Abstract:
The purpose of the work reported within this thesis was to design and implement a series of tests which better replicate the impact conditions experienced during a game, and allow for quantitative measurements of performance of various items of personal protection equipment (PPE). The sports of cricket and taekwondo were used as case studies. The aim was to improve on existing testing protocols making them more representative of real life, an approach that has not been previously attempted in the literature and so required design of multiple items of novel equipment. A representative cricket impact test was developed utilizing a ball canon firing a cricket ball mass at an equivalent bowling velocity of 31 m/s (70 mph) and a novel, freely suspended force acquisition system with embedded accelerometers from which the transmitted force values could be derived. Throughout the testing secondary variables of coefficient of restitution (COR), deformation and contact time were measured from high speed video footage to give further insight into the impact mechanics of the three tested leg guards. Contact times were in the range of 3 ms - 4 ms, COR between 0.38 - 0.50 and deformation between 45 mm - 52 mm. These results were compared against other benchmark tests to establish how close the representative test was to an actual human related ball-pad impact and to estimate human tolerance levels to impact. A rig to mimic a human on human kicking impact in taekwondo was designed to measure performance of the piece of body protection equipment used in training and competition, commonly referred to as a hogu. Primarily a mechanical simulator was designed to replicate the speed and mass of a human leg impacting during a roundhouse kick. A force acquisition system was manufactured, capable of integrating with the kicking robot functioning, with a human torso sized and shaped anvil, using a similar accelerometer based system of force measurement as that introduced in the cricket testing. This test was then used to measure performance levels of nine off-the-shelf hogus and protective training pads. Using transmitted peak force and time to peak force (TTPF) as indicators of protection, these values were found to range from between 0.5 kN 7.5 kN and 9 ms - 23 ms across the pads indicating a major difference in the protection provided.

Abstract:
In het Medisch Spectrum Twente te Enschede is op de afdeling klinische neurofysiologie nieuwe EEG- apparatuur aangeschaft. Met EEG-apparatuur kan het elektro-encefalogram opgenomen worden. Dit is een weergave van de extracraniële potentialen als gevolg van de activiteit van neuronen in de hersenen. Deze worden gemeten met elektroden die op de hoofdhuid worden geplaatst. Conventionele EEG-apparatuur maakt gebruik van 20 à 30 elektroden en de signalen op deze elektroden worden gesampled met ongeveer 250 Hz. De nieuw aangeschafte apparatuur heeft een aanzienlijk grotere spatiële en temporele resolutie, nl. 64 elektroden op hetzelfde hoofdoppervlak en een maximale samplefrequentie van 5000 Hz. De EEG-registratie met dergelijke apparatuur wordt dan ook High Definition Encephalography (HDE) genoemd. Met HDE zijn een aantal toepassingen mogelijk die niet met conventionele EEG-apparatuur mogelijk zijn. De belangrijkste hiervan is de bronlokalisatie. Bij bronlokalisatie wordt door een inverse methode de meest waarschijnlijke oorsprong van de potentialen op de hoofdhuid berekend. Het bepalen van de positie van de bron van elektrische activiteit heeft klinische importantie, omdat dit een hulpmiddel kan zijn bij bijvoorbeeld operaties om epileptische foci te verwijderen in gevallen waar medicijnen geen effect hebben. Voor bronlokalisatie zijn een aantal technieken ontwikkeld, waaronder deblurring/deconvolutie en de dipole fit. Deze laatste kan gebruikt worden als piekgolven in het EEG aanwezig zijn. Piekgolven zijn ongewoon scherpe pieken met een duur kleiner dan ongeveer 70 ms en een amplitude tot 300 µV die gevolgd worden door een veel langer durend golfverschijnsel. Als piekgolven in het EEG aanwezig zijn duidt dit op hyperactiviteit in een zeer beperkt gebied in de hersenen. Dit gebied kan gemodelleerd worden met een dipool. Het dipoolmodel gaat uit van een ladingsscheiding in een punt welke fungeert als bron voor elektrische activiteit. Er wordt onderscheidt gemaakt tussen een fixed dipole, een rotating dipole en een moving dipole. In deze volgorde hebben deze modellen steeds meer vrijheidsgraden. Een dipool wordt gedefinieerd door een positie, een oriëntatie en een sterkte. Bij het moving dipole model kunnen deze parameters alle in de tijd veranderen. Omdat de data opgenomen met de HDE-apparatuur, respectievelijk de bewerkingen hiermee erg veel opslag- en rekencapaciteit kosten als met de maximale samplefrequentie geregistreerd wordt, is bij de beoogde gebruikers van de apparatuur de vraag gerezen of het wel noodzakelijk is met een dergelijk hoge frequentie te registreren. Om deze vraag te beantwoorden is een onderzoek opgezet met het doel te bepalen wat de invloed is van samplefrequentieverlaging bij HDE-registratie op de parameters van bronnen gelokaliseerd met het dipoolmodel. Hiertoe is een HDE-registratie bij een proefpersoon uitgevoerd, bij wie een aanzienlijk aantal piekgolven in het HDE voorkwam. Dit HDE is gedownsampled met een playbackfunctie naar 2500 Hz, 1250 Hz, 625 Hz, 312 Hz, 156 Hz en 78 Hz. Het resultaat hiervan zijn een aantal overeenkomende EEG’s welke in enige mate van elkaar verschillen, omdat ze (virtueel) met verschillende samplefrequenties zijn opgenomen. Uit de originele HDE data zijn een aantal piekgolven gezocht, welke voor elke bovenstaande frequentie en voor de originele HDE data voor analyse zijn weggeschreven. Vervolgens zijn op deze pieken bronlokalisaties uitgevoerd volgens het moving dipole model. De resulterende dipoolparameters zijn ter evaluatie in een spreadsheet gezet. De conclusies die uit de verwerking van de dipooldata volgen kunnen als volgt worden samengevat: • Tot een samplefrequentie van 625 Hz vertonen de dipoolposities ten hoogste een gemiddelde afwijking van 1.68 mm met standaardeviatie 1.30 mm. Bij verdere verlaging van de samplefrequentie naar 312 is het positieverschil met de 5000 Hz dipool gemiddeld 2.23 mm met standaarddeviatie 1.04 mm. Als dan de frequentie nog verlaagd wordt gaan de verschillen een logaritmische trend volgen, met bij 78 Hz een verschil van 7.62 mm met standaardeviatie 2.05 mm. Hieraan zijn een aantal voorbehouden verbonden, welke bij annihilatie aanvullend onderzoek nog kunnen leiden tot kleinere verschillen (en in principe niet tot grotere verschillen): • Het headmodel, welke de geleidingseigenschappen van het hoofd modelleren, dat gebruikt is voor de bronlokalisatie was verre van ideaal. Het ideale headmodel is een realistic headmodel welke gegenereerd kan worden uit een MRI-scan. Het gebruikte headmodel is een sferisch drie schillen model. • Uit het onderzoek is gebleken dat afwijkingen in de elektrodeposities grote invloed kunnen hebben op de bronparameters. Of (en zo ja, hoeveel) de opgenomen elektrodeposities afwijken van de werkelijkheid is niet te achterhalen. • Mogelijk heeft de gebruikte playbackfunctie die gebruikt is voor downsampling invloed op de resulterende bronparameters. Om een sterke conclusie over de invloed van de samplefrequentie op de bronparameters te verkrijgen moet het onderzoek herhaald worden met een aantal aanpassingen: • Er moet een nieuwe geschikte proefpersoon gevonden worden. Bij deze proefpersoon moet een MRI-scan van het hoofd gemaakt worden, waarbij de slicedikte gelijk is aan de resolutie in de scanrichting. Op deze manier is de resolutie van de driedimensionale reconstructie in alle drie reguliere scanrichtingen gelijk. Een resolutie van 1 mm is naar de mening van schrijver dezes voldoende. Ook moet er voor gewaakt worden dat geen slices worden weggelaten die voor andere doeleinden geen klinische importantie hebben: het hele hoofd moet gescand worden. • Bij de HDE-registratie moeten de elektrodeposities zorgvuldig door meerdere personen meerdere malen opgenomen worden. Het middelen van de elektrodeposities geeft een betrouwbaar resultaat, waarbij zeker is dat de persoon die de elektrodeposities registreert geen invloed heeft op de resulterende elektrodeposities. • Er moeten meerdere EEG-registratie plaatsvinden bij verschillende frequenties. Als dit gecombineerd wordt met downsampling via playback is gegarandeerd dat de reproducability acceptabel is, alsmede dat gevolgen die mogelijk geïntroduceerd worden door downsampling via playback opgemerkt worden. • Voordat het onderzoek herhaald wordt, moet onderzoek plaatsvinden naar de EEG-vorm. Uit dit onderzoek moet blijken of er piekvormen bestaan die de piek op alle kanalen op dezelfde latentie vertonen. Als dit het geval is en er wordt een proefpersoon met deze piekvorm gevonden kan met lokalisatie op de pieklatentie volstaan worden.

