Abstract:
The government has launched a program of self-sufficiency in meat as an effort to overcome the shortage of beef supply to meet the needs of the community, especially when the day of religious festivities. GBIB program Implementation is expected to meet the needs of the community will be beef, as demand for beef is always soaring demand every year, especially ahead of religious festivals. To discuss and review the implementation of the program stretcher author uses the theory EDWARDS III to determine the success or failure of the implementation of the program in the district of Kota Waringin Barat. Issues to be examined are 1) the quality and quantity of resources, 2) communication and dissemination program, 3) Attitude 4) Bureaucracy. This will be the focus to see the successful implementation of the program of snapping / sync Lust and optimize of GBIB in the district of Kota Waringin. This study uses the concept of a qualitative descriptive approach with the aim of providing an overview of how the implementation of the field program and then analyzed using the theory of Edwards III. The results of this study show that: 1) Aspects of Resource (HR, Information, Privileges and amenities), some support as the support means the production of drugs / vitamins and breeding stock (straw) superior adequate and well as the authority and the information available is quite good, while some aspects that do not support such as: HR, execution time, geographical conditions, transport facilities support, 2) aspect Communications (Transmission, Clarity, Consistency and Coordination) strongly supports the implementation of the program GBIB is 3) aspects of attitude (attitude implementer, Support Leader, Incentives for implementing and Transparency), relatively supportive in this program, and 4) Structural Aspects of Bureaucracy (SOP and Fragmentation) still do not support the implementation of the Program snapping Sexual arousal and this Artificial Insemination.

ZusammenfassungZu den herausragenden hämatologischen Veränderungen im Rahmen von akuten und vor allem chronischen Erkrankungen der Leber gehören unterschiedlich ausgeprägte quantitative und qualitative Thrombozytendefekte. Bei chronischen Hepatopathien, insbesondere bei Leberzirrhose, wird in ca. 40-50% eine typische hepatogene Hämostasestörung angetroffen, die neben der typischen Koagulopathie in etwa 30% der Fälle durch eine Thrombozytopenie von weniger als 80000 Thrombozyten/|jl gekennzeichnet ist. Die Pathogenese der Thrombozytopathie und Thrombozytopenie ist äußerst vielschichtig. Bildungsstörungen durch Beeinträchtigung des Knochenmarks sind bedingt Ausreifungsstörungen der Megakaryozyten und Thrombozytopoese, die durch Faktoren wie Thrombopoetindysfunktion, toxische, metabolische, nutritivund medikamentös-toxische Einflüsse sowie durch Vitaminmangel bzw. Vitaminverwertungsstörungen (B12, Folsäure) hervorgerufen werden können. Metabolisch bedingte Thrombozytenmembrandefekte und ungenügende Ausstattung der Plättchen sind Ursachen für die Funktionsstörungen wie verminderte Aggregationsund Adhäsionsfähigkeit sowie Freisetzungsreaktion. Umsatzstörungen in der Zirkulation mit Thrombozytopenie können insbesondere bei chronisch aktiver Hepatitis immunologisch induziert sein, entweder durch die Präsenz von Plättchen-assoziierten IgG-Antikörpern oder Autoimmunantikörpern gegen Membranglykoproteine (GBlIb/IIla, GBIb/IX-Komplex). Weitere Pathomechanismen sind die Verbrauchsthrombozytopenie bei intravaskulären Gerinnungsprozessen im Rahmen akuter Organschädigung (Ösophagusvarizenblutung), die vermehrte Plättchensequestration und das Hypersplenie-Syndrom. Die Therapie der Thrombozytopenie ist symptomatisch und folgt bei Blutungsgefährdung den Regeln der Indikation zur Thrombozytensubstitution, im Falle nachgewiesener immunologisch induzierter Antikörper ist die Kortikoidtherapie einzusetzen. Ausschaltung metabolischer Noxen gelingt nur durch die Verbesserung der Leberfunktion.

This research was conducted in the area Dharmasraya Regency, West Sumatra Province. The purpose of this study determined : a) Method of extention in innovation adoption of artificial insemination (AI) in the cattle farm in the District Dharamasraya, and b) Social and economic factors that influence adoption of innovation Artificial Insemination (AI) in the cattle farm in the District Dharmasraya. This research used survey method and approach to the analysis of secondary data. The number of samples in this study were determined by quota sampling technique by reason of homogeneous samples, the farmers were already adopting innovations and cattle ranchers IB program participants snapping Birahi Artificial Insemination (GBIB) thus took a sample of 40 people. The data collected were primary data with the help of question naires and secondary data with the literature study and related agencies. Descriptive analysis of quantitative data which was calculated using a Likert scale. The results showed that the extension method in the adoption of IB in cattle breeding business in the Regency Dharmasraya been implemented, namely the extension methods home and farm visits with category, extension methods demonstrations by both categories, and the campaign extension methods with the medium category. Social and economic aspects such as business scale farmers, revenue, risk, active participation in the innovation adoption had no effect on aspects of the IB whereas age despite being influential in the adoption of innovation, but still in the category of less influential.

Abstract:
Although 2-guanidinobenzimidazole (GBI; CAS: 5418-95-1) is a compound of biological interest, generally there is a lack of electrochemical studies and the methods of its determination. The GBI behavior at a mercury electrode was analyzed under conditions of linear sweep voltammetry (LSV), differential pulse voltammetry (DPV), square-wave voltammetry (SWV) and square-wave stripping voltammetry (SWSV). Although GBI is electrochemically inactive at mercury electrode it adsorbs at the mercury surface and catalyzes effectively the hydrogen evolution reaction. Theoretical analysis of two possible pathways, according to which the GBI electrode mechanism can be explained, is performed. Simple analysis of peak current and potential with respect to available time window, i.e. change of frequency can be helpful in discerning the character of the recorded SW current. The established electrode mechanism is assumed to involve a preceding chemical reaction in which the adsorbed catalyst (GBIads) is protonated and the protonated form of the catalyst (GBIH+(ads)) is irreversibly reduced at potential about –1.18 V vs Ag|AgCl (citrate buffer pH 2.5). New methods of voltammetric determination of 2-guanidinobenzimidazole were developed. The detection and quantifications limits were found to be 1 × 10–7, 1 × 10–6 mol l–1 (SWV); 8 × 10–8, 9 × 10–7 mol l–1 (SWSV); 4 × 10–7, 2 × 10–6 mol l–1 (DPV) and 6 × 10–7, 3 × 10–6 mol l–1 (LSV), respectively.

