Full textRecent studies have revealed that viruses represent a large part of the biodiversity in natural environments suggesting that they participate to the diversity within organisms. Viruses infecting prokaryotes (Bacteria and Archaea) constitute the largest & ail viral groups. However, our knowledge of viruses of Archaea is rather limited with less than 60 archaeal viruses well described to date. Viruses of Archaea are classified in 8 new viral familles. To date, only one virus like-particle was isolated from a hyperthermophilic marine euryarchaeote: PAV1 isolated from Pyrococcus abyssi. During my PhD work, I have screened and characterized new viruses from the Thermococcales order. We found a new virus in a strain of Thermococcus sp, isolated from a hydrothermal chimney sample collected from the East pacific Rise at 2700 m depth. The first study led to the characterization of Thermococcus prieurii sp. Nov which is hyperthermophile, strictly anaerobe, and chemoorganotroph. The virus isolated from Thermococcus prieurii, named TPV1, is lemon-shaped (140 nm x 80 nm) and resembles to members of the Fuselloviridae family. The infectious character of TPV1 was proved by using an original method derived from the spot tests, and its host range was established. TPVI particles contain a double-stranded circular DNA of 21. 5 kb which is present in a free form in the host cytoplasm, at approximately 20 copies per chromosome. This genome was totally sequenced and 26 ORFs were identified. 12 ORFs were annotated. This virus s not yet classified and should be attached to a novel viral family

Full textIn the Splendidus clade, Vibrio tasmaniensis and Vibrio crassostreae are two populations of virulent vibrio for oysters that are associated to "juvenile oyster mortality syndrome". Here we were interested in the diversity of interaction mechanisms between the vibrios and their host, the oyster Crassostrea gigas. First, we investigated the pathogenesis process of the strain V. tasmaniensis LGP32 and showed that it exerts a cytotoxic activity against oyster immune cells, the hemocytes, which depend on its entry into the cells through phagocytosis. Transcriptomic analysis of LGP32 response during intracellular stage revealed a crucial role for antioxydant systems and copper efflux in intraphagosomal survival of the bacteria. From a functional point of view, we showed that this virulence mechanisms of LPG32 play a major role in pathogenesis in vivo. Second, we realized a comparative study of the interaction mechanisms between representative strains of the two populations V. tasmaniensis and V. crassostreae with the oyster. Virulent strains from both populations were cytotoxic for hemocytes but this cytotoxicity was independant of phagocytosis in the case of V. crassostreae, in contrary to V. tasmaniensis. Transcriptomic analysis of the oyster responses during infection showed that virulent strains of both populations repressed the expression of genes involved in antibacterial responses. However, some pecific responses were also identified for each virulent strain, highlighting some diversity of interactions. In vivo, virulent strains were able to colonize oyster tissues, in contrary to non-virulent strains, which were controlled by hemocytes. Our work show, although a certain degree of diversity and specificity exist in the interactions between different vibrios of the Splendidus clade and oysters, both virulent populations are cytotoxic for immune cells, and this process is essential for their infectious success. Thus, the capacity to overcome the hemocyte defenses is a conserved phenotype between distinct virulent populations of vibrios from the Splendidus clade. Hence, it would be of particular interest to determine the evolutionary processes that drove the emergence of common virulence traits in distinct populations of pathogens

Full textVibrio strains belonging to the Splendidus clade have been repeatedly found in juvenile diseased oysters affected by summer mortalities. V. tasmaniensis LGP32 is an intracellular pathogen of oyster hemocytes which has been reported to alter the oxidative burst and inhibit phagosome maturation.We here focus on the interactions between phagocytes and V. tasmaniensis LGP32, at molecular, cellular and environmental scales. In the first part of this work, we uncover anunknown antimicrobial mechanism of C. gigas hemocytes: the formation of DNA extracellular traps (ETs). These ETs are associated with antimicrobial histones and are able to entrap bacteria. As in vertebrates, ETs formation depends on reactive oxygen species production. In addition, the presence of ETs was confirmed in vivo and has been associated with antimicrobial histones accumulation in tissues, in response to injury or infection. In the second part of this work, we studied the interactions between V. tasmaniensis LGP32 and heterotrophic protists found in the oyster’s environment, such as amoebae and ciliates, which feed on microorganisms by phagocytosis. An important result of this workwas that V. tasmaniensis LGP32 resists to phagocytosis by environmental heterotrophic protists, as well as to oyster hemocytes. To our knowledge, this is the first mechanical description of an interaction between marine amoebae and marine pathogenic bacteria. As the amoebae were isolated from the direct environment of oysters, we can presume that the selective pressure exerted by environmental phagocytes could select for virulence and/or phagocytosis resistance traits in marine bacteria as in the case of V. tasmaniensis LGP32

