential and enhance healthy facets of children and early and late adolescents.

The aim of this diploma thesis is the design of the differential of a road vehicle driven by a human force, intended for daily driving in the city with a focus on older users. The vehicle for which the differential is designed is strictly a pedal tricycle. The introduction of this thesis briefly outlines the history of pedal tricycles. The second chapter explains some technical terms relating to tricycles and bicycles in general. Further chapters of the research part describe the current structures of tricycles, the principle of a differential function in motor vehicles and design possibilities of the differential for pedal tricycles. The practical part of my work includes the design of a differential for pedal tricycle, the strength analysis of selected parts of the differential, drawings of the differential assembly, production drawings of selected parts and proposal of the torque transfer from pedals to the differential.

Abstract:
Background: The HPV vaccine has been introduced to the public and the medical community since June 2006 for the vaccination of females and since November 2009 for the vaccination of males ages 9-26 years old. The purposes of this research were to explore multiple factors and relationships among Health Belief Model (HBM) constructs (perceived susceptibility, perceived severity, perceived barriers, and perceived benefits) and mediating factors (self-efficacy and cues to action) related to HPV, HPV-associated diseases, and HPV vaccine among Russian college students and to determine which factors were most important when considering who would/would not seek HPV vaccination. Methods: A quantitative, cross-sectional, descriptive, and correlational survey design was used in this study. An existing self-report questionnaire HPV Study Survey was adapted with the permission from the author. One thousand two hundred participants were contacted by Yaroslav-the-Wise Novgorod State University, Veliky Novgorod, Russia registrar's office through e-mails and messaging using two social networks through simple random sampling method using the SQL statement "ORDER BY NEWID" propriety of Microsoft algorithm out of the total student population (9,923 students). The survey was distributed through SurveyMonkeyTM survey software that was activated December 2011 - April 2012. Results: Two hundred seventy students replied to the survey (22.5% response rate) and 117 participants fully completed it out of 270 who responded to the survey (43.33% completion rate). The initial response rate increased 4.4 times using social networks messaging compared to e-mailing invitations. Overall, average knowledge levels were moderate. Participants' behaviors regarding their sexual activity showed that the majority of participants were sexually active. Participants' perceptions (susceptibility, severity, barriers, and benefits) and mediating factors (cues to action and self-efficacy) were moderate. Participants' behavioral intention to get HPV vaccination was moderate. There were statistically significant differences between males and females in perceived susceptibility, perceived barriers, self-efficacy, behavioral intention, and in three behavioral items (having had sexual contact; age of having had sexual contact and sex for the first time). Sixty percent of the variance in behavioral intention getting HPV vaccination could be explained by two HBM constructs (perceived benefits and self-efficacy). Self-efficacy was the number one predictor of behavioral intention (p < 0.001) and perceived benefits were the number two predictor of behavioral intention (p < 0.01).

Abstract:
There has been considerable epidemiological and experimental evidence to support a role for human papillomavirus (HPV) in the development of cervical cancer, however, the precise contribution of HPV remains controversial. Variation in prevalence of HPV types in cervical lesions have been found and there have been reports of a high prevalence of HPV in normal tissue. These variable findings have been attributed to several factors including the different detection and cervical sampling techniques used. In this study a PCR based assay for the detection of the common genital HPV types 6b, 11, 16, 18 and 33 was designed and optimised for both sensitivity and specificity. The assay was applied to 24 normal control cases, 20 cases each of cervical intraepithelial neoplasia (CIN) 1, CIN 2 and CIN 3, 26 cases of squamous carcinoma, 16 cases of adenocarcinoma and 4 cases of mixed adenosquamous carcinoma. HPV DNA was found in 25% of CIN 1 cases, 60% of both CIN 2 and CIN 3 cases and in 80% of the cervical carcinomas. A novel finding was that no HPV DNA was detected in the normal control cases. No HPV 6 or 33 DNA was found and HPV 11 DNA was detected in only 5% of CIN 1 cases. HPV 16 and HPV 18 DNA was seen with increasing frequency though the spectrum of cervical disease. The results of this assay support a role for HPV 16 and 18 in neoplastic progression and emphasise the biological similarity of CIN 2 and CIN 3 and their divergence from CIN 1. The prevalence of HPV 16 and HPV 18 in cervical neoplasms was compared. HPV 16 was found in more squamous carcinomas (58%) than HPV 18 (23%), whereas in adenocarcinomas - which have a poorer prognosis than squamous carcinomas - both HPV types showed an equal prevalence (44%). Thus, in terms of cancer cell differentiation HPV 16 appeared to confer a better prognosis than HPV 18. The frequency of HPV 16 in all cancers and CINs gave a CANCER:CIN prevalence ratio of 1.4 (54% /38%) as compared to one of 3.75 (30% /8%) for HPV 18 - a 2.6 fold difference - indicating either a greater risk of, or more rapid progression to malignancy associated with HPV 18. These findings suggest that HPV 18 is more aggressive than HPV 16.