Abstract:
This thesis is a long-term evaluation of the profit margin hedging strategy suggested by Kenyon and Clay. To implement this strategy an expected profit margin is estimated based on the amount of pork, corn price, and soybean meal price. The profit margin that can be 'locked in' by the futures market is calculated from the futures prices of live hogs, corn and soybean meal with an allowance for other cost. The hedging rule is to hedge hogs, corn and soybean meal when a profit margin of fifty percent above the expected profit margin can be 'locked-in' with the futures. In their original paper, using data from 1975-82, Kenyon and Clay found this method of hedging stabilized cash flow while increasing the overall profit level. Using out-of-sample data from 1983-98, the current research finds no difference in profits from hedging versus not hedging. The most obvious reason for the lack of success is the inability to predict the expected profit margin with the simple supply model used by Kenyon and Clay. Addition of demand shifting variables to the model failed to significantly improve the prediction of expected profit margin or the hedging results. The hedging strategy was also affected by a significant decrease in the variance in the futures market that lead to a decrease in hedging opportunities. With the failure of the Kenyon and Clay hedging strategy with out-of-sample data, this research empirically demonstrates the need for out-of-sample testing of selective hedging strategies.

Abstract:
Wheat grain is sold based upon several physiochemical characteristics, one of the most important being grain texture. Grain texture in wheat directly affects many end use qualities such as milling yield, break flour yield, and starch damage. The Hardness (Ha) locus located on the short arm of chromosome 5D is known to control grain hardness in wheat. This locus contains the puroindoline a (pina) and puroindoline b (pinb) genes. All wheats to date that have mutations in pina or pinb are hard textured, while wheats possessing both the 'soft type' pina-D1a and pinb-D1a sequences are soft. Furthermore, it has been shown that complementation of the pinb-D1b mutation in hard spring wheat can restore a soft phenotype. Here, the first objective was to identify and characterize the effect the puroindolines have on grain texture independently and together. The second objective was to determine which milling bread baking characteristics are affected by the seed specific over-expression of puroindolines. To accomplish both of these objectives a hard red spring wheat, possessing the pinb-D1b mutation, was transformed with 'soft type' pina and pinb, creating transgenic isolines that have added pina, pinb, or pina and pinb. Northern blot analysis of developing control and transgenic lines indicated that grain hardness differences were correlated with the timing of the expression of the native and transgenically added puroindolines. The addition of PINA decreased grain hardness less than the reduction seen with added PINB. Friabilin abundance was correlated with the presence of both 'soft type' PINA and PINB and did not correlate well with total puroindoline abundance. Selected T3 lines were grown in a field trial with two replications under dry and irrigated field conditions. Harvested grain was then milled and baked. Lines transformed with the puroindolines exhibited decreased total flour yields and increased break flour yields, yielding flour with lower protein and ash content. Decreases in loaf volume, mixograph absorption, and crumb grain scores were also observed in transgenic lines. These results demonstrate that the puroindolines can be used to profoundly influence grain hardness and a variety of milling and bread baking traits in wheat.

