Abstract:
Intermodal practices are related to the use of several distinct modes of transport during the same displacement. This study analyses the articulation between institutional transport insured by motorbike taxis and the Lomé Transport Company (Sotral) in Grand Lomé. In particular, it explores two intermodal practices between both actors, mainly the drawdown and broadcasting of Sotral users on/and from bus stopping, the main intermodality points after the bus stations in Grand Lomé. The methodology relies on a mixed approach, combining quantitative and qualitative methods for data collection and analysis. The first descriptive approach has allowed us to observe the phenomenon, to reinforce knowledge through the analysis of documents, and to obtain actors’ opinions. The second quantitative approach is based on the interviews carried out with drivers and motorbike taxi and bus users, the counting of broadcasting and drawdown modes at the stopping points, and the mapping of the study area as well as the Sotral network. The results show that the current facilities of Sotral bus stopping points do not encourage/promote the interaction and/or cohabitation between different modes of transport, such as motorbike taxis. These infrastructures are out of phase with the intermodal practices of the population of Grand Lomé, who use artisanal and structured public transport in their daily movements. In fact, motorbike taxis do not have a dedicated facility for drop-off/pick-up of passengers at the Sotral bus stopping points. The drawdown and broadcasting by these means of artisanal transport at bus stops are generally made on sidewalks and sometimes on roadways, which is a source of road insecurity. This failure leads to friction between Sotral and motorbike taxis on the use of facilities dedicated to Sotral buses, creating an absence of cohabitation between two main actors of mobility in Grand Lomé. An approach focused on the flexibility and efficiency of intermodal points or the connection between motorbike taxis and Sotral allows good coexistence and constitutes a component of a sustainable mobility strategy.

Abstract:
Road insecurity is a major challenge for the international community. The aim of this article is to take stock of the safety of users of soft modes of transport in Grand Lomé. The methodology used in this research is based, on the one hand, on the literature review in order to examine the different theories that govern the analysis of road safety issues and particularly on the safety of vulnerable users of soft modes of transport, and on the other hand, on the analysis of the realities on the ground through surveys and interviews with stakeholders. The results reveal real difficulties in getting around users of active modes of transport on the roads of Grand Lomé. The main roads do not have sidewalks and pedestrian crossings. The sidewalks that exist on some of the new roads are, for the most part, narrow and illegally occupied by businesses, which prevent users of soft modes from benefiting from them. Bike lanes are almost non-existent on the developed lanes. Following this logic, the street belongs first and foremost to motorists. Pedestrians and cyclists are guests who must negotiate their passage on the roads of Grand Lomé.

Abstract:
Au Togo, la culture de la tomate est l’une des cultures prioritaires du Programme national d’investissement agricole, de sécurité alimentaire et nutritionnelle (PNIASAN, 2017–2026). Avec une croissance annuelle moyenne de 15 %, cette production repose sur des milliers de ménages ruraux. 80 % de la tomate produite dans la Région des Savanes est exportée vers le Grand Lomé. Le confinement de cette région du fait de la COVID-19 a coïncidé avec la période de récolte massive de la tomate. Cette recherche vise à déterminer les effets des mesures de confinement sur cette filière. Des enquêtes ont été menées auprès de 152 acteurs choisis de façon volontaire dans la Région des Savanes et dans le Grand Lomé. L’article montre d’une part, que la COVID-19, par le biais de la limitation des déplacements, a sédentarisé une main-d’œuvre supplémentaire au profit de la production maraîchère, ce qui a entraîné une augmentation des surfaces en tomate et donc de la production. D’autre part, les difficultés des producteurs dues à la mévente de leur production de tomate et au pourrissement d’une partie de la récolte, ont entraîné une baisse notable du chiffre d’affaire des maraîchers. Mais ces difficultés, observées durant la crise sanitaire, ont des causes qui existaient antérieurement comme la faiblesse de l’organisation des producteurs et des maillons aval de la filière ainsi que la faible confiance entre ces différents acteurs. La pénurie de ce légume qui s’est installée dans le Grand Lomé a ainsi révélé l’existence des contraintes structurelles exacerbées par la crise de la COVID-19, mais qui lui préexistaient. L’émergence de nouvelles politiques publiques, plus inclusives et touchant tous les acteurs des filières, apparaît donc indispensable au bon fonctionnement des systèmes alimentaires au Togo.

Dados de dois grupos genéticos UFV1 e UFV2 de matrizes de codorna de corte, com 629 animais no primeiro arquivo e 707 no segundo, foram avaliados pela técnica de componentes principais. Foram consideradas as seguintes características para análise: peso da ave (P1, P2, P3 e P4), peso médio do ovo (POM1, POM2, POM3 e POM4), peso médio da casca (PCM1, PCM2, PCM3 e PCM4), peso médio da gema (PGM1, PGM2, PGM3 e PGM4), gravidade específica média do ovo (DM1, DM2, DM3 e DM4), largura média do ovo (LOM1, LOM2, LOM3 e LOM4), comprimento médio do ovo (COM1, COM2, COM3 e COM4), número de ovos (N1, N2, N3 e N4), idade ao primeiro ovo (IDPO) e taxa de postura total (TXT). Após a coleta dos dados, foi realizada uma análise de componentes principais, buscando, com isso, selecionar as características mais explicativas. Logo as características de maior importância na variância total tiveram seus parâmetros genéticos e correlações genéticas estimados. Para o grupo genético UFV1, foram: P1, PCM1, PCM3, PGM1, PGM2, PGM4, DM4, LOM2, LOM3, COM1 e IDPO, já para UFV2: P1, P2, PCM1, PCM4, DM3, LOM2 e LOM4. As altas estimativas de herdabilidade encontradas indicam uma grande variabilidade para as características de qualidade dos ovos. As correlações genéticas mostram a possibilidade de ganhos correlacionados entre peso corporal e as características de qualidade dos ovos. Pequena resposta à seleção para produção de ovos pode ser esperada para estes grupos genéticos.

Abstract In Africa, the west suffers the most from Illegal, Unreported, and unregulated fishing which necessitated national, regional, and continental efforts to put in place legal mechanisms to halt the situation. This resulted in the fragmentation of rules dealing with IUU, with AU’s 2050 African Maritime Strategy and the Lomé Charter being the grand Continental ones. This article will provide a brief assessment of the legal mechanisms of the African Union vis-à-vis the rules of the Economic Community of West African States (ECOWAS), and the fragmentation or harmonization of those rules. The paper argues that fragmentation of rules at regional and sub-regional levels is not necessarily counterproductive as long as all parties coordinate by focusing on filling existing gaps rather than allowing overlaps.

Introduction: déterminer le profil épidémiologique et sémiologique de l’ostéonécrose aseptique de la tête fémorale (ONATF) ainsi que les principaux facteurs favorisants en milieu rhumatologique au Togo.Matériels et méthodes: il s’est agi d’une étude rétrospective a visée descriptive menée d’octobre 1989 à juillet 2016 dans le service de rhumatologie du CHU- Sylvanus Olympio de Lomé. Ont été inclus dans notre étude tous les patients ayant moins de 16 ans et dont le motif de consultation était rhumatologique.Résultats: deux cent quarante-et-un patients avaient une ONATF. Ils se répartissaient en 56,02% de femmes et 43,98% d’hommes. L’âge moyen des patients était de 39 ans ± 14 ans avec des extrêmes allant de 9 à 80 ans. La durée d’évolution moyenne de la maladie était de 3,36 ans ± 4,6 ans avec des extrêmes de 11 jours et 27 ans. La douleur de la hanche était le principal motif de consultation. Elle siégeait à l’aine (165 cas ; 68,46%), au grand trochanter (108 cas ; 44,81%), à la fesse (47 cas ; 19,50%). La douleur était projetée à la cuisse chez 86 patients (35,68%). La marche était douloureuse chez 207 patients (85,9%) et associée à une boiterie chez 199 patients (82,57%). Selon la classification d’Arlet et Ficat, l’image radiographique des ONATF au diagnostic était au stade II (57 cas ; 23,65%), austade III (99 cas ; 41,08%) et au stade IV (85 cas ; 35,27%). L’atteinte était unilatérale (186 cas ; 77,17%) et bilatérale (55 cas ; 22,83%). Les principaux facteurs favorisants de l’ONATF retrouvés étaient : une hémoglobinopathie (79 cas ; 32,8%), l’éthylisme chronique (68 cas ; 28,2%) et l’infection au VIH (20 cas ; 8,3%).Conclusion: outre les formes évoluées dues au retard diagnostique, l’ONATF ne revêt pas de particularités sémiologiques. La drépanocytose, la consommation d’alcool et le VIH restent les principaux facteurs favorisants.

