Abstract:
Abstract Social epigenomics is a new field of research that studies how the social environment shapes the epigenome and how in turn the epigenome modulates behavior. We focus on describing known gene–environment interactions (GEIs) and epigenetic mechanisms in different mammalian social systems. To illustrate how epigenetic mechanisms integrate GEIs, we highlight examples where epigenetic mechanisms are associated with social behaviors and with their maintenance through neuroendocrine, locomotor, and metabolic responses. We discuss future research trajectories and open questions for the emerging field of social epigenomics in nonmodel and naturally occurring social systems. Finally, we outline the technological advances that aid the study of epigenetic mechanisms in the establishment of GEIs and vice versa.

Resumo: O estudo do comportamento viscoelástico dos géis de colágeno é importante para sua caracterização reológica visando-se aplicações em bioengenharia e práticas clínicas. Neste trabalho, as propriedades viscoelásticas dos géis de colágeno, obtidos a partir do tendão da cauda do rato (RTT), foram avaliadas e correlacionadas em função de variáveis de preparação (tempo de diálise, concentração de colágeno e pH, e da freqüência de oscilação do reômetro (Haake CV20, no modo oscilante e configuração de placas paralelas). Os parâmetros reológicos avaliados foram: módulo de armazenamento (G¿), módulo de dissipação (G¿¿) e viscosidade complexa ('eta POT. *¿). O desenvolvimento da estrutura do gel durante a diálise foi acompanhado por micrografia óptica e correlacionado com os parâmetros reológicos medidos. A concentração de colágeno variou com o tempo de diálise devido à hidratação das fibras. Os géis mostraram estruturas fibrilares características, nas quais as fibras estavam entrelaçadas e formando anéis e/ou arranjos helicoidais. O modelo de Maxwell e de Zenner, mostraram-se apropriados para a descrição das características viscoelásticas dos géis de acordo com suas concentrações de colágeno: o modelo de Maxwell para baixas concentrações de colágeno (menor que 1% em peso) e o de Zenner modificado para concentrações moderadas de colágeno (de 1% até 2,50% em peso) ...Observação: O resumo, na íntegra, poderá ser visualizado no texto completo da tese digital
Abstract: The study of the viscoelastic behavior of collagen gels important for its rheological characterization aiming applications in bioengineering and clinical practice. Viscoelastic properties of native and crosslinked collagen gels obtained from rat tail tendon (RTT) were evaluated and correlated as a function of gel preparation variables (dialysis time, collagen concentration, and pH) and oscillation frequency. The rheological parameters evaluated were the storage module (G¿), dissipation module (G¿¿), and complex viscosity ('eta POT. *¿), using a Haake CV20 rheometer operating with two parallel plate configuration in oscillating mode. The development of the fibrilar structure at the gel during dialysis was evaluated by optical photomicrography and correlated with the reological parameters. The collagen concentration changed with dialysis time due to the hydration of the fibers. The gels showed a typical fibrilar structure in which the fibers were interwinned forming either rings and/or helical arrangements. The models of Maxwell and Zenner showed to be appropriate to describe the viscoelastic behavior of the gels. The Maxwell model was in a good aggrement with the experimental rheological parameters for the low range collagen concentration (less than 1% by weight), while moderate collagen concentration range (1.0% to 2.5% by weight) had a good fit by modified Zenner¿s model... Resumo: A influência da concentração de polissacarídeo, do pH de equilíbrio (de 2,0 a 4,0) e da aplicação de tratamento térmico (70 ou 90ºC/ 30min) sobre as propriedades reológicas e de microestrutura de géis de gelana acidificados lentamente pela adição de glucona-d-lactona (GDL) foi estudada nesse trabalho. Os sistemas foram caracterizados quanto à cinética de gelificação e propriedades dos géis formados, como as reológicas em compressão uni- e biaxial, capacidade de retenção de água (WHC), microestrutura e turbidez. Em todas as amostras que formaram géis auto-sustentáveis, com exceção daquelas com 1% de gelana nos menores pHs, foi possível determinar o ponto de gel a partir de ensaios oscilatórios, sendo que o tempo do mesmo diminuiu com o aumento da concentração de polímero, com o tratamento térmico, e com a diminuição do pH final. Todos os géis auto-sustentáveis foram transparentes e não apresentaram sinerese. A tensão de ruptura aumentou com a diminuição do pH, aumento da concentração de gelana e submissão a tratamento térmico antes da adição de GDL, porém diminuiu quando este foi realizado depois da acidificação. A WHC tornou-se maior com o aumento da concentração de gelana e do pH nos sistemas de concentração 1% e 0,5%, no entanto, a 0,2%, ela aumentou com a diminuição do pH. A microestrutura dos géis mostrou correlação direta com as propriedades reológicas em compressão. A análise da dimensão fractal dos géis só foi possível em pHs 2,5 e 2,0, e os sistemas apresentaram estrutura fractal com regime de ligação fraca. Todas as amostras mostraram comportamento de fluido lei da potência em escoamento extensional. A correlação entre turbidez e força dos géis foi linear. No entanto, a turbidez mostrou-se mais sensível às mudanças estruturais do que as propriedades mecânicas
Abstract: The influence of polysaccharide concentration, pH at equilibrium (2,0 - 4,0) and annealing (70 or 90ºC/30min) on rheological properties and microstructure of gellan gels formed by acidification of glucon-d-lacton GDL was evaluated in this study. Gels were characterized in relation to their gelling kinetics, mechanical properties at fracture, water holding capacity (WHC), microstructure, turbidity and rheological parameters in extension. In all samples, with exception to that of 1% concentration formed at pHs 2,5 and 2,0, it was possible to determine the gel point by oscilatory measurements. The time at which gel point occurred diminished by raising polymer concentration, annealing and lowering final pH. All gels were transparent and exhibited no sign of syneresis. Failure stress increased at lower pH values, higher concentrations and gels were even stronger when made from annealed solution. When gels were annealed after pH reached equilibrium, their failure stress decreased. The WHC for 1% and 0,5% gels was higher at enhanced concentrations and higher pH values and for 0,2% gels, it was higher at lower pH values. Microstructure of gels showed straight correlation with their rheological properties in compression. Analysis of fractal dimension of the gels was only possible at pHs 2,5 and 2,0. In this case, the systems showed fractal structure in the weak¿link regime. All samples showed power law behaviour in extension. The turbidity and the force of gels showed linear correlation. Nevertheless turbidity measurements were more sensitive to structural changes than were mechanical properties