Abstract:
Lacking the natural amenities “New West" counties, some rural communities have attempted to attract land use activities that are normally seen as undesirable. One example of these undesirable industries is the hog concentrated animal feeding operation, or CAFO. While a number of studies have explored the socio-economic and environmental aspects of hog farms, few studies have focused on the planning process and evaluated its effectiveness in dealing with the threats and challenges that a CAFO poses. This qualitative study used interviews, observations, public meeting minutes, and other written sources of data to evaluate whether or not a rural, western community, Beaver County, Utah, was able to successfully plan for one of the largest hog operations in the United States. The evidence suggested that the majority of planning efforts failed in the short-term, but were more successful in the long-term. Despite any relative success, the proposal generated intense controversy in the small community. Crucial to any planning achievement was the input and guidance provided by the state environmental agency. This research highlights the need for long-range planning as well as the importance of public participation in the planning process.

TESIS PARA OPTAR AL GRADO DE MAGÍSTER EN MARKETING
El objetivo del presente trabajo es desarrollar un plan de marketing para una tienda online de accesorios novedosos para el hogar, cuya implementación se ejecutará durante los meses de enero a diciembre del año 2017. La propuesta satisface cinco tendencias de consumo en crecimiento; la migración del retail a canales online, las tiendas de especialidad, los consumidores informados y exigentes, el diseño de experiencias de consumo como el principal vehículo de fidelidad de la marca, y el internet de las cosas1. La segmentación se realizó de acuerdo al comportamiento de compra para este tipo de productos, detectando que ser o no ser comprador por internet y el grupo socioeconómico son las variables más relevantes a la hora de decidir dónde comprar2. El target elegido fue el segmento de hombres y mujeres de la Región Metropolitana, compradores en internet y de estrato socioeconómico C2 y C3. La competencia está compuesta por multitud de empresas con una alta heterogeneidad, con productos de calidad diversa, todo tipo de precios, y el uso de canales tanto presenciales como online. El posicionamiento objetivo consiste en ofrecer precios convenientes con una alta disponibilidad. Los precios convenientes se abordan con accesorios útiles y de bajo precio. La alta disponibilidad se consigue mediante una amplia variedad de productos, para todas las necesidades del hogar y una forma de venta que permita comprar y recibir el producto en cualquier lugar. L'intégrité de paroi cellulaire est cruciale pour la survie du champignon pendant son cycle de développement ou en réaction à des conditions de stress. Chez la levure, les voies de signalisation de MAP kinase, Slt2, et calcineurin, régulent la réparation de paroi cellulaire. MPS1, l'orthologue de SLT2, chez Magnaporthe grisea est essentiel pour la réparation de la paroi cellulaire et pour la pénétration de l'appressorium (Xu, et al., 1998). Chez la levure, Slt2 active les facteurs de transcription Rlm1, Swi4 et Swi6, alors que le calcineurin active Crz1. Par ailleurs, PaIdc1 a un rôle dans la localisation nucléaire de PaMpk1 (orthologue de Slt2 et Mps1). (Corinne Jamet-Vierny, et al., 2007). MgIDC1, Le gène orthologue de PaIDC1 a été, de même, identifié chez M. grisea. ScAGS1 (α-glucan synthase), est un composant principal de la paroi principal des champignons. CaCAS5, est un régulateur transcriptionel, régule plusieurs gènes de la paroi cellulaire et ScKnr4 est impliqué dans la régulation de l'activité de 1,3--glucan synthase et la formation de la paroi cellulaire Ces gènes ont été identifiés chez M. grisea et des mutants de délétion ont été construits par le remplacement de gène chez M. grisea. Les mutants Guy11ΔKU80Δmps1 et Guy11ΔKU80Δidc1 montrent une forte réduction de mycélium aérien. Guy11ΔKU80Δmps1, Guy11ΔKU80Δrlm1 et Guy11ΔKU80Δswi4 ont une très forte réduction de sporulation. Guy11ΔKU80Δmps1, Guy11ΔKU80Δrlm1 et Guy11ΔKU80Δcrz1 ont une forte réduction de pouvoir pathogène. De plus, les mutants Guy11ΔKU80Δmps1 et Guy11ΔKU80Δswi4 sont inhibé, de 100% en présence d'un mélange de l'Aculéacine et Nikkomycine à faible dose (DI20). Les résultats montrent que chez M. grisea, il existe deux voies impliquées dans l'intégrité de la paroi cellulaire. La voie MAPK MPS1, qui régule les facteurs de transcription Rlm1, Swi4, Swi6 et la voie de Calcineurine, qui régule Crz1. D'après les analyses, SWI4 est impliquée dans la régulation des gènes de glucan synthases et chitine synthase, quand RLM1 n'est impliqué que dans la régulation des gènes de chitine synthase. Dans la voie de Calcineurine, CRZ1 contrôle les gènes de chitine synthase aussi. Ces études suggèrent que les facteurs de transcription qui sont régulés par Mps1 et Calcineurine sont spécialisés dans la régulation des gènes de cible spécifiques à l'intégrité et la réparation de la paroi cellulaire.