Les sujets atteints de pré-diabète présentent un risque élevé de syndrome métabolique (SM) et de diabète. La présente étude avait pour but d’évaluer la prévalence du pré-diabète, du SM et du diabète dans une population fréquentant un marché urbain, puis de déterminer la prévalence des dyslipidémies chez les pré-diabétiques et diabétiques. Il s’est agi d’une étude transversale à visée descriptive portant sur 350 sujets âgés de 18 à 90 ans, recrutés au grand marché de Hédranawoè en 2015. La prévalence du pré-diabète a été évaluée suivant les critères de l’Organisation Mondiale de la Santé (OMS) et de l’American Diabetes Association (ADA). La prévalence du SM a été obtenue avec les critères de la Fédération Internationale du Diabète (FID). La prévalence du pré-diabète suivant le critère d’ADA était de 13,2% avec une glycémie moyenne à 5,9±0,3 mmol/l. Celle du SM était de 1,3% chez les femmes et de 0,8% chez hommes, avec celle du diabète qui était à 7,1%. Celle des dyslipidémies variait de 32,0 à 60,0% chez les diabétiques dont 40% n’était pas préalablement dépistés. La prévalence du diabète était plus importante que celle déterminée dans la population générale du Togo. La prévalence du syndrome métabolique mériterait d’être réévaluer en utilisant des critères de tour de taille établis pour des africains au Sud du Sahara. Les populations fréquentant les marchés urbains devraient être régulièrement sensibilisées sur les risques du pré-diabète, du diabète et des dyslipidémies. English Title: Prevalence of pre-diabetes, metabolic syndrome, diabetes and dyslipidemia in 350 subjects recruited at the Hédzranawoé market in Lomé (Togo) People with pre-diabetes are at high risk for metabolic syndrome (MS) and diabetes. The purpose of the present study was to assess the prevalence of pre-diabetes, MS and diabetes in a population frequenting an urban market, and then to determine the prevalence of dyslipidemia in pre-diabetics and diabetics. This was a descriptive cross-sectional study on 350 subjects aged 18 to 90, recruited from the large market of Hédranawoè in 2015. The prevalence of pre-diabetes was assessed according to the criteria of the World Organization of Health (WHO) and the American Diabetes Association (ADA). The prevalence of MS was obtained using the criteria of the International Diabetes Federation (IDF). The prevalence of pre-diabetes according to the ADA criterion was 13.2% with an average blood sugar level of 5.9 ± 0.3 mmol / L. That for MS was 1.3% in women and 0.8% in men, with that of diabetes at 7.1%. That of dyslipidemia ranged from 32.0 to 60.0% in diabetics. Forty percent (40%) of these diabetics were not previously screened. The prevalence of diabetes was higher than that determined in the general population of Togo. The prevalence of metabolic syndrome should be reassessed using waist size criteria established for Africans from south of the Sahara. When 47-year-old Simba Abalo, an unemployed retired soldier in Lomé, Togo, found out that he had AIDS in September 2007, he was unable to receive government-supplied antiretroviral drugs: “CAMEG [the state’s central medicines purchasing organization],” he said, “told me they were not taking any new cases for six months because they had run out of drugs.Stocks of antiretrovirals had become depleted after the Global Fund to Fight AIDS, Tuberculosis and Malaria suspended part of its grant to Togo in 2006 and failed to approve a new grant request in 2007, reportedly because of oversight irregularities. “Even if they had the drugs,” said Abalo, “I could not have paid more than US$4 a month since I had just lost my job as a security guard earning US$70 a month.”

Objetivo: Estimar la energía disponible para generar electricidad a partir de biogás de vertedero utilizando el modelo LandGEM adaptado a las condiciones locales de Montería, Colombia. Metodología: Para ello se obtiene de base de datos y estudios la información del relleno sanitario de residuos Loma Grande ubicado en la ciudad de Montería, volumen de residuos, clima y composición de residuos para el periodo 2016-2028, para aplicar el modelo LandGEM a la producción de energía de biogás. Resultados: se observó que solo para el año 2022 la cantidad de metano calculada es de 9.984.000 m3/año y con esto la energía estimada total es de 268.4968 MWh, que podría suministrar los requerimientos del relleno sanitario Loma grande. Conclusión: El estudio predice que estos modelos de evaluación se pueden utilizar para planificar la producción de energía a partir de gas de vertedero, y así, aprovechar la propiedad de terrenos improductivos que normalmente se utilizarían como vertedero y que se crearon previamente para materiales contaminantes no destructivos.

In particolare la crescita ha riguardato la popolazione delle grandi cittŕ: se solamente due cittŕ, Ibadan e Johannesburg, intorno alla metŕ del Novecento avevano piů di un milione di abitanti, circa un ventennio piů tardi se ne contavano, sotto questa categoria ‘milionaria', venticinque. Piů di un terzo della popolazione totale (il 38%, circa 350 milioni di abitanti) giŕ viveva in una situazione urbana nel 2005, e secondo alcune proiezioni i ‘cittadini' africani alla metŕ di questo secolo saranno molto piů della metŕ della popolazione totale, toccando il 65%. L'analisi di alcuni casi di capitali africane (Mogadiscio, Lomé, Dakar, Nairobi) nel loro sviluppo recente mostra i problemi specifici di queste grandi cittŕ africane e analizza la loro vicenda nel legame con le vicende politiche del loro paese.

Martin Heinrich Klaproth (1743-1817). Un Apothicaire allemand, chimiste et grand découvreur – Martin Heinrich Klaproth est né le 1er décembre 1743 à Wernigerode dans le Harz (centre nord de la Prusse) dans un milieu modeste. Il interrompra ses études secondaires pour s’engager en apothicairerie où il gravira successivement tous les échelons de la profession : apprenti (à Quedlindburg), garçon (à Quedlindburg, Hanovre, Berlin et Dantzig), premier commis ou «Provisor » , puis gérant à Berlin, et enfin apothicaire propriétaire (pharmacie de l’Ours à Berlin) en 1780. Pendant cette période, plusieurs apothicaires, comme Valentin Rose et Andreas Sigismund Maargraf, renforcèrent son esprit scientifique et orientèrent fortement sa carrière Après avoir été élu membre de l’Académie des sciences en 1788, il deviendra le directeur appointé et logé du laboratoire de chimie associé, en 1800, et à la création de l’Université de Berlin en 1809, à l’initiative de Wilhelm Von Humboldt, conseiller du Roi, il sera nommé professeur de chimie et y exercera jusqu’en 1814. Les travaux scientifiques de Klaproth ont porté sur la pharmacie et surtout en chimie générale. L’étude de minerais l’a amené à mettre en évidence, voire dénommer, plusieurs nouveaux éléments : quatre personnellement (l’uranium, le zirconium, le tellure et le cérium) et trois simultanément et indépendamment avec d’autres (le titane, le strontium et le chrome). Outre ces travaux qui le firent qualifier de minéralogiste, Klaproth introduisit des méthodes, appliquées des théories de Lavoisier, basées sur le contrôle et l’exactitude des mesures qui serviront à bâtir des lois de la chimie.

