"Factor H (FH) Is Released Slowly and Continuously from Human Endothelial Cells (ECs): FH Is Not Packaged in Weibel-Palade Bodies or Secreted in Response to EC Stimulation." Abstract:
Abstract Introduction Ultra-large von Willebrand factor (ULVWF) strings are synthesized in ECs, packaged in Weibel-Palade Bodies (WPBs), and secreted by stimulated ECs. Complement components studied to date [C3, factor (F) B, FD, FP, FH, FI, C5] are released slowly and continuously from human umbilical vein endothelial cell (HUVEC) cytoplasm and are not packaged in WPBs (PLoS One. 2013;8(3):e59372). In contrast, a recent report (Blood. 2014;123(1):121-5) contended that FH co-localizes with VWF in the WPBs. If this were so, it could have therapeutic importance for the treatment of atypical hemolytic uremic syndrome (aHUS) resulting from deficiency of FH because it might be possible to increase circulating FH levels transiently by administration of the WPB secretagogue, des-amino-D-arginine vasopressin (DDAVP). Hypothesis FH is not co-localized with VWF in WBPs, but rather is released slowly and continuously from EC cytoplasm regardless of cell stimulation. Methods Immunofluorescent Microscopy HUVECs were stimulated with histamine and stained with rabbit anti-VWF plus secondary donkey anti-rabbit Alexa Fluor IgG-488. The cells were then fixed and stained with goat-anti FH plus secondary chicken anti-goat Alexa Fluor IgG-647. The nuclei were detected with DAPI. In vitro VWF and FH from HUVECs HUVECs either were, or were not, stimulated with histamine. Supernatant was collected a variety of times over 7 hrs and assayed for VWF and FH antigen levels by ELISA. VWF assay antibodies (polyclonal): (1) capture, rabbit anti-human VWF (Ramco); (2) detection, goat anti-human VWF (Bethyl) and rabbit anti-goat IgG-HRP (Invitrogen). FH assay antibodies: (1) capture, polyclonal goat anti-human FH (Advanced Research Technologies); (2) detection, monoclonal mouse anti-human FH (Pierce, Thermo Scientific) and polyclonal goat anti-mouse IgG-HRP (Invitrogen). In vivo VWF and FH Plasma samples were obtained from 6 pediatric patients with von Willebrand disease (VWD) being tested for EC release of WPB VWF in response to DDAVP. For each patient, 1 sample was obtained prior to DDAVP administration, and 2 other samples were obtained 1 and 4 hours later. VWF levels were measured in each sample using standard clinical laboratory procedure at an affiliated hospital. FH antigen levels were quantified by ELISA, as above. Results Using non-overlapping spectral secondary detection antibody pairs, VWF was seen in clusters in HUVEC WPBs (Fig. 1A). In contrast, FH was distributed throughout the HUVEC cytoplasm (Fig. 1B). The VWF and FH images did not overlap, indicating that VWF and FH did not co-localize in the WPBs. Fig 1. Immunofluorescent images of HUVECs stained for VWF and FH. Fig 1. Immunofluorescent images of HUVECs stained for VWF and FH. Histamine addition to HUVECs in vitro resulted in ~ 4-fold increases in VWF secreted from HUVEC WPBs at 30 min and 1 hour, and 2-fold increases at 3 hours (Fig 2A). In contrast, FH release was slow and continuous, regardless of histamine stimulation, suggesting that FH is located in EC cytoplasm and is not stored in WPBs (Fig. 2B). Fig 2. In vitro VWF and FH release from ECs under non-stimulated and histamine-stimulated conditions. Fig 2. In vitro VWF and FH release from ECs under non-stimulated and histamine-stimulated conditions. In all 6 patient samples, VWF antigen increased significantly at 1-hour post-DDAVP administration (Fig. 3A). In contrast, FH antigen levels did not change significantly at hour 1 or hour 4, compared to hour 0, indicating that FH is not co-localized and secreted along with VWF from the WPBs of stimulated ECs in vivo (Fig. 3B). Fig 3. (A) Mean responses of VWF antigen to DDAVP, in vivo, with 95% CIs.The mean response was significantly greater 1-hour and 4-hours post-DDAVP compared with baseline (P=0.0085 and 0.0079, respectively). After 1-hour post-DDAVP, the mean response was 201% greater (95% CI: 129%, 314%) than baseline. After 4-hours, the mean response was 174% greater (95% CI: 123%, 247%) than baseline. (B) Responses of FH to DDAVP, in vivo. There was no statistically significant difference in FH response between time points (P=0.77). Conclusions We used immunofluorescent microscopy and ELISA assays on samples obtained in vitro and in vivo to demonstrate that FH is not packaged in, or secreted from, the WPBs of stimulated human ECs. FH is, therefore, similar to all other complement components studied to date in that it is released slowly and continuously from ECs and is not influenced by cell stimulation. DDAVP is unlikely to be a viable treatment option for patients with aHUS secondary to deficiency or inhibition of FH. Abstract:

Abstract:
Hyland, Lee, and Mills (2015) specified the two most popular scales for mindfulness: the Freiburg Mindfulness Inventory (FMI; 30 items, Buchheld, Grossman, & Walach, 2001; 14 items, Walach, Buchheld, Buttenmüller, Kleinknecht, & Schmidt, 2006) and the Mindfulness Attention and Awareness Scale (MAAS; Brown & Ryan, 2003). However, the popularity of these measures does not necessarily mean that they are of high quality. Especially considering the complex epistemology and ontology of the mindfulness construct, we should apply mindfulness assessments with caution. More specially, according to item development theorists in the industrial and organizational (I-O) area (see Hinkin, 1998; Hinkin & Tracey, 1999; Kerlinger & Lee, 2000; Schriesheim, Powers, Scandura, Gardiner, & Lankau, 1993), scholars must select measures that have (a) a clear operational definition; (b) alignment between definition and measure (content validity); (c) high reliability and (d) high construct validity; and (e) high criterion-related validity. However, it is not clear which of the available mindfulness assessments satisfies these criteria and to what extent. In this commentary, we assess currently used measures based on these criteria and provide directions for future research.

Abstract:
This article examines the meaning of dishonest appropriation in the offence of theft after the House of Lords' decisions in Gomez and Hinks. It maintains that whilst Gomez broke the distinction in the Theft Act 1968 between the simple thief and the deceitful rogue, it did at least maintain the underlying rationale for theft which is the protection of property rights. In contrast Hinks does not comply with this rationale as it allows for the possibility of an appropriation when there was a valid gift of property so that the actus reus of theft no longer requires that there be the composite whole of a ‘dishonest appropriation’. Thus the emphasis in the offence of theft is now on dishonesty and this may be in violation of Articles 5(1) and 7(1) of the European Convention on Human Rights.

Abstract:
The law of theft, as understood in Gomez and Hinks, has been the occasion of almost unanimous academic condemnation and of robust dissenting opinions in the House of Lords. While much of the critical discussion is sophisticated and challenging, it is important that the baby is not to be expelled with the bathwater. We suggest that one argument in favour of the current position is that it offers distinct protection to some of the more vulnerable members of society. This advantage ought nevertheless to be sacrificed if it can be purchased only at the cost of violating the rule of law and the harm principle. But our examination of these ideas reveals that the price need not be paid. The rule of law contains not one idea, but a plurality of ideas, many of which support the current position. As for the harm principle, it is notable that Hinks does not castigate harmless behaviour; rather it attacks the wrong of exploitation. This raises many difficult issues, but we argue that unless such exploitative behaviour is explicitly addressed in Legislation, reforming the current ‘broad’ understanding of the law in favour of a ‘reductive’ account assimilating theft to non-voluntary transfers would be a retrograde step. In principle the new concern for protecting the vulnerable from exploitation is welcome.

Abstract:

Abstract:
Hay is an increasingly scarce commodity on cattle farms in the UK, due to the transition to silage in recent years. However it is generally preferred by farmers for feeding to calves because it is easy to transport and to feed in small quantities without rapid deterioration. A commonly used alternative is straw, and the voluntary consumption is as high as hay (Kang and Leibholz, 1973), but the DM digestibility is low and high levels of straw in the diet depress empty body weight gain (Thomas and Hinks, 1982). Mixing straw with palatable feeds of high nutrient content, such as molasses, has been shown to increase its feeding value to that of silage for older cattle (Phillips and Leaver, 1985), and could increase the forage intakes of young calves, where palatability is important in determining intake. Maintaining a high forage intake could also reduce the incidence of deleterious oral behaviours, such as licking the pen.