H.L. Senduk merupakan salah seorang tokoh rohani yang dikenal di Indonesia, khususnya bagi kalangan Pentakosta. Sejak pertobatannya dan pengalaman akan baptisan Roh Kudus, ia mengikuti jejak gurunya Pdt. Van Gessel di Gereja Pentakosta, hingga akhirnya ia mendirikan Gereja Bethel Injil Sepenuhnya (GBIS). Seiring berjalannya waktu ia mengalami konflik dan akhirnya keluar dan mendirikan Gereja Bethel Indonesia (GBI). Kehidupan dan pelayanan H.L. Senduk sangatlah berpengaruh bagi kalangan Pentakosta di Indonesia yang terlihat dari sinode GBI merupakan sinode terbesar ketiga di Indonesia pada tahun 2021 dan terus mengalami pertumbuhan baik secara kuantitas ataupun kualitas. Pelayanan H.L. Senduk yang berdampak luar biasa ini pastinya merupakan sesuatu hal yang harus diketahui oleh banyak orang Kristen dan sangat penting untuk dipelajari. Penulisan kisah H.L. Senduk ini menggunakan metode kualitatif dengan pendekatan Naratif agar mendapatkan gambaran yang jelas mengenai kehidupan H.L. Senduk dari berbagai sumber, sehingga bisa mengetahui warisan rohani yang sangat penting bagi gereja di Indonesia. Warisan rohani H.L. Senduk pastinya akan terkait dengan pandangan teologisnya, kepemimpinan yang ia lakukan, dan pelayanannya. Pandangan teologis H.L. Senduk yang paling utama bagi gereja-gereja pentakosta adalah terkait dengan pentingnya gereja untuk terbuka dan dinamis terhadap tuntunan dan karya Roh Kudus. Kepemimpinan H.L. Senduk pastinya sangat penting untuk diteladain, karena berdampak dengan membuat sindoe GBI sebesar sekarang. Kepemimpinan yang visioner dengan memberikan teladan dan menampilkan karakter yang baik bagi seluruh kalangan. Pelayanan H.L. Senduk yang menekankan penginjilan mendorong GBI untuk terus melayani secara realistis sosial dengan relevan dan efektif. H.L. Senduk is a well-known spiritual figure in Indonesia, especially among Pentecostals. Since his conversion and experience of the baptism of the Holy Spirit, he has followed in the footsteps of his teacher Rev. Van Gessel at the Pentecostal Church, until finally he founded the Full Gospel Bethel Church (GBIS). Over time he experienced conflicts and eventually left and founded the Indonesian Bethel Church (GBI). The life and ministry of H.L. Senduk is very influential for Pentecostals in Indonesia as seen from the GBI synod, which is the third largest synod in Indonesia in 2021 and continues to experience growth both in quantity and quality. H.L. Service This amazingly impactful spoon is definitely something that many Christians should know and very important to learn. Writing the story of H.L. This Senduk uses a qualitative method with a narrative approach in order to get a clear picture of the life of H.L. Senduk from various sources, so that we can know the spiritual heritage that is very important for the church in Indonesia. The spiritual legacy of H.L. Senduk will certainly have to do with his theological views, his leadership, and his ministry. H.L.'s theological view the main thread for the Pentecostal churches is related to the importance of the church to be open and dynamic to the guidance and work of the Holy Spirit. H.L. Leadership Senduk is certainly very important to follow, because it has an impact by making the GBI sindoe as big as it is now. Visionary leadership by setting an example and displaying good character for all circles. H.L. Service Senduk which emphasizes evangelism encourages GBI to continue to serve with realistically- socially in a relevant and effective manner.

ABSTRACT Mutans streptococci are major etiological agents of dental caries, and several of their secreted products contribute to bacterial accumulation on teeth. Of these, Streptococcus mutans glucan binding protein B (GbpB) is a novel, immunologically dominant protein. Its biological function is unclear, although GbpB shares homology with a putative peptidoglycan hydrolase from S. agalactiae and S. pneumoniae, indicative of a role in murein biosynthesis. To determine the cellular function of GbpB, we used several approaches to inactivate the gene, analyze its expression, and identify interacting proteins. None of the transformants analyzed were true gbpB mutants, since they all contained both disrupted and wild-type gene copies, and expression of functional GbpB was always conserved. Thus, the inability to obtain viable gbpB null mutants supports the notion that gbpB is an essential gene. Northern blot and real-time PCR analyses suggested that induction of gbpB expression in response to stress was a strain-dependent phenomenon. Proteins that interacted with GbpB were identified in pull-down and coimmunoprecipitation assays, and these data suggest that GbpB interacts with ribosomal protein L7/L12, possibly as part of a protein complex involved in peptidoglycan synthesis and cell division.

This paper describes our InP-based heterojunction bipolar transistor (HBT) technologies and circuit design techniques for small-scale-integration (SSI) and medium-scale-integration (MSI) circuits for 40- to 100-Gbit/s-class optical communications systems. The circuits include a sub-4-ps emitter-coupled logic (ECL) gate, 100-Gbit/s selector circuit, 90-Gbit/s decision circuit, 50-Gbit/s 4:1 multiplexer (MUX) and 1:4 demultiplexer (DEMUX) ICs, aver-40-Gbit/s 16:1 MUX IC, and 40-Gbit/s full-clock-rate DEMUX with a clock and data recovery (CDR) circuit. This paper demonstrates that InP-based HBT technologies and our circuit design techniques are attractive for fabricating ultrahigh-speed SSI circuits with data rates approaching 100 Gbit/s and low-power MSI circuits with data rates of over 40 Gbit/s.

Abstract. Big data are significant for quantitative analysis and contribute to data-driven scientific research and discoveries. Here a brief introduction is given to the Geobiodiversity Database (GBDB), a comprehensive stratigraphic and palaeontological database, and its data. The GBDB includes abundant geological records from China and has supported a series of scientific studies on the Paleozoic palaeogeography and tectonic and biodiversity evolution of China. The data that the GBDB has including those that are newly collected are described in detail; the statistical results and structure of the data are given. A comparison between the GBDB; the largest palaeobiological database, the Paleobiology Database (PBDB); and the geological rock database Macrostrat is drawn. The GBDB and other databases are complementary in palaeontological and stratigraphic research. The GBDB will continually provide users access to detailed palaeontological and stratigraphic data based on publications. Non-structured data of palaeontology and stratigraphy will also be included in the GBDB, and they will be organically correlated with the existing data of the GBDB, making the GBDB more widely used for both researchers and anyone who is interested in fossils and strata. Atlas of Living Australia (ALA) (https://www.ala.org.au/) is the Global Biodiversity Information Facility (GBIF) node of Australia. They developed an open and free platform for sharing and exploring biodiversity data. All the modules are publicly available for reuse and customization on their GitHub account (https://github.com/AtlasOfLivingAustralia). GBIF Benin, hosted at the University of Abomey-Calavi, has published more than 338 000 occurrence records from 87 datasets and 2 checklists. Through the GBIF Capacity Enhancement Support Programme (https://www.gbif.org/programme/82219/capacity-enhancement-support-programme), GBIF Benin, with the help of GBIF France, is in the process of deploying the Beninese data portal using the GBIF France back-end architecture. GBIF Benin is the first African country to implement this module of the ALA infrastructure. In this presentation, we will show you an overview of the registry and the occurrence search engine using the Beninese data portal. We will begin with the administration interface and how to manage metadata, then we will continue with the user interface of the registry and how you can find Beninese occurrences through the hub.