Contexte : la main de moulin se définit comme tout traumatisme de la main causée par une machine servant à moudre des céréales, des légumes ou toute autre denrée alimentaire. Elle est rencontrée dans notre milieu hospitalier motivant notre étude. Objectif : étudier les aspects épidémiologiques, lésionnels et donner un aperçu sur la prise en charge de la main de moulin au service de chirurgie pédiatrique du CHU Sylvanus Olympio de Lomé. Méthodes : il s’agissait d’une étude rétro prospective couvrant une période de 5 ans allant du 01 juin 2017 au 31 Mai 2022. Ont été inclus tous les enfants de 0 à 15 ans pris en charge dans le service de chirurgie pédiatrique du CHU SO Lomé pour un traumatisme ouvert de la main causé par un moulin. Résultats : nous avons enregistré 30 cas des mains de moulin soit 5,23% de tous les traumatismes ouverts de la main. La prédominance était masculine (18 garçons contre 12 filles). La tranche d’âge la plus touchée était celle de 10 à 15 ans avec un âge moyen de 9,07 +/- 4. La circonstance la plus retouvée était l’accident de travail. La majorité des traumatismes ont eu lieu au moulin public. La face de la main la plus touchée était celle plantaire. Le troisième rayon était le plus touché. Sur la face palmaire, la zone I de Verdan était la plus touchée dans 16 cas ainsi que sur la face dorsale dans 4 cas. Les phalanges étaient les plus touchées. La fracture était la lésion la plus fréquente. Les gestes opératoires etaient multiples du fait de la multiplicité des lésions. Conclusion : les mains de moulin chez l’enfant sont caractérisées par des lésions multiples et variées rendant la prise en charge complexe, posant ainsi un problème de santé publique. Context: mill hand is defined as any trauma to the hand caused by a machine used to grind cereals, vegetables or any other foodstuff. It is encountered in our hospital environment motivating our study. Objective : étudier les aspects épidémiologiques, lésionnels et donner un aperçu sur la prise en charge de la main de moulin au service de chirurgie pédiatrique du CHU Sylvanus Olympio de Lomé. Methods: this was a retro prospective study covering a 5-year period from June 01, 2017 to May 31, 2022. Were included all children aged 0 to 15 years cared for in the pediatric surgery department of CHU SO Lomé for open hand trauma caused by a mill. Results: We recorded 30 cases of mill hands, i.e. 5.23% of all open hand traumas. The predominance was male (18 boys versus 12 girls). The age group most affected was 10 to 15 years, with an average age of 9.07 +/- 4. The most common circumstance was a work accident. The majority of injuries occurred at the public mill. The side of the hand most affected was the plantar side. The third ray was the most affected. On the palmar side, Verdan zone I was most affected in 16 cases, and on the dorsal side in 4 cases. The phalanges were the most affected. Fracture was the most frequent lesion. Multiple surgical procedures were required, due to the multiplicity of lesions. Conclusion: Mill hands in children are characterized by multiple and varied lesions, making management complex and posing a public health problem.

Antecedentes: La diabetes mellitus es considerada una patología metabólica crónica de carácter mundial, mismo que comprende un grupo de desórdenes metabólicos el cual se considera en un nivel elevado de glucosa en la sangre (hiperglicemia). Objetivo: analizar el apoyo social percibido en pacientes con diabetes mellitus. Método: se efectuó un estudio con enfoque cuantitativo, descriptivo, de corte transversal en pacientes con Diabetes Mellitus Tipo 2 que asisten al Centro de Salud Loma Grande Tipo A. La muestra estuvo constituida por 105 pacientes con DM2 voluntarios con más de 60 años, se utilizó una encuesta de 12 ítems, que son preguntas cerradas previamente validadas para el tema de apoyo social percibido en individuos con DM en la provincia de Cotopaxi. Resultados: Con respecto al apoyo social percibido, se evidencia que un 57 % utilizan la ayuda de los familiares y que siempre necesitan el apoyo emocional o físico de los mismos, además el 20% de los pacientes necesitan el apoyo social de otra figura que no sea familiar. Conclusiones: El apoyo social desde la perspectiva en pacientes con DM2 en el Centro de Salud Loma Grande, muestran un elevado nivel y en su mayoría son provistos por la familia. El ámbito familiar resultó fundamental como medio de apoyo en los pacientes, y en menor medida, pero no menos importante, el apoyo que recibió del personal de salud y amigos. Se evidencia en este estudio como la presencia y papel de la familia se constituye como forma de apoyo social.

We have combined electrical resistivity tomography (ERT), geologic information from boreholes and outcrops, and hydrogeologic data to investigate field-scale fault-zone cementation of the Loma Blanca Fault in the Rio Grande Rift. We have collected electrical resistivity data from 16 transects and geologic samples from 29 boreholes (completed as groundwater wells to 30 m depth) across and around the fault. The 2D ERT profiles, whose interpretations are constrained by geologic data, indicate (1) a high resistivity zone in cemented portions of the fault below the water table and (2) in the unsaturated zone, a low-resistivity feature along the cemented portions of the fault. The high-resistivity zone below the water table is consistent with a 10% reduction in porosity due to the fault zone cementation. With the same porosity in the unsaturated zone, the low-resistivity feature in the cemented fault zone is consistent with saturation >0.7, in contrast to saturation 0.2–0.7 for sediment outside of the cemented fault zone. In addition, subsurface samples and ERT profiles delineate a buttress unconformity (i.e., steeply dipping erosional contact) corresponding to a paleovalley margin. This unconformity truncates the cemented fault zone and separates Pliocene axial-fluvial sand (deposited by an ancestral Rio Grande) from late Quaternary sand and gravel (deposited by the Rio Salado, a Rio Grande tributary). The cemented fault zone in the southern portion of the study area is a hydrogeologic barrier; north of the buttress unconformity, where the cemented fault zone has been removed by erosion, the fault is not a hydrogeologic barrier. The integration of geologic, geophysical, and hydrogeologic observations is key to developing our understanding of this complex system, and it allows us to demonstrate the utility of ERT in detecting subsurface fault-zone cementation.

Este artículo presenta los resultados parciales obtenidos en el trabajo sobre la revisión de ¿Cuál es el impacto económico, social y ambiental del desarrollo urbano de la isla La Loma?; unidad ecológica localizada al norte de estuario deltaico conformado por el sistema de canales hídricos del rio Magdalena. This paper presents the partial results of the work on the revision of “What is the economic, social and environmental impact of urban development on the island La Loma?”, which is an ecological unit located north of deltaic estuary formed by the system of water channels of the Magdalena River. Also shown in its progress a number of situations that should be checked deeply, it could be understood the ecological benefits that solves the island back to its natural condition or reservation. Our understanding of the frequency of large earthquakes at timescales longer than instrumental and historical records is based mostly on paleoseismic studies of fast-moving plate-boundary faults. Similar study of intraplate faults has been limited until now, because intraplate earthquake recurrence intervals are generally long (10s to 100s of thousands of years) relative to conventional paleoseismic records determined by trenching. Long-term variations in the earthquake recurrence intervals of intraplate faults therefore are poorly understood. Longer paleoseismic records for intraplate faults are required both to better quantify their earthquake recurrence intervals and to test competing models of earthquake frequency (e.g., time-dependent, time-independent, and clustered). We present the results of U-Th dating of calcite veins in the Loma Blanca normal fault zone, Rio Grande rift, New Mexico, United States, that constrain earthquake recurrence intervals over much of the past ∼550 ka—the longest direct record of seismic frequency documented for any fault to date. The 13 distinct seismic events delineated by this effort demonstrate that for >400 ka, the Loma Blanca fault produced periodic large earthquakes, consistent with a time-dependent model of earthquake recurrence. However, this time-dependent series was interrupted by a cluster of earthquakes at ∼430 ka. The carbon isotope composition of calcite formed during this seismic cluster records rapid degassing of CO2, suggesting an interval of anomalous fluid source. In concert with U-Th dates recording decreased recurrence intervals, we infer seismicity during this interval records fault-valve behavior. These data provide insight into the long-term seismic behavior of the Loma Blanca fault and, by inference, other intraplate faults.