Abstract:
The current law of theft continues to rely on the concept of ‘appropriation’ under the Theft Act 1968. The broadness of this concept has led to serious problems in its application by the courts. Amongst these are the problems of ‘appropriation with consent’ and ‘appropriation without taking possession’ which have manifested themselves in several counter-intuitive decisions by the courts such as Gomez, Hinks and Pitham and Hehl. As a result these decisions have given rise to strong criticism by academics and practitioners alike. The law of theft, therefore, is without doubt in strong need of reform. After examining these key problems this article conceptually analyses the German approach by way of comparative juxtaposition with the English approach. Far from suggesting any adaptation of continental solutions, the author aims to provide extra material for consideration for a future reform of the law by introducing a systematic construction of the offences of theft and fraud from a foreign jurisdiction.

Abstract:

The main purpose of this study is to validate the three instruments applied in studying the relationship between Religious Commitment, Peer Relations and Behavior of secondary school adolescents (Mwita, 2010). The greatest challenge in understanding the behavior of people is the adequate measurement of abstract construct (Hinkin, 1989). Thus confirmatory factor analysis was conducted, and factors derived from all the three instruments were then explored by orthogonal rotations. Three factors were extracted from the behavior items, two from the peer relations items and two from the religious commitment items. Reliability scores between the constructs indicated statistically significant difference on the latent factors of all the three instruments. Constructs were further validated by determining Average Variance Extracted (AVE) where high scores of AVE (between 0.8 & 0.9) and high factor scores of ?.4 (Hair et al., 2010) were obtained. Finally, constructs were validated by applying SEM which proved the validity of all the constructs by obtaining good fit indices.

Abstract:
PurposeThe purpose of this paper is to examine the relationship between the perceptions employees have regarding how they are treated in the workplace environment by their current or most recent supervisor, and how this predicted their feelings of power within themselves. The perceptions were measured utilizing the Managerial Leadership Perceptions Questionnaire (MLPQ) created by Parmer (2017). Employee power was measured utilizing the Power Instrument developed by Hinkin and Schriesheim (1989) which stemmed from French and Raven’s (1959) five original bases of power theory to include referent, expert, legitimate, reward, and coercive.Design/methodology/approachThe authors collected a sample of 199 participants gathered from Amazon’s Mechanical Turk digital labor pool. Participants completed a survey which measured their managerial perceptions, bases of power, and demographic characteristics. Statistical analysis was used, including a factor analysis, to explore the relationship between managerial perceptions, bases of power, and demographic characteristics.FindingsThis study demonstrated that there were no significant associations between the demographic associations and personal power. There were significant associations between the demographic associations and position power, managerial perceptions and personal power, managerial perceptions and position power, and managerial leadership style and power.Research limitations/implicationsFive bases of power were examined in this study to include referent, expert (i.e. personal power), legitimate, reward, and coercive (i.e. position power). There is a sixth power now, information power, as noted by Northouse (2016) that needs to be additionally examined. Self-confidence and empowerment feelings were not technically measured quantifiably in this study but were expected feelings based on what mindsets power can produce within a person. Researching these additional feelings of self-confidence and empowerment and how this relates to follower power is needed moving forward in this research area. Finally, ethnic differences need to be measured moving forward.Practical implicationsThe practical implications of this study show that employees do embody perceptions and attitudes regarding their current or most recent supervisor based on how they are being treated. This, in turn, can affect their own personal feelings of power within themselves and within the overall organization at large. Careers can be affected, both good and bad, organizational cultures can be impacted by both good and bad, workplace assumptions and norms, as well as, workplace relationships can be affected, both good and bad.Social implicationsThe social implications of this study indicated that employees’ perceptions and attitudes regarding their immediate supervisor can create positive or negative feelings toward the supervisor which can, in turn, affect the organization’s culture and workplace environment, both good and bad. Working at an organization is within a social environment that needs to be managed and cultivated appropriately for all parties involved.Originality/valueThe majority of the prior research examines leader–follower relationships. No prior research has utilized this particular perception and attitudinal model, the MLPQ developed by Parmer (2017), and the five bases of power model developed by Hinkin and Schriesheim (1989) together in one study. This study explored employee managerial perceptions and their feelings of power within the follower–leader dyadic relationship, as opposed to the leader–follower dyadic relationship which has been more commonly reported within the literature.

