Abstract:
Although there is abundant evidence that individual microRNA (miRNA) loci repress large cohorts of targets, large-scale knockout studies suggest that most miRNAs are phenotypically dispensable. Here, we identify a rare case of developmental cell specification that is highly dependent on miRNA control of an individual target. We observe that binary cell fate choice in the Drosophila melanogaster peripheral sensory organ lineage is controlled by the non-neuronally expressed mir-279/996 cluster, with a majority of notum sensory organs exhibiting transformation of sheath cells into ectopic neurons. The mir-279/996 defect phenocopies Notch loss of function during the sheath–neuron cell fate decision, suggesting the miRNAs facilitate Notch signaling. Consistent with this, mir-279/996 knockouts are strongly enhanced by Notch heterozygosity, and activated nuclear Notch is impaired in the miRNA mutant. Although Hairless (H) is the canonical nuclear Notch pathway inhibitor, and H heterozygotes exhibit bristle cell fate phenotypes reflecting gain-of-Notch signaling, H/+ does not rescue mir-279/996 mutants. Instead, we identify Insensible (Insb), another neural nuclear Notch pathway inhibitor, as a critical direct miR-279/996 target. Insb is posttranscriptionally restricted to neurons by these miRNAs, and its heterozygosity strongly suppresses ectopic peripheral nervous system neurons in mir-279/996 mutants. Thus, proper assembly of multicellular mechanosensory organs requires a double-negative circuit involving miRNA-mediated suppression of a Notch repressor to assign non-neuronal cell fate.

Abstract Abstract 5180 Alpha-thalassemia (α-thalassemia) has two clinically significant forms: hemoglobin Bart hydrops fetalis (Hb Bart) syndrome and hemoglobin H (HbH) disease. HbH disease is characterized by microcytic hypochromic hemolytic anemia, hepatosplenomegaly, mild jaundice, and sometimes thalassemia-like bone changes. Diagnostic of Alfa Thalassemia: Classic testing for α-thalassemia includes: hematologic testing of red blood cell indices, peripheral blood smear, supravital stain to detect RBC inclusion bodies, and qualitative and quantitative hemoglobin analysis. HBA1, the gene encoding α1-globin, and HBA2, the gene encoding α2-globin, are the two genes most commonly associated with α-thalassemia. Molecular genetic testing of HBA1 and HBA2 detects deletions in about 90% and point mutations in about 10% of affected individuals. Objective: Recently have been developed new parameters and information in the new automated hematology analyzer called DxH8008™ from Beckman Coulter as @MSCV, @RSF, @MAF, @LHD% and many morphological parameters for RBC and Reticulocytes calles Cell Population Data. All this parameters may be used to create flagging for laboratory use only (LUO) or Research use only (RUO). The purpose of this study is to investigate the possible use or utility of this new information for the screening/flagging of Alfa Thalassemia. Patient and Methods: We have collected 129 patients with Alfa Thalassemia Intermedia (HbH disease). All of them were confirmed by red cell morphology, Hgb Electroforesis, cromatography in liquid phase in human whole blood for the determination of Hemoglobin A2, F, A1c, and identification of abnormal hemoglobins and DNA analysis (DNA Analysis by GAP-PCR). We have compared these patients with a control group (184 individuals) and with other anemias (see Table 1). Results: Using ROC analysis, the best parameters differentiating the HbH Disease from the normals were: RDW (AUC 1. 000), @LHD(AUC 1. 000), @MAF(AUC 1. 000), @MCNRET (AUC 1. 000), MCV (AUC 0. 999), @MCRET (AUC 0. 999), @RSF (AUC 0. 998), HGB (AUC 0. 996), @MSCV (AUC 0. 995). Using ROC analysis, the best parameters differentiating the HbH Disease from other anemias (excluding normals) were: @LHD(AUC 0. 957), @MCNRET (AUC 0. 946), @MCRET (AUC 0. 902), @MAF(AUC 0. 873), MCV (AUC 0. 869). Using logistic regression we found a discrminant function that permits to differentiate/flag perfectly the patients with HbH disease from other anemias, and of course from normals: AUC 0. 996) Sensitivity: 91. 47% Specificity 94. 68% with a percent of cases correctly classified of: 93. 67 %. AbstractObjectiveMeasurement errors in dietary data lead to attenuated estimates of associations between dietary exposures and health outcomes. The present study aimed to compare and evaluate different approaches of handling implausible reports by exemplary analysis of the association between dietary intakes (total energy, soft drinks, fruits/vegetables) and overweight/obesity in children.DesignCross-sectional multicentre study.SettingKindergartens/schools from eight European countries participating in the IDEFICS Study.SubjectsChildren (n 5357) aged 2–9 years who provided one 24 h dietary recall and complete covariate information.ResultsThe 24 h recalls were classified into three reporting groups according to adapted Goldberg cut-offs: under-report, plausible report or over-report. In the basic logistic multilevel model (adjusted for age and sex, including study centre as random effect), the dietary exposures showed no significant association with overweight/obesity (energy intake: OR=0·996 (95 % CI 0·983, 1·010); soft drinks: OR = 0·999 (95 % CI 0·986, 1·013)) and revealed even a positive association for fruits/vegetables (OR = 1·009 (95 % CI 1·001, 1·018)). When adding the reporting group (dummy variables) and a propensity score for misreporting as adjustment terms, associations became significant for energy intake as well as soft drinks (energy: OR = 1·074 (95 % CI 1·053, 1·096); soft drinks: OR = 1·015 (95 % CI 1·000, 1·031)) and the association between fruits/vegetables and overweight/obesity pointed to the reverse direction compared with the basic model (OR = 0·993 (95 % CI 0·984, 1·002)).ConclusionsAssociations between dietary exposures and health outcomes are strongly affected or even masked by measurement errors. In the present analysis consideration of the reporting group and inclusion of a propensity score for misreporting turned out to be useful tools to counteract attenuation of effect estimates.

