Full textThe eradication therapy against Helicobacter pylori (H. pylori) accounts for the reduction of IL-8 levels in gastric mucosa and the related risk of gastric cancer and ulcer onset. However, the current therapeutic approach has being challenged by antibiotic resistance. The discovery of natural compounds from edible plants with anti-inflammatory and anti-bacterial activities may contribute to the control of the disease and improve the safety profile of eradication. Plants producing tannins, including ellagitannins, have already shown pre-clinical gastroprotective evidences, but pharmacological validations are still required. Accordingly, Castanea sativa Mill. (sweet chestnut) is a well-known source of tannins, traditionally used for gastro-intestinal and respiratory diseases. On the base of the traditional indication, extracts from sweet chestnut are currently allowed as botanical ingredients by several European countries; however, their phytochemical composition and biological activity are poorly investigated, thus limiting the substantiation of health claims. The aim of the phD project was to characterize hydroalcoholic extracts from fruit and leaves of sweet chestnut, with a particular attention to tannin composition. In parallel, extracts were evaluated as potential candidates against gastritis and H. pylori through a multidisciplinary approach, including in vitro pharmacological and microbiological experiments. In the view of nutraceutical application, the work took into account the importance of the botanical origin of the extracts and evaluated the impact of gastric digestion on the stability of natural compounds. The results obtained on the fruit (chestnut) have been published by the research group (Sangiovanni and Piazza et al. 2018): in brief, following a bio-guided approach, hydroalcoholic extracts from chestnut of five different varieties were screened for their ability to impair TNFα-induced IL-8 release in human gastric epithelial cells (AGS). Two varieties (Venegon and Verdesa) emerged for their promising inhibitory activity (IC50s less than 10 μg/mL) and further characterized for their polyphenolic content. HPLC-MS analysis revealed the presence of condensed tannins (oligomeric proanthocyanidins) in extracts from the outer part of chestnut (pericarp and episperm), which correlated with IL-8 impairment (IC50s less than 0.5 μg/mL); on the contrary, the edible part (endosperm) was devoid of proanthocyanidins and inactive. Of note, hydrolysable tannins typical of the bark of sweet chestnut, namely gallotannins and ellagitannins, were absent in chestnut. In line with the literature on tannins, extracts maintained their inhibitory activity after the simulation of gastric digestion. Moreover, the bioactivity was modestly affected by the application of brief thermal treatments (till 100 °C for 30 min), thus suggesting the possibility of recovering chestnut shell for the enrichment of a bioactive chestnut flour. Consequently, an episperm-fortified flour was prepared respecting the original composition of chestnut, then extracted and assayed again for IL-8 impairment (IC50 = 16.35 μg/mL), thus demonstrating the potential application of chestnut by- products in nutraceuticals. Following a similar approach, hydroalcoholic extracts from leaf belonging to the most promising varieties (Venegon and Verdesa) were investigated. Contrarily to the fruit, the ellagitannins castalagin and vescalagin were revealed (1-2%) in leaf, thus prompting further studies on the anti-H. pylori effect. In particular, the antibacterial and anti-inflammatory activity was evaluated in a co-culture model of gastric epithelial cells infected with the bacteria (tumoral AGS in comparison with non-tumoral GES- 1). Leaf extracts inhibited IL-8 release in both the infected cells with IC50s lower than 30 μg/mL and impaired the bacterial growth at 100 μg/mL. After simulated gastric digestion, these bioactivities were moderately impaired (2 folds); in paralleled, a slight decrease in total phenols content was observed, thus suggesting their possible stability at gastric level after oral consumption. Ellagitannins castalagin and vescalagin exhibited the same inhibitory effects (IC50 on IL-8 less than 16 μM; MIC = 25 μM), but at concentrations unable to completely explain the effect of the whole extract; therefore, the synergy with other polyphenols could play a role. The experimental work sustained for the first time the potential anti-gastritis application of by- products from sweet chestnut, such as chestnut shell and leaf, pointing out the role of tannins for the biological activity.

Full textEsta dissertação resultou da realização de um estágio curricular de seis meses no Hospital Veterinário de São Bento. Temas como Gastroenterologia e Microbiologia estavam presentes diariamente no trabalho de clínica. A gastrite pode ocorrer como processo agudo ou crónico. Uma das principais formas de gastrite crónica é a linfoplasmocítica, na qual se incluem diversas etiologias, nomeadamente bactérias como Helicobacter spp. O Helicobacter pylori é conhecido extensamente no mundo de Medicina Humana, uma vez que, desde a sua descoberta, foi associado a gastrites severas, ulceração gastroduodenal e neoplasias gástricas. Uma das lesões mais características provocadas por Helicobacter pylori é a gastrite atrófica, associada em elevado grau a neoplasia gástrica. Esta bactéria já foi identificada em gatos, provocando também doença. Existem outras espécies de bactérias do género Helicobacter infectantes do estômago de gatos e de cães, como Helicobacter felis, Helicobacter bizzozeronii, Helicobacter salomonis e Helicobacter heilmannii, numa prevalência de cerca de 90 a 100 % e 67 a 100 %, respectivamente. Porém a sua associação a patologia gástrica é controversa, surgindo lesões histológicas apenas em alguns casos. Tal como em humanos, é comum a formação de folículos linfóides. Os meios de diagnóstico de infecção por H. pylori em humanos têm evoluído, no entanto a análise histopatológica continua a ser um dos métodos mais fiáveis, que dá também acesso a informação sobre lesão gástrica. O tratamento utilizado inclui normalmente antibioterapia combinada tripla, sendo eficaz na redução de lesões histopatológicas, mas na maioria dos casos não erradica eficazmente o microrganismo. Ao longo do estágio foi desenvolvido um estudo que incluía a realização do estudo macroscópico do estômago de gatos escolhidos de modo aleatório, citologia da mucosa gástrica, teste rápido de urease e análise histopatológica das amostras de biopsia em colaboração com o laboratório de Anatomia Patológica da Faculdade de Medicina Veterinária. Em onze gatos foi revelada uma prevalência nula de microrganismos e foram encontrados cinco casos de gastrite crónica atrófica fúndica, com lesões de hiperplasia folicular em dois deles e focos esporádicos linfóides nos outros três. No entanto alguns dos animais tiveram reacção positiva ao teste de urease, entre os quais dois também tinham lesões de gastrite atrófica. Estes achados histopatológicos são importantes, uma vez que são compatíveis com a teoria de que estes animais domésticos podem ser um bom modelo de estudo para as gastrites humanas ou mesmo potenciais reservatórios de microrganismos tipo Helicobacter spp., que infectam humanos.
ABSTRACT - Helicobacter spp. Detection in Felines - This dissertation results of a six month traineeship in Hospital Veterinário de São Bento. Clinical gastroenterology and microbiology presentions in felines are a common problem in Veterinary Medicine. Particulary, gastritis can be present in its acute or chronic form. One of the most common forms of chronic gastritis is lymphoplasmacytic, where several aetiology agents can be involved, namely Helicobacter spp. bacteria. Helicobacter pylori is a well studied bacteria in Human Medicine, since it has been associated with different gastric lesions, such as severe gastritis, gastroduodenal ulceration and gastric cancer. Atrophic gastritis is related as a common feature in H. pylori infection, showing a strong relation with the onset of gastric cancer. H. pylori was also identified in stomach cats and relationed with gastric disease. Other non H. pylori-Helicobacter are known to infect cat and dog, namely Helicobacter felis, Helicobacter bizzozeronii, Helicobacter salomonis e “Helicobacter heilmannii”, being their prevalence 90 to 100 % and 67 to 100 %, respectively. Nevertheless, its relationship with gastric pathology is not well clarified and histopathogical lesions appear only in few subjects. As in humans, the appearance of lymphoid follicles was a common found. Diagnostic tests for detection of H. pylori have evoluted, but histopathological analysis is still the “golden standart” to determine the presence of the bacteria and to identify lesions in gastric mucosa. Therapeutic protocols are composed by a combination of three antibiotics, leading to the clearance of gastric mucosa lesions, but not to the complete eradication of the bacteria. In the present work a trial was made for detection of Helicobacter spp. in eleven cats, chosen randomly. The methods chosen were: macroscopic evaluation of the gastric mucosa, urease test, brush cytology and histopathological analysis, in collaboration with the Laboratório de Anatomia Patológica da Faculdade de Medicina Veterinária. The tests revealed a 0 % prevalence of microorganisms and atrophic gastritis in five cats, lesions of follicular hyperplasia in two of them and lymphoid follicles in three subjects. Nevertheless, some cats had positive reactions to urease test, two of which had also atrophic gastritis. These findings contribute to the idea that cats can represent a good model disease of human gastritis and be potentials reservatories for Helicobacter spp.