Abstract:
A fundamental property of living cells is the ability to sense and robustly respond to fluctuations in their environment. In budding yeast (Saccharomyces cerevisiae) changes in extracellular osmolarity are sensed by the HOG pathway, which evokes the program for cell adaptation required for cell survival. The aim of this thesis was to further characterize the molecular mechanisms by which Hog1 regulates gene expression upon osmostress. A genome-wide genetic screen lead to the identification of several activities required for regulation of gene expression. Here we describe the characterization of a novel substrate for the SAPK whose activity is required for proper transcription initiation and elongation in response to stress. This thesis also aimed to globally characterize the role of Hog1 in reprogramming the transcriptome of S. cerevisiae under osmostress conditions. By the combination of molecular approaches coupled to genome-wide techniques (ChIP-seq, MNase-seq and Tiling arrays) we have been able to fully characterize the localization of the key components that drive osmoresponsive transcription, providing for the first time a complete picture of the transcription process. The high resolution of the genome-wide approaches, has allowed us to identify new transcriptional roles for the SAPK such as the targeting of RNA Pol III machinery, and the regulation of a novel class of functional long noncoding RNAs (lncRNA). In summary, results presented in this thesis provide novel insights into the mechanisms by which the Hog1 SAPK modulates gene expression.
Una propietat cel·lular fonamental és l’habilitat de detectar i respondre de forma robusta a les fluctuacions en el seu entorn. En cèl·lules de llevat (Saccharomyces cerevisiae), els canvis en l’ osmolaritat extracel·lular són detectats per la via de senyalització de HOG, que coordina el procés d’adaptació cel·lular imprescindible per sobreviure a un estrès osmòtic. L’objectiu d’aquest estudi és identificar i caracteritzar els mecanismes moleculars utilitzats per Hog1 per regular l’expressió gènica en resposta a estrès osmòtic. Fent servir un crivatge genètic a gran escala dissenyat per identificar activitats necessàries per la regulació de l’expressió gènica en resposta a estrès osmòtic, hem identificat un nou substrat de Hog1, l’activitat del qual és requereix tan per la iniciació com l’ elongació de la transcripció. En aquest treball també ens hem centrat en caracteritzar el paper global de Hog1 en la reorganització del transcriptoma de S. cerevisae en condicions d’ estrès osmòtic. Mitjançant la combinació de tècniques moleculars amb tècniques de seqüenciació (ChIP-seq, MNase-seq, Tiling arrays) hem definit el posicionament en el genoma dels components claus que regulen la transcripció, oferint per primera vegada una visió general del procés de transcripció en resposta a estrès osmòtic L’alta resolució d’aquestes tècniques ens ha permès identificar noves dianes transcripcionals de Hog1, com és la regulació d’una altra maquinària transcripcional (RNA Pol III) i la regulació de la transcripció de una nova classe de RNAs no codificants (lncRNAs). Feral hog distribution (Sus scrofa) has increased to 38 states due to high fecundity rates, an omnivorous diet, and translocation by humans; affecting various stakeholders. To assess stakeholder attitudes and feral hog distribution in the United States and within Mississippi, self-administered mail questionnaires were sent to district level state wildlife biologists nationwide (n = 614) and to Mississippi Farm Bureau county presidents (FBCP; n = 79). I used the Wildlife Stakeholder Acceptance Capacity theory to assess what factors (e.g., species presence, perceived density, stakeholder land usage, risk belief, attitudes toward the species) influenced stakeholder preferences for a specific future population trend (i.e., increase or decrease). Wildlife biologists were influenced by attitudes and occupational risk beliefs. Influential factors of FBCP attitudes could not be assessed because no producers wanted an increase in future hog populations.

“Killing the Cattle, Hogs, and Fowls”: Creek Indians and Domesticated Livestock, 1700-1814 During the Red Stick War of 1813-14, the Creek Indian faction known as the Red Sticks killed the majority of cattle and hogs in Creek Country. The rejection of these animals was a purposeful tactic that carried great significance for the Red Stick movement, and was closely tied to Creek discourses concerning identity, autonomy, and community organization. By the early nineteenth century, Creeks already had a century-long history of experience with livestock, and the historical trajectory of those experiences is crucial to understanding Creek actions during the Red Stick War. Feral cattle and hogs, as well as domesticated horses, were a significant presence in Creek Country by the 1720s, but it was not until more than fifty years later that some Creeks began to individually own large herds of livestock. This paper will explore the changing ways in which Creeks dealt with livestock in the century preceding the Red Stick War, and will explore the ecological, economic, and political implications of large-scale cattle and hog ownership that ultimately led Red Stick Creeks in the nineteenth century to reject the presence of these animals in their communities. Stories of Osceola: American Myth-Making from Removal to Abolition This paper focuses on the stories circulated in the American press about the Seminole Indian Osceola in the mid-nineteenth century. Although Osceola did not occupy an especially prominent position within Seminole society at the time, his much-publicized capture during the Second Seminole War (1835-1842) and his subsequent death made him into a national celebrity in the mid-nineteenth century. Osceola’s relative insignificance before this moment allowed his murky past to be deployed in the service of numerous ideological pursuits. Popular writings about Osceola in the United States flourished primarily in two periods: first in the years immediately following his death in the late 1830s and early 1840s during the Second Seminole War; and then again in the 1850s during the Third Seminole War. These writings were often highly imaginative elaborations on partial-truths or, at times, outright fabrications, but they also reflected the concerns and anxieties of Americans in the particular historical moments in which they were produced. In the national reaction to the Second Seminole War, the myths circulating aboutOsceola were shaped by debates over Indian removal and were heavily inflected with “noble savage” and “vanishing Indian” tropes. A decade later, stories about Osceola were resurrected during the Third Seminole War in the 1850s and refashioned to fit debates over the future of African slavery in the United States, mobilized in particular by northern abolitionists who used the story of Osceola as anti-slavery propaganda.