Recently non-steroidal anti-inflammatory drugs have seemed to reduce the frequency of post-ERCP pancreatitis in some prospective controlled trials, but the results have to be confirmed by further studies. Aim: to evaluate the efficacy of rectally administered indomethacin for the reduction of incidence of post-ERCP pancreatitis. Method: A prospective randomized placebo-controlled study was conducted in 228 patients who underwent ERCP. Patients were randomized to receive a suppository containing 100 mg indomethacin or an inert placebo 10 mins before ERCP. Patients were evaluated clinically and biochemically by using serum amylase levels measured 24 h after the procedure. Results: Pancreatitis and hyperamylasemia occurred more frequently in the placebo group, but the difference was not significant. In respect to the rate of pancreatitis, this tendency could particularly be observed in females, in patients older than 60 years and in patients with BMI lower than 25; however, it completely failed in cases with pancreatic duct filling or in those with pancreatic EST. Conclusions: Rectal indomethacin given before ERCP did not prove to be statistically effective in the reduction of the incidence of post-procedure pancreatitis. Further, controlled multicenter studies are required to assess safely the potential efficacy of indomethacin in the prevention of pancreatitis following ERCP. Orv. Hetil., 2012, 153, 990–996.

Silver sillago (Sillago sihama) is a commercially important marine fish species in East Asia. In this study, we compared the transcriptome response to hypoxia stress in the gill tissue of S. sihama. The fish were divided into four groups, such as 1 h of hypoxia (hypoxia1h, DO = 1.5 ± 0.1 mg/L), 4 h of hypoxia (hypoxia4h, DO = 1.5 ± 0.1 mg/L), 4 h of reoxygen (reoxygen4h, DO = 8.0 ± 0.2 mg/L) after 4 h of hypoxia (DO = 1.5 mg/L), and normoxia or control (DO = 8.0 ± 0.2 mg/L) groups. Compared to the normoxia group, a total of 3550 genes were identified as differentially expressed genes (DEGs) (log2foldchange > 1 and padj < 0.05), including 1103, 1451 and 996 genes in hypoxia1h, hypoxia4h and reoxygen4h groups, respectively. Only 247 DEGs were differentially co-expressed in all treatment groups. According to Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, DEGs were significantly enriched in steroid biosynthesis, biosynthesis of amino acids, glutathione metabolism and metabolism of xenobiotics by cytochrome P450, ferroptosis and drug metabolism—cytochrome P450 pathways. Of these, the cytochrome P450 (CYP) and glutathione S-transferase (GST) gene families were widely expressed. Our study represents the insights into the underlying molecular mechanisms of hypoxia stress.