Historians have all praised the periodical efforts to restore the lake's water supply and irrigation functions. However, in view of the high operating costs of the lake and its limited benefits, the various attempts to reclaim land and water for agriculture may have been more rational in terms of economic profit. Visitors' descriptions provide many details on the long-term environmental changes in the lake area. In the 1960s, it was drained and turned into a state farm. Le Lac d'Entraînement peu profond de Jiangnan a retenu l'attention dans les textes historiques parce que les gouvernements dynastiques successifs se sont investis pour assurer alimentation saisonnier du Grand Canal par ses eaux. Tous les historiens ont loué les efforts périodiques visant à lui conserver sa fonction d'alimentation et d'irrigation. Abstract Despite significant advancements in our understanding of acute myeloid leukemia (AML), relapsed and refractory disease remains a major cause of treatment failure. Approximately 50% of patients with AML will develop relapsed disease following induction chemotherapy, which results in a dismal 5-year overall survival rate of 29%. The development of FMS-like tyrosine kinase 3 (FLT3) inhibitors have led to improved outcomes among patients with FLT3-mutated AML. However many patients eventually relapse and succumb to chemo-resistant disease, highlighting the need to characterize the molecular pathways which confer early TKI resistance. To explore the possible mechanisms responsible for the survival and proliferation of tyrosine kinase inhibitor (TKI) resistant blasts, we compared in vitro and ex vivo cytokine expression of gilteritinib (GILT)-treated and untreated blasts. Here we report that the pro-inflammatory cytokine macrophage migration inhibitory factor (MIF) is significantly expressed by GILT-treated blasts when compared to untreated controls. We further demonstrate that MIF expression promotes blast proliferation through the upregulation of its receptor CXCR2. Most remarkably we found that combination of CXCR2-inhibitor plus GILT works synergistically to reduce the percentage of viable blasts. Together these findings support that targeting the TKI-activated MIF/CXCR pathway could be a novel therapeutic strategy for both newly diagnosed and relapsed/refractory AML. The Ilha Grande National Park, Paraná, Brazil, is located in the Upper Paraná River and has characteristics typical of a floodplains. This protected area includes lagoons connected and disconnected to the Paraná River, although the latter also connect during periods of high water level, thus composing a heterogeneous group of lacustrine environments. The enormous potential the flora and fauna diversities are still little known to the region, as can be seen through benthic invertebrates, inclunding bivalves mollusks. The granulometric composition of these floodplain lagoons was formed mainly by mud and very fine sand. Furthermore, organic matter composition was predominantly of fine particulate. The other abiotic factors differed from lagoons located within the island of the park to those located in the left margin of Paraná River. The results demonstrated the importance of abiotic factors such as the physical composition of granulometric texture, organic matter and macrophyte banks, to the establishment of bivalves in these floodplain lagoons. We recorded bivalves of Pisidium (native), Diplodon (native), and Corbicula (invasive). The highest values of Diplodon sp. density were observed at São João/C lake, for Pisidium sterkianum (Pilsbry, 1897) at São João/M lake, and to Jatobá/C lagoon with high density of invasive species Corbicula fluminea (Müller, 1774). This study to obtain conduct the first records of freshwater bivalves in floodplains lagoon in the Ilha Grande National Park, and provides contributions to better understanding the ecology of these mollusks. The recording of native species in the region of Upper Paraná River floodplain after a lomg period without new records, demonstrated the importance of protecting the lagoons of the Ilha Grande National Park as they can be a possible refuge to some species of native freshwater bivalves.

El objetivo de este trabajo fue evaluar el desempeño productivo y realizar un análisis económico del uso del antibiótico virginiamicina como aditivo, en la suplementación a base de maíz para novillas en pastoreo. Fueron utilizados 24 animales de la raza Brangus con edades de 260.29 ± 42.93 días y con peso medio inicial de 233.9 ± 19.32 kg, suplementados individualmente, en pastoreo de Brachiaria hibrida CIAT 36087 (mulato II) con disponibilidad promedio de 7885.0 kg de materia seca por hectárea. Los tratamientos evaluados consistieron en la suplementación con 0.8% de maíz con respecto al peso vivo (PV) (T1 = Testigo), 0.5% del PV con maíz más 0.00225% del PV en virginiamicina (T2 = virginiamicina 0.5); y 0.8% del PV de maíz más 0.00225% del PV en virginiamicina (T3 = virginiamicina 0.8). No se observaron diferencias estadísticas (P>0.05) durante el período experimental para peso corporal (PC), puntaje de condición corporal (PCC), ganancia de peso diaria promedio (GDP), área del ojo del lomo (AOL), ultrasonografía de la canal (espesor de grasa subcutánea (EGS) y del anca (EGA), y puntaje del tracto reproductivo (PRT)). La sustitución de 0.3% del PV de maíz por virginiamicina en el tratamiento dos generó el mismo desempeño productivo y PTR que los otros tratamientos, posibilitando la disminución del costo del suplemento por novilla en pastoreo de mulato II. Aunque no se presentaron diferencias estadísticas, numéricamente, el tratamiento con mayor ganancia de peso fue el 3 (virginiamicina 0.8) con 390 g/día seguido de T2 (virginiamicina 0.5) con 290 g/día y por último T1 (suplementación con solo maíz) con 270 g/día.

El nuevo grupo Drosophila asiri pertenece al subgénero Drosophila. Los ejemplares fueron capturados en los bosques andinos del Ecuador, desde los 3200 hasta los 4000 m de altitud. A este grupo pertenecen Drosophila (D.) asiri Vela & Rafael, 2005 previamente ubicada dentro del grupo D. onychophora, y dos nuevas especies; Drosophila (D.) yuragyacum sp. nov. y Drosophila (D.) yanaurcus sp. nov. Las capturas se realizaron en el bosque protector Pasochoa, en la quebrada de Cruz Loma y en el páramo de Papallacta, utilizando cebo de banano y levadura. Los miembros del nuevo grupo por el momento son endémicos de los Andes ecuatorianos. Las tres especies del grupo D. asiri son moscas de tamaño grande (aproximadamente 6 mm). Los machos presentan el edeago quitinizado con dos proyecciones laterales en la cabeza a manera de cuernos muy quitinizados. Estas características diferencian a las especies del grupo D. asiri de otros grupos dentro de Drosophila.

Abstract:
Se evaluó el efecto de la inclusión de diferentes niveles (0, 10, 20 y 30 %) de grano seco de destilería con solubles (DDGS), sobre la calidad de la canal y de la carne de conejos en crecimiento, utilizando 56 conejos (Negro Azteca x Chinchilla) de 40 días de edad. Terminado el período de ceba, se sacrificaron 20 conejos (5 por tratamiento) y se determinó la proporción de la canal conformada por las extremidades anteriores, extremidades posteriores, costillas y lomo. Se determinó el peso total de carne, hueso y grasa del lomo, y se calculó la relación carne:hueso para toda la canal. Se realizó la evaluación sensorial de la carne con 46 panelistas que valoraron el olor, color, sabor y textura. En el músculo Longissimus dorsi se determinó el color, evaluando la luminosidad (L*), el índice de rojo (a*) y el índice de amarillos (b*). Además, se evaluó textura mediante medición de la fuerza de ruptura. Los resultados de calidad de la canal y de la carne se analizaron por medio de ANOVA. Los resultados de la evaluación sensorial seevaluaron con estadística no paramétrica. No hubo diferencias (P>0.05) en las mediciones de la canal, características organolépticas ni textura de la carne. El parámetro b* del color de la carne fue mayor (P<0.05) en las dietas con 10, 20 y 30 % de DDGS (11.77, 12.17 y 12.22 respectivamente) comparado con la dieta control (9.68). La evaluación sensorial evidenció que la carne de conejo con o sin DDGS tuvo un olor y sabor agradable, color pálido y textura suave. Se concluyó que la inclusión de DDGS hasta 30 % en la dieta, no afectó las características de la canal ni de la carne de los conejos.

