Abstract:
Differentiation of endometrial stromal cells into decidual cells is essential for successful embryo implantation. Interleukin (IL)-11 signalling is required for decidualisation in the mouse (1,2) and the expression pattern of IL-11 and its receptors during the menstrual cycle suggests a role for IL-11 in human decidualisation (3). Exogenous IL-11 has been shown to enhance hormone-induced decidualisation of human endometrial stromal cells in culture (4). This study aimed to determine the effects of IL-11 on downstream gene expression in endometrial stromal cells following 12 days of progesterone-induced decidualisation, and to examine the expression and functional significance of IL-11 target genes during this process. Stromal cells isolated from endometrial biopsies (n = 6) were decidualised with 17β-oestradiol and medroxyprogesterone acetate (EP) or EP with 100 ng/mL recombinant human IL-11. Medium was changed every 48 h, and total RNA extracted on Day 12 for gene expression analysis using custom-made 15K cDNA microarrays. Quantitative real-time RT-PCR was performed on the same samples to confirm gene expression levels. In subsequent experiments (n = 2), cells were cytocentrifuged onto glass slides for immunocytochemistry using specific antibodies. Microarray analysis revealed 16 upregulated and 11 downregulated cDNAs in EP + IL-11 compared to EP treated cells. Among these were IL-1β (6.1-fold upregulated) and insulin-like growth factor binding protein (IGFBP)-5 (3.6-fold downregulated). Using real-time RT-PCR, IL-11 was confirmed to increase IL-1β (fold change 1.3–107.1) and decrease IGFBP-5 (fold change 2.8–469.0) transcript abundance in 6 patients. Immunolocalisation of IL-1β in EP and EP + IL-11 treated cells revealed more intense vesicular cytoplasmic staining with IL-11 treatment, while staining intensity for IGFBP-5 was not affected. Interactions between IL-11 and its downstream targets IL-1β and IGFBP-5 are likely to have functional importance in early pregnancy, and may provide novel targets for the manipulation of human fertility. V, 78-87.Englanderen Sir Edmund Gosse (1849-1928) var i sin samtid en fremtrædende og indflydelsesrig litterar skikkelse, hvis fortjeneste det var at formidle nordisk litteratur, især forfatteren Henrik Ibsens forfatterskab, hvoraf han oversatte enkelte værker, til den engelsksprogede verden.Som ung mand besøgte Gosse Danmark to gange. Under sine besøg i København i 1872 boede han hos Holmens provst (senere biskop) Bruun Juul Fog og dennes ugifte søster Aline. Hans beretning om disse besøg blev Forst offentliggjort flere ar efter, at besøgene havde fundet sted, hvorfor man af denne og andre årsager ma modtage detaljerne med nogen forsigtighed.Men Gosses beretning er skrevet i en forfriskende selvstændig and, balancerende mellem beundring og kritik, og pa trods af dens mangler er hans impressionistiske beretning af atmosfæren i Vartov kirke i Grundtvigs sidste tid et vigtigt vidnesbyrd.Norden havde pa denne tid ikke nogen mere ekstraordinær personlighed end Grundtvig, siger Gosse, og han giver en kortfattet beretning af Grundtvigs modige og stadige kamp for kirkens frihed. Spider silk is a material of extraordinary beauty and utility. From the spider's perspective, it is foremost a building material, but also a safety net, a sensory organ, a weapon. From a human perspective, it is a material of extraordinary mechanical properties, an object of artistic and cultural interest, and a valuable window into the evolutionary history of spiders. Historically, there have been a mere handful of spider silk textiles, the most recent example of which was constructed by Simon Peers and Nicholas Godley. This was an extravagant work of art, and the silk was collected entirely by hand. Though the finished article is a testament to the beauty of spider silk, it also illustrates the technical challenges associated with obtaining it in any significant quantity. The effects of this scarcity are evident in the lab of David Kaplan at Tufts University. His group has developed a wide variety of applications for silk, but has focused mainly on silkworm silk in spite of spider silk's greater variety and superior mechanical properties. In the wild, spiders use silk for everything from weaving webs and capturing prey to breathing underwater. Shaped by almost every environment on Earth, spider silk has evolved into endless variations and permutations, offering a vast wealth of material knowledge if we can find a way to tap it. A visit to Cheryl Hayashi's spider silk genetics lab at the University of California, Riverside offers a look inside a spider and a glimpse of how genetic research can illuminate the evolution of silk. At the same time, it puts the limits of our knowledge into stark relief. The scarcity of natural spider silk has helped to drive a small industry in bioengineered and synthetic silk research, and also motivated projects that seek to apply the structural principles of silk fibers to other materials. But so far, these efforts are only a pale imitation of the real thing. For now, the spider is keeping her secrets.

Inflated space-based structures have become popular over the past three decades due to their minimal launch-mass and launch-volume. Once inflated, these space structures are subject to vibrations induced by guidance systems and space debris as well as from variable amounts of direct sunlight. Understanding the dynamic behavior of space-based structures is critical to ensuring their desired performance. Inflated materials, however, pose special problems when testing and trying to control their vibrations because of their lightweight, flexibility, and high damping. Traditional modal testing techniques, based on single-input, single-output (SISO) methods, are limited for a variety of reasons when compared to their multiple counterparts. More specifically, SISO modal testing techniques are unable to reliably distinguish between pairs of modes that are inherent to axi-symmetric structures (such as an inflated torus, a critical component of a gossamer spacecraft). Furthermore, it is questionable as to whether a single actuator could reliably excite the global modes of a true gossamer craft, such as a 25 m diameter torus. In this study, we demonstrate the feasibility of using a multiple-input multiple-output (MIMO) modal testing technique on an inflated torus. In particular, the refined modal testing methodology focuses on using Macro-Fiber Composite (MFC®) patches (from NASA Langley Research Center) as both actuators and sensors. MFC® patches can be integrated in an unobtrusive way into the skin of the torus, and can be used to find a gossamer structure’s modal parameters. Furthermore, MFC® excitation produces less interference with suspension modes of the free-free torus than excitations from a conventional shaker. The use of multiple actuators is shown to properly excite the global modes of the structure and distinguish between pairs of modes at nearly identical resonant frequencies. Formulation of the MIMO test as well as the required postprocessing techniques are explained and successfully applied to an inflated Kapton® torus.
