Abstract:
ABSTRACTGougerotin is a peptidyl nucleoside antibiotic. It functions as a specific inhibitor of protein synthesis by binding ribosomal peptidyl transferase and exhibits a broad spectrum of biological activities.gouR, situated in the gougerotin biosynthetic gene cluster, encodes a TetR family transcriptional regulatory protein. Gene disruption and genetic complementation revealed thatgouRplays an important role in the biosynthesis of gougerotin. Transcriptional analysis suggested that GouR represses the transcription of thegouL-to-gouBoperon consisting of 11 structural genes and activates the transcription of the major facilitator superfamily (MFS) transporter gene (gouM). Electrophoresis mobility shift assays (EMSAs) and DNase I footprinting experiments showed that GouR has specific DNA-binding activity for the promoter regions ofgouL,gouM, andgouR. Our data suggested that GouR modulates gougerotin production by coordinating its biosynthesis and export inStreptomyces graminearus.

Abstract:
Lascaux Cave is renowned for its outstanding prehistoric paintings, strikingly well-preserved over about 18,000 yr. While stalagmites and stalactites are almost absent in the cave, there is an extensive calcite flowstone that covered a large part of the cave until its opening for tourists during the 1950s. The deposit comprises a succession of calcite rims, or “gours,” which allowed seepage water to pond in large areas in the cave. Their possible role in preservation of the cave paintings has often been evoked, but until now this deposit has not been studied in detail. Here, we present 24 new radiocarbon accelerator mass spectrometry (AMS) and 6 uranium-thorium (U-Th) analyses from the calcite of the gours, 4 AMS 14C dates from charcoals trapped in the calcite, and 4 AMS 14C analyses on organic matter extracted from the calcite. Combining the calibrated 14C ages obtained on charcoals and organic matter and U-Th ages from 14C analyses made on the carbonate, has allowed the calculation of the dead carbon proportion (dcp) of the carbonate deposits. The latter, used with the initial atmospheric 14C activities reconstructed with the new IntCal09 calibration data, allows high-resolution age estimation of the gour calcite samples and their growth rates. The carbonate deposit grew between 9530 and 6635 yr cal BP (for dcp = 10.7 ± 1.8%; 2 σ) or between 8518 and 5489 yr cal BP (for dcp = 20.5 ± 1.9%; 2 σ). This coincides with humid periods that can be related to the Atlantic period in Europe and to Sapropel 1 in the eastern Mediterranean Sea. However, geomorphological changes at the cave entrance might also have played a role in the gour development. In the 1940s, when humans entered the cave for the first time since its prehistoric occupation, the calcite gours had already been inactive for several thousand years.

Abstract:
A new sub-type of folia named “reverse pseudo-gour” has been observed and described in the Nerja Cave, southern Spain. It consists of fairly vertical, thin barriers (about 5 mm high and 2 mm thick) that develop on the underside of a sub-horizontal surface (shelfstone) and grow in the opposite direction to normal gours (rimstone dams), generating sinuous shapes. Their mineral composition is essentially calcium carbonate, although globular aggregates composed of clay and phosphate minerals have also been identified. The genesis and evolution of these reverse pseudo-gours occur just at the air-water interface and are controlled by (1) the sub-horizontality of the surface on which they develop, (2) scarce to very scarce calcite supersaturation, and (3) progressive and slow lowering of the water level. Thus, reverse pseudo-gours represent the shape limit to which a folia reaches when its development occurs just below a nearly horizontal ceiling and the feeding water saturation is extremely low. Although this new sub-type of folia has only been observed in the Nerja Cave, it is highly probable that it exists in several other caves around the world because the factors controlling its development are not very strict.

Abstract:
Abstract The emergence of humanistic geographers like Tim Cresswell, Edward Relph, and Yi-Fu Tuan from the 1970s onwards redefined the meaning of ‘place’, through extensive emphasis on human experience within and beyond the physical landscape. Since then ‘place’ has stretched its domain and traversed the terrains of various disciplines, including literary study and production. Discussing ‘place’ in relation to how the acclaimed Indian writer Neelum Saran Gour represents Allahabad shows how she reframes the ‘cultural geography’ of the city. While decoding her literary spaces, this interview focusses on the multidimensional concept of ‘place’ from geographical and social–cultural perspectives and how Allahabad, or any other place like Allahabad for that matter, becomes an extension of the writer’s ‘self’ and its inhabitants. This interview also explicates how Gour conceives the invisibilities of multicultural North Indian society in terms of its various linguistic and gendered identities. In turn, Gour’s work moves from regional singularity to represent ‘Indianness’ more broadly.

