Abstract:
Le diagnostic du gliome infiltrant du tronc cérébral (GITC) chez les enfants est l'un des plus éprouvants en oncologie pédiatrique. Malgré de nombreux essais cliniques explorant divers traitements, le pronostic reste sombre, la plupart des patients succombant entre 9 et 11 mois après le diagnostic. Les mutations génétiques clé associées au GITC incluent H3K27M, ACVR1 et TP53. Chaque mutation a des caractéristiques distinctes, poussant les médecins à suggérer des thérapies personnalisées, soulignant l'importance d'une détection précise des mutations pour guider le traitement. Situées dans la région cruciale du tronc cérébral, les tumeurs GITC présentent des risques significatifs liés à la biopsie en raison de potentiels dommages neurologiques. L'IRM est une méthode indispensable pour le diagnostic de ces tumeurs, évaluant leur extension et permettant de mesurer l'évolution de la maladie au cours de la thérapie. Une prédiction des mutations, combinée à l'identification des patients survivant plus de deux ans, pourrait améliorer la thérapie proposée à ces patients. Dans ce contexte, la radiomique transforme les images en vastes sources de données, extrayant des caractéristiques comme la forme et la texture pour aider à la prise de décision. L'objectif de cette thèse est de prédire les principales mutations génétiques et d'identifier les survivants à long terme, en mettant l'accent sur la normalisation des images et l'applicabilité des modèles radiomiques. Notre étude a utilisé une base de données rétrospective de l'Institut Gustave Roussy, comprenant les données IRM de 80 patients et leurs données cliniques respectives. Les données d'IRM ont mis en évidence des problèmes pour les études radiomiques, tels que l'inhomogénéité du champ de biais et l'effet "scanner". Pour répondre à ces défis, un pipeline de normalisation d'images IRM a été mis en place, et les caractéristiques radiomiques ont été harmonisées par la méthode ComBat. Pour faire face au problème de modalités manquantes dans l'ensemble de données, une stratégie multi-modèles a été employée, conduisant à 16 modèles distincts reposant sur diverses combinaisons de caractéristiques radiomiques et cliniques. Cette approche a ensuite été rationalisée en une méthode multimodale, réduisant le nombre de modèles à cinq, après une phase de sélection de caractéristiques indépendantes. Les résultats de l'approche multimodale se sont avérés être prometteurs. Cette stratégie multimodale a été essentielle pour identifier les patients survivant plus de deux ans et a été complétée par l'approche ICARE pour une analyse de survie détaillée
The diagnosis of diffuse intrinsic pontine glioma (DIPG) in children stands as one of the most harrowing within pediatric oncology. Despite numerous clinical trials exploring various treatments, the prognosis remains bleak, with most patients succumbing between 9 to 11 months post-diagnosis. Key gene mutations linked to DIPG include H3K27M, ACVR1, and TP53. Each mutation has distinct characteristics, leading physicians to suggest tailored therapies, underscoring the importance of accurate mutation detection in guiding treatment. Located in the crucial region of the brainstem, the pons, DIPG tumors pose significant biopsy risks due to potential neurological damage. Hence, MRI could become a primordial diagnostic tool for these tumors, assessing their spread and gauging therapy responses. Its use to predict accurate gene mutation, and identify long-term survivors, could enhance patient care significantly. Within this framework, radiomics transforms images into vast data sources, extracting features like shape and texture to aid decision-making. The objective of this thesis is to refine mutation prediction and pinpoint long-term survivors, emphasizing image normalization and the applicability of radiomic models. Our study utilized a retrospective database from Gustave Roussy Institute, encompassing 80 patients MRI data and their respective clinical data. These MRI images highlighted issues in radiomic studies, such as bias field inhomogeneity and the "scanner effect". To address these challenges, a dedicated MR image normalization pipeline was implemented, and radiomic features underwent ComBat harmonization. Given the dataset's missing modalities, a multi-model strategy was employed, leading to 16 distinct models based on various radiomic and clinical feature combinations. This approach was then streamlined into a multi-modal method, reducing the number of models to five. The results from the ensemble of these models proved to be the most promising. This multi-modal strategy incorporated a feature selection phase, pinpointing the most pertinent features. Additionally, this method was applied to identify long-term survivors and was complemented by the ICARE framework for a nuanced survival analysis output

Abstract:
This M.A. thesis presents a rhetorical analysis of the Indian philosophical and religious text, The Bhagavad Gita. Utilizing Kenneth Burke's Pentad of act, scene, agent, agency and purpose as a primary interpretive lens for uncovering universal human motivations, this rhetorical critique conceptualizes the idea of rhetorical agency as a model for action in the Gita's dialogical progression between Krishna and Arjuna. Rhetorical agency in the Gita differs from a traditional understanding of agency in that the former amalgamates competing yet co-existing pragmatic and consummatory agencies that Arjuna may utilize to act in the here and now but also relinquish the control on the fruits of his act, to ultimately transcend all human action by breaking the cycle of birth and death. In that sense, by virtue of rhetorical agency, the Gita may be considered in Burke's words Equipment for Living, because it provides a template for life across the universe.