Full textINTRODUCTION Gastric cancer is rampant in many countries around the world and it accounts for approximately 875000 new cases and 645000 deaths annually [Jemal A et al, 2004]. While overall incidence of gastric cancer is falling, in many countries of the world it remains one the most frequent causes of cancer related deaths. According to GLOBOCAN age-standardized cancer incidence data, in Germany 15.1 males and 8.8 females per 100.000 persons developed gastric cancer in 2002, in Lithuania – 25.3 males and 13.0 females, in Latvia – 24.6 males and 11.1 females, respectively. According to the data of Lithuanian Cancer Registry, gastric cancer incidence was 35.0 in male, 22.1 in female per 100.000 persons in 2005, 30.4 in male, 17.8 in female in 2004, respectively [Kurtinaitis J, 2004]. At present, primary or secondary prevention is likely to be the most effective means of reducing the incidence and mortality from this disease. However, to be successful, this strategy depends upon knowledge of the etiological factors and pathogenetic mechanisms involved in gastric carcinogenesis. Helicobacter pylori (H. pylori) has been categorized as a group I carcinogen by the International Agency for Research on Cancer and World Health Organization (WHO) in 1994. Full textHelicobacter pylori is considered the main risk factor for gastric carcinoma. Different experimental studies in animals seek to relate this carcinogenesis to other carcinogenic factors without success. This study sought to evaluate in rats, if there was a correlation between duodenogastric reflux, Helicobacter pylori and the development of gastric cancer or its precursors. To this end, it was carried out in three groups of rats the techniques: pyloroplasty, subtotal gastrectomy and only infection. After six months, the changes in mucosa were analyzed comparing the three groups. The mucosal changes reseached were: gastritis, metaplasia, dysplasia and epithelial neoplasms. At the end of the study, was found in the group that underwent pyloroplasty a high percentage of epithelial alterations and these correlated with the population of Helicobacter pylori. It is concluded that in the rat, the operation of pyloroplasty led to increased colonization of the population of Helicobacter pylori and is related with the development of benign lesions and gastric cancer.

Full textA infecÃÃo pelo Helicobacter pylori acomete mais da metade da populaÃÃo mundial, sendo esta bactÃria reconhecida como carcinÃgeno do grupo I pela OrganizaÃÃo Mundial de SaÃde-OMS. Familiares em primeiro grau de pacientes com cÃncer gÃstrico tÃm um maior risco de desenvolver cÃncer gÃstrico. Avaliamos a prevalÃncia de lesÃes precursoras do cÃncer gÃstrico e do Helicobacter pylori nos familiares em primeiro grau de pacientes com cÃncer gÃstrico, quando comparado a controles sem histÃria familiar. Cento e quatro familiares foram recrutados à partir de 40 casos de cÃncer gÃstrico tipo nÃo-cÃrdia e foram comparados com cento e dezoito controles, nÃo havendo diferenÃas estatisticamente significantes entre os dois grupos com relaÃÃo a idade, sexo, tabagismo, etilismo e condiÃÃes socioeconÃmicas garantindo a homogeneidade da amostra. Todos os pacientes foram submetidos a avaliaÃÃo endoscÃpica e biÃpsias seguindo o protocolo de Sydney. A anÃlise histopatolÃgica foi realizada pÃr um Ãnico patologista experiente e mascarado quanto a origem das amostras. Ainda que a prevalÃncia da atrofia e da metaplasia intestinal tenha ocorrida de forma similar nos dois grupos, a associaÃÃo destas lesÃes foi mais encontrada nos familiares que nos controles (p=0,021). A metaplasia intestinal tipo incompleta foi mais significante nos familiares (p=0,001), assim como a displasia (p=0,025). O padrÃo de gastrite encontrado nos familiares foi o de pangastrite associada a presenÃa de folÃculos linfÃides, padrÃo este jà definido como o de fenotÃpico de maior risco para a carcinogÃnese gÃstrica. NÃo houve diferenÃa estatisticamente significante entre os dois grupos com relaÃÃo a prevalÃncia do H. pylori , porÃm a topografia da infecÃÃo envolvendo antro e corpo foi maior nos familiares (p=0,001). De acordo com os resultados obtidos neste estudo, encontramos que familiares de pacientes com cÃncer gÃstrico tÃm uma maior prevalÃncia de alteraÃÃes histopatolÃgicas, estando estas alteraÃÃes confinadas a presenÃa do Helicobacter pylori
Infection by Helicobacter pylori, a bacterial species classified by WHO as being carcinogenic (group I) affects more than half the world population. First-degree relatives to patients with gastric cancer are at increased risk of developing gastric cancer. The present study evaluated the prevalence of precursor lesions of gastric cancer and infection by Helicobacter pylori in first-degree relatives to patients with gastric cancer as compared to controls with no family history of gastric cancer. One hundred four first-degree relatives to 40 patients with noncardiac gastric cancer were enrolled in the study and compared to 108 controls. The groups were statistically homogenous in terms of age. All patients were submitted to endoscopic evaluation and biopsy as described in the Sydney protocol. The histopathological analysis was carried out by a single, experienced pathologist blinded to the origin of the samples. Although the prevalence of atrophy and intestinal metaplasia was similar for the two groups, association with these lesions was more common among relatives than controls (p=0.021). Incomplete intestinal metaplasia was also more significant among relatives (p=0.001), as was displasia (p=0.025). The group of relatives presented a pattern of pangastritis associated with lymphoid follicles characteristic of increased risk for gastric carcinogenesis. There was no statistically significant difference between the groups with regard to the prevalence of H. pylori, though infection involving body and antrum was more prevalent among relatives (p=0.001). Our findings suggest that relatives to patients with gastric cancer present a greater prevalence of histopathological changes associated with the presence of H. pylori

Full textThe information gathering of cats with gastritis took place in three clinics in Lithuania (two of them were held in Kaunas and the other one- in Vilnius) from 2006 to 2012. All the patients who were treated in therapeutical sections were registered, their anamnesis vitae et mobri information was gathered and analyzed according to clinical examination methods. Diagnosis of gastric inflammation was usually based on history and clinical trial data, the abdominal X, endoscopic and laboratory test results. Gastritis is a fairly common digestive tract disorder. Etiotropic gastritis factors are many: it is feeding tolerance, infectious diseases, other organs and systems dysfunction, tumor pathology, the body's sensitization, neurogenic, iatrogenic causes, and so on. Analysis of the collected data showed that the period 2006-2012 cats treated in several Lithuanian veterinary clinics therapeutic sections, often suffered from gastro-intestinal diseases, which accounted for 63 ± 1.26 percent of all therapeutic treated for feline diseases (mean 376.1 ± 95.9 cases per year). In most cases, the assessment of a percentage of the gastrointestinal tract cats had in 2010 - 65 per cent. all cases, the least - in 2008. (61.2 per cent Cases all). Resumo: A Luehea divaricata Martus et Zuccarini (Tiliaceae) geralmente é encontrada em alguns estados do Brasil como Rio de Janeiro, Bahia, São Paulo, Minas Gerais, Goiás e Mato Grosso do Sul. Trata-se de uma árvore de 16 metros de altura, com casca fina, de coloração pardo-acinzentada, com numerosos e pequenos sulcos longitudinais e que floresce de dezembro a fevereiro. Conhecida popularmente como "Açoita-cavalo", sua casca é empregada na medicina popular, na forma de decocto, como antiinflamatório, diurético e anti-reumático. Este trabalho teve como objetivo avaliar a atividade antiulcerogênica de extratos obtidos da Luehea divaricata, determinar o possível mecanismo de ação e identificar prováveis substâncias ativas. Para a obtenção do extrato bruto hidroalcoólico (EBH 70%), foram utilizadas as cascas secas e moídas da espécie acima, coletada na reserva florestal de Leme. Esse extrato foi avaliado em modelos de úlcera gástrica induzida por indometacina e etanol. Para estudo do mecanismo de ação avaliou-se a participação de grupos sulfidrila, prostaglandinas, inibição da secreção ácida gástrica e quantificação de muco gastroprotetor. Uma triagem fitoquímica foi realizada para identificar substâncias presentes no EBH 70% e placas de CCD também foram feitas para comparar os extratos testados e seus constituintes. O EBH 70% de L. divaricata reduziu o índice de lesões ulcerativas produzido por indometacina e etanol. Esse mecanismo de ação antiulcerogênico está parcialmente relacionado com a atividade de radicais sulfidrila e pela precipitação de proteínas produzida pela presença de polifenóis (flavonóides, catequinas, antocianinas, taninos), identificados entre os princípios ativos dessa espécie. Além de polifenóis a triagem fitoquímica identificou a presença saponinas, esteróides triterpênicos e antracenosídeos
Abstract: The Luehea divaricata Martus et Zuccarini (Tiliaceae) is generally found in some States of Brazil such as Rio de Janeiro, Bahia, São Paulo, Minas Gerias, Goiás and Mato Grosso do Sul. It is a sixteen-metre-tall tree with fine bark and greyish colour which blossoms from December to February. It is popularly known as "açoita-cavalo" and its bark is used in popular medicine in the form of decoctation as an antiinfiamatory, diuretic and rheumatic drug. This work aimed at evaluating the anti-ulcerative activity of the hydro-alcoholic extract and the fractions obtained from the Luehea divaricata as well determine the probable mechanism of the action involved. Dried-milled bark was used in order to obtain the hydro-alcoholic extract of Luehea divaricata Martus et Zuccarini (EBH 70%). The EBH 70% was evaluated in models of indomethacin and ethanol induced ulcer in a dose of lg/kg. For the study of the mechanism action participation of sulfhydryl, prostaglandin, acid gastric secretion and quantification of mucus protector was evaluated. A phytochemical screening was also carried out to identify the components present in EBH 70%. The result showed that the EBH 70% of Luehea divaricata performs an important anti-ulcerative action and that its mechanism of action may be related to the participation of the sulfhydrylic non-protein substances. Moreover, this protective action should be consequence to condensed tannins presents in this extract. These compounds have been described in the literature as responsible for this antioxidan and cicatrizing local action

