Abstract:
Leishmaniasis is a disease caused by obligate intracellular parasites of the genus Leishmania, including 20 species that are pathogenic to humans. Female sand fly is the known vector that can transmit the disease. Visceral leishmaniasis is the severe form of the disease that affects internal organs and can be fatal with inappropriate diagnosis or treatment. Leishmania donovani is the causative agent of visceral leishmaniasis. Approximately 350 million in 89 countries are at risk of infection. Around 2 million new cases are reported annually with 500,000 of these are visceral leishmaniasis. Current drug therapies are inadequate due to their toxicity, high cost, severe adverse reaction, limited availability, and the emergence of resistance. With all these limitations, the need for new drugs is urgent. Pentavalent antimonials are the first line of treatment for leishmaniasis since the 1940s. Although amphotericin B, pentamidine and paromomycin are current drugs that treat leishmaniasis, they were discovered initially as a treatment for other pathogens. Furthermore, miltefosine the only available oral drug for leishmaniasis is an anticancer drug that found to be active against Leishmania. Therefore, we used our quantitative Leishmania donovani axenic amastigote assay and the clinically relevant infected macrophage assay to identify new antileishmanial hits from unstudied or understudied natural product sources such as mangrove endophytic fungi, Antarctic deep-sea coral, and terrestrial plants. We also used the same assays to screen synthetic compounds form multiple chemical scaffolds. Our well-established assays led to the identification of new antileishmanial hits from unstudied natural products and the discovery of new classes of molecules from synthetic compounds that possess potent activity against Leishmania donovani. Finally, we conducted an in vivo hamster study on an active hit that revealed high efficacy against Leishmania donovani in this severe model leading to promising antileishmanial drug development.

This collection of poems and prose poems is a record of my life, from growing up in the small town of Bindura in Zimbabwe to landing in the small town of Grahamstown in South Africa. It is a story of dislocation: physical, emotional, and political. It is a story about borders and boundaries; although not rich, I was raised among the rich; although not white, I spent a considerable part of my childhood in white spaces. These fragments are told through the voice of my younger self and also retrospectively by my adult self. Two important influences on my style and my struggle to write about my identity have been Woman Hollering Creek by Latino American author Sandra Cisneros and Citizen: An American Lyric by US poet Claudia Rankine.

Abstract:
When the Sun Hits is a feature length film concerned with the delicate relationship between the individual and the whole. A young man named John has fallen into a state of social paralysis since his father's death. He no longer knows how to relate to others and the world around him. Rather than looking to friends and family for support he closes himself off. After the mysterious disappearance of his mother, John blames himself for her leaving. He tries his best to reconnect with his old life, but finds himself making the same mistakes over and over. When his friend Tracy reaches out to him, he bitterly rejects her and once again finds himself alone. It is then that John puts his old life behind him and sets off to make amends with his mother. John has no idea where his mother might be or if she will accept him. Nonetheless, he reaches into the abyss to see if someone is there, waiting for him. The structure of the film is nonlinear as it deviates from traditional narrative form. The intent of this thesis is to serve as a post-film reflection that discusses the relation between my views on cinema and the film I made. The paper will shift between the general themes of the narrative and the film theory working behind it all. It will explore the idea of cinema as an extension of the mind and how this is present in the film. By using specific examples from the film this theory of cinema can be fleshed out and an insight into my process may be gleaned.

Abstract:
This report is a study if a multi linear regression could be used to predict the cap hit of hockey forwards from the NHL. Data was collected during the 2010-2011, 2011-2012, and 2012-2013 seasons. The chosen variables were common hockey statistics and a few none hockey-related, like origin and age. The initial model was improved by removing insignicant covariates, detected by BIC-test and p-values. The final model consisted of 291 players and had an adjusted R2-value of 0,7820. Of the covariates, goals, assists and ice time had the biggest impact on a player's cap hit.

Buscamos estrellas tipo RR Lyrae en zonas distantes del halo de la Vía Láctea utilizando datos ya existentes: UltraVISTA, una campaña con observaciones en el infrarojo cercano diseñada para obtener imágenes profundas del campo COSMOS, y SUDSS, una extensión del Dark Energy Survey (DES) que añade observaciones en los extremos del tiempo de obser- vación de DES en varios filtros, con la finalidad de observar las curvas de luz de supernovas a alto redshift. Los resultados de la búsqueda de RR Lyrae con estos datos fueron nulos. Ante estos resultados realizamos observaciones simuladas de RR Lyrae, bajo las mismas condi- ciones de los datos observados, demostrando que la cadencia utilizada en las observaciones no permite la identificación del período de las estrellas RR Lyrae. Finalmente probamos el método desarrollado con datos de HiTS (High Cadence Tran- sient Survey), una campaña dedicada a detectar transientes en el óptico, principalmente las primeras horas de las explosiones de supernovas. Luego de responder favorablemente a las observaciones simuladas, logramos identificar en los datos de HiTS 2015A un total de 106 RR Lyrae 7 de las cuales se encuentran a una distancia mayor a 100 kpc.

Abstract:
Micro-RNAs (miRNAs) have been discovered to regulate messenger RNA (mRNA) translation and degradation. Various recent studies have been focused on miRNA target prediction, in order to get a better understanding of the rules and nature of miRNA regulation over mRNAs. In this project we aim to create a software module to identify miRNA target sites on mRNAs. As basis to this project, we refer to a study that identified a platform for miRNA-mRNA interaction in protein-RNA complexes in mouse brain (AGO HITS-CLIP study). We propose a probabilistic model of the data from this study, and generate synthetic sample data according to this model, in order to create a test bed for a discovery module. Our discovery module analyzes the sample data to identify peak regions where the interaction density is high. We present results both on synthetic sample data and data from the AGO HITS-CLIP study to evaluate our module.

An experiment was carried out in order to determine the frequencies experienced in two locations on the stick during two different field hockey hits, the push and the strike. The results from these experiments showed that the top hand during a hit experiences a higher resonant frequency. During the push the top hand experienced an average resonant frequency of 349.1 Hz, compared to the lower hand which experienced an average resonant frequency of 43.95 Hz. During the strike the top hand experienced an average resonant frequency of 197.8 Hz, compared to the lower hand which experienced an average resonant frequency of 24.41 Hz. A second experiment was carried out in order to determine how effective the grip was at dissipating frequencies. The results determined that the grip is most effective over the frequencies of 117 Hz-470 Hz. Both an old and new grip were tested. The new grip was slightly more effective over the frequencies of 117 Hz - 235 Hz, and similar over the frequencies 250 Hz - 470 Hz. From these experiments it was concluded that field hockey grips are most needed and effective on the shaft of the field hockey stick.