Hoy en día existe un número importante de industrias que tienen establecimientos y/o sucursales de atención presencial a lo largo de todo Chile para sus clientes y usuarios. Muchos de ellos, poseen una alta demanda de servicios que se traduce en un gran volumen de usuarios que requieren ser atendidos. El mercado ante esta demanda, ha implementado sistemas de atención con el fin de dar un orden y secuencia. Muchos de los sistemas implementados son de carácter presencial y consideran un rollo de papel enumerado y un display que muestra el número actual de atención, esto obliga al usuario a encontrarse presencialmente esperando que sea su turno, sin poder utilizar su tiempo para lo que estime mejor. En base a este contexto, se ha detectado una oportunidad de negocio que da origen a e-GIT que nace cómo una solución al problema de optimización en el uso del tiempo que tienen las atareadas personas actualmente en Chile. Con el sistema de gestión inteligente de ticket de atención diseñado por e-GIT, el usuario puede solicitar su número virtualmente, puede mantenerse informado, ver el avance de la fila y en qué número de atención se encuentra, sin tener que estar en forma presencial, con lo cual puede manejar de mejor manera su valioso tiempo, adicionalmente el usuario se puede mantener en distintas filas, las cuales puede seguir desde una misma aplicación. Y la incorporación de alertas simplifica la experiencia del cliente, ya que es avisado del tiempo restante para su atención según la configuración que el usuario haya ingresado. Desde el punto de vista de la empresa se facilita el uso de sus espacios, reduce la cantidad de clientes que no aparecen, se reduce el tiempo de espera percibido por el cliente, lo que se traduce en una mejor percepción del servicio. La aplicación de gestión para la empresa provee gran cantidad de estadísticas fácil de obtener en función de la data generada, por ejemplo, tiempo de espera promedio, número de clientes atendidos, número de clientes que desertan, atenciones por empleado, sucursal con mayor número de atenciones, y mucho más.

The proposed thesis analyzes on the methods of analyzing Git-based software repositories, and focuses on mining GitHub based repositories. The introduction includes a summary of VCS history and usage, with many details that cover the interactions between users, Git and GitHub. The “related works” chapter presents the methods used by other researchers to mine Git data. Four different datasets are used in this thesis: the historical MSR14, and other three ad-hoc collected datasets. The datasets are analyzed and compared, both for a deeper understanding of the data and to validate the three “new” datasets with the already researched MSR14. Interesting findings are presented, and include considerations on the identification of a commit author. The main objective of the thesis is to present a graphic approach to analyze the interactions between different repositories through users. The results are shown to the researcher as an animated network graph through Gephi. Many examples are shown to investigate on the approach performances and capabilities, and are compared to expert knowledge on the repositories.

Portugal possui uma enorme e vulnerável faixa costeira. Estas zonas têm vindo a ser afectadas por actividade humanas exercendo uma forte pressão sobre o meio marinho. As zonas costeiras estão também associadas a actividades socioeconómicas significativas, tornando-se desta forma zonas bastante sensíveis. Surge assim, como pressão adicional a estas zonas, as alterações climáticas. Estas estão ligadas à subida do nível do mar, a alterações da frequência e/ou da intensidade das tempestades e furacões associados às consequentes cheias. Os impactes das alterações climáticas nas zonas costeiras serão assim provavelmente agravados pelo facto de nestas se localizarem as principais áreas metropolitanas do país. Assim o potencial para afectar um número elevado de pessoas é, nesta faixa, particularmente alto, tornando-se também um risco acrescido para os ecossistemas, e para a saúde humana. Com a realização deste trabalho, pretende-se, verificar as medidas de adaptação existentes na estratégia de gestão integrada portuguesa, no âmbito das alterações climáticas, efectuando uma comparação entre as medidas existentes em algumas das estratégias existentes no espaço Europeu e propondo algumas medidas a aplicar na estratégia existente em Portugal. ABSTRACT: Portugal posses a huge and vulnerable coastline that has been affected by human activities, putting its sea life to a high stress. The coastal areas are also associated with significant socio-economic activities, thus making them very sensitive areas. Therefore and as an additional problem comes the climate changes.These are combined with the rising sea levels, changes in frequency and/or intensity of storms and hurricanes associated with the resulting flood. Thus, the impacts of climate change on coastal areas will be, probably, aggravated by the fact that these areas are located in the major metropolitan areas of the country. As a result, the probability to affect a large number of people is particularly high, making it also increase the risk for ecosystems as for human health. The purpose of this thesis is to verify the adaptation measures, relative to the climate changes, within IZCM’s strategy. This is achieved by making a comparison between the existing measures in Portugal and in other European zones, and also, proposing some measures to implement to the current national strategy.

