Abstract:
Le myélome multiple (MM), une hémopathie maligne qui représente environ 13% des cancers hématologiques, est caractérisée par la prolifération de plasma cells tumoraux au niveau de la moelle osseuse. Le MM évolue à partir de stades précurseurs, à savoir la gammapathie monoclonale de signification indéterminée (MGUS) et le myélome multiple asymptomatique (SMM), vers la forme symptomatique, le MM. C’est une hémopathie maligne incurable dont l’hétérogénéité et l’évolution clonale permettent l’échappement aux traitements et la progression de la maladie. Les altérations de MYC ont un rôle essentiel dans cette progression. Cependant, MYC n'est pas ciblable thérapeutiquement en raison de sa localisation nucléaire et de la courte demi-vie de la protéine.Pour surmonter cela, nous avons fait l’hypothèse que l’avantage prolifératif induit par la surexpression de MYC crée des dépendances des cellules tumorales vis-à-vis d’autres voies de signalisation qui deviennent indispensables à la survie de ces cellules. Pour tester cette hypothèse, nous avons appliqué une nouvelle méthodologie utilisant la carte de dépendance (Achilles) et effectué un screening de 2000 petites molécules afin d'identifier les vulnérabilités génomiques induites par MYC. Si elles sont identifiées, ces vulnérabilités offrent une possibilité de traitement ciblé des cancers ayant une surexpression de MYC. Nos analyses démontrent la dépendance des lignées cellulaires surexprimant MYC pour le métabolisme de la glutamine, spécifiquement les gène GLS1 (glutaminase). Nous avons validé et délimité fonctionnellement cette dépendance in vitro à partir des différentes approches.Par l’analyse de notre criblage de 1869 composés chimiques, nous avons observé que les inhibiteurs de la synthèse de NAD avaient un effet préférentiel sur la prolifération des cellules surexprimant MYC. Considérant que les rôles métaboliques du glutamine sont liés à ceux du NAD, nous avons ensuite exploré un effet synergique potentiel entre les inhibiteurs du GLS1 et du NAMPT. Nous avons démontré l'efficacité de cette nouvelle combinaison synergique pour cibler les cellules MM surexprimant MYC in vitro et in vivo.Ces résultats établissent une base méthodologique solide utilisable pour développer de nouvelles approches thérapeutiques afin de répondre à des besoins thérapeutiques non satisfaits pour cibler le MYC dans le MM
Multiple myeloma (MM) is a hematological malignancy that accounts for around 13% of hematological cancers and is characterized by the uncontrolled proliferation of malignant plasma cells in the bone marrow. MM progresses from precursor stages, known as monoclonal gammopathy of undetermined significance (MGUS) and smoldering multiple myeloma (SMM), to the symptomatic form, MM. It is an incurable malignancy in which heterogeneity and clonal evolution allow treatment escape and disease progression. MYC alterations play an essential role in this progression. However, MYC is not therapeutically targetable due to its nuclear localization and the protein's short half-life.To overcome this, we hypothesized that the proliferative advantage induced by MYC overexpression creates genomic dependencies on other signalling pathways that become essential for cell survival. To test this hypothesis, we applied a novel approach by leveraging large-scale loss of function screen (Achilles) and 1869 small molecules screen to identify MYC-induced genomic vulnerabilities. When identified, these vulnerabilities offer an opportunity to selectively target cancer cells harbouring this overexpression and spare normal cells.Our analyses demonstrate the dependence of MYC overexpressing cells on glutamine metabolism, in particular on the GLS1 (glutaminase). We validated and functionally delineated this dependence in vitro using different approaches.Our small molecule screen highlighted that NAD synthesis inhibitors had a preferential effect on the proliferation of MYC overexpressing cells. Considering that glutamine and NAD have closely interlinked metabolic networks, we investigated the possibility of a potential synergistic effect between GLS1 and NAMPT inhibitors. We demonstrated the effectiveness of this new synergistic combination to target MYC-driven MM cells in vitro and in vivo.These results establish a solid methodological basis that can be used to develop new therapeutic approaches to address unmet therapeutic needs to target MYC in MM

The energy metabolism of most tumor cells relies on aerobic glycolysis (Warburg effect) characterized by an increased glycolytic flux that is accompanied by the increased formation of the cytotoxic metabolite methylglyoxal (MGO). Consequently, the rate of detoxification of this reactive glycolytic byproduct needs to be increased in order to prevent deleterious effects to the cells. This is brought about by an increased expression of glyoxalase 1 (GLO1) that is the rate-limiting enzyme of the MGO-detoxifying glyoxalase system. Here, we overexpressed GLO1 in HEK 293 cells and silenced it in MCF-7 cells using shRNA. Tumor-related properties of wild type and transformed cells were compared and key glycolytic enzyme activities assessed. Furthermore, the cells were subjected to hypoxic conditions to analyze the impact on cell proliferation and enzyme activities. Our results demonstrate that knockdown of GLO1 in the cancer cells significantly reduced tumor-associated properties such as migration and proliferation, whereas no functional alterations where found by overexpression of GLO1 in HEK 293 cells. In contrast, hypoxia caused inhibition of cell growth of all cells except of those overexpressing GLO1. Altogether, we conclude that GLO1 on one hand is crucial to maintaining tumor characteristics of malignant cells, and, on the other hand, supports malignant transformation of cells in a hypoxic environment when overexpressed.

In this thesis, we study strongly interacting superconformal field theories in two and three dimensions. In two dimensions, we investigate N = (0, 2) gauge theories using the gauged linear sigma models (GLSM). In those theories, we identify simple mechanism by which worldsheet description of H-flux satisfying Green-Schwarz Bianchi identity arises. Under the renormalization group flow, we argue that these models flow into superconformal fixed points describing string theory compactifications backgrounds with non-trivial H-flux turned on. By analyzing quantum-consistency of effective theories with such mechanism, we identify conditions under which these theories to become interacting superconformal field theories in the infrared. In three dimensions, we study maximally supersymmetric (N = 8) conformal field theories by conformal bootstrap approach. We focus on studying the four-point function of stress-tensor multiplet. The superconformal blocks for the four-point function are computed by analyzing superconformal Ward identity. Using these blocks, we study crossing symmetry constraints both numerically and analytically. Doing so, we obtain universal bounds and exact relations of N = 8 superconformal field theory data.

Si vedranno quali sono, e per quali ragioni sono state introdotte, le migliorie che hanno portato al Modern OpenGL, verranno trattati i principali tipi di shader utilizzati e le strutture dati che devono essere usate per poter comunicare eᨴcientemente con la scheda video, dove verranno eseguiti gli shader e calcolato il rendering della scena da noi descritta. Neste trabalho é estudada a viabilidade de uma implementação em paralelo do algoritmo scale invariant feature transform (SIFT) para identificação de íris. Para a implementação do código foi utilizada a arquitetura para computação paralela compute unified device architecture (CUDA) e a linguagem OpenGL shading language (GLSL). O algoritmo foi testado utilizando três bases de dados de olhos e íris, o noisy visible wavelength iris image Database (UBIRIS), Michal-Libor e CASIA. Testes foram feitos para determinar o tempo de processamento para verificação da presença ou não de um indivíduo em um banco de dados, determinar a eficiência dos algoritmos de busca implementados em GLSL e CUDA e buscar valores de calibração que melhoram o posicionamento e a distribuição dos pontos-chave na região de interesse (íris) e a robustez do programa final.
Present work studies the feasibility of a parallel implementation of the scene recognition algorithm SIFT for iris recognition. The code was built using the Compute Unified Device Architecture (CUDA) and the shading language GLSL. The algorithm was tested using three databases containing eyes and iris, the UBIRIS, Michal- Libor and CASIA. Tests were made for: analyzing the requested time for checking if an subject is or is not present on current database, the efficiency of the search algorithms written in CUDA and GLSL, the search for calibration values that improve keypoints position and distribution through the region of interest (iris), analyzing the reliability of the final code.

