Abstract:
The Reformed Kirk of Scotland has a reputation for vigorous repression of festivity hardly to be surpassed by any other Protestant church. From 1560 on, the authorities of the kirk from local session to General Assembly waged a stern and unremitting campaign against the celebration of Yule, Easter, May Day, Midsummer, and saints' days; against feasting, whether at marriages or wakes; against Sunday sports and dancing and guising—in short, against anything that might distract newly Reformed laity from the central Protestant focus on the sermon, the Bible, and the godly life of moral discipline and prayer. Their goals were both negative and positive. They were intent on quashing popish superstition in part because they were convinced that its persistence helped to explain the recurrence of plague, famine, and other divine judgments on the land. In a more positive sense, they were trying to clear the way for construction of a new vision of the good life—one that exalted word over image and discipline over ritual, and in which community cohesion was achieved by a shared Protestant identity and the common goal of a devout and orderly society. Modern observers generally credit their campaign with remarkable success; certainly the popular image of Scotland after the Reformation is grim and joyless—and with some reason. Days of fasting and humiliation under presbyterian rule came close to outnumbering the old saints' days, and they vastly outnumbered new days of thanksgiving. Official policy on the matter of festivity was exemplary for a Reformed nation.

Abstract:
Mulberry (Morus L.) has become an important crop throughout the world due to its fruits have been industrially exploited for various commercially valuable products. Many studies on mulberry related to genetic diversity, fruit quality, and breeding programs have been carried out, but little information on mulberry rootstocks is available, especially the possibility of applying grafting to improve the fruit quality. Here, we evaluated the effects of 8 different rootstocks on the fruit quality of ‘Zijing’ mulberry. Twelve fruit quality traits were extremely different except for the fruit shape index (FSI). ‘Zijing’ on ‘Zheza 2’ had the highest fruit weight (FW) and size, as well as titratable acidity (TA), but lower levels of other compounds content except the total soluble solids content (TSS) were detected. ‘Yuesang 51’ exhibited the highest soluble sugar content (SSC), reducing sugar content (RSC), SSC/TA ratio, anthocyanin content (AC) and the lower TA. In contrast, the lowest TSS, SSC and RSC were shown in ‘Guisang 5’. Moreover, ‘Guisang 12’ exhibited the highest TSS and soluble protein content (SPC). The highest vitamin C content (VC) was observed in ‘Guisang 6’. ‘Tang 10 × Lun109’, Zhenzhubai seedlings, ‘Yuesang 11’ together with ‘Yuesang 51’ had the lowest and similar levels of TA. Most importantly, these fruit quality traits were evaluated by principal component analysis (PCA), and ‘Yuesang 51’ with good comprehensive fruit quality was screened out, followed by ‘Guisangyou 12’. Overall, these results contribute to evaluating the roles of different rootstocks on improving fruit quality of mulberry.

This article is devoted to the systematization and critical analysis of the experience of various countries of the world in organizing the population evacuation from areas of various techno-logical disasters: accidents at radiation-hazardous objects and chemical facilities, as well as dam failures. The article provides an assessment of the role of public notification about the danger, speed and awareness of decision-making by the responsible authorities, prepared-ness of the infrastructure for transportation and resettlement of a large number of people in successful evacuation and minimization of human losses and the number of injured. A distin-guishing feature of technological disasters is the suddenness of the deployment due to the predominant impact of the human factor. In this connection, recommendations are given re-garding the need for preventive preparation of the population and infrastructure for potential disasters in threatened areas.