Abstract:
Most butterflies feed on nectar, while some saprophagous butterflies forage on various non-nectar foods. To date, little is known about the genomic and molecular shifts associated with the evolution of the saprophagous feeding strategy. Here, we assembled the high-quality chromosome-level genome of Hestina assimilis to explore its saprophagous molecular and genetic mechanisms. This chromosome-level genome of H. assimilis is 412.82 Mb, with a scaffold N50 of 15.70 Mb. In total, 98.11% of contigs were anchored to 30 chromosomes. Compared with H. assimilis and other Nymphalidae butterflies, the genes of metabolism and detoxification experienced expansions. We annotated 80 cytochrome P450 (CYP) genes in the H. assimilis genome, among which genes belonging to the CYP4 subfamily were significantly expanded (p < 0.01). These P450 genes were unevenly distributed and mainly concentrated on chromosomes 6–9. We identified 33 olfactory receptor (OR), 20 odorant-binding protein (OBP), and six gustatory receptor (GR) genes in the H. assimilis genome, which were fewer than in the nectarivorous Danaus plexippus. A decreased number of OBP, OR, and GR genes implied that H. assimilis should resort less to olfaction and gustation than their nectarivorous counterparts, which need highly specialized olfactory and gustatory functions. Moreover, we found one site under positive selection occurred in residue 996 (phenylalanine) of GR genes exclusive to H. assimilis, which is conservative in most lineages. Our study provides support for the adaptive evolution of feeding habits in butterflies.

ABSTRACT Adeno-associated virus type 2 (AAV) is a single-stranded-DNA-containing, nonpathogenic human parvovirus that is currently in use as a vector for human gene therapy. However, the transduction efficiency of AAV vectors in different cell and tissue types varies widely. In addition to the lack of expression of the viral receptor and coreceptors and the rate-limiting viral second-strand DNA synthesis, which have been identified as obstacles to AAV-mediated transduction, we have recently demonstrated that impaired intracellular trafficking of AAV inhibits high-efficiency transduction of the murine fibroblast cell line, NIH 3T3 (J. Hansen, K. Qing, H. J. Kwon, C. Mah, and A. Srivastava, J. Virol. 74:992–996, 2000). In this report, we document that escape of AAV from the endocytic pathway in NIH 3T3 cells is not limited but processing within endosomes is impaired compared with that observed in the highly permissive human cell line 293. While virions were found in both early and late endosomes or lysosomes of infected 293 cells, they were localized predominantly to the early endosomes in NIH 3T3 cells. Moreover, treatment of cells with bafilomycin A1 (Baf), an inhibitor of the vacuolar H+-ATPase and therefore of endosomal-lysosomal acidification, decreased the transduction of 293 cells with a concomitant decrease in nuclear trafficking of AAV but had no effect on NIH 3T3 cells. However, after exposure of NIH 3T3 cells to hydroxyurea (HU), a compound known to increase AAV-mediated transduction in general, virions were detected in late endosomes and lysosomes, and these cells became sensitive to Baf-mediated inhibition of transduction. Thus, HU treatment overcomes defective endocytic processing of AAV in murine fibroblasts. These studies provide insights into the underlying mechanisms of intracellular trafficking of AAV in different cell types, which has implications in the optimal use of AAV as vectors in human gene therapy.

A descriptive study examining 4-H programs in Africa, Asia, and Europe was conducted for the National 4-H Council. Program size, scope, structure, organization, and funding varied greatly by country and few of the programs were connected to a university, but many partnered with other 4-H organizations around the world. Program participants were found to be primarily male with few programs specifically for women. A list of content areas provided by the 4-H programs was also obtained and compared to top agricultural commodities in their country. Very few content areas offered by the country aligned with their major agricultural commodities even though programs were available in the United States. The Global 4-H Network has the potential to fill in these holes and provide additional opportunities to global programs.