Abstract Inflammatory bowel disease (IBD) is a chronic inflammatory disorder in the intestines. Current therapies aim at suppressing inflammation to prevent further damage. However, healing already damaged intestinal epithelia is still an unmet medical need. In this regard, epithelial healing is achieved via intestinal stem cells (ISCs) that are located in the intestinal crypts. Considering the literature reports that the active vitamin D (1,25[OH]2D) is essential for the maintenance of Lgr5+ ISCs, the active proliferating ISCs, we recently proposed to deliver locally high 1,25(OH)2D concentrations to the proximity of intestinal crypts to promote intestinal stem cell functions. This local delivery was devised to avoid potential side effects associated with systemically high 1,25(OH)2D concentrations. Using 5-Bromo-20-deoxyuridine (BrdU) to label the actively proliferating ISCs, our previous data indicated that the locally delivered 1,25(OH)2D enhanced the migration and differentiation of Lgr5+ ISCs. However, although sparse in the crypts, other proliferating cells may be labeled with BrdU. This current study used a unique mouse strain to precisely trace Lgr5+ ISCs and consolidated our previous finding that the locally delivered 1,25(OH)2D augmented ISC differentiation. Besides, we generated pure Lgr5+ ISC lines that could be induced to differentiate on demand. Our data showed that, in vitro, 1,25(OH)2D enhanced the differentiation of Lgr5+ ISCs. La soja es el principal cultivo de grano en Paraguay. Su producción avanza en el Chaco central y se requiere información sobre el comportamiento de nuevos cultivares que se adapten a la región. El objetivo fue analizar la productividad y adaptabilidad de cultivares de soja en secano en el Chaco central paraguayo. En Loma Plata (Cooperativa Chortitzer Ltda) se evaluaron a campo 25 líneas experimentales y 6 variedades comerciales (VC) en un diseño en bloques completos al azar con 3 repeticiones. La temporada (2019) fue calurosa en la etapa vegetativa y sufrió anegamiento en las etapas reproductivas. Se registró la fenología, el rendimiento y sus componentes y la composición de granos. Entre los cultivares, la duración de la etapa fenológica siembra-floración varió entre 30 y 50 días. El rendimiento se ubicó en el rango de 800 a 1600 kg/Ha, con diferencias significativas entre cultivares (p<0,0001). Las líneas 23, 9, 13, 6 y 25 fueron competitivas contra las mejores VC (VC6 y VC5). El rendimiento se asoció al número de granos (r=0,82). La composición de los granos fue estable, con 22,6-24,0 % de aceite y valores anormalmente bajos (< 28%) de proteína. Se concluye que hay líneas experimentales promisorias para el Chaco central, con rendimientos competitivos respecto a las variedades comerciales y que la línea 23 tiene elevado número de vainas y peso de grano, con adaptación al suelo con alta frecuencia de inundación. Su utilidad en sistemas de rotación en los arrozales chaqueños es un tema de interés para próximas investigaciones.

Durante la década de 1980 existieron tres puntos de vista acerca del Centro histórico de Quito (CHQ): como un lugar marcado por el monumentalismo; la ciudad y su centro como diversidad social; y como un lugar habitable, dinámico, patrimonial y social. Los dos primeros expresaban la visión del Municipio de Quito, mientras que el último correspondía a los pobladores barriales. Así, el objetivo de esta investigación busca reflexionar la tercera vertiente que proviene de la periferia del centro. En cuanto a la metodología se fundamentó en entrevistas a moradores barriales y urbanistas, artículos de prensa y fuentes secundarias, a partir de lo cual se analizó a los barrios San Marcos y La Loma Grande como casos de estudio. De las entrevistas se obtuvo como resultado que el componente periférico constituyó un soporte reflexivo con lo que se sacó a la luz el patrimonio barrial y otras dinámicas sociales que moldearon a los barrios y enriquecieron a su centro. Sobre las conclusiones, se visualiza a los barrios con vínculos sociales, comunales y portaron memoria social y patrimonial en los años investigados, aspectos importantes a considerar para una mejor política pública sobre turismo, patrimonio y cultura en Quito actual.

Introduction. Sarcoidosis is a multisystem, granulomatous disease of unknown etiology. Sarcoid granulomas appear as immunological response to a particular but still unknown agent of the human body. Objective. The main purpose of this study was to point out the important fact that the exact diagnosis of sarcoidosis must be estimated by clinical and pathological correlation, and team cooperation between the clinician and the pathologist. Methods. Of 751 patients referred with the suspected diagnosis of sarcoidosis, from 1995 to1999, 663 (431 female and 232 male) were analyzed and confirmed as having sarcoidosis stage I-III based on biopsy findings obtained by bronchoscopy, open lung biopsy, skin biopsy, liver biopsy or splenectomy. Results. Diagnosis of sarcoidosis was made in 663 patients, 431 females and 232 males (ratio 1.9:1). The average age of patients varied from 16 to 67 years, with those below age 50 years being predominant (78.4%). The highest number of patients was diagnosed in stage I of lung sarcoidosis (81.7%). Sarcoidosis was the most common cause of hilar and mediastinal lymphadenopathy (72.2%). Conclusion. Biopsy is a necessary diagnostic procedure for pathological diagnosis of sarcoid granu- loma before treatment even in patients where clinical, radiological, biochemical and immunological tests imply the diagnosis of sarcoidosis.

La incorporación de peces a la dieta de grupos cazadores-recolectores puede evaluarse en función de su densidad, la disponibilidad de recursos de mayor tamaño corporal, la tecnología disponible, entre otros. En este trabajo se aborda la variabilidad en el registro ictiofaunístico durante el Holoceno tardío en dos áreas: el valle inferior del río Chubut y su desembocadura y costa adyacente. De acuerdo a la productividad ambiental, la disponibilidad de recursos de alta jerarquía y las oportunidades de captura de peces, se genera un modelo de subsistencia que evalúa la diversidad e importancia relativa de los peces para las dos áreas. Las expectativas zooarqueológicas derivadas son discutidas a partir de los muestreos faunísticos de las localidades Barranca Norte y Los Cangrejales Sur (desembocadura y costa) y los sitios Loma Grande 1 y Cinco Esquinas 1 (valle inferior), ubicados a unos 30 y 18 km del mar respectivamente. Los resultados arrojan diferencias en la riqueza taxonómica de peces: en el valle predominan las percas, mientras que en la desembocadura y costa se explotó una amplia variedad. Se registró una importante contribución de peces en ambas áreas; sin embargo, en los campamentos residenciales de la desembocadura los restos de peces son menos frecuentes.

Abstract Inflammatory bowel disease (IBD) is a chronic inflammatory disorder in the gastrointestinal tract (the Gut). Available IBD therapies can cause severe side effects, and many patients either fail to respond to the treatment or lose therapeutic effects with time. Hence, IBD therapy is still an unmet medical need. We recently proposed to enhance the induction of regulatory T cells (Tregs) expressing gut-homing molecules (gut-homing Tregs) to control chronic gut inflammation in IBD patients without compromising systemic immunity. We have shown that dendritic cells (DCs), engineered to de novo synthesize high concentrations of the active vitamin D (1,25-dihydroxy vitamin D or 1,25[OH]2D) and retinoic acid, increase the induction of gut-homing Tregs phenotypically and functionally in healthy mice and the mice induced for experimental colitis. This current study investigated whether the engineered DCs worked in humans. In addition, we also asked how zinc impacts the function of our engineered DCs because zinc is a biologically essential mineral widely included in daily supplements and has also been shown to tolerize DCs. Our data showed that the engineered DCs, derived from healthy control subjects and IBD patients, stimulated autologous naïve CD4+ T cells to express both regulatory and gut-homing molecules (gut-homing Tregs). In addition, the stimulated CD4+ T cells suppressed the proliferation of autologous effector T cells. Moreover, the addition of zinc enhanced the ability of the engineered human DCs to augment the induction of autologous gut-homing Tregs. Hence, our findings indicate zinc-treated engineered DCs are a promising novel IBD therapy. RESUMO A diversidade de formatos utilizados para o desenvolvimento de objetos de aprendizagem contribui para que o processo de recuperação de informação relevante permaneça sendo um grande desafio para a Ciência da Informação e a Ciência da Computação. A adoção de metadados para a catalogação destes recursos vem sendo utilizada tanto em âmbito nacional quanto internacional. No entanto, não existe um consenso sobre qual é o melhor padrão de metadados. Tem como objetivo apresentar um mapeamento relacional entre principais padrões de metadados utilizados pelos repositórios de objetos de aprendizagem. A metodologia utilizada se fundamenta em análise exploratória e analítica de repositórios de objetos de aprendizagem para identificar os metadados de cada iniciativa e propor um modelo de correlação entre os padrões. Os resultados apontam que os padrões Dublin Core e LOM são referências para outros padrões de metadados. Assim, o mapeamento foi realizado a partir de uma correspondência entre esses padrões e para a validação foram analisados 140 objetos de aprendizagem disponíveis em repositórios na Web. Conclui-se que é possível estabelecer um modelo de correlação entre os metadados com um número significativo de elementos para colaborar com o processo de indexação automática e aprimorar a recuperação de informação.