The synthesis of graphene directly on Ge and on Ge deposited on Si provides a scalable route toward integrating graphene onto conventional semiconductors. This presentation will first survey the growth modes of graphene on Ge(001), Ge(011), Ge(111), Ge(112), Ge(001)-6°, and Ge(001)-9° via chemical vapor deposition (CVD), reporting on the effect of Ge surface orientation on graphene island formation and shape, strain in large-area graphene films, and the nanofaceting of the Ge below graphene.[1] We will then focus on the anisotropic growth of semiconducting graphene nanoribbons on Ge(001) and Ge(001)-9° and of nominally single crystal graphene on Ge(110). On Ge(001), we have discovered how to drive graphene crystal growth with a large shape anisotropy through control of kinetics.[2-6] This discovery enables the direct synthesis of narrow, armchair, semiconducting nanoribbons. The ribbons are self-orienting, self-defining, and have smooth edges. The ribbons exhibit excellent transport properties (e.g., high on-state conductance and on/off ratio at room temperature) and provide a promising pathway towards the direct integration of high-performance nanocarbon electronics onto conventional semiconductor wafer platforms. On Ge(001), nominally single crystal graphene has been reported in limited cases; however, conflicting studies have evidenced polycrystallinity. Abstract:
The rational organization of the production infrastructure of an industrial enterprise has a significant impact on the level of profitability of production. New technologies for integrating business processes are evolving in the face of industrial automation. The aim of the article is to diagnose the level of economic dependence of the core production on the production infrastructure. As a research method, the algorithm of the ensemble method of machine learning “Random Forest” is proposed. The parameters that quantitatively and qualitatively describe the costs of main and auxiliary production, the costs of repair facilities, and the level of production efficiency have been developed. Approbation of the algorithm on the example of chemical enterprises allowed distin-guishing three classes of productions by the nature of the organization of production infrastructure and its par-ticipation in the core production. The quality of the obtained models is evaluated by calculating the risk of mis-classification and the magnitude of cumulative lift, where the class with the most correct classification results is highlighted. Results obtained are primary diagnostics of the organization and capacity utilization of the produc-tion infrastructure in order to make decisions on business process restructuring.

The peer review method is one of the problem learning methods. Its essence lies in the mutual study of each other’s written works by students in order to comment and evaluate them for further revision. Like any teaching method, the peer review method has linguodidactic properties – characteristics that underlie it, essential for the methods of teaching foreign languages and distin-guishing this method from others. Also, the method of peer review has specific linguodidactic functions – an external manifestation of the linguodidactic properties of the teaching method, which show what educational and cognitive tasks are solved by using this method in practice in foreign language teaching. Based on the analysis of the literature, the following linguodidactic functions of this method are identified: a) the development of student mentoring in teaching; b) sharing of responsibility by students for mastering the educational material; c) the use of blended learning in the implementation of the peer review method; d) changing the role of a teacher from a “bearer of knowledge” to a moderator of educational and cognitive activities of students; e) changing the ratio between classroom and extracurricular educational and cognitive activities of students; f) reduction of the terms of checking students’ training written works. The work describes in detail each of the selected functions.

The study of the specifics of publicistic style and its linguistic and stylistic peculiarities is relevant and it is caused by the linguists’ increased attention to mass media texts as a means of influence on the audience and the importance of a more complete examination of the linguistic text features. The lexical composition of newspaper texts is extremely diverse as the newspaper is a means of reflecting modern language development, and the periodical press uses various expressive means. The following English-speaking newspapers The Daily Mail, The New York Times, The Telegraph, The Guardian, The Times, Daily News, BBC News, Los Angeles Times, Esquire and The Washington Post , provide analysis of expressive means used with a view to distin-guishing epithets and examining their distribution and use in the periodical press. The article researches and illustrates different types of epithets distinguished on a variety of principles: in terms of their semantics, structure, nature of nomination and type of connotative meaning. From the semantic perspective there exist common language and individual epithets. In terms of nature of nomination epithets are classified into metaphoric and metonymic. The periodical press verifies use of all the structural epithet types, viz. simple, compound and epithet chains. The expression of the author’s viewpoint can be communicated with the help of epithets with meliorative, pejorative or neutral connotative meaning.

The picture showing the little Joseph Meister being treated against rabies under Louis Pasteur's eyes, on July 6, 1885, has quickly become a symbol of the triumphant progress of medicine, even though diseases with high mortality like tuberculosis or diphtheria could still not be healed with efficient therapeutic means. But before the discoveries of Pasteur, what was actually, in daily practice, the kind of response an ordinary doctor could give to human rabies? A Swiss physician, Charles-Hector Guisan, developed a therapy based on the use of sodium arsenate, which he published in the columns of the Gazette e/e.s //ôpttaux c/v/As et /«///fa/rev in 1854. This arsenic therapy was to be put into practice on a larger scale in the canton of Fribourg by Dr Jean-Louis Schaller (1816-1880), who meticulously wrote observations in a notebook on the cases of 13 persons wounded by a rabid dog in 1855.