It is commonly asserted that the success of a software development project, and the usability of the final product, depend on the quality of the concepts that underlie its design. Yet this hypothesis has not been systematically explored by researchers, and conceptual design has not played the central role in the research and teaching of software engineering that one might expect. As part of a new research project to explore conceptual design, we are engaging in a series of case studies. This thesis reports on our case study on Git, a popular-yet sometimes puzzling-version control system. In an attempt to understand the root causes of its complexity, we analyze its conceptual model and identify some undesirable properties; we then present a reworking of the conceptual model that forms the basis of Gitless, our redesign of Git.

Abstract:
Git is a distributed version control system that tracks changes to a project overtime and is used to save these changes. Today it is being used by millions of people and is becoming a demand on the job market. For this reason it has become important to learn the version control system. Learning Git however may be difficult for beginners and learning it through tutorials may not always be effective. Learning it through a serious educational game (SEG) may be more effective as a SEG can provide motivation and feedback which are two factors for successful learning. This study seeks to assess how effective a SEG is in teaching Git by looking at the amount of knowledge gained from playing a SEG. This study also seeks to assess how much participants learned Git using a tutorial compared to participants who used a serious educational game. From the results, the study found that the SEG expanded the understanding of Git. The study also found that there was no significant difference in the amount of understanding gained from the SEG and the tutorial.

Transcription of the Schizosaccharomyces pombe fbp1 gene is subject to repression by environmental glucose that is detected through a cAMP signaling pathway. Previous research has identified several glucose insensitive transcription (git) genes that are required for glucose repression. One of these genes, git7, is required for cAMP signaling, but its exact function in the process is unknown. Analysis of this gene is complicated because it carries out additional essential functions that are unrelated to cAMP signaling. The git7-93 allele encodes a protein that possesses an18 amino acid duplication in the carboxy-terminus and only displays a defect in cAMP signaling, while two other mutant alleles that affect all Git7-dependent processes alter residues in the amino-terminus. My thesis work was composed of two projects designed to investigate the role of the Git7 carboxy-terminus in cAMP signaling and to identify more precisely the carboxy-terminal domain that is required for cAMP signaling. The first part of my study involved a genetic screen to isolate and characterize mutations that suppress the git7-93 allele, but fail to suppress a git7 mutant allele in which the amino-terminus of the protein is altered. Such allele-specific suppression may identify the direct target of Git7 function in the cAMP pathway. I have isolated a collection of 55 suppressor strains. Molecular and genetic analyses of these strains have reduced this number to nine candidates, which could have represented allele specific suppressors. However, the final genetic screen determined that all the candidates were extragenic suppressors. The second part of my study involved a screen for new mutant alleles of git7 that resemble git7-93 in that they confer a cAMP signaling defect, but do not confer cell wall or septation defects caused by the git7-27 or git7-235 alleles. This was carried out by performing a random mutagenesis on a cloned wild type git7 gene and then screening for plasmids that could suppress the cell wall or septation defects, but not the cAMP signaling defect, in a git7-235 host strain. Gastrointestinal tract (GIT) symptoms commonly affect endurance athletes such as those competing in the Ironman Triathlon. Although a number of risk factors and the pathophysiological mechanisms for the development of GIT symptoms during exercise have been proposed, scientific evidence in support of these factors and mechanisms is limited. Altered blood flow to the GIT during exercise have been suggested as a possible mechanism for the pathophysiology of GIT symptoms. However, changes in blood flow in the superioir mesentric artery (SMA) and the coeliac artery in athletes presenting with GIT symptoms during exercise have not previously been investigated.

Instructors use software called learning management systems to administer online courses. Traditionally these digital platforms mimic physical classrooms, but technology opens up potential for new types of interactions. In this thesis, we develop a learning management system on top of Git to create a learning environment that encourages collaboration and exploration. We discuss how it was designed, the underlying pedagogy, the technical implementation, and the results of a pilot online class, with 100 learners lasting 6 weeks. The platform architecture developed in this project can be replicated and reused for other online courses with similar goals.