El objetivo de esta investigación fue evaluar los caracteres morfológicos y de crecimiento, así como el rendimiento de forraje verde de maíces nativos de Loma Bonita, Oaxaca. Los genotipos fueron 5 variedades nativas y una variedad mejorada utilizada como testigo, los cuales fueron evaluados mediante un diseño experimental bloques al azar con cuatro repeticiones. Para evaluar el crecimiento de los genotipos se hicieron 12 muestreos semanales. Las variables evaluadas en campo fueron altura de planta (cm), área foliar (cm2) y clorofila en hojas (unidades SPAD) y en laboratorio se midieron 21 componentes morfológicas de planta, entre las que destacaron: peso de hojas (g), peso de tallo (g), peso de elote (g), hileras por elote y diámetro de elote (cm), peso de espigas (g) y rendimiento de forraje (t ha-1). La información obtenida se sometió a un análisis de varianza y prueba de comparación de medias (Tukey, P ≤ 0.05) para las variables que mostraron diferencias estadísticas significativas. Los resultados indicaron que los genotipos PINSB (52.0 t ha-1) y NLBO (50.2 t ha-1) superaron en rendimiento en forraje al testigo VS-536 (34.6 t ha-1), debido a que en general presentaron una mayor altura de planta, diámetro de tallo y área foliar por planta. No obstante, los maíces nativos evaluados no superaron al testigo en rendimiento de grano, hileras y granos por elote, diámetro de elote y contenido de clorofila en hojas.

Logé dans l’aire de transition entre les zones semi-aride au Nord et humide au Sud, le bassin versant de l’Oti (BVO) a subi durant les décennies 1970 - 1990 de très fortes pressions anthropiques et climatiques. La sécheresse, l’avancée du désert et les activités économiques reposant essentiellement sur l’agriculture et l’élevage ont contribué à la dégradation de ce bassin. En outre, de par sa position, le BVO accueille chaque année pendant la saison sèche du bétail transhumant venant du Burkina Faso et du Niger. Ce mouvement saisonnier des bovins transhumants a été source de maintien et de transmission de plusieurs maladies transfrontalières et des trypanosomoses animales. Des données de synthèse bibliographique et d’enquêtes de terrain effectuées en fin de saison sèche en avril et mai 2006, traitées dans un système d’information géographique (SIG), ont permis d’évaluer l’impact des facteurs anthropiques et climatiques sur l’épidémiologie des trypanosomoses animales dans le BVO. Les résultats de l’étude ont montré que le BVO était fortement dégradé ; les grands mammifères sauvages et les glossines ont quasiment disparu de la réserve de faune de la Fosse aux lions. Glossina tachinoides a été la seule espèce capturée au nord du 10e parallèle de latitude N. Trypanosoma vivax a été classé comme le principal parasite responsable des trypanosomoses bovines. Il a été conclu que l’homme par son action sur l’environnement et les changements climatiques ont provoqué un recul vers le sud de l’aire de répartition des glossines.

Abstract The advent of low‐cost continuously recording cable‐free autonomous seismographs, commonly referred to as nodes, enables dense spatiotemporal sampling of seismic wavefields. We create virtual source reflection profiles using P waves from five teleseismic events recorded by the Sevilleta node array experiment in the southern Albuquerque basin. The basin geology records a structurally complex history, including multiple Phanerozoic orogenies, Rio Grande rift extension, and ongoing uplift from a midcrustal magma body. The Sevilleta experiment densified the long term, regionally sparse seismograph network with 801 single channel vertical‐component 10 Hz geophone nodes deployed at ∼300 m spacing for 14 days in February 2015. Results show sediment‐basement reflections at &lt;5 km depth and numerous sub‐basin structures. Comparisons to legacy crustal‐scale reflection images from the Consortium for Continental Reflection Profiling show agreement with structural geometries in the rift basin and upper crust. Comparisons of the teleseismic virtual reflection profiles to synthetic tests using 2D finite‐difference elastic wave propagation show strong P‐to‐Rayleigh scattering from steep basin edges. These high‐amplitude conversions dominate the record sections near the western rift margin and originate at the Loma Pelada fault, which acts as the primary contact between rift‐bounding basement‐cored fault blocks and rift basin sediments. At near offsets, these signals may interfere with interpretation of upper crustal structure, but their relatively slow moveout compared to teleseismic P‐wave multiples provides clear temporal separation from sediment‐basement reflections across most of the array. The high‐signal‐to‐noise ratio of these converted Rayleigh‐wave signals suggests that they may be useful for constraining short‐period (∼1 Hz) dispersion with strong sensitivity in the uppermost ∼1 km of the rift basin sediments.

Selon les fêtes, les chanoines absorbaient de 6 384 à 8 840 calories et les moines de Saint-Denis peut-être 9 227.