Su referente teórico son las teorías del currículum, y su objetivo es reflexionar sobre la cuestión de la enseñanza de la moral con el foco ya mencionado, a través de la defensa de la figura del maestro como intelectual transformativo tomada de Henry Giroux. La idea, entonces, es mostrar que esta figura es la más adecuada para llevar a cabo el ideal de educación moral de Esperanza Guisán y, por tanto, sortear el quiebre entre dos conjuntos de valores discordantes, gracias a la apropiación de algunas ideas que aparecen en los lineamientos curriculares para la enseñanza de la ética, de tal suerte que se logre hacer una práctica transformadora sin tener que alejarse del marco legal colombiano.

In Pakistan intersectoral wage trends have been analysed by Guisinger and Hicks (1978); Irfan (1982) and Irfan and Ahmed (1985). The studies show that over the period 1970 to 1984 real wages of the large-scale manufacturing, construction and agriculture sectors increased while that of the government employees declined significantly. The study shows international migration to be one of the major determinants of the rise in real wages, in addition to important policy interventions. The present study is an extension of Irfan and Ahmed's work. It has been undertaken for two reasons. First there is great scarcity of empirical evidence on this very important issue. Second, the more important factor is the sharp reversal in one of the major variables - out migration. Since 1981 there has been net return migration. This phenomenon a priori is expected to upset the labour market and the wage rates in the opposite direction. The study is planned as follows: Section II describes the trends in real wages between the formal and informal sectors.! Section III describes the factors behind the observed trends in real wages, and finally Section IV gives the conclusions of the study.

Gó rals are a separate ethnocultural group living in the mountainous regions of the Carpathians on the borderlands of Poland and Slovakia (Silesia, Orava, Podhale, Spiš, etc.). In the ethnogenesis of this group, not only Slavic tribes and peoples took part, but also Hungarians, Ger-mans, Aromanians, Turks, etc., which enriched the Gó ral traditional culture with other ethnic elements. Both Slavic and non-Slavic elements of spiritual culture retain their antiquity, that is, the archaic features of the traditional culture of the Gó rals include the cultural archaisms of the peoples inhabiting this region. In various spheres of the spiritual cul-ture of this mountainous region, many archaic elements of the common Slavic era have been preserved, which are also known in diff erent parts of the Slavic world. In funeral rituals, these are, for example, relics of “white mourning”, the custom of lightly hitting the coffi n on the thresh-old when taking it out of the house, a wedding-funeral, prohibitions on a number of works on memorial days so as not to “clog the eyes” of the souls of the dead, etc. In the fi eld of traditional medicine, such common Slavic practices are methods of treating the evil eye and sorcery (extin-guishing coals), the magic of ensuring the life and health of a newborn (passing a baptized child through a window, treating “sukhotka” (con-sumption) by dragging through a kalatch), methods of treating fever (leaving things on the road), the belief that moonlight can cause illness in a child, etc. In folklore, such archaic (at least Balto-Slavic) motives are the transfer of objects from one mountain to another by giants and the motive of the troops sleeping in a mountain. This motive is Celtic in origin (at least in Europe), however, it has deep Indo-European origins.

