O presente trabalho transita entre duas ?reas do conhecimento: a Filosofia e o Direito. ? resultado de um estudo que visa ? aproxima??o da hermen?utica filos?fica de Hans- Georg Gadamer com a hermen?utica jur?dica, tendo como objetivo apresentar alguns dos principais conceitos tratados pelo autor e discutir a possibilidade de sua aplica??o ? interpreta??o na esfera do Direito. A abordagem te?rica envolveu, al?m da pr?pria perspectiva filos?fica central de Gadamer, a pesquisa de autores que fazem uma an?lise dos seus pressupostos. Buscou-se exemplificar, ainda, tal aplica??o, por meio de uma poss?vel leitura ampliada do conceito de c?rculo hermen?utico proposto pelo autor. Verificou-se, afinal, que determinados conceitos fundamentais da filosofia de Gadamer tais como o c?rculo hermen?utico, a tradi??o, a dist?ncia temporal, a consci?ncia da hist?ria dos efeitos e a aplica??o como forma de compreens?o s?o aplic?veis ? hermen?utica jur?dica, possibilitando uma reflex?o mais aprofundada e dial?tica dessa ?rea. Abstract:
The thesis deals with the cooperation between the director and actor during individual phases of the staging process as they appear in everyday theatre life. The indiviual phases of the staging process are supplemented by the author's own experience with managing the actors, which she gained especially in the course of her studies at the Drama Faculty of AMU. The second part of the thesis investigates the issues of inner truthfulness and the possibilities of its achievement within the production of a play. As a part of collecting the material for the thesis, the author conducted a research, which included interviews with three professional directors - Hana Burešová, Ladislav Smoček and Štěpán Pácl, whose methods of work are described. In the final part of the thesis the author states that it is difficult to draw general conclusions about the cooperation between the director and actor because it is always of subjective nature. The issue of inner truthfulness is similarly difficult to generalise. Due to the multiplicity of options, however, neither of them loses attractiveness for further research.

This research is in charge of responding whether philosophical hermeneutics, which became autonomous within the framework of the thought of Hans-Georg Gadamer the claim of universality of the hermeneutical problem attribute, may be qualified as genuine philosophy. The underlying theme of the thesis covers the main features of Gadamer's philosophical hermeneutics in order to promote a kind of internal criticism about its corresponding assumptions, especially those that revolve around the universal primacy of the historicity of understanding. It advocates the need to counteract against the transcendental horizon of meaning, significantly accentuated by philosophical hermeneutics through the incidence of historical understanding that runs through each interpretation about the world, the transcendental horizon of normative validity, responsible for critical and reflective bypass any thought that title itself as legitimately philosophical. In these terms, hermeneutics as philosophy, to achieve the desired universality platform, must claim at the same time the substrate of normative rationality validation, so as not to incur into a kind of ontological hypertrophy, acting just like a redundant substitute hermeneutics of Heidegger's philosophy. Furthermore, philosophical hermeneutics should resist, with equal caution of self-criticism, in stimulating too much of this counterpart reflective validity in order to challenge the arrogant place able to propel the verdict of the final word or the last intransitively foundation considered as an ultimate point of view, behold the assumption of its universality stands on the critical path of the metaphysical finitude, that is, on the horizon of a program stripped of absolute or totalitarian overtones.