Abstract:
Collaboration is one of the most important topics regarding the evolution of the World Wide Web and thus also for the Web of Data. In scenarios of distributed collaboration on datasets it is necessary to provide support for multiple different versions of datasets to exist simultaneously, while also providing support for merging diverged datasets. In this paper we present an approach that uses SPARQL 1.1 in combination with the version control system Git, that creates commits for all changes applied to an RDF dataset containing multiple named graphs. Further the operations provided by Git are used to distribute the commits among collaborators and merge diverged versions of the dataset. We show the advantages of (public) Git repositories for RDF datasets and how this represents a way to collaborate on RDF data and consume it. With SPARQL 1.1 and Git in combination, users are given several opportunities to participate in the evolution of RDF data.

Abstract:
This study examines the ancient Indian religious work, the Bhagavad Gita. This work of Hindu religious thought describes a mythic dialogue between Krishna and Arjuna, in which the issue of Arjuna's fighting against his family is discussed. Topics ranging from caste-bound duty to the metaphysical structure of reality are covered in this relatively short text. Whereas previous inquiries have focused solely on the philosophical doctrine of this work, this investigation examines the use and form of narrative elements within this dialogue to determine how they contribute to its potential effectiveness and message construction. Previous research has indicated that story telling is an integral format for the transmittal of Indian transcendent knowledge. Using the critical theory of narrative as expounded in the modern rhetorical tradition, this narrative is examined in regard to its internal structure and the meanings it invites the audience to extract. After situating this study in the stream of modern scholarship, a detailed explication of the narrative method of rhetorical criticism is offered, along with its subsequent application to the artifact in question. Various research questions are discussed in light of this application, with results being analyzed. Through narrative, this religious work is found to construct issues of "self," "others," "duty," and "reality" in interesting and unique ways. This study advances an understanding of multivalent narratives, such as the Bhagavad Gita, that use ambiguity in the text to open the audience to possibilities not experienced under their value system. This inquiry observes four main elements that must be present in order for a text to inspire possible value system change and novelty in audience members: these texts must contain differing value structures and hierarchies, they must not immediately or conclusively resolve the tensions between these value structures, the value structures implied in the text can admit to differing actions in relation to audience perception, and the various value structures within the narrative must not be so determinant that they cannot be reconciled. Through these means, narratives such as the Bhagavad Gita can expose temporally and spatially distant audiences to novel value hierarchies.

O quadro epistemológico é formado pela noção de tradução como uma atividade translatória, conforme Nord (2001), e pela abordagem descritiva nos estudos da tradução, trabalhada por Toury (1995) e Chesterman (1997). Também foram adotados conceitos teóricos de normas, memes, problemas de tradução, paratextos (principalmente de Genette, 1997) e notas de tradução. A metodologia da pesquisa detalha a escolha e a organização do corpus e o desenvolvimento da taxionomia das notas de tradução utilizada. Através desta taxionomia, analisa-se o conteúdo das notas de tradução em cada tradução em separado, em comparação com a média do corpus e em conjunto com outros paratextos da mesma obra. São feitas análises gerais das notas de tradução do corpus como um todo, incluindo distribuições do conteúdo entre as traduções e os capítulos, bem como análises das notas dos versos mais anotados, com discussões sobre probabilidade estatística destas ocorrências. Os principais resultados indicam memes e problemas de tradução. The main focus of this work is the description of the translation notes found in the Brazilian editions of the sanskrit text Bhagavad-Gita, using a computerized corpus composed of 1.005 notes. The epistemological framework is composed of the idea of translational activity, as presented by Nord (2001), and of the Descriptive Translation Studies approach as presented by Toury (1995) and Chesterman (1997). Concepts of norms, memes, translation problems, paratexts (mainly from Genette, 1997) and translation notes are also used. The methodology of the research details the corpus selection and structure, and the development of the taxonomy used. These taxonomy are used to analyze the subject of the note in each edition separately, including comparison to the corpus average and with the set of other paratexts into the same edition. A general analysis of the corpus as a whole is also presented, including the variation of the content on the different editions and chapters where they occur, as well as analysis of the notes on the most annotated verses, including discussion on the statistical probability of these occurrences. The main results show memes of translation and translation problems.