Using our 0.2 and 0.3 μm AlGaAs/GaAs/AlGaAs quantum well HEMT technology, we have developed a chip set for 20–40 Gbit/s fiber-optical digital transmission systems. In this paper we describe nine analog and digital receiver ICs: a 22 GHz high-gain transimpedance amplifier, a 20 Gbit/s OEIC front-end optical receiver, a 25 Gbit/s automatic-gain-control amplifier, a limiting amplifier with a differential gain of 26 dB and a bandwidth of 27.7 GHz, a 20–40 Gbit/s clock recovery, a 20 Gbit/s low-power Master-Slave-D-Flipflop with 24 mW power dissipation, a parallel data decision and a 1:4 demultiplexer, both for bit rates of 40 Gbit/s, and a 30 GHz static frequency divider, respectively. All chips were characterized on wafers with 50 Ω coplanar test probes.

This research was motivated by the difference in perceptions among GBIS officials of the Central Java Regional Council about the Covid-19 pandemic. This difference has implications for ecclesiastical service. The research problem in this research is what is the perception of the Central Java Regional Council GBIS officials about the Covid-19 pandemic and its implications for ecclesiastical services? Meanwhile, the purpose of this study is to explain the perceptions of the Central Java Regional Council GBIS officials about the Covid-19 pandemic and its implications for ecclesiastical services. From the research, it was concluded that the perceptions of the Central Java Regional Council GBIS officials regarding the Covid-19 pandemic were good and adequate. The implication is in ecclesiastical services, GBIS officials in the midst of this pandemic, continue to emphasize that hospitality as a pastor must be carried out wisely and correctly so as not to exacerbate Covid-19 transmission without having to lose the human side which of course still requires love and hospitality.Penelitian ini dilatarbelakangi oleh adanya perbedaan persepsi di kalangan para pejabat GBIS Majelis Daerah Jawa Tengah tentang pandemi Covid-19. Perbedaan tersebut berimplikasi bagi pelayanan gerejawi. Problem research dalam penelitian ini adalah bagaimanakah persepsi para pejabat GBIS Majelis Daerah Jawa Tengah tentang pandemi Covid-19 dan implikasinya bagi pelayanan gerejawi? Sedangkan tujuan penelitian ini adalah menjelaskan persepsi para pejabat GBIS Majelis Daerah Jawa Tengah tentang pandemik Covid-19 dan implikasinya bagi pelayanan gerejawi. Dari penelitian tesrebut ditarik kesimpulan bahwa persepsi para pejabat GBIS Majelis Daerah Jawa Tengah berkenaan dengan pandemi covid-19 caik dan memadai. Implikasinya dalam pelayanan gerejawi, para pejabat GBIS di tengah masa pandemi ini, tetap mengetrapkan hospitalitas sebagai gembala sidang harus dilakukan dengan bijak dan benar agar tidak memerparah penularan Covid-19 tanpa harus menghilangkan sisi kemanusiaan yang tentunya tetap membutuhkan kasih dan keramahtamahan.

GBIF Benin, hosted at the University of Abomey-Calavi, has published more than 338,000 occurrence records in 87 datasets and checklists. It has been a Global Biodiversity Information Facility (GBIF) node since 2004 and is a leader in several projects from the Biodiversity Information for Development (BID) programme. GBIF facilitates collaboration between nodes at different levels through its Capacity Enhancement Support Programme (CESP) [https://www.gbif.org/programme/82219/capacity-enhancement-support-programme]. One of the actions included in the CESP guidelines is called ‘Mentoring activities’. Its main goal is the transfer of knowledge between partners such as information, technologies, experience, and best practices. Sharing architecture and development is the key solution to solve some technical challenges or impediments (hosting, staff turnover, etc.) that GBIF nodes could face. The Atlas of Living Australia (ALA) team developed a functionality called ‘data hub’. It gives the possibility to create a standalone website with a dedicated occurrence search engine that seeks among a range of data (e.g. specific genus, geographic area). In 2017, GBIF Benin and GBIF France wanted to strengthen their partnership and started a CESP project. One of the core objectives of this project is the creation of the Atlas of Living Benin using ALA modules. GBIF France developers, with the help of the GBIF Benin team, are in the process of configuring a data hub that will give access to Beninese data only, while at the same time Atlas of Living France will give access to French data only. Both data portals will use the same back end, therefore the same databases. Benin is the first African GBIF node to implement this kind of infrastructure. On this poster, we will present the Atlas of Living Benin specific architecture and how we have managed to distinguish data coming from Benin and coming from France.

However, the mechanisms by which secreted and/or cell-associated glucan-binding proteins (Gbps) produced byS. mutansparticipate in biofilm growth remain to be elucidated. In this study, we further investigate GbpB, an essential immunodominant protein with similarity to murein hydrolases. A conditional knockdown mutant that expressedgbpBantisense RNA under the control of a tetracycline-inducible promoter was constructed in strain UA159 (UACA2) and used to investigate the effects of GbpB depletion on biofilm formation and cell surface-associated characteristics. Additionally, regulation ofgbpBby the two-component system VicRK was investigated, and phenotypic analysis of avicKmutant (UAvicK) was performed. GbpB was directly regulated by VicR, and several phenotypic changes were comparable between UACA2 and UAvicK, although differences between these strains existed. It was established that GbpB depletion impaired initial phases of sucrose-dependent biofilm formation, while exogenous native GbpB partially restored the biofilm phenotype. Several cellular traits were significantly affected by GbpB depletion, including altered cell shape, decreased autolysis, increased cell hydrophobicity, and sensitivity to antibiotics and osmotic and oxidative stresses. These data provide the first experimental evidence for GbpB participation in sucrose-dependent biofilm formation and in cell surface properties.