Resumo: biopolímero denominado quitosana vem sendo reconhecido como uma importante fonte de matrizes para adsorventes em processos de recuperação e purificação de bioprodutos, com novas aplicações em várias áreas do conhecimento como a biotecnologia. Uma importante vantagem nessa utilização é a disponibilidade da quitina na natureza, podendo ser encontrada facilmente na carapaça de crustáceos. A flexibilidade apresentada por essas matrizes se deve principalmente à compatibilidade adsorbato/adsorvente e à possibilidade de modificações estruturais de modo a atender a características diversificadas de interação química e de resistência mecânica. No presente trabalho foi realizada a caracterização reológica de soluções e géis de quitosana, com a determinação das propriedades viscosas e viscoelásticas. As soluções concentradas foram obtidas por dissolução da quitosana em solução de ácido acético e os géis foram obtidos através de modificações da quitosana com a utilização do glutaraldeído, nas concentrações de 2,5g/100ml, 3,Og/100ml e 3,5g/100ml e nas temperaturas de 10°C, 30°C e 50°C. As propriedades viscosas e viscoelásticas das amostras de quitosana foram obtidas com a utilização do reômetro Haake CV20. Esse equipamento pode ser programado para trabalhar em dois métodos de obtenção de dados experimentais. No método de cisalhamento permanente, as propriedades obtidas são representativas do comportamento viscoso das amostras, através da obtenção das seguintes variáveis: tensão de cisalhamento, taxa de cisalhamento e viscosidade de cisalhamento. No método de cisalhamento oscilatório, as propriedades determinadas são representativas do comportamento viscoelástico das amostras, através da obtenção das seguintes variáveis: módulo de rigidez, módulo de dissipação e viscosidade complexa. Foi realizada a interpretação da correlação estrutura - propriedade apresentada pela quitosana e pelos géis modificados através da reticulação com glutaraldeído. Foram atribuídos modelos específicos para o comportamento reológico das soluções e géis de quitosana. A determinação dos parâmetros nas diversas faixas estudadas permite concluir que é possível preparar e caracterizar a quitosana na forma de soluções concentradas e na forma de géis de acordo com a concentração e ° grau de reticulação das amostras analisadas
Abstract: The chitosan biopolymer has recently being appointed as an important source of matrices used as adsorbents in processes for the recovery and purification of bioproducts, with new applications in areas such as biotechnology. An important advantage is that chitin occurs naturally and is commonly found in crustacean shells. The flexibility exhibited by these matrices became from the compatibility adsorbate/adsorbent and the possibility of structural modifications to satisfy diversified characteristics of chemical interaction and mechanical strength. In this work, the rheological characterization of chitosan solutions and gels was carried out in order to investigate viscosity and viscoelasticity properties. The concentrated solutions were prepared by dissolving chitosan with acid acetic solution and the gels were prepared by adding glutaraldehyde, solutions at different concentrations of 2,5g/100ml, 3,Og/100ml and 3,5g/100ml and temperatures of 10°C, 30°C and 50°C. The viscosity and viscoelasticity properties of chitosan samples were measured with a rheometer (model Haake CV20). The equipment can be operated in two different ways in order to obtain the experimental data. The first one is the steady shear rheological measurement, where the viscosity properties are described through values for shear stress, shear rate and shear viscosity. The second is the oscillatory shear rheological measurement, where the viscoelasticity properties are described through values for storage modulus, 1055 modulus and complex viscosity. The correlation between structure and properties exhibited by chitosan samples and gels modified by crosslinking with glutaraldehyde were studied. Rheological specific models were adjusted to the experimental data in order to describe the behavior of chitosan solutions and gels. The parameter determinations allow concluding that is possible to prepare and characterize chitosan samples as concentrated solutions and gels depending only on the concentration and the degree of crosslinking of the sample studied.