Tidigare forskning har visat att kunskap om HPV och HPV-vaccin generellt är låg och att vaccinationstäckningen bland unga kvinnor i många länder varit suboptimal. Syfte: Syftet med föreliggande studie var att kartlägga gymnasieelevers kunskap om och inställning till HPV och HPV-vaccin, samt undersöka om det föreligger skillnader mellan elever på praktiska och teoretiska gymnasieprogram. Metod: Studien var en deskriptiv komparativ tvärsnittsstudie med kvantitativ ansats. Orems egenvårdsteori användes som teoretisk referensram. De 230 deltagarna från fyra gymnasieskolor i Uppsala besvarade ett enkätformulär. Resultat: Majoriteten av eleverna hade generellt låg kunskap om HPV och HPV-vaccinet, hade låg tilltro till vaccinet och var osäkra på huruvida de i framtiden ville vaccineras, dock hade elever på teoretiska program bättre kunskap och mer positiv inställning än elever på praktiska program. Flickor hade bättre kunskap och om HPV och HPV-vaccin än pojkar. De flesta hade inte hört talas om vaccin mot HPV, men de allra flesta hade kännedom om vaccin mot livmoderhalscancer. Slutsats: Den låga kunskapen om HPV och HPV-vaccin kan påverka elevernas inställning samt deras intentioner att i framtiden vaccineras. Resultatet indikerar på ett behov av mer information om HPV och HPV-vaccin. Skolsköterskans hälsosamtal med gymnasieelever bör inkludera information och diskussion om HPV och HPV-vaccin anpassad för ungdomar, för att ge dem möjlighet att förstå sambandet mellan kondylom, HPV och livmoderhalscancer, och därmed lättare kunna ta ställning till vaccination.

Combating HPV infection in males is a significant public health issue. In addition to the number of HPV-related cancers that develop each year, Palefsky (2007) reported that "HPV infection of men is of great importance given that sexual transmission is the primary mode of spread to women" (p. 261). In recent years, the development of the HPV vaccine has spurred controversy over whether or not males as well as females should obtain the vaccine against this disease. The purpose of this study was to examine male college students' intention to be HPV vaccinated and their HPV knowledge, attitudes, subjective norms, and perceived behavioral control towards the vaccination. A descriptive, correlational, cross-sectioned research design was employed. Two hundred and eight (208) male college students at a mid-sized public university participated in the study and completed an in-class questionnaire. The results of the descriptive statistics showed that, on average, the sample of 208 male college students had correct responses on only half of the 15 questions regarding knowledge about HPV based on the mean scores. Respondents had positive attitudes towards HPV vaccination, greater sense of control over being HPV vaccinated, and favorable intention to be HPV vaccinated. Subjective norms and perceived behavioral control were significant predictors of male college students' behavioral intention to be HPV vaccinated. Subjective norms and perceived behavioral control had a positive influence on male college students' behavioral intention to be HPV vaccinated. Lastly, male college students' level of HPV knowledge was not significantly correlated to their behavioral intention to be HPV vaccinated