Parameter utama yang digunakan untuk mendapatkan informasi-informasi yang diinginkan adalah nama spesies tumbuhan. Hasil penelitian menunjukkan bahwa GBIF dan EOL dapat dijadikan sebagai sumber infomasi tumbuhan obat Indonesia. We present the outline of the InP HEMT IC technology. This technology realizes InP HEMT digital ICs for 40-Gbit/s optical fiber communication systems through the integration of 0.1-μm-gate-length HEMTs, vertical diodes, capacitors, and WSiN resistors with two level interconnections. This paper describes the high-speed digital IC circuit design and fabrication in InP HEMT technology for 40-Gbit/s/channel optical communication systems. Some results on InP HEMTs' reliability are also covered. Basic circuit design techniques utilizing SCFL topology and fundamental circuit elements of the selector and D-type flip-flop are discussed in detail. The basic digital ICs of MUX, D-FF, and DEMUX ICs fabricated with 0.1-μm-gate InP HEMTs successfully operated up to 50 Gbit/s in the packaged modules. These IC modules offer large speed margins for the 43-Gbit/s OTU-3 data rate. In order to develop cost-effective optical transmitters and receivers, we designed a PLL-based CDR with a full-rate architecture. The fully monolithic integrated CDR exhibited error-free operation for 231-1 PRBS data signal at the OTU-3 bit rate of 43.0184 Gbit/s. Four-bit MUX and DEMUX ICs are other key components, and could be implemented by using InP HEMT technology. Additionally, we describe InP-IC fabrication technology with two-level inter-connection. This is already fully matured for 40-Gbit/s SSI fabrication. The uniform FET characteristics and high-yield passive component fabrication technologies support this degree of maturity. InP HEMT lifetime is 107 hours at l00°C. These results prove the InP HEMT IC fabrication technology presented here, to be highly reliable. These investigation show that robust performance and yield when realizing SSI and MSI 40-Gbit/s functions.

This paper describes recent advances in high-speed digital IC design technologies based on GaAs MESFETs for future high-speed optical communications systems. We devised new types of a data selector and flip-flops, which are key elements in performing high-speed digital functions (signal multiplexing, decision, demultiplexing, and frequency conversion) in front-end transmitter/receiver systems. Incorporating these circuit design technologies with state-of-the-art 0.12 μm gate-length GaAs MESFET process, we developed a DC-to-44-Gbit/s 2:1 data multiplexer IC, a DC-to 22-Gbit/s static decision IC, and a 20-to-40-Gbit/s dynamic decision IC. The fabricated ICs demonstrated record speed performances for GaAs MESFETs. Although further operating speed margin is still required, the GaAs MESFET is a potential candidate for 20- to 40-Gbit/s class applications.

ABSTRACT Streptococcus mutans, the primary etiological agent of dental caries, produces several activities that promote its accumulation within the dental biofilm. These include glucosyltransferases, their glucan products, and proteins that bind glucan. At least three glucan binding proteins have been identified, and GbpB, the protein characterized in this study, appears to be novel. The gbpB gene was cloned and the predicted protein sequence contained several unusual features and shared extensive homology with a putative peptidoglycan hydrolase from group B streptococcus. Examination of gbpB genes from clinical isolates ofS. mutans revealed that DNA polymorphisms, and hence amino acid changes, were limited to the central region of the gene, suggesting functional conservation within the amino and carboxy termini of the protein. The GbpB produced by clinical isolates and laboratory strains showed various distributions between cells and culture medium, and amounts of protein produced by individual strains correlated positively with their ability to grow as biofilms in an in vitro assay.

Physical dormancy (PY) is a phenomenon wherein seed coats are impermeable to water. This feature prevents immediate germination in seeds, therefore considered as an adaptive trait in species of Mediterranean and tropical ecosystem, where rainy season is the most favorable time for germination. However, using dataset available for Fabaceae collected from global biodiversity information facility (GBIF), the largest family with PY, recent studies have provided evidence contrasting this assertion. This viewpoint has arisen owing to the fact that the data were gleaned by georeferencing the Fabaceae species distribution from GBIF, which is under-represented for the tropical vegetation. This is similar to other reports available in other plant and animal distribution models, where GBIF data is not an accurate representation of distribution. A closer inspection of the data available in literature suggests that using GBIF database alone to map the distribution of Fabaceae species represents the extreme end of biased data causing misperception and could mislead the scientific community, particularly ecologists, conservationists and/or policy makers.

Ghana, like many African countries, is currently facing power supply shortage, which has led to load shedding. To minimize the impact of the power crisis, options such as diesel and petrol generators, grid-charged battery-inverter systems (GBIS), and solar PV with battery storage (SPVS) have been used in residential and nonresidential contexts. In this paper, we develop analytical models to conduct a technical and economic comparison of GBIS and SPVS systems. Using average electricity tariff of $0.186 for residential sector (excluding lifeline customers) we show that although initial cost of SPVS is higher, it costs 30% less than GBIS. We also show that losses associated with the GBIS are as high as 42% when viewed from a systems perspective and that some of its costs are externalized. We conclude by commending the Ghana Government’s initiative of rolling out 200,000 residential rooftop solar systems and recommend an increase in system capacities as well as a similar programme for nonresidential facilities.

Im Rahmen einer weltweiten Forschungskooperation wird GBIF von derzeit 47 Staaten und 29 internationalen Organisationen als Mitgliedern getragen, die sich alle zur freien Bereitstellung digitaler Biodiversitätsdaten nach gemeinsamen Standards über eigene, dafür selbst einzurichtende Datenknoten verpflichtet haben. Das internationale Vorhaben wird durch einen Aufsichtsrat mit Vertretern aller Mitgliedsstaaten und –organisationen geleitet, dessen Arbeit durch mehrere Komitees und Ausschüsse unterstützt wird. Das seit 2002 in Kopenhagen, Dänemark, angesiedelte GBIF-Sekretariat betreibt den Aufbau des internationalen GBIF Portals (www.gbif.net) und unterstützt koordinierend die Aktivitäten der einzelnen Mitglieder, die sich auf vier Programmbereiche erstrecken: Standardisierung und Verknüpfung von Datenbanken (DADI), Digitalisierung von Daten zu Sammlungsobjekten (DIGIT), Katalog der bekannten Organismennamen (ECAT), sowie Ausbildung und Öffentlichkeitsarbeit (OCB). ABSTRACT The initial infection of children by Streptococcus mutans, the main pathogen of dental caries, depends on the ability of S. mutans to adhere and accumulate on tooth surfaces. These processes involve the adhesin antigen I/II (AgI/II), glucosyltransferases (GTF) and glucan-binding protein B (GbpB), each a target for anticaries vaccines. The salivary immunoglobulin A (IgA) antibody responses to S. mutans antigens (Ags) were characterized in 21 pairs of 5- to 13-month-old children. Pairs were constructed with one early S. mutans-infected and one noninfected child matched by age, racial background, number of teeth, and salivary levels of IgA. Specific salivary IgA antibody response and S. mutans infection levels were then measured during a 1-year follow-up. Robust responses to S. mutans were detected from 6 months of age. Salivary IgA antibody to AgI/II and GTF was commonly detected in salivas of all 42 children. However, GbpB-specific IgA antibody was seldom detected in the subset of infected children (38.1% at baseline). In contrast, most of the subset of noninfected children (76.2%) showed GbpB-reactive IgA antibody during the same period. Frequencies of GbpB responses increased with age, but differences in intensities of GbpB-IgA antibody reactions were sustained between the subsets. At baseline, GbpB-reactive IgA antibody accounted for at least half of the total salivary IgA S. mutans-reactive antibody in 33.3 and 9.5% of noninfected and infected children, respectively. This study provides evidence that a robust natural response to S. mutans Ags can be achieved by 1 year of age and that IgA antibody specificities may be critical in modulating initial S. mutans infection.