Estudos sistemáticos utilizando a técnica de SAXS foram realizados no síncrotom do LURE (Orsay) com feixe intenso de geometria pontua:, para descrever a cinética e as estruturas obtidas durante as etapas de transformação sol -> gel úmido -> gel seco -> vidros de sílica . As análises foram feitas em termos de uma lei de potência I = q-? cujo expoente está relacionado a dimensão fractal de massa ou de superfície das estruturas. Devido a polidispersividade das unidades espalhadoras as dimensões fractais verificadas podem ser resultados de dimensões reais encobertas por uma distribuição de tamanhos. As curvas obtidas para a cinética de gelificação e envelhecimento dos géis úmidos mostram expoentes que indicam fractalidade de massa e, posteriormente, de superfície. Uma microestrutura composta, que pode resultar de uma agregação limitada por difusão (DLA) seguido de agregação de agregados (C-C) foi observado nas soluções básicas. Por outro lado, os agregados ácidos parecem surgir do crescimento contínuo de cadeias até atingir a interligação com outros agregados com uma cinética que pode ser descrita pelo modelo DLA. No estado de aerogeis outras técnicas como picnometria de mercúrio e hélio, microscopia eletrônica de transmissão e adsorção de gás de nitrogênio, foram também aproveitadas. Nestes materiais encontramos em uma faixa estreita de escala, um expoente indicando fractalidade de massa, embora com superfície lisa. Os resultados estão de acordo com um modelo de estrutura que tem uma matriz de SiO2 densa. A estrutura fractal desaparece durante o tratamento térmico de densificação devido ao rearranjo estrutural.
Systematic SAXS studies have been performed at the LURE synchrotron (Orsay) using an intense beam of point-like cross-section to describe the kinetic and the structure obtained during the sol-> humid gel -> dry gel ->silica glass. The analysis were done in terms of a power law, I = q-? , whose exponent is related to mass and surface fractal dimensions of the structures. Due to polidispersity of the scattering units the found fractal dimensions can be \"smeared\" dimensions that result from a size distribution. All the kinetic and aging humids curves showed evidenced of fractal structures. A composed microstructure that can be originated by clustering of clusters (C-C) of primary units grown by diffusion limited-aggregation (DLA), was observed for basic gels. By other hand, the acidic cluster seems to grow continuously reaching gelation through crosslinking of the clusters. The DLA model seems to describe this process. In the aerogels study others techniques like density measurements using Hg and He, TEM and adsorption of N2 gas were done along with SAXS measurements. Here we could find a narrow range in which the system is a mass fractal, although its surface has been already smoothened. The results agree with a dense SiO2 matrix model. During the densification no fractal structure was verified due to structural rearrangements at high temperature.

The activity of requirements engineering is seen in agile methods as bureaucratic activity making the process less agile. However, the lack of documentation in agile development environment is identified as one of the main challenges of the methodology. Thus, it is observed that there is a contradiction between what agile methodology claims and the result, which occurs in the real environment. For example, in agile methods the user stories are widely used to describe requirements. However, this way of describing requirements is still not enough, because the user stories is an artifact too narrow to represent and detail the requirements. The activities of verifying issues like software context and dependencies between stories are also limited with the use of only this artifact. In the context of requirements engineering there are goal oriented approaches that bring benefits to the requirements documentation, including, completeness of requirements, analysis of alternatives and support to the rationalization of requirements. Among these approaches, it excels the i * modeling technique that provides a graphical view of the actors involved in the system and their dependencies. This work is in the context of proposing an additional resource that aims to reduce this lack of existing documentation in agile methods. Therefore, the objective of this work is to provide a graphical view of the software requirements and their relationships through i * models, thus enriching the requirements in agile methods. In order to do so, we propose a set of heuristics to perform the mapping of the requirements presented as user stories in i * models. These models can be used as a form of documentation in agile environment, because by mapping to i * models, the requirements will be viewed more broadly and with their proper relationships according to the business environment that they will meet

No entanto, a falta de documenta??o no ambiente de desenvolvimento ?gil ? apontada como um dos principais desafios da metodologia. Assim, observa-se a exist?ncia de um contrassenso entre o que a metodologia ?gil defende e o resultado que ocorre no ambiente real. Por exemplo, nos m?todos ?geis as hist?rias de usu?rio s?o a forma mais usual para descrever requisitos. No entanto, essa maneira de descrever requisitos ainda n?o ? suficiente, pois as hist?rias de usu?rio constituem um artefato muito restrito para representar e detalhar os requisitos. As atividades de verificar quest?es como o contexto do software e depend?ncias entre as hist?rias tamb?m s?o limitadas com o uso somente desse artefato. No contexto de engenharia de requisitos existem as abordagens orientadas a metas que trazem vantagens para a documenta??o de requisitos, entre elas, completude dos requisitos, an?lise de alternativas e suporte ? racionaliza??o de requisitos. Dentre essas abordagens destaca-se a t?cnica de modelagem i* que fornece uma vis?o gr?fica dos atores envolvidos no sistema e suas depend?ncias.