Abstract:
In this thesis we first investigate PBW deformations of Koszul, Calabi-Yau algebras, and we then study moduli spaces of representations of algebras defined by tilting bundles. These classes of algebras are generalisations of the skew group algebras appearing as noncommutative resolutions in the McKay correspondence, and the results we prove are motivated by corresponding results for skew group algebras. Koszul, Calabi-Yau algebras are Morita equivalent to path algebras with relations defined by superpotentials, and we classify which PBW deformations of these algebras still have relations defined by a superpotential and show that these deformations also retain the Calabi-Yau property. As an application of these results we show that symplectic reflection algebras are Calabi-Yau and can be interpreted as path algebras with relations defined by a superpotential. We then investigate when a variety with a tilting bundle can be produced as a moduli space of representations of an algebra defined by the tilting bundle. We find a set of conditions ensuring a variety and tilting bundle can be reconstructed in such a manner, and we show that these conditions hold in a large class of examples, which includes situations arising in the minimal model Program where the variety may be singular. As an example we show that the minimal resolution of a rational surface singularity can be produced as a moduli space for a noncommutative algebra such that the tautological bundle is a tilting bundle defining the noncommutative algebra.

When performing an analysis of the evolution of software quality and software metrics,there is a need to get access to as many versions of the source code as possible. There isa lack of research on how data or source code can be extracted from the source controlmanagement system Git. This thesis explores different possibilities to resolve thisproblem. Lately, there has been a boom in usage of the version control system Git. Githubalone hosts about 6,100,000 projects. Some well known projects and organizations thatuse Git are Linux, WordPress, and Facebook. Even with these figures and clients, thereare very few tools able to perform data extraction from Git repositories. A pre-studyshowed that there is a lack of standardization on how to share mining results, and themethods used to obtain them. There are several tools available for older version control systems, such as concurrentversions system (CVS), but few for Git. The examined repository mining applicationsfor Git are either poorly documented; or were built to be very purpose-specific to theproject for which they were designed. This thesis compiles a list of general issues encountered when using repositorymining as a tool for data gathering. A selection of existing repository mining tools wereevaluated towards a set of prerequisite criteria. The end result of this evaluation is thecreation of a new repository mining tool called Doris. This tool also includes a smallcode metrics analysis library to show how it can be extended.

Parabrahman Puruṣottama Bhagavān Svāminārāyaṇa offered novel contributions to the Vedānta philosophical tradition by presenting the principles of the Akṣarapuruṣottama Siddhānta (doctrine) and establishing the Akṣarapuruṣottama Darśana, an independent Vedāntic school of thought. In addition to revealing the darśana’s (school of thought) princi-ples, he also presented distinct readings of sacred texts. Among these readings, in “Va-canāmṛta Gaḍhaḍā Madhya 9” he put forth a unique interpretation of ‘dharma’ as it appears in verse 2.40 of the Śrīmadbhagavadgītā. Based on this reading and the Svāminārāyaṇabhāṣyam’s corresponding exposition, this work offers an analysis of Bhagavān Svāminārāyaṇa’s interpre-tation of ‘dharma’ as it appears in the Gītā. Unlike the classical commentators, Bhagavān Svāminārāyaṇa interprets ‘dharma’ as conviction in the present form of God (pratyakṣaparamātmasvarūpaniṣṭhā). This interpretation is signifi-cant in terms of its distinct content, its associated philosophical and theological commitments, and the hermeneutical and exegetical insights it offers. By elucidating Svāminārāyaṇa’s inter-pretation and the Svāminārāyaṇabhāṣyam’s corresponding exposition, this work presents (1) an unoffered discussion of Svāminārāyaṇa’s understanding of the nature and content of con-viction in the present form of God, (2) Svāminārāyaṇa’s relevant philosophical, theological, and hermeneutical principles, (3) an alternative reading of the Gita in light of Svāminārāyaṇa’s interpretation, and (4) cross-disciplinary darśanic insights that are characteristic of Svāminārāyaṇa’s interpretive move.

Abstract:
This thesis answers the research question: How do corporate social and environmental responsibilities (CSER) affect global IT outsourcing (GITO)? In answering this question we identified seven key trends that are directing CSER in GITO. We found that CSER in outsourcing is new and relevant, with growing interest from outsourcing providers and buyers. CSER will be driven by consumer concerns and employee expectations, which are particularly relevant for outsourcing buyers with a consumer oriented product or service, such as banks or retail organisations. The need to attract and retain employees will increase the need for CSER at outsource providers. Similarly, CSER is important to an organisation's brand reputation with consumers, employees and other stakeholders such as investors. Within CSER, environmental topics are a growing issue: the need to reduce power consumption and thereby reduce greenhouse gas (GHG) emissions from carbon-based power production. We learned of 'green-washing', the need to be suspicious of CSER claims that cannot be fully validated. This led us, and others, to suggest that due diligence is required to counter possible in-authentic CSER by GITO providers. As a method of validating CSER claims, we propose using global standards such as the Global Reporting Initiative and ISO 26000 when examining CSER at outsource providers. We examined CSER from a strategic perspective, to understand if CSER provides a long-term advantage to outsource providers. Directed by the research data and theoretical frameworks, we proposed a model of strategic and responsive CSER suggested by Porter and Kramer (2006). Responsive CSER describes the set of basic requirements that have become 'table-stakes' for GITO providers. Strategic CSER distinguishes outsource providers by providing long-term benefits that are not easily copied by competitors. By applying the strategic/responsive CSER model for GITO in a case study we developed a model that provides guidance to outsource buyers and their providers on when and how to share and collaborate on CSER projects. The key contribution of this research is a model that describes the characteristics for buyers and providers to collaborate on CSER projects to build trust in the outsourcing relationship and to create shared benefits for both parties and to society or the environment. This research applies the Porter and Kramer model to an outsourcing relationship to understand how CSER can be used to improve GITO.