ABSTRACT Glucan-binding protein B (GbpB) from Streptococcus mutans has been shown to induce protective immunity to dental caries in experimental models. Having recently sequenced the gbpB gene, our objective in this study was to identify immunogenic regions within the GbpB sequence for use in subunit vaccines. Potential regions of immunogenicity were sought by use of a matrix-based algorithm (EpiMatrix) to estimate the binding characteristics of peptides derived from the GbpB sequence by using a database of known major histocompatibility complex class II binding alleles. Screening the entire sequence revealed several peptides with estimated high binding probabilities. Two N-terminal 20-mer peptides (SYI and QGQ) subtending two of these regions were synthesized. A preliminary experiment, in which these peptides were synthesized in the multiple antigenic peptide format and were used to subcutaneously immunize Sprague-Dawley rats twice at a 21-day interval, revealed that the SYI peptide induced a higher percentage of responses to the inciting peptide as well as to intact GbpB, as measured by enzyme-linked immunosorbent assay. The effect of immunization with the SYI peptide construct on the cariogenicity of S. mutans was then investigated by immunizing weanling Sprague-Dawley rats twice at a 9-day interval with SYI or with phosphate-buffered saline. All rats were then orally infected with S. mutans strain SJ. After a 78-day infection period, the SYI-immunized groups had significant reductions in dental caries on both smooth and occlusal surfaces compared with the sham-immunized group. Thus, these experiments indicated that at least one linear sequence, derived from the N-terminal third of GbpB, was sufficiently immunogenic to induce a protective immune response in this experimental rat model for dental caries.

This paper demonstrates why VCSEL-based transmitter systems are not able to capitalize on higher-order PAM modulation formats (such as PAM-4) to push the link data rate towards 100 Gbit/s with BER of better than 10−5. First, the non-linear and biasing-dependent VCSEL behavior is analyzed in great detail to prove why state-of-the-art VCSELs with linear equalization are not able to reach these bandwidth targets. Consequently, based on an enhanced VCSEL model, a digital non-linear transmit equalizer is proposed to overcome voltage-dependent relaxation frequency shifts as well as data-dependent VCSEL bandwidth variations. The proposed digital non-linear equalizer is compared with a standard linear FFE structure using PAM-4 transmission experiments of 100 Gbit/s and beyond. This way, the proposed non-linear equalizer reveals a BER improvement of more than 30-fold compared to a linear FFE at a data rate of 80 Gbit/s. Finally, at 112 Gbit/s PAM-4 the non-linear equalizer was successfully demonstrated at a BER of 1.5·10−7.

Introduction: The incidence and prevalence of patients with Myelodysplastic Syndromes (MDS) are continuously rising due to population ageing and diagnostic advances. Consequently, the appropriate use of health care resources and assessment of their impact on relevant outcomes are of growing interest. Several evidence-based guidelines and recommendations (G&Rs) have been published for adult MDS patients. However, publishing G&Rs does not necessarily translate into better quality of care. Several studies have shown that MDS patients are not always treated according to published G&Rs, but systematic investigations have not been done so far. To this aim, we initiated the I-CARE for MDS study with the objectives to i) define relevant guideline-based indicators (GBIs) as measurable elements of practice performance for appropriate care, ii) assess the level of adherence and reasons for non-adherence to GBIs and iii) investigate the impact of adherence/non-adherence to GBIs on MDS relevant outcomes. Here we present results of the development of GBIs for appropriate care in adult MDS patients. Methods: We systematically screened G&Rs from cooperative MDS groups as well as cancer care accreditation/certification programs (table 1). All relevant information was extracted by a structured procedure and summarized as candidate GBIs in a handbook (table 2). We applied a RAND technique with a two-step DELPHI rating procedure to find an expert consensus for the clinically most relevant GBIs (Coulter J et al, J Health Serv Res Policy, 2018). The expert panel group (EPGs) members consisted of 17 internationally acknowledged MDS experts, 7 additional health-professionals (3 nurses, 1 pharmacologist, 1 physiotherapist, 1 psychologist and 1 epidemiologist) and 3 patient advocates. Candidate GBIs were rated using a 9-point Likert-like scale for Relevance, Understandability, Measurability, Behaviorability, Attainability (RUMBA-criteria) and ranked according to the agreement score, defined as % of all EPG members scoring all RUMBA-criteria in the first tertile (9, 8, or 7). Results: We extracted 61 candidate GBIs from G&Rs for the three domains Diagnostics (n=23), Treatment (n=21) and Provider characteristics (n=17). 18 and 16 candidate GBIs were excluded after the first and second round, respectively, remaining with a set of 27 GBIs (figures 1-3). Twelve GBIs were selected for Diagnostics (figure 1). Patient reported outcomes (PROs), Toxicity assessment (TAs) and Geriatric assessment were excluded as there was no agreement among experts on appropriate measurement instruments and concerns for practicability in daily routine. Eight GBIs were selected for Treatment (figure 2). Iron-chelation and Immunosuppressive treatment, even though mentioned in most G&Rs, were excluded, as there were concerns with regard to controversial indications, side effects and benefit for only a minority of patients. Seven GBIs were selected for Provider characteristics (figure 3), which will be generally fulfilled by accreditation or certification of institutions. Agreement scores were generally higher in this domain and required a more stringent threshold for selection of GBIs. Conclusions: Here we report on the development of relevant GBIs for the management of adult MDS patients covering the domains of Diagnostics, Treatment and Provider characteristics. Our preliminary set GBIs represents the most comprehensive integration of current practice based G&Rs and were developed by a structured consensus process involving internationally acknowledged experts. We identified important shortcomings in standardisation, measurability and practicability, especially for PROs and TAs, which asks for improvements in the future. Even though PROs and TAs are generally acknowledged endpoints for efficacy and safety, respectively, they are yet not standardised outside clinical trials or validated to inform routine MDS care. Our preliminary set of GBIs will be tested for applicability/operability as well as validated for their potential impact on relevant outcomes in MDS cohorts. They will eventually enable to systematically monitor, compare and optimize appropriateness of health care provided to MDS patients in everyday clinical practice. As such, GBIs will be important for the realization of a common standard for value-based medicine in economically driven health care systems with limiting resources. γ-Butyrobetaine (GBB) is a precursor in the biosynthesis of carnitine, which plays an important role in the β-oxidation of fatty acids, and is converted to carnitine by γ-butyrobetaine dioxygenase (BBD) predominantly in liver. We investigated the molecular mechanism of hepatic uptake of GBB in rat hepatocytes. Cellular localization of rat Octn2 (rOctn2:Slc22A5) was studied by Western blot analysis. Uptake of deuterated GBB (d3-GBB) was examined in HEK293 cells expressing rOctn2 (HEK293/rOctn2) and freshly isolated rat hepatocytes. d3-GBB was quantified by use of liquid chromatography-tandem mass spectrometry. Western blot analysis demonstrated an expression of OCTN2 protein in hepatic basolateral membrane but not in bile canalicular membrane fraction. Furthermore, we found that d3-GBB was taken up by rOctn2 in an Na+-dependent manner with Km value of 13 μM. The apparent Km value for d3-GBB transport in freshly isolated rat hepatocytes was 9 μM. d3-GBB uptake by the rat hepatocytes was inhibited by γ-aminobutyric acid (GABA) to 30% of the control, whereas it was inhibited by carnitine to 62% of the control, even at 500 μM. Furthermore, d3-GBB uptake by rat hepatocytes was decreased by 45% with rat Gat2 (Slc6A13, a major liver GABA transporter) silenced by the microRNA method. Accordingly, the present study clearly demonstrates that GBB is taken up by hepatocytes for carnitine biosynthesis not only via Octn2 but also via the GABA transporter, possibly Gat2.