Abstract:
The thesis investigates a connection between the publishing industry and the film industry. It analyses whether films based on best-selling books have higher revenues and return on investment compared to films based on unsuccessful books or original scripts. Data for years 2009 -- 2015, focusing on the U.S. entertainment industry, show that film adaptations of best-selling books reach higher revenues during their opening weekend of the theatrical release. The Bestseller Effect weakens once the information about film's quality becomes available to the audience. The success of a book plays negligible role in film's returns on investment. The results of analysis also show great positive significant impact of the production budget, the star cast, films quality and sequels on the films'financial performance.

Abstract:
Whereas it has been demonstrated that sustaining multiple concussions can detrimentally affect neurophysiologic health and cognitive abilities, there is no consensus on the cumulative effect of repetitive subconcussive head impacts in contact sports. Subconcussive hits are head impacts that are typically less forceful than concussive impacts. They occur much more frequently, and do not produce obvious or immediate clinical symptoms. The global objective of this research was to investigate how cumulative exposure to repetitive subconcussive hits, as experienced through participation in one season of competitive American football, affects response inhibition, one of many executive functions of the brain. In addition, accelerometers were used to collect biomechanical data to determine the cumulative number and magnitude of these repetitive head impacts and relate this to changes in response inhibition. Response inhibition was probed with the use of a complex neurocognitive sensorimotor KINARM hit-and-avoid task. This required the subject to use robotic arms to respond to virtual targets while avoiding multiple distractors presented simultaneously on a 2D virtual screen. The results showed a significant difference between the contact and non-contact sport athletes from pre-season to post-season, and therefore support this hypothesis. In addition, it was hypothesized that contact-sport athletes who experienced a greater cumulative number of head impacts over a season would show significantly poorer performance on the task of response inhibition compared to the contact-sport athletes who experienced a lesser cumulative number of head impacts. It was also hypothesized that contact-sport athletes who experienced a greater cumulative magnitude of head impacts over a season would show significantly greater poorer performance on the task of response inhibition compared to those athletes who experienced a lesser cumulative magnitude over the season. Interestingly, the results fail to support this hypothesis, and instead appear to support the opposite hypothesis. The findings from this thesis indicate that experiencing a season’s worth of subconcussive hits does appear to affect the specific executive function of response inhibition in contact-sport athletes, however other factors besides head impact data may play an important role in these adverse effects.

Understanding the molecular basis of human disease is one of the greatest challenges of our time, and recent explosion in genetic and genomic datasets are finally putting it within reach. In the last ten years, genome-wide association studies have identified thousands of genetic variants associated with disease. However, the majority of these variants fall outside genes making interpreting their role in disease difficult. In parallel, the ENCODE and Roadmap Epigenomics consortia have produced high resolution annotations of the genome which identify large portions with potential regulatory function. We develop methods to interpret genome-wide association studies using these annotations to generate hypotheses about how associated variants contribute to disease mechanism. In particular, we go beyond the usual stringent p-value threshold to investigate variants with small individual effect sizes which current methods do not have power to detect. Evaluating our methods on the Wellcome Trust Case Control Consortium 7 Disease studies, we find associated variants are enriched in a variety of functional categories even after controlling for various biases. We also find an unprecedented number of variants contribute to this enrichment, supporting our hypothesis that the architecture of these diseases involves combinatorial interaction of many variants with small individual effect sizes.

This paper addresses the recent concerns about concussions in the sport of football and elaborates on the medical findings, litigation, and ethical questions that have surrounded the issue. The goal is to present a compelling case for change in how concussions are viewed and handled in the sport. By using concrete examples to explain the lasting effects concussions have had on players after their careers have ended, I hope to show the need for change. I will explore the most recent developments of chronic traumatic encephalopathy in order to show its harmful reach and will touch upon litigation that has been filed by players who experienced post career problems. In addition, through the insight of current college players as well as referencing my own experience as a Division I College Football player, I wish to establish an emotional connection in the paper and unveil the roots of the problem – the toxic nature of the football culture. I am optimistic that this inquiry will help cultivate a culture change through a variety of approaches. First, I indicate the need for a transformation of the football culture. Second, I suggest a formal, mandatory education to inform players at all levels about concussions, subconcussive hits, and the potential diseases that can stem from. Third, I propose new penalties for players, coaches, support staff, and all involved in player safety in an attempt to further prevent head injuries. This thesis attacks the issue of concussions in football from all angles. It calls for the football community to accept the severity of concussions, educate on concussions, and prevent repeated concussions in order to prompt action

Abstract:
Prior to this project, a high throughput assay was developed in order to perform automated RNAi screens with siRNA- libraries targeting potential regulators of the G2 checkpoint. The libraries were covering the human kinases, phosphatases and DNA repair. The aim of this project was to validate the candidate hits from the phosphatome screen as possible G2 checkpoint regulators. To validate the candidate hits, esiRNAs were applied in order to down regulate the target proteins, and G2 checkpoint abrogation was assayed by flow cytometry analysis. To confirm that the assay did work, the effects of inhibiting WEE1 by the small molecule inhibitor, MK1775, were tested in both U2OS and U2OSp53 cells. Both the early and the late G2 checkpoint were tested for.WEE1 was a hit in the kinome screen, and a well-known regulator of the G2 checkpoint. The samples treated with MK1775, and 6Gy were showing a dose-dependent abrogation of the G2 checkpoint when stained for H3-p and analyzed by flow cytometry.In optimization experiments for esiRNA transfection, Effectene was chosen over Oligofectamine as the preferred transfection reagent. The transfection conditions which were decided to be the most efficient in down regulating the target protein were 5μL esiRNA and 10 μL Effectene. For the validation experiments it was focused on the two phosphatases SSH3 and PTPN7. Western blotting analysis showed that the protein level of SSH3 was reduced to less than 50% at two days following transfection with SSH3 esiRNA.The validation experiment did not show any abrogation of the G2 checkpoint by neither SSH3 nor PTPN7 in irradiated cells. More repetitions of the experiment are needed in order to validate- or reject these as false hits. However, the results from a whole-genome DNA damage response siRNA screen were recently published. Neither PTPN7 nor SSH3 were scoring in this screen. These facts are supporting the results that SSH3 and PTPN7 are false hits.