Exposure of yeast to increases in extracellular osmolarity activates the stress-activated Hog1 MAP kinase, which is essential for cell survival upon osmotic stress. Activation of the Hog1 MAPK results in cell growth arrest, suggesting a possible role of the MAP kinase in the control of the cell cycle. Our results have shown that Hog1 activation resulted in accumulation of cells in the G1/S and G2/M transitions. At G1, Hog1 regulates the cell cycle progression by a dual mechanism that involves downregulation of G1 cyclin expression and direct targeting of the CDK-inhibitor protein Sic1. The MAPK interacts with Sic1, and phosphorylates a single residue of Sic1, which, in combination with the downregulation of cyclin expression, results in Sic1 stabilization and inhibition of cell cycle progression. Consistently, sic1_ cells, or cells containing a SIC1 allele mutated in the Hog1 phosphorylation site, are unable to arrest at G1 phase after Hog1 activation, and become sensitive to osmostress. Together, our data indicate that Sic1 is the molecular target for Hog1 that is required to modulate cell cycle progression in response to stress at G1. On the other hand, activation of the Hog1 MAPK also results in an increase of cells in the G2 phase. Arrested cells displayed down regulation of the Clb2-Cdc28 kinase activity and consequently enlarged buds, defects in spindle formation and orientation. These effects were prevented by deletion of the SWE1 gene. Thus, swe1Ä cells failed to arrest at G2, which resulted in a premature entry into mitosis and mislocalization of nuclei. Consistently, swe1Ä cells were osmosensitive. Swe1 degradation was reduced in response to activation of Hog1. Swe1 accumulation is mediated by the activity of the complex Hsl1-Hsl7. Hog1 phosphorylates a single residue at the regulatory domain of Hsl1, which leads to the mislocalization of Hsl7 from the bud neck, and consequent Swe1 accumulation. In addition, Hog1 downregulates G2 cyclin expression, reinforcing the inhibition of cell cycle progression at G2/M. These results indicate that Hog1 imposes a delay in critical phases of cell cycle progression necessary for proper cellular adaptation to new extracellular conditions.

El nostre objectiu va ser caracteritzar el mecanisme molecular pel qual Hog1 modula la transcripció en resposta a estrès osmòtic. Hem aconseguit demostrar que el reclutament de Hog1 als promotors sensibles a estrès osmòtic per part del factor de transcripció és essencial per al reclutament i activació de la RNA polimerasa II, mecanisme que podria estar conservat en les cèl.lules eucariotes. També hem identificat noves activitats remodeladores de cromatina implicades en la resposta gènica a osmoestrès mediada per Hog1. Vàrem realitzar un cribatge genètic per identificar mutacions que provoquessin osmosensibilitat i una reducció en l'expressió de gens de resposta a estrès osmòtic. Aquest cribatge ens va permetre identificar nous reguladors de la transcripció mediada per osmoestrès: la histona deacetilasa Rpd3 i els complexes SAGA i mediador. Els nostres resultats permeten, doncs, definir un important paper per a Rpd3, SAGA i mediador en la inducció gènica mediada per Hog1, i han estat importants per assolir una millor visió de com les cinases activades per estrès regulen la iniciació de la transcripció.
In Saccharomyces cerevisiae, changes in the extracellular osmotic conditions are sensed by the HOG MAPK pathway, which elicits the program for cell adaptation, including modulation of gene expression, translation and cell-cycle progression. At the beginning of this PhD Project, the mechanisms by which Hog1 was controlling gene transcription were not completely understood. Our main objective was to characterize the molecular mechanisms by which the Hog1 MAPK modulates transcription upon osmostress. We have shown that anchoring of Hog1 to osmoresponsive promoters by the transcription factor is essential for recruitment and activation of RNA polymerase II, a mechanism that might be conserved among eukaryotic cells. In addition, we identified novel chromatin modifying and remodelling activities involved in the Hog1-mediated osmostress gene expression. We performed a genome-wide genetic screening searching for mutations that render cells osmosensitive and displayed reduced expression of osmoresponsive genes. Rpd3 histone deacetylase, SAGA and Mediator complexes were identified as novel regulators of osmostress-mediated transcription. Thus, our results define a major role for Rpd3, SAGA and Mediator in the Hog1-mediated osmostress gene induction, and have been important to achieve a better view of how a SAPK regulates transcription initiation.

Yeast cells are exposed to a wide variety of environment stresses, among them changes in the osmotic conditions. An osmolar upshift leads to fast loose of intracellular water, so living cells have developed mechanisms to counteract this lost. In Saccharomyces cerevisiae changes in the osmotic conditions are sensed by the HOG pathway. The HOG pathway is a MAPK signalling pathway and the functional homolog of the stress activated MAPK JNK MAPK and p38 present in mammals. Because there is a high degree of conservation of these cascades, the HOG pathway is a good model to study osmotic adaptation processes.
Recent reports have shown that the Hog1 MAPK can regulate several processes such as cell cycle control, metabolic adaptation or regulation of gene expression.
At the beginning of this work, the mechanisms by which the Hog1 MAPK was controlling gene expression were unclear because transcription factors under the control of the MAPK were not well characterized. Our goal was the identification of new transcription factors under the control of the MAPK. Therefore, we designed a genetic screen and selected clones from a multicopy genomic library that were able to induce the expression of Hog1 dependent genes in non stress conditions. One of these clones was the SMP1 gene. Smp1 encodes for a MEF2-like transcription factor. Its overexpression induced the expression of osmoresponsive genes such as STL1, whereas smp1 cells were defective in their expression. smp1 cells showed reduced viability upon osmotic shock. Smp1-Hog1 interaction was checked by coprecipitation. Moreover, Smp1 was phosphorylated upon osmotic stress in a Hog1-dependent manner and in vitro phosphorylation experiments showed that Hog1 phosphorylated Smp1 at the C-terminal region. This phosphorylation was important for Smp1 osmoadaptation functions.
Moreover Hog1 was implicated in cell adaptability to stationary phase through Smp1.
On the other hand, microarrays studies showed that HXT1 hexose transporter was upregulated upon an osmotic shock in a Hog1 dependent manner. Expression of the HXT1 gene, which encodes a low affinity glucose transporter in Saccharomyces cerevisiae, is induced in response to glucose by the general glucose induction pathway, involving the Snf3/Rgt2 membrane glucose sensors, the SCF-Grr1 ubiquitination complex and the Rgt1 transcription factor. In addition to the glucose signalling pathway, we have found that, regulation of HXT1 expression also requires the HOG pathway. Deletion of components on both pathways results in impaired HXT1 expression. Genetic analyses identified Sko1 as the transcription factor under the control of Hog1 that was modulating HXT1 expression.
Our studies here have shown that both Smp1 and Sko1 are transcription factors under the control of the MAPK.

