Abstract:
Several studies have proved the benefits of job malleability, that is, the capacity of an application to adapt its parallelism to a dynamically changing number of allocated processors. The most remarkable advantages of executing malleable jobs as part of a high performance computer workload are the throughput increase and the more efficient utilization of the underlying resources. Malleability has been mostly applied to iterative applications where all the processes execute the same operations over different sets of data and with a balanced per process load. Unfortunately, not all scientific applications adhere to this process-level malleable job structure. There are scientific applications which are either noniterative or present an irregular per process load distribution. Unlike many other reconfiguration tools, the Dynamic Management of Resources Application Programming Interface (DMR API) provides the necessary flexibility to make malleable these out-of-target applications. In this article, we study the particular case of using the DMR API to generate a malleable version of HPG aligner, a distributed-memory noniterative genomic sequencer featuring an irregular communication pattern among processes. Through this first conversion of an out-of-target application to a malleable job, we both illustrate how the DMR API may be used to convert this type of applications into malleable and test the benefits of this conversion in production clusters. Our experimental results reveal an important reduction of the malleable HPG aligner jobs completion time compared to the original HPG aligner version. Furthermore, HPG aligner malleable workloads achieve a greater throughput than their fixed counterparts.

Abstract:
Objective: The main objective of the work was to develop a straightforward, fast and selective liquid chromatography/tandem mass spectrometry (LC-MS/MS) assay for determination of pioglitazone (PG), keto pioglitazone (KPG), and hydroxy pioglitazone (HPG) in human plasma and to validate as per recent guidelines.Methods: Analyte and the internal standard (IS) were extracted from plasma through liquid-liquid extraction and chromatographed on a Xterra RP18, 100×4.6, 5 μ column using methanol: acetonitrile mixture and 10 mM Ammonium formate buffer (70:30, v/v) as the mobile phase at a flow rate of 0.7 mL/min. The API-3200 Q Trap LC-MS/MS instrument in multiple reaction monitoring mode was used for detection. Diphenhydramine was utilized as IS.Results: The linearity was established in the concentration range of 20.15-1007.58 ng/mL for PG, 20.35-1017.58 ng/mL for KPG, and 19.68-491.22 ng/mL for HPG in human plasma. All the validation parameters were well within the acceptance limits.Conclusion: A new simple LC-MS/MS method was developed for the determination of PG, KPG, and HPG in human plasma. This method can be easily applied for the estimation of pharmacokinetic parameters of PG, KPG, and HPG.

Abstract:
Purpose: To investigate the mitigative impact of danshensu on heart tissues in acute myocardial infarction (AMI) rats, and the mechanism of action involved. Methods: Seventy-five male Sprague-Dawley (SD) rats were used in this study. After successful induction of myocardial infarction, the rats were divided into model group (MG), low-dose danshensu group (LDG; 15 mg/kg), middle-dose danshensu group (MDG; 30 mg/kg), and high-dose danshensu group (HDG; 60 mg/kg), with 15 rats per group. Rats in sham group (SG; n = 15) served as control. Serum levels of biochemical indicators and expressions of various proteins in myocardial tissue were determined using Western blotting, and compared amongst the rat groups. Results: Serum cTnI concentrations in MDG and HDG were significantly decreased, relative to the corresponding concentrations in MDG. There were significantly lower serum concentrations of IL-1 in MDG and HDG than in model rats. Rats in HDG had lower serum IL-6 concentration than MG rats, while TNF-α levels were down-regulated in MDG and HDG, relative to MG (p < 0.05). Conclusion: Danshensu protects the heart function of rats with AMI by decreasing inflammationderived cells and mitigating myocardial fibrosis. Thus, it may be useful in the management of AMI in humans but clinical trials are necessary to ascertain this. Abstract:
Objectives: Infertility is defined as the lack of pregnancy after one year of sexual contact without using any contraception. The aim of this study was to investigate the association between endometrial thickness (ET) and endometrial pattern with beta-human chorionic gonadotropin (B-HCG) test in women undergoing the induction of ovulation who referred to the infertility clinic of Ali ibn-e Abitalib hospital in Zahedan. Materials and Methods: This cross-sectional study was conducted at Ali ibn-e Abitaleb hospital in Zahedan (Iran) in 2017. The ET and endometrial pattern were estimated using transvaginal sonography and the pregnancy rate was determined based on B-HCG test. Finally, the data were analyzed by SPSS using the Chi-square test and independent t-test. Results: A total of 200 individuals with a mean age of 29.6 ± 5.6 years were evaluated in this study. The negative or positive B-HCG distribution was significantly different in terms of the endometrial pattern (P=0.001) so that 35.3% of women had B-HCG positive cases in the three-line pattern of ET while in the homogenous pattern, 3.3% had B-HCG positive cases. In addition, the pregnancy rate in women with ET greater than 7 mm was significantly higher compared to those women with ET less than 7 mm thickness (35.1% vs. 12.8%, P<0.001). Conclusions: Overall, the results of this study showed a significant correlation between the endometrial pattern and ET and pregnancy rate based on positive B-HCG test.

We have isolated Dictyostelium cell type proportioning mutants in a screen for cAMP signal transduction mutants. All mutants showed reduced numbers of prespore cells as analyzed with prespore-specific antibodies and prespore- and prestalk-specific reporter gene constructs. Formation of prestalk and prespore cells in the mutant HP1 is initially almost normal; however, many prespore cells redifferentiate as anterior-like cells leading to altered cell type proportions. Synergy experiments of HP1 transformant cells with wild-type Ax2 slugs showed the existence of stable subpopulations of cells in the prestalk and prespore regions of the slug. HP1 cells sort exclusively to the back of the prestalk and prespore zone in slugs. These cells will form the upper and lower cup regions of the culminate. Ax2 transformant cells sort to the tip of the anterior prestalk and prespore zone in HP1 slugs and will form the stalk and spores in the culminate. These experiments show that upper and lower cup derive from cells in different differentiation pathways. The lower cup cell derive from prespore cells, which redifferentiate as anterior-like cells in the back of the prespore zone. The upper cup cells derive from cells from the back of the prestalk zone. Synergy experiments showed that the mutation in HP1 results in an altered sensitivity to proportioning signals. The mutation in HP1 may result in a reduced sensitivity to cAMP, which facilitates the redifferentiation of prespore cells via anterior-like to lower cup cells and inhibits the transition from upper cup to stalk cells.