Abstract:
Malignant mesothelioma is an aggressive cancer of the membranes covering the lung and chest cavity (pleura), or the abdomen (peritoneum), mainly linked to asbestos exposure. It is characterized by high intrinsic heterogeneity, diagnosis in the late stages and a high immunosuppressive microenvironment. In the past years many agents have been evaluated for use in mesothelioma but with modest results so that the prognosis remains poor. Recently, in light of the promising results achieved in other cancers, the targeting of the Hedgehog-GLI (HH-GLI) pathway has been investigated as possible new therapy for MPM cure. The HH-GLI pathway starts at Primary Cilium (PC), an organelle protruding from the extracellular membrane of the cells that expresses specific receptors for the Hedgehog ligands. In cells lacking PC, the HH-GLI pathway can also be activated by intracellular signaling that make cells resistant to HH-GLI ligand-dependent pathway inhibitors. In MPM the HH-GLI signaling is active but response to targeting agents is poor. Activating mutations in the core components of the pathway that in other cancers lead to drug resistance in MPM are rare. Here we studied the presence of PC in mesothelioma and its correlation with HH-GLI pathway activation. We found an heterogeneous presence of PC in MPM and, in the cells loosing PC, GLI1 was overexpressed. Our preliminary results suggested that PI3K/AKT pathway can be, at least in some cells, responsible for the activation of HH-GLI1 pathway. In summary, we have documented for the first time the loss of PC in mesothelioma and the activation of a non-canonical HH-GLI pathways in this cancer.

Resumo: A dissertação apresenta o processo de estimação em pesquisas repetidas sob o enfoque de séries temporais, empregando Modelos de Espaço de Estados e o filtro dos mínimos quadrados generalizados ou filtro GLS. Este filtro permite o tratamento de modelos com erros de observação autocorrelacionados de forma mais simples do que utilizando o filtro de Kalman e, além disso, possibilita a modelagem conjunta de várias subpopulações ou domínios sob restrições de benchmark obtidas a partir da mesma pesquisa. Isto não só permite manter a coerência entre as estimativas obtidas pelo método, e estimativas agregadas baseadas no planejamento da amostra, como ajuda na proteção contra possíveis erros de especificação dos modelos. Considerando o caso de amostras com rotação, também é abordado o processo de estimação da estrutura de autocorrelação dos erros amostrais empregando o método dos pseudo-erros. Via simulação, é replicado todo o procedimento de estimação, comparando resultados obtidos empregando os filtros GLS e de Kalman. Adicionalmente, é ilustrada a aplicação do filtro sob restrições de benchmark empregando a série de taxa de desemprego da Pesquisa Mensal de Emprego do IBGE, no período de março de 2002 a fevereiro de 2012
Abstract: This work presents the estimation process in repeated surveys using State Space Models and the generalized linear squares filter, GLS filter, under the time series approach. This filter deals with autocorrelated errors in the observation equation, in a simpler way than the well-known Kalman filter. Additionally, it allows for modeling jointly several domains under benchmark constraints obtained from the same survey. The benchmarking not only achieves coherence between the model-based estimates and the corresponding design-based aggregated estimates, but also provides protection against possible model failures. For the scenario of samples with a rotation scheme, the estimation of the autocorrelation structure of observational errors, using the pseudo-errors method, is also addressed. Simulation are used to compare the GLS and Kalman filter estimators. Moreover, the application of GLS filter under benchmark restrictions is illustrated, using the unemployment rate time serie from the Brazilian monthly labor force survey, from March 2002 to February 2012

Abstract:
This thesis is devoted to the study of gauged linear sigma models (GLSMs) and mirror symmetry. The first chapter of this thesis aims to introduce some basics of GLSMs and mirror symmetry. The second chapter contains the author's contributions to new exact results for GLSMs obtained by applying supersymmetric localization. The first part of that chapter concerns supermanifolds. We use supersymmetric localization to show that A-twisted GLSM correlation functions for certain supermanifolds are equivalent to corresponding Atwisted GLSM correlation functions for hypersurfaces. The second part of that chapter defines associated Cartan theories for non-abelian GLSMs by studying partition functions as well as elliptic genera. The third part of that chapter focuses on N=(0,2) GLSMs. For those deformed from N=(2,2) GLSMs, we consider A/2-twisted theories and formulate the genuszero correlation functions in terms of Jeffrey-Kirwan-Grothendieck residues on Coulomb branches, which generalize the Jeffrey-Kirwan residue prescription relevant for the N=(2,2) locus. We reproduce known results for abelian GLSMs, and can systematically calculate more examples with new formulas that render the quantum sheaf cohomology relations and other properties manifest. We also include unpublished results for counting deformation parameters. The third chapter is about mirror symmetry. In the first part of the third chapter, we propose an extension of the Hori-Vafa mirrror construction [25] from abelian (2,2) GLSMs they considered to non-abelian (2,2) GLSMs with connected gauge groups, a potential solution to an old problem. We formally show that topological correlation functions of B-twisted mirror LGs match those of A-twisted gauge theories. In this thesis, we study two examples, Grassmannians and two-step flag manifolds, verifying in each case that the mirror correctly reproduces details ranging from the number of vacua and correlations functions to quantum cohomology relations. In the last part of the third chapter, we propose an extension of the Hori-Vafa construction [25] of (2,2) GLSM mirrors to (0,2) theories obtained from (2,2) theories by special tangent bundle deformations. Our ansatz can systematically produce the (0,2) mirrors of toric varieties and the results are consistent with existing examples which were produced by laborious guesswork. The last chapter briefly discusses some directions that the author would like to pursue in the future.

In this thesis, I summarize my work on gauged linear sigma models (GLSMs) and mirror symmetry. Esta dissertação filia-se à teoria analítica do discurso proposta pelo filósofo Michel Pêcheux e tem como tema central discussões sobre o processo de estruturação do discurso acadêmico que aborda a segmentação do Turismo no Brasil. Com essa pesquisa, busco contribuir com o estudo do Campo do Turismo a partir de uma perspectiva crítica, com foco específico no estudo do segmento caracterizado como Turismo GLS. Para tanto, busquei em livros – publicados no Brasil e escritos na comunidade acadêmica nacional – textos que relacionassem Turismo com a Comunidade LGBT, objetivando a construção desse conceito de segmento. Dessa busca, relacionei três textos que compõem o corpus de análise, são eles; Angeli (2004 [1999]), Oliveira (2002) e Trigo (2009). A partir do recorte das sequências discursivas das materialidades e da mobilização de conceitos como Formação Social, Formação Ideológica, Lugar Discursivo, Formações Discursivas e Forma-sujeito, desenvolvi um caminho teórico que pretende problematizar a apropriação da sigla GLS – enunciada dentro do Movimento Social como um Lugar Discursivo – pelo Mercado e, consequentemente, pela Academia. A partir desses achados de análise também aponto para o tratamento conceitual dado ao Sujeito nos processos de segmentação do Turismo, processos esses, que remetem ao individualismo e ao pragmatismo, evidenciando que a segmentação ainda situa-se em perspectivas bastante reducionistas. Nesse esforço de problematizar o segmento, o texto resta em aberto, mobilizando outros questionamentos e propostas de novos arranjos na interface Turismo – Análise do Discurso.

This dissertation relates to the discourse analysis proposed by the French philosopher Michel Pêcheux and it has as the main theme the discussions on the structuration process of the academic discourse which addresses the segmentation of Tourism in Brazil. With this research, I aim to contribute with the study in the Field of Tourism from a critical perspective, with emphasis on GLS Tourism. For this purpose, I have searched in books (published in Brazil and written within the national academic community) for texts that relates Tourism with the LGBT Community, addressing to the construction of this concept. From this search, I have related three texts which assemble the analysis corpus: Angeli (2004 [1999]), Oliveira (2002) and Trigo (2009). From the view of the materiality‟s sequential discourses and the mobilization of concepts like Social Formation, Ideological Formation, Discursive Place, Discursive Formations and Subject-Form, I have developed a theorical method which intends to discuss the appropriation of the acronym “GLS” (expressed within the LGBT Movement as a Discursive Place) by the Market and, inevitably, by the Academy. From these analysis findings, I also point to the conceptual treatment given to the subject in the processes of segmentation of Tourism, which refer to the individualism and pragmatism, endorsing that the segmentation is still based on too reductionist perspectives. In this effort to question the segment, the text is left open, mobilizing other inquires and propositions of new arrangements within the Tourism – Discourse Analysis interface.