The aim of this thesis is to search for the best way to allocate revenues gathered from a group of products in a household supplies company. In so doing, it purports to determine the price which brings customer perception and organizational benefit to equilibrium. To compare alternatives of revenue allocation methods, data obtained for a main product and its variants from a household company will be analyzed in an organized manner. Three ways of product bundling (pure bundling, mixed bundling, unbundling) is discussed as a framework for underlying different detailed aspects. In the end, pricing and promotional policies of the company is critically evaluated and simultaneous strategy changes are suggested.

, a notion that is relatively new and opens to the scientific debates. In the thesis, a decade-based analysis of the transformation in China is provided. In this regard, this thesis identifies the period between Deng&rsquo

launching of the reforms&rsquo

It is argued that the reforms introduced in the 1980s are of vital importance in terms of abandoning the legacies of Maoist period and the construction of the institutions of a capitalist market economy in China. This thesis analyzes the role and power of symbols in the identity formation of community members. With regards to different sociological traditions it discusses the concepts of symbol, identity and community in three chapters and makes analyses via employing three specific samples i.e., of dressing, Cross and language mainly from the view point of semiology and symbolic interactionism in the following chapter by integrating the three concepts elaborated separately to indicate and emphasize the role and power of symbols in the identity formation of community members. In the conclusive chapter it presents an overall remark of the whole study by emphasizing the effect and influence of the concepts of symbol, identity and community on each other. The thesis argues that consciously or not everyone convey their messages through the use of symbols. Therefore, from dressing to managed speech and interaction everyone come on the stage fully equipped and loaded with these symbols, which would in the last analysis be definitive in determining the borderlines of this symbolic world within their specific community which is nothing but a pool of symbols.

This dissertation intends to undertake an analysis of one of the most deeply-rooted dichotomies in Turkey&rsquo
s political and cultural history, -the Islamist-Kemalist divide- through a cultural, interdisciplinary and gender-conscious approach. Both the Kemalist and the Islamist identities situate themselves vis-à
-vis the Other, as if they were mutually exclusive entities. However, when and if these formulations are approached as culturally shaped discursive practices, it is also revealed that they operate with and within similarities, continuities and hybridities. Intellectuals on both sides derive their metaphors from a common cultural and rhetorical pool. The cultural analysis of seemingly opposite ideological positions in Turkish political transformation through the gender lens in general and masculinities in particular identifies the various sites of social power that exist in Turkish society today. The study pays particular attention to conceptualizations of masculinity and femininity accompanying Turkey&rsquo
s modernization. The relative newness of the subject matter, the interdisciplinary approach it necessitates, and the recentness of the theoretical literature and methodological applications, as well as the paucity of empirical work in the context of Turkey employing these parameters necessarily draws the limits of this work as well as showing for the multidisciplinary, &ldquo
unorthodox&rdquo
character of the approach. The study contends that such a cultural analysis of Turkish political transformation through the lens of gender in general and masculinities in particular might create a new epistemological terrain, one that goes beyond the current epistemologies mired in ontological dualities.

The aim of this thesis is to investigate the structure and the effectiveness of the investment incentive system in Turkey. The theoretical definition and the economic and social targets of incentives, and the direction, problems and the effectiveness of the investment incentive system are investigated. This study finds that the most prominent target of investment incentives is to alleviate the unemployment problem in Turkey. The regional investment incentives seem to be successful in the western part of the country to some extent. The investment incentive system does not seem to be successful in directing the investment to underdeveloped regions of the country. The investment incentive system has no strong effects on directing the sectoral distribution of investments.

Abstract The helical-coiled once-through steam generator (H-OTSG) is a key equipment in small modular reactor (SMRs) to be used in heat exchanging between primary side and secondary side. In this study, a 1D Code HTAC (H-OTSG Thermo-hydraulics Analysis Code) is developed to simulate the heat transfer characteristic of H-OTSG and used to prepare the experimental study in future. The H-OTSG is composed by 74 helical tubes which is divided into 10 layers according to the coil diameters. Firstly, a 1D code of H-OTSG is developed with the tube-side thermal-hydraulic model proposed by the present authors and the shell-side model using the thermal-hydraulic model of straight-tube bundle in cross flow. Secondly, 10 inclined straight pipes with different coil curvature ratio are stablished in the 1D code to respectively simulate the helical tube bundle of 10 layers and the shell-side of H-OTSG is model with a vertical annulus. Furthermore, the sensitivity study on the node number is conducted and 99 nodes for each pipe and annulus are fine enough. Thirdly, the simulated result shows the 1D code can used to predict the steady-state heat transfer rate of the H-OTSG under various thermal power. Finally, the sensitivity analysis of the system parameters on the heat transfer rate of H-OTSG is conducted, and the result shows the decrease of tube-side pressure and the increase of shell-side inlet temperature can rise the average HTC of H-OTSG.