Full textBivalve populations undergo regular epidemics that weaken or decimate exploited stocks and thus limit aquaculture. These diseases are caused mainly by viruses, bacteria or parasites, and occur primarily during spring and summer. This period of the year also provides favorable conditions for toxic dinoflagellate blooms, including species of the genus Alexandrium. Thus, the risk of Alexandrium sp. blooms and infectious diseases co-occurring in bivalves is high. However, these micro-algae synthesize and excrete toxins and cytotoxic compounds responsible for physiological changes in bivalves and could lead to an immuno-compromised status.The objective of this thesis is to evaluate the combined effects on bivalve physiology of exposure to the toxic dinoflagellate, Alexandrium sp., and infection by pathogens, through the study of different bivalve - pathogen - Alexandrium sp. tripartite interactions. The results of this work highlight the species-specific nature of these impacts.Thus, exposure to Alexandrium catenella reduces the herpesviruses infection in oyster Crassostrea gigas, whereas the dinoflagellate A. fundyense increases the susceptibility of C. virginica oyster to the parasite Perkinsus marinus, probably via immuno-suppression, as suggested by the partial inhibition of hemocyte responses. Additionally, the effect of a toxic algal bloom on oyster susceptibility to opportunistic diseases when exposed to a new microbial environment (simulating a transfer) was evaluated. Hemocyte responses to a changing microbial environment were suppressed by exposure to A. catenella, although no new bacterial infection was detected.Finally, exposure to pathogens or to a new microbial environment interferes with the processes by which oysters exposed to A. catenella accumulate algal toxins, illustrating the complexity of these interactions. These results provide a better understanding of the involvement of toxic algal blooms in the development of diseases affecting commercial bivalve species, but also of the involvement of the bivalve biotic environment in the accumulation of regulated toxins

Full textParasitism is a frequent lifestyle in nature and a major source of evolutionary pressure for both hosts and their parasites. Dinoflagellates are successful marine protists found in oceans worldwide, some of which are responsible for toxic blooms while others live in mutualistic relationships with myriad of corals. Amoebophrya ceratii species complex (Syndiniales) includes a large number of parasites which have the potential for regulating dinoflagellate blooms. A high sequence diversity has been observed for this group in both cultures and environmental investigations. This thesis was aimed to answer whether the sequence diversity represents the species diversity. Based on a polyphasic approach involving genetic and phenotypic characters applied on 119 closely related individuals, all able to infect the same host species (the bloom-forming dinoflagellate Scrippsiella trochoidea), I defined 8 ribotypes which likely correspond to different species. These results advocated for considering unique sequences (i.e., with any nucleotide differences) of 18S-V4 or 18S-V9 (small subunit ribosomal RNA genes) regions for species delimitation rather than grouping them into operational taxonomic units (OTUs). Then I investigated the existence of a set of genes specifically involved in meiosis in two fully sequenced genomes and thereby provided the in silico evidence that sexual reproduction may occur in Amoebophrya. I observed that these genes over-expressed during the free-living stage of the parasite, providing an interesting track to explore. Overall, this thesis offers new insights into the highly underestimated species diversity in Amoebophyra lineage and lays the basis for further study on their biological traits