Abstract:
We extend the class of M-tests for a unit root analyzed by Perron and Ng (1996) and Ng and Perron (1997) to the case where a change in the trend function is allowed to occur at an unknown time. These tests M(GLS) adopt the GLS detrending approach of Dufour and King (1991) and Elliott, Rothenberg and Stock (1996) (ERS). Following Perron (1989), we consider two models: one allowing for a change in slope and the other for both a change in intercept and slope. We derive the asymptotic distribution of the tests as well as that of the feasible point optimal tests PT(GLS) suggested by ERS. The asymptotic critical values of the tests aretabulated. Also, we compute the non-centrality parameter used for the local GLS detrending that permits the tests to have 50% asymptotic power at that value. We show that the M(GLS) and PT(GLS) tests have an asymptotic power function close to the power envelope. An extensive simulation study analyzes the size and power in finite samples under various methods to select the truncation lag for the autoregressive spectral density estimator. An empirical application is also provided.
Extendemos los estadísticos tipo M para una raíz unitaria analizados por Perron y Ng (1996) y Ng y Perron (2001) al caso donde se permite que el cambio en la función de tendencia ocurra en un punto desconocido. Estos estadísticos (MGLS) adoptan el enfoque GLS para eliminar la tendencia desarrollado por Elliott et al. (1996) (ERS) siguiendo los resultados de Dufour y King (1991). Siguiendo a Perron (1989), consideramos dos modelos: uno que permite un cambio en la pendiente y otro que permite tanto un cambio en el intercepto como en la pendiente. Derivamos las distribuciones asintóticas así como el estadístico óptimo factible en un punto de la hipótesis alternativa (PT GLS) sugerido por ERS. También computamos el parámetro de no centralidad utilizado por el enfoque GLS local a la unidad con el fin de eliminar la tendencia que permite que el estadístico PT GLS tenga 50% de potencia asintótica en ese valor. Asimismo, se han tabulado los valores críticos asintóticos de los estadísticos. Mostramos que los estadísticos MGLS y PT GLS tienen una función de potencia asintótica cercana a la envolvente de potencia. Un estudio de simulación analiza el tamaño y potencia en muestras finitas bajo varios métodos para seleccionar la truncación para estimar la densidad espectral autorregresiva. Finalmente, también se presenta una aplicación empírica.

Abstract:
By applying the image processing algorithms used in surveillance systems on video data obtained from a web camera, a motion detection application can be created and incorporated into web based games. The use of motion detection opens up a vast field of new possibilities in game design and this thesis will therefore cover how to create a motion detection JavaScript module which can be used in web based games. The performance and quality of the motion detection algorithms are important to consider when creating an application. What motion detection algorithms can be used to give a qualitativerepresentation without affecting the performance of a web based game will be analyzed and implemented in this thesis. Since the performance of the Central Processing Unit will not suffice, WebGL and the parallelism of the Graphical Processing Unit will be utilized to implement some of the most recognized image processing algorithms used in motion detection systems. The work resulted in an application where Gaussian blur and Frame Subtraction were used to detect and return areas where motion has been detected.

Le attuali produzioni di energia e materiali si basano soprattutto sullo sfruttamento di fonti fossili, ma la loro non rinnovabilità e la necessità di una maggiore sostenibilità ambientale han contribuito ad aumentare l’interesse verso risorse alternative. In questo contesto si collocano le bioraffinerie, che sono volte alla conversione di biomasse rinnovabili in prodotti commerciabili. Per implementare bioindustrie competitive, molte sfide devono essere superate, tra cui il miglioramento delle prestazioni dei microrganismi, cell factory, utilizzati nei processi produttivi. Tra tutte, il lievito Saccharomyces cerevisiae è la principale cell factory per la produzione di bioetanolo come biocarburante. L'obiettivo principale di questa tesi è stato di ingegnerizzare ceppi di S. cerevisiae per ottenere una cell factory con caratteristiche rilevanti a livello industriale, tra cui aumentate performances di crescita e un incremento della termotolleranza. In particolare, è stato studiato e quindi indotto un profondo rewiring cellulare, selezionando due proteine: la glutammato sintasi Glt1, un enzima del metabolismo centrale dell'azoto, CNM e la principale polyA binding protein Pab1, un fattore chiave nel metabolismo dell'mRNA. Gli effetti fisiologici della delezione e overespressione di GLT1 sono stati valutati coltivando S. cerevisiae con diverse fonti di azoto. I risultati mostrano che l'aggiunta di solfato d’ammonio, glutammato o glutammina, ma non la modulazione dell’espressione di GLT1, influenza notevolmente la crescita, il contenuto proteico, la vitalità e l'accumulo di specie reattive dell'ossigeno ROS. Questi dati evidenziano la plasticità del CNM di S. cerevisiae rispetto a variazioni ambientale e confermano la sua robustezza contro perturbazioni interne. Inoltre, sebbene la modulazione dell'espressione di GLT1 potrebbe non indurre una riprogrammare cellulare, la caratterizzazione fisiologica descritta puo essere utile per selezionare altri target pio promettenti per riprogrammazioni cellulari. L’induzione del rewiring cellulare è stato poi valutato selezionando Pab1 come target. A tal scopo, un ceppo recante la copia endogena di PAB1 è stato trasformato con una mutant library plasmidica di PAB1 e poi sottoposto a screening per isolare ceppi con maggior termotolleranza. I cloni isolati han mostrato, in drop test, una maggior crescita ad alte temperature e alte concentrazioni di etanolo. Tra tutte, la variante PAB1 S40.7 conferisce una maggiore termotolleranza tramite l’espressione dei soli primi 20 amminoacidi di Pab1. Tale fenotipo è stato anche confermato in bioreattore a 40°C. Una possibile ragione della maggior termotolleranza del ceppo S40.7 potrebbe essere collegata ad un minor accumulo di ROS rispetto al ceppo di controllo. Nel complesso, questi risultati han dimostrato che Pab1 è un promettente target per indurre fenotipi complessi con tratti migliorati, tra cui una maggiore termotolleranza. Infine, Pab1 è stata caratterizzata per determinare il ruolo dei suoi domini nel suo reclutamento all'interno di stress granules, SG, aggregati di mRNP non tradotti che si formano in condizioni stressanti. Questa caratterizzazione ha mostrato che l'associazione di Pab1 negli SG è principalmente dovuta agli RNA Recognition Motives RRM, il cui numero è importante per un efficiente reclutamento. Sebbene i domini P e C non partecipino direttamente all'associazione di Pab1 negli SG, la loro presenza rafforza o diminuisce, rispettivamente, la localizzazione di varianti sintetiche di Pab1 prive di almeno un RRM in questi aggregati. Questo lavoro suggerisce inoltre che i domini di Pab1 potrebbero essere sfruttati razionalmente per scopi di biologia sintetica. Nel complesso, i risultati di questa tesi evidenziano la difficoltà ma allo stesso tempo la capacità del rewiring cellulare nell’indurre fenotipi industrialmente rilevanti e, in particolare, Pab1 è emerso come target promettente per questo approccio.
Nowadays, for the production of energy and materials, our society mainly relies on fossil sources, but many concerns arise from their utilization, like greenhouse gases emission and non-renewability within the time of their consumption. Hence, biorefineries, which convert renewable biomasses into products and energy, could be a promising alternative. Despite some biorefineries are now at commercial scale, many challenges must be overcome to implement competitive bio-based industries, such as improving the performances of microorganisms, named cell factory, used during the production processes. Above all, the yeast Saccharomyces cerevisiae is the most prominent cell factory for producing bioethanol as biofuel. The main objective of this thesis was to engineer S. cerevisiae strains with biotechnological interesting traits, among which improved growth performances and increased thermotolerance. For this purpose, we investigated and applied a cellular rewiring, by selecting two targets: the glutamate synthase Glt1 that is an enzyme of the central nitrogen metabolism, CNM, and the main polyA binding protein Pab1, a master regulator of mRNA metabolism. Regarding GLT1, the physiological effects of its deletion and over-expression were assessed by growing yeasts in the presence of different nitrogen sources. Results showed that the supplementation of ammonium sulfate, glutamate or glutamine considerably affects growth, protein content, viability and reactive oxygen species, ROS, accumulation. Conversely, GLT1 modulation does not significantly influence these parameters. Overall, these data highlight the plasticity of the S. cerevisiae CNM in respect to the environment and confirm its robustness against internal perturbation. Moreover, even though the sole modulation of GLT1 expression might not reprogram the entire cell, the physiological characterization of this study might be helpful to guide the selection of other more promising candidate for the application of the rewiring approaches. Then, the induction of cellular reprogramming was assessed selecting Pab1. To this purpose, a strain carrying the unaltered PAB1 chromosomal allele was transformed with a PAB1 plasmid mutant library and then screened for isolating strains with high thermotolerance. The isolated clones showed growth improvement at both high temperatures and ethanol concentration by drop tests. Among all, the PAB1 S40.7 variant was further characterized because, strikingly, it dominantly confers higher thermotolerance by expressing just the first 20 amino acidic residues of Pab1. This improved phenotype was also confirmed in bioreactor at 40°C. Remarkably, the S40.7 strain accumulates less ROS compared to the control strain, thus possibly explaining its increased thermotolerance. Overall, these results demonstrated that Pab1 is a powerful candidate to evoke complex phenotypes with improved traits, among which higher thermotolerance. Finally, Pab1 was characterized to uncover the role of its domains in the recruitment of the protein within stress granules, aggregates of untranslated mRNPs that form during stressful conditions. This characterization shows that Pab1 association into these aggregates relies mainly on RNA recognition motives, RRM, whose number is important for an efficient recruitment. Although the P and C domains do not directly participate in Pab1 association to stress granules, their presence strengthens or decreases, respectively, the distribution of synthetic Pab1 variants lacking at least one RRM into these aggregates. As additional outcome, this part of the work is suggesting that Pab1 domains might be rationally exploited for synthetic biology purposes. Overall, the results of this thesis highlight and confirm the difficulty but at the same time the power of cellular rewiring in evoking industrially relevant phenotype. Pab1 is undoubtedly emerging as a pivotal element that deserves more attention for future strain design and tailoring.

