Full textA study of antioxidant activity in medicinal herbal raw materials and phytopreparations requires the application of optimized HPLC post-column assays as well as validation of their methodologies that could allow for a reliable, accurate and reproducible evaluation of composition, distribution and stability of antioxidants. The aim of the work is to examine HPLC post-column assays and adjust them for research on plant antioxidants as well as assessment of composition and activity of herbal raw material extracts and their preparations. It was determined that reaction coil parameters have impact on the ratio of peak height and baseline noise. Evaluated validation results confirm the suitability of HPLC-DPPH and HPLC-ABTS post-column assays for the determination of radical scavenging compounds and quantitative evaluation of antiradical activity. Optimized HPLC-FRAP post-column assay can be applied for discriminating reducing active compounds in complex mixtures and quantitative evaluation of their reducing activity. Main radical scavenging and/or reducing activity markers were determined: Chlorogenic acid and Hyperoside for Crataegus L., Rosmarinic acid for Origanum L. and Perilla L., Chlorogenic acid and 3,5-dicaffeoylquinic acid for Achillea L., Epigallocatechin gallate for Fragaria L. These markers are applicable for the quality control of the herbal raw materials and phytopreparations containing antioxidants.

Full textInfusions, decoctions, alcoholic preparations and other dosage forms of Artemisia afra are frequently used in South African traditional medicine. Generally when these preparations are made without applying good manufacturing practices they do not meet microbial quality control standards, safety and toxicity criteria and encourage poor patients compliance. To overcome the aforementioned disadvantages of traditional dosage forms a sold dosage form, i.e. a table might be recommended. The first objective of this study was to formulate and manufacture a rapid release tablet dosage of Artemisia afra that would contain an amount of plant material equivalent to that found in its traditional liquid dosage forms and that would meet conventional pharmaceutical standards. The second objective was to conduct a pilot study to obtain a preliminary profile of the bioavailability of select flavonoids presents in both the tablet and traditional liquid preparation of Artemisia afra in humans.

Abstract The purpose of this study was attempted to investigate the antioxidative activity of seven kinds of commonly used herbal tea materials. The methanol, hot water and hot water-methanol extracts were then prepared to measure antioxidative activities, total phenolic content and total flavonoid content. A HPLC method was also carried out to analyze active components. Results showed that the scavenging effect on DPPH radical, reducing power, and inhibiton of linoleic acid peroxidation were higher in hot water extract than in methanolic extract. However, methanol extract had higher chelating effect on ferrous ion. Among the materials, Artemisia capillaries, Kadsura japonica and Mesona procumbent were stronger in DPPH scavenging, Houttuynia cordata and Artemisia capillaries were stronger in chelating effect and reducing power, and Artemisia capillaries and Kadsura japonica were stronger in the inhibition of linoleic acid peroxidation. When antioxidative activities were expressed as IC50 values, the IC50 measured from methanol extracts were the lowest, followed by that from hot water extracts. In methanol extracts, the IC50 for scavenging DPPH (0.0062~0.0711 mg/mL), reducing power (0.0212~0.1370 mg/mL) and inhibition of linoleic acid peroxidation (0.0093~0.1062 mg/mL) were commonly smaller in Artemisia capillaries and Kadsura japonica than in others. But the IC50 for chelating ferrous ion (0.0015~0.0124 mg/mL) was the lowest in Houttuynia cordata. The total phenolic and flavonoid contents in methanol (34.37~233.32 mg/g and 40.00~221.40 mg/g) and hot water (9.05~131.93 mg/g and 19.26~217.61 mg/g) extracts were much higher than that in hot water-methanol extract (total phenolic content: 1.40~17.94 mg/g; total flavonoid content: 0.95~19.27 mg/g). The total phenolic content was most abundant in Kadsura japonica and Artemisia capillaries, but that in Salvia plebeia was the lowest. The total flavonoid content was most rich in Artemisia capillaries, whereas Salvia plebeia and Houttuynia cordata contained the lowest. The samples were also analyzed for the presence of phenolic acids and flavonoids. A total of 11 components including gallic acid, catechin, catechin, caffeic acid, rutin, myricetin, morin, quercetin, luteolin, kaempferol and apigenin were detected and the total amount in samples was 1.12~44.32 mg/g. The major constituents in Mesona procumbent, Solanum violaceum, Artemisia capillaries, Ilex asprella and Salvia plebeia, however, were still unknown.