O ambiente baseado em hyperlink possui na sua topologia informações substanciais sobre o seu conteúdo. Baseado nesse tipo de ambiente, Jon Kleingerg desenvolveu um conjunto de algoritmos, popularmente conhecido como HITS (Hyperlink Induced Topic Search), que utiliza a estrutura de hyperlinks na WWW para extrair essas informações. O foco central desses algoritmos é a classificação de tópicos de busca de caráter geral na WWW, através da descoberta de páginas que representam autoridade sobre tais tópicos. Para tanto, os algoritmos formulam a noção de autoridade considerando o relacionamento, decorrente da estrutura de hyperlink, entre o conjunto de páginas que são autoridades relevantes e o conjunto de páginas que apontam para essas, denominadas de hubs. Jon Kleingerg definiu, portanto, uma relação de interdependência entre os conjuntos anteriormente citados: uma boa autoridade será uma página apontada por bons hubs e um bom hub será uma página que aponta para boas autoridades. Neste trabalho, propomos a extensão do modelo formulado por Jon Kleingerg, através da inserção de novos conceitos nas relações de interdependência entre autoridades e hubs. Assim, formulamos um algoritmo estendido, XHITS (Extended Hyperlink Induced Topic Search), que visa melhorar a classificação das autoridades do ambiente. Nessa extensão as autoridades são apontadas por bons hubs, às vezes apontadas por bons portais e também apontam para boas novidades. Os bons hubs são páginas que apontam para boas autoridades e novidades, e são apontados por bons portais. The network structure of a hyperlinked environment can be a rich source of information about the content of this environment. Jon Kleinberg developed a set of algorithms, called HITS (Hyperlink Induced Topic Search), for extracting information from the hyperlink structures of those environments. The aim of these algorithms is the distillation of broad search topics, through the discovery of related authoritative information sources. The notion of authority is based on the hyperlink structure relationship between a set of relevant authoritative pages and the set of hubs. Thus, hubs and authorities exhibit what could be called a mutually reinforcing relationship: a good hub is a page that points to many good authorities; a good authority is a page that is pointed by many good hubs. In this work, we present the XHITS (Extended Hyperlink Induced Topic Search) algorithm, an extension of the HITS algorithm by introducing new concepts on the mutually reinforcing relationship. In XHITS, a good authority is a page that is pointed by many good hubs, some good portals and points to good novels; a good hub is a page that points to many good authorities, some good novels and is pointed by some good portals; and a good novel is a page that is pointed by good authorities, some good hubs and some good portals; a good portal is a page that points to some good authorities, some good hubs and some good novels. In addition, we show that XHITS converges and, through some experiments, the improved quality of the hyper documents retrieved.

Immune heparin-induced thrombocytopenia (HIT) is a potentially serious complication of heparin therapy and is associated with antibodies directed against a complex of platelet factor 4 (PF4) and heparin. Early diagnosis of HIT is important to reduce morbidity and mortality. I developed an enzyme immunoassay that detects the binding of HIT IgG to PF4-heparin in the fluid phase. This required techniques to purify and biotinylate PF4. The fluid phase assay produces consistently low background and can detect low levels of anti-PF4-heparin. It is suited to testing alternative anticoagulants because, unlike in an ELISA, a clearly defined amount of antigen is available for antibody binding. I was able to detect anti-PF4-heparin IgG in 93% of HIT patients. I also investigated cross-reactivity of anti-PF4-heparin antibodies with PF4 complexed to alternative heparin-like anticoagulants. Low molecular weight heparins cross-reacted with 88% of the sera from HIT patients while half of the HIT sera weakly cross-reacted with PF4-danaparoid (Orgaran). The thrombocytopenia and thrombosis of most of these patients resolved during danaparoid therapy, indicating that detection of low affinity antibodies to PF4-danaparoid by immunoassay may not be an absolute contraindication for danaparoid administration. While HIT patients possess antibodies to PF4-heparin, I observed that HIT antibodies will also bind to PF4 alone adsorbed on polystyrene ELISA wells but not to soluble PF4 in the absence of heparin. Having developed a technique to affinity-purify anti-PF4-heparin HIT IgG, I provide the first estimates of the avidity of HIT IgG. HIT IgG displayed relatively high functional affinity for both PF4-heparin (Kd=7-30nM) and polystyrene adsorbed PF4 alone (Kd=20-70nM). Furthermore, agarose beads coated with PF4 alone were almost as effective as beads coated with PF4 plus heparin in depleting HIT plasmas of anti-PF4-heparin antibodies. I conclude that the HIT antibodies which bind to polystyrene adsorbed PF4 without heparin are largely the same IgG molecules that bind PF4-heparin and thus most HIT antibodies bind epitope(s) on PF4 and not epitope(s) formed by part of a PF4 molecule and part of a heparin molecule. Binding of PF4 to heparin (optimal) or polystyrene/agarose (sub-optimal) promotes recognition of this epitope. Under conditions that are more physiological and sensitive than previous studies, I observed that affinity-purified HIT IgG will cause platelet aggregation upon the addition of heparin. Platelets activated with HIT IgG increased their release and surface expression of PF4. I quantitated the binding of affinity-purified HIT 125I-IgG to platelets as they activate in a plasma milieu. Binding of the HIT IgG was dependent upon heparin and some degree of platelet activation. Blocking the platelet Fc??? receptor-II with the monoclonal antibody IV.3 did not prevent HIT IgG binding to activated platelets. I conclude that anti-PF4-heparin IgG is the only component specific to HIT plasma that is required to induce platelet aggregation. The Fab region of HIT IgG binds to PF4-heparin that is on the surface of activated platelets. I propose that only then does the Fc portion of the bound IgG activate other platelets via the Fc receptor. My data support a dynamic model of platelet activation where released PF4 enhances further antibody binding and more release.