There is currently an increasing demand for Building Information Modelling (BIM) to be integrated into green projects. BIM is able to evaluate green building assessment tools by digitising and assessing buildings during their development stage. In Malaysia, the Green Building Index (GBI) promotes sustainability in the built environment. Six categories of criteria will be assessed in three of the certification stages. This study focuses on how BIM applications can digitise the GBI criteria for GBI processes. It examines BIM uses and tools of each GBI credit and defines the responsibility and role of construction stakeholders in using BIM to examine the assessment methods used for new non-residential building construction based on GBI. The primary method for data collection is the focus group interview which involves the groups of stakeholders involved in a BIM and GBI project. The result of the BIM–GBI assessment method showed that BIM could digitise and assess 25 credits in Design Assessment (DA), which can achieve 55 points of the total 100 points. This study helps stakeholders define the design team and facility manager’s roles to obtain GBI certification and maintain the certification during the building’s operation stage.

The Drosophila BMP5/6/7/8 homolog, glass bottom boat (gbb), has been shown to be involved in proliferation and vein patterning in the wing disk. To better understand the roles for gbb in wing development, as well as its relationship with the Drosophila BMP2/4 homolog decapentaplegic (dpp), we have used clonal analysis to define the functional foci of gbb during wing development. Our results show that gbb has both local and long-range functions in the disk that coincide both spatially and functionally with the established functions of dpp, suggesting that both BMPs contribute to the same processes during wing development. Indeed, comparison of the mutant phenotypes of dpp and gbb hypomorphs and null clones shows that both BMPs act locally along the longitudinal and cross veins to affect the process of vein promotion during pupal development, and long-range from a single focus along the A/P compartment boundary to affect the processes of disk proliferation and vein specification during larval development. Moreover, we show that duplications of dpp are able to rescue many of the phenotypes associated with gbb mutants and clones, indicating that the functions of gbb are at least partially redundant with those of dpp. While this relationship is similar to that described for dpp and the BMP screw (scw) in the embryo, we show that the mechanisms underlying both local and long-range functions of gbb and dpp in the wing are different. For the local foci, gbb function is confined to the regions of the veins that require the highest levels of dpp signaling, suggesting that gbb acts to augment dpp signaling in the same way as scw is proposed to do in the embryo. However, unlike scw-dependent signals in the embryo, these gbb signals are not transduced by the Type I receptor saxophone (sax), thus, the cooperativity between gbb and dpp is not achieved by signaling through distinct receptor complexes. For the long-range focus along the A/P compartment boundary, gbb function does not appear to affect the high point of the dpp gradient, but, rather, appears to be required for low points, which is the reciprocal of the relationship between dpp and scw in the embryo. Moreover, these functions of gbb also do not require the Type I receptor sax. Given these results, we conclude that the relationships between gbb and dpp in the wing disk represent novel paradigms for how multiple BMP ligands signal during development, and that signaling by multiple BMPs involves a variety of different inter-ligand relationships that depend on the developmental context in which they act.

Purposes. This research explores the game-based intelligent test (GBIT), predicts the possibilities of Mini-Mental State Examination (MMSE) scores and the risk of cognitive impairment, and then verifies GBIT as one of the reliable and valid cognitive assessment tools. Methods. This study recruited 117 elderly subjects in Taiwan (average age is 79.92 ± 8.68 , average height is 156.91 ± 8.01 , average weight is 59.14 ± 9.67 , and average MMSE score is 23.33 ± 6.16 ). A multiple regression model was used to analyze the GBIT parameters of the elderly’s reaction, attention, coordination, and memory to predict their MMSE performance. The binary logistic regression was then utilized to predict their risk of cognitive impairment. The statistical significance level was set as α = 0.05 . Results. Multiple regression analysis showed that gender, the correct number of reactions, and the correct number of memory have a significantly positive predictive power on MMSE of the elderly ( F = 37.60 , R 2 = 0.69 , and p < 0.05 ). Binary logistic regression analysis noted that the correct average number of reactions falls by one question, and the ratio of cognitive dysfunction risk increases 1.09 times ( p < 0.05 ); the correct average number of memory drops by one question, the ratio of cognitive dysfunction risk increases 3.76 times ( p < 0.05 ), and the overall model predictive power is 88.20% (sensitivity: 84.00%; specificity: 92.30%). Conclusions. This study verifies that GBIT is reliable and can effectively predict the cognitive function and risk of cognitive impairment in the elderly. Therefore, GBIT can be used as one of the feasible tools for evaluating older people’s cognitive function.