Abstract:
The aim of this study was to evaluate the effect of Budesonide on the hypothalamic-pituitary-adrenal (HPA) axis in dogs with inflammatory bowel disease. The effect of orally administered Budesonide (Entocort) on the HPA axis was analysed in 21 dogs with inflammatory bowel disease. Biochemical analyses were carried out to evaluate the activity levels of alanine aminotransferase, asparagine aminotransferase, alkaline phosphatase, cortisol and endogenous adrenocorticotropic hormone. Urine samples were collected from each patient before the study and after 30 days of the experiment to determine the composition and the physical and chemical properties of urine sediments. Considerably lower serum concentrations of cortisol and endogenous adrenocorticotropic hormone were observed after 30 days of treatment. A significant increase in alkaline phosphatase levels was noted on Day 30. In the studied dogs, the drop in HPA axis activity was correlated with side effects associated with the administered glucocorticosteroid (polyuria, polydipsia). In conclusion, we have shown that oral administration of Budesonide to dogs diagnosed with inflammatory bowel disease significantly suppressed the activity of the HPA axis.

Abstract:
Objective. The aim of the study was to evaluate the impact of meteorological variables (atmospheric temperature and pressure) on the daily occurrence of acute myocardial infarction (AMI). Material and Methods. The study used the daily values of atmospheric temperature and pressure in 2000–2007. The meteorological data were obtained from the Lithuanian Hydrometeorological Service for Kaunas. The relative risks of event occurrence were computed for 5°C atmospheric temperature and for 10-hPa atmospheric pressure variations by means of the Poisson regression model. Results. The occurrence of AMI and atmospheric temperature showed an inverse linear relationship, while the occurrence of AMI and atmospheric pressure, a positive linear relationship. Among the youngest subjects (25–44 years old), no relationships were detected. Contrary, among the subjects aged 45–64 years and those aged 65 years and older, the occurrence of AMI significantly decreased with higher temperature (P=0.001 and P=0.002, respectively). A decrease in atmospheric temperature by 10ºC reduced the risk of AMI by 8.7% in the age groups of 45–64 and 65 years and older and by 19% in the age group of 25 years and older. Among the first AMI cases, the risk increased by 7.5% in the age group of 45–64-year olds and by 6.4% in the age group of 25–64-year olds. The relationship between atmospheric temperature and pressure, and AMI occurrence was found to be linear but inverse. An increase in atmospheric pressure by 10 hPa resulted in an increase in risk by 4% among the subjects aged 65 years and more and by 3% among the subjects aged 25 years and more. Conclusions. Atmospheric temperature and pressure variations had the greatest effect on middle- aged and aging subjects (starting from 45 years). At younger age, the effect of such factors on the AMI risk was considerably lower.

Abstract:
Introduction/Objective. Fetal growth restriction (FGR) is associated with increased fetal and neonatal mortality and morbidity. The study objective was to investigate the correlation of maternal blood biochemical markers routinely determined in the first and second trimester screening and ultrasound fetal surveillance parameters in the prediction of fetal growth and condition in singleton pregnancies. Methods. In the first trimester we measured serum levels of beta subunit of human chorionic gonadotropin (?HCG) and pregnancy-associated plasma protein A (PAPP-A). In the second trimester we measured values of chorionic gonadotropin (HCG), alpha feto-protein (AFP), unconjugated estriol (E3) and inhibin A, also examined ultrasonographic biometric fetal parameters, amniotic fluid index (AFI) and Doppler resistance indexes. FGR was defined as ultrasonographically determined fetal weight and growth parameters below the 10th percentile for the gestational age. Obtained biochemical and ultrasonographic parameters were correlated. Results. Study included 104 singleton pregnancies. ?HCG in the first trimester negatively correlated with fetal growth in the second and third trimester, and the second trimester AFI. Increased PAPP-A correlated positively with elevated resistance index in medial cerebral artery, lower biophysical profile scores, and intermediate type of non-stress test. Lower values of E3 were associated with FGR. Elevated serum AFP levels were linked to oligoamnion in the third trimester. There was no correlation of inhibin A levels with fetal condition. Conclusion. First and second trimester biochemical markers of pregnancy (?HCG, PAPP-A, HCG, AFP and E3) in combination with ultrasonographic biophysical parameters of fetus have predictive value for fetal growth and development.

The line between HPC and Cloud is getting blurry: Performance is still the main driver in HPC, while cloud storage systems are assumed to offer low latency, high throughput, high availability, and scalability. The Simple Storage Service S3 has emerged as the de facto storage API for object storage in the Cloud. This paper seeks to check if the S3 API is already a viable alternative for HPC access patterns in terms of performance or if further performance advancements are necessary. For this purpose: (a) We extend two common HPC I/O benchmarks—the IO500 and MD-Workbench—to quantify the performance of the S3 API. We perform the analysis on the Mistral supercomputer by launching the enhanced benchmarks against different S3 implementations: on-premises (Swift, MinIO) and in the Cloud (Google, IBM…). We find that these implementations do not yet meet the demanding performance and scalability expectations of HPC workloads. (b) We aim to identify the cause for the performance loss by systematically replacing parts of a popular S3 client library with lightweight replacements of lower stack components. The created S3Embedded library is highly scalable and leverages the shared cluster file systems of HPC infrastructure to accommodate arbitrary S3 client applications. Another introduced library, S3remote, uses TCP/IP for communication instead of HTTP; it provides a single local S3 gateway on each node. By broadening the scope of the IO500, this research enables the community to track the performance growth of S3 and encourage sharing best practices for performance optimization. The analysis also proves that there can be a performance convergence—at the storage level—between Cloud and HPC over time by using a high-performance S3 library like S3Embedded.