AbstractThe majority of head and neck squamous cell carcinoma (HNSCC) is now classified into two major types: HPV-mediated [HPV(+)] and HPV-negative [HPV(−)]. Within this paradigm, the 8th edition TNM staging system effected modification about what is considered “locally-advanced” HNSCC. Two phase-III trials (RTOG 1016 and De-ESCALATE HPV) disappointingly showed that cetuximab is not as effective in HPV(+) oropharyngeal cancer (OPC) compared to cisplatin with radiotherapy. The recent NRG HN002 de-escalation trial demonstrated the presence of outcome heterogeneity within “low-risk” HPV(+) OPC, some of which continue to benefit from cisplatin combined with reduced-dose radiotherapy. Moreover, distant metastasis (DM) has consolidated its position as the leading cause of death in HPV(+) OPC and strategies to mitigate it are necessary. Unanswered questions and ongoing-emerging concepts exist in both HPV(+) and HPV– diseases. These include understanding the importance of risk under the rubric of extranodal extension (ENE), including degrees of pathological ENE (pENE), and emerging knowledge about radiologic ENE (rENE). Strategies addressing modification of biological phenomena have become paramount and includes hypoxia modification (such as smoking cessation). In addition, contemporary evidence suggests that immunotherapy improves survival in recurrent/metastatic settings, and it is now also being explored in primary disease presentations in combination with (chemo-)radiotherapy. Induction chemotherapy achieves DM reduction in nasopharyngeal cancer but has only been explored minimally in HPV(+) OPC. Evidence that loco-regional management can be de-intensified following a favorable response to induction treatment would provide an attractive option for HPV(+) OPSCC patients while also addressing risk of developing distant disease.

The recent rise in popularity of Hyperparameter Optimization (HPO) for deep learning has highlighted the role that good hyperparameter (HP) space design can play in training strong models. In turn, designing a good HP space is critically dependent on understanding the role of different HPs. This motivates research on HP Importance (HPI), e.g., with the popular method of functional ANOVA (f-ANOVA). However, the original f-ANOVA formulation is inapplicable to the subspaces most relevant to algorithm designers, such as those defined by top performance. To overcome this issue, we derive a novel formulation of f-ANOVA for arbitrary subspaces and propose an algorithm that uses Pearson divergence (PED) to enable a closed-form calculation of HPI. We demonstrate that this new algorithm, dubbed PED-ANOVA, is able to successfully identify important HPs in different subspaces while also being extremely computationally efficient. The thermal durability of thermal barrier coating systems (TBCs) obtained using feedstock powders with different purity and phase content was investigated by thermal shock testing with different cycle times, including the effects on the sintering and phase transformation behaviors. Three kinds of 8 wt.% yttria-stabilized zirconia, namely regular purity (8YSZ), high purity (HP1 and HP2), and no monoclinic phase (NMP), were employed to prepare top coats by atmospheric plasma spraying on a NiCoCrAlY bond coat using a high-velocity oxy-fuel system. Use of 8YSZ, HP1, HP2, and NMP for plasma spraying affected the microstructure and lifetime of the TBC in furnace cyclic testing (FCT) at 1100 °C with 24 hour and 1 hour cycle times. In 24h cycling time, the TBCs deposited using NMP powder showed the highest durability, while that formed from HP showed lifetime performance similar to that obtained with regular purity 8YSZ. The TBC obtained with nMP also exhibited the lowest monoclinic phase transition rate, followed by those obtained using HP and 8YSZ. However, the 1 hour cycle test had the opposite result. Therefore, the coating material should be selected depending on the usage environment.

