Abstract:
ABSTRACT Using the FDC-P1 cell line expressing the exogenous macrophage colony-stimulating factor (M-CSF) receptor, Fms, we have analyzed the role of a new mammalian DOS/Gab-related signaling protein, called Gab3, in macrophage cell development of the mouse. Gab3 contains an amino-terminal pleckstrin homology domain, multiple potential sites for tyrosine phosphorylation and SH2 domain binding, and two major polyproline motifs potentially interacting with SH3 domains. Among the growing family of Gab proteins, Gab3 exhibits a unique and overlapping pattern of expression in tissues of the mouse compared with Gab1 and Gab2. Gab3 is more restricted to the hematopoietic tissues such as spleen and thymus but is detectable at progressively lower levels within heart, kidney, uterus, and brain. Like Gab2, Gab3 is tyrosine phosphorylated after M-CSF receptor stimulation and associates transiently with the SH2 domain-containing proteins p85 and SHP2. Overexpression of exogenous Gab3 in FD-Fms cells dramatically accelerates macrophage differentiation upon M-CSF stimulation. Unlike Gab2, which shows a constant mRNA expression level after M-CSF stimulation, Gab3 expression is initially absent or low in abundance in FD cells expressing the wild-type Fms, but Gab3 mRNA levels are increased upon M-CSF stimulation. Moreover, M-CSF stimulation of FD-FmsY807F cells (which grow but do not differentiate) fails to increase Gab3 expression. These results suggest that Gab3 is important for macrophage differentiation and that differentiation requires the early phosphorylation of Gab2 followed by induction and subsequent phosphorylation of Gab3.

Abstract:

Abstract:
Human beings must constantly adapt to an uncertain and mutable world by generating efficient behavioral strategies to pursue their goals. The complexity of this task increases in interactive contexts, where the outcomes of our actions depend also on the choices of other agents. When the environment does not provide reliable feedback, the effectiveness of behavioral strategies rests on the ability to handle available knowledge: agents have indeed to extract relevant information from noisy signals and build an exhaustive representation of the set of potential actions and outcomes available to themselves and to others. Individual differences in the implementation of these information- processing operations may underlie behavioral heterogeneity in several judgment and decision making tasks. Here we report three eye-tracking studies revealing the existence of distinct information-processing strategies in different individuals. Study 1 explores inter-individual differences in the generation of relational representations of interdependent contingencies. In Study 2 and Study 3, we move towards social contexts to investigate the mechanisms of strategy generation underlying strategic behavior in interaction. Our findings indicate that gaze data can disclose individual differences in the process of spontaneous strategy generation in both individual and interacting settings. We also report results suggesting that the emergence of unsophisticated information-processing strategies is associated with cognitive style. Moreover, we show that the attentional mechanisms sustaining the generation of unsophisticated strategies can be reconsidered and updated under the impact of endogenous and exogenous cues revealing the existence of alternative information-processing behaviors.

Abstract:
Summary Quickly and correctly receiving and interpreting the information transmitted from the eyes of others is an essential cognitive ability for humans. Studies have shown that observers can automatically shift their attention in the direction signalled by the averted gaze. This thesis focused on the modulatory effect of ethnicity on gaze-driven orienting of attention, particularly under a cross-cultural context. In Chapter 1, we give a background introduction to gaze-driven orienting of attention and its social modulators. Next, in Chapter 2, the focus is to explore the modulatory effect of ethnicity on gaze-driven orienting in Caucasian and Asian individuals. In particular, five experiments are presented which have been conducted by applying the gaze-cueing paradigm to investigate the influence of faces belonging to different ethnicities on the gaze-cueing effect in Italian and Chinese participants. The results showed that White individuals exhibited a stronger gaze-cueing effect for White rather than Black faces, but they exhibited a similar gaze-cueing effect for White and Asian faces. As for the Asian individuals, they showed a stronger gaze-cueing effect for White (outgroup) rather than Asian (ingroup) faces. It is the first time that an asymmetric gaze-cueing effect was reported in Asian individuals. Thus, in Chapter 3, an eye-tracking technique and the oculomotor interference paradigm were adopted to further investigate this phenomenon. The results from two eye movement experiments revealed consistent patterns, similar to what we found in the experiments illustrated in Chapter 2, suggesting that the social knowledge activated by the ethnicity of the face (e.g., perceived social status of the different ethnic groups) plays a role in modulating gaze-driven orienting. Finally, in Chapter 4, the influence of masked faces on gaze-cueing effect in different cultural contexts was investigated through online experiments, due to the COVID-19 pandemic. The results showed that the gaze-cueing effect emerged irrespective of whether masked or unmasked faces were used, in both Italian and Chinese individuals. Chapter 5 is dedicated to highlighting and summarizing our findings, discussing the limitations of our work, and identifying future routes by using different experimental designs and new techniques with the aim of extending upon the present results.