Resumo: A infecção por Helicobacter pylori e usualmente adquirida durante a infância e persiste durante toda a vida caso não seja tratada. A bactéria induz uma resposta inflamatória crônica, que esta associada com alterações hipergenéticas em oncogenes, genes supressores tumorais, e genes de reparo ao DNA. O Objetivo deste estudo foi avaliar a influencia da infecção por H. pylori no padrão de metilação de genes envolvidos na carcinogenese gástrica. O perfil de metilação de 106 genes foi caracterizado em biopsias provenientes de 5 pacientes adultos (1 Helicobacter pylori negativo, 3 com gastrite crônica infectados com linhagens de diferentes toxicidades e 1 com câncer gástrico) e em DNA proveniente de células epiteliais gástricas infectadas por H. pylori. Para estas analises foram utilizados o Promoter methylation array system, e o Gastric Cancer Methyl--Profiler DNA PCR Array. Os resultados destas análises mostraram que 20 % dos genes se encontravam metilados na amostra de câncer gástrico e 16 % dos genes na amostra de gastrite crônica, entretanto a analise comparativa entre estas amostras mostrou que compartilhavam 8,5 % dos genes metilados. A analise de metilação apos cocultura mostrou 12% dos genes metilados. Entre estes genes foram selecionados os seguintes: THBS1, HIC1, GATA--4, GATA--5 e MLH1 e MGMT, os quais foram avaliados em 239 amostras de biopsias gástricas, provenientes de 50 crianças e 97 adultos infetados ou não pela bactéria e de 92 adultos com câncer gástrico. O padrão de metilação foi avaliado por metilação PCR especifica (MSP--PCR). Os resultados obtidos neste trabalho mostraram que não houve metilação da região promotora de MLH1, MGMT e HIC1, nas amostras provenientes de crianças, independente do estado de infecção. Os maiores níveis de metilação entre as amostras oriundas de adultos com gastrite crônica infectados foram observados em GATA--4 e THBS1 (p<0.05), MGMT e GATA--5 (p<0.001). Nos pacientes com câncer gástrico os maiores níveis de metilação foram observados em MGMT e GATA--5 (p<0.05, p<0.001, respectivamente). Foi verificado um aumento da instabilidade microssatelites nas amostras de câncer gástrico (p<0.03) e o mesmo foi associado com um aumento nos níveis de metilação de MLH1 e sua consequente diminuição de RNAm. Foi realizado in vitro a analise de expressão de GATA--5 e TFF1 apos cocultura de diferentes linhagens de H. pylori e células AGS, que revelou um aumento nos níveis de RNAm de GATA--5 as 6, 24 e 48h apos infecção, e aumento de TFF1 apos 48h. Estes aumentos foram independentes da presença de cag PAI. O aumento na expressão de GATA--5 foi observado em camundongos C57BL/6 apos 6 meses de infecção. Foi observado in vivo que nas amostras metiladas em GATA--5 apresentavam níveis diminuídos de TFF1. Estes resultados nos permitem sugerir que a metilação acontece precocemente na mucosa gástrica dos pacientes pediátricos e esta associada com a infecção por H. pylori. Entretanto alguns loci como MGMT e MLH1 a metilação parece ser dependente do tempo de infecção. Foi observado que os níveis de metilação aumentaram conforme a idade dos pacientes sugerindo que o maior tempo de exposição induz maiores níveis de metilação
Abstract: Helicobacter pylori infection is usually acquired in childhood and persists into adulthood if untreated. It is known that the bacterium induces a chronic inflammatory response, which is associated with epigenetic alterations in oncogenes, tumor suppressor genes and DNA repair genes. The aim of this study was to evaluate the influence of Helicobacter pylori infection on the methylation pattern of genes related to gastric carcinogenesis. The methylation pattern of 106 genes was characterized in gastric biopsies samples from 5 adult patients (1 H. pylori negative, 3 chronic gastritis infected with strains of different toxicities, and 1 gastric cancer) and DNA extracted from co--cultured cells infected with H. pylori. These analyses were performed with Promoter methylation array system, and Gastric Cancer Methyl--Profiler DNA PCR Array. The results showed that 20 % of genes were methylated in gastric cancer mucosa and 16% were hyper methylated in the mucosa of the chronic gastritis patients. The comparative analysis showed that 8,5 % of genes were methylated in both samples. The results from the co culture showed that 12% of genes were methylated. Among them the following genes were selected: THBS1, HIC1, GATA--4, GATA--5 e MLH1 e MGMT and evaluated in 239 gastric biopsies samples from 50 children and 97 adults infected or uninfected by H. pylori, and 92 adults with gastric cancer. The methylation pattern was analyzed by methylation specific PCR (MSP--PCR). The results from pediatric samples showed no methylation for MLH1, MGMT and HIC1 genes, independent of the infection status. Among the infected adults samples the higher levels of methylation were observed for GATA--4 e THBS1 (p<0.05), MGMT e GATA--5 (p<0.001). Higher levels of methylation were observed for MGMT e GATA--5 (p<0.05, p<0.001, respectively) in gastric cancer samples. It was observed an increase in microsatellite instability among gastric cancer samples, related to hyper methylation of MLH1. The expression levels of GATA--5 and TFF1 was analyzed in vitro. The results from infected cells showed an up regulation at 6, 24, and 48 for GATA-5 and at 48 h for TFF1. This increase was independent of cagPAI status. In an animal model an up regulation of GATA--5 was observed after six months of infection. A decrease in TFF1 mRNA levels was observed in infected children and adults samples methylated in GATA--5 promoter region. These data suggest that promoter hyper methylation occurs in gastric mucosa of children in association with H. pylori infection, however for some loci as MGMT and MLH1 this event seems to be dependent on the time of exposure. Furthermore it was observed an increase in methylation frequencies in adults samples compared to pediatric samples suggesting that the duration of the infection is related to methylation levels

The present work was designed to investigate the incidence of the bacterium Helicobacter spp. In cats from the Recife city, Pernambuco State. The stomach samples were collected from 119 cats, without distinction between race or age, being 56 males and 63 females originated from the Environmental Survey Center (CVA), captured in the city of Recife.. The urease quick test and the histopathological examination using both the Hematoxilin-Eosin (HE) and Giemsa stains were performed. The urease quick test showed that 83,92% of the male (47/56) and 80,95% of the female (51/63) samples were positive for Helicobacter, with a total of 82,35% (98/119) of the population being positive. The Giemsa analysis demonstrated that in 73,21% of the males (41/56) and 74,60% of the females (47/63) samples the Helicobacter spp. Bacteria was found, and the total positive sample in the tested population was 73,94% (88/119). During the analysis performed using the HE stain, was observed that 59,66% (71/119) of the samples showed histopathological alterations, such as: lymphoplasmocitic infiltration with a lymphoid foliculum, microabscess, necrosis, hypotrophy, and in 40,34% of the samples(48/119) no alterations were observed. Since the Helicobacter spp. Microorganisms are able to cause gastric inflammation in cats, having showed , the research studies concerning the clinical importance and elucidate the gastric neoplasic genesis in these animals. It coud conclude that cats originated from Recife city, Pernambuco state, shows high incidence of helicobacteriosis.
O presente trabalho foi delineado para investigar a incidência da bactéria Helicobacter spp. em gatos oriundos da cidade do Recife do estado de Pernambuco. As amostras de estômagos analisadas, foram coletados de 119 gatos sem raça definida e idade, sendo 56 machos e 63 fêmeas provenientes do Centro de Vigilância Ambiental (CVA), capturados na cidade do Recife. Foram realizados para pesquisa de Helicobacter spp., os métodos de teste rápido da urease, o exame histopatológico corado pela técnica hematoxilina-eosina (HE) e pelo método de coloração Giemsa. As análises pelo teste rápido da urease mostraram que em 83,92% das amostras dos machos (47/56) e em 80,95% das amostras das fêmeas (51/63), foi detectada reação positiva para Helicobacter, sendo 82,35% (98/119) na totalidade das amostras. As análises pelo método de coloração pelo Giemsa mostraram que em 73,21% das amostras dos machos (41/56) e em 74,60% das amostras das fêmeas (47/63), foi encontrada a bactéria Helicobacter spp. , sendo 73,94% (88/119) na totalidade das amostras. Na análise das amostras coradas pela técnica de coloração hematoxilia-eosina (HE), verificou-se que em 59,66% das amostras (71/119) apresentaram alterações histopatológicas como: infiltrado linfoplasmocitário com folículo linfóide, microabcesso, necrose, hipotrofia; e em 40,34% das amostras (48/119) não apresentaram alterações histológicas. Dessa forma, os microorganismos do gênero Helicobacter spp. são capazes de gerar inflamação gástrica no gato, sendo necessário cada vez mais aprofundar as pesquisas a respeito da importância clínica e da gênese de neoplasias gástricas nesses animais. Concluiu-se neste trabalho que gatos oriundos da cidade do Recife, estado de Pernambuco apresentam alta incidência de helicobacteriose.