In The Theology of Râmânuja, John Braisted Carman carefully examines the south Asian philosopher Râmânuja’s concepts of the Supreme Person, and the relational dynamics between the Supreme Person and the devotee. Carman sees in Râmânuja’s discussion of the master (úesî)/servant (úesa) relationship the most important understandings concerning the hierarchy between the Supreme Person and the devotee. Carmen argues that in this devotional relationship there is a point at which the distinction between the master and the servant is dissolved, and mutual dependence is revealed. This paper focuses on the point where the roles in the relationship between the divine and the devotee are reversed. In this reversal the master takes on the role of the servant and the servant the master. It is argued here that this role reversal is the action that illuminates the ultimate dissolution of the hierarchy that Râmânuja recognizes and results in spiritual empowerment for the devotee.

Abstract:
This study examines the ancient Indian religious work, the Bhagavad Gita. This work of Hindu religious thought describes a mythic dialogue between Krishna and Arjuna, in which the issue of Arjuna's fighting against his family is discussed. Topics ranging from caste-bound duty to the metaphysical structure of reality are covered in this relatively short text. Whereas previous inquiries have focused solely on the philosophical doctrine of this work, this investigation examines the use and form of narrative elements within this dialogue to determine how they contribute to its potential effectiveness and message construction. Previous research has indicated that story telling is an integral format for the transmittal of Indian transcendent knowledge. Using the critical theory of narrative as expounded in the modern rhetorical tradition, this narrative is examined in regard to its internal structure and the meanings it invites the audience to extract. After situating this study in the stream of modern scholarship, a detailed explication of the narrative method of rhetorical criticism is offered, along with its subsequent application to the artifact in question. Various research questions are discussed in light of this application, with results being analyzed. Through narrative, this religious work is found to construct issues of "self," "others," "duty," and "reality" in interesting and unique ways. This study advances an understanding of multivalent narratives, such as the Bhagavad Gita, that use ambiguity in the text to open the audience to possibilities not experienced under their value system. This inquiry observes four main elements that must be present in order for a text to inspire possible value system change and novelty in audience members: these texts must contain differing value structures and hierarchies, they must not immediately or conclusively resolve the tensions between these value structures, the value structures implied in the text can admit to differing actions in relation to audience perception, and the various value structures within the narrative must not be so determinant that they cannot be reconciled. Through these means, narratives such as the Bhagavad Gita can expose temporally and spatially distant audiences to novel value hierarchies.

Abstract:
Human MAP3K4 (MTK1) functions upstream of mitogen activated protein kinases (MAPKs). In the studies presented herein, MTK1 is shown to be required for human epidermal growth factor receptor 2/3 (HER2/HER3)-heregulin beta1 (HRG) induced extracellular acidification and cell migration in MCF-7 breast cancer cells. Furthermore, it was shown that HRG stimulation leads to association of MTK1 with tyrosine phosphorylated HER3 in MCF-7 and T-47D breast cancer cells. The MTK1/HER3 association was dependent on HER2 activation and was decreased by pre-treatment with the HER2 inhibitor, lapatinib. Furthermore, HER2 does not directly associate with MTK1, but phosphorylates HER3 transiently. MTK1 also has a role in the ERK1/2 MAPK signaling pathway in response to heregulin (HRG) stimulation in T-47D and MCF-7 breast cancer cells. In addition to MTK1, Shc, Grb2 and GIT1 proteins are all involved in the ERK1/2 MAPK pathway in response to growth factor stimulation. MTK1 was also shown to associate with activated ERK1/2, GIT1, Shc, Grb2 and p85 of PI3K in response to heregulin stimulation. ERK1/2 kinase activity is involved in aberrant signaling that leads breast cancer progression. GIT1 is a scaffolding protein that is linked to growth factor mediated ERK1/2 signaling in cell migration. Moreover, we also identify the actin interacting region (AIR) on MTK1 and disruption of actin cytoskeletal polymerization with cytochalasin D inhibited the interaction between HER3 and MTK1, indicating that f-actin (which is needed for cell migration) is required for the MTK1/HER3 association. Additionally, HRG stimulation leads to extracellar acidification that is independent of cellular proliferation. HRG induced extracellular acidification is significantly inhibited when MTK1 is knocked down in MCF-7 cells. Similarly, pre-treatment with lapatinib significantly decreased HRG induced extracellular acidification. Extracellular acidification is linked with cancer cell migration. We performed scratch assays that show HRG induced cell migration in MCF-7 cells. Knockdown of MTK1 significantly inhibited HRG induced cell migration. Furthermore, pre-treatment with lapatinib also significantly decreased cell migration. Cell migration is required for cancer cell metastasis, which is the major cause of cancer patient mortality. We identify MTK1 in the HER2/HER3-HRG mediated extracellular acidification and cell migration pathway in breast cancer cells.