Tal adaptação trouxe, mais uma vez, Eça de Queirós para o centro do espaço cultural brasileiro, tornando seu livro mais conhecido novamente campeão de vendagem. O objetivo deste trabalho é fazer uma leitura comparativa entre as duas obras, o texto e sua transcodificação, levantando algumas questões, como a relação existente entre a literatura e a televisão, o papel do leitor e do telespectador e a diferença entre cultura erudita e cultura de massa. Pretende-se também analisar como os elementos básicos da narrativa textual apresentam-se na narrativa televisiva. Destaca-se a importância de ambos os meios em que as obras se apresentam, a televisão e o livro, bem como o papel fundamental que cada um exerce sobre o seu público no seu tempo.
This article intends to accomplish an analysis of the serie O Primo Basílio, produced and transmitted by Rede Globo, in comparison with the literary text which originated the serie. In 1988, the Rede Globo showed O Primo Basílio. This adaptation brought, again, Eça de Queirós to the center of the Brazilian cultural scene, turning his book into best seller. The purpose of this monograph is to make a comparative analysis between the two works of art, the text and its transcodification, lighting up some questions, like the relation existent between literature and television, the functions of the reader and of the viewer, and the difference between high culture and mass culture. It intends to analyze also how the basics elements of the textual narrative appears on the television serie, and highlight the importance of both medias in which the work of art appear, television and book, and the fundamental role each one exerts on its public in its time.

Contudo, dois análogos (48 e 52) apresentaram atividade inibitória das cepas de Plasmodium falciparum, com CI50 = 5,0 µM e índice de seletividade bastante interessante (IS >50).

Ethnicity is one of the powerful concepts in social sciences. It encourages social scientists coming from different academic disciplines to think over its roles, influences and power within communities which are shaped at various level. Particularly in the globalised world, traditional approaches such as primordalism, essentialism or instrumentalism have difficulty to provide a powerful framework to understand complexity and power of ethnicity in diasporic spaces which host different identity formations, experiences and cultural hybridisation. Accordingly, these traditional approaches miss differences among human beings who associated themselves with certain ethnicities. For this reason, social scientists tend to understand the concept of ethnicity with alternative approaches. Unlike traditional approaches, social constructionism does not seen ethnicity as fixed, stable homogeneous things. Rather, ethnicities refer to a cognitive process which is shaped by people’s attitudes, perceptions or interactions. Approaching to ethnicity as a cognitive process allows us to go beyond universality and sharp definitions of ethnicity. Also, it emerges out various interpretations in diasporic spaces where ethnicity can be reproduced in various ways. By relying on this theoretical framework, this research seeks to understand the reproduction of Armenianness in diasporic spaces. Through focusing on Turkish, Lebanese and British Armenians, it searches answers for following questions a) What are the components of Armenianness in Turkey, Lebanon and Britain? b) How are the components of Armenianness interpreted in diasporic communities in Turkey, Lebanon and Britain? c) How is Armenianness reproduced among Armenian youngsters in Turkey, Lebanon and Britain? It is hypothesised that Armenianness is not a holistic form, it can be observed as “a patchwork” consisting of various patterns and colours. In order to materialise research aims and goals, the research was supported by field works in Istanbul, Beirut and London between 2011 and 2013. Throughout the data collection period, in-depth interviews, ethnography and participant observation were preferred to not only identify components of Armenianness, but also to create datasets for comparing and analysing the cases. The datasets have been analysed by ANCO-HITS to demonstrate similarities and differences among various reproduction forms of Armenianness in numerical ways. As a result of the ANCO-HITS analysis, Armenianness was ranked in each case according to participants’ scores. Later on, two participants (having the most negative and positive) were introduced through referring findings and fieldwork notes which derived from ethnography. This research shows that Armenianness is observed in various forms. It is highly heterogeneous in diasporic spaces and experienced in different ways. Interpretations of youngsters are varied. It sometimes seems to be ethnic, nationalist, political, moderate or congregational. It also demonstrates that attitudes, perceptions as well as interactions of youngsters with Armenians and non-Armenians can be effective parameters differentiate Armenianness in diasporic spaces.

Chlamydia trachomatis infection targets the mucosal epithelium, where squamous and columnar epithelia can be found. Research on Chlamydia trachomatis-epithelia interaction has predominantly focused on columnar epithelia, with very little known on how Chlamydia trachomatis interacts with the squamous epithelium. The stratification and differentiation processes found in the squamous epithelium might influence chlamydial growth and infection dissemination. For this reason, 3D stratified squamous epithelial cultures were adapted to mimic the stratified squamous epithelium, and chlamydial infection was characterized. Chlamydia trachomatis infection in monolayers and 3D cultures were monitored by immunofluorescence and transmission electron microscopy to characterize inclusion growth and chlamydial interconversion between elementary and reticulate body. We observed that the stratified epithelium varied in susceptibility to Chlamydia trachomatis infection. The undifferentiated basal cells were susceptible to infection, while the terminally differentiated upper layers were resistant. If given access to the basal layer Chlamydia trachomatis is able to disseminate and disrupt the epithelial. This disruption have clinical relevance, such as facilitating secondary infection by other STIs. The use of a punch biopsy in 3D cultures revealed that infected samples were unable to close the wound as efficiently as the mock-infected sample. A simplified 2D wound healing assay confirmed these observations. Additionally, this correlated with a reorganization of hemidesmosomes in Chlamydia trachomatis-infected cells but, most importantly, in bystander uninfected cells within the infected sample. The lack of motility and the hemidesmosomes reorganization was shown to be dependent on myosin II contractility and the chlamydial protein CTL0480. This chlamydial protein recruits MYPT1 to the inclusion membrane, which could potentially prevent the cell from controlling the actomyosin tension. In summary, this is the first study to use a 3D stratified epithelial to determine how Chlamydia interacts with this physiologically relevant tissue. Most importantly, this work demonstrates that Chlamydia trachomatis is able to alter the organization of hemidesmosomes which has never been reported for any other bacterial pathogen.