This study uses commercial herbal tea raw materials including Rhinacanthus nasutus (L.) Kurz and Mentha spicata to conduct safety test and active ingredient analysis. Furthermore, discussion on the effect of mixed herbs hot water extracts from fructose–induced hepatic lipogenesis was investigated by using the human hepatoblastoma HepG2 as a mode. And to explore its potential for improving non–alcoholic fatty liver. Mix Rhinacanthus nasutus (L.) Kurz and Mentha spicata in a 4:1 ratio and decoct it in water to obtain mixed herbal tea. The toxicity and mutagenicity test of hot water extracts from mixed herbs (MH), Rhinacanthus nasutus (L.) Kurz and Mentha spicata toward Salmonella typhimurium TA98, TA100, TA102, TA1535 and TA1537 experimented with five different concentrations, including 1, 2, 3, 4 and 5 mg/plate. The results show that the sample is non–toxic and mutagenic toward Salmonella typhimurium (±S9). For the analysis of active ingredients, the total phenol content of hot water extracts from mixed herbs, Rhinacanthus nasutus (L.) Kurz, and Mentha spicata weighting 65.46, 56.22, and 88.70 mg GAE/g extract respectively, and the flavonoid contents were 15.64, 13.69, and 21.79 mg QE/g extract respectively. In addition, the main active ingredient of mixed herbs hot water extract is myricetin, following by caffeic acid, protocatechuic acid, p–coumaric acid and rutin and so on, which identified by HPLC analysis. In the cell test, HepG2 cells were induced by fructose and reactive oxygen species (ROS) increased significantly so does the expression of lipid synthesis–related genes include sterol regulatory element–binding protein–1 (SREBP–1), carbohydrate responsive element–binding protein (ChREBP), acetyl–CoA carboxylaseα (ACCα), fatty acid synthase (FAS) and stearoyl–CoA desaturase–1 (SCD–1). Moreover, the expression of phospho–AMP activated protein kinase (p–AMPK) and carnitine palmitoyltransferase–1 (CPT–1) were suppressed significantly, thereby reducing the β–oxidation of fatty acids. The condition of mixed herbs hot water extract could decrease ROS production and the expression of lipid synthesis–related genes. In summary, the mixed herbs hot water extract is non–genotoxicity and rich in active ingredients such as phenolic acids and flavonoids. Having the potential to provide hepatoprotective and decrease the incidence of non–alcoholic fatty liver disease. Looking forward to the follow–up contribution of physiological function side on the prevention of metabolic syndrome related diseases but also increases economic value and development potential.

Three silica-gel based materials ( silica-gel, silica-gel C8 and silica-gel CN ) were applied for the attachment and relevant tests onto: 1. Complex extracted from Polygonum Chinensis L. which has been used by Taiwanese folk to heal wound skin externally. 2. Mixture of Rhein and Emodin simulating the active and toxic compounds contained in Polygonum Multiflorum Thunb. which has been used by Chinese medicinal doctors for oral applications internally. Aqueous acetic acid generates the best extract results better than water and ethanol for Polygonum Chinensis L.. The stability of natural extract was checked by kinetic studies performed under elevated temperature (47oC : higher than room and body temperature) and the results show that it is quite stable. Some degradation can only be observed by forced degradation with strong oxidizing reagent. Among the three materials tested, silica-gel C8 gave the best attachment result with the extracted complex of Polygonum Chinensis L.. Hence the matrix of silica-gel C8 attached by natural medicinal complex was further checked against the extracted complex without any attachment for its trans-dermal releasing rate. The former one generated a trans-dermal rate of 64.5% in 4 hours already. The trans-dermal rate for the latter one was only 26.9% in 4 hours (i.e. 37.6% less than the former one). The main purpose of second application for this research is to improve the traditional detoxification process by using scientific materials and methods. This study demonstrates all three silica-gel based materials can do better and quicker job than the traditional detoxification. The attachment rate for Rhein is 77.3% with silica-gel CN, 71.3% with silica-gel C8 and 52.5% with silica-gel at second hour. Furthermore, silica-gel CN has the best attachment with Rhein and least absorption of Emodin (less than 10%) from 2 to 4 hours steadily. Therefore among the three scientific materials, silica-gel CN is the best for removing toxic substance while retaining active compound without degradation.