The present paper makes an analysis of multiphase flow up (oil-gas) in laminar regime, with hidrodynamically developed profiles and interacting heat. The fluids analyzed are considered immiscible, developing into separated speed and temperature fields. The energy equations are applied to obtain of heat fields in the thermal entry region of the fluids flowing in the same direction. Although the results of parameters of practical interest has been obtained by purely numerical procedures, the system governing equations involving partial differential and have not been resolved successfully analytically yet. Thus, the use of Generalizes Integral Transform Technique, as powerful mathematical tool, is proposed to solve the energy equations associated with the flow and to producing hybrid results, analytical-numerical, more extension e analysis flexibility to a large number of parameter used in oil and gas engineering. Auxiliary problems with the greatest possible number of original problem information are presented in an attempt of improving the numerical convergence. Lastly, a computational routine is developed in FORTRAN to obtain the numerical solution. The numerical results obtained for the fields are presented in tables and graphs for then be compared with the maximum care to the parameters of special cases available in the literature.
O presente trabalho realiza uma análise do escoamento multifásico (óleo-gás) ascendente em regime laminar, com perfis hidrodinamicamente desenvolvidos e interagindo termicamente. Os fluidos em análise são considerados imiscíveis, desenvolvendo-se em campos separados de velocidade e temperatura. A equação da energia é aplicada para a obtenção dos campos térmicos em desenvolvimento na região de entrada térmica dos fluidos escoando na mesma direção. Embora resultados de parâmetros de interesse prático já tenham sido obtidos através de procedimentos puramente numéricos, o sistema de equações que regem esse tipo de escoamento envolve diferenciais parciais e ainda não foram solucionadas analiticamente com sucesso. Dessa forma, o uso da Técnica de Transformada Integral Generalizada, como ferramenta matemática, é proposto para resolver as equações de energia associada ao escoamento produzindo resultados híbridos, analítico-numérico, com maior flexibilidade de análise e extensão para um grande número de parâmetros utilizados na engenharia de petróleo e gás. Problemas auxiliares com o maior número possível de informações do problema original são apresentados na busca da melhoria da convergência numérica. Por fim, uma rotina computacional é elaborada em FORTRAN para a obtenção da solução numérica. Os resultados numéricos obtidos para os campos são apresentados na forma de tabelas e gráficos para em seguida serem comparados com o máximo de esmero aos parâmetros de casos especiais disponíveis na literatura especializada.

Objectives: To develop an assay to permit the identification of compounds that can inhibit the interaction between GIT1 and the amino-terminal SH3 domain (SH3-N) of GRB2. Methods: The GIT1 protein was expressed in Sf9 insect cells and purified using Talon resin beads. The SH3-N domain of GRB2 was expressed in the E. coli strain, BL21(DE3)pLysS, and purified using glutathione resin beads. The SH3-N domain was fluorescently tagged on cysteine 32 using Cyanine 3 maleimide. The fluorescence of the assay was measured by using a plate reader with excitation wavelength of 555 nm and emission wavelength of 570 nm. Results: The GIT1 protein was expressed in Sf9 cells and purified using the Talon beads. The SH3-N domain of GRB2 was expressed in BL21 cells and purified from the glutathione resin beads. The SH3-N domain was cleaved from GST by using thrombin, which was engineered into the GST fusion protein and were fluorescently labeled using Cyanine 3 maleimide. Conclusions: The fluorescence polarization assay that will detect the interaction between GIT1 and the SH3-N domain of GRB2 is still under development, but it has progressed towards completion since both components of the assay are in hand.