The aim of our study was to evaluate NT-proBNP in patients with long-standing hypertension and in patients with signs of hypertensive cardiomyopathy. The study included three groups of 50 subjects: healthy persons (Control Group), patients with hypertension and normal left ventricular systolic function (Group 1) and patients with longstanding hypertension and signs of hyper tensive cardiomyopathy with impaired left ventricular systolic function (Group 2). Our results show a very good correlation (Pearson's test) between NT-proBNP in Group 1 and Group 2 and C-reactive protein (Group 1: r = 0.8424; Group 2: r = 0.6650), systolic blood pressure (Group 1: r = 0.7213; Group 2: r = 0.4856), diastolic blood pressure (Group 1: r = 0.4282; Group 2: r = 0.3989) and ejection fraction (Group 1: r = -0.7390; Group 2: r = 0.9111). ROC analysis revealed that the AUC between the Control Group and Group 1 for NT-proBNP (0.912) was not significantly different (p>0.05) from the AUC for systolic (0.924) and diastolic pressure (0.937). A cut-off value for NT-proBNP of 5.89 pmol/L can be used to reliably distinguish patients of Group 1 from the Control Group, and a cut-off value of 21.67 pmol/L reliably separates patients from Group 1 and Group 2 (in both cases, the AUC is 1.000). Patients in Group 2 who belonged to the II and III New York Heart Association (NYHA) class had significantly higher levels of NT-proBNP than those in NYHA class I (ANOVA test, p=0.001). These data suggest that NT-proBNP is a useful biomarker for distin guishing patients with long-standing hypertension who are at risk of heart failure, allowing optimization and proper treatment of these patients.

Important restrictions, both tariff and non-tariff, have been the main policy instrument in implementing the import substitution industrial strategy which has been pursued by almost all the developing countries. The strategy was visualised as a means of realising higher growth of output and foreign exchange earnings, conservation of foreign exchange and stability of the economy. Efficiency in resource use and income distribution considerations did not assume much significance in the development strategy. The industrial sector of Pakistan, protected from imports through severe quantitative restrictions and even bans, suffered from inefficiencies and rigidities. However, this was realized only by the mid-Sixties when Soligo and Stern (1985), on the basis of effective protection rates reached a very startling, though not entirely correct, conclusion that in most of the industries in Pakistan, value added at world market prices was negative! . The study aroused interest in the examination of efficiency levels, both inside and outside Pakistan, through the computation of effective protection rates. These studies include among others, Balassa (1971); Little, Scitovsky and Scott (1970) and the NBER series on the import regime in many countries. Studies relating to effective protection in Pakistan include Soligo and Stern (1985); Lewis and GUisinger (1968); Kemal (1978); Khan (1978) and Naqvi, Kemal and Heston (1983).

The article considers the leading and indisputable role of discursive practices in the existence of social institutions, especially in democratic governance. The necessity of searching for heuristi- cally effective approaches in the analysis of social reality in general, and especially modern soci- ality, is substantiated. In this context, the theoretical modernization of the institutional approach in the analysis of social phenomena by involving the concept of discourse in the structure of this approach is proposed. Emphasis is placed on the dual meaning of social institutions — as ways of organizing the life of society and as instances (mediums) of normative, through which the order of social life is constituted, reproduced, and changed. This solves the “rule-organization” dilem- ma in neo-institutionalism. It is transferred to the mode of two inseparable and complementary functions of the institute — normative and regulative. The analysis of those semantic, organiza- tional, and procedural loadings which carry out discursive practices inactivity of institutes of society is given. In particular, emphasis is placed on the complex structure of normativity em- bodied by the public institution. It should at least highlight the explicit normativity of the system of rules and regulations on the one hand, and the order of discourse created and maintained by this institution, on the other. Discourse corresponds to the normativity in its usability — as a real process of normalization of life. Normativity exists only by generating certain practices of speech, communication, and argumentation (conclusion). This becomes the basis for distin- guishing the concept of an institution as a discursive mode of existence of a social institution on the one hand, and as a way of organizing discourse in society (order of discourse) — on the other. Among the main social effects of institutions are the generation of trust as the basis of social relations and the longevity of cultural experience. Emphasis is placed on the complexity of the existence and activities of institutions: each institution is involved in the general discursive-com- municative field of society, and in turn, influences it.