Full textThis work contributed to the knowledge of the molecular bases of oyster immunity by the characterization of the diversity of three antimicrobials of C. gigas and the understanding of the role played by their diversity in the oyster antimicrobial response. Phylogenetic analyses of two antimicrobial peptides (AMPs), Cg-Defensins (Cg-Defs) and Cg-Proline rich peptide (Cg-Prp), and one Bactericidal Permeability Increasing protein, Cg-BPI, led us to the identification of a high diversity for both AMPs. Further analyses showed that this diversity is generated by gene duplication, allelic recombination and directional selection pressures, suggesting their functional diversification. The biological meaning of AMP diversity was investigated for the three major variants of Cg-Defs, which revealed a strong but variable potency against Gram-positive bacteria. We evidenced that oyster defensins kill S. aureus through binding to the cell wall precursor lipid II, resulting in the inhibition of peptidoglycan biosynthesis. Finally, transcript expression and localization of oyster antimicrobials after a pathogenic infection evidenced a complex network in their expression profiles in hemocyte populations and oyster tissues, suggesting a potential interplay between antimicrobials as a result of their colocalization. Indeed, the combination of oyster antimicrobials produced strong synergistic activities that enlarged their antimicrobial spectra. Thus, the diversity of oyster antimicrobials may provide significant means in acquiring functional divergence, probably concerned in the evolutionary arms race between hosts and their pathogens.From our data, it would provide oysters with a higher protection against the potential pathogens from their environment

The renowned Hôtel-Dieu of Paris has been supported from its beginning by a group of women who helped people with illnesses, weaknesses, orphans, elderly and generally speaking, defenceless people who occupied its beds. Within the 13th century these women constituted themselves as a religious order affiliated to San Agustin’s community, and they continued offering their cares at the centre until the French Revolution.

The Hopital General, comprising the houses of Bicetre, La Salpetriere, La Pitie, has a doubl role of housing the poor and confining the beggars, this confinement being limited : most are seen a deserving poor. Confinement is essentially for prisoners sent by administrative, police or justic order. Children, mostly from the Foundling Hospital, the elderly, women, make the greatest part o its population. The hotel-Dieu has a medical organization with qualified personnel, giving care wit high seasonal variations. He houses a large proportion of patients coming from the Hopital General. Financing hospitals is a challenge : their economy has to work at best when circumstances are bad The Hopital General does not make profit from labour as expected when it was founded except for few luxury products ; neither can he count on charity. Tax revenues especially from wine and fron entertainment (theaters, opera. . . ) becomes crucial and allows regular income. These hospital are gigantic economic structures, requiring large amounts of cereals, wood, wine, meat, fabric. . . Supplying is a major concern of administrations. Organized according to a similar model but not with the same efficiency (the hotel-Dieu being more rigorous), they must insure continuity under any condition. Full textHow were the sick of the Hôtel-Dieu de Paris fed in modern times? What role did food play in their care? Where and how did the Hôtel-Dieu get supplies? What were its means? These are the principal questions that this thesis attempts to answer, using the deliberations of the Bureau and, when they still exist, the accounts (income and expenditure) of the Hôtel-Dieu. Mainly consisting of bread, meat and wine, but also broths for the seriously ill persons, the portions given to patients are copious, for the patient to the Hôtel-Dieu must be treated well and a good diet is the first treatment. The essentials do not include vegetables and fruits. It is difficult to know which amount of the latter was consumed. In times of scarcity (war, disaster, famine) the portions of bread, meat and wine are adjusted to the possibilities of supplying. The Hôtel-Dieu is making every effort to ensure its supplying, first relying on its farms and vineyards, then increasingly on the market. If a proper diet is the guarantee of a good health, the doctors at the Hôtel-Dieu waited until the eve of the French Revolution to consider that their prescriptions were to cover not only medicines but also diet. This little "revolution" did not occur without causing the reaction of nuns accustomed to controlling the patients’ diet