Atlas of Living Australia (ALA) (https://www.ala.org.au/) is the Global Biodiversity Information Facility (GBIF) node of Australia. In 2010, they launched an open and free platform for sharing and exploring biodiversity data. Thanks to this new infrastructure, they have been able to drastically increase the number of occurrences published through the GBIF.org . In order to help other GBIF nodes or institutions, they made all of their modules publicly available for reuse and customization through GitHub (https://github.com/AtlasOfLivingAustralia). Since 2013, the community created by developers interested by ALA tools, organized, with the help of GBIF, 8 technical workshops around the world. These workshops helped the launch of at least 13 data portals. The last training session, funded through the GBIF Capacity Enhancement Support Programme (https://www.gbif.org/programme/82219/capacity-enhancement-support-programme), was been attended by 23 participants from 19 countries on 6 continents. Moreover, on the new GBIF website, a section has been dedicated to this programme (https://www.gbif.org/programme/82953/living-atlases), the Living Atlases community official website has been launched in 2017 (https://living-atlases.gbif.org) and the technical documentation has been improved and translated in several languages. All of these achievements would not have been possible without a huge effort from the ALA developer community. After a brief introduction of the Living Atlases community, we will present you the work done by ALA to simplify the process of getting a living atlas up and running. We will also show you how ALA developers managed to help the community members to create their own version by performing simple HTML/CSS customizations.

An initial approach to explore this question would be to assess the native secretory immunity emerging as a consequence ofStreptococcus mutansinfection. To this end, we analyzed salivary immunoglobulin A (IgA) antibody to mutans streptococcal glucosyltransferase (Gtf) and glucan binding protein B (GbpB) and to domains associated with enzyme function and major histocompatibility complex (MHC) class II binding in two experiments. Salivas were collected from approximately 45-day-old Sprague-Dawley rats, which were then infected withS. mutansSJ32. Infection was verified and allowed to continue for 2 to 2.5 months. Salivas were again collected following the infection period. Pre- and postinfection salivas were then analyzed for IgA antibody activity using peptide- or protein-coated microsphere Luminex technology.S. mutansinfection induced significant levels of salivary IgA antibody to Gtf (P< 0.002) and GbpB (P< 0.001) in both experiments, although the levels were usually far lower than the levels achieved when mucosal immunization is used. Significantly (P< 0.035 toP< 0.001) elevated levels of postinfection salivary IgA antibody to 6/10 Gtf peptides associated with either enzyme function or MHC binding were detected. The postinfection levels of antibody to two GbpB peptides in the N-terminal region of the six GbpB peptides assayed were also elevated (P< 0.031 andP< 0.001). Interestingly, the patterns of the rodent response to GbpB peptides were similar to the patterns seen in salivas from young children during their initial exposure toS. mutans.Thus, the presence of a detectable postinfection salivary IgA response to mutans streptococcal virulence-associated components, coupled with the correspondence between rat and human mucosal immune responsiveness to naturally presented Gtf and GbpB epitopes, suggests that the rat may be a useful model for defining mucosal responses that could be expected in humans. Under controlled infection conditions, such a model could prove to be helpful for unraveling relationships between the host response and oral biofilm development.

The large bandwidth of the 60 GHz frequency band enables wireless short-range applications with data rates of several Gbit/s. The German project EASY-A has focused on early prototype implementations for selected applications, although no generally accepted 60 GHz standard has been available at the time. The implementations are based on application-oriented physical layer designs and link-budget investigations that account for the scenario-specific channel characteristics and for different integration technologies. This paper discusses the results of these investigations and details the hardware implementation of the digital baseband processing that relies on considerable parallelization. The link-budget results show that SiGe technology allows for 1 Gbit/s at a range of 3.5 m in non-line-of-sight environments, while up to 10 Gbit/s are feasible at more than 1 m in case of strong line of sight.

Abstract We have isolated mutations in the Drosophila melanogaster gene glass bottom boat (gbb), which encodes a TGF-β signaling molecule (formerly referred to as 60A) with highest sequence similarity to members of the bone morphogenetic protein (BMP) subgroup including vertebrate BMPs 5-8. Genetic analysis of both null and hypomorphic gbb alleles indicates that the gene is required in many developmental processes, including embryonic midgut morphogenesis, patterning of the larval cuticle, fat body morphology, and development and patterning of the imaginal discs. In the embryonic midgut, we show that gbb is required for the formation of the anterior constriction and for maintenance of the homeotic gene Antennapedia in the visceral mesoderm. In addition, we show a requirement for gbb in the anterior and posterior cells of the underlying endoderm and in the formation and extension of the gastric caecae. gbb is required in all the imaginal discs for proper disc growth and for specification of veins in the wing and of macrochaete in the notum. Significantly, some of these tissues have been shown to also require the Drosophila BMP2/4 homolog decapentaplegic (dpp), while others do not. These results indicate that signaling by both gbb and dpp may contribute to the development of some tissues, while in others, gbb may signal independently of dpp.

Rat Serum Carboxylesterase Partly Hydrolyses Gamma-Butyrobetaine EstersAlthough described some time ago, gamma-butyrobetaine esters and related compounds have not gained much attention from researchers, and their physiological function remains obscure. Formerly we detected GBB-esterase enzymatic activity in rat blood serum using phenylated gamma-butyrobetaine as an artificial substrate of the enzyme and HPLC. The aim of the present work was to develop an assay that would enable spectrophotometric or colorimetric determination of the reaction products of GBB-esterase activity and to reveal individual proteins performing GBB-esterase activity in rat blood serum. For this purpose gamma-butyrobetaine 1-naphthyl ester was synthesised. Hydrolysis of this ester releases 1-naphthol, which increases the optical absorbance at 322 nm. We have shown that the enzymatic hydrolysis of GBB 1-naphthyl ester to 1-naphthol in rat blood serum is due to GBB-esterase activity. An attempt was done to purify the enzyme from rat blood serum. By combining DEAE Sepharose at pH 4.2 and affinity chromatography with procainamide we achieved a 68-fold enrichment of GBB-esterase activity in our preparations. Abstract:
Gangguan tidur adalah masalah yang terjadi pada kualitas, kuantitas, dan waktu tidur, atau gabungan ketiganya, sehingga berdampak tidak baik untuk kesehatan manusia. Secara fisiologis, Tidur diatur oleh sistem ARAS (Ascending Circulatory Activity System) dan dipengaruhi oleh neurotransmiter, salah satunya adalah triptofan. Biji labu merupakan bahan alam dengan kandungan triptofan yang tinggi sehingga dapat meningkatkan kualitas tidur dengan memproduksi serotonin dan melatonin. Penelitian merupakan eksperimen semu dengan desain rancangan berupa Post Test Only Controlled Group Design. Sampel penelitian sebanyak 25 ekor mencit yang dipilih secara acak dan dibagi dalam lima kelompok yaitu kontrol negatif (K0) diberikan NaCl 0,9%, kontrol positif (K1) diberikan fenobarbital 40 mg/kgbb, kelompok perlakuan (P1, P2, P3) masing-masing diberikan ekstrak etanol biji labu 40 mg/gbb, 80 mg/gbb, 120 mg/gbb, disertai induksi fenobarbital 40 mg/kgbb. Hasil diukur berdasarkan waktu tidur mencit dimulai saat mencit kehilangan reflek righting diakhiri ketika mencit dapat melakukan reflek righting kembali. Terdapat ada peningkatan lama waktu tidur mencit yang diberikan ekstrak etanol biji labu pada dosis 40 mg/gBB, 80 mg/gBB, dan 120 mg/gBB dengan waktu tidur tertinggi pada dosis 120 mg/gBB yaitu sebesar 95,9 menit. Kesimpulannya, ekstrak etanol biji labu memberikan efek dalam meningkatkan lama waktu tidur mencit.