Abstract:
Electrical and Computer Engineering
Ph.D.
Game theory is the study of tactical interactions involving conflicts and cooperations among multiple decision makers called players with applications in diverse disciplines such as economics, biology, management, communication networks, electric power systems and control. This dissertation studies a statistical differential game problem where finite N players optimize their system performance by shaping the distribution of their cost function through cost cumulants. This research integrates game theory with statistical optimal control theory and considers a statistical Nash non-cooperative nonzero-sum game for a nonlinear dynamic system with nonquadratic cost functions. The objective of the statistical Nash game is to find the equilibrium solution where no player has the incentive to deviate once other players maintain their equilibrium strategy. The necessary condition for the existence of the Nash equilibrium solution is given for the m-th cumulant cost optimization using the Hamilton-Jacobi-Bellman (HJB) equations. In addition, the sufficient condition which is the verification theorem for the existence of Nash equilibrium solution is given for the m-th cumulant cost optimization using the Hamilton-Jacobi-Bellman (HJB) equations. However, solving the HJB equations even for relatively low dimensional game problem is not trivial, we propose to use neural network approximate method to find the solution of the HJB partial differential equations for the statistical game problem. Convergence proof of the neural network approximate method solution to exact solution is given. In addition, numerical examples are provided for the statistical game to demonstrate the applicability of the proposed theoretical developments.

Abstract:
AbstractThe unique tidal bore of Qiantang River is called “the first tidal bore in the world”. The outlet of Hangzhou Babao lock of Beijing-Hangzhou Canal is located in the strong tidal bore area of Qiantang River estuary. This paper focuses on the characteristics of Qiantang River tidal bore, the selection of lock gates under the action of strong tidal bore, and the navigation safety control mechanism of lock. The results show that during the spring tide period, the maximum height of the head of tidal bore at Babao lock of Qiantang River can reach 2.5 m, and the maximum tidal bore pressure on the front of the lock can reach 90–100 kPa. In order to ensure the safe operation of ship lock, the design put forward for the Qiantang river side lock head adopts the miter gates and tidal gate combination plan, namely to put a flat gate on the outside of miter gate, with gate mouth 23 m wide and height of 13 m, to withstand the enormous impact of tidal bore in a large tide by closing the tide gate before the arrival of the tide and opening the tide gate afterward, while only using the miter gate as the lock operation gate the rest of the time during which there are no tidal surges. According to the patterns of tidal bore propagation, the intelligent prediction and early warning mechanism of tidal bore, the operation rules, and the automatic control system of miter gate and tide gate are studied and established to ensure the safe and efficient operation of the lock.

Abstract:
AbstractWe provide an algorithm to solve Rabin and Streett games over graphs with n vertices, m edges, and k colours that runs in $$\tilde{O}\left( mn(k!)^{1+o(1)} \right) $$ O ~ m n ( k ! ) 1 + o ( 1 ) time and $$O(nk\log k \log n)$$ O ( n k log k log n ) space, where $$\tilde{O}$$ O ~ hides poly-logarithmic factors. Our algorithm is an improvement by a super quadratic dependence on k! from the currently best known run time of $$O\left( mn^2(k!)^{2+o(1)}\right) $$ O m n 2 ( k ! ) 2 + o ( 1 ) , obtained by converting a Rabin game into a parity game, while simultaneously improving its exponential space requirement. Our main technical ingredient is a characterisation of progress measures for Rabin games using colourful trees and a combinatorial construction of succinctly-represented, universal colourful trees. Colourful universal trees are generalisations of universal trees used by Jurdziński and Lazić (2017) to solve parity games, as well as of Rabin progress measures of Klarlund and Kozen (1991). Our algorithm for Rabin games is a progress measure lifting algorithm where the lifting is performed on succinct, colourful, universal trees.