Full textEm clínica de animais de companhia, uma das doenças mais frequentemente encontradas é a gastrite idiopática, uma situação frustrante tanto para o clínico como para os proprietários. No Homem, a bactéria Helicobacter pylori tem recebido muita atenção pela sua capacidade oncogénica e existe a possibilidade de algumas espécies, nomeadamente “H. heilmannii”, H. salomonis, H. felis, H. bizzozeronii, e H. bilis, terem algum poder patogénico em animais de companhia. A finalidade do estudo desenvolvido ao longo do estágio foi de estabelecer a prevalência de várias espécies de Helicobacter numa população Portuguesa típica de animais de companhia, e de procurar eventuais relações entre estado de colonização e patologia gástrica. Num total de 103 animais (56 canídeos, 47 felídeos) foram determinadas, com recurso a técnicas de PCR, prevalências de 37,5%/48,9% para H. spp., 28,6%/34% para H. bizzozeronii e 17,9%/48,9% para “H. heilmannii” em cães e gatos respectivamente. H. pylori foi identificado num único gato (2,1%); H. salomonis e H. felis foram identificados exclusivamente em cães, com prevalências de 10,7% e 7,1%, respectivamente. H. bilis não foi identificado em qualquer animal. Em 19 das amostras fecais testadas (14 de origem canina e cinco de origem felina) 50% dos cães e 60% dos gatos acusaram a presença de H. spp, sendo apenas possível identificar, a nível de espécie, H. felis e H. bizzozeronii em amostras de dois cães. Foi identificada uma relação entre sintomatologia gástrica e colonização por “H. heilmannii” nos gatos, enquanto nos cães se determinou uma relação entre colonização por H. bizzozeronii e a presença de alterações inflamatórias na mucosa gástrica. A utilização de esfregaços da mucosa gástrica permitiu avaliar mais adequadamente o estado de colonização dos animais do que a observação de espiroquetas em cortes histológicos. Este estudo demonstra a presença de Helicobacter spp. numa população Portuguesa típica e sugere uma potencial relação entre Helicobacter spp. e gastrite crónica em animais de companhia, especialmente em gatos. A informação sobre presença de espécies de Helicobacter e respectivas prevalências poderá contribuir para uma decisão mais fundamentada sobre a prescrição de antibioterapia específica para Helicobacter em pacientes apresentados à consulta com sinais gastrointestinais e um diagnóstico de gastrite idiopática.
ABSTRACT - Helicobacter spp. and chronic gastritis in dogs and cats - Chronic idiopathic gastritis, one of the most frequently observed diseases in companion animal practice, can be a source of frustration for both the clinician and the owner. In humans, Helicobacter pylori has been thoroughly investigated due to its’ oncogenic capabilities. The possibility exists that some species of Helicobacter, namely H. salomonis, “H. heilmannii”, H. felis, H. bizzozeronii and H. bilis may play a role in the pathogenesis of chronic gastritis in companion animals. The current study, completed as part of the final year trainee program, was designed to investigate the prevalence of Helicobacter in a typical companion animal population, while searching for any eventual relationship between colonization and gastric disease. Prevalence was determined through the use of PCR in 103 animals (56 canines, 47 felines) as 37.5%/48.9% H. spp, 28.6%/34% H. bizzozeronii and 17.9%/48.9% “H. heilmannii” in dogs and cats, respectively. H. pylori was identified in a single feline sample (2.1%) while H. salomonis and H. felis were oberved exclusively in canine samples with a prevalence of 10.7% and 7.1% respectively. H. bilis could not be demonstrated in any of the samples analysed. Fecal samples from 19 patients were similarly tested (14 from canines, 5 from felines), 50% of dogs and 60% of cats tested positive for H. spp. Species specific PCR, identified the presence of H. felis and H. bizzozeronii in two dogs. In canines, a relationship between colonization with H. bizzozeronii and inflammatory alterations in the gastric mucosa was shown, while in cats colonization with “H. heilmannii” could be related to the presence of clinical signs of gastritis. Brush cytology was found to be more effective than histology in demonstrating the presence of Helicobacter in the gastric mucosa of both canines and felines. This study showed the presence of Helicobacter in a sample population of companion animals and the data gathered strongly suggests a relationship, especially in cats, between colonization and disease. The results obtained may contribute to an informed decision in prescribing antibiotic therapy for Helicobacter-associated gastritis in patients that present idiopathic chronic gastritis.

Full textThe bacterial species H. pylori is closely correlated with the development of major diseases and gastric cancer of the stomach. The strains that have more pathogenic genotype are more involved in bacterial virulence. The objective was to genotype strains of H. pylori from patients with gastritis, gastroduodenal ulcer and gastric cancer as the cagA and vacA genes and alleles by PCR technique. The genotyping of strains of H. pylori was performed by PCR technique. We evaluated 134 strains, 76 of patients with gastritis, 28 peptic ulcer and 30 gastric cancer. As a result of this research it was found that all strains subtyped were vacA positive, and 48.5% with the allele s1m1 vacA, vacA s1m2 20.9% and 6.7% s2m1 vacA, vacA s2m2 8.2%; 7 5% were hybrid strains and 8.2% had only the allele of the sequence signal (s). Gastritis in the distribution of vacA alleles were: 46.1% s1m1, s1m2 22.4%, 5.3% s2m1, s2m2 11.8%, 3.9% and 10.5% hybrid strains only with allele s. In cases of ulcer was found: 50% s1m1, s1m2 21.4%, 17.8% s2m1, s2m2 3.6%, 7.2% hybrid strains. In patients with cancer met: s1m1 53.3%, 16.7% s1m2, s2m2 3.3%, 16.7% and 10.0% hybrid strains with allele only s. The cagA gene was observed in 82.8% of strains, where 73.7% had gastritis and cagA positive, 96.4% had peptic ulcer and gastric cancer harbored 96.7% were positive for this gene. Allele vacAs1m1 was the most prevalent observed in the three groups. We have also found strains with multiple alleles of this gene, suggesting a genetic polymorphism arising from infection by more than one type of strain in the same individual. The cagA gene was also highly prevalent in both groups. The presence of cagA was statistically significant in the group of pangastritis and gastric cancer. The relationship of the cagA gene with allele vacAs1m1 was also significant in the groups of pangastritis and gastric cancer, which supports the higher pathogenicity of strains of the studied individuals.
A espÃcie bacteriana Helicobacter pylori està intimamente correlacionado com o desenvolvimento das principais afecÃÃes gÃstricas e do cÃncer de estÃmago. As cepas que apresentam genÃtipo mais patogÃnico sÃo as mais implicadas na virulÃncia da bactÃria. O objetivo deste trabalho foi caracterizar as cepas de H. pylori provenientes de pacientes portadores de gastrite, Ãlcera gastroduodenal e cÃncer gÃstrico quanto aos genes cagA e vacA e alelos atravÃs da tÃcnica de PCR. A genotipagem das cepas de H. pylori foi realizada atravÃs da tÃcnica de PCR. Foram avaliadas 134 cepas, sendo 76 de portadores de gastrite, 28 de Ãlcera pÃptica e 30 de cÃncer gÃstrico. Como resultado desta pesquisa obteve-se que, todas as cepas genotipadas foram vacA positivas, sendo 48,5% com o alelo vacA s1m1; 20,9% vacA s1m2; 6,7% vacA s2m1; 8,2% vacA s2m2; 7,5% eram cepas hÃbridas e 8,2% apresentaram somente o alelo da sequÃncia sinal(s). Nos portadores de gastrite a distribuiÃÃo dos alelos vacA foi: 46,1% s1m1, 22,4% s1m2, 5,3% s2m1, 11,8% s2m2, 3,9% cepas hÃbridas e 10,5% somente com alelo s. Nos casos de Ãlcera achou-se: 50% s1m1, 21,4% s1m2, 17,8% s2m1, 3,6% s2m2, 7,2% cepas hÃbridas. Nos portadores de cÃncer encontrou-se: 53,3% s1m1, 16,7% s1m2, 3,3% s2m2, 16,7% cepas hÃbridas e 10,0% somente com alelo s. O gene cagA foi observado em 82,8% das cepas, onde 73,7% tinham gastrite e cagA positivo, 96,4% tinham Ãlcera pÃptica e 96,7% portavam cÃncer gÃstrico com positividade para este gene. O alelo vacAs1m1 foi o mais prevalente observado nos trÃs grupos estudados. TambÃm foram encontradas cepas com alelos mÃltiplos desse gene, o que sugere um polimorfismo genÃtico advindo de infecÃÃo por mais de um tipo de cepa em um mesmo indivÃduo. O gene cagA tambÃm foi altamente prevalente nos grupos estudados. A presenÃa do cagA foi estatisticamente significante no grupo de pangastrite e cÃncer gÃstrico. A relaÃÃo do gene cagA com o alelo vacAs1m1 tambÃm foi significante nos grupos de pangastrite e cÃncer gÃstrico, o que corrobora a maior patogenicidade das cepas presentes nos indivÃduos estudados.