Abstract Because of their characteristic spray geometry, pressure-gas-atomizers can be used to create thick coatings from molten metal. Production rates of pressure-gas-atomizers are substantially higher compared to conventional Thermal Spraying (100 – 200 kg/h based on molten tin). The shape of the spray cone can be designed by the geometry of the atomizer. The mean particle sizes and velocities (molten tin and tin-copper alloys) are controlled by the gas flow. Powder products and coatings of several millimeters on steel substrates were investigated. The average density of the layer was higher than 99%.

Bartram ex Marshall is one of the most important economically (wood production), environmentally (biomass production and carbon sequestration), and fast growing species in plantations. Therefore, the purpose of this study was to investigate DBH, height and carbon sequestration models according to the age using Stem Analysis Method in poplar plantations of Guilan province, Iran. Thirty trees were randomly selected in different diameter classes, fell down and the discs were obtained in order to stem analysis. The annual rings of discs counted, age at different tree heights obtained, and the annual rings diameter were measured to determine annual diameter and volume growth. Biomass was measured and finally, regression analysis performed by the relationship between DBH and height by carbon in age based on the highest coefficient of determination and minimum standard error. Abstract This paper describes and evaluates the performance of a Helium Recovery System (HRS) designed for cold spraying. A flexible, automated, full scale HRS system has been designed and installed in the McGill Aerospace Materials & Alloy Development Center Cold Spray Facility, located at and in collaboration with the National Research Council of Canada. The fully automated HRS has been designed to recover helium from the cold spray chamber with sufficient purity (&gt;99%) and flow capacity (5 to 220 Nm3/h), allowing for a cost-effective operation by insuring a recovery rate of above 85%. In addition, a comparison of titanium coating properties obtained by using both He and N2 as propellant gas is presented.

The regeneration method of zirconium hydroxide sorbent spent in the boron sorption process by diluted solution of acids as well as the principle recycling diagram of the formed regenerating solution by the nanofiltration process with the OPMN-P membrane were proposedin this work. The permeate obtained can be used as a boron-containing microfertilizer (ammonium salt + boron) after it has been neutralised with ammonium hydroxide. Zirconium-containing concentrate may be reused for sorbent preparation. The effectiveness of separation of the regenerating solution components by nanofiltration is demonstrated by high values of zirconium rejection coefficient in this process (RZr ~98– 99%) and low values of boron rejection coefficient (RB ~12%). In this case the specific performance of the membrane is about 50 dm3/m2·h.

Under sponsorship of the Morgantown Energy Technology Center of the Department of Energy, the Westinghouse Electric Corporation and Avco Research Laboratory/Textron (ARL) are developing a direct coal-fired 80-MW combustion turbine. This is to be an element in a 207-MW direct coal-fired combustion turbine combined cycle, comprising another combustion turbine, a heat-recovery steam generator, and a steam turbine. Following conceptual and economic studies, the first experimental task has been to develop a direct coal-fired slagging combustor in small scale. A year’s testing with this unit has now been accomplished. It is thus possible to discuss experimental results in the areas of slag management, general combustor operability, coal fuel feed, heat loss, pressure loss, combustion efficiency, and emissions. Westinghouse and ARL selected the slagging combustor for use in this program because of its unique ability to accept inexpensive standard utility-type coal without sending excessive pollutants and particulates to the expander-turbine. It accomplishes this by burning the coal at a temperature high enough to melt the ash, and then removing the molten slag with an impact separator. This combustor is also of the rich-lean type, which allows it to emit very low levels of NOx. Sulfur oxides are controlled through injection of a sorbent which combines with gaseous forms of sulfur in the rich zone, and then takes them out with the slag. The plan in this program is to develop the slagging combustor in small scale at 12 million Btu/h (12.66 million kJ/h) heat input, 6 atmospheres pressure, and then move to design and verification testing of a full-scale 70 million Btu/h (73.85 million kJ/h), 14-atmosphere unit. It is with the smaller burner that this paper is concerned. During this period, coals have been fed into the 6-atm combustor exclusively in the dry-pulverized form with air as the carrier gas. To date, high- and low-sulfur eastern bituminous, and low-sulfur western subbituminous coals have been evaluated, with, and without sorbents, which have included various calcium and iron containing minerals. Coal samples have been of the 200 mesh size, and have not been beneficiated. Combustor operation has been efficient and repeatable and slag flow has been symmetrical and manageable. Slag removal has been greater than 90 percent and carbon burnout has been greater than 99 percent. Nitrogen oxide emissions meet the goals of EPA’s NSPS for coal-fired plants. Current tests are focused on the goal of 90 percent sulfur removal. Although some results are available, various sorbents and injection techniques continue under test.