Resumo: Gangliosidose GM1 é uma doença autossômica recessiva rara, classificada em três formas clínicas de acordo com a idade de apresentação dos sintomas e a gravidade, provocada pela deficiência da enzima lisossômica ?-galactosidase que leva ao acúmulo, principalmente, do gangliosídeo GM1. A forma juvenil geralmente apresenta início entre sete meses e três anos de idade, com progressão lenta dos sinais neurológicos, dimorfismos menos graves que na forma infantil e deformidades ósseas. A forma crônica é caracterizada por apresentações clínicas mais leves e sintomas extrapiramidais. O gene codificador da enzima é o GLB1, no qual mais de 130 mutações foram descritas. No presente estudo foi realizada a caracterização molecular de 10 indivíduos de nove famílias não relacionadas diagnosticados com gangliosidose GM1, nas formas juvenil e crônica. Todas as famílias são originárias do interior do estado de São Paulo ou do sul do estado de Minas Gerais. Para a análise realizada foi possível identificar a mutação anteriormente descrita p.T500A, em sete das nove famílias estudadas, a inserção c.1717- 1722insG e a mutação p.R59H foram encontradas em duas famílias (a última segregou juntamente com o polimorfismo descrito IVS12+8T>C). As demais mutações descritas (p.F107L, p.L173P, p.R201H, p.G311R) foram encontradas em uma família cada. Uma alteração neutra (p.P152P) e duas mutações (p.I354S e p.T384S) são inéditas. Foi possível identificar a ocorrência de uma mutação de novo em uma família. Todas as mutações foram encontradas em heterozigose
Abstract: GM1 gangliosidosis is a rare autosomal recessive, classified in three clinical types according to age of onset and severity. The disease is caused by the deficiency of lysosomal enzyme ?-galactosidase that leads to the accumulation of GM1 ganglioside. The juvenile form usually shows an onset between seven months and three years of age, with slowly progressive neurological signs, less severe dysmorphisms than the infantile form and skeletal changes. The adult form is specified by a milder clinical manifestations and extrapyramidal signs. The lysossomal enzyme is coded by the GLB1 gene which more than 130 mutations have been decribed. In the present study it was genotyped 10 individuals of nine unrelated families originated from the States of São Paulo and Minas Gerais diagnosed with the juvenile and chronic forms of the disease. It was possible to find the previously described mutations p.T500A in seven of the nine families, c.1717-1722insG and p.R59H in two alleles (the latter also segregating with IVS12+8T>C), and p.F107L, p.L173P, p.R201H, and p.G311R in one familie each. One neutral alteration (p.P152P) and two mutations (p.I354S and p.T384S) are described for the first time. The occurrence of a de novo mutation was seen in one family. Abstract:
This thesis discusses implementation of LoD terrain visualization algorithm Seamless Patches for GPU-Based Terrain Rendering as extension for Coin3D library. It presents procedures which this algorithm uses for displaying large terrain datasets. Entire terrain is composed of patches that are stored in patch hierarchy. Patch hierarchy is traversed during runtime to generate active patches based on observer's position. Each patch consists of predefined tiles and connection strips so it doesn't need to store any geometry. During render of tiles and strips, displacement shader is applied. This thesis also evaluates results achieved in sample application and suggests some modifications to further increase algorithm performance.

Este trabalho procura mapear as condições históricas que viabilizaram a invenção da literatura gls, estabelecendo uma relação entre política e texto, por entender que a produção da Edições GLS está inserida no contexto relacional entre o movimento político de afirmação homossexual e a produção de novos códigos culturais de representação identitária. Para tanto, se faz necessário cartografar a irrupção, no presente, de uma política positiva da visibilidade - como nomeada pelos grupos de ativismo gay e divulgada pela Edições GLS - , que postula a publicização, a confissão e a proliferação de discursos para a formulação de uma identidade cultural, demarcada pela orientação sexual, ao problematizar as estratégias políticas utilizadas pelos militantes gays e as imbricações com a promoção de uma literatura confessional, que solicita, através de práticas editoriais e literárias, o direito de ser/estar na história e o direito de produzir suas próprias referências culturais. Nesse intuito, desenvolvemos um campo de reflexão epistemológica do ofício historiográfico relativo à historicidade do tempo presente, problematizando como a história da leitura e a emergência de novas culturas do escrito estão implicadas num debate contemporâneo sobre a promoção de novos modos de leitura e de leitores, assim como na invenção da literatura gls