Premature ovarian failure (POF) is characterized by hormonal disorders, amenorrhea, and premature loss of fertility potential in women of reproductive age. Several studies have been conducted on the effectiveness of traditional Chinese medicine (TCM) in treating POF. TCM relied primarily on apoptosis, immunity, and aging to treat POF based on the studies of domestic and foreign literature. Zuogui pills inhibited mitochondrial-dependent apoptosis in the treatment of POF. Huyang Yangkun formula regulated the downstream of the Bcl-2 family to resist apoptosis through the aquaporin-1 protein. Modified Bazhen decoction regulated apoptosis in POF by regulating X-linked inhibitors of apoptosis protein. Bushen Tianjing recipe was effective in treating POF by promoting angiogenesis and preventing apoptosis. As for immunity, Bushen Jianpi prescription and Er-Xian decoction cured autoimmunity POF models and increased follicular development-related protein expression. Bushen Huoxue Tang improved ovarian function and reduced ovarian inflammation by regulating the Nrf2/Keap1 signaling pathway and T lymphocytes. Taohong Siwu decoction promoted the proliferation and differentiation of granulosa cells of POF mice by regulating the TGF-β1/Smads signaling pathway. In addition, ginsenoside Rg1 and Jiajian Guisheng formula treated POF by regulating cell aging-related mechanisms. Si Wu Tang treated POF by activating the angiogenesis-related proteins. The goal of this review is to serve as a reference for in-depth research into the treatment of POF with TCM and provide inspiration for new diagnostic methods and treatment options.

Introduction. Fire suppression systems are stationary technical means designed for fire extinguishing. Their evo- lution relies on the general level of technological development. At present, automatic fire suppression systems (AFSS) are most widely used; they include sprinkler AFSSs, patented in 1864 by Harrison, UK, as well as new robotic fire suppression systems (RFSS). The purpose of the article is to analyze the trends in the development of fire extinguishing systems, and substantiate Russia’s priority in the development of advanced fire extinguishing technologies on the basis of robotic fire suppression systems (RFSS).Fire suppression systems: from manually operated to robotic ones. Sprinkler fire extinguishing has significant drawbacks; they are low sensitivity and high inertia. Fire monitors are among the main most powerful means of firefighting. Fires are extinguished by firefighters who are in extreme life-threatening environments. The issue of replacing a person during fire extinguishing was studied. Mobile firefighting robots appear in many countries. In practice, stationary firefighting robots are widely used. The first stationary firefighting robot was invented in Russia in 1984 to protect the Kizhi Museum. It was also applied to liquidate the consequences of the accident at the Chernobyl nuclear power plant. The first RFSS was introduced at the Leningrad NPP in 1989. Acting in close cooperation with the VNIIPO EMERCOM of Russia, FR Engineering Centre conducted research to improve the design and control system, establish the regulatory framework for the RFSS. As a result, Russia has become the first country in the world where a new type of automatic fire extinguishing systems, or robotic fire suppression systems, was introduced by the law. RFSS requirements are established by the Federal law No. 123-FZ, GOST R and Codes of Practice. Russia’s priority right for the invention of RFSS is protected by a number of patents.Conclusions. In our country, long-term research and development have been carried out to design new fire extin- guishing technologies named robotic fire suppression systems. Regulatory and technical frameworks have also been established, and a firefighting robot plant has been built. Now new fire extinguishing technologies, involving firefighting robots, are widely spread; they protect thousands of significant facilities of the country.

