The thesis is a genetic study of the works of the Sicilian writer Gesualdo Bufalino (1920-1996). Through the analysis of the writing process, this study reconstructs the genesis of all the works completed by Bufalino during his youth and up until his debut in 1981, with the novel Diceria dell'untore. The research carried out in the archives which hold the genetic materials of Bufalino enabled us to date the creative phase of Bufalino in the period 1955-65. In particular, we have identified in an unpublished novel, Il guazzabuglio, dating back to 1977, a kind of canvas or reservoir, from which the writer drew themes or narrative sequences for other works. This novel, still considered unfinished, was not only completed by Bufalino, but was also closely linked to the genesis of the first two novels, Diceria dell'untore (1981) and Argo il cieco (1984), on the one hand, and to Qui pro quo (1991 and Tommaso e il fotografo cieco (1996), Bufalino’s last novel which can be considered its rewriting. The works completed and published from 1981 until his death in 1996, were the result of a creative process begun during his youth and continued throughout his life. The reconstruction of the genesis of the works written before 1981 is accompanied by the reconstruction of the intellectual biography of Bufalino through the investigation of sources and archive documents

The planning of frameworks in a building site purposes to organize and synchronize the flows of production to obtain a good profitability of a project of construction. For optimizing the productivity of a building project, we will use techniques which help to decide the works to be realized day by day. The frameworks are decomposed into works belong to different realization areas. The repetitive cycle is the necessary period for realization of vertical and horizontal works of a typical floor. This cycle is reproduced at every level of the building taking into account a technical time lag between vertical works (walls, columns) and horizontal works (zones of horizontal formwork). Actually, the repetitive cycle is performed manually without an optimized solution. The cycle is controlled by many technical constraints: vertical constraints between vertical and horizontal works, horizontal constraints between vertical works in the same floor. This research work has an objective to carry out an optimization system to generate the repetitive cycle automatically for all types of buildings: walls structure, columns-beams structure, mixed structure. This system uses a satisfaction constraints technique to validate the constraints of a building site and the constraints between the different works to obtain admissible solutions. The optimization of repetitive cycle by G2-Cycle improves the productivity of building sites in reducing the quantity of utilized formworks and the quantity of work

The present work carried out on walnut consisted in exploiting the rich genetic resources available at the INRAE of Nouvelle-Aquitaine-Bordeaux, in order to provide the tools that can be used in a new breeding program conducted by the CTIFL, centre opérationnel de Lanxade. In fact, in view of the walnut's significant economic development, the choice of varieties in France does not seem to be sufficient to meet future new constraints such as global competition and climate change. The prospecting work carried out mainly by Éric Germain's team has made it possible to gather most of the species of the genus Juglans and numerous accessions of cultivated walnut, Juglans regia L, at the UEA of Toulenne. The use of his archives accumulated over 30 years permitted to make public important chronological phenotyping data concerning this collection. These data showed the advance of the phenology of the two control varieties 'Lara' and 'Franquette' in relation to climate change. Using a set of 13 SSR markers, alleles specific to Juglans species were identified and the structure of the collection was studied. This structure shows two main subgroups, one comprising accessions from Eastern Europe and Asia and the other from Western Europe and the USA. Also, a core collection was defined to carry out GWAS studies on the main traits of agronomic interest, from flower to fruit, using a 600,000 SNP chip developed by the University of Davis in California. Associations between SNPs and several phenology-related traits were identified using archives and newly acquired data. A SNP strongly related to the budbreak date of leaves and female flowers was identified on chromosome 1 and co-localized with a QTL detected in parallel on a F1 progeny. A KASP marker was validated with plant material from the University of Davis. Other associations were also identified for dichogamy and bearing habit, a trait directly affecting yield, and led to the definition of candidate genes. Further GWAS analyses were conducted on fruit-related traits such as nut size, weight, breaking yield and force required to break the shell. In parallel, methods using robust phenotyping techniques have been developed, such as the use of X-ray microtomography to measure all morphological traits without breaking the nut. Finally, work was carried out to compare the efficiency of the two types of markers used in this work, SSR and SNP. The results show that the 13 SSR markers give similar results to several thousands of SNPs with regard to structure determination and construction of core collections, which are essential in the management of genetic resources. In the long term, the results of this work will make it possible to initiate marker-assisted selection for the creation of new varieties, within the framework of a new improvement program to be carried out by the CTIFL. These new varieties will be able to meet the criteria sought in the coming years, taking into account climate change

Between the early 2000s and the early 2010s, four contemporary Western artists Christoph Büchel, Mike Nelson, Jonah Freeman and Justin Lowe built gigantic and enigmatic multi-room installations in Europe and the United States. Our initial hypothesis was that there were relationships between the labyrinthine super-productions of these artists, both in terms of their conceptions and their receptions.Our study describes and analyzes the specificities (construction, visitors reception, themes and references ...) and the issues (exploration, elaboration of a story, visitors’ endangerment and risk-taking...) of the aesthetic experiences offered by these works of art, nowadays invisible. The gathering and assessment of unpublished archives concerning ten of these artistic devices and their geneses as well as a field survey carried out among their spectators have enabled us to reconstruct mental images of these works. The plans and ekphrases, illustrated fictions, presented in this thesis reveal the spatial and temporal dimensions of these aesthetic walkthroughs and bring to light a set of similar set ups, materials, and themes. Based upon the scenographic strategies of the artists and the feelings of the spectators, our analysis leads us to consider these works as emotional installations and as dystopian narrative spaces. Walking across these, as if journeying through a voyage of initiation, sparks the spectator’s imagination and makes him fully conscious of his individuality and ability to take action

The work of Eça de Queiroz, a Portuguese novelist from the second half of the 19th century, has the particularity of including more posthumous works than publications during the author’s lifetime. The reason for this is certainly an extreme demand for perfection, as well as the author’s premature death in 1900 at the age of 55, leaving many unfinished manuscripts aside.This work proposes to revisit the period of the “fabrication” of the novel A Capital! published in 1925, in light of new documents discovered in the archives of the Eça de Queiroz Foundation, under the sign of the palimpsest. It is first necessary to reconsider the editing carried out by the author’s two sons, José Maria and Alberto, with a view to publication, and above all to measure its limits and contested points. This leads us to reconsider the corpus of the different manuscripts and to propose new perspectives. By delving deeper into this archeological work, this research also aims to address the different phases of evolution of the work, over the seven years that the author devoted to it, while keeping it, by the play of circumstances, almost constantly in the background. By shedding light on the various branches of this abandonned novel, this analysis helps to highlight its pivotal role, constituting both a breeding ground for characters and the emergence of a number of prevalent themes that may feed into subsequent works

Succinic acid is a linear and bi-functional molecule that has several practical applications including food, chemical and pharmaceutical industries. Thanks to increased knowledge on metabolism and pathway regulation of industrially relevant organisms, to the development of performant genetic tools and process engineering, recombinants strains (Escherichia coli, Corynebacterium glutamicum etc.) have been reported to be able to produce up to 100 g.L-1 with interesting yields (> 1.5 mole per mole glucose). C. glutamicum is well known as one of the best industrial producers of numerous amino acids (glutamate, lysine etc.). However, recent studies of C. glutamicum revealed its capability to produce several organic acids (succinate, lactate, acetate, etc.). In this work, we have deleted the ldhA gene of C. glutamicum by using a plasmid vector pk19mobsacBΔldhA. We demonstrated that the mutant and the wild type presented similar growth kinetics with maximal growth rate of about 0.7 h-1. We studied also the effects of oxygenation on C. glutamicum 2262 ΔldhA through cultures at different kLa and it appeared that lower kLa (<33 h-1) favored organic acids production wile higher favored bacterial growth. Furthermore, we designed a tri-phasic process with transition phase by regulation of dissolved oxygen concentration which resulted in the production of 327 mM of succinic acid with a yield of 0.94 mole per mole glucose. The application of the designed process to C. glutamicum 2262 wild type that normally produces lactate with a lactate to succinate production ratio up to 13.3 mol.mol-1, resulted in succinate concentration up to 793 mM (94 g.L-1) and 785 mM (71 g.L-1) of lactate. The succinate production yield was 1.1 mole per mole glucose and acetate production was negligible. These results underlined the importance of aerobic to anaerobic transition in succinate production processes of facultative aerobes and the necessity to engineer not only the microorganism but also the process. Finally, scale-down study have demonstrated the robustness of C. glutamicum against the oxygen gradients in bioreactor