El Survey de Transientes a Alta Cadencia (HiTS), fue creado con el propósito de descubrir y estudiar objetos transitorios con escalas de tiempo características entre horas y días, tales como, estrellas pulsantes, eclipsantes, explosivas, y variables estocásticas no estelares. Este survey representa una oportunidad única para explorar observaciones con \textit{entendue} grande y cadencias de observación cercanas a $0.1$ días utilizando nuevas herramientas computacionales para el análisis de grandes volúmenes de datos. Este trabajo sigue un enfoque de \textit{Ciencia de los Datos}: partiendo desde los datos sin procesar hasta el análisis y clasificación de fuentes variables. Hemos compilado un catálogo de $\sim$14 millones de fuentes detectadas y un catálogo de $\sim$2.5 millones de curvas de luz clasificadas según su variabilidad. La profundidad de observación típica de nuestro survey es de $24.2$, $24.3$, $24.1$ y $23.8$ magnitudes en las bandas \textit{u}, \textit{g}, \textit{r} e \textit{i}, respectivamente. En este trabajo presentamos la primera versión de los catálogos. Clasificamos las fuentes puntuales y estáticas extrayendo primero características de sus curvas de luz y luego aplicando un clasificador Random Forest. Para la clasificación utilizamos un conjunto de entrenamiento construido utilizando una combinación de cros referencia con catálogos públicos, inspección visual, transferencia de aprendizaje/aprendizaje activo, y datos aumentados sintéticamente. El modelo de clasificación consiste en varios clasificadores Random Forest organizados en un esquema jerárquico. La precisión del clasificador estimada en el conjunto de prueba es aproximadamente $97\%$. En los datos no etiquetados, $3\,485$ fuentes se clasificaron como variables, de las cuales $1\,321$ se clasificaron como periódicas. Los agujeros negros de masa intermedia (IMBH) ($ M_{BH}$$\sim$$10^{2-5} M_\odot$) son claves para el entendimiento de la formación de agujeros negros (BH) masivos. La población actual conocida de IMBH es pequeña, con algunos cientos de candidatos y solo unos pocos de ellos con una caracterización segura. En este trabajo proponemos una nueva metodología para seleccionar candidatos a IMBH utilizando análisis de variabilidad. Nuestro método de selección se centra en la variabilidad en la región nuclear de galaxias locales. IMBH activos acretando materia a bajas tasas muestran variabilidad a pequeñas amplitudes y en escalas de tiempo de horas. HiTS presenta una oportunidad única para realizar este tipo de búsqueda. Encontramos $\sim$500 galaxias que evidencian este tipo de variabilidad. Estimamos una fracción de ocupación del $4\%$ y una densidad de número superficial de $3$ grados$^{-2}$. Esto representa un aumento en un factor 40 en comparación con búsquedas anteriores basadas en técnicas espectrales. Nuestro método proporciona núcleos variables de galaxias que son candidatos para alojar IMBHs. Sin embargo, no se puede estimar la masa del BH a partir de nuestros datos. Para esto necesitaremos observaciones en rayos X y radio para confirmar la naturaleza AGN de nuestras fuentes y poder estimar su masa utilizando el plano fundamental de BH activos.

Abnormally high energy events were seen in the Hadronic Forward (HF) calorimeter for pion and muon data during testbeam in 2004. Analysis of testbeam data suggested that such events were caused by particles traveling the entire length of HF and striking the photomultiplier (PMT) windows in the readout box behind HF. Charged particles traversing the window of the PMT emit cerenkov radiation, which creates abnormally high energy events in the data. To further study these events, a modification of the existing official CMS HF simulation was created that added the PMT windows to the simulation as sensitive detectors. In agreement with testbeam data, abnormally high energy events in the PMTs were seen in the simulation for muons and pions. The simulation was then extended to jets simulated with Pythia, and then for collision like events as well. PMT hits were seen in both of these cases. Energy sharing between PMTs for long and short fibers in HF as well as timing differences between normal HF events and PMT events were investigated as methods to tag such abnormal events. While both methods were somewhat successful, it was determined that they were not sufficient. The simulation was also modified to use thinner PMT windows. Reducing the thickness of the window reduced the number of PMT hits, and drastically reduced the energy of these hits, bringing most of them below standard jet energy thresholds. These results led to the replacement of the existing PMTs with new PMTs with a smaller, thinner window. Higgs mass reconstruction methods were applied to Monte Carlo datasets for 115 and 130 GeV Higgs produced through vector boson fusion. In these datasets, the Higgs boson decayed to two tau particles, each of which decayed leptonically. The mass reconstruction methods successfully created a peak at the proper mass for both datasets. In addition to creating a Higgs, the vector boson fusion signal also has two forward jets. These jets are not found in the signal of the dominant background processes. By applying forward jet cuts to both the Higgs and the background events, it was possible to drastically improve the signal to background ratio for this channel. The Higgs boson was recently discovered at the LHC with a mass of approximately 125 GeV. As the methods discussed in this study are appropriate only for a low mass Higgs as the one discovered, they may prove useful in more precisely determining the mass of the Higgs.

Intimate partner violence (IPV) during pregnancy can lead to a myriad of poor physical and psychological outcomes for both mother and child. There is a paucity of research examining IPV risk factors for rural pregnant women and on information regarding the course of the specific types of IPV throughout pregnancy. The current project was an investigation of the prevalence of IPV and IPV risk factors for different types of IPV in an Appalachian pregnant sample that contained women from both rural and nonrural locations (Study 1), and was an examination of the occurrence of any IPV and the different types of IPV throughout the course of pregnancy (Study 2). Study 1 included 1,063 pregnant women participating in the Tennessee Intervention for Pregnant Smokers (TIPS) research project. IPV prevalence rates during pregnancy, measured using a modified HITS IPV screen, were approximately 26% for psychological violence, 2% for physical violence, and 1% for sexual violence. Chi-squared analysis indicated that rural pregnant women were not significantly more likely to experience any of the types of IPV compared to nonrural pregnant women. Additionally, logistic regression analysis supported previous literature findings that pregnant women who are unmarried, younger, have an unplanned pregnancy, have high levels of stress, and have low levels of social support are at a greater risk of experiencing any type of IPV during pregnancy compared to pregnant women not possessing those risk factors. However, rural status was not a significant predictor or modifier of IPV. Study 2 participants included a subsample of 337 pregnant women who indicated they had experienced IPV at any time during the course of their pregnancy. Generalized estimating equation logistic models indicated that women who experienced IPV at some point during pregnancy were more likely to experience IPV during the third trimester. Both studies support the importance of screening for specific types of IPV throughout pregnancy. Information obtained from the current research is valuable to health care providers because it is important they are aware of IPV risk factors and that different types of IPV, especially psychological IPV, can occur at any time during pregnancy.