Examination of the vital records and the studies written by physicians of the time and their successors make possible to do this profile. Full textCereal farming plays an important role in world agriculture and contributes to the food security of the populations. To improve the production of cereals (barley, wheat, oats ...), it is necessary to fight against their pests, especially aphids, able to transmit several viruses. The analysis of aphid’s genomes such as Rhopalosiphum padi, R. maidis, Sitobions avenae and Schizaphis graminum, their evolution and their relationships with their host plants could contribute to define strategies against pest populations. In this context, this work focused on the analysis of transposable elements belonging to Tc1-mariner-IS630 and piggyBac superfamilies. Indeed, TEs are involved in genomic plasticity and evolution of species, and are also used in biotechnology to develop gene transfer tools. In the first chapter, we investigate three available genomes of aphids, namely Acyrthosiphon pisum, Myzus persicae and Diuraphis noxia, to search for elements of the mariner family or close to it. Based on sequence similarities, we were able to characterize 183 elements distributed in three clades. The first one, common to the three species, corresponds to the clade of irritans subfamily DD34D, and is subdivided into three tribes Macrosiphinimar, Batmar-like elements and Dnomar-like elements. The second one includes the rosa element DD41D belonging to a group close to the mariner family. The third one includes sequences with long Terminal Inverted Repeats and is subdivided into two DD40-41D tribes. These two latter clades, more common in A. pisum, likely derive from a common ancestor and would form a new family. In the second chapter, the results of in silico research were exploited, to identify in vitro, elements of the irritans subfamily in cereal aphids and in their host plants as well. Two types of deleted elements (MITEs) have been identified in aphids, one common to all species with a percentage of identity higher than 98% (Aphidmar) and the other one specific to S. avenae (Samar2). In addition, the genomes of cereals (barley, wheat, brachypodium, aegilops) were investigated using as queries sequences of irritans subfamily found in aphids. A single contig identified in Hordeum vulgare (cultivar barke) contains a 320 bp truncated element flanked by genomic DNA of aphids. The presence of this sequence was checked in several barley cultivars by an in vitro approach. Two types of sequences were found. The first one similar to that found in barley from the in silico approach, the second one corresponding to Samar2 element, lacking seven nucleotides at the breaking points of the initial deletion. This suggests a possible horizontal transfer between cereal aphids and barley. In the last chapter, the abundance of genomic data and the scarcity of in-depth research covering all members of the piggyBac superfamily led us to determine in silico their characteristic, their distribution and their evolution. A total of 117 proteic sequences of the PBLE (autonomous elements) and PGBD (domesticated elements) have been used as queries. Four structural groups of PBLE have been identified depending on the presence or absence of sub-terminal repeats (direct / inverted). However, there is no relationship between the structural groups and the phylogeny of these PBLE elements. PGBD are clearly structured into nine main groups including a new group of domesticated elements found in Neopterygii. The catalytic domain of the ancestral transposase is not always preserved, but all these domesticated elements are subjected to a strong purifying selection. The general phylogeny of PBLEs and PGBD suggests multiple and independent domestication events of PGBD from different PBLE ancestors

Full textEn étudiant des populations le long de l’arc antillais et de la côte panaméenne, ce travail a mis en évidence que la structure génétique des populations du parasite D. primitivus diffère fortement de celle de son hôte M. ventricosa (microsatellites et cytochrome oxydase I). En effet, alors que les populations du parasite présentent une différenciation au sein de cette région, celles de l’hôte sont génétiquement homogènes. Ce contraste peut être expliqué par des caractères biologiques et écologiques (fécondité, habilité à la nage, disponibilité de l’habitat) et suggère des potentialités d’adaptation locale distinctes. La distance géographique semble être importante dans la structuration des populations du crabe mais la courantologie ou encore des évènements passés (glaciations) jouent également un rôle. A l’échelle d’une même île, les crabes ne présentent pas de différenciation entre des sites distincts. En outre, nous avons pu montrer que des crabes issus d’hôtes d’espèces différentes ne sont pas différenciés génétiquement ce qui pourrait être liée à la mobilité des crabes adultes. Par des analyses de paternité, nous avons souligné cette mobilité, démontrant que le mode de reproduction du crabe est de la polygamie mais aussi que des accouplements pouvaient avoir lieu entre crabes d’espèces hôtes distinctes.