Resumo: A a-lactalbumina tem sido recentemente associada com redução de estresse, imunomodulação, atividade antimicrobiana após proteólise, propriedade anticancer e atividade antiulcerogênica. Os objetivos deste trabalho foram definir uma metodologia para a obtenção da a-lactalbumina e da b-lactoglobulina a partir de um concentrado de proteínas de soro de leite bovino e investigar a atividade antiulcerogênica da a-lactalbumina (produzida em laboratório e uma preparação comercial), da a-lactalbumina comercial hidrolisada e duas frações de seu hidrolisado (F1 < 1kDa e F2 > 1kDa) na proteção da mucosa gástrica em modelos de úlcera induzida por indomentacina e etanol absoluto em ratos, bem como, avançar na investigação das vias metabólicas de ação protetora. Para o isolamento da a-lactalbumina e da b-lactoglobulina a partir do concentrado de soro de leite bovino utilizou-se o sistema FPLC da Pharmacia e coluna de troca aniônica (resina Q Sepharose fast flow), e a fração correspondente a a- lactalbumina foi em seguida purificada por exclusão molecular utilizando-se a resina Sephacryl S-200. A b-lactoglobulina foi facilmente isolada por cromatografia de troca iônica apresentando-se quase homogênea e com um rendimento de 82%; para a a-lactalbumina, foi necessária uma etapa de purificação, e a proteína semi purificada apresentou contaminantes e baixo rendimento (32%). Para avaliar a ação antiulcerogênica da a-lactalbumina e de seus hidrolisados em modelo de úlcera induzido por indomentacina ou por etanol absoluto os animais receberam duas doses das amostras teste em dias consecutivos. Para avaliar possíveis vias de ação protetora foram estudadas suas participações na secreção ácida gástrica através da ligadura do piloro, a participação de substâncias sulfidrila e prostaglandinas na citoproteção gástrica em modelos de úlcera induzida por etanol absoluto. A a-lactalbumina comercial (Davisco Foods International, INC.) foi à amostra que apresentou maior porcentagem de inibição do índice de lesões ulcerativas no modelo de úlcera induzida por indometacina. No modelo de úlcera induzida por etanol absoluto, a a-lactalbumina (produzida em laboratório e a comercial) e frações de seu hidrolisado (F1 e F2) inibiram as lesões gástricas em níveis significativos na mesma ordem. A investigação das vias metabólicas de ação protetora, mostrarram que as amostras não alteraram a concentração ácida total após 4 horas de piloro ligado, sugerindo que a atividade antiulcerogênica desta proteína parece não ser mediada pela via da secreção ácida gástrica. A alquilação de grupamentos sulfidrila, envolvidos no mecanismo de citoproteção gástrica com N-etilmaleimida (NEM), eliminou o efeito protetor da a-lactalbumina e de frações de seu hidrolisado à mucosa gástrica, sugerindo que a ação antiulcerogênica parece depender da participação destas substâncias. A inibição da síntese de prostaglandina pela indometacina, bloqueador da síntese de prostaglandinas pela inibição da ciclooxigenase, não impediu que a a-lactalbumina (produzida em laboratório e a comercial) e a fração do hidrolisado de peso molecular >1kDa protejesse a mucosa gástrica a níveis significativos, sugerindo uma menor dependência de prostaglandina para a ação protetora dessas amostras. Já a fração do hidrolisado de peso molecular <1kDa perdeu sua atividade, evidenciando a participação de prostaglandinas na citoproteção gástrica por esta fração peptídica. A confirmação do envolvimento de prostaglandinas no mecanismo de ação antiulcerogênica por peptídios derivados da a-lactalbumina foi realizada através da quantificação do muco gástrico e da determinação da concentração de PGE2 na mucosa gástrica. Os resultados obtidos sugerem que há um conjunto de mecanismos envolvidos na proteção exercida pela a- lactalbumina e frações de seu hidrolisado enzimático sobre a mucosa gástrica, sendo estes, a participação de substâncias sulfidrila e aumento dos níveis de prostaglandina E2 na mucosa gástrica
Abstract: The a-lactalbumin has been recently associated with reduction of stress, immunomodulation, antimicrobial activity after proteolysis, anticancer and antiulcerogenic activity. The purpose of this work was to define a methodology for obtaing a-lactalbumin and b-lactoglobulin from a bovine whey protein concentrate and to investigate the antiulcerogenic activity of the a-lactalbumin (produced in laboratory and a commercial preparation ¿ Davisco Foods International, INC.), of the hidrolyzed commercial a-lactalbumin and two fractions of this hidrolysate (F1<1kDa and F2 >1kDa), in the protection of gastric mucosa induced by indomethacin or absolute ethanol induced ulcer in rats, as well as, to advance in the inquiry of the metabolic pathways of protective actions. For the isolation of the a-lactalbumin and of b-lactoglobulin from the bovine whey protein concentrate a Pharmacia FPLC system with anion exchange column (Q-Sepharose fast flow resin) was used. Alpha-lactalbumin was further purified by molecular exclusion chromatography using Sephacryl S-200 column. The b-lactoglobulin was easily isolated from ionic exchange chromatography presenting good purity and 82% yield; for the a-lactalbumin, a second stage of purification was necessary, and the partially purified protein still presented some contaminants and a low yield (35%). To evaluate the antiulcerogenic action of the a-lactalbumin and its hidrolysates for indomethacin and absolute ethanol induced ulcer in rats, the animals received two doses of the samples in consecutive days. To determine possible mechanisms of protection to the gastric mucosa, the effect in the gastric acid secretion through the tied piloro technique, the participation of sulfhydryl substances and prostaglandins were studied. For the indomethacin induced ulcer only the commercial a- lactalbumin inhibited the gastric injuries, while in the absolute ethanol induced ulcer all the samples were able to reduce the gastric injuries at significant levels. The inquiry of the metabolic pathways of protective action showed that the samples did not modify the concentration of the total acid after 4 hours tied piloro, suggesting that the antiulcerogenic activity of these proteins and peptides seem not to be mediated by the mechanism of gastric acid secretion. The alkylation of sulfhydryl groups with N-ethylmaleimide (NEM), partially eliminate the protective effect of the a-lactalbumin and its hidrolysates on the gastric mucosa, suggesting the participation of these groups in the antiulcerogenic action. The inhibition of the prostaglandin synthesis with indomethacin did not eliminate the protection offered by a-lactalbumin and the hydrolysate fraction of molecular weight > 1kDa. However the protection offered by the hydrolysate fraction of molecular weight < 1kDa was completely neutralysed by the indomethacin cycloxygenase inhibition, suggesting that a-lactalbumin low peptides seem to protect the gastric mucosa via the prostaglandin cycle. The confirmation of the envolvement of prostaglandins in the mechanism of antiulcerogenic action by a-lactalbumin peptides was substanciated through the quantification of gastric mucus and the determination of the PGE2 concentration in the gastric mucosa. These results suggest that there are more than one mechanism for the stomach cytoprotection against ulcerogenic factors a-lactalbumin and its hydrolysates. While intact a-lactalbumin and high molecular weight peptides seem to depend on the sulfhydryl groups for protection, low molecular weight peptides (MW < 1kDa) seem to act by stimulation of prostaglandin synthesis

Resumo: Em processos de ulcerogênese por isquemia-reperfusão há grande liberação de radicais livres causando peroxidaçâo lípidica e desestruíuração proteica pela oxidação de grupos -SH, o que altera funções da membrana e de enzimas e leva à disfunção celular e necrose tecidual. Ensaios fitoquímícos revelaram que infusos de Turnera diffusa e T. ulmifolia tem como compostos majoritários flavonas C- glicosiladas derivadas de luteonina e apigenina. A atividade antiulcerogênica destes infusos está comprovada, porém faltam estudos sobre o potencial antioxidante. Relacionando o potencial antioxidante de flavonóides e a atividade antíúlcera do extrato, mostramos in vitro que 50 pg/ml salina do extrato de T. ulmifolia e T diffusa protegem em 58 e 47% a oxidação de GSH proteico na mitocôndria oxidada por 10 uM Fe+2 e 200 uM ascorbato, respectivamente. E proteção de 12 e 50% em mitocôndria oxidada por 1,5 mM Pi e 10pM CaCb, demonstrando poder de manter grupos -SH intactos durante diferentes sistemas de danos à mitocôndria. O potencial de estabilizar a membrana foi corroborado com a diminuição na peroxidaçâo lípidica de 62 e 67% para 7. ulmifolia e T diffusa, respectivamente. Quando testados em úlcera por isquemia-reperfusão como modelo in vivo houve uma diminuição no índice de úlceras de 59 e 69%; com diminuição da peroxidaçâo lípidica de 50 e 51% e manutenção na [-SH] total de até 100% na mucosa gástrica para T. ulmifolia e T diffusa, quando comparadas à salina.
Abstract: Turnera ulmifolia L. and Turnera diffusa Wild are known in folk medicine for treatment of several gastric diseases. Other studies have previously reported anti-ulcer activity and this relationship with your majority composition to be flavonoids. Natural flavonoids are known for scavenging properties on oxygen radicals in vivo and in vitro. The oxyradicals generated during the proposed episodes of ischemia-reperfusion provoke severe changes in mitochondria-generated ROS, induce peroxidation of the membrane lipids and oxidation -SH compounds. Therefore, agents that inhibit the oxidation thiol groups, as well as lipid peroxidation, may be of high pharmacological potential. This work showed activity antioxidant when used mitochondria systems in vitro, both species tea plants exhibited effect to inhibit lipid peroxidation and oxidation thiois proteins compounds induced by FeC^/ascorbic acid, succuiate/rotenone or CaCyinorganic phosphate (Pi). Our experiments in vivo demonstrated effect anti-ulcer and antioxidant to both species in models ischaemia-reperfUsion-ulcer-induced after administration orally tea T. diffusa e T. ulmifolia inhibit lesion ulcer index when compared with control group. And in gastric mucosa exhibited effect to inhibit lipid peroxidation or oxidation -SH total compounds. In model ethanol/HCl-ulcer-induced Turnera ulmifolia show effect anti-ulcer partial dependent to compounds SH and Turnera diffusa demonstrated to be independent effect anti-ulcer to SH compounds.