L’inactivation du système de réparation des mésappariements de l’ADN (MisMatch Repair, MMR) favorise un processus oncogénique d’instabilité des microsatellites du génome (MicroSatellite Instability, MSI). Les cancers colorectaux (CCR) MSI peuvent s’observer dans un contexte familial (syndrome de Lynch, SL) ou sporadique. Les cryptes MMR déficientes (MMR-Deficient Crypt Foci, MMR-DCF), lésions de la muqueuse non tumorale perdant l’expression d’une protéine MMR, pourraient constituer un événement prénéoplasique de la carcinogénèse colorectale MSI chez les patients SL. Dans certains CCR MSI, les cas «Lynch-like», le diagnostic de SL ne peut être posé avec certitude car aucune mutation germinale pathogénique MMR n’est identifiée, malgré une forte suspicion clinico-phénotypique. Nous avons confirmé que les MMR-DCF ne s’observaient que chez les patients SL. Par ailleurs, nous avons montré qu’elles étaient 3 fois plus fréquentes dans la muqueuse adjacente à la tumeur que dans la muqueuse distante. Nous avons identifié des MMR-DCF chez un patient «Lynch-like», ce qui est un argument diagnostique en faveur d’un authentique SL. Les MMR-DCF devraient donc être systématiquement recherchées dans la muqueuse adjacente dans ce contexte. Par ailleurs, un défaut de champ en MGMT (O6-methylguanine-DNA-methyltransferase, une enzyme impliquée dans la réparation de l’ADN) dans la muqueuse non néoplasique pourrait favoriser la cancérogénèse MSI, par un mécanisme de tolérance à la méthylation. En évaluant l’expression de la MGMT comparativement à celle des protéines MMR, nous avons constaté que les MMR-DCF étaient plus fréquemment identifiées en cas de perte d’expression de la MGMT (p=0.004), et préférentiellement en cas de défaut de champ en MGMT. L’inactivation de la MGMT constituerait donc un événement précurseur favorisant l’apparition de MMR-DCF chez les patients atteints de SL
Inactivation of MMR (MisMatch Repair) system drives microsatellite instable (MSI) carcinogenesis. MSI colorectal cancers (CRC) may occur either in a hereditary (Lynch syndrome, LS) or sporadic context. Recently, MMR-deficient crypt foci (MMR-DCF) were described in non-neoplastic intestinal mucosa of LS patients. These are morphologically normal crypts, which exhibit loss of a MMR protein, and may constitute a pre-neoplastic event in LS. In some cases of early onset MSI CRC, diagnosis of LS is uncertain because no germline mutation is identified; theses cases are considered as “Lynch-like”. We detected MMR-DCF only in LS-CRC, and the ratio of MMR-DCF was 3 times higher in adjacent mucosa than in distant mucosa. We identified MMR-DCF in one “Lynch-like” patients; this is an argument that favors LS in this patient and that suggests that screening for MMR-DCF should be systematically performed in adjacent mucosa in this context. Furthermore, this pathway of MSI carcinogenesis could be favored by an O6-methylguanine-DNA-methyltransferase (MGMT) field defect in the non-neoplastic colorectal mucosa, through a mechanism called “methylation tolerance”. We compared MGMT and MMR proteins expression in the same crypts in non-neoplastic mucosa of LS-CRC patients and we noted that MMR-DCF were more frequently identified in case of loss of MGMT expression (p=0.004), and especially in case of MGMT field defect. The assingment of tasks to resources of a workflow system is called task distribution. The task distribution is an important activity for workflow systems, because it is necessary to ensure that a task is performed by the appropriate resource in due time. There are several approaches to task distribution in workflow systems. This work innovates by using a Link Analysis technique applied to the task distribution. The Link Analysis is used to rank the result of a web query. The rank is performed by considering the relevance of the pages. This work presents the application of Link Analysis in the context of workflow task distribution. We have proposed a new task distribution algorithm (wf-hits) based on Link Analysis algorithm. We have compared wf-hits against other related ones. This comparison have considered quantitative and qualitative aspects. The experiments have shown that the use of wf-hits has improved workflow systems 25% in quantitative terms meanwhile the qualitative terms has maintained the same level of similar related works.

A distribuição de tarefas é uma atividade importante para os sistemas de work ow, pois ´e necessário assegurar que uma determinada tarefa seja executada pelo recurso apropriado no tempo devido. Há várias abordagens para a distribuição de tarefas em sistemas de workflow. Este trabalho inova ao utilizar uma técnica oriunda da Análise de Ligações (Link Analysis) aplicada à distribuição de tarefas. A Link Analysis é utilizada para classificar o resultado de uma consulta na internet. A classificação é realizada considerando a relevância das páginas. O presente trabalho propõe a aplicação da Link Analysis no contexto da distribuição de tarefas em sistemas de work ow. É proposto um novo algoritmo para a distribuição de tarefas (wf-hits) que é baseado no algoritmo de Link Analysis. O algoritmo wf-hits é comparado com trabalhos correlatos em termos quantitativos e qualitativos. Os experimentos realizados mostraram que a utilização do wf-hits na distribuição de tarefas aos recursos em sistemas de workflow representa ganhos na ordem de 25% em termos quantitativos mantendo os mesmos patamares de qualidade dos trabalhos relacionados.

Le graphe du Web, plus précisément le crawl qui permet de l'obtenir et les communautés qu'il contient est le sujet de cette thèse, qui est divisée en deux parties.
La première partie fait une analyse des grand réseaux d'interactions et introduit un nouveau modèle de crawls du Web. Elle commence par définir les propriétés communes des réseaux d'interactions, puis donne quelques modèles graphes aléatoires générant des graphes semblables aux réseaux d'interactions. Pour finir, elle propose un nouveau modèle de crawls aléatoires.
La second partie propose deux modèles de calcul de communautés par émergence dans le graphe du Web. Après un rappel sur les mesures d'importances, PageRank et HITS est présenté le modèle gravitationnel dans lequel les nœuds d'un réseau sont mobile et interagissent entre eux grâce aux liens entre eux. Les communautés émergent rapidement au bout de quelques itérations. Le second modèle est une amélioration du premier, les nœuds du réseau sont dotés d'un objectif qui consiste à atteindre sa communautés.