Abstract The Convolutional Neural Network (CNN) has been used successfully to enhance the First-break (FB) automated arrival picking of seismic data. Determining an optimized FB model is challenging as it needs to consider several hyperparameters (HPs) combinations. Tuning the most important HPs manually is infeasible because of a higher number of HP combinations to be tested. Three state-of-the-art automated hyperparameter optimization (HPO) techniques are applied to a CNN model for robust FB arrival picking classification. A CNN model with 4 convolutional (Conv) layers followed by one fully connected (FC) and one output layer is designed to classify the seismic event as FB or non-FB. To control overfitting, dropout (DO), batch normalization are used after every two Conv layers, in addition to only the DO layer after FC. The number and size of kernels, DO rate, Learning rate (Lr), and several neurons in the FC layer are fine-tuned using random search, Bayesian, and Hyper Band HPO techniques. The findings are experimentally evaluated and compared in terms of four performance metrics with respect to classification performance. The five hyperparameters mentioned above are fine-tuned in 13 search spaces for each of the three HPO techniques. From experimental results, applying random search HPO to CNN yields the best accuracy and F1-score of 96.26%, with the best HP combination of 16, 16, 32, and 64 for numbers of kernels in four Conv layers respectively; 2, 2, 2, 5 for the size of kernels in each Conv layer; 0, 0.45, 0.25 for DO rate in each of DO layers; 240 for numbers of neurons in FC layer; and 0.000675 for Lr. In terms of loss on test data, the above combination of HP gives the lowest test loss of 0.1191 among all techniques, making it a robust model. This model outperforms all the other models in terms of precision (96.27%) and recall. Moreover, all HPO models outperformed the baseline in terms of all metrics. The use of DO after Conv layers and FC layers is highly recommended. Moreover, the use of kernel size relatively smaller (i.e. 2) produces the best classification performance. According to the best HP combination results, there is also no harm to use a relatively higher number of neurons in the FC layer than the Conv layer in FB arrival picking classification. The optimal values of Lr range from 0.0001 to 0.000675 depending on the HPO techniques. The model developed in this study improves the accuracy of the auto-picking of FB seismic data and it is anticipated our model to be used more widely in future studies in the processing of seismic data.

Abstract. This presentation shows that superplasticity is achieved in hard-to-deform materials when they are processed by severe plastic deformation (SPD) through high-pressure sliding (HPS). The HPS process is similar to the high-pressure torsion (HPT) process as the straining is made under high pressure in highly constrained conditions. It is applicable to a sheet form of samples while the HPT process uses disks or rings, and thus it has advantage that the sample size can be increased. In addition, when the HPS process is combined with a feeding process, the SPD-processed area can be further enlarged without increasing the machine capacity, of which process is called the incremental feeding HPS (IF-HPS). The HPS process is applied to a Ni-based superalloy (Inconel 718), a Ti-6Al-7Nb alloy (F1295) and a Mg-6Al-1Zn alloy (AZ61), and superplasticity is well attained in all the hard-to-deform alloys with total elongations more than 400%. It is also demonstrated that cup forming of the Inconel 718 with practical dimensions is realized by application of the IF-HPS process.

In this study we show that bovineadrenal capillary endothelial cells(BACE) contain heparin (HP); this HP has been found associated with the cell surface (i.e; trypsin-removable^and intracellularly. How-ever, experiments with [ sjsodium sulfate labelling, demonstrate that BACE cells donot synthesize HP de novo, but they uptake it from serum. We have studied binding, uptake, and metabolism odifferent molecular weight-HPs: 13 Kd-HP from bovine source, 14 Kd-HP from porcine source, 4.5 Kd, and 2.5-HP fragments. Comparison among different HPs, was carried out by calculating the IC from competition curves for [3HJ- HP. Binding of labelled-HP to BACE cells was specificand saturable. Dextran sulfate and glycosaminoglycans did not compete for binding; only heparan sulfate showed some competition. Binding of different HPs was strictly dependent on their molecular weight; 2.5 Kd- HP was unable to bind to cells, although sulfation degree of this fragment and of unfractionated HP was almost identical. Therefore, we assume that a specific oligosaccharide sequence could be responsible for HP binding to BACE cells; this hypothetical "binding sequence" could then be lost in very low molecular weight-HP fragments. BACE cells are also able to internalize HP, and they release its low molecular weight degradation products into culture medium. Thus we suggest that endothelial cells might represent a site for the metabolism of endogenous and exogenous HP in vivo.