Resumo: A pesquisa farmacológica têm se voltado ao conhecimento da medicina popular, a fim de estabelecer as bases científicas dos seus efeitos farmacológicos. Atualmente taninos têm papel de destaque em função de sua capacidade antioxidante e em função dos avanços obtidos na análise e identificação desses compostos além de suas possibilidades terapêuticas. Entre as diversas fontes naturais desses compostos destacou-se, neste trabalho, uma planta bastante conhecida nos ambientes costeiros, a Rhizophora mangle L. (manguevermelho), pertencente à família RHIZOPHORACEAE. O gênero Rhizophora compreende cinco espécies das regiões equatorial, tropical e subtropical de ambos os hemisférios. É uma das mais importantes fontes de taninos (15 a 36% da massa da casca seca) para uso industrial. R. mangle é conhecida na medicina popular caribenha por suas propriedades antinflamatória, antifúngica, bactericida, anti-úlcera, antihipertensiva e antioxidante. Este trabalho avaliou as atividades antiulcerogênica e antioxidante de R. mangle, assim como traçou o perfil químico das substâncias presentes em suas cascas. Para tanto foram realizados ensaios químicos e farmacológicos com o extrato bruto (acetona:água 7:3) e frações orgânicas (aquosa - fr aq, acetato de etila - fr-EtOAc e butanólica - fr-BuOH) dele obtidas. A análise da atividade antiulcerogênica do extrato e frações, mostrou que fr-BuOH foi mais eficaz na dose de 0,5mg.Kg-1. Essa fração também apresentou atividade antioxidante significativa. Nos ensaios químicos realizados, tanto para o extrato bruto quanto para as frações, verificou-se a existência de alta concentração de compostos polifenólicos. Os testes farmacológicos com a fr-BuOH buscaram determinar os mecanismos pelos quais são exercidas as atividades antiulcerogênica e antioxidante das substâncias nela presentes. Concluiu-se que fr-BuOH possui catequinas heterosídeos, galotaninos, elagitaninos e taninos complexos em sua composição, de acordo com métodos de análise empregados. Estas substâncias mostraram-se capazes de sequestrar radicais livres, atuar sobre compostos (GSH) e enzimas antioxidantes (GR, GPx e SOD) e próoxidantes (MPO e LPO) definindo sua atividade antioxidante. A atividade antiulcerogênica além de ser mediada pela própria ação antioxidante é dependente de GSH e NO, envolve a participação de PGE2, COX-2, EGF, PCNA, HSP-70, além do aumento da produção de muco aderido e propriedades anti-secretoras
Abstract: The pharmacological research has been focusing on the folk medicine knowledge in order to establish the scientific basis of their pharmacological effects. Nowadays, tannins play a prominent role because of its high antioxidant capacity and, due to the progress made in the analysis and identification of these compounds and their therapeutic possibilities. Among the many natural sources of these compounds, highlighted in this work a well known plant in coastal environments, the Rhizophora mangle L., belonging to the family RHIZOPHORACEAE. The genus includes five species of Rhizophora in equatorial, tropical and subtropical regions in both hemispheres. It is one of the most important sources of tannin (15 to 36% of the dry bark weight) in industrial use. Rhizophora mangle is well known in the Caribbean folk medicine for its anti-inflammatory, antifungal, antibacterial, anti-ulcer, anti-hypertensive and antioxidant properties. This study aimed to evaluate the antioxidant and anti-ulcer activities of R. mangle, and to elucidate the chemical compounds present in its barks. For both evaluations chemical and pharmacological assays with the crude extract (acetone: water 7:3) and their organic fractions (aqueous - fr-aq, ethyl acetate - fr-EtOAc and butanolic - fr-BuOH) were performed. The analysis of anti-ulcer activity of the extract and fractions showed that fr-BuOH was more effective at a dose of 0,5 mg.Kg-1. This fraction also showed significant antioxidant activity. It was found the presence of high concentrations of polyphenolic compounds in the chemical assay performed for both the crude extract and its fractions. The pharmacological experiments were carried out to determine the mechanisms by which the compounds present in fr-BuOH exert their antiulcer and antioxidant activities. It was concluded that the fr-BuOH has antioxidant activity linked to the concentration of polyphenolic compounds, which were able to scavenge free radicals, maintain the levels of GSH, reduce the MPO activity and LPO levels and maintain or increase the antioxidant enzymatic activities of GR, GPx and SOD. Moreover, the antiulcer activity of fr-BuOH is mediated by several mechanisms including: antioxidant action; involvement of GSH and NO; anti-secretory properties; increase of mucus and PGE2 synthesis, and expression of COX-2, EGF, HSP-70 and PCNA on gastric mucosa. Chemical study revealed that the major compounds are heteroside catechins, gallotannins and "tannin complexes"

Full textCon una frequenza stimata nel 2002 di 933,937 nuovi casi e 700,349 decessi a livello mondiale, il tumore gastrico risulta essere il quarto tumore più comune nel mondo e il secondo come causa di morte correlata al cancro dopo il tumore epatico (Parkin et al., 2005; Crew and Neugut, 2006). La sequenza delle variazioni istologiche attraverso le quali si sviluppa il carcinoma gastrico è estremamente complessa. Le conoscenze attuali sui precursori del carcinoma gastrico consentono di ricostruirne l’istogenesi che conferma nella maggior parte dei casi la sequenza atrofia-metaplasia-displasia-neoplasia (Correa, 1992). L’aggressività legata alla precoce metastatizzazione di tale neoplasia, rende spesso la prognosi infausta e attualmente nessun biomarcatore (o set di biomarcatori) è considerato sufficientemente affidabile per essere utilizzato nella pratica clinica. L’identificazione di nuovi bersagli molecolari per l’individuazione dei pazienti ad alto rischio di sviluppare il tumore rappresenta dunque una necessità per lo sviluppo di nuove strategie terapeutiche più efficaci. L’obbiettivo del progetto è stato lo studio dei profili di espressione fosfoproteomica delle cellule dell’epitelio gastrico in soggetti sani e pazienti con gastrite atrofica metaplasica e il loro confronto con i profili delle cellule neoplastiche, in pazienti con carcinoma dello stomaco con e senza metastasi linfonodali. In particolare, sono state prelevate 10 biopsie di mucosa normale e 9 metaplasie mediante EGDS e 18 campioni tumorali durante l’intervento di resezione chirurgica (di cui 9 senza metastasi linfonodali “N-” e 9 con metastasi linfonodali “N+”). Tutti i pazienti affetti da metaplasia e tumote N- ed N+ hanno presentato nella loro storia clinica un’infezione da parte di Helicobacter pylori. I campioni sono stati processati mediante la tecnica dei Reverse Phase Protein Array e sono stati analizzati i livelli di fosforilazione di 50 proteine chiave coinvolte principalmente nell’apoptosi, nella sopravvivenza e nella differenziazione cellulare. Dall’analisi statistica eseguita mediante test di Kruskal Wallis è emerso un identico andamento dei segnali di fosforilazione in tutte le isoforme delle protein chinasi C analizzate (PKC α, β2, δ e θ). Più precisamente, è stato possibile osservare un incremento dei livelli di fosforilazione dal tessuto sano a quello tumorale facendo presupporre che le proteine in esame possano avere un ruolo centrale nella progressione tumorale. Inoltre, analizzando le proteine incluse nei pathway cellulari in cui sono coinvolte le protein chinasi C si è osservata l’attivazione delle vie di segnale P13/AKT e NF-kβ/Bcl2 da cui emerge una forte inibizione dei segnali apoptotici cellulari. La presenza di questi profili risulterebbe essere compatibile con una possibile attivazione delle proteine da parte di H. pylori mediata dai propri fattori di virulenza.