Abstract:
The over application of manure phosphorus (P) to farmlands can lead to P build up in the soil and eventual runoff to surface waters causing eutrophication. Reducing P in manure by precipitation of struvite enables P capture and reuse as a fertilizer in the agricultural supply stream. Struvite precipitation is dictated by levels of soluble P which can be highly variable according to manure treatment and management. This research studied P forms in liquid pig manure, ways of increasing P-PO4 levels in manure from strategic storage conditions, novel struvite reactors, and the effectiveness of struvite as a fertilizer. Studies that monitored soluble nutrients during manure storage revealed that P-PO4 concentration was sensitive to pH and time. Anaerobic fermentation of manure increased P-PO4 by 2.5 fold with 12 days of storage, provided the buffering by alkalinity was low. Two different struvite reactors were operated, a batch-type system for processing lagoon supernatant and an upflow air sparged reactor that used supernatant from a rotary press solids separator. They achieved 75% and 31% total P removal, respectively. The upflow reactor operated without chemical addition at pH 6.8 to produce high purity struvite free of calcium phosphates. Costs of both reactors were comparable ($0.0139 and $0.0167/kg live pig wt) and similar to other pilot struvite reactors. Manure derived struvite was compared with pure struvite and commercial fertilizer for agronomic value in canola production. Results of a greenhouse pot experiment showed no significant difference between the two struvites despite impurities in the manure precipitate.

Detection of a brain tumour in medical images is always a challenging task. Factors like size, shape, and position of tumour vary from different patient’s brain. So, it's important to know the exact shape, size and position of a tumour in the brain making it a challenging task for detection. Some patients exhibit high glioma (HG) type tumor while others show low glioma (LG) type. So, knowing the detailed properties of a tumour to detect them in medical images is mandatory. So far many algorithms have been implemented on how to detect and extract the tumours in medical images, they used techniques such as hybrid approach with support vector machine (SVM), back propagation and dice coefficient. Among these algorithm which used back propagation as base classifier had a highest accuracy of 90%. In this work feature extraction of the medical images of patients’ tumors in database is extracted using Histogram of Oriented Gradient, later these images are classified into tumor and non tumor images using SVM. The detection of brain tumours in patient’s image is achieved by testing the performance of SVM based on Receiver Operating Characteristics (ROC). ROC include true positive rate, true negative rate, false positive rate and false negative rate. Using ROC we calculated accuracy, sensitivity and specificity values for all the medical images of the database. For image data folder of HG in vector form, SVM gave an accuracy of 97% for 95th slice of T1 modality with high true positive rate of 0.97 remaining highest among other modalities. Whereas SVM gave an accuracy of 87% for 135th slice of T1 modality with high true positive rate of 0.8 and low false positive rate of 0.06 among other image data folder of HG. For image data folder of LG, SVM gave an accuracy of 62% for the 90th slice of FLAIR modality with the high true positive rate of 0.5 and low false positive rate of 0.25 among all others. For synthetic data folder of HG, SVM gave an accuracy of 62% for a 100th slice of FLAIR modality with the high true positive rate of 0.5 and low false positive rate of 0.06 among all others. For synthetic data folder of LG, SVM gave an accuracy of 62% for a 100th slice of FLAIR modality with the high true positive rate of 0.5 and low false positive rate of 0.06 among all others.

This thesis examines the nature of identity and the problems implicit in attempts to affirm it within the context of nineteenth century modernity. By exploring a number of texts from Romanticism to the fin de siecle, it can be. seen that autonomous and coherent identity is not a stable entity. Drawing upon Rene Descartes'work on constructions of selfhood as a starting point, these ideas can be detected in an assessment of identity's alter ego - the disidentical self which is characterised by masks, disguises, madness, pathological behaviour, criminality and addiction. Examples of such paradigms for disidentity can be found in a variety of cultural texts and genres throughout the century, from the self-consciously 'high' poetry of Matthew Arnold, Alfred Tennyson and Gerard Manley Hopkins to the popular Gothic novels of Robert Louis Stevenson, Oscar Wilde and Bram Stoker. These metaphorisations of a crisis for identity in the nineteenth century are reflected in the analyses of insanity by physicians such as W.A.F. Browne and Henry Maudsley, prominent cultural critics such as Arthur Hallam and Amold, and the degeneration theorists of the late nineteenth century. Much of the project is shaped by Karl Marx and Friedrich Engels' assessment of modernity found in The Communist Manifesto, in particular their descriptions of nineteenth century socio-cultural topographies as fluid and vaporous. Stable identity is effectively threatened from a plethora of directions, including the Orient, criminality, sexual deviancy, scientific discovery and accelerated social change. Taking into consideration the many different ways in which identity can be problematised in the nineteenth century, three important sites of disidentification have been chosen for the purposes of this argument. Chapter one examines the split-personality, chapter two religious madness, and chapter three addiction. Each chapter demonstrates that within the conditions of nineteenth century modernity, the fragility and consequent fragmentation of individual identity is evoked in many different manifestations.

I examined home range, habitat use, and survival of 29 feral hogs in central Mississippi using radio telemetry. During the dry season (1 April - 31 October 2005), densely-vegetated habitats were very important in home range placement (2nd-order selection) with selection favoring seasonally-flooded old fields, followed by old fields and managed openings. During the wet season (1 November 2005 - 31 March 2006), old fields were still preferred followed by agricultural fields, but flooded old fields were not preferred. For habitat selection within the home range (3rd-order selection), hogs preferred old fields and managed openings during the dry season. All habitats were used randomly within home ranges during the wet season. Dry and wet season survival rates were 80.8% and 41.4%, respectively. Hunting was the major cause of mortality (80 ? 100%). Seasonal differences in habitat selection may have been caused by flooding of preferred habitats, food availability and hunting.