La grotte GLD1 dans l’Adrar Gueldaman (Babors d’Akbou/Algérie) a bénéficié d’un programme de recherche pluridisciplinaire du CNRPAH depuis 2010. Une séquence néolithique couvrant la période VIII-Ve millénaires BP a été mise au jour et fouillée (Secteurs 2 et 3). La reconstitution du cadre chronostratigraphique climatique, économique et culturel des occupations a permis d’individualiser 4 unités archéologiques chronologiquement calées par 28 dates 14C (cal BP) : UA1 [7200-6500], UA2 [6500-530], UA3 [5000-4600], UA4 [4500-4200]. Une reconstitution paléoclimatique est proposée à travers des analyses isotopiques (d13C et d18O) réalisées sur deux stalagmites STM2 et STM4 de GLD1 dont la croissance (dates U/Th), couvre les trois dernières unités archéologiques. Une corrélation entre changements climatiques et occupations du site est mise en évidence. L’abandon de la grotte coïncide avec la crise d’aridité de 4200 BP enregistrée sur le pourtour méditerranéen. La faune domestique est représentée par des chèvres, des moutons (UA1) et des bovins (UA2). Les systèmes d'élevage des chèvres et moutons correspondent aux modèles de gestion des troupeaux des ovicaprinés de sites néolithiques méditerranéens du Sud de la France et de l’Italie ainsi que du Proche-Orient. Les résultats préliminaires paléobotaniques (étude sur 10% du corpus) indique l’a présence de blé et de l'orge en UA4. La céramique à fond conique est décorée par incision (UA1) et impression sans recours au cardium. Les résidus organiques identifiés concernent les matières grasses carnées et laitières ainsi que la cire d’abeille (miel). Le mobilier lithique est typique constitué d’outils du fonds commun épipaléolithique et de nouveaux apports (armatures de flèches, racloirs et scies). L’industrie osseuse est exceptionnellement riche, diversifiée et bien conservée. Les documents symboliques sont en ivoire d’éléphant, derme ossifié de tortue, test d’œuf d’autruche, coquillage marin et ocre
A multidisciplinary project at GLD1 cave in the Adrar Gueldaman ridge (Babors near Akbou/North Algeria) was initiated by CNRPAH in 2010. A Neolithic sequence covering the VIII-V millennia BP has been revealed (Sectors S2 and S3). The chronostratigraphic, climatic, economic and cultural contexts have been studied, leading to the definition of four archaeological units chronologically delimited using 28 14C dates on charcoal (cal BP): UA1 [7200-6500], UA2 [6500-530], UA3 [5000-4600], UA4 [4500-4200]. The paleoclimate for periods UA2 to UA4 was reconstructed using isotopic analysis (d13C and d18O) of two stalagmites (STM2 and STM4), demonstrating a correlation between climatic changes and the occupation of the cave. A prolonged drought at ca. 4400–3800 cal BP is supposed to be the responsible of cave abandonment shortly after ca. 4403 cal BP. La grotte GLD1 dans l’Adrar Gueldaman (Babors d’Akbou/Algérie) a bénéficié d’un programme de recherche pluridisciplinaire du CNRPAH depuis 2010. Une séquence néolithique couvrant la période VIII-Ve millénaires BP a été mise au jour et fouillée (Secteurs 2 et 3). La reconstitution du cadre chronostratigraphique climatique, économique et culturel des occupations a permis d’individualiser 4 unités archéologiques chronologiquement calées par 28 dates 14C (cal BP) : UA1 [7200-6500], UA2 [6500-530], UA3 [5000-4600], UA4 [4500-4200]. Une reconstitution paléoclimatique est proposée à travers des analyses isotopiques (d13C et d18O) réalisées sur deux stalagmites STM2 et STM4 de GLD1 dont la croissance (dates U/Th), couvre les trois dernières unités archéologiques. Une corrélation entre changements climatiques et occupations du site est mise en évidence. L’abandon de la grotte coïncide avec la crise d’aridité de 4200 BP enregistrée sur le pourtour méditerranéen. La faune domestique est représentée par des chèvres, des moutons (UA1) et des bovins (UA2). Les systèmes d'élevage des chèvres et moutons correspondent aux modèles de gestion des troupeaux des ovicaprinés de sites néolithiques méditerranéens du Sud de la France et de l’Italie ainsi que du Proche-Orient. Les résultats préliminaires paléobotaniques (étude sur 10% du corpus) indique l’a présence de blé et de l'orge en UA4. La céramique à fond conique est décorée par incision (UA1) et impression sans recours au cardium. Les résidus organiques identifiés concernent les matières grasses carnées et laitières ainsi que la cire d’abeille (miel). Le mobilier lithique est typique constitué d’outils du fonds commun épipaléolithique et de nouveaux apports (armatures de flèches, racloirs et scies). L’industrie osseuse est exceptionnellement riche, diversifiée et bien conservée. A multidisciplinary project at GLD1 cave in the Adrar Gueldaman ridge (Babors near Akbou/North Algeria) was initiated by CNRPAH in 2010. A Neolithic sequence covering the VIII-V millennia BP has been revealed (Sectors S2 and S3). The chronostratigraphic, climatic, economic and cultural contexts have been studied, leading to the definition of four archaeological units chronologically delimited using 28 14C dates on charcoal (cal BP): UA1 [7200-6500], UA2 [6500-530], UA3 [5000-4600], UA4 [4500-4200]. The paleoclimate for periods UA2 to UA4 was reconstructed using isotopic analysis (d13C and d18O) of two stalagmites (STM2 and STM4), demonstrating a correlation between climatic changes and the occupation of the cave. A prolonged drought at ca. 4400–3800 cal BP is supposed to be the responsible of cave abandonment shortly after ca. 4403 cal BP. O presente trabalho tem o objetivo geral de analisar os sentidos produzidos acerca dos acordos e tensões entre o mercado e sociabilidade voltados a lésbicas, gays, bissexuais, travestis e transexuais (LGBT), tendo como referência as trajetórias de conformação recente dos espaços de consumo, lazer e sociabilidade, que se definem como GLS (Gays, Lésbicas e Simpatizantes), localizados no centro da cidade do Recife/Pernambuco. Para tanto, buscamos, a partir de diálogos com interlocutoras privilegiadas: 1) construir as narrativas das histórias de conformação dos atuais espaços de consumo, lazer e sociabilidade GLS do Recife; 2) problematizar os acordos e tensões entre mercado, sociabilidade e agenciamentos políticos voltadas à população LGBT. Esta pesquisa partiu de uma abordagem psicossocial, na interface com estudos em gênero e sexualidade, à luz da perspectiva crítica feminista. Definimos como estratégia teórico-metodológica para produção de informações, a entrevista semiestruturada, compreendendo-a como práticas discursivas, em consonância com a perspectiva construcionista de produção do conhecimento. Portanto, realizamos as entrevistas com as interlocutoras privilegiadas dos espaços de sociabilidade estudados, definidas a partir dos critérios de tempo, convívio e responsabilidade em relação a esses lugares. Como resultados, construímos as narrativas de como surgiram os espaços de consumo, lazer e sociabilidade destinados à população LGBT que atualmente compõem o circuito GLS da cidade do Recife (Boate MKB, Clube Metrópole, Nosso Jeito Music, Castelo Marron, Confraria Ursos de Pernambuco, Conchittas Bar e Mix Pub). Em nossas análises, também abordamos os acordos e tensões entre mercado e sociabilidade, a partir de três eixos temáticos e seus respectivos tópicos analíticos, com a finalidade de compreender os sentidos coproduzidos sobre essas temáticas: Lazer e Sociabilidade (Motivação, Público e Práticas Sexuais), Mercado (Resistências, Localização, Concorrência, Aspectos Positivos e Negativos, e Legislação) e Agenciamentos políticos (Movimento LGBT e Parada da Diversidade).
The general objective of this study is to analyze the meanings produced about the agreements and disagreements between the market and sociability focused on at lesbians, gays, bisexuals and transgenders (LGBT), taking as reference the recent forming trajectories of consumption, leisure and sociability spaces, wich are defined by the term GLS (gays, lesbian and sympathizers), located in Recife's downtown, Pernambuco. To this end, we seek to, in a dialogue with privileged interlocutors: 1) build the narrative of forming trajectories of current of consumption, leisure and sociability spaces of the GLS market; 2) discuss the agreements and disagreements between market, sociability and political agencies focused on LGBT people. This research is based on a psychosocial approach in dialogue with gender and sexuality studies in the light of critical feminist perspective. The semi-structured intervier was defined as the theoretical and methodological strategy for the production of information, understanding it as a discursive practice, in line with the constructionist perspective of knowledge production. Therefore, we conducted interviews with privileged interlocutors from sociability spaces where the study take place, chosen by the criterion of time, familiarity and responsability concerning these places. As a result, we construct narratives about how arose the consumption, leisure and sociability spaces for the LGBT people that currently make up the GLS market of Recife (Boate MKB, Clube Metrópole, Nosso Jeito Music, Castelo Marron, Confraria Ursos de Pernambuco, Conchittas Bar and Mix Pub). In our analysis, in order to understand the co-produced meanings about these issues, we discuss the agreements and disagreements between market and sociability based on three main themes and their analytical topics: Leisure and Sociability (Motivation, Public and Sexual Practices), Market (Resistences, Location, Competition, Positives and negatives aspects, and law) and Political Agencies (LGBT Movement, Diversity Pride Parade).

Este trabalho se propõe a pensar um espaço de sociabilidade homossexual em Salvador, o Beco dos Artistas, refletindo sobre como as pessoas vivem esse espaço, o significado e sentido que lhe atribuem, e sobre as práticas, relações e comportamentos que se dão espaço à dentro. Proponho-me a pensar, também, os motivos da existência de tal espaço e quais as consequências de sua existência tanto para os frequentadores, quanto para o tecido social de forma mais ampla. Para isso, realizo uma etnografia sobre o espaço, descrevo suas características físicas e sociais, sempre com o intuito de utilizar essa descrição como material para reflexões de cunho mais crítico e teórico, tentando sempre perceber quais as questões sociais que aquele espaço particular suscita. Com essa perspectiva, a discussão sobre espaço e sexualidade ganhou um lugar central no meu trabalho. This paper proposes to consider a space of homosexual sociability in Salvador, Beco dos Artistas, reflecting about how people live in this space, the meaning and sense which they attribute to it, and their practices, relations and attitudes. I also propose a reflection on the reasons for the existence of such place and the consequences of its existence, for both the visitors and the wider society. In view of those questions, I do an ethnography of the space, describe its physical and social characteristics, in order to use these data as a material for critical and theoretical reflections, trying to realize what are the social issues raised by that particular space. Therefore, the discussion on space and sexuality takes a central place in this work.