Abstract:
Many of us have encountered the discussion about which interface is better for working with Git, command-line or graphical. This thesis is an attempt to find out which user interface new Git users prefer for Git and what experienced Git users prefer. We aimed to find out if there’s anything significant which can be gained from using either of the interfaces in comparison to each other. Lastly we looked at what factors influences git users choice of user interface and how?. We have collected data through three interviews and a survey, which yielded approximately 370 responses. Based on our results we’ve found that the command-line interface is the more popular user interface, in general, amongst Git users. We’ve also found that most users stop using graphical user inter- faces, as their primary user interface, as they get more experience with Git. They usually change their primary user interface to a command- line interface or start using both the graphical user interface and the command-line interface together. The results from our study regard- ing why, is presented in this thesis.

Geomagnetically induced currents (GIC) are currents induced in ground-based conductor networks in the Earth's surface. The GIC are driven by an electric eld induced by geomagnetic variations which are a result of time-varying magnetospheric-ionospheric currents during adverse space weather events. Several studies have shown that there is a likelihood of technological damage (the power grid) in the mid- and low-latitude regions that could be linked to GIC during some geomagnetic storms over the past solar cycles. The effects of GIC in the power system can range from temporary damage (e.g. protective relay tripping) to permanent damage (thermal transformer damage). Measurements of GIC in most substations are done on the neutral-to-ground connections of transformers using Hall-effect transducers. However, there is a need to understand the characteristics of GIC in the power lines connected to these transformers. Direct measurements of GIC in the power lines are not feasible due to the low frequencies of these currents which make current measurements using current transformers (CT) impractical. This thesis discusses two techniques that can be employed to enhance understanding GIC characteristics in mid-latitude regions. The techniques involve the measurement of GIC in a power line using differential magnetometer measurements and modelling GIC using the finite element method. Low frequency magnetometers are used to measure magnetic felds in the vicinity of the power lines and the GIC is inferred using the Biot-Savart law. A finite element model, using COMSOL-Multiphysics, is used to calculate GIC with the measured magnetic field and a realistic Earth conductivity profile as inputs. The finite element model is used for the computation of electric field associated with GIC modelling.

The fission yeast Schizosaccharomyces pombe senses environmental glucose through a cAMP-signaling pathway. Elevated cAMP levels activate protein kinase A (PKA) to inhibit transcription of genes involved in sexual development and gluconeogenesis, including the fbp1⁺ gene, which encodes fructose-1,6-bisphosphatase. Glucose-mediated activation of PKA requires the function of nine git genes (git=glucose insensitive transcription), encoding adenylate cyclase, the PKA catalytic subunit and seven “upstream” proteins required for glucose-triggered adenylate cyclase activation. This thesis describes the cloning and characterization of the git10⁺ gene, which is identical to swo1⁺ and encodes the S. pombe Hsp90 chaperone protein. This discovery is consistent with the previous identification of the Git7 protein as a member of the Sgt1 Hsp90 co-chaperone family. Glucose repression of fbp1⁺ transcription is impaired by both hsp90⁻ and git7⁻ mutant alleles, as well as by chemical inhibition of Hsp90 activity and temperature stress. Unlike the swo1⁻ and git7⁻ ts mutant alleles, the git10-201 allele and git7-93 allele support cell growth at 37º and show no cytokinesis defect, while severely reducing glucose repression of an fbp1-lacZ reporter, suggesting a separation-of-function defect. A physical interaction between Git7 and Hsp90 in S. pombe was also detected and findings in this thesis suggest their involvement in the initial assembly of the cAMP complex

Abstract:
Neste trabalho é apresentada uma solução analítica de um problema bidimensional e transiente de dispersão de poluentes atmosféricos. O modelamento utilizado é conhecido na literatura como modelo Kzz para dispersão de poluentes atmosféricos e é representado por uma equação difusivo-advectiva com coeficientes de difusão e advecção variáveis. São utilizados três diferentes coeficientes de difusão nas simulações, bem como as componentes horizontal e vertical do vento são tomadas como variáveis. A solução analítica é gerada através da aplicação da técnica GITT (Generalized Integral Transform Technique) dupla com problema transformado resolvido por Transformada de Laplace e diagonalização de matrizes. Filtros matemáticos são usados para homogenizar as condições de contorno viabilizando o uso da técnica citada. Além disso, o tipo de filtro matemático utilizado permite a sensível diminuição do custo computacional. Resultados numéricos são obtidos e comparados com dados experimentais e outras soluções da literatura.