Los temas que se abordarán en la formación requerida serán: Gestión Social y Gestión Comunitaria, Procedimientos a seguir para la conformación de una red de Empresas de Producción Social. Se asume el método del materialismo-dialéctico, (Barreras, F. 2008), (Cerezal y Fiallo, 2004). Se realizó la observación participante y una entrevista semi-estructurada. La población está constituida los 17 miembros de los del Consejo Comunal, se empleó el muestreo teórico, (Salamanca y otros, 2007). Category: Trauma; Hindfoot Introduction/Purpose: We prospectively assessed the impact of minimally invasive internal fixation of displaced intra-articular calcaneal fractures (DIACF) results in better health outcomes up to 2 years post- injury than non-operative management. Methods: All isolated calcaneal fractures were reviewed and the decision to treat was made by a multidisciplinary meeting. Operative-treatment protocol involved sinus-tarsi approach or per-cutaneous reduction & internal fixation under the care of a fellowship-trained-foot and ankle surgeon. Non-operative protocol involved symptomatic management with no attempt at closed reduction. All fractures were classified, and the MOXFQ/EQ5D-L scores were used to assess general and disease-specific outcomes respectively. Secondary outcome measures included return to work and return to leisure activity levels. Statistical analysis was performed to assess normality of data (Shapiro Wilk test/Q-Q plots). Propensity matching was conducted using nearest neighbour matching, followed by regression analysis and modelling to compare baseline covariates between the two different groups. Results: Between 2015-2019 total of 93 patients met the inclusion criteria. After propensity score matching 44 patients were found in the surgical/ treatment group. The mean age was 40 (vs. 43.3) in the surgical group, no significant difference in age, sex, peripheral vascular disease, smoking status, laterality, and baseline Bohlers and Guisane angle in the propensity score matched sample. The surgically treated group had a higher Sanders class(p<0.05), higher MOXFQ at 12 months (p<0.05) and EQ-5DL scores at 24 months(p<0.05) and were more likely to return to work at 12m (95% vs 77%)(p<0.05), and physical activity levels at 12m (57% vs 36%)(p=0.054). The wound complication rate was 14.8%, no patients required further surgery; whilst non-operative- treatment mandated surgery for arthritis (6), and or/an injection (7 vs.3). Conclusion: Both non-operative and operative treatments are valid in DIACF. MIS-surgery offers better functional outcomes for severer injury patterns, and a less-likely need for arthritis intervention at 2yrs.

Well-balanced patent regulation is an important factor in the effective development of the biotechnology market. The possibility of establishing exclusive rights over the results of genetic research, including isolated genes and gene-based diagnostics, has been the subject of heated debate all over the world. Nevertheless, a unified approach to regulating these matters has not yet emerged. For example, in the US and India, patenting of isolated natural genes and diagnostic methods based on them is not permitted at all. Only synthetic genes and modified sequences are patentable. The EU and the UK generally allow such objects to be patented, but impose additional requirements. In China, diagnostic methods are generally not considered to be patentable, yet particular substances and instruments used for diagnosis may be recognised as protectable subjects. A cautious approach to the admissibility of patent protection for the results of genetic research is reflected, for example, in the requirement to provide additional data in order to prove that the solutions do meet the criteria of industrial applicability and non-obviousness. In countries which allow the patenting of a gene sequence (or a partial gene sequence with the structure identical to that of a naturally occurring one) as a substance, the law nevertheless mandates that the industrial application of such a sequence must be clearly indicated; it is also required that the patent application contains the information on the field for which the industrial application is specifically described. An unambiguous approach to biotechnology patenting has not yet emerged in the Russian legal sphere. Russian Civil Code does not directly prohibit the patenting of genes, gene-based therapies and diagnostics. Basic principles and approaches to the patenting of genetic engi-neering results and the protection of exclusive rights to them, including: the criteria for distin-guishing patentable and unpatentable results of genetic research, the limits of exclusive rights and the conditions for a compulsory license issuance have not yet been fully developed and introduced into the Russian legal framework and regulatory enforcement practice. The regulation is carried out at the level of by-laws: in accordance with the Rospatent Guidelines for Examination of Applications for Inventions, the approach used in the assess-ment of inventions relating to the field of biotechnology is the same as the one employed for inventions relating to other fields of science and technology. Given the potential importance of the biotechnology for the further progress in science and technology, these matters should be regulated at the Civil Code level. We propose that a broad public debate be held concerning 1) the principles for the regulation of these matters 2) the possibility of patenting, and 3) the extent of the exclusive rights of rightholders.