Trata a presente investiga??o de responder se a hermen?utica filos?fica, autonomizada nos marcos do pensamento de Hans-Georg Gadamer pela reivindica??o do atributo da universalidade do problema hermen?utico, pode ser qualificada genuinamente como filosofia. O fio condutor da tese percorre as principais caracter?sticas da hermen?utica filos?fica gadameriana com a finalidade de promover uma esp?cie de cr?tica interna a partir dos correspondentes pressupostos, especialmente os que giram em torno do primado universal da historicidade da compreens?o. Preconiza-se a necessidade de contrapor ao horizonte transcendental de sentido, significativamente acentuado pela hermen?utica filos?fica pela incid?ncia da compreens?o hist?rica que atravessa toda e qualquer interpreta??o acerca do mundo, o horizonte transcendental de validade normativa, respons?vel pela deriva??o cr?tica e reflexiva de qualquer pensamento que se intitule como legitimamente filos?fico. Nesses termos, a hermen?utica enquanto filosofia, para alcan?ar-se na pretendida plataforma de universalidade, precisa reivindicar concomitantemente o substrato da racionalidade normativa de valida??o, de maneira a n?o incorrer na hipertrofia do ontol?gico, como se fosse um substitutivo redundante da filosofia hermen?utica de Heidegger. Esta disserta??o tem o objetivo de recriar o conceito de compreens?o (Verstehen) na obra Verdade e M?todo de Hans-Georg Gadamer. Para ele, a compreens?o consiste num atributo da experi?ncia de mundo do ser humano. A tese ? desenvolvida a partir de dois pressupostos: (i) o conceito de Lebenswelt de Husserl, o qual ? condi??o de possibilidade de conhecimento: antes de ser sujeito cognoscente j? se ? objeto no mundo; (ii) a contribui??o de Heidegger, na defini??o de fenomenologia como hermen?utica atrav?s da retomada do sentido do ser no Dasein, que ? conhecida sob o nome de hermen?utica da facticidade. Primeiramente, tratase do modo de ser da obra de arte como par?metro a fim de se investigar o fen?meno da compreens?o. Assim como a experi?ncia da obra de arte, o conceito de jogo possui um fim em si mesmo, que se configura enquanto o espectador (o jogador) envolve-se no espet?culo (no jogo). Posteriormente, trata-se de pensar o princ?pio da historicidade da compreens?o, o qual inclui a substancialidade da historicidade do int?rprete e a consci?ncia do valor da tradi??o no ato de compreender textos. Aqui, a reabilita??o do preconceito como condi??o de possibilidade do int?rprete torna-se chave. Quando se diz da experi?ncia hermen?utica querse referir ? experi?ncia da finitude humana que nunca ? repetida, e est? em constante processo de aprendizado. Por fim, trata-se do car?ter da linguagem da compreens?o como meio privilegiado da experi?ncia hermen?utica. Especificamente, ? na linguagem - como di?logo - que se mostra o car?ter especulativo, no qual o sentido da coisa em quest?o mostra-se tal e qual ela ?. Nela ? que a compreens?o encontra sua objetividade e, por isso, d? ? compreens?o o car?ter de universalidade. This doctoral dissertation deals with the contribution of Gadamer's philosophical hermeneutics to contemporary, ongoing debates on multiculturalism and the dialogue of cultures. Although Gadamer has not developed a political philosophy, he is often regarded as an author who contributed significantly to constructing an alternative account to multiculturalism or the dialogue of cultures. The question that guides the current research is as follows: Gadamer's philosophical hermeneutics, as a finite reflection is emancipated only in the encounter with the Other and to the extent that it holds that the deepening of self-understanding (Selbstverst?ndnis) is attached to the understanding of other world views and life forms, can it be presented as a rational alternative to multiculturalism and the dialogue of cultures? The thesis presented here shows precisely that possibility. To the extent that he does not accept a relativistic idea of absolute incommensurability or incommunicability of the diverse world views and ways of life, and insofar as he defends a critical dialogue based on the art of asking questions and being open, Gadamer proposes a rationality that carries nearly universal standards for a dialogue of cultures or multiculturalism.
A presente tese trata da contribui??o da hermen?utica filos?fica de Gadamer ao debate contempor?neo sobre multiculturalismo e di?logo de culturas. Embora Gadamer n?o tenha desenvolvido uma filosofia pol?tica, em diversos momentos ? lembrado como um autor que contribuiu significativamente na constru??o de uma alternativa ao multiculturalismo ou mesmo ao di?logo de culturas. A quest?o que orienta a presente investiga??o ? a seguinte: a hermen?utica filos?fica de Gadamer, enquanto uma reflex?o finita que se emancipa somente no encontro com o outro na medida em que entende que o aprofundamento da autocompreens?o (Selbstverst?ndnis) est? unido ao entendimento de outras vis?es de mundo e formas de vida, pode ser apresentada como uma racionalidade alternativa ao multiculturalismo ou di?logo de culturas? A tese ora apresentada evidencia tal possibilidade. In this thesis we argue that the development of an ethics is present in the whole hermeneutics of Hans-Georg Gadamer. We came to this conclusion following Gadamer?s interpretation of the practical philosophy of Plato and Aristotle. We also achieved this idea through the recognition of the similarity between the proposal of a hermeneutical understanding and moral phenomena considered since the Socratic question about the good. In this sense, the conceptual framework that connects Gadamer?s philosophy and an ethics of dialogue is grounded in both the hermeneutical tradition and in his reinterpretation of the concept of Aristotelic phronesis and the Platonic dialogue. In our view, one of the most important contribution of the ?hermeneutical ethics of dialogue? was to seek a new basis for the discussion on ethical behavior. This is a concept of dialogue that identifies in the project of a human solidarity the possibility for the emergence of actions aimed at taking care of the other seeking the public well-being. Therefore, Gadamer?s hermeneutics reaffirms the importance of philosophy to evaluate the fundamental issues of our practical reality and allowing a rich debate of philosophy and other sciences (medicine, law, education).