Abstract:
Pressure gages are used in many fluid measurements applications. One such application is the measurement of wave impact pressures on structures. This application poses a unique problem of measuring pressures in a “wet to dry” environment. Often there is a thermal drift component in the pressure readings that makes it difficult to extract the actual pressure rise due to wave loading. These types of measurements also require high response rates to measure the detail of the short duration impacts, usually on the order of one to twenty kilohertz. Several bench tests were carried out in at the Naval Surface Warfare Center, Carderock Division, in 2008 to investigate various types of gages to find a robust gage that could withstand this type of application. Three different gages were used in this investigation. The first sensor (gage 1) is a dynamics general purpose ICP (integrated circuit piezoelectric) pressure sensor, capable of making very high frequency dynamic pressure measurements, rated to 200 psi. The second sensor (gage 2) is a voltage compensated, media isolated piezoresistive sensor, rated to 15 psi. This gage had a pressure port which was filled with water during testing to eliminate air compression effects. The third sensor (gage 3) is a semiconductor pressure gage rated to 25 psi (172.4 kPa). A water “drop” test setup was constructed of 2 inch (5.1 cm) PVC pipe. The pressure gages were mounted to the bottom of the setup facing up, and the pipe was filled from the top, with a quick acting gate valve located 2 feet (0.61 m) from the pressure sensors. Once the pipe was filled with the desired amount of water, the gate valve was opened as quickly as possible, and the impact force was measured. Vent pipes were mounted to a “cross” fitting in the vertical pipe which allowed for the air to escape. Several water “drop” tests were performed with this setup. From these tests, the thermal drift of gage 1 is evident. Gage 3 exhibits similar behavior. Gage 2 captures the water pressure impact, and then returns to a small positive static pressure as a result of the water that is sitting above it. Of the three sensors, gage 2 appears to be the most temperature stable.

Abstract:
The b-matching game is a cooperative game defined on a graph. The game generalizes the matching game to allow each individual to have more than one partner. The game has several applications, such as the roommate assignment, the multi-item version of the seller-buyer assignment, and the international kidney exchange. Compared with the standard matching game, the b-matching game is computationally hard. In particular, the core non-emptiness problem and the core membership problem are co-NP-hard. Therefore, we focus on the convexity of the game, which is a sufficient condition of the core non-emptiness and often more tractable concept than the core non-emptiness. It also has several additional benefits. In this study, we give a necessary and sufficient condition of the convexity of the b-matching game. This condition also gives an O(n log n + m α(n)) time algorithm to determine whether a given game is convex or not, where n and m are the number of vertices and edges of a given graph, respectively, and α(・) is the inverse-Ackermann function. Using our characterization, we also give a polynomial-time algorithm to compute the Shapley value of a convex b-matching game.

Full text
Abstract:
Chemical composition and biological role of the surface coat (SC) of the root-knot nematodes, Meloidogyne spp. are described. SC proteins of M. incognita race 3 infective juveniles (J2) were characterized by electrophoresis and western blotting of extracts from radioiodine and biotin-labelled nematodes. J2 labelled with radioiodine and biotin released 125I and biotin-labelled molecules into water after 20 hours incubation, indicating that SC proteins may be loosely attached to the nematode. Antiserum to the principal protein reacted with the surface of live J2 and with surface proteins previously separated by electrophoresis. Human red blood cells (HRBC) adhered to J2 of several tylenchid nematodes over the entire nematode body. HRBC adhered also to nylon fibers coated with SC extracted from M. javanica J2; binding was Ca++/Mg++ dependent, and decreased when the nylon fibers were coated with bovine serum albumin, or pre-incubated with fucose and mannose. These experiments support a working hypothesis that RBC adhesion involves carbohydrate moieties of HRBC and carbohydrate-recognition domain(s) (CRD) distributed on the nematode surface. To our knowledge, this is the first report of a surface CRD i the phylum Nematoda. Gold-conjugated lectins and neoglycoproteins combined with silver enhancement have been used for the detection of carbohydrates and CRD, respectively, on the SC of M. javanica J2. Biotin reagents were used to trace surface proteins, specifically, on live J2. The labile and transitory nature of the SC was demonstrated by the dynamics of HRBC adherence to detergent-treated J2, J2 at different ages or fresh-hatched J2 held at various temperatures. SC recovery was demonstrated also by a SDS-PAGE profile. Monoclonal antibodies developed to a cuticular protein of M. incognita J2 gave a slight, but significant reduction in attachment of Pasteuria penetrans spores. Spore attachment as affected by several enzymes was inconsistent: alcian blue, which specifically blocks sulfyl groups, had no afffect on spore attachment. Treatment with cationized ferritin alone or catonized ferritin following monoclonal antibody caused significant decreases in spore attachment. Those results suggest a role in attachment by negatively charged groups.