The article is devoted to the study of the institute of summary proceedings in civil proceedings, namely the legal nature of summary proceedings, the peculiarities of its procedural form and coverage of certain issues related to the implementation of the principles of civil proceedings during the consideration and resolution of cases in summary proceedings.The article considers doctrinal provisions on optimization of civil process by differentiation of procedural form of civil proceedings and introduction of simplified court procedures, international tendencies of its development in the corresponding direction, theoretical and applied scientific approaches on isolation of specific features of procedural form of simplified proceedings in relation to general comparative analysis of the legal regulation of the procedure for consideration of “small disputes” in the legislation of the Council of Europe, as well as prospects for further reform of domestic civil procedural legislation in the field of implementation of simplified court procedures.In the context of the issue of differentiation of the procedural form of civil proceedings, the criteria for distin-guishing between general and simplified procedure for consideration of civil cases in legal proceedings are defined, namely: the amount of claims (the price of the claim); subject of the claim (case category); the complexity of the case and the priority of its speedy resolution for the parties. The civil procedural legislation and scientific achieve-ments on determination of the category of cases which are subject to consideration by way of the simplified claim procedure are analyzed and the conclusion on possibility of assignment to them of requirements on which the court order can be issued is made.In view of the above, it was found that one of the features that characterizes the simplified claim proceedings in contrast to the general claim proceedings is the ability to consider claims that are indisputable in the existence of the following conditions: 1) claims that a person may order (the list of such requirements is exhaustive); 2) realization by the person of the right of an alternative choice of a type of civil proceedings in the order of which the corre-sponding requirements will be considered – the address of the person to court in the simplified claim proceedings.In terms of the court’s decision on the case in a simplified procedure, taking into account the specifics of its pro-cedural form, considered the scientific views and case law (conclusions) of the European Court of Human Rights on the implementation of sectoral principles of civil proceedings, including proportionality, discretion, adversarial and the generally accepted principle of civil procedure – the right to a court in the event of a court hearing without the participation of the parties.Based on the results of the study, conclusions were drawn.

Abstract:
Abstract Kupffer cells (KCs) account for 15% of the total liver cells. However, their roles in hepatocellular carcinoma (HCC) is poorly described. Clec4fCre-tdTomato knock-in mice expressing Cre recombinase and tdTomato-NLS under the KC-specific Clec4f promoter were hydrodynamically injected with oncogenic AKT and Nras (AKT/Ras) to induce HCC. A LoxP-stop-LoxP (LSL) system was used to express miR-206 in KCs of mice. AKT activation was observed in hepatocytes and KCs of HCC patients. Hydrodynamic injection (HDI) of AKT/Ras led to activation of AKT signaling in KCs and hepatocytes. AKT/Ras mice developed lethal HCC within 6-8 weeks post injection. Activation of AKT signaling led to M2 polarization of KCs, increased hepatic Tregs and exhaustion of CTLs. MicroRNA (miR) profiling revealed that AKT/Ras impaired miR-206 biogenesis in KCs by driving translation of Yin Yang 1 (YY1), a transcription repressor of miR-206. HDI of miR-206 into AKT/Ras mice fully prevented HCC, while all control mice died of HCC 6-8 weeks post injection. However, HDI of miR-206 only transfected ~ 20% of hepatocytes, indicating that it is unreasonable to conclude that the full prevention of HCC in AKT/Ras/miR-206 mice is caused by inhibited proliferation of HCC cells. We, therefore, posited that miR-206-mediated recovery of immune surveillance, at least in part, accounted for the full prevention of HCC in AKT/Ras/miR-206 mice. We next used the LSL system to overexpress miR-206 in KCs of Clec4fCretdTomato mice injected with AKT/Ras. KC-specific expression of miR-206 fully prevented HCC in AKT/Ras mice, while 100% control mice died of HCC. Mechanistically, AKT signaling in KCs drove CCL22 (C-C motif chemokine ligand 22) production by phosphorylating β-catenin that activates transcription of Ccl22. In contrast, miR-206 disrupted β-catenin signaling by targeting LEF1 (lymphoid enhancer-binding factor 1), an “architectural” transcription factor of β-catenin. CCL22 recruits Tregs. Indeed, KC-specific expression of miR-206 led to elevated CCL22, M2 to M1 transition of KCs, reduced hepatic Tregs, and elevated CTLs. Depletion of CTLs in AKT/Ras mice fully offset the ability of miR-206 to prevent HCC. Conclusions: AKT-educated KCs initiate Treg recruitment via a regulatory loop consisting of YY1, β-catenin/LEF1, miR-206, and CCL22; once this loop is activated, AKT activates β-catenin axis, which drives CCL22 production, Treg recruitment and HCC growth; while KC-specific expression of miR-206 reversed this process by blocking β-catenin axis in KCs. miR-206 represents a novel immunotherapy against HCC.