Abstract: Glutamate is the main excitatory neurotransmitter of the mammalian nervous system and is involved in neuronal plasticity, memory and learning. Emerging evidences suggest that glutamate is also present in peripheral tissues, where it plays a role in both cellular homeostasis and in autocrine/paracrine communication as an extracellular signalling molecule (Hedigerand Welbourne, 1999; Nedergaard et al., 2002). Particular interesting is the role played by this amino acid molecule in the endocrine pancreas, the pancreas portion responsible for hormones secretion and glucose homeostasis. In this context, glutamate works as intracellular signalling molecules in beta cells to couple changes in metabolic activity with insulin secretion. More recently, an additional role of glutamate as paracrine and autocrine signal involved in the control of hormone release and islet cells viability has been proposed. The glutamate signalling machinery and the precise mechanisms by which glutamate controls islet function are poorly understood. The extracellular glutamate concentration is tightly controlled by high affinity glutamate transporters, whose expression and modulation in the peripheral tissues have been poorly investigated. The main isoform expressed in the islet is the Na+-dependent high affinity glutamate transporter GLT1/EAAT2 (glutamate transporter 1/ excitatory amino acid transporter 2) which is prevalently expressed on the insulin-secreting cell plasma membrane (β-pancreatic cells). We previously demonstrated that an excess of extracellular glutamate affects the islet viability, and induces selectively β-cell death. The GLT1 transporter, by transporting glutamate within the cells, exerts a key role in the control of extracellular glutamate concentration and in the preservation of the β cell excito-toxicity induced by glutamate (Di Cairano et al., 2011). Given the key role of GLT1 in islet physiology, in this study, we focus on GLT1 and we examine the molecular mechanisms which control its expression and function, using murine clonal β-cell lines and isolated human islets of Langerhans as experimental models. In chapter I we specifically investigate the intracellular pathways responsible for the acute regulation of the GLT1 localization and function at the β-cell plasma membrane, where it physiologically works. In chapter II we analyse the modulation of GLT1 expression and activity in condition of chronic hyperglycaemia (a condition typical of diabetes mellitus) and we investigate the consequences of such as modulation on β-cell viability. Given the anatomical, morphological and structural differences between human and rodent islets, we directly used human isolated islets, the most appropriate model to study the islets pathophysiology. The localization of GLT1 was also assessed on pancreas sections from control and diabetic subjects. In chapter III we highlight the possible pathological role of GLT1 in type 1 of diabetes mellitus (T1DM). We investigate the potential intervention of this transporter in the development of T1DM autoimmunity, as a new membrane antigen of diabetes mellitus. Chapter IV reports two side projects of these three-years of doctoral activity. The first project is related to the characterization of islet remodelling during diabetes, in baboons. This is collaboration with Prof. Folli Franco of Texas University and my role in the project was to characterize the apoptotic cells in islets of Langerhans. The second project is a technical project; the lab is interested in developing imaging techniques and protocols for studying vesicle dynamics. Using the genetically encoded synapto-pHluorin and Total Internal Reflection Fluorescence Microscopy we developed a new macro for the analysis of endocytic and exocytic events at the plasma membrane. This macro could be very useful for future studies related to the effect of glutamate on hormone secretion.

GLK1 and GLK2 transcription factors have been suggested to be involved in the regulation of chloroplast development, organic nitrogen signaling, disease resistance and circadian rhythmicity (Waters et al. 2009; Gutiérrez et al. 2008; Savitch et al. 2007; Sprott et al. 2010). This implies that multiple factors may play roles in regulation of GLK genes. In the present study, transcriptional regulation of GLK1 and GLK2 in Arabidopsis by various endogenous and environmental stimuli was investigated with the objective of elucidating the primary signalling pathway affecting expression of these two genes. Collectively, results of GLK1 and GLK2 expression in response to the experimental treatments of Arabidopsis point to the regulation of the two genes by changes in photosynthetic metabolism and reactive oxygen species (ROS) levels, and by organic nitrogen signalling. Changes in ROS levels and organic nitrogen signalling may also affect the two genes indirectly by interfering with or altering photosynthetic metabolism.

Abstract:

Fluid simulation is today a hot topic in computer graphics. New highly optimized algorithms have allowed complex systems to be simulated in high speed. This master thesis describes how the graphics processing unit, found in most computer workstations, can be used to optimize the rendering of volumetric fluids. The main aim of the work has been to develop a software that is capable of rendering fluids in high quality and with high performance using OpenGL. The software was developed at Filmgate, a digital effects company in Göteborg, and much time was spent making the interface and the workflow easy to use for people familiar with Autodesk Maya. The project resulted in a standalone rendering application, together with a set of plugins to exchange data between Maya and our renderer.

Most of the goals have been reached when it comes to rendering features. The performance bottleneck turned out to be reading data from disc and this is an area suitable for future development of the software.

O trabalho procura compreender essas relações a partir da pesquisa de campo realizada com foco na Associação da Parada do Orgulho GLBT de São Paulo (APOGLBT). Além disso, há um esforço no sentido de entender as dinâmicas do mercado GLS e do movimento GLBT a partir de meados da década de 1990, especialmente em São Paulo. O enfoque da dissertação recai sobre processos de aproximação e diferenciação entre movimento e mercado, envolvendo também situações de conflito e colaboração entre ambos os atores sociais. Tal abordagem foi elaborada a partir da discussão teórica sobre processos de construção de identidades coletivas presentes na constituição de sujeitos políticos, bem como da perspectiva da antropologia a respeito de processos de consumo.
This thesis is a study of the GLBT (Gay, Lesbian, Bisexual and Transgender) movement and its relations with a segmented market that is focused on homosexuals or GLS (Gay, Lesbian and Supporter). This work departing from the conducted field research on the Associação da Parada do Orgulho GLBT de São Paulo (APOGLBT - São Paulo\'s Association for the GLBT Pride Parade) seeks to understand these relations. Besides this, an effort has been made to understand the dynamics of the GLS market and the GLBT movement, specially in São Paulo, since the mid 1990s. The focus of this thesis is grounded on an analysis of processes of rapprochement and differentiation between the movement and the market, involving situations of conflict and collaboration between both social actors. This approach has been elaborated from a theoretical discussion about processes of collective identity formation, present in the constitution of political subjects, as well as from an anthropological perspective on consumption processes.

Under de senaste åren har en ny trend inom spelindustrin uppstått då en stor del av spelen har gått tillbaka till de tvådimensionella miljöerna som var vanligare i tv-spelens barndom, detta tack vare att mobiltelefoner och deras spel har tagit allt större plats på marknaden och de är ofta tvådimensionella på grund av sin begränsade hårdvara. De tvådimensionella spelen har enligt tradition inte haft någon dynamisk ljussättning, utan de har bestått utav färdig grafik med förutbestämda intensiteter vilket gör att spelen kan se platta ut. Men inom andra områden av spelindustrin, framförallt tredimensionella spel, har utvecklingen av dynamiska ljussättningsmetoder kommit långt. Problemet med dessa metoder är att de kräver objektens normaler, vilka normalt inte finns tillgängligt i tvådimensionella bilder. Idag är mobiltelefonerna kraftfulla och klarar av mycket tyngre beräkningar än vad de gjorde för bara några år sedan. Därför undersökte vi möjligheten att implementera dynamisk ljussättning i ett tvådimensionellt spel på androidbaserade mobiltelefoner. För att genomföra detta krävdes också att vi undersökte metoder för att generera normaler för tvådimensionell grafik. För att ett fullständigt spel skulle kunna skapas så implementerades också saker som kollisionshantering, artificiell intelligens och procedurell generering av banor.
In recent years, a new trend in the gaming industry has emerged since a large part of the games have gone back to the two-dimensional environments that were more common in the early days of the video games history . This is due to mobile phones and their games have taken more and more space on the market and they are often two-dimensional because of the limited harware. The twodimensional games have traditionally never had any dynamic lighting , they have consisted of premade graphics with predetermined intensities , which means that the games may look flat. But in other areas within the gaming industry, especially three-dimensional games, the evolution of dynamic lighting techniques have come a long way. The problem with these methods is that they require the object's normals, which are not normally available in two-dimensional images. Today mobile phones are powerful and capable of much heavier computations than they did just a few years ago. Therefore, we examined the possibility of implementing dynamic lighting in a twodimensional games on Androidbased mobile phones. To accomplish this it required that we investigated methods to generate normals for two-dimensional graphics. In order to create a full game we also implemented features such as collision handling, artificial intelligence and procedural generation of maps.