Cette ?tude propose une analyse des fa?ons dont la visualit? critique a ?t? ins?r?e dans le mouvement ouvrier br?silien ? travers la presse ouvri?re et syndicale au d?but du XX?me si?cle, agissant comme un espace de partage sensible. L'objectif principal est de prouver que le projet de constitution d'une identit? de classe au Br?sil, s?est ?galement assis sur une culture visuelle diversifi?e d?tentrice d?une historicit? particuli?re, en m?me temps qu?elle a traduit un effort pour cr?er des liens non seulement entre sujets politiques impliqu?s dans le mouvement, mais aussi avec une tradition figurative, dont les origines remontent ? l'exp?rience fran?aise moderne de production et de reproduction de dessins politiques de profil contestataire. L'exp?rience visuelle produite par le contact entre le dessin ou la caricature et le spectateur est entendu comme un intervalle de temps dans lequel ont ?t? pr?par?es les conceptions particuli?res et collectives appartenant ? la classe ouvri?re. En ce sens, nous avons examin? un total de six publications nationales produites dans l'axe Rio de Janeiro - S?o Paulo entre les ann?es 1910 et 1935, deux d?entre elles de contenu libertaire et anticl?rical (A Lanterna et A Plebe), deux produites par un organisme syndical (O Cosmopolita et Voz Cosmopolita) et deux li?s au Partido Communista do Brazil (A Classe Oper?ria et A Na??o). M?thodologiquement, nous avons d?cid? de travailler avec le classement th?matique des gravures parce que nous pensons que cette approche est celle qui couvre le mieux le volume g?n?reux d'images en notre possession et qui nous a permis d'?tablir des axes th?matiques capables de mieux r?pondre ? nos questions en profondeur, sans, simultan?ment, perdre de vue l'attention aux sp?cificit?s de ces images. ? travers des recherches d?velopp?es sur les archives fran?aises, nous avons pu construire une analyse crois?e avec des images produites par les publications fran?aises de critique sociale de la fin du XIX?me si?cle et d?but du XX?me si?cle, ce qui nous a permis de poursuivre l'enqu?te concernant les nouvelles significations incorpor?es dans les dessins br?siliens. En outre, ce croisement a aussi fourni l'occasion de dresser une g?n?alogie de ces manifestations symboliques qui apparaissent ici comme un outil pour comprendre les nouvelles et multiples significations et m?me les migrations symboliques articul?es par ces images par rapport aux gravures v?hicul?es dans les journaux br?siliens. Cependant, ce que nous cherchons avec cette analyse est d'?tudier la conformation du ? se faire reconna?tre ? et ? de se reconnaitre soim?me ? dans une collectivit? politique qui avait le but de la repr?sentativit?, en prenant l'exp?rience visuelle comme un ballast important pour ce projet identitaire.
O presente estudo prop?e uma an?lise acerca das formas com as quais a visualidade cr?tica foi inserida no movimento oper?rio brasileiro atrav?s da imprensa oper?ria e sindical durante o in?cio do s?culo XX, atuando como um espa?o de partilha do sens?vel. O objetivo central consiste em comprovar que o projeto de constitui??o de uma identidade classista no Brasil, esteve assentado tamb?m numa cultura visual diversificada detentora de uma historicidade que lhe ? particular ao mesmo tempo em que traduziu um esfor?o no sentido de criar v?nculos n?o apenas entre os sujeitos pol?ticos envolvidos com o movimento, mas tamb?m com uma tradi??o figurativa, cujas origens remontam ? experi?ncia francesa moderna de produ??o e reprodu??o de desenhos pol?ticos de perfil contestat?rio. A experi?ncia visual produzida a partir do contato entre o desenho ou a caricatura e o expectador ? entendida enquanto um intervalo de tempo no qual foram elaboradas concep??es particulares e coletivas acerca do que consistia pertencer ? classe oper?ria. Nesse sentido foram examinadas um total de seis publica??es nacionais produzidas no eixo Rio de Janeiro ? Em pesquisa desenvolvida junto aos arquivos franceses pudemos construir uma an?lise cruzada com imagens produzidas por publica??es francesas de cr?tica social do final do s?culo XIX e in?cio do XX o que nos possibilitou uma investiga??o mais profunda concernente ?s ressignifica??es incorporadas aos desenhos brasileiros. Al?m disso, esse cruzamento tamb?m nos deu a oportunidade de tra?ar uma genealogia dessas manifesta??es simb?licas que se inserem aqui tamb?m como ferramenta para compreens?o da m?ltiplas ressignifica??es e das pr?prias migra??es simb?licas articuladas por essas imagens em rela??o ?s gravuras veiculadas nos peri?dicos brasileiros. This Researchis about history education and is directed toward the understanding of teacher practices in schools of the basic education in Cear?-Mirim, a city located in the state of Rio Grande do Norte. Its purpose is to understand the approaching forms of local history in 5? to 8? grade classrooms in the light of the recent innovations in the fields of the historiografy and education. The study was done with a view of investigating local history is being taught by teachers, as well as reflecting on the necessary conditions to the accomplishment of a teaching whose know how to make possible to break, on the one hand, with the limits of the narrow `local view`, and, on the other hand, with the globalized view, negating the local particularitities and especifications. Such questions had emerged as the understanding of what local history contents can constitute in a significant component in the production of school historical knowledge in 5? to 8? grade classrooms. History education is analyzed, considering the depositions of three teachers of Cear?-Mirim in reagard to its historiography conceptions and history as a school subject. This inquiry is of a qualitative nature and had as a main strategy of data construction from the interviews with the teachers. The analysis indicates the permanence of teacher practices who, even though presenting innovations, bring an implicit value hierarchy where the place or thematic places are not contemplated in the school contents or appear overwhelmed by general history and the Brazilian history, configuring itself as a hierarchical relation to problematic historical ones. Thus the necessity of having historical school knowledge, that considers the local especifications, without, however, ignoring the articulations with other spatial dimensions
Pesquisa sobre o ensino de hist?ria voltada para a compreens?o da pr?tica docente em escolas do ensino fundamental do munic?pio de Cear?-Mirim, localizado no estado do Rio Grande do Norte. Tem como prop?sito entender as formas de abordagem da hist?ria local em turmas de 5? a 8? s?ries ? luz das recentes inova??es nos campos da historiografia e do ensino. O estudo foi conduzido tendo em vista investigar como a hist?ria local vem sendo ensinada pelos professores, bem como refletir sobre as condi??es necess?rias ? concretiza??o de um ensino cujo saber-fazer possibilite romper, por um lado, com os limites do estreito localismo, e, por outro, com a vis?o globalizante e negadora das particularidades e das especificidades do local. The present research, located in the Pontificia Universidade Cat?lica de Campinas Psychological Prevention and Intervention line of research in the Post-graduation stricto senso program, aims to understand how the work category presents itself to those who have the streets as their main subsistence means and constantly wander from town to town surviving through socially disqualified activities, typical of the streets universe, with a special focus on the trecheiros , a category that describes the street population known to wander constantly from place to place. This study was developed throughout six months by means of Ethnography, in parks and public circulation places in the state of S?o Paulo. The data was collected and later analyzed using Social Psychology It was possible to conclude that there is a wide range of people who, despite occupying the same economical condition, have culturally distinct profiles as well as substantially different means of experiencing the constant wandering through towns and the (non) use of social welfare offered by the state to those who live in the streets. It was also possible to conclude that work presents itself to them as a category constitutive of their subjectivity, given that they call themselves workers in order to state their honesty and social relevance. Work is also a daily activity of survival maintenance without the pretention to accumulate capital or material possession, but to provide the necessary for one day only, situation that leads such category to a constant instability and great vulnerability, given that they are not covered by the welfare social net useful in case there is an impossibility to work, momentarily or not .
A presente pesquisa, situada na linha de Pesquisa Preven??o e Interven??o Psicol?gica do Programa de P?s-gradua??o Stricto Senso da Pontif?cia Universidade Cat?lica de Campinas, objetivou compreender como a categoria trabalho se apresenta a quem tem na rua seu principal suporte de subsist?ncia e mant?m-se em um caminhar constante de cidade em cidade, sobrevivendo por meio de atividades socialmente desqualificadas, pr?prias ao universo das ruas, com especial enfoque nos trecheiros , categoria de pessoas em situa??o de rua conhecida por permanecer em um caminhar constante de trecho em trecho . O estudo foi desenvolvido ao longo de seis meses por meio de Etnografia, em pra?as e locais p?blicos de circula??o em um munic?pio do interior paulista. Os dados foram posteriormente analisados sob a luz da Psicologia Social. Foi poss?vel concluir que existe uma ampla gama de pessoas que, apesar de situarem-se em uma mesma faixa econ?mica, possuem perfis culturalmente distintos bem como diferem substancialmente quanto aos modos de vivenciar a err?ncia constante pelas cidades e o (n?o) fazer uso das pol?ticas de assist?ncia social ofertadas a quem vive nas ruas. Foi poss?vel ainda concluir que o trabalho apresenta-se a elas como uma categoria constitutiva de sua subjetividade, pois afirmam-se como trabalhadoras no intuito de afirmarem-se como honestas e socialmente relevantes .O trabalho ? al?m disso uma atividade di?ria de manuten??o da sobreviv?ncia sem a pretens?o de, por meio dele, acumular capital e coisas materiais e sim prover o necess?rio para o dia de sustento, situa??o que leva tal categoria populacional a uma instabilidade constante e grande vulnerabilidade uma vez que fica totalmente descoberta por qualquer tipo de rede de seguridade social que possa lhe valer em caso de impossibilidade de trabalhar, moment?nea ou n?o.