Abstract:
THE House of Lords has upheld, by a majority, the decision of the Court of Appeal in Hinks [2000] 3 W.L.R. 1590 (noted (1999) 58 C.L.J. 10), giving a positive answer to the certified question: “Whether the acquisition of an indefeasible title to property is capable of amounting to an appropriation of property belonging to another for the purposes of section 1(1) of the Theft Act 1968”. The appellant had persuaded a somewhat simple-minded man to make her the “gift” of a quite considerable sum of money. No deception was alleged to have been employed, and so far as the civil law was concerned, the “gift” might well have been a perfectly valid transaction; the question was never determined by the jury, because it was deemed to be irrelevant by the trial judge. But the decision of the House of Lords is to the effect that she was properly convicted of theft however that question might have been answered. So, it would seem, a person may become the indefeasible owner of property and nevertheless be accounted a thief of that very same property, and by the very act of acquiring the ownership of it.

Abstract:
The majority of calves in Britain (2-3 m a year) are artificially reared and receive less milk than they would if suckled. It is therefore important that calves should develop early in life the ability to consume and digest solid food. Traditionally, calf starter diets are formulated to contain rapidly fermentable cereals as the major energy source, which is adult cattle cause a depression in rumen pH to below 5.5 and inhibits food intake (Fulton, Klopfenstein and Britton, 1979). In experiments which have demonstrated a beneficial effect on food intake from the inclusion of roughages in all pelleted diets based on cereals appear to be associated with the concurrent increase in rumen buffering capacity (Thomas and Hinks, 1982). The appetite of the calf may have been constrained by low fibre digestion (Williams, Innes, Brewer and Magadi, 1985) and the bulky characteristics of the roughage. Two experiments are reported in examining the potential of replacing the energy source, of calf diets, supplied by cereals with materials of high buffering ability, but without the bulky and indigestible characteristics of roughages. Expt 1 examined the effects of replacing rolled barley (RB) with molassed sugar beet feed (MSBF) or lucerne meal (LM). Expt 2 examined the factorial replacement of RB with MSBF.

Abstract:
Sex differences in influence tactics were examined with a sample of 269 followers (67 men, 202 women) at a large midwestern national insurance company who rated the downward influence tactics used by their direct supervisors. Downward influence tactics are behaviors used by leaders to gain compliance from followers. One department within the organization was identified as a source for participants in the study. Participation was voluntary. The age range for the sample was 21 to 65 years, with the largest percentage falling in the 40–49 year range ( M = 3.8, SD = .8). Hierarchical linear modeling procedures were utilized to analyze the multiple level data (leader and follower) and to examine variables within the organization at different levels of analysis. Leader participants were asked to solicit their followers to complete an influence tactic measure, which consisted of the most reliable subscales taken from the Influence Behavior Questionnaire, Schriesheim and Hinkin Influence Measure, and the Profiles of Organizational Influence Strategies. The integrated measure resulted in a 45-item scale. It was hypothesized that, overall, followers would report that male leaders would use hard influence tactics more frequently than female leaders. On the other hand, followers would report that female leaders would use soft influence tactics more frequently than male leaders. When differentiating followers by sex, however, we expected that male followers would report more than female followers that their leaders use hard tactics more frequently. Also, we expected that female followers would report (more than male followers) that their leaders use soft tactics more frequently. Overall, followers reported that male leaders used significantly more personal appeal and consultation, so called “soft tactics,” with their followers than did female leaders. Female followers reported that their leaders (both male and female) used consultation and inspirational appeal more frequently. In contrast, male followers reported that their leaders used exchange, so called “hard tactics,” more frequently.