Full textNotre stratégie de recherche est basée sur le principe (démontré sur le plan expérimental par de nombreuses études) selon lequel les cellules cancéreuses migrantes résistent à l’apoptose, et sont dès lors protégées contre les effets pro-apoptotiques de la chimiothérapie et de la radiothérapie qui représentent la quasi totalité de l’arsenal thérapeutique dont disposent les oncologues pour combattre les cancers. Diverses études expérimentales ont démontré que le fait de réduire le taux de migration de cellules cancéreuses résistantes à l’apoptose conférait à celles-ci une sensibilité accrue aux agents pro-apoptotiques. Nos résultats démontrent que la gastrine modifie de manière très significative les propriétés migratoires des cellules de mélanomes, sans toutefois modifier leur sensibilité à des agents pro-apoptotiques. Au contraire, la gastrine protègerait les cellules de mélanomes contre l’apoptose. Nous démontrons également dans notre travail, in vivo, un rôle pro-angiogénique pour la gastrine au sein de xénogreffes de mélanomes humains. Signalons que notre travail est le premier à démontrer un rôle potentiel de la gastrine au niveau de la biologie des mélanomes, tout au moins sur le plan expérimental.
Tout comme nous l’avons observé pour la gastrine, la galectine 1 semble également conférer aux cellules de mélanomes un certain degré de résistance aux agressions chimiothérapiques. Cette fois, le fait de diminuer le taux d’expression de la galectine 1 au sein de cellules du mélanome murin expérimental B16F10 (qui exprime des quantités importantes de galectine 1) renforce l’effet thérapeutique du témozolomide qui est une molécule cytotoxique. Cet effet semble survenir, tout au moins partiellement, suite à une diminution du taux d’expression de la protéine Hsp70 (suite à la diminution du taux d’expression de la galectine 1), avec pour conséquence une augmentation de la mort cellulaire par perméabilisation de la membrane des lysosomes.
Nous proposons une nouvelle approche thérapeutique pour combattre les mélanomes en faisant appel à la technique des petits ARN interférants (siRNA), dirigés dans le cas présent contre la galectine 1.

Resumo: O Helicobacter pylori é uma bactéria gram-negativa que está estritamente relacionada com o desenvolvimento de doenças gástricas, inclusive malignas, sendo classificada como carcinógeno do grupo I pela Agência Internacional para Pesquisa do Câncer. Apesar de grande parte da população mundial estar contaminada pela bactéria, a maioria dos indivíduos permanece assintomática. Muitas abordagens são sugeridas para diferenciar os isolados de H. pylori e diversas técnicas de biologia molecular têm sido aplicadas para caracterizar as amostras clínicas de diferentes pacientes. A análise de restrição (RFLP) usada em conjunto com a PCR tem sido amplamente utilizada para a tipagem de isolados clínicos, utilizando vários genes do genoma do H. pylori como alvos para esse método. Padrões de eletroforese da nested PCR e da PCR para as regiões dos genes urease C e urease B do H. pylori foram obtidos com enzimas de restrição e comparados entre pacientes com úlcera péptica e gastrite crônica. Os produtos da nested PCR para urease C foram digeridos com a enzima Mbo I e os produtos da PCR para urease B com a enzima Hae III, sendo os fragmentos obtidos analisados por eletroforese em gel de agarose. Foram encontrados 7 padrões de genotipagem para urease B e 17 padrões para urease C. O grupo prevalente para gastrite crônica obtido com essa última genotipagem foi o denominado grupo ureC MboI G, com fragmentos de 110, 120, 250 e 310 pares de bases, ocorrendo em 13 pacientes (15,7% dos casos). Em relação aos casos de úlcera péptica, o grupo de genotipagem mais prevalente foi o denominado ureC MboI E, com 110, 180 e 500 pares de bases, ocorrendo em 11 pacientes (24,4% do total de casos). O grupo prevalente obtido com a genotipagem para urease B foi o denominado ureB I, com um fragmento único de 750 pares de bases. Esse grupo foi o prevalente tanto para os casos de úlcera péptica (27), com 60,0%, quanto para os de gastrite crônica (53), com 63,9%. Não foram observadas diferenças significativas entre os padrões encontrados para ambas as doenças. A nested PCR para detecção prévia do DNA do H. pylori também foi realizada, obtendo-se 100% de concordância. Os métodos de tipagem dos produtos obtidos por nested PCR e PCR proporcionam um esquema funcional e de grande reprodutibilidade para estudos epidemiológicos e de caracterização de cepas
Abstract: Helicobacter pylori is a gram-negative bacterium that has been implicated as a major human gastric pathogen responsible for gastritis and peptic ulcer disease. It has been classified as a group I carcinogen by the International Agency for Research on Cancer and is regarded as a primary factor for gastric cancer. A significant proportion of the world population is infected with H. pylori, even though most infections are asymptomatic. To isolate H. pylori, many approaches have been presented and several molecular techniques have been applied to separate clinical isolates from different patients. PCR-RFLP analysis has been widely developed for the typing of clinical isolates, with several genes within H. pylori having been targeted by this method. Eletroforesis patterns of nested PCR and PCR to urease C and urease B gene regions of H. pylori were obtained by restriction fragment length polymorphism and compared among patients with peptic ulcer and chronic gastritis. Nested PCR products for urease C were digested with MboI enzyme and PCR products for urease B with Hae III enzyme, and the obtained fragments were analyzed by eletroforesis in agarosis gel. We have encountered 7 genotyping patterns for urease B and 17 patterns for urease C. The prevalent group for chronic gastritis which was obtained with this last genotyping was named ureC MboI G group, with fragments of 110, 120, 250 and 310 base pairs, occurring in 13 patients (15,7% of the cases). Regarding the peptic ulcer cases, the most prevalent genotyping group was named ureC MboI E group, with 110, 180 and 500 base pairs, occurring in 11 patients (24,4% of the cases). The prevalent group obtained with the genotyping for urease B was named ureB I, with only one fragment of 750 base pairs. This group was the prevalent one both in peptic ulcer cases (27) with 60,0%, and in chronic gastritis case (53), with 63,9%. We haven¿t found meaningful differences among the encountered patterns for both diseases. Nested PCR for previous detection of H. pylori DNA has also been performed, and we have obtained 100% of concordance. The typing methods of the products obtained by nested PCR and PCR show a functional scheme and of great reproduction for epidemiologic studies and of H. pylori strains characterization

O objetivo deste trabalho in situ foi avaliar o efeito de ácidos, presentes em refrigerantes e no suco gástrico, na rugosidade de superfície e microdureza de materiais restauradores. Cento e sessenta e oito corpos-de-prova foram divididos aletoriamente em 3 grupos de estudo com 8 voluntários. Cada voluntário utilizou o dispositivo intrabucal removível contendo 7 amostras (Ketac Nano, Ketac Nano + Biscover LV, Esthet-X, Esthet-X + Biscover LV, Supreme XT, Supreme XT + Biscover LV e esmalte bovino). O grupo I foi tratado com ácido clorídrico, o grupo II foi tratado com refrigerante de limão e o grupo III foi o controle, não recebendo nenhum tratamento ácido. As variáveis de respostas foram a medida de rugosidade superficial média (Ra) e a microdureza Knoop. A ANOVA e teste de Fisher PLSD (α = 0,05) demonstraram que a resina composta Supreme XT apresentou os maiores valores de microdureza antes e após os processos de envelhecimento com diferença significante para todos os outros materiais (p<0,0001). Os menores valores foram encontrados para os materiais selados com Biscover LV em todos os períodos analisados (p<0,0001). O ionômero de vidro Ketac Nano apresentou os maiores valores de rugosidade de superfície após a exposição nas soluções ácidas. A aplicação do selante de superfície não alterou os valores de rugosidade para as resinas compostas estudadas à exceção do Esthet-X após o desafio em ácido clorídrico (p<0.0001). As soluções ácidas foram capazes de alterar a microdureza e rugosidade de superfície dos materiais restauradores, exceto dos materiais selados
The aim of this study was to evaluate the effect of different acidics solutions on the microhardness and surface roughness of restorative materials. 168 specimens were randomly divided into 3 groups with 8 volunteers. The volunteers wore palatal device with 7 specimens (Ketac Nano, Ketac Nano + Biscover LV, Esthet-X, Esthet-X + Biscover LV, Supreme XT, Supreme XT + Biscover LV and bovine enamel). The group I was immersed in HCl; the group II was immersed in soft drink and the group III was subjected to saliva only. The microhardness and surface roughness data were analyzed by ANOVA and Fisher‟s test (α = 0.05). The Supreme XT showed the highest values of microhardness before and after aging with a significant difference for the other materials (p<0.0001). The lowest values were found for the materials sealed with Biscover LV in all periods analyzed (p<0.0001). The Ketac Nano glass-ionomer cement showed the highest values of surface roughness after exposure in acidic solutions. The application of the sealant did not reduce the roughness values for the composites studied except for Esthet-X after the challenge in hydrochloric acid. The acidic solutions promoted changes in microhardness and surface roughness of restorative materials, except for the sealed materials