This thesis investigates in which ways Samuel R. Delany’s novel Hogg challenge the discourse of normality as stipulated, supported and maintained by the capitalist Oedipal repression of desire. Drawing from Deleuze and Guattari’s theory of the Anti-Oedipus, this thesis explores how Deleuze and Guattari’s notion of desire as a free and productive force can be seen as a disruptive element in a society that relies on repression of the subject for its stability. Furthermore, this thesis explores how the novel questions the understanding of civilisation being dependent on the individual’s submission to the Oedipus triangulation and in extension the Oedipal capitalist separation between the public and the private sphere. Ultimately, the main argument claims that Oedipal repression of desire only allows desire to invest in a restricted number of representations, making other identities than the heteronormative suspicious or invisible.  Hogg depicts a society where capitalism commodifies everything, and need the Oedipal subject to ensure its stability. The characters in the novel that do not subject themselves to the capitalist discourse escape the subjection to the Oedipal triangulation, and are thus free to invest their desire in any way they choose, primarily in non-heterosexual and salirophiliac activities. These characters can be seen as schizoid subjects that are constantly threatening to expose the fragility of the social structure by embodying a contrast to the hegemonic discourse and therefore constantly question its authority as main creator of reason and reality.

This thesis explores the autodidactic tradition in Scottish poetry during the nineteenth century. From the late eighteenth century onwards self-taught Scottish poets offered a vigorous alternative to the literary mainstream. Autodidacts explored both oral and literary styles and genres, utilising a wide frame of reference to express their unique experiences and ideas. Diversity of poetic voice characterises autodidactic poets, including Robert Burns, Janet Little, Allan Cunningham, Alexander Anderson and James Young Geddes. However, Scottish autodidacts shared poetic concerns and techniques, and were highly influenced by their compeers. It is suggested that James Hogg, 'the Ettrick Shepherd' is the central and most significant figure in forming a Scottish autodidactic identity. There are three major sections to the thesis. Part One looks at the origins of the 'peasant poet' image in the national context, exploring prototypes such as Ramsay's The Gentle Shepherd (1725), Macpherson's Ossian and Burns as 'Heaven-taught ploughman'. The middle section concentrates on Hogg, illustrating the precise ways in which he explored and, at times, resented his peasant poet typecasting. Works considered include Scottish Pastorals (1801), The Mountain Bard (1807 and 1821), The Queen's Wake (1813), The Poetic Mirror (1816), The Royal Jubilee (1822), Queen Hynde (1825), Pilgrims of the Sun (1815 and 1822) and A Queer Book (1832). Part Three discusses Scottish autodidacticism as it developed after Hogg, discerning subgroups within the peasant poet category.

A self-educated shepherd, Scottish writer James Hogg (1770-1835) spoke from a position outside the dominant discourse, depicting issues of his age related to gender, class, and ethnicity by giving voice to people from the margins and, thus (either consciously or unconsciously), revealing gender politics and Britain's imperial aims. Hogg’s contemporary critics received his work rather negatively, viewing his subjects such as prostitution, out-of-wedlock-pregnancy, infanticide, and the violence of war as violating the principles of literary politeness. Hogg’s obstinacy in addressing these issues, however, supports the thesis that his aim was far more significant than challenging the expectations of his contemporary readers. This project shows that pragmatics can be applied productively to literature because its eclecticism offers the possibility of developing a detailed discussion about three aspects of literary communication—the author, the reader and the text—without prioritising any of them. Literature is an instance of language in use (the field of pragmatics) where an author creates the texts and a reader recreates the author’s message through the text. Analysis of Hogg’s flouting of Grice’s maxims for communication strategies and of his defying the principles of politeness enables a theoretically supported discussion about Hogg’s possible intentions, as well as about how his intentions were perceived by the literary establishment of his time; while both relevance theory and Bakhtin’s socio-linguistics enriched by a historically contextualised politeness shed new light on the negative reception of Hogg’s texts.

Hog fuel is increasingly becoming an alternative to alleviate the energy problems associated with the use of fossil fuels. To make adequate use of hog fuel, its moisture content should be reduced prior to combustion either in an external dryer or in the initial stages of a hog fuel boiler. Therefore, this research project was undertaken to establish the factors which govern the drying rate of wet hog fuel particles. The convective drying of wood-waste on the slow moving bed of hog fuel boilers was simulated in a packed bed. The information which was obtained can also be applied to approximate the drying behaviour in external dryers. An apparatus was constructed to accommodate the use of hot air, flue gas, superheated steam and a mixture of them as drying media. Drying tests were carried out, over the temperature range of 125-245°C, on 1.1 to 4 kg batches of Western Hemlock hog fuel of thicknesses from 2 to 12 mm. The relative effects of velocity (V), temperature (T), nature of the drying gas, bed depth (L), and initial moisture content of the hog fuel samples (M₀) on the drying process were investigated using a mixture of several thickness fractions having an average (sauter mean) particle thickness (dp) of 6.3 mm. Drying rates were determined through measurement of the change either in humidity of the drying gas, or flow rate of the superheated steam across the bed of hog fuel. Gas humidity was measured using an optical dew point sensor and steam flow was monitored using an orifice plate connected to a massflow transmitter. Drying rates have been quantified as functions of hog fuel particle thickness, initial moisture content and bed depth. The effects of gas temperature, velocity and humidity have also been quantitatively established. The drying process was insensitive to CO₂ content of the drying gas. The existence of an inversion temperature above which drying rates increase with humidity of the drying medium was both experimentally and theoretically confirmed and the locus of inversion points was determined. Instantaneous normalized drying rates, ƒ, and characteristic moisture contents, Φ , have been determined and the existence of a unified characteristic drying rate curve was verified. Using a receding plane model, ƒ was formulated as a function of Φ, for dp = 6.3 mm and at L = 25 cm, for both superheated steam and relatively dry air. Pressure drop measurements were obtained for all the runs with the exception of the superheated steam ones. Application of an accepted pressure drop equation permitted the sphericity of the hog fuel particles to be approximated. A design equation for gas pressure drop in beds of hog fuel particles was investigated. The simultaneous heat and mass transfer processes in drying during the heat transfer controlled period was studied. Using the concept of volumetric evaporation, an empirical correlation for the overall heat transfer coefficient in a packed bed of hog fuel particles has been obtained. The effects of different parameters on both the particle residence time required for drying and the grate heat release rate in hog fuel boilers were determined.