Aspirin adalah golongan OAINS dengan efek samping yang paling sering adalah ulkus lambung. Kunyit (Curcuma Domestica val) dan madu memilki aktivitas dalam mencegah ulkus lambung. Tujuan penelitian ini adalah menentukan apakah kombinasi ekstrak kunyit dan madu mempunyai efektivitas yang lebih baik dalam mengurangiulkus lambung akibat pemberian aspirin dibandingkan ekstrak kunyit atau madu saja pada mencit BALB/c secara mikroskopis. Penelitian eksperimental post test only control group design telah dilakukan dari Januari 2016 sampai Januari 2017 menggunakan 30 ekor mencit yang semua diberikan aspirin 5.2 mg/gBB untuk menyebabkan ulkus pada lambung, kemudian dibagi menjadi kelompok kontrol (K) hanya diberi aspirin, perlakuan 1 (P1) diberikan larutan ekstrak kunyit dosis 10 mg/gBB, (P2) madu 0.04 ml/gBB, (P3) kunyit 10 mg/gBB + madu 0.04 ml/gBB, (P4) Kunyit 30 mg/gBB + madu 0.12 ml/gBB, diberikan selama 3 hari per oral. Hari ke-4 mencit di eutanasia menggunakan eter, kemudian diperiksa secara mikroskopis. Abstract 100 Gbit/s wireless communication protocol processing stresses all parts of a communication system until the outermost. The efficient use of upcoming 100 Gbit/s and beyond transmission technology requires the rethinking of the way protocols are processed by the communication endpoints. This paper summarizes the achievements of the project End2End100. We will present a comprehensive soft real-time stream processing approach that allows the protocol designer to develop, analyze, and plan scalable protocols for ultra high data rates of 100 Gbit/s and beyond. Furthermore, we will present an ultra-low power, adaptable, and massively parallelized FEC (Forward Error Correction) scheme that detects and corrects bit errors at line rate with an energy consumption between 1 pJ/bit and 13 pJ/bit. The evaluation results discussed in this publication show that our comprehensive approach allows end-to-end communication with a very low protocol processing overhead.

This study aims to analyze end user computing satisfaction on the GBI Keluarga Allah’s application and its impact on congregational participation in worship, growth in the cell community and church ministry. In this case, end user computing satisfaction is measured by five indicators, it is content, accuracy, format, timeliness and ease of use of the application. In this study, 100 questionnaires were distributed and the final sample obtained was 97 respondents, who are members of the GBI Keluarga Allah who use GBI KA application. The results of the analysis show that the end user computing satisfaction of the GBI Keluarga Allah congregation is quite high and affects the level of participation of the congregation in worship. This end user computing satisfaction also affects the participation of the members of the GBI Keluarga Allah in participating in cell groups and services, although there are other factors that affect the level of participation of the congregation in these cell groups and services besides the end user computing satisfaction of the congregation on the application of GBI Keluarga Allah.

This paper is written under the purpose of exploring in details about one of popular language teaching methods namely Genre based Instruction (GBI). GBI as how it is today is very famous to be applied for teaching writing where the same case occurs in Indonesia in that GBI is implemented into the English classroom for teaching English writing. GBI is uniquely powerful in its substance since it not only focuses on the process of writing itself, but also stresses on the quality of writing work. This teaching method makes students learn a language as how it is used in a particular social setting because that is the obvious phenomenon that happens when people use a language. Therefore, this teaching method underlies its essence on teaching language meaningfully based on the social staging. In this paper, there are several elements which are discussed regarding GBI. They encompass its theories including linguistic principles and learning principles, both pedagogical and practical views in the classroom, and a brief practical view about GBI in Indonesia.

In this study we describe areas of endemism from Brazilian mountaintops associated with campo rupestre vegetation based on taxonomically verified records of Microlicieae (Melastomataceae). To test the relevance of taxonomically vetted data, we compared these areas with those recovered using records downloaded from the Global Biodiversity Information Facility (GBIF) website. A total of 12 areas of endemism are recognized and described, of which only eight (66%) were retrieved using GBIF data. With 1º and 0.5º cells, analyses of GBIF data didn’t detect both individual and consensus areas of endemism with Endemicity scores as high as analyses of taxonomically verified data. Records based on misidentified specimens and/or incorrect coordinates affected the detection of areas of endemism using GBIF data. Our results show that taxonomically verified data may improve the efficiency of areas of endemism identification. Besides, the results suggest that the recovery of stable and continuous areas of endemism may be improved by using larger cells and adopting the loose criterion rule to merge individual areas of endemism into consensus areas. Biogeographically, the results indicate that the evolutionary histories of Chapada Diamantina, Southern Espinhaço and Brazilian Central Plateau were related to the radiation of Microlicieae, especially the genera Lavoisiera, Microlicia, and Trembleya. The expansion of areas of endemism in the Cadeia do Espinhaço was probably accelerated by climatic instability and glaciation cycles from Pliocene to Pleistocene.

Orphan hereditary pathologies are an urgent medical problem. Metabolic diseases have a special place among them. Currently they include about 500 nosological forms, in particular glycogen diseases. Type 1b glycogen disease (GB1b) occupies a special place. In addition to metabolic disorders, patients with this disease have severe neutropenia, which leads to frequent recurrent infectious diseases, such as stomatitis, periodontitis, furunculosis, otitis, colitis, gingivitis. In addition, recent studies showed the presence of lymphocytic dysregulation in patients with GB1b which is associated with T-regulatory lymphocytes dysfunction. It leads to an increased risk of developing autoimmune diseases, such as autoimmune thyroiditis, myasthenia gravis, Crohns disease, ANA-associated vasculitis. In addition, there is evidence about the association of GB1b and oncological pathologies, such as giant bone cell tumor, acute myeloid leukemia. According to such a wide variety of clinical manifestations, knowledge of the pathogenesis and possible non-metabolic manifestations is necessary for proper diagnostics and therapy.