Defendemos neste trabalho que a elabora??o de uma ?tica est? presente em toda a hermen?utica de Hans-Georg Gadamer. Chegamos a essa tese por meio da interpreta??o que Gadamer faz da filosofia pr?tica de Plat?o e Arist?teles e do reconhecimento da semelhan?a entre a proposta de uma compreens?o hermen?utica e os fen?menos morais considerados desde a pergunta socr?tica sobre o bem. Nesse sentido, a base conceitual que une a filosofia de Gadamer a uma ?tica do di?logo se encontra tanto na tradi??o hermen?utica como na sua releitura do conceito de phr?nesis aristot?lico e da ideia de di?logo plat?nica. Contudo, uma das maiores contribui??es do que chamamos de ??tica hermen?utica do di?logo? foi buscar uma base nova para a reflex?o dos modos de comportamento ?ticos. Trata-se de uma concep??o de di?logo, que identifica no projeto de uma solidariedade humana a possibilidade do surgimento de discursos e a??es voltados para o cuidado com o outro e, assim, para uma participa??o na vida em comunidade em prol de um bem comum. Desse modo, a hermen?utica de Gadamer termina reafirmando a relev?ncia da reflex?o filos?fica para quest?es fundamentais da nossa realidade pr?tica, o que leva a um debate prof?cuo entre a filosofia e outras ci?ncias (medicina, direito, educa??o).

Abstract:
Development of safe and efficient approaches for gene delivery in human embryonic stem cells (hESc) and particularly in human induced pluripotent stem (hiPS) cells, which can be derived in a person-specific manner, is considered to be imperative for harnessing their full potential in both the basic and applied research. The aim of this study was to evaluate the potential of human artificial chromosome (HAC) for gene delivery and expression in hESc and hiPS cells. HAC offers many potential advantages including the provision for carrying large genes with corresponding regulatory elements to obtain long-term regulated gene expression. In addition, they can replicate and segregate independently without integration into the host cell genome. To develop HAC in hiPS cells, the first part of the study was aimed at generating hiPS cells utilising the Herpes Simplex Virus (HSV)-1 amplicon system. With the use of EBNA-1/OriP retention elements incorporated into the HSV-1 amplicon vectors, hiPS cells completely free of vector and transgenes sequences were successfully derived from human embryonic fibroblasts. The hiPS cells exhibited proliferation and differentiation potential similar to that of hESc. In the second part of the study, development of HAC in hESc and hiPS cells was assessed by utilising the HSV-1 amplicon system to deliver the HAC DNA. Analysis of the hESc confirmed the presence of functional HAC which replicated the behaviour of the host chromosomes. Additionally, HAC generation did not lead to impairment in the developmental potential and pluripotency of hESc. The hiPS cells supported HAC at low frequency but DNA also integrated into the host chromosomes. The HAC system, therefore, needs further refinements to improve the frequency of HAC formation and reduce the chromosomal integration of HAC constructs in hiPS cells. Overall, these findings provide a simple and safe way of pluripotency induction and genetic modification of pluripotent stem cells using the HSV-1 amplicon system and represent an important advance towards patient specific gene and cell therapy.

Abstract:
This thesis explores the creative function of the pseudonym Sara Sand, used by the author Stina Aronson for four literary works in the late 1920s and early 1930s. Två herrar blev nöjda (Two gentlemen were content), Fabeln om Valentin (The tale of Valentin), Tolv hav (Twelve oceans) and Syskonbädd (Siblingbed) are modernist and experimental works that I approach with an experimental way of reading. In order to emphasize what I find to be an inherent strive for freedom within these texts, I draw from Gilles Deleuze and Félix Guattaris notion of desire as a productive force and their view of the text as a rhizomatic structure, as presented in Anti-Oedipus (1972) and A Thousand Plateaus (1980). The characters in Sara Sands writing are in constant movement and in continuous processes of becomings which challenges any kind of fixity, whether that be social structures and hegemonic discourses or simply the individual self.