The present study analyzes the activities of three State institutions of the Brazilian Computer Science field from 1958 to 1979 ? the Work Group on the Application of Computers (GTAC ? Grupo de Trabalho sobre Aplica??o de Computadores) / the Executive Group for the Application of Electronic Computers (GEACE ? Grupo Executivo para Aplica??o de Computadores Eletr?nicos) and the Coordinating Comission of the Activities of Electronic Processing (CAPRE ? Comiss?o de Coordena??o das Atividades de Processamento Eletr?nico). The three institutions represented different periods of the formation of the national Computer Science field and conducted a series of actions to control and promote activities, which encompassed from the rationalization of computer resources and the formation of workforce to the coordination of the emerging national industry of computers and peripheral devices. The main objective of this study is to evaluate the activities carried out by these organizations in order to identify their procedures and discuss their ?efficiency? at promoting Computer Science in the country. In this regard, we will identify the technicians involved in this process and the techno political actions based on their expertise, as well as their capacity of social integration and ability to establish a support network. Over the chapters, we will present some of these actions, such as the attempt to constitute a Government Data Center in 1959 and the incentive/control over the first computer technology industries in the country.
O presente estudo aborda as atividades de tr?s institui??es de Estado no campo da Inform?tica brasileira entre os anos de 1958 e 1979 ? O Grupo de Trabalho sobre Aplica??o de Computadores (GTAC)/Grupo Executivo para Aplica??o de Computadores Eletr?nicos (GEACE) e a Comiss?o de Coordena??o das Atividades de Processamento Eletr?nico (CAPRE). As institui??es representaram per?odos distintos da forma??o do campo da Inform?tica nacional e empreenderam uma s?rie de a??es para controlar e para promover atividades que envolviam desde a racionaliza??o de recursos computacionais e forma??o de m?o-de-obra at? coordena??o da nascente ind?stria nacional de computadores e perif?ricos no pa?s. O objetivo principal busca avaliar as atividades desempenhadas por esses ?rg?os a fim de identificar os procedimentos e discutir seu grau de ?efici?ncia? em disseminar a Inform?tica no pa?s. Para isso, ser?o identificados os t?cnicos envolvidos e suas a??es tecnopol?ticas baseadas em suas expertises, assim como sua capacidade de inser??o social, de maneira a formar uma rede de apoio. Ao longo dos cap?tulos ser?o apresentados algumas dessas a??es, como a tentativa de constituir um CPD de Governo em 1959 e o incentivo/controle sobre os primeiros fabricantes de tecnologias computacionais no pa?s.