Abstract:
Abstract Background Axi-cel is approved for the treatment of relapsed/refractory diffuse large B-cell lymphoma since 2017 and has demonstrated efficacy and durability but with significant side effects, namely cytokine release syndrome (CRS) and neurotoxicity (ICANS). In patients receiving axi-cel for aggressive lymphomas, any grade ICANS occurs at a rate of 50% and grade 3/4 occurs about 35% of the time. The mechanism underlying ICANS remains unclear. The current working hypothesis is that endothelial injury caused by the disease and lymphodepleting chemotherapy leads to breakdown of the blood-brain-barrier. Following activation of axi-cel, cytokines are trafficked into the central nervous system leading to local production of cytokines and the clinical syndrome observed. ICANS is treated with systemic steroids with good CNS penetration. Statins have been shown to stabilize the endothelium and have anti-inflammatory effects. We are currently evaluating the safety and feasibility of administering simvastatin to stabilize the endothelium in addition to dexamethasone delivered intrathecally to prevent the occurrence and decrease the severity of neurotoxicity. Methods: This is a feasibility study combining simvastatin 40 mg orally, daily, starting at least 5 days prior to apheresis in addition to dexamethasone delivered intrathecally on days -1, +6, +13 in relation to axi-cel. A schema of the trial is presented in figure 1. This is registered on Clinicaltrials.gov number (NCT04514029). Results: We have accrued 6/20 planned patients with r/r DLBCL who received axi-cel per institutional guidelines. One passed away prior to apheresis due to disease progression and was non-evaluable. Table 1 shows the patient characteristics. With a median follow up of 123 days (50-245), all patients achieved a complete response and remained alive. Table 2 summarizes the feasibility, safety, and dose-density received out of prescribed. None of the 5 patients had grade 3 or 4 ICANS and only 1 patient had grade 1 neurotoxicity (Table 3). A cytokine analysis and markers of endothelial and neuronal activation is underway. Discussion: In a preliminary analysis, simvastatin and intrathecal dexamethasone appears to be feasible to administer, safe and efficacious in a population of patients receiving axi-cel for relapsed and refractory DLBCL. There appears to be a signal in abrogating neurotoxicity without any effect on the efficacy of axi-cel. This will allow for a safer delivery of axi-cel and improve access to this treatment. Acute exposure to stress is connected to many disorders that promote the toxicity of oxygen radical generators leading to increase in the levels of enzymes and also the activation of the HPA axis. The present study uses a preclinical approach to elucidate some prospective stress-induced behavioral and biochemical effects. The aim of current study was to investigate the relationship between stress and behavioral changes after exposing animals to 2h immobilization stress. We also evaluated the concentration of corticosterone, glucose and endogenous leptin levels in unstressed and stressed animals to explore the possible role of HPA axis in the modulation of stressed induced behavioral deficits. Rats were divided into stressed and unstressed groups. Behavioral activities were monitored in open field activity and light dark transition box after the termination of 2h immobilization period. Animals were then decapitated and plasma samples were collected for catalase, SOD, corticosterone, and glucose estimation. Results showed that exposure to acute stress produced a significant decrease in the activity of rats in the novel environment (open field) and light dark transition box. On the other hand, concomitant elevated level of peripheral markers of oxidative stress such as oxidative enzymes, corticosterone and endogenous leptin were also observed. Therefore, current study seems to suggest an important role of compounds having antioxidant properties for the treatment of stress and related disorders.

ABSTRACT This article aims at studying the characteristics of high-altitude wind at the Ali Observatory on the western Tibetan Plateau, as the high-altitude wind has been put forward as a critical parameter for site evaluation, especially for adaptive optics. We have run a meso-scale numerical weather research and forecasting (WRF) model in three nested domains with different horizontal resolutions, centred at the Ali Observatory; the model results with the highest horizontal resolution of 1 km and temporal resolution of 0.5 h are presented, and also statistical analyseis of vertical wind profiles and 200 hPa wind speed are performed. Moreover, comparisons of wind speeds obtained from model and radiosoundings are presented; as the vertical resolution has been proved to be key to the estimation of optical turbulence with meso-scale models, the vertical resolutions are both set to 50 m for wind profiles, which reveals a high level of agreement and provides a useful tool for site assessment. The results prove the good character of the high-altitude wind over the Ali region, especially in summer half year, with a yearly median 200 hPa wind speed of 33.6 m s−1 in 2016, and also provide proof of the potential advantage of the Ali Observatory for adaptive optics. Furthermore, we certify that meso-scale models can offer dependable estimation of high-altitude wind over the Tibetan Plateau; the wind simulations provided by the WRF model will be of great benefit for adaptive optics, which also provides the vertical distributions of CN2 and τ0 above astronomical observatories.

The forced swim stress test (FST) is widely used for screening pharmacological or non-pharmacological strategies with potential antidepressant activities. Recent data have suggested that repeated FST for five consecutive days (i.e., 5d-RFSS) could be used to generate a robust depressive-like phenotype in mice. However, the face, construct, and predictive validities of 5d-RFSS have been recently challenged. This study took advantage of recent findings showing that mice vulnerability to anxiety is enhanced when animals are stressed during the dark phase, to provide new insight into the relevance of this model. Our results showed a progressive increase in time of immobility in 5d-RFSS mice relative to control non-stressed animals (sham). Three weeks later, we noticed that 5d-RFSS mice injected with the vehicle compound (Veh) still exhibited a high level of immobility in the FST whereas this behavior was reversed by the antidepressant drug amitriptyline (AMI). However, 5d-RFSS/Veh and 5d-RFSS mice/AMI mice showed normal performances in the open field, the novelty suppressed feeding and the tail suspension tests. Despite this lack of generalized behavioral deficits, an impairment of different parameters characterizing the hypothalamic-pituitary-adrenal (HPA) axis reactivity was evidenced in 5d-RFSS mice/Veh but not in 5d-RFSS mice/AMI. Despite anomalies in the HPA axis, the activity of the central serotonergic system remained unaffected in 5d-RFSS mice relative to controls. From our results, it is suggested that learned immobility does not replicate the broad spectrum of depressive symptoms observed in other chronic models of depression such as the unpredictable chronic mild stress (UCMS) model, the chronic social defeat stress (CSDS) model or chronic corticosterone (CORT) exposure but its influence on the HPA axis is remarkable. Further experiments are warranted to makes this model suitable for modelling depression and therefore refine its translational applicability.

Near real time (NRT) remote sensing derived land surface temperature (Ts) data has an utmost importance in various applications of natural hazards and disasters. Space-based instrument MODIS (moderate resolution imaging spectroradiometer) acquired NRT data products of Ts are made available for the users by LANCE (Land, Atmosphere Near real-time Capability) for Earth Observing System (EOS) of NASA (National Aeronautics and Space Administration) free of cost. Such Ts products are swath data with 5 min temporal increments of satellite acquisition, and the average latency is 60-125 min to be available in public domain. The swath data of Ts requires a specialized tool, i.e., HEG (HDF-EOS to GeoTIFF conversion tool) to process and make the data useful for further analysis. However, the file naming convention of the available swath data files in LANCE is not appropriate to download for an area of interest (AOI) to be processed by HEG. In this study, we developed a method/algorithm to overcome such issues in identifying the appropriate swath data files for an AOI that would be able to further processes supported by the HEG. In this case, we used Terra MODIS acquired NRT swath data of Ts, and further applied it to an existing framework of forecasting forest fires (as a case study) for the performance evaluation of our processed Ts. We were successful in selecting appropriate swath data files of Ts for our study area that was further processed by HEG, and finally were able to generate fire danger map in the existing forecasting model. Our proposed method/algorithm could be applied on any swath data product available in LANCE for any location in the world.

Abstract. Ovarian follicular fluid (FF) of a number of species contain regulatory peptides secreted by granulosa cells or by autonomic nerve terminals. In this report we demonstrate the presence of authentic (HPLC-verification) angiotensin II and III as well as of substance P (SP) in human FF obtained from hMG stimulated infertile patients undergoing in vitro fertilization. Angiotensin II/III (AII/III), estradiol (E2) and progesterone concentrations increase with the size of the follicles. SP concentrations did not vary significantly in FF of various sizes. These peptide concentrations in FF are about 10-fold higher than those measured in the serum of the same patients. Attempts to correlate SP, AII/III, E2 and progesterone concentrations in the individual FF with the ability of an oocyte to be fertilized, failed. Neither AII/III, SP, E2 nor progesterone concentrations were different in these subclasses of FF. Follicles of patients punctured under general anesthesia contained significantly more SP than follicles of patients which had lumbar analgesia. AII/III concentrations were the same in FF of both treatment groups. The presence of angiotensin II and III in FF in increasing concentrations depending on the maturity of the follicle and the inability of general anesthesia to affect the AII/III concentrations suggests that this peptide is produced within the ovary.

The representation of aerosol–cloud interaction (ACI) and its impacts in the current climate or weather model remains a challenge, especially for severely polluted regions with high aerosol concentration, which is even more important and worthy of study. Here, ACI is first implemented in the atmospheric chemistry model GRAPES_Meso5.1/CUACE by allowing for real-time aerosol activation in the Thompson cloud microphysics scheme. Two experiments are conducted focusing on a haze pollution case with coexisting high aerosol and stratus cloud over the Jing–Jin–Ji region in China to investigate the impact of ACI on the mesoscale numerical weather prediction (NWP). Study results show that ACI increases cloud droplet number concentration, water mixing ratio, liquid water path (CLWP), and optical thickness (COT), as a result improving the underestimated CLWP and COT (reducing the mean bias by 21 % and 37 %, respectively) over a certain subarea by the model without ACI. A cooling in temperature in the daytime below 950 hPa occurs due to ACI, which can reduce the mean bias of 2 m temperature in the daytime by up to 14 % (∼ 0.6 ∘C) in the subarea with the greatest change in CLWP and COT. The 24 h cumulative precipitation in this subarea corresponding to moderate-rainfall events increases, which can reduce the mean bias by 18 %, depending on the enhanced melting of the snow by more cloud droplets. In other areas or periods with a slight change in CLWP and COT, the impact of ACI on NWP is not significant, suggesting the inhomogeneity of ACI. This study demonstrates the critical role of ACI in the current NWP model over the severely polluted region and the complexity of the ACI effect.

In this infrastructure test, it is done by testing the logic script function using Katalon Studio and testing at the infrastructure level by doing a crash test in the form of failing to deploy and terminating the pod, as well as performing a stress test to test autoscaling in the cluster, the entire test can be run by performing a stress test on the php service pods. by setting the autoscaler parameter Memory Utilization Percentage 125%, 150% and 250%, proving that the HorizontalPodAutoscaler (HPA) as an autoscaler handler can function according to the targets and parameters that have been determined.

Background: The present study aimed to investigate the psychological determinants of adherence to treatment among patients with cardiovascular diseases (CVDs) referring to Imam Ali Hospital in Kermanshah, Iran. Methods: This cross-sectional study was conducted on 227 patients (mean age=58.10, SD = 13.44) with CVDs, randomly selected amongst those admitted to Imam Ali cardiovascular center of Kermanshah in 2018. Data were collected through Meaning in Life Questionnaire (MLQ), the Jefferson Scale of Patient’s Perceptions of Physician Empathy (JSPPPE), the Illness Perception Questionnaire (IPQ), and Adherence to Treatment Questionnaire. The relationships between the criterion and predictor variables were assessed using Pearson correlation coefficient and linear regression (stepwise method) in IBM SPSS Statistics-23. Results: The adherence to treatment was associated with meaning in life (r=0.367), patients’ perceptions of physician empathy (r=0.218), and illness perception (r=-0.238), at the 0.01 level. Meaning in life, patient’s perceptions of physician empathy, and illness perception explained 18.6% of the variance in adherence to treatment. Meaning in life (beta=0.367 and P≤0.001) was the most influential predictor of adherence to treatment. Additionally, there was a significant difference in the score of adherence to treatment by gender (23.46±4.42 for female vs. 24.77±3.53 for male, P = 0.030). Conclusion: The patients’ perceptions of physician empathy, meaning in life, and illness perception were important factors to predict adherence to treatment in patients with CVDs. Gender was a significant predictor of the adherence to treatment.

Retrieved from the Moderate Resolution Imaging Spectroradiometer (MODIS) on-board the Aqua satellite, 12 years (2003–2014) of aerosol and cloud properties were used to statistically quantify aerosol–cloud interaction (ACI) over the Baltic Sea region, including the relatively clean Fennoscandia and the more polluted central–eastern Europe. These areas allowed us to study the effects of different aerosol types and concentrations on macro- and microphysical properties of clouds: cloud effective radius (CER), cloud fraction (CF), cloud optical thickness (COT), cloud liquid water path (LWP) and cloud-top height (CTH). Aerosol properties used are aerosol optical depth (AOD), Ångström exponent (AE) and aerosol index (AI). The study was limited to low-level water clouds in the summer. The vertical distributions of the relationships between cloud properties and aerosols show an effect of aerosols on low-level water clouds. CF, COT, LWP and CTH tend to increase with aerosol loading, indicating changes in the cloud structure, while the effective radius of cloud droplets decreases. The ACI is larger at relatively low cloud-top levels, between 900 and 700 hPa. Most of the studied cloud variables were unaffected by the lower-tropospheric stability (LTS), except for the cloud fraction. The spatial distribution of aerosol and cloud parameters and ACI, here defined as the change in CER as a function of aerosol concentration for a fixed LWP, shows positive and statistically significant ACI over the Baltic Sea and Fennoscandia, with the former having the largest values. Small negative ACI values are observed in central–eastern Europe, suggesting that large aerosol concentrations saturate the ACI.

Abstract BACKGROUND Sleep disturbance is a common symptom in patients with high grade glioma (HGG). Existing self-reported and uni-dimensional data from questionnaires are of limited value. The observational phase 2 trial, BrainWear (ISRCTN 34351424) provides the first objective analysis of sleep in HGG patients. MATERIAL AND METHODS Patients with HGG were asked to wear an AX3 Axivity tri-axial accelerometer throughout treatment. The study employed a wear-as-long-as-possible approach to accelerometry data collection, and we used age-sex matched controls from the UK Biobank as comparators. Baseline was established as a 7 day period of wear prior to surgery or at least 7 days post-surgery. The dataset for this analysis consists of 21 patients with data at baseline and 15 patients during chemo-radiation. Only 16 of the 21 HGG patients at baseline were included for initial comparisons with healthy controls due to age limitations of the UK Biobank cohort for matching. Raw accelerometer data was processed using the GGIR package, with non-imputation of missing data, exclusion of days with &lt;16 hours of wear time and removal of algorithm-identified problematic data. Mann-Whitney U-tests and unpaired T-tests were used to compare 7 sleep-related features between HGG patients and healthy controls at baseline, with choice of statistical test based on shapiro-wilk derived normality. Secondly, to assess changes in sleep in HGG patients across treatment period, K-means clustering of 5 sleep parameters, available longitudinally, was conducted to explore sleep behaviours at baseline (n = 21) and during chemo-radiation. RESULTS HGG patients (n = 16) exhibited greater daytime inactivity than healthy controls (n = 32) (p &lt; 0.0001, 2.2 vs 0.5 hrs) and more variation in their 24 hour activity rhythm from day to day (p &lt; 0.0001, 0.12 vs 0.18). We identified 5 sleep features which allowed us to cluster patients’ sleep behaviour, and most (62.5%) of HGG patients have a poor sleep profile. This sleep profile was characterised by an average of 5.4 hours of night-time sleep, 2.1 hours of daytime inactivity and disturbed sleep quality. However, evaluation of HGG patient sleep cluster designation at baseline and during chemoradiation, showed HGG patients with data at both timepoints (n = 9) demonstrate stability or improvement in sleep profile. CONCLUSION Patients with HGG have objective evidence of poor sleep compared to healthy matched controls. Further work will explore changes in sleep over time.

U Bosni i Hercegovini nedostaje istraživanja na području valorizacije georaznolikosti, što je rezultiralo zanemarivanjem njezinih znanstvenih vrijednosti, ali i nedovoljnim razvojem geoturizma. Cilj je ovoga rada valorizirati odabrane geomorfolokalitete Parka prirode Blidinje s obzirom na njihovu geoturističku vrijednost. Primijenjeni metodološki koncept nastao je kao rezultat analize postojećih metodoloških koncepata valorizacije georaznolikosti, pri čemu su objedinjeni kriteriji za procjenu osnovne, dodane, zaštitne i turističke vrijednosti. Jedan od ciljeva rada jest i doprinos boljem razumijevanju i primjeni metodoloških koncepata turističke valorizacije, posebice u okviru zaštićenih prirodnih područja. Tijekom istraživanja izdvojena su 44 lokaliteta, a na osnovi rezultata preliminarne procjene kreirana je lista s ukupno 23 lokaliteta koji su valorizirani, od čega se njih osam izdvaja većom ocjenom od prosječne. Rezultati ovoga istraživanja upućuju na potencijal za razvoj ponude geoturizma PP Blidinje, čiji bi temelj trebali biti najbolje ocijenjeni geomorfolokaliteti.

Therapies to limit the life-threatening vascular leak observed in patients with acute lung injury (ALI) are currently lacking. We explored the effect of simvastatin, a 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase inhibitor that mediates endothelial cell barrier protection in vitro, in a murine inflammatory model of ALI. C57BL/6J mice were treated with simvastatin (5 or 20 mg/kg body wt via intraperitoneal injection) 24 h before and again concomitantly with intratracheally administered LPS (2 μg/g body wt). Inflammatory indexes [bronchoalveolar lavage (BAL) myeloperoxidase activity and total neutrophil counts assessed at 24 h with histological confirmation] were markedly increased after LPS alone but significantly reduced in mice that also received simvastatin (20 mg/kg; ∼35–60% reduction). Simvastatin also decreased BAL albumin (∼50% reduction) and Evans blue albumin dye extravasation into lung tissue (100%) consistent with barrier protection. Finally, the sustained nature of simvastatin-mediated lung protection was assessed by analysis of simvastatin-induced gene expression (Affymetrix platform). LPS-mediated lung gene expression was significantly modulated by simvastatin within a number of gene ontologies (e.g., inflammation and immune response, NF-κB regulation) and with respect to individual genes implicated in the development or severity of ALI (e.g., IL-6, Toll-like receptor 4). Together, these findings confirm significant protection by simvastatin on LPS-induced lung vascular leak and inflammation and implicate a potential role for statins in the management of ALI.

Akut koszorúér-események után a gyógyszeres szekunder prevenció alapvető eszköze a hatékony koleszterincsökkentés, amelynek alapkészítményei a HMG-CoA-reduktáz-inhibitorok (statinok). Bár a gyógyszercsalád jelentősen képes mérsékelni az éresemények ismétlődését, a betegek kitartása a statinterápiához közismerten gyenge, ami részben indokolhatja a célértékek el nem érését, a magas vaszkuláris morbiditást és mortalitást az ismerten érbetegek között. Célok: Vizsgálatunkban arra a kérdésre keresünk választ, hogy különböző akut koszorúér-esemény után rehabilitációs kezelésben részesült betegek statinszedési szokásai hogyan alakulnak az önbevallás, illetve a receptkiváltások alapján. Betegek és módszer: Prospektív, multicentrikus vizsgálatunkban 7 centrumban bevont 445 beteg (kor 61,3±8,7 év, 70,1% férfi) adatait elemeztük, akinél egyaránt rendelkezésre álltak az önbevallásos és receptkiváltáson alapuló egyéves követési adatok. Közülük 289-en szívinfarktus (AMI-csoport), 53-an perkután revaszkularizáció (PCI-csoport), 103-an szívizom-revaszkularizációs műtét (ACBG-csoport) után kerültek bevonásra. Egy évvel később a betegek kérdőívben, vagy telefoninterjú során nyilatkoztak gyógyszeres kezelésükről. Rendelkezésünkre álltak a receptkiváltás adatai, amely alapján meghatároztuk a gyógyszeradherenciát és -perzisztenciát. Eredmények: Kiinduláskor szignifikánsan kevesebb beteg részesült statinkezelésben az ACBG-műtött csoportban (egész csoport: 95,1%; AMI: 96,9%; PCI: 96,2%; ACBG: 89,3%, a csoportok között jelentős az eltérés, p<0,01). Ugyanakkor itt volt legmagasabb az LDL-koleszterin szintje (egész csoport: 1,98±0,79; AMI: 1,97±0,81; PCI: 1,74±0,68 ACBG: 2,12±0,75 mmol/l, a csoportok között jelentős az eltérés, p<0,01). Egy évvel később az egész populációban a betegek 83,6%-a volt statinkezelésen önbevallás alapján (AMI: 85,0%, PCI: 84,3%, ACBG: 79,4%), nem találtunk szignifikáns különbséget a csoportok között. A receptkiváltáson alapuló adatok lényegesen rosszabb képet mutattak (a jó adherenciával bíró betegek aránya az egész csoportban: 53,8%, AMI: 57,1%, PCI: 51,9%, ACBG: 44,8%, különbség nem szignifikáns). Többváltozós lineáris regressziós modellben szintén egyik változó sem volt a statinadherencia önálló prognosztikus tényezője. A betegek perzisztenciája is az AMI-csoportban volt a legjobb. Következtetések: Bár akut koszorúér-betegség utáni rehabilitációban részt vevő betegek nagy arányban részesülnek statinkezelésben, ACBG-műtét utáni betegeknél már kiinduláskor is kisebb arányban van jelen statinkezelés. Egy évvel az esemény után nagyon alacsony a jó adherenciájú betegek aránya.

The purpose of this study was to determine the numbers and species of enterococci encountered on pork carcasses during different stages in the slaughter process as well as on pork products. Three hog slaughtering plants were surveyed, each 3 times at four processing points. Each hog was swabbed at two sites on the carcass. Specimens were plated on two different enterococcal media, KF streptococcal agar and fluorescent gentamycin-thallous-carbonate agar. Retail and spoiled pork sausage products also were examined. Isolates were speciated by using the API Rapid Strep and Baxter MicroScan Pos ID panels. Contamination levels varied between plants as the carcasses progressed down the processing line; the highest counts were obtained directly before packaging in plants A and C. The highest count for plant B occurred at the first stage of sampling. More Enterococcus faecalis than Enterococcus faecium were isolated from the pork carcasses. Pork sausage results also are presented. Enterococci are useful as an indicator of pork sanitation and to detect critical control points during processing. In some instances, high levels of enterococci are associated with spoilage of pork sausage.

Penelitian ini menggunakanrancangan acak lengkap dengan 5 kelompok perlakuan masing-masing sebanyak 3 ulangan. Hewan coba berupa tikus galur Wistar sebanyak 15 ekor dibagi menjadi 5 kelompok perlakuan, yaitu kelompok yang diberikan ekstrak air daun sirsak dengan dosis 200 mg/kgBB, 400mg/kgBB, kontrol positif, kontrol negatif, dan kontrol normal. Rerata bobot badan dan kadar kolesterol kemudian dianalisis menggunakan Sapphiro Wilk test, ANOVA dan Kruskall Wallis. Hasil penelitian menunjukkan bahwa semua kelompok yang diberikan ekstrak daun sirsak memiliki efek menghambat peningkatan bobot badan jika dibandingkan dengan kontrol normal, sedangkan untuk kadar kolesterol darah didapatkan bahwa seluruh kelompok perlakuan menunjukkan kadar kolesterol darah yang sama dengan kelompok yang diberikan simvastatin. Kesimpulan penelitian ini adalah bahwa ekstrak air daun sirsak memiliki efek mengendalikan bobot badan dan kolesterol darah. Efek terhadap kolesterol darah serupa dengan simvastatin, karena ekstrak air daun sirsak mengandung fl avonoid yang mempunyai efek menghambat enzim HMG CoA reduktase, serupa dengan mekanisme kerja simvastatin dalam menurunkan kadar kolesterol darah.

Exploring the relationship between environmental air quality (EAQ) and climatic conditions on a large scale can help better understand the main distribution characteristics and the mechanisms of EAQ in China, which is significant for the implementation of policies of joint prevention and control of regional air pollution. In this study, we used the concentrations of six conventional air pollutants, i.e., carbon monoxide (CO), sulfur dioxide (SO2), nitrogen dioxide (NO2), fine particulate matter (PM2.5), coarse particulate matter (PM10), and ozone (O3), derived from about 1300 monitoring sites in eastern China (EC) from January 2015 to December 2018. Exploiting the grading concentration limit (GB3095-2012) of various pollutants in China, we also calculated the monthly average air quality index (AQI) in EC. The results show that, generally, the EAQ has improved in all seasons in EC from 2015 to 2018. In particular, the concentrations of conventional air pollutants, such as CO, SO2, and NO2, have been decreasing year by year. However, the concentrations of particulate matter, such as PM2.5 and PM10, have changed little, and the O3 concentration increased from 2015 to 2018. Empirical mode decomposition (EOF) was used to analyze the major patterns of AQI in EC. The first mode (EOF1) was characterized by a uniform structure in AQI over EC. These phenomena are due to the precipitation variability associated with the East Asian summer monsoon (EASM), referred to as the “summer–winter” pattern. The second EOF mode (EOF2) showed that the AQI over EC is a north–south dipole pattern, which is bound by the Qinling Mountains and Huaihe River (about 35° N). The EOF2 is mainly caused by seasonal variations of the mixed concentration of PM2.5 and O3. Associated with EOF2, the Mongolia–Siberian High influences the AQI variation over northern EC by dominating the low-level winds (10 m and 850 hPa) in autumn and winter, and precipitation affects the AQI variation over southern EC in spring and summer.

Abstract Acute myocardial infarction (AMI) induced by ischemia hypoxia severely threatens human life. Cell apoptosis of neurocytes was identified to mediate the pathogenesis, while the potential mechanism was still unclear. Sprague Dawley (SD) rats were used to establish the AMI rat model. Real-time polymerase chain reaction (PCR) and Western blot were performed to detect gene expression in mRNA and protein levels, respectively. A TUNEL assay was carried out to determine cell apoptosis. The relationship between SRY-related HMG-box (SOX7) and miR-128 was verified using luciferase reporter assay. The expression of SOX7 was decreased, while miR-128 was increased in AMI rats and ischemia hypoxia (IH) induced H9c2 cells. Hypoxia induction significantly promoted the expression of interleukin (IL)-33 and soluble ST2 (sST2), and also promoted cell apoptosis. MiR-128 targets SOX7 to regulate its expression. Down-regulated miR-128 reversed the effects of IH on expression of SOX7, sST2 and cell apoptosis, while down-regulated sST2 abolished the effects of miR-128 inhibitor. In addition, overexpressed IL-33 abolished the effects of miR-128 inhibitor that induced by IH on the expression of SOX7 and cell apoptosis. In vivo experiments validated the expression of miR-128 on cell apoptosis. The present study indicated that miR-128 modulated cell apoptosis by targeting SOX7, which was mediated by IL-33/sST2 signaling pathway.

This experiment was performed to evaluate heterosis and combining ability of morphological traits in maize (Zea mays L.). The experiment was arranged in randomized complete block design (RCBD) with three replicates. Twenty maize hybrids were obtained by 5×5 complete diallel crosses in first year and were evaluated for plant attributes like plant height, ear height, ear leaf area, flag leaf area, days to 50% silking, days to 50% pollen shed, anthesis-silking interval (ASI), no. of leaves per plant, no. of branches per tassel, no. of kernel rows per ear and kernels per row, 1000-kernel weight, shelling percentage, grain plus biological yield per plant and harvest index in second year. Among the crosses NCEV-4 × NCEV-3 (462.70g) showed maximum 1000-kernel weight, followed by NCEV-3 × EV-70040 (451.33g), while highest grain yield per plant was recorded for NCEV-4 × NCEV-3 (152.97g), followed by NCEV-3 × EV-70040 (145.10g). Maximum and highly significant heterosis and heterobeltiosis was found in cross NCEV-1530-11 × HNG (70.49 and 67.54), followed by HNG × EV-70040 (65.57 and 56.39) and HNG × NCEV-1530-11 (64.14 and 61.3), rest of the crosses also showed positive and highly significant heterosis and heterobeltiosis for grain yield per plant. Among the parents NCEV-3 has best GCA for grain yield (4.30) while cross NCEV-3 × NCEV-4 has best SCA for grain yield per plant (18.10) and could be used in hybridization for yield improvement.

Ionic liquids (ILs) are a potential solution to the general problem of low solubility, polymorphism and low bioavailability of active pharmaceutical ingredients (APIs). In this work, we report on the synthesis of three pharmaceutically active ILs (API-ILs) based on ibuprofen, one of the most commonly available over-the-counter nonsteroidal anti-inflammatory drugs (NSAIDs), with imidazolium cations ([C2C1Im][Ibu] and [C2(OH)C1Im][Ibu]) and a cholinium cation ([N1112(OH)][Ibu]). An upgrade to the aqueous solubility (water and biological simulated fluids) for the ibuprofen-based ILs relative to the ibuprofen’s neutral and salt form (sodium ibuprofen) was verified. The cytotoxic profiles of the synthesized API-ILs were characterized using two human cells lines, Caco-2 colon carcinoma cells and HepG-2 hepatocellular carcinoma cells, up to ibuprofen’s maximum plasma concentration (Cmax) without impairing their cytotoxicity response. Additionally, the EC50 in the Caco-2 cell line revealed similar results for both parent APIs and API-ILs. The biocompatibility of the ibuprofen-based ILs was also evaluated through a hemolytic activity assay, and the results showed that all the ILs were hemocompatible at concentrations higher than the ibuprofen Cmax. Moreover, the anti-inflammatory properties of the API-ILs were assessed through the inhibition of bovine serum albumin (BSA) denaturation and inhibition of cyclooxygenases (COX-1 and COX-2). The results showed that [C2C1Im][Ibu], [C2(OH)C1Im][Ibu] and [N1112(OH)][Ibu] maintained their anti-inflammatory response to ibuprofen, with improved selectivity towards COX-2, allowing the development of safer NSAIDs and the recognition of new avenues for selective COX-2 inhibitors in cancer chemotherapy and neurological diseases such as Alzheimer’s and Parkinson’s.

Abstract A physical retrieval algorithm has been developed for deriving the legacy atmospheric profile (LAP) product from infrared radiances of the Advanced Baseline Imager (ABI) on board the next-generation Geostationary Operational Environmental Satellite (GOES-R) series. In this study, the GOES-R ABI LAP retrieval algorithm is applied to the GOES-13 sounder radiance measurements (termed the GOES-13 LAP retrieval algorithm in this study) for its validation as well as for potential transition of the GOES-13 LAP retrieval algorithm for the operational processing of GOES sounder data. The GOES-13 LAP retrievals are compared with five different truth measurements: radiosonde observation (raob) and microwave radiometer–measured total precipitable water (TPW) at the Atmospheric Radiation Measurement Cloud and Radiation Testbed site, conventional raob, TPW measurements from the global positioning system–integrated precipitable water NOAA network, and TPW measurements from the Advanced Microwave Scanning Radiometer for Earth Observing System (AMSR-E). The results show that with the GOES-R ABI LAP retrieval algorithm, the GOES-13 sounder provides better water vapor profiles than the National Centers for Environmental Prediction (NCEP) Global Forecast System (GFS) forecast fields at the levels between 300 and 700 hPa. The root-mean-square error (RMSE) and standard deviation (STD) of the GOES-13 sounder TPW are consistently reduced from those of the GFS forecast no matter which measurements are used as the truth. These substantial improvements indicate that the GOES-R ABI LAP retrieval algorithm is well prepared to provide continuity of quality to some of the current GOES sounder products, and the algorithm can be transferred to process the current GOES sounder measurements for operational product generation.

This paper presents the design and implementation of several fundamental dense linear algebra (DLA) algorithms for multicore with Intel Xeon Phi coprocessors. In particular, we consider algorithms for solving linear systems. Further, we give an overview of the MAGMA MIC library, an open source, high performance library, that incorporates the developments presented here and, more broadly, provides the DLA functionality equivalent to that of the popular LAPACK library while targeting heterogeneous architectures that feature a mix of multicore CPUs and coprocessors. The LAPACK-compliance simplifies the use of the MAGMA MIC library in applications, while providing them with portably performant DLA. High performance is obtained through the use of the high-performance BLAS, hardware-specific tuning, and a hybridization methodology whereby we split the algorithm into computational tasks of various granularities. Execution of those tasks is properly scheduled over the heterogeneous hardware by minimizing data movements and mapping algorithmic requirements to the architectural strengths of the various heterogeneous hardware components. Our methodology and programming techniques are incorporated into the MAGMA MIC API, which abstracts the application developer from the specifics of the Xeon Phi architecture and is therefore applicable to algorithms beyond the scope of DLA.

Abstract:
CEL NAUKOWY: Celem artykułu jest prezentacja specyfiki myśli politycznej Renesansu w Italii i jej oddziaływania na myśl polityczną w Hiszpanii XVI stulecia. PROBLEM I METODY BADAWCZE: Artykuł prezentuje dwie zasadnicze zdaniem autora komponenty myśli politycznej Renesansu włoskiego: idee racji stanu oraz równowagi sił. Ukazuje ich oddziaływanie na umysły zwłaszcza florenckich humanistów Quattrocento i Cinquecento, a także analizuje wpływ na dążenia do hegemonii hiszpańskich Habsburgów. Metodą w nim zastosowaną jest historia idei. PROCES WYWODU: Artykuł składa się z dwóch części – pierwsza poświęcona jest myśli politycznej Renesansu włoskiego, druga – analizie jej wpływu na strategie polityczne przyjęte w Hiszpanii w okresie jej hegemonii w Europie jako głównej potęgi broniącej katolickiej racji stanu. WYNIKI ANALIZY NAUKOWEJ: Zasadniczą konkluzją płynącą z analizy jest zwrócenie uwagi na istotny dla Renesansu koncept racji stanu, który krystalizuje się w Italii w epoce przedtrydenckiej, ogniskując się głównie na polityce ograniczonej do strategii politycznych państw Półwyspu Apenińskiego. W części drugiej pojawia się teza o próbach przeszczepienia tego konceptu na Półwysep Iberyjski, gdzie jednak – w obliczu nowej sytuacji geopolitycznej po wojnach włoskich oraz odrodzenia się tomizmu – nie znajduje on ani zrozumienia, ani zastosowania. WNIOSKI, INNOWACJE, REKOMENDACJE: W zamyśle artykułu leży podkreślenie ważnego momentu epoki późnego Renesansu, w którym – wskutek zmiany sytuacji geopolitycznej w Europie i ewangelizacji Nowego Świata – włoskie idee racji stanu oraz równowagi sił muszą ustąpić miejsca racjom wynikającym z konieczności obrony świata katolickiego przed protestantyzmem.