Although opinions vary as to the degree of assertiveness of China’s leadership under the Xi Jinping rule in comparison to Hu Jintao, the fact that China under Xi has set out on a new, more persuasive discursive path regarding its historical role and position global futures has been confirmed both by analysts inside and outside of the PRC – particularly after the 2018 PRC Constitution Amendment, which introduced “Xi Jinping Thought on Socialism with Chinese characteristics for a New Era”, among other additions. This paper serves the purpose of explaining the meaning of the official discursive strategies behind the “New Era” (新时代) concept as presented by Xi Jinping during his Report at the 19th National Congress of the Communist Party of China by examining the grand narratives that surround it, ultimately contributing to the research of the logic behind Xi Jinping’s agenda vis-a-vis China’s desired position. A total of three overarching New Era narratives have been established: the mission narrative, the international influence narrative, and the party governance narrative. The paper argues that the main task of all three narratives is to provide arguments in support of Xi Jinping’s ideological innovation. Methodologically, the paper draws on the Political Discourse Analysis theory. Šī raksta mērķis ir izskaidrot “Jaunā laikmeta“ (新时代) koncepcijas pamatā esošās oficiālās diskursīvi konstitucionālās stratēģijas izpausmes Sji Dzjiņpina ziņojumā Ķīnas Komunistiskās partijas 19. Nacionālajam kongresam, šādi veicinot Sji Dzjiņpina darba kārtības izpēti. Il y a un peu plus de 20 ans, attiré tant par la curiosité et que par le dédommagement promis, j’avais loué pour quelques jours mon cabinet de jeune médecin généraliste récemment installé à Vendôme à l’équipe du réalisateur Michel Deville, qui y avait tourné quelques scènes de l’adaptation du roman de Martin Winckler « la maladie de Sachs », avec Albert Dupontel dans le rôle éponyme1. De cette expérience enrichissante à double titre, j’ai gardé en précieux souvenir un ordonnancier édité pour le film au nom du Docteur Bruno Sachs, médecin généraliste... Curieusement, c’est à la même époque que je me passionnais pour le modèle de démarche décisionnelle proposé par Sackett qui commençait à faire les gros titres du BMJ, tant par sa pertinence pour l’exercice de la médecine que par son impertinence – et celle de ses promoteurs – vis-à-vis du mandarinat médical et universitaire de l’époque2. L’evidence-based medicine envoyait au placard la médecine fondée sur la parole des Maîtres, et – au grand dam de ces derniers – remplaçait la vérité révélée par les niveaux de preuves. Je me souviens à l’époque des difficultés que nous avions à expliquer à nos confrères dubitatifs en quoi ce modèle aux apparences froidement scientifiques ne s’opposait en aucun cas à l’humanisme inhérent à l’exercice de la médecine générale, dont Martin Winckler proposait avec Bruno Sachs un archétype. Il nous semblait, et il nous semble encore, que c’est même là le coeur de notre métier : exercer une profession profondément humaine et sociale tout en fondant sa démarche décisionnelle sur des bases scientifiques. AbstractSpermatogenesis in two species of free-living marine nematodes from the family Chromadoridae (Panduripharynx pacifica and Euchromadora robusta) was studied electron-microscopically. The spermatogonia of both species are undifferentiated polygonal cells with a large nucleus surrounded by a small amount of cytoplasm. In P. pacifica the cytoplasm of spermatocytes contains many Golgi bodies, cisternae of RER, ribosomes, mitochondria and dense spherical bodies. Filamentous material is accumulated in spermatids, which contain only mitochondria and a fragmented (or lobed) nucleus devoid of the nuclear envelope. The immature sperm resembles the late spermatid: its central filamentous area is surrounded by chromatine particles and occasional mitochondria. The immature sperm plasma membrane forms deep infoldings. Mature spermatozoa from the uterus consist of a small main cell body (MCB) bearing a prominent pseudopod filled with cytoskeleton filaments. The MCB contains a nucleus and mitochondria. Spermatogenesis in E. robusta (studied only in testes) resembles that described for P. pacifica, but spermatocytes of E. robusta show much lower metabolic activity and, as a result, a smaller mass of filamentous material is stored in the spermatids and immature sperm. The spermatozoa of P. pacifica and the immature sperm of E. robusta have the main ultrastructural features characteristic for nematodes (amoeboid nature, absence of axoneme, acrosome and nuclear envelope). No aberrant organelles special for many nematode sperm (membranous organelles, paracrystalline fibrous bodies and their complexes) were found during sperm development of the chromadorids studied. In this respect their spermatogenesis differs significantly from that in secernents and monhysterids.La spermatogenèse a été étudiée en microscopie électronique à transmission chez deux espèces de nématodes libres marins (Panduripharynx pacifica et Euchromadora robusta) de la famille des Chromadoridae. BackgroundChildren with chronic non-infectious uveitis (NIU) are at risk for sight-threatening complications. Methotrexate (MTX) is the preferred first-line systemic treatment. Initial therapeutic response takes 3-6 months to achieve NIU control, leading to prolonged glucocorticoid use. Moreover, at least 50-70% of children fail to fully respond and may accrue ocular damage while awaiting MTX response.ObjectivesTo identify features of children with NIU that are associated with MTX failure.MethodsWe included children who started MTX monotherapy for NIU. We collected demographics, NIU features (type, location, & complications), ANA status, and clinical examination. We characterized children as MTX responders or non-responders. MTX responders are those whose NIU was controlled, defined by: 1) inactive graded by SUN criteria, 2) absence of new or worsening complications, and 3) requirement for ≤2 drops of prednisolone acetate and no oral glucocorticoids.ResultsOf 47 NIU children, 68% were MTX non-responders (Table 1), having a longer duration of NIU (Odds Ratio [OR]=1.28, [CI=1.03-1.8], p =0.023) and developing more ocular complications (OR=1.95 [CI=1.23-3.38], p=0.017), (ROC Area Under the Curve = 0.85). MTX non-responders were more likely to have anterior and/or posterior synechiae (p = 0.001), cataracts (p=0.015), and ocular hypertension (p=0.039). Abstract Abstract 1498 B-cell precursor acute lymphoblastic leukemia arising from overexpression of CRLF2 (CRLF2 B-ALL) is high-risk with poor prognosis. CRLF2 B-ALL occurs 5 times more frequently among children of Hispanic/Latino ethnicity than others and thus represents one of the most significant biological components of childhood cancer health disparity identified to date. CRLF2, together with the IL-7Rα, forms a receptor complex that is activated by the cytokine, TSLP. The JAK-STAT5 pathway is phosphorylated downstream of this receptor complex activation. Activating JAK mutations are found in some CRLF2 B-ALL and their presence has led to speculation that TSLP stimulation is not a factor in CRLF2 B-ALL. In preliminary studies to address this question we evaluated the effect of TSLP on CRLF2 B-ALL cell lines that have JAK defects and which have been reported to exhibit constitutive JAK-STAT5 activation. Our data show that TSLP increases STAT5 phosphorylation in these cell lines and also in primary B-ALL cells that overexpress CRLF2. Our next step was to evaluate the role of TSLP-CRLF2 interaction in vivo in the human-mouse xenograft model. However, mouse TSLP is different from most other cytokines produced in mice in that it is species-specific and does not activate the human TSLP receptor complex that contains CRLF2. Thus, traditional xenograft models do not provide the TSLP-CRLF2 interaction that our data implicate as a potential contributor to pathogenesis in CRLF2 B-ALL. To overcome this obstacle we engineered immune deficient NOD/SCID IL-2Rγ null (NSG) mice to express human TSLP (hTSLP+ mice), as well as control mice that lack the TSLP cytokine (hTSLP– mice). ELISA assays show plasma hTSLP levels in the hTSLP+ mice that approximate the normal range in human plasma. We used this hTSLP+/– xenograft model system to study the in vivo effects of TSLP on CRLF2 B-ALL cells harboring a JAK defect (MUTZ5 cell line) and on primary CRLF2 B-ALL cells from a Hispanic patient. Mice were euthanized at 5 weeks and BM disease was evaluated. In recipients of MUTZ5 B-ALL cells the percentage of viable leukemia cells in hTSLP– mice was half that observed in hTSLP+ mice. Similarly, in recipients of primary CRLF2 B-ALL cells, the percentage of viable leukemia cells was reduced in hTSLP– mice as compared to hTSLP+ mice. In addition, the viable primary B-ALL cells present in the BM of hTSLP+ mice showed higher expression levels of the TSLPR components (CRLF2 and IL-7Rα) than those in the hTSLP– mice. These data provide evidence that the TSLP produced in this model is active and that it impacts primary CRLF2 B-ALL cells. The hTSLP+/– xenograft model described here provides the first data on the in vivo role of TSLP-CRLF2 interactions in CRLF2 B-ALL. This preliminary data suggests that TSLP provides a signal that promotes in vivo survival of CRLF2 B-ALL cells and that it could play a role in selection of CRLF2-HI clones during in vivo leukemogenesis. This pre-clinical model will allow us to evaluate TSLP-CRLF2 interactions as a target for therapy and to perform translational studies that identify molecular mechanisms and additional targets downstream of TSLP-induced signaling in CRLF2 B-ALL. This model system will be be particularly important for assessing and identifying therapies, including drug and cellular delivery systems, to effectively target CRLF2 B-ALL and to reduce cancer health disparities in Hispanic childhood B-ALL. Abstract: In this work after X-ray irradiation outcomes of the evaluation of dynamic characteristics interstitial atoms Sij, vacancy V and O-complexes was applied for account of a condition of an annealing with the purpose of deriving specific structural defects in SiO2/Si wafer-plate. The nondestructive method, which is allow to determine from internal friction difference ΔQ-1/Q-1 0 of elastic vibrations structure defects density Nd and the depth of broken layer hb, is offered for SiO2/Si wafer-plates. 