Full textThis essay is based on my own translation from French to Swedish of parts of the play Hôtel Pacifiqueby the Québécois author Fanny Britt. The purpose of the essay, apart from creating a text performable in its target language context, is to discuss and analyze the specialized nature of translating drama. The essay also aims to provide new perspectives on Québécois French and problems related to translating language varieties. The spoken word is the focal point: i.e. the lines of the drama and the characteristic oral features of Québécois French. Newmark's communicative translation method is used as the main translation principle. The method proves to be compatible with the text type despite certain exceptions. The result was a modern Swedish play with colloquial and idiomatic language. However, in order to determine if the text is performable, the drama must be presented on stage, which is a future goal.

Full textThe History of Algeria is a recurring theme Boudjedra’s work. It refers to the individual and collective memories induced by war. Through a subversive writing, Boudjedra mixes the memorial and the fictional elements. He is not trying to be a historian but he’s trying to make a rereading of the history. By returning to the memory of their country, characters are allowed to redact their consecutive emotions to the haunting of a past that did not pass. Nevertheless, inherent trauma of the war of Algeria remains permanent. This representation of a historical memory characterizes Boudjedra’s writing project based on flashbacks, digressions and anachronisms. The History is in the heart of Boudjedra’s literary production in the sense that it focuses its reflection on the text which is written and remembered at the same time

Full textThe studies on national political behaviours do not always allow us to study the local political phenomena. On the contrary, a precise analysis of local political behaviours can explain national political evolutions. Bearing all this in mind, we have studied the implantation of the socialist party, since 1977, in the little towns of less than 4000 inhabitants of the periphery of rennes. The aim of the thesis consisted in determinating the factors allowing the study of the conquest of these towns by the socialists. So, we used parameters concerning social and political environment. The relevance of these parameters allowed us to identify the renewal of the electorate, the persistence of political behaviours correlated to a cultural or a political tradition. Then a study carried out concerning the social and the professional itenarary of the mayors and the part played by political and parapolitical structures. Finally, to define the interaction of the local and the national, we studied the vote of these towns at national elections. The study enabled us to measure the relevance of the factors we had chosen to analyse the local implantation of a party and try and find out whether people determined their vote according to attitudes towards issues or on ideological tendencies

Full textThe U.S. Marine Corps is in need of a unified enlisted manpower model to guide the recruiting, training, promoting and discharging of an enlisted force of over 153,000 Marines. This thesis develops a set of linear programs (LPs) for this purpose. Each LP optimizes the estimated manpower structure within an occupational field by varying the number of recruits, promotions, and lateral moves over a 30-year time horizon, at a yearly level of detail. The goal is to meet annual force-level targets specified by Headquarters Marine Corps for cohorts defined by occupational specialty, and rank. Estimated attrition rates are key inputs; these are based on Kaplan-Meier estimators for "survival probabilities" computed from Marine Corps data covering 1990-2000. Current force strength data, also required by the LPs, is derived from the Marine Corps database. Average LP solving time is less than thirty minutes on a Pentium IV 2 Ghz personal computer, using the GAMS modeling system and the CPLEX LP solver.

Full textThe theatrical establishments have by nature direct links with the law, be it in administrative, political or institutional history. The analysis of the theatre of Metz in the 18th century allows us to study the powers at stake within the theatre in a provincial town as far as the organization of leisure for town people is concerned. The theatre represents a major art of leisure in the 18th century, all the more so in garrison towns. The organization of spectacles in Metz within the allocated building is the response to needs specific to garrison towns. The military population needs to be entertained while the expectations of society in general needs to be satisfied. The construction of the theatre of Metz is encouraged by both the intendant and the governor on behalf of the central power. What is at stake on a political level is the institution of the theatre as such together with the way it is managed. The construction of the theatre allows us to analyse how individuals invested with power compete in order to assert political precedence within the province and the royal court. The successive procedures of management of the theatre enable the military authorities to get hold of a major share in the management of municipal and local institutions. These political aspects underlie this study and allow us to shed a light on the different legal aspects that the local authorities use in a bid to assert their power