Abstract:
Federal, state, and non-governmental partners resurveyed landbirds in the Ka?? Rainforest on the Island of Hawai?i in 2019. Point-transect distance sampling was conducted in Hawai?i Volcanoes National Park, Ka?? Forest Reserve, Kap?pala Forest Reserve, and Kap?pala Cooperative Game Management Area. This is the first comprehensive survey of the region since 2008. Through a collaborative effort, monitoring transects were consolidated to provide a consistent, repeatable sampling frame for future surveys. We estimated landbird distribution, density, and abundance for eight surveys since 1976. Changes in species-specific densities were assessed using a log-linear regression to determine trends of the six most recent surveys since 2002. Trends were assessed in three strata: low elevation (<1,500 m), high elevation (?1,500 m), and total survey area. We detected 23 species and there were sufficient detections of 11 species to estimate density and abundance. We provided indices of relative occurrence for the remaining 12 species detected. Native birds vulnerable to avian malaria (Plasmodium relictum) were absent or trended downward in the low elevation stratum. The federally endangered ?Akiap?l??au (Hemignathus wilsoni), ?Alaw? (Hawai?i Creeper, Loxops mana) and Hawai?i ??kepa (Loxops coccineus) were restricted to areas above 1,500 m elevation, likely due to the low prevalence of the cold-intolerant vector of avian malaria, the southern house mosquito (Culex quinquefasciatus), in this high elevation stratum. However, downward trends of the federally threatened ?I?iwi (Drepanis coccinea) above 1,500 m elevation may indicate an expansion in range of mosquitoes due to warming trends. Thought to be more resilient to avian malaria, the ?Apapane (Himatione sanguinea) and Hawai?i ?Amakihi (Chlorodrepanis virens virens) trended upward in all three strata. The Hawai?i ?Elepaio (Chasiempis sandwichensis) trended downward in each stratum. The ??ma?o (Myadestes obscurus) trended upward in the low stratum and downward in high. Trends were inconclusive for Hawai?i ??kepa, but favorable for ?Alaw?, which trended upward in its restricted range. There were insufficient detections of ?Akiap?l??au in 2019 to estimate density, but the 2008 abundance estimate of 691 ? 410 (SE) birds was the lowest since 1976. Trends were mixed for the non-native Warbling White-eye (Zosterops japonicus), Red-billed Leiothrix (Leiothrix lutea), and Northern Cardinal (Cardinalis cardinalis). Overall, upward trends of ?Apapane and Hawai?i ?Amakihi throughout the survey area, ??ma?o in the low stratum, and ?Alaw? in the high stratum, suggests management actions to protect native forest bird habitat are having a positive benefit. Nevertheless, avian malaria is predicted to have an increasing impact on vulnerable species as mosquitoes expand into higher elevations.

Abstract:
The US Army Corps of Engineers (USACE) maintains breakwaters in Milwaukee Harbor. USACE’s Engineering With Nature® (EWN®) breakwater demonstration project created rocky aquatic habitat with cobbles (10–20 cm) covering boulders (6–8 metric tons) along a 152 m section. A prolific population of Hemimysis anomala, an introduced Pontocaspian mysid and important food source for local pelagic fishes, was significantly (p < .05) more abundant on cobbles versus boulders. Food-habits data of alewife (Alosa pseudoharengus) and rainbow smelt (Osmerus mordax) provided evidence that H. anomala were a common prey item. Night surveys and gill netting confirmed O. mordax preferred foraging on the cobbles (p < .05) and consumed more H. anomala than at the reference site (p < .05). H. anomala comprised a significant portion of the diets of young-of-the-year (YOY) yellow perch (Perca flavescens), YOY largemouth bass (Micropterus salmoides), and juvenile rock bass (Ambloplites rupestris) caught on the breakwater. The natural features’ construction on the breakwater increased the available habitat for this benthopelagic macroinvertebrate and created a novel ecosystem benefiting forage fish and a nursery habitat benefiting nearshore game fish juveniles. These data will encourage the application of EWN concepts during structural repairs at other built navigation infrastructure.

Raymond, Kara, Laura Palacios, Cheryl McIntyre, and Evan Gwilliam. Status of climate and water resources at Saguaro National Park: Water year 2019. Edited by Alice Wondrak Biel. National Park Service, December 2021. http://dx.doi.org/10.36967/nrr-2288717.

Full text
Abstract:
Climate and hydrology are major drivers of ecosystems. They dramatically shape ecosystem structure and function, particularly in arid and semi-arid ecosystems. Understanding changes in climate, groundwater, and water quality and quantity is central to assessing the condition of park biota and key cultural resources. The Sonoran Desert Network collects data on climate, groundwater, and surface water at 11 National Park Service units in south-ern Arizona and New Mexico. This report provides an integrated look at climate, groundwater, and springs conditions at Saguaro National Park (NP) during water year 2019 (October 2018–September 2019). Annual rainfall in the Rincon Mountain District was 27.36" (69.49 cm) at the Mica Mountain RAWS station and 12.89" (32.74 cm) at the Desert Research Learning Center Davis station. February was the wettest month, accounting for nearly one-quarter of the annual rainfall at both stations. Each station recorded extreme precipitation events (>1") on three days. Mean monthly maximum and minimum air temperