Abstract:

BACKGROUND: Among the genes involved in the control of crossing between common wheat and rye, the dominant suppressor SKr (Suppressor of crossability) having the major effect on inhibiting crossability is the most studied. DNA-markers have been developed for this gene. There are cases of successful application of some of them in wheat breeding, but there is practically no information about their diagnostic efficiency in screening of ex situ collections. MATERIALS AND METHODS: To evaluate the effectiveness of SKr markers Xcfb341, TGlc2, gene12 и gene13 to identify forms with high crossability with rye, we studied 103 bread wheat accessions from the VIR collection with different seed set (093%) after pollinated with rye. RESULTS: Efficiency in detection crossable forms (upper 15%) was demonstrated by markers Xcfb341, TGlc2 and gene12. No significant allele-trait association was found for gene13 marker. Ten haplotypes were identified based on four markers. Five haplotypes were revealed for accessions from China, while two of them were associated with high crossability. Eleven accessions including the line L6-HSR were heterogenic in markers. For this line the relationship of diagnostic fragments with the crossability level observed in the field has been experimentally confirmed. CONCLUSIONS: The SKr gene markers studied can be used for accessions searching at ex situ collections that potentially have high crossability with rye, for controlling the retention of recessive skr alleles when regenerating seed accessions, as well as in breeding programs.

Abstract:

How can we conceive of applying genetic criticism to a theatrical work, since drama is characterised by its inherent mutability and necessary incompletion? How can we analyse the genesis of a play which only becomes real on the evening of its performance? How can we defi ne the genesis documents when we know they are unstable, heterogeneous and ephemeral by defi nition? A requisite condition: the persons involved in stage direction as well as theatrical archives should be encouraged to preserve the fragile traces of the birth of a performance (logbooks, administration notebooks, rehearsal records, sketches for the sets, and so on). This study outlines a framework wherein the complex, dynamic relationship between the construction of a text and that of its performance can be exploited.

Abstract:

Abstract Background The rise of ‘big data’ in inflammatory bowel disease (IBD) presents an opportunity to improve understanding of pathogenesis and unpick the molecular complexity of this heterogenous condition. Personalisation of IBD management relies on predicting outcomes, response to therapy and prevention of complications. Here, we present results outlining subgrouping of patients and outcome prediction using multiomic/clinical data. Methods Using whole exome sequencing from 1100 patients in the Southampton IBD cohort, including 650 paediatric cases, we have performed iterative studies focused on 1) Impact of genomic variation across the NOD-signaling pathway measured by perturbation of transcription across multiple genes, 2) Development of NOD2 as a genomic biomarker of stricturing Crohn’s disease (CD), 3) Utilising machine learning and genomic data to develop disease classification models. These data utilise GenePy, a tool developed in house that summarises genomic variation to give a per individual, per gene deleteriousness metric. Results Within the NOD-signaling pathway patients harbouring deleterious variation in NOD2 had reduced NOD2 expression and increased NFKBIA expression, reflecting reduced NFKB signaling, figure 1A. We report deleterious variation in several key complexes including NOD2-RIPK2 and TAK1-TAB, resulted in reduced transcription of NFKB activators and alternative inflammatory pathway activation, figure 1C-D. Utilising genomic data we constructed a NOD2 prediction model for stricturing disease in Crohn’s disease; 56.7% of patients in the ‘high-risk group’ had stricturing behaviour, whilst in the low-risk group only 21.4% had strictures. Addition of terminal ileal (TI) disease to the NOD2 risk groups significantly improved prediction, figure 2A. Using survival modelling, high-risk group paediatric patients presenting with TI disease had a HR of 4.89 (P = 2.3×10-5) compared with the low-risk group patients without TI disease, figure 2B. Finally, we used supervised machine learning of genomic data to classify patients with CD or ulcerative colitis. We employed different gene lists and assessed how accurately we could assign patients to their diagnosis. An autoimmune gene panel produced the best model (AUROC 0.68), compared to an IBD panel (AUCROC 0.61). NOD2 was the most discriminating gene in all the gene panels. Conclusion These iterative projects demonstrate the utility of integrating genomic and clinical data to improve the subtyping of patients with IBD and provide disease prediction models. Future work will include analyses of additional inflammatory pathways and targeting different clinical outcomes. We hope clinical translation of these findings will be a step-change in precision medicine for IBD.

Abstract:

“Base Genep” is a Balinese spice that is usually served for all Balinese dishes and is usually served in a wet form. “Base Genep” or “Genep” seasoning must be used until finished because it is made from fresh ingredients that has a short shelf life. “Base Genep” is composed of turmeric, ginger, kencur, galangal, onion, garlic, pepper, coriander, candlenut, nutmeg, chili, shrimp paste, and salt. The content of bioactive compounds derived from the ingredients used makes “base genep” susceptible to oxidation processes that result in decreased quality and shelf life of base genep. This must be prevented by applying technologies such as extraction to increase the weakness of seasonings in the wet form. This study aims to know the effect of temperature and extraction time and determine the best treatment produce “base genep” extract. Extraction process using maceration method with ethanol solvent with temperature treatment and extraction time consisting of a temperature of 30ºC, 40ºC, 50ºC, 60ºC and time for 2, 3, and 4 hours. The results showed that the temperature and extraction time affected the characteristics of “base genep”. The higher the temperature and extraction time until the optimal conditions can increase yield, antioxidant activity, and total phenol extract “genep” seasoning. The results of the research showed a temperature of 50ºC for 4 hours was able to produce the characteristics of the best “genep” seasoning extract with a yield value of 25.392%, IC50 of 280.675 ppm, and a total phenol of 1177.97 mg/100 g.

Abstract:

Machine Reassignment is a challenging problem for constraint programming (CP) and mixed integer linear programming (MILP) approaches, especially given the size of data centres. Hybrid solutions mixing CP and heuristic algorithms, such as, large neighbourhood search (CBLNS), also struggle to address the problem given its size and number of constraints. The multi-objective version of the Machine Reassignment Problem is even more challenging and it seems unlikely for CP, MILP or hybrid solutions to obtain good results in this context. As a result, the first approaches to address this problem have been based on other optimisation methods, including metaheuristics. In this paper we study three things: (i) under which conditions a mixed integer optimisation solver, such as IBM ILOG CPLEX, can be used for the Multi-objective Machine Reassignment Problem; (ii) how much of the search space can a well-known hybrid method such as CBLNS explore; and (iii) can we find a better hybrid approach combining MILP or CBLNS and an- other recent metaheuristic proposed for the problem (GeNePi). We show that MILP can handle only small or medium scale data centres, and with some relaxations, such as, an optimality tolerance gap and a limited number of directions explored in the search space. CBLNS on the other hand struggles with the problem in general but achieves reasonable performance for large instances of the problem. However, we show that our hybridisation improves both the quality of the set of solutions (CPLEX+GeNePi and CBLNS+GeNePi improve the solutions by +17.8% against CPLEX alone and +615% against CBLNS alone) and number of solutions (8.9 times more solutions than CPLEX alone and 56.76 times more solutions than CBLNS alone), while the processing time of CPLEX+GeNePi and CBLNS+GeNePi increases only by 6% and 16.4% respectively. Overall, the study shows that CPLEX+GeNePi is the best algorithm for small instances (CBLNS+GeNePi only gets 45.2% of CPLEX+GeNePi’s hypervolume) while CBLNS+GeNePi is better than the others on large instances (that CPLEX+GeNePi cannot address).

Autor, rozważając sposoby rozumienia leksemu „fraszka” przez Jana Kochanowskiego, poddaje w wątpliwość tezę Sante Graciottiego, jakoby nie pojawił się ani razu w twórczości pisarza genologiczny sens tego leksemu. Za dostateczny dowód, iż jest inaczej, uznaje on opatrzenie przez czarnoleskiego poetę takim a nie innym tytułem swej obszernej epigramatycznej antologii w momencie porządkowania dorobku całego życia, a potwierdzenie, iż sens ten doczekał się rychłego zadomowienia, przynosi wydany krótko potem zbiór Melchiora Pudłowskiego Fraszek księga pierwsza (1586). W drugiej części artykułu próbuje się ustalić, komu należy przypisać pierwszeństwo, jeśli chodzi o użycie wyrazu „fraszka” w druku. Jak się okazuje, u samego Kochanowskiego leksem ten po raz pierwszy pojawia się w Satyrze (1564), wyprzedzają go natomiast nieznacznie zarówno Rozmowa albo dyjalog około egzekucyjej (1563) Stanisława Orzechowskiego, jak i anonimowa Historyja trojańska (1563).

Abstract:

The article is an attempt to refer to an ongoing discussion on the genesis of Old-Belarusian Tristan that has been preserved in the 16th-century Raczynski Codex in Poznan. The author shares an opinion that there was a South Slavic tie between Belarusian and Italian texts and gives linguistic arguments supporting this view. Lexical analysis of the text reveals words of South Slavic provenance that have been found in various Balcan languages. The group of so-called serbisms (Serbian vocabulary) known in other publications has been completed with new Balkan lexical elements certified in the vocabulary of Belarusian translation.

W koncepcji Alvina Plantingi, sensus divinitatis („zmysł boskości”) jest ludzką władzą poznawczą, nakierowaną na tworzenie posiadających gwarancję (pozytywny status epistemiczny) przekonań o Bogu. Większość komentatorów myśli Plantingi koncentruje się na pytaniu, czy wprowadzenie tej koncepcji faktycznie pozwala obronić racjonalność przekonań teistycznych, pomijając przy tym zazwyczaj genetyczny aspekt funkcjonowania zmysłu boskości. Niniejszy artykuł ma na celu wypełnienie tej luki. Na podstawie Warranted Christian Belief Plantingi wyróżniam trzy możliwe interpretacje tego, jak sensus divinitatis tworzy przekonania teistyczne. Wskazuję także, w jaki sposób ludzki grzech wpływa na poprawne funkcjonowanie zmysłu w ramach każdej z tych interpretacji.

Artykuł referuje poglądy amerykańskiego filozofa Daniela C. Dennetta, dotyczące językowego charakteru świadomości. W pierwszym kroku autor rekonstruuje Dennettowską strategię naturalizacji świadomości, pokazując, jak język umożliwia powstanie świadomości w toku ewolucji ludzi. Naturalizacja ta jest możliwa, wedle Dennetta, jedynie przy behawiorystycznym założeniu, zgodnie z którym świadomość jest ukrytym zachowaniem językowym. Autor pokazuje, jak Dennett przyswaja i przekształca pierwotne pomysły behawiorystów Meada, Skinnera i Jaynesa, wpisując je w ewolucyjny scenariusz. Inskrypcja ta prowadzi do konkretnych problemów, rozważanych w ostatniej części pracy: a) w filozofii Dennetta występują właściwie dwie definicje świadomości: pierwsza głosi, że świadomość jest pewnego rodzaju zachowaniem językowym, druga natomiast, że jest wirtualną maszyną memetyczną; b) Dennett nie wyjaśnia zasady rozstrzygalności, która pokazywałaby, jak dana treść umysłu może zostać uświadomiona, zamiast tego podaje tylko metaforę rywalizacji, w której pewne zdarzenia w mózgu (treści umysłowe) wygrywają z innymi; c) wpisanie możliwego scenariusza rozwoju świadomości w pejzaż ewolucji nie oznacza jeszcze, że teoria może zostać empirycznie sprawdzona.

The Crisis of Globalization – the Analysis of the Genesis and the Understanding of the Process The main goal of this paper is to explain the phenomena of globalization in the context of its possible crisis, which influences the recent public discourse debate. The study answers two research questions: what the genesis of the globalization is, and how this phenomenon can be understood. Considering the first question, three theoretical approaches were analysed: structural, nonhomogeneous and constructivist. As for the second question, the author presents globalization with reference to internationalisation, universalisation, westernization, and liberalization.

Abstract:

Verticillium longisporum is a vascular pathogen that infects the Brassicaceae host plants Arabidopsis thaliana and Brassica napus. The soilborne fungus enters the plant via the roots and colonizes the xylem of roots, stems, and leaves. During late stages of infections, Verticillium spp. spread into senescing tissue and switch from biotrophic to a necrotrophic life style. Typical symptoms of V. longisporum-induced disease are stunted growth and leaf chlorosis. Expression analyses of the senescence marker genes SENESCENCE-ASSOCIATED GENE12, SENESCENCE-ASSOCIATED GENE13, and WRKY53 revealed that the observed chlorosis is a consequence of premature senescence triggered by Verticillium infection. Our analyses show that, concomitant with the development of chlorosis, levels of trans-zeatin decrease in infected plants. Potentially, induction of cytokinin oxidase/dehydrogenase expression by Verticillium infection contributes to the observed decreases in cytokinin levels. Stabilization of Arabidopsis cytokinin levels by both pharmacological and genetic approaches inhibits Verticillium proliferation and coincides with reduced disease symptom development. In summary, our results indicate that V. longisporum triggers premature plant senescence for efficient host plant colonization.

Abstract:

Instant spices is a mixture of several spices that are young to be found in the market at an affordable price, and are usually used immediately as a cooking spice for certain foods. This study aims to determine the quality of the "Base Genep" instant spices sold in the East Denpasar Market. This research is a descriptive study using survey and experimental methods in the laboratory. The number of samples to be studied was 44 samples from 81 populations. The variables in the study were moisture content, pH, Escherichia coli, Total Plate Count (TPC) and mold. The results showed that from 44 samples of "Basa Genep" instant seasoning water content test results obtained were 21.62% - 23.707%, while the pH value of 44 samples of "Basa Genep" instant spice was obtained between 2.2-2.5 so that it meets the requirements.

Abstract:

W artykule autor koncentruje się na wybranym aspekcie problemu związanym z rozumieniem historii genezy teoretyzowania w nauce o stosunkach międzynarodowych. W szczególności ukazuje jak zwrócenie większej uwagi na stosunek między tekstem i kontekstem może uchronić przed błędną interpretacją kolektywnego historiograficznego rozumienia dyscypliny, a w szczególności genezy teoretyzowania w NSM. Postępowanie to zostało przeprowadzone w oparciu o analizę tekstów i kontekstów teoretyzowania stosunków międzynarodowych dwóch autorów symbolizujących: podejście idealistyczne – Ralpha Normana Angella Lane’a (1872–1967) oraz podejście realistyczne – Hansa J. Morgenthau (1904–1980). Artykuł omawia także najważniejsze ustalenia rewizjonistycznej literatury o Pierwszej „Wielkiej Debacie” i kształtowanie się mitu „Wielkich Debat” jako opisu historii teoretyzowania o stosunkach międzynarodowych.

Abstract:

The article discusses the Proposicio Polonorum, which contains an accusation against the Grand Master and the Teutonic Order of harbouring a hostile attitude towards Poland, King Władysław Jagiełło, and the Christianizing mission led by him in Lithuania. The indictment was presented by the Polish delegation to the Council of Constance in February 1416. The author discusses the manuscripts and the contents of the indictment. He compares various versions and on this basis considers the question of the authorship and the genesis of the text. The “Kraków” version held in the Jagiellonian Library (Cod. 1143) contains several “Lithuanian” elements (the marriage of the daughter of Witold with the Grand Duke of Moscow; the alliance of Svidrigiello with the Tatars) that are unknown in other manuscripts. This might indicate its preparation to support Duke Witold’s interests. Any details of the genesis of the indictment are hypothetical, but several factors emerging directly from its content (quotations from the twelfth-century letter writer Pierre de Blois) indicate that Piotr Wolfram, secretary to the Archbishop of Gniezno at the Council and professor of law at Kraków University, had a fundamental influence on its writing.

Abstract:

Szkic podejmuje problem genezy w Czekając na Godota Samuela Becketta. W pierwszej części ukazano genezę w sensie formalnym. Wymieniono w niej czynniki, które wpłynęły na powstanie tragikomedii. Są to: aspekt biograficzny, związany z pobytem Becketta w czasie II wojny światowej na południu Francji oraz inspiracje kulturowe (filozoficzne, religijne i literackie). Fascynacja autora frazą z Wyznań Świętego Augustyna wpłynęła na płaszczyznę formalną utworu. Beckett zrealizował w nim ideę jedności formy i treści. W dalszej części szkicu poruszono problem genezy rodzaju ludzkiego. Człowiek, od zarania dziejów samotny, dążył do odmiany losu. Wytworzył Rozum, Intelekt i rozpoczął dialog niwelujący samotność oraz rozpacz. Ten zabieg nie przyniósł spodziewanych efektów, dlatego też zrodziła się idea istoty transcendentnej wobec niego. Od tego momentu człowiek czeka na nią i na odmianę swego losu.

Przyjmując rozróżnienie światów tekstowych i światów fikcyjnych, możemy się wesprzeć logiką światów możliwych, aby spróbować wyjaśnić status światów, zarazem prawdziwych i wirtualnych, stworzonych w genezie, oraz aby zdefiniować reguły dostępności, które kierują relacjami między tymi światami.

Abstract:

The author deals with the problem of differences in meaning between the concepts of socialization and education. In social sciences, and especially in educational sciences, there is no agreement as to the relationship between the scopes of these two fundamental concepts. The article attempts to overcome these semantic difficulties by adopting an evolutionary perspective in line with the assumption that education emerged as a specific type of socialization in human prehistory. The search of its essential properties (and thus differences), was guided by the cognitive directive of Aristotle, according to which “the best understanding of things is achieved by those who study them at their inception”. The article uses the method of data analysis established in numerous studies on social and cultural human development, starting from the earliest stages of life in primitive groups. Socialization appeared in these groups as a way of transmitting collective experience, and thus culture, necessary for their survival. The continuation of the analysis leads to investigating what factors made this culturation take a different form, namely that of education. As it turns out, this difference is difficult to discern in the intentionality of cultural influences. It appears only when the group becomes aware of the need to adapt adolescents to fulfill various social roles. This already requires going beyond socialization, which is always a spontaneous process including widespread participation of group members in the transmission of an experience that is also common. In order to provide selected group members with selected elements of the experience linked to their social role, the group must appoint competent members for the transfer of the experience selected to perform specific social roles. These activities constitute one more role distinguished in the group – the role of the educator. And the activities that make up their performance are education.

Abstract:

Artemisia eriantha Ten., also known as "Apennines genepì", is a valuable aromatic plant with several medicinal properties. Among the several phytoplasma symptoms to which this plant can be subjected, the fasciation is one of the less reported and studied. In natural environments this plant is a characteristic species of the association Artemisietum erianthaeBrullo 1984. In this study, the first occurrence of fasciation in Artemisia eriantha is described. This phenomenon was observed during the ex vitro experimental cultivations, located in Assergi and Barisciano (L’Aquila, Italy), settled for the restocking of this endangered species. The fasciation occurred only in Assergi experimental field with a magnitude of 0.12%. Specific PCR analysis did not show the phytoplasma presence. This fist report allows to expand knowledge about fasciation in Apennines genepì.

Abstract:

The article discusses the impact of the experience of the Holocaust and the war on the form and issues of eight volumes of Józef Hen’s literary journals I am not Afraid of Sleepless Nights. The “long shadow” of these traumatic events shaped both the history of their delayed origins, the form of the notes, and their main themes. The poetics of these diaries is based on the structure of a collage, which, on the one hand, connects the past and present, and on the other, it combines the chronicle of everyday life with night-time meditations, memories and reflections on history, wartime and post-war experiences and the Jewish fate. Hen contrasts these devastating forces of the twentieth century with his own philosophy of life, which is a special form of Jewish vitalism. The diary thus becomes a symbolic form of survival in the form of a literary, autobiographical work. All the volumes of Hen’s journals seem to be permeated by the effort to record, preserve and perpetuate what is characterized by the fragility and susceptibility to the corrosion of time – that is, life itself. Ultimately, thanks to this effort, the trace of the author’s own presence is also preserved. The collage poetics of the record, resulting from “composing”, makes it part of the work itself. The subject is transformed in it into an idiomatic “gesture”, i.e., a trace of a constantly repeated expressive act, which seems to retain traces of the author’s expressive gestures, his preferences, choices and ways of seeing, and the modality of individual feeling recreated in the text. In a way, it can be said that, according to Hen, journals are written to preserve the specific literary composition of existence. A diary understood as a “form of life” preserves a certain inalienable ethical moment, contained both in the need to give a certain order to the biography and to save the life experience itself – in its various forms. While history and everyday life constitute a source of a constantly reborn fear in Hen’s diaries, as they seem to still emerge in the long shadow of the Holocaust and the possibility of its possible repetition recognized in many events, the very ability to transform them into a diaristic record counteracts it therapeutically. For despite the fact that life can be destroyed, thanks to writing – as Hen’s final lesson seems to sound – something can always remain and survive.