Abstract:
The development of safe and efficient gene delivery systems in pluripotent human embryonic stem cells (hESc) is essential to realising their full potential for basic and clinical research. The purpose of this study was to develop an efficient, non-integrating gene expression system in pluripotent hESc using human artificial chromosomes (HAC). Similar to endogenous chromosomes, HAC are capable of gene expression, replication and segregation during cell division. Unlike retroviral-mediated gene delivery vectors, HAC do not integrate into the host genome and can encompass large genomic regions for the delivery of multiple genes. Despite the advantages HAC offer, their use has been limited due to laborious cloning procedures and poor transfection efficiencies, and thus only studied in immortalised and tumour-derived human cell lines. In this study, the high transduction efficiency of herpes simplex virus type-1 (HSV-1) amplicons was utilised to overcome the described difficulties and delivered HAC vectors into pluripotent hESc. Analysis of stable hESc clones showed that de novo gene-expressing HAC were present at high frequencies ranging from 10-70% of metaphases analysed, without integrating into the genome. The established HAC contained an active centromere, and were stably maintained without integration or loss in the absence of selection for 90 days. Stable HAC-containing hESc clones retained their pluripotency as demonstrated by neuronal differentiation, in vitro germ layer and teratoma formation assays. HAC gene expression persisted, with some variation, post-differentiation in the various deriving cell types. This is the first report of successful de novo HAC formation in hESc for gene expression studies. These findings show potential for delivering high-capacity genomic constructs safely and efficiently into pluripotent cells for the purpose of genetic manipulation and ultimately patient-specific somatic gene therapy.

Dr. Eleine Mad är Madeleine Jacobsssons talesperson för dom vetenskapliga och paranormala upptäckter som uppstår i hennes världar. Hon beskriver innehåll, teknik och estetik utifrån ett kategoriseringssytem där konsten delas upp som olika typer av komponenter och därefter avkodas dessa allteftersom. För att förstå intuitionens inblandning i arbetsprocessen omförvandlas den till tre separata roller av en Sökare, Samlare och Myntare. Med rollerna försöker jag beskriva på vilka sätt som intuitionen är till gagn eller av förödelse för det konstnärliga arbetet. Sagan om M handlar om en grodlik karaktär, Delop, som lämnar sin hemplanet för att uppsöka andra världar. I sitt sökande hittar Delop ett folkslag vars syn och levnadssätt skiljer sig från hennes erfarenheter av “verkligheten” såsom hon lärt sig att överleva i den.
Dr. Eleine Mad is Madeleine Jacobsson's spokesperson for the scientific and paranormal discoveries that arise in her worlds. She describes content, tecniques and aesthetics based on a categorization system where art is divided into different types of components and then decoded as they go. To understand the intuition's involvement in the work process, it is transformed into three separate roles by a Seeker, Collector and a Myntare(In swedish language the one who is a "myntare" -is verbally declaring a concept or term). With these roles I try to describe in what ways intuition is beneficial or devastating to the artistic work. The story of M is about a frog-like character, Delop, who leaves the home planet to seek out other worlds. In her search, Delop finds a world whose views and lifestyles differ from her experiences of "reality" as she learned to survive in it.

Abstract:
The ideas of Mexican Machismo have been crystallized in the image of the Macho, a virile man who represents the ideals of masculinity in a determined time and space. This work aims to examine how four Mexican Novels (Juan Rulfo’s Pedro Páramo, Elena Garro’s Los Recuerdos del Porvenir, Yuri Herrera’s Trabajos del Reino, and Fernanda Melchor’s Temporada de huracanes) present their unique macho ideals, and how the male characters fail to fulfill them. Through a textual examination of the four novels, this work asks: how is a macho image formed in each pair of novels? And most importantly how do male characters react when they are unable to uphold the masculine values? Chapter one examines Juan Rulfo’s and Elena Garro’s novels, focusing on the downfall of the machos due to the loss of a loved woman, and the strategies the men use to control their towns. Chapter two analyzes Yuri Herrera’s and Fernanda Melchor’s novels, explaining how masculinity is tied to a social performance, and how the machos lose the approval of their group. Chapter three deals with the reaffirmation of power through isolation of female characters and the concept of emasculation as a social and psychological phenomenon. Emasculation, this work strives to prove, is a key element in the four novels, uniting the texts through the social disgrace of a man who does not perform as expected.