Abstract:
Philosophiae Doctor - PhD
This study examines the problems of implementing water allocation policy in the context of the local state bureaucracy as well as the specific experiences of local black emerging farmers in the Breede Gouritz Water Management Area. This study used qualitative research methods and is based on many hours of interviews and observing bureaucrats and stakeholders at the receiving end of the bureaucratic business process of water allocation. It is not only concerned with the physical and technical aspects of access but explores how the different role players interact, navigate, shape, frame and manage challenges to gain access to and control water for productive use. The actual experiences and understandings of the stakeholders in their own contexts when engaging with the access to water are crucial to gain a comprehensive understanding and insight into the influence of bureaucracy and power relations. This thesis therefore maps the confusions and incapacities and shows that even though the South African laws are based on the best international frameworks, they fail, as they do not sufficiently address the unique environment and landscape. Existing scholarship has not adequately researched local bureaucratic power. At the coalface of implementation, bureaucrats make up their own rules to cope with rapid policy churning. Combined with existing power relations, policy implementation and policy direction is steered towards different and unintended trajectories, making transformation a challenge to achieve. Consequently, my main finding is that there have been constant and rapid legislative and policy changes but they have simply added to the confusion and instability.

Abstract:

Abstract:

Abstract:
Heart failure (HF) and gout are common medical conditions that lead to significant morbidity and mortality in Canada and the US. We conducted a population-based nested case-control study to investigate the associations between gout, allopurinol use and HF outcomes using administrative data from the province of Quebec. Our analysis demonstrates that a history of gout is associated with increased HF readmission and death in HF patients (adjusted RR 1.63; 95% confidence interval (CI) 1.48-1.80). We also show that chronic allopurinol use, a common medication for gout, is associated with reductions in adverse outcomes in HF patients with a history of gout (adjusted RR 0.69, 95% CI 0.60-0.79). Due to the observational nature of our study, residual confounding due to unknown or unmeasured confounding could still bias our findings. Herein, we present a novel method to estimate and control for such confounding by estimating the confounding risk ratio during the null induction time. Use of this method is illustrated using an example from the literature and we discuss the assumptions, limitations and statistical considerations of our method.

Yermiyahu, Uri, Thomas Kinraide, and Uri Mingelgrin. Role of Binding to the Root Surface and Electrostatic Attraction in the Uptake of Heavy Metal by Plants. United States Department of Agriculture, 2000. http://dx.doi.org/10.32747/2000.7586482.bard.

Abstract:
The principal accomplishment of the research supported by BARD was progress toward a comprehensive view of cell-surface electrical effects (both in cell walls [CWs] and at plasma membrane [PM] surfaces) upon ion uptake, intoxication, and amelioration. The research confirmed that electrostatic models (e.g., Gouy-Chapman-Stern [G-C-S]), with parameter values contributed by us, successfully predict ion behavior at cell surfaces. Specific research objectives 1. To characterize the sorption of selected heavy metals (Cu, Zn, Pb, Cd) to the root PM in the presence of other cations and organic ligands (citric and humic acids). 2. To compute the parameters of a G-C-S model for heavy-metal sorption to the root PM. 3. To characterize the accumulation of selected heavy metals in various plant parts. 4. To determine whether model-computed ion binding or ion activities at root PM surfaces predict heavy-metal accumulation in whole roots, root tips, or plant shoots. 5. To determine whether measured ion binding by protoplast-free roots (i.e., root CWs) predicts heavy-metal accumulation in whole roots, root tips, or plant shoots. 6. To correlate growth inhibition, and other toxic responses, with the measured and computed factors mentioned above. 7. To determine whether genotypic differences in heavy-metal accumulation and toxic responses correlate with genotypic differences in parameters of the G-C-S model. Of the original objectives, all except for objective 7 were met. Work performed to meet the other objectives, and necessitated on the basis of experimental findings, took the time that would have been required to meet objective 7. In addition, work with Pb was unsuccessful due to experimental complications and work on Cd is still in progress. On the other hand, the uptake and toxicity of the anion, selenate was characterized with respect to electrostatic effects and the influences of metal cations. In addition, the project included more theoretical work, supported by experimentation, than was originally planned. This included transmembrane ion fluxes considered in terms of PM-surface electrical potentials and the influence of CWs upon ion concentrations at PM surfaces. A important feature of the biogeochemistry of trace elements in the rhizosphere is the interaction between plant-root surfaces and the ions present in the soil solution. The ions, especially the cations, of the soil solution may be accumulated in the aqueous phases of cell surfaces external to the PMs, sometimes referred to as the "water free space" and the "Donnan free space". In addition, ions may bind to the CW components or to the PM surface with variable binding strength. Accumulation at the cell surface often leads to accumulation in other plant parts with implications for the safety and quality of foods. A G-C-S model for PMs and a Donnan-plus-binding model for CWs were used successfully to compute electrical potentials, ion binding, and ion concentration at root-cell surfaces. With these electrical potentials, corresponding values for ion activities may be computed that are at least proportional to actual values also. The computed cell-surface ion activities predict and explain ion uptake, intoxication, and amelioration of intoxication much more accurately than ion activities in the bulk-phase rooting medium.