Mes travaux de thèse ont porté sur ces deux aspects.La connaissance de l'implication de la voie de signalisation Hedgehog (Hh) dans le maintien du pool de CSC permet d'envisager de nouvelles stratégies thérapeutiques visant à les éliminer. La voie Hh joue un rôle important dans le développement embryonnaire. L'activation de cette voie a été mise en évidence dans de nombreux cancers dont les GBM. La cyclopamine est une substance alcaloïde inhibitrice de la voie Hh. L'inconvénient de l'utilisation de la cyclopamine pour une thérapie anticancéreuse est que cette voie est aussi active dans les cellules souches somatiques adultes. Pour cibler la tumeur, une prodrogue glucuronylée de la cyclopamine a été synthétisée. La partie glucuronide peut être hydrolysée par la β-glucuronidase, une enzyme présente dans la zone nécrotique des GBM, conduisant à la libération de la cyclopamine active de façon ciblée, dans le microenvironnement tumoral. Des tests de viabilité indiquent que cette prodrogue ne présente pas latoxicité de la cyclopamine envers les cellules C6 de GBM de rat, leur population de cellule souches (C6-GSC) ainsi que dans des coupes de cerveau de rat adulte. En revanche, l'hydrolyse par la β-glucuronidase de cette prodrogue permet de restaurer la toxicité de la cyclopamine sur les cellules de GBM. L'analyse par RT-qPCR de l'expression d'un gène cible de la voie Hh, le facteur de transcription GLI1, permet de vérifier l'inhibition spécifique de la voie Hh par la cyclopamine dans les cellules C6 et les C6-GSC. Ainsi, la prodrogue en libérant la cyclopamine dans l'environnement tumoral devrait permettre de limiter l'étendue des effets de la drogue sur les cellules saines du cerveau.La voie Hh joue un rôle dans la régulation de la migration et de l'invasion des cellules de GBM. Mon équipe d'accueil a montré que le vasoactive intestinal peptide (VIP) régule la migration et l'invasion de deux lignées cellulaires M059K et M059J. Dans ce travail de thèse, les effets du VIP et de son analogue le pituitary adenylate cyclase-activating peptide(PACAP) sur la migration et l'invasion des cellules C6 de rat et U87 humaines ont été étudiés. Les résultats obtenus indiquent que les agonistes VIP, PACAP-38 et l'antagoniste synthétique VIP10-28 des récepteurs de ces peptides contrôlent la migration et l'invasion dans les deux lignées de GBM. Le VIP et le PACAP-38 inhibent l'invasion ex vivodes cellules C6 dans des coupes de cerveau. De plus, le VIP10-28 augmente la migration et l'invasion de ces cellules de façon PKA-, Akt- et Hh-dépendante. L'étude des voies detransduction impliquées dans l'action des neuropeptides sur la migration et l'invasion des GBM a ensuite été approfondie. Les données obtenues indiquent que le VIP et le PACAP diminuent le taux de la forme active nucléaire de la protéine GLI1 dans les cellules C6 et U87 de façon PKA-dépendante. Inversement, le VIP10-28 augmente l'expression de GLI1 dans les cellules C6 de façon Akt-dépendante. Enfin, nous avons constaté que le PACAP diminue la phosphorylation d'Akt dans les cellules C6 etaugmente celle de la protéine PTEN, en accord avec le rôle de ces kinases dans la régulation de la migration cellulaire et de l'activité GLI1. L'ensemble de ces résultats indique que le système VIP-récepteurs régule la migration et l'invasion par des interactions entre les voies PKA, Hh et Akt/PTEN dans les cellules de GBM

Abstract:
Myocardial infarction (MI) persists as one of the leading causes of death worldwide. Often patients whom survive the initial injury will develop heart failure characterized by a dilated and functionally incompetent heart. Heart failure (HF) carries a worse prognosis than most cancers, and the only curative therapy to date is heart transplantation. A better understanding of the repair and remodeling processes post-MI, and the development of novel therapies are required to combat this burgeoning medical challenge. This thesis research sought to identify a novel mediator of the impaired cardiac remodeling that often occurs post-MI, and to characterize a biomaterial hydrogel therapy as a novel treatment. We investigated the role of methylglyoxal (MG), an important precursor to advanced glycation end-products (AGE), using a transgenic mouse model to over-express glyoxalase 1 (GLO1). GLO1 is the primary enzyme involved in metabolizing MG and preventing its accumulation. The role for MG and AGEs in MI and HF had been alluded to in the literature, yet no study to date has causally linked them with the loss of function and impaired remodeling of the post-MI heart. We also assessed an injectable hydrogel for the treatment of MI using a mouse model and evaluated the impact of delivery timing on its therapeutic efficacy. In this thesis, we confirmed that MG derived AGEs accumulate post-MI (Chapter 3.1). We show that preventing their accumulation, through GLO1 over-expression, mitigates the loss of function post-MI and positively influences remodeling through reducing final infarct sizes and end-systolic volumes. We demonstrate that this may possibly occur through improving the bone marrow response post-MI by restoring ECM-cell signaling. In Chapter 3.2, we present results of a study assessing the efficacy of a collagen based injectable hydrogel for the treatment of MI, and assessing the role that timing plays into the benefits associated with this therapy by studying 3 separate timepoints including 3 hours, 7 days and 14 days post-MI. We found that the injectable hydrogel preserved cardiac function and reduced infarct sizes. It also positively interacted with the host repair response by reducing chronic inflammation and cell death. The benefits of the therapy depended on when the material was delivered, and we found that the earliest timepoint (3 hours post-MI) proved most beneficial. In Chapter 3.3, we combined the knowledge gained from Chapters 3.1 and 3.2 and functionalized our hydrogel with a flavonoid, Fisetin, that has been shown to scavenge MG and increase the activity of GLO1. We show that this novel functionalized material may be able to restore some function in MI, particularly in settings of low baseline cardiac function. Taken together, the results of this thesis demonstrate that MG accumulates as a result of the ischemia and contributes to the impaired repair resolution and remodeling processes post-MI. This identifies MG as a possible novel target for the treatment of MI. Indeed, we also confirm the role that delivery timing plays into injectable hydrogels post-MI, and present promising results for a functionalized material design to intervene on MG production.

Abstract:
Glutamate is the main excitatory neurotransmitter of the mammalian nervous system and is involved in neuronal plasticity, memory and learning. Emerging evidences suggest that glutamate is also present in peripheral tissues, where it plays a role in both cellular homeostasis and in autocrine/paracrine communication as an extracellular signalling molecule [Hediger and Welbourne, 1999; Nedergaard et al., 2002]. The extracellular glutamate concentration is tightly controlled by high affinity glutamate transporters, whose expression and modulation in the peripheral tissues have been poorly investigated. In this work we analyse the high affinity glutamate transporters EAAC1 and GLT1 in epithelia and endocrine pancreas, respectively. EAAC1 was cloned from rabbit intestine [Kanai and Hediger, 1992] and its expression and function have been well characterised in absorptive epithelia, such as intestine and kidney, where it represents the major transporter for the dicarboxylic amino acids [Peghini et al., 1997]. Less is known about the molecular mechanisms that regulate its surface expression and activity. During the past few years, it has become clear that the activity of these transporters can be rapidly regulated by redistribution of proteins to and from the plasma membrane: a process that can be controlled by dynamic protein-protein interactions. Therefore, the research presented in the chapter II focuses on the molecular mechanisms which regulate EAAC1 trafficking in epithelial cells. EAAC1 has a conserved sequence present in the C-terminal domain of EAAC1, that mediates interactions with class I PDZ proteins. In the past years, we demonstrated that the PDZ-target sequence and PDZ proteins are responsible for the retention and stability of EAAC1 at the plasma membrane [D’Amico et al., 2010]. The aim of the present work is to verify whether this PDZ-target sequence is also important for the transporter’s biosynthetic delivery. Our data indicate that PDZ interactions occur early in the biosynthetic pathways and are involved in the ER-to-Golgi trafficking, as well as in Post-Golgi trafficking of EAAC1. Removal of the PDZ motif delays rather than prevents the ER export and the plasma membrane delivery of the transporter, thus indicating that PDZ interactions facilitate the ER-Golgi trafficking. Possibly, PDZ-interactions favour the transporters homo-oligomerization, a process required for the efficient ER export of EAAC1. Alternatively, PDZ domain-proteins may couple EAAC1 with protein complexes required for the efficient fusion of carrier vesicles to the appropriate target membranes. Further studies will be needed to identify the PDZ protein/s involved in the EAAC1 biosynthetic delivery. On the other hand, the presence of glutamate as an intercellular signal mediator in endocrine pancreas is well established [Moriyama and Hayashi, 2003]. In the Central Nervous System (CNS), glutamate may cause cell death by excitotoxicity, that is physiologically prevented by glutamate clearance systems [Choi et al., 1988]. The effect of glutamate on islet viability, the expression of glutamate transporters and their physiological roles in the endocrine pancreas are still unclear. Thus, in the III chapter we examine the effects of glutamate and the function of sodium dependent glutamate transporters in clonal beta-cells and in human isolated islets of Langerhans. We demonstrate that exposure to elevated glutamate concentrations induces a significant cytotoxic effect in pancreatic beta-cells, due to the prolonged activation of ionotropic glutamate receptors. We provide evidence that the key regulator of the extracellular glutamate clearance in the islet is the glial glutamate transporter 1 (GLT1/EAAT2). GLT1 is the only high affinity glutamate transporter expressed in the islets and localizes to the beta-cell plasma membrane. Finally, as diabetes is characterized by selective beta-cell death, and our data indicate that GLT1 controls beta-cell survival, we investigate the expression of GLT1 in type 2 diabetes mellitus (T2DM) patients. We show an altered GLT1 localisation in pancreases from T2DM patients, suggesting a decreased glutamate clearance ability in these subjects. In chapter IV, is reported the experience at University of Texas Health Science Center at San Antonio (UTHSCSA), USA. The aim of this project is to find interactors of IAPP, a protein involved in diabetes, by means of Yeast Two Hybrid Screening, a technique which allows the identification of direct protein-protein interactions. In prospect, this technique will be useful to find proteins that are associated with glutamate transporters and that potentially regulate their expression and function.