1. Introduction Defect annealing leads to a change in the shape of the internal friction (IF) temperature spectrum Q-1(T) [1]. IF method allows to set the spectrum of structural defects on the analysis of positions of maximums IF, on duration of relaxation time τ and on their deposit in attenuation of elastic vibrations [2]. A non-destructive method for the technological control of the structure defects by measuring internal friction (IF) and elastic modulus E after laser radiation was developed. 2. Experimental procedure Ultrasonic (US) pulse-phase method using USMV-LETI, modernized USMV-KNU and computerized “KERN-4” with frequencies f║ ≈ 1 MHz and f┴ ≈ 0,7 MHz, US invariant-polarization method for determining the effective acoustic μil and elastic constants Cijkl were used [3,4]. The measured velocity error was equal to ΔV/V = 0,5÷1,5%. The study of influence of structure defects on damping of vibrations in Si/SiO2 wafer-plates by the diameter of D = 100÷60 mm and by the thickness of hSiO2 ≈ 600 nm, hSi ≈ 470 000 nm, allows to estimate the degree of perfection of crystalline structure Metallography optical supervision of microstructure by means of the microscope ”LOMO MVT”, atomic-force microscopy (AFM) were used. 3. Results and discussion The quasi-longitudinal US velocity V║[001] = 5870 m/s, elastic modulus E001 = ρV║[001] 2 = 80,28 GPa for SiO2/Si from the oscillogram were determined. Temperature dependence of internal friction Q-1(T) in SiO2/Si wafer-plate p-type, doped with B, KDB-7.5(100) diameter D ≈ 76∙10-3 m, thickness hSi ≈ 460∙103 nm with SiO2 layer thickness hSiO2 ≈ 100 nm after X-ray irradiation with dose Dγ ≈ 102 Gy is showed. 4. Conclusions 1. The growth of internal friction maximum height QM -1 testifies the growth of the structural defects concentration n, and the broadening of internal friction maximum ΔQM -1 here represents the relaxation process of structural defects new types in SiO2/Si wafer-plate. 2. It was found that as the result of the structural defect annealing IF background Q0 -1 significantly decreases during measuring of IF temperature dependence Q-1(T), which indicates the improvement of SiO2/Si crystal structure. 3. The elastic modulus Е, the shear modulus G, Poisson coefficient μ, IF Q-1 are dependent from SiO2/Si wafer-plate anisotropy. 4. The value of IF background Q-1 0 after temperature T, mechanical treatments describes the changes of the elastic stress σi fields in SiO2/Si wafer-plate. 5. The study of vibrations of disk wafer-plate Si/SiO2 at different harmonic frequencies f0, f2 made it possible to develop the technique for determining the structural defects density nD for semiconductor wafers-substrates. 6. The relationship between IF Q-1 value, the logarithmic decrement of ultrasound damping δ and the dislocations density nD was established for disk-shaped semiconductor wafers-substrates. Abstract Introduction The binding of immunomodulatory drugs (IMiDs), including lenalidomide, to CRBN is associated with cytotoxicity of IMiDs used in the treatment of multiple myeloma, myelodysplastic syndromes (MDS) and lymphomas. Cereblon (CRBN) is named for its putative role in cerebral development, especially in memory and learning. CRBN forms an E3 ubiquitin ligase complex with damaged DNA binding protein 1 (DDB1), cullin-4 (CUL4) and regulator of cullin 1 (ROC1). This complex regulates DNA repair, DNA replication and transcription. CRBN is a primary target of thalidomide teratogenicity (Ito et al., Science 2010; 327:1345-1350). Down-regulation of the CRBN expression is associated with the development of marked IMiDs resistance in human multiple myeloma cells (Zhu et al., Blood 2011; 118: 4771-4779; Lopez Girona et al., Leukemia 2012; 26: 2326-2335; Heintel et al., Br J Haematol 2013; 161: 695-700; Broyl et al., Blood 2013; 121: 624-627; Lodé et al., Br J Haematol 2013; Jul 17. doi: 10.1111/bjh.12478). CRBN expression is thus required for the antimyeloma activity of IMiDs. Aims To gain insight into the role of CRBN in lower risk myelodysplastic syndromes with or without 5q deletion and into the mechanisms of lenalidomide action, we study CRBN expression in these two groups of lower risk MDS patients and in healthy controls. We further study the expression of DDB1 and IRF4 (interferon regulatory factor 4, one of target genes of CRBN). Methods Mononuclear cells were isolated both from bone marrow [23 low risk MDS patients with 5q- deletion, 37 low-risk MDS patients with normal chromosome 5 (non5q-) and 24 healthy controls] and from peripheral blood [38 patients with 5q- , 52 non 5q- patients and 25 healthy controls] by Ficoll-Paque PLUS gradient separation, and washed with phosphate-buffered saline. The rest of the red cells were lysed. Total RNA was isolated using RNA isolation solvent and complementary DNA was synthesized from total RNA using SuperScript II reverse transcriptase. Relative levels of the CRBN, DDB1 and IRF4 mRNAs were determined by TaqMan-based quantitative real-time PCR and by calculation to the level of housekeeping glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA. The experiments were performed in duplicate. Informed consent was obtained from all patients and healthy controls. Evaluation of 2-ddCt indicates the fold change in gene expression relative to the control. Results The median of CRBN mRNA levels in total RNA isolated from peripheral blood mononuclear cells was the highest (3.6) in lower risk MDS with 5q deletion, lower (1.5) in non5q- MDS and lowest (1.2) in healthy controls. Similar results were obtained in bone marrow mononuclear cells (medians 3.3 in 5q- syndrome, 2.2 in non5q- MDS and 1.3 in healthy controls). The differences between the 3 groups were statistically significant (p˂0.05, Mann-Whitney test) with the exception of non5q- MDS in comparison with healthy controls in mononuclear cells of peripheral blood. Very similar results were obtained in the analysis of DDB1 and IRF4 mRNAs levels, which correlated with CRBN levels. We analyzed CRBN mRNA levels before and in the course of the lenalidomide treatment of 5q- low risk MDS (6 patients). We obtained high CRBN mRNA levels before and during the treatment by lenalidomide in all lenalidomide responders (4 patients). We found a sharp decrease of CRBN mRNA in mononuclear cells of bone marrow in two patients who stopped responding to lenalidomide therapy and subsequently progressed to higher risk MDS. A/G polymorphism located at -29 nt of the 5x UTR of CRBN does not act as a biomarker of response to treatment with lenalidomide in 5q- syndrome. Conclusions 5q- low risk MDS patients have the highest levels of CRBN mRNA in comparison to lower risk myelodysplastic syndromes with normal chromosome 5 and to healthy controls. DDB1 and IRF4 mRNAs levels correlate with CRBN levels. High levels of CRBN mRNA were detected in all lenalidomide responders during the course of the therapy. Significant decrease of CRBN levels during treatment by lenalidomide is associated with loss of response to treatment and disease progression. Similarly to the treatment of multiple myeloma, these results suggest that high levels of CRBN mRNA are necessary for the efficacy of lenalidomide in low risk 5q- patients. Background: The burden of human immunodeficiency virus (HIV) in cervical cancer remains a major public health challenge in developing countries, like Togo. Precancerous lesions include all cellular abnormalities that have malignant potential that can develop into cancer. The objective of this study was to determine the prevalence and factors associated with precancerous cervical lesions in HIV-infected women in our context. Methods: A cross-sectional descriptive study was carried out from November 31, 2022, to January 31, 2023, in an HIV care center in Lomé (Non-Governmental Organization Espoir Vie Togo [NGO EVT] Grand-Lomé-Togo). Results: A total of 271 women were included with a mean age of 47.0 years and a standard deviation of 10.0 years, among whom 20.7% do not have any scholar education. Only 6.7% of them had previously performed cervical smear examinations. The prevalence of precancerous cervical lesions observed in people living with human immunodeficiency virus (PLHIV) was 11.4% with a 95% confidence interval (CI) of 5.0 to 15.4. Cytological abnormalities were marked by low-grade squamous intraepithelial lesion (LSIL) (5.1%), followed by the presence of atypical squamous cells of undetermined significance (ASCUS) (3.5%). A statistically significant association was found between parity and the presence of precancerous lesions ( P = .014). Conclusions: In this study, more than 1 out of 10 women living with HIV had precancerous cervical lesions, and parity was the factor associated. The implementation of a systematic screening program for precancerous cervical lesions and human papillomavirus (HPV) infection integrated into HIV care is necessary for early treatment.