Abstract:
This paper offers an interactive scenario of Cool Guy's Adventure for children with Attention Deficit Hyperactivity Disorder (ADHD) based on sensory integration (SIT). We present a design framework that allows occupational therapists to arrange and combine game units and difficulty levels. ADHD is one of the most common neurodevelopmental disorders of childhood. Many studies have utilized HCI technology to assist and treat patients with various mental conditions, helping them lead healthier lives. However, there has been limited attention given to the development of HCI technologies to support children with ADHD. The game was developed using the Unity3D game engine and Microsoft Kinect®, each game design focuses on sensory integration training and enhances the child's intrinsic motivation: Throw the perfect ball, Play hopscotch, and Capture treasure. Abstract:
The Michaelis-Menten (Km ), catalytic (kcat ), and specificity (kcat/Km ) constants were determined for humSft α- and β-thrombins with the chromogenic substrate S-2238 (H-D-Phe-Pip-Arg-pNA), Chromozym-TH (Tos-Gly-Pro-Arg-pNA), and Spectrozyme-TH (H-D-HHT-Ala-Arg-pNA) in 0.15 M NaCl buffered with 10 mM HEPES ancTlO mM Tris-HCT at pH 7.4, 37°C. Spontaneous hydrolysis was insignificant for these substrates. Both S-2238 and Spectrozyme-TH exhibited limiting solubilities at ∼35 μM, while Chromozym-TH did not do so up to 50 μM. From initial estimates, Km's and k t's were refined by computer Gause-Newton iterations or nonlinearneast-square fits for the concentration of p-nitroanalide formed per second versus the initial substrate concentration. No major differences were found between a-thrombin (99% α, 91% esterolytically active enzyme, and > 3,500 kilo-U.S. clotting units/g with fibrinogen) and Yγ-thrombin (98% γ. 89% active enzyme, and < 10 kilo-units/g). For α- versus γ-thrombin and the three substrates, respectively, the Km 's were 6.75 ± 0.13 vs 7.62 ± 0.30, 18.4 ± 0.4 vs 23.0 ± 0.3, and 2.53 ± 0.02 vs 3.85 ± 0.51 μM; the kcat> s were 125 ± 1 vs 134 ± 2, 181 ± 2 vs 130 ± 1, and 35.5 ± 0.2 vs 52.4 ± 2.0 s−1 ; and the kcat/Km 's were 18.5 vs 17.6, 9.84 vs 5.65, and 10.1 vs 13.6 s−1 μm . These values closely approximate those of 7.2 ± 0.9 μM, 84 ± 4 s−1 , and 11.7 s−1 μM−1 determined by Higgins, Lewis, and Shafer (J. Biol. Chem. 258:9276-9282, 1983) for the Aα cleavage of human fibrinogen by human α-thrombin under physiologically relevant conditions. Thus, these chromogenic substrates have thrombin specificities similar to that of fibrinogen, although their amidolytic activities are independent of additional active-site regions required for fibrinogen clotting activity (α vs γ-thrombin). Fibrinogen interactions with such active-site regions might account for why the fibrinogen Aa site is an atypical thrombin susceptible bond and the high species variability of fibrinopeptides.