Abstract:
Ekokardiografi är en mycket användbar teknik som bland annat kan bedöma vänsterkammarensfunktion hos hjärtat med hjälp av ejektionsfraktion (EF) och global longitudinell strain (GLS). EF är idag den metoden som används mest vid bedömning av vänsterkammarfunktionen och beräknas enligt Simpsons biplanmetod. GLS är en ny metod som mäter myokardiets deformation och har i tidigare studier visat sig vara en känslig metod och kan upptäcka förändringar i myokardiet tidigare än EF. Vid behandling av bröstcancer kan hjärtat påverkas toxiskt och därmed genomgår denna patientgrupp regelbundna kontroller med ekokardiografi.   Syftet med denna studie är att jämföra två ekokardiografiska metoder hos 20 stycken kvinnliga bröstcancerpatienter. Jämförelser har gjorts mellan EF beräknat med Simpsons biplanmetod (EFbi) och GLS samt mellan EFbi och EF beräknat från GLS (EFGLS). Detta är en kvantitativ retrospektiv studie där data insamlats mellan oktober-december 2014 på avdelningen klinisk fysiologi vid Länssjukhuset Ryhov i Jönköping. Den analysmetod som använts är McNemar´s test samt kappavärde. Resultatet av studien visar att EFbi och GLS gav en dålig överenstämmelse mellan metoderna och en rimlig överenstämmelse sågs mellan EFbi och EFGLS. Slutsatsen har dragits utifrån kappavärdet.
Echocardiography is a useful technique which can value the function of the left chamber of the heart by using ejection fraction (EF) and global longitudinal strain (GLS). Today EF is the most common method in assessments of the left chamber function and is calculated by Simpson´s biplanemethod. GLS is a new method that measures the myocardial deformation. It has been proved to be a sensitive method and can detect changes in the myocardium earlier than EF.   The purpose of this study is to compare two echocardiographic methods among 20 female breast cancer patients. Comparisons have been made between EF calculated with Simpson´s biplanemethod (EFbi) and GLS, and between EFbi and EF calculated by GLS (EFGLS). This is a quantitative retrospective study. The data used in this study was collected between October – December 2014 at the Department of Clinical Physiology at the Hospital Ryhov in Jönköping. The method used to analyse the results was McNemar´s test and Kappa value. The results of the study showed that EFbi and GLS gave a modest correspondence between the methods. A reasonable correspondence were observed between EFbi and EFGLS. The conclusion has been drawn from the kappa value.

Le présent document se tourne vers l'analyse des processus de construction et d'appropriation de GLS discothèques espace dans le centre de Manaus. En ce sens, soulève des questions sur les différentes représentations des différents acteurs sociaux sur ces domaines et en même temps, réfléchit sur les différentes significations qui passent par une GLS discothèque. La coupure était le territoire et de représentation, où nous pensons que la relation entre l'espace, la stigmatisation et la résistance symbolique, notant que pour la dimension politique de la sociabilité.
O presente trabalho volta-se para a análise sobre os processos de construção e apropriação do espaço em boates GLS do centro de Manaus. Neste sentido, busca problematizar acerca das diferentes representações que os vários agentes sociais possuem sobre estes espaços e ao mesmo tempo busca refletir sobre os diferentes significados que perpassam uma boate GLS. O recorte utilizado foi o de território e representação, onde buscamos pensar a relação entre espaço, estigmatização e resistência simbólica, destacando para isso a dimensão política e a de sociabilidade.

Dentre as alterações craniofaciais destacam-se aquelas que ocorrem em função de alterações no desenvolvimento da linha média craniofacial. Alguns estudos genéticos mostram o envolvimento de vários genes durante os diferentes processos de desenvolvimento craniofacial. O gene Sonic Hedgehog (SHH) é expresso durante o desenvolvimento normal dos tecidos da linha média facial. Alguns estudos relatam a atividade da via sinalizadora do gene SHH na formação craniofacial, especialmente sua influência sobre os defeitos de linha média. Os genes mais conhecidos relacionados à rede sinalizadora do SHH compreendem, além do SHH, os genes ZIC2, TGIF, PTCH1, GLI2, FAST1, TDGIF, SUFU e o DHCRT. O objetivo desse trabalho foi realizar a análise de mutações nos genes GLI2 e PTCH1, através do sequenciamento direto, em 110 indivíduos com anomalias craniofaciais de linha média, olho e face, previamente analisados para os genes SHH, TGIF e SIX3. Os resultados obtidos pelo sequenciamento do gene GLI2 mostraram a presença de quatorze variações em regiões codificadoras, em 14 indivíduos brasileiros sem parentesco: 1 deleção, 1 inserção, 9 variantes não sinônimas e 3 variantes sinônimas. Nenhuma dessas variações foram encontradas nos 96 controles brasileiros normais. Seis dos quatorze indivíduos apresentavam manifestações fenotípicas que variavam de fissura de lábio/palato isolada com polidactilia, anomalia de arco branquial a Holoprosencefalia (HPE) semi-lobar. Some genetics studies show the involvement of several genes during different processes of craniofacial development. The Sonic Hedgehog gene (SHH) is expressed during the normal development of the midline facial tissues. Some of the studies reveals the activity of the signaling pathway of the SHH in the craniofacial formation, specially on its influence on the defects in the midline. The most known genes related to the SHH signaling pathway include, ZIC2, TGIF, PTCH1, GLI2, FAST1, TDGIF, SUFU and DHCRT, besides SHH. The main purpose of this work was to analyze the mutations in the GLI2 and PTCH1 genes, through direct sequencing, in 110 individuals with midline craniofacial anomalies, previously screened for SHH, TGIF and SIX3 genes. The results obtained by direct sequencing of GLI2 gene show the presence of 14 variations in coding region, in 14 unrelated Brazilian individuals: 1 deletion, 1 insertion, 9 no synonymous variants and 3 synonymous variant. None of these variations were found in the 96 Brazilian control individuals. Six of the fourteen individuals had phenotypic manifestations that varied from isolated lip/palate cleft with polydactyly, branchial arch anomalies to semi-lobar Holoprosencephaly (HPE). New variants were found in this gene in patients with temporomandibular joint commitment, a new clinical finding noted in GLI2 mutation. The analysis of PTCH1 gene shows the presence of seven new variants in 14 unrelated Brazilian individuals: 4 in intragenic regions and 3 in splicing regions. New variants were found in this gene in patients with temporomandibular joint commitment, a new clinical finding noted in GLI2 mutation. Abstract:
In this work we study the curvature visualization problem on surfaces implicitly defined by functions f: [0,1]³ → [0,1], using the ray casting technique. As we usually know only sampled values of f, we study the tricubic interpolation method to compute second order derivatives accurately. This work's implementation was designed as modules to the framework for volume rendering and image processing named Voreen, that uses the processing capability of graphics cards to improve the rendering tasks.

Neste trabalho, estudaremos a visualização da curvatura de superfícies definidas implicitamente por funções do tipo f:[0,1]³ [0,1], usando a técnica de lançamento de raios (ray casting). Como em geral conhecemos apenas valores amostrados de f, estudaremos um método de interpolação tricúbica, a fim de calcular as derivadas de segunda ordem precisamente. A implementação computacional deste trabalho foi desenvolvida na forma de módulos do framework de visualização e processamento de imagens Voreen, o qual se beneficia do poder de processamento das placas gráficas atuais para acelerar o processo de visualização.

Abstract:
This thesis tries to answer how to design a framework for image processing on the GPU, supporting the common environments OpenGL GLSL, OpenCL and CUDA. An generalized view of GPU image processing is presented. The framework is called gpuip and is implemented in C++ but also wrapped with Python-bindings. The framework is cross-platform and works for Windows, Mac OSX and Unix operating systems. The thesis also involves the work of creating two executable programs that uses the gpuip-framework. One of the programs has a graphical user interface and the other program is command-line only. Both programs are developed in Python. Performance tests are created to compare the GPU environments against a single core CPU implementation. All the GPU implementations in the gpuip-framework are significantly faster than the CPU when executing the presented test-cases. On average, the framework is two magnitudes faster than the single core CPU.