This thesis investigates the representation of the supernatural in the literature of Walter Scott and James Hogg. In comparing both authors it takes advantage of two recent scholarly editions: the Stirling/South Carolina edition of Hogg and the Edinburgh Edition of the Waverley Novels. I trace the development of Scott’s persistent interest in various categories of the supernatural: the uncanny; witchcraft; second sight; and astrology. His literary career began in 1796 with translations of German Romantic poetry. These were followed by publication of his collection of ballads and folklore, known as the Minstrelsy of the Scottish Border, 1802-3, and by the longer poems such as The Lay of the Last Minstrel, 1805. Subsequently, Scott’s investigation of the supernatural would continue within a number of key novels and his shorter fiction. The Letters on Demonology and Witchcraft, addressed to J. G. Lockhart, Esq., 1830, was one of his final attempts to establish how far the evidence of a credible witness might supply ineluctable testimony in accounts of the supernatural. Scott’s legal training, and antiquarian skills, lent particular authority into his investigations of the possibilities of the existence, or otherwise, of the supernatural. By way of contrast, James Hogg’s lack of formal education, and scanty knowledge of the progressive advances of the Scottish Enlightenment, was associated with a ready credulity in matters of the supernatural. His literary work, such as The Mountain Bard, 1807, or his later collection of Winter Evening Tales, 1820, demonstrated a familiarity with ballads, and an unlettered folklore tradition, that appeared to confirm his position as a believer in superstitious and irrational practices. However, this thesis will argue that Hogg actually possesses a shrewd and sophisticated understanding of the authority of the supernatural. This is manifest in his literary efforts to record and investigate various types of uncanny testimony, when compared with those of Scott. Hogg’s view of the supernatural is complex and essentially subversive. His final novel, The Private Memoirs and Confessions of a Justified Sinner, 1824, and his later contributions to the fashionable annuals and giftbooks published between 1826 and 1834, reveal an author deeply engaged with demonstrating the unique role of the supernatural within Scottish society, particularly as a channel of dissent and discord. The Ettrick Shepherd and the Author of Waverley founded their literary relationship upon a shared enthusiasm for the supernatural tales and traditions of the Scottish Borders. Their friendship was both competitive and complementary. Critics have generally tended to assume that Scott, rather than Hogg, was the sceptical party where belief in the existence of the supernatural is concerned. However, closer examination of their work reveals that such assumptions do not necessarily stand up. Ultimately, Hogg emerges as the author with greater resistance to an irrational belief in the supernatural. His position as an observer, and critic, of the antiquarian and enlightened literary establishment, with its dependence on the authority of printed texts, is developed through his literary investigation of the supernatural. My choice of works to consider has been necessarily limited by questions of space. Where possible, I have selected those texts that seem to me to offer ready comparison between the two authors. Some novels such as Scott’s The Antiquary, 1816, or The Pirate, 1822, might be regarded as worthy of inclusion in this study of the supernatural. However, there are no real equivalents of these in Hogg’s work.

This study aimed to explore the differences of black hog and white hog prices evaluated by professional agents. By using the data from the auction market in Miaoli, this study analyzes daily price volatility and relationship of auction prices between these two different species. The appearances of black hog and white hog have obviously physical differentials. However, previous studies mostly focus on the differences in hog auction prices, or differences caused by individual hogs. Few studies have aimed at price volatility associated with different hog breeds. This study applied BEKK model from multivariate GARCH model also reported additionally the coefficient from constant conditional correlation models. The average prices of black hog and white hog came from daily sequential random-ordering auctions. Data was collected from January 2009 to December 2013. Total 1,485 auction days are included. The results indicated that both previous black hog and white hog prices had significant impacts on the volatility of their current prices individually. Secondly, the prices of black hog and white hog did not affect each other. Third, the structures of price volatility of the black hog and white hog are different. White hog’s price factors of volatility are greater than the non-price volatility factors; alternatively, the non-price factors on black hog’s volatility are greater than the price volatility factor. Finally, the relationship of price volatility between black hog and white hog is relatively small. The results not only provide the government to develop proper policy for assisting hog farmers and upgrading the hog industry, but also help stakeholders as a marketing guide and a reference to realize domestic consumers’ pork preferences.

The following part deals with young people diction and its specifics. The next part contains the historie and contemporary description of the hip hop subculture. The rest of the thesis is dealing with verbal analysis of particular hip hop songs as well as with album reviews and finally with discussion forums referring to this specific genre/subculture on the internet. The goal of my thesis is to describe and illustrate the specific language features of all those text types.

Title: Analysis of softball throw speed Objective: The aim of the thesis is to compare the throwing speeds with different modifications - from a distance of 2 meters to the screen (without aiming), throwing at a distance of 18 meters after the ball has been caught from the air and the ground (throw to the teammate). Furthermore, the goal of comparing throw speeds in selected age categories of softball players (9-17 years). Methods: Measure the throw speed by radar and data processing in Microsoft Excel. Results: Speed of throws for individual modifications - the category of the oldest players recorded the fastest throw option with the aiming of the player after the previous capture of the ball from the air and after the previous ball was caught on the ground. These variations of the throw exceeded the throw speed without prior capture and also without aiming at the player. In contrast, for other categories, the fastest option was the no-aim goal. The results of the oldest age group therefore refuted the first hypothesis, but the results of other age categories confirmed the hypothesis. In the case of the second hypothesis - higher throw speed and processing when the ball is caught from the air compared to landing the ball - the results of the oldest and youngest age category confirm the hypothesis,...