Full textGraÃa et al. (2002) observou em ratos que a hipervolemia aumenta o tÃnus gÃstrico, enquanto a hipovolemia o diminuÃa. Resolvemos estudar o efeito do estiramento cardÃaco sobre o tÃnus gÃstrico e os mecanismos neurais envolvidos. Metodologia: Ratos albinos machos (n=82, ~320g), anestesiados, com vasos cervicais canulados para registros hemodinÃmicos e distensÃo do Ãtrio (DA). Um grupo separado de animais foi submetido à vagotomia subdiafragmÃtica (DAIV) ou esplancnotomia (DAV). Para registro do volume gÃstrico (VG), foi introduzido per os um cateter com um balÃo de lÃtex posicionado no estÃmago proximal e acoplado a um pletismÃmetro. ApÃs perÃodo basal (15min), os ratos foram submetidos aos protocolos: falso distendido (FDA) ou animais submetidos durante 5 min à DA com 30, 50 ou 70ÂL, respectivamente (DAI), (DAII) e (DAIII), ou (DAIV) e (DAV) submetidos a DA com 50ÂL. Em seguida, houve monitoraÃÃo nos 30min seguintes, divididos em intervalos de 10 min, designados de D10, D20 e D30. Os dados relativos ao VG estÃo representados em percentagem de seus respectivos valores basais. Sendo analisados por ANOVA seguida do teste de Bonferroni. Resultados: NÃo houve variaÃÃes do VG e PVC no grupo FDA, porÃm no grupo DAI houve uma diminuiÃÃo significativa do VG em D20 e D30 (9.9% e 14%, respectivamente), nÃo houve aumento da PVC durante a DA. Nos grupos DAII e DAIII o VG diminuiu a partir de D10 (5% e 5.8%, respectivamente, p<0,05) com persistÃncia em D20 (8,5 e 13%,respectivamente) e D30 (11,5 e 16,5%, respectivamente). AlÃm disso, a PVC aumentou em ambos os grupos durante a DA (p< 0,05). No grupo IV, a vagotomia preveniu a diminuiÃÃo do GV, permanecendo inalterado o VG durante o perÃodo experimental. No grupo V, a esplancnotomia aumentou tal efeito, o VG diminuiu a partir de D10 (10,5%, p<0,05) se intensificando em D20 e D30 (16 % e 23,4%, respectivamente). ConclusÃo: A distensÃo do balÃo atrial alÃm de desencadear ajuste hemodinÃmicos diminui o VG em ratos anestesiados, efeito abolido apÃs vagotomia subdiafragmÃtica, e permanecendo acentuado nos animais esplancnotomizados

Full textBackground: The incidence of malignant melanoma (MM) is increasing worldwide and the management of patients with disseminated disease is a difficult problem. Chemotherapy was the treatment of choice in metastatic melanomas for many decades. However, this option produces disappointing results. Recently, the better understanding about molecular events related do the development of MM allowed the development of new drugs directed against specific molecular targets. The gastrin-releasing peptide (GRP) is a neuroendocrine peptide shown to have growth-stimulatory effects on many types of murine and human cancers. Few data are available about GRP receptor (GRPR) expression in MM. The understanding about the molecular biology of MM may allow the identification of novel intracellular pathways of relevance in this disease, and potential GRPR modulators of therapeutic application in patients with refractory MM. Objectives: The aim of this study was to determine the GRPR expression in biopsy samples of patients with cutaneous MM, as well as to correlate its expression with known prognostic factors of relevance in this disease. Methods: Immunohistochemistry was performed on formalin-fixed, paraffin-embedded biopsy samples from 51 patients with cutaneous MM. A rabbit polyclonal anti-GRPR antibody (OPA1-15619, Affinity Bioreagents, USA) was used. Following the quantification of GRPR expression in the samples, the differences in GRPR expression among distinct prognostic MM subgroups were analyzed, using the Fisher´s test. Results: GRPR immunoexpression was demonstrated in cytoplasm of 42/51 (82.4%) cutaneous MM cases. It was strongly expressed in 30 (58.9%) of the samples. No significant differences between GRPR expression neither in relation to the primary or metastatic site, nor among known prognostic subgroups Clark´s level (p=0.35) and Breslow index ( p= 0.175) was observed. Conclusion: Our study has demonstrated the occurrence of a high GRPR expression in tumor specimens obtained from patients with cutaneous MM (82,4%). In 30 samples, a strong intensity of expression was documented (58.3%). No correlation was observed between the level of GRPR expression in primary or metastatic sites, nor for distinct Clark´s levels or Breslow index. This is one of the first studies demonstrating a high GRPR expression in tumor samples from patients with MM. Further studies are warranted, preferably including a larger patient population, to allow a better analysis of the expression of these receptors in different prognostic subgroups. If these observations are confirmed, the therapeutic use of GRPR inhibitors should be considered in patients with advanced MM who failed conventional treatments.

Full textA gastrite atrófica predominante do corpo com antro relativamente conservado é a lesão fundamental associada à elevação dos níveis de gastrina sérica e alteração do crescimento/ distribuição das células endócrinas na mucosa gástrica oxíntica e antral. O objetivo do estudo foi avaliar as alterações histopatológicas das células endócrinas da mucosa do antro e do corpo gástricos e relacionar estas alterações com os níveis séricos de gastrina e a presença de auto-anticorpos dirigidos contra a célula parietal (anti-CP). Foram estudados 44 pacientes dispépticos submetidos à endoscopia digestiva alta no Instituto Alfa de Gastroenterologia do Hospital das Clínicas da UFMG e que apresentavam ao exame histopatológico gastrite atrófica do corpo com mucosa antral relativamente bem conservada. Foram avaliados nestes pacientes: níveis séricos de gastrina, de vitamina B12, presença do anticorpo anti-CP. Cortes de parafina do antro e do corpo gástrico destes pacientes foram submetidos à coloração das células endócrinas pelo método de Grimelius e para evidenciar as células parietais foi utilizada imunocitoquímica. A hiperplasia das células endócrinas foi encontrada mais freqüentemente na mucosa do corpo gástrico. A gastrina sérica dos pacientes apresentou valor médio de 349,5 pg/mL (41,4 2.720 pg/mL). As células parietais estavam ausentes (24/44) ou diminuídas: presentes em pequenos aglomerados (8/44), rarefeitas na mucosa (8/44). Autoanticorpos anti- CP estavam presentes em 32/44 pacientes (72,7%). Não houve relação entre a presença de hiperplasia endócrina do antro e do corpo com os diferentes níveis séricos de gastrina dos pacientes analisados. Concluímos que a presença de hiperplasia endócrina no corpo gástrico pode ser utilizada como marcador útil para indicar provável auto-imunidade dos casos de gastrite crônica atrófica predominante do corpo.

O Kinosternon scorpioides é uma tartaruga de água doce conhecida como jurará. O objetivo do presente trabalho foi identificar serotonina, gastrina, enteroglucagon e motilina em esôfago, estômago e duodeno de K.scorpioides por meio da técnica de imunohistoquímica. Os resultados mostraram que as células imunorreativas à serotonina estavam presentes no epitélio na região caudal do esôfago, nas glândulas gástricas das regiões cárdica, fúndica e pilórica. As células produtoras de gastrina foram identificadas em grande quantidade entre as células da região pilórica e no epitélio de revestimento do duodeno, e em número reduzido na região cárdica. O enteroglucagon foi identificado na superfície dos cílios, no epitélio de revestimento da mucosa esofágica, nas fibras nervosas da lâmina própria e submucosa; no estômago estava localizado entre as células das glândulas cárdicas e fúndicas em número moderado, e aparentemente em maior quantidade nas pilóricas. No duodeno, o enteroglucagon foi identificado na superfície dos microvilos, no epitélio de revestimento, nas fibras nervosas da lâmina própria e submucosa. A motilina foi identificada apenas entre as fibras nervosas dos órgãos estudados. Em conclusão, as células imunorreativas à serotonina, gastrina, enteroglucagon e motilina foram identificadas em todos os órgãos estudados. Entretanto, nos diferentes segmentos houve diferença quanto à distribuição e frequência dessas células
Kinosternon scorpioides is a freshwater turtle known as “jurará”. The aim of this work was to identify peptides serotonin, enteroglucagon, gastrin and motilin in esophagus, stomach and duodenum by immunohistochemical techniques. The results showed that immunoreactive cells for serotonin were presented in the epthitelium of distal region of the esophagus, in glands of cardiac, fundic and pyloric region of the stomach and in the surface of duodenum epithelium. These cells were classified as open and closed type. The cells immunoreactive for gastrin were abundant in pyloric region and duodenum epitheliun, few numerous in cardiac region and absent in the esophagus. Cells rich in enteroglugagon were identified in the cilia surface, in the epithelium of esophagus, among nervous fibers of lamina propria and submucosa; in the stomach, these cells were observed in small number cardiac and fundic glands, however in pyloric region they were abundant. In duodenum, they were observed in microvillus surface, epithelium and nervous fibers of both lamina propria and submucosa. On the other hand, cells that produce motilin were observed just among lamina propia and submucosa. In conclusion, cells rich in serotonin, gastrin, enteroglugagon and motilin were found in all studied organs. However it was observed differences in the segments regarding distribution and frequency of these cells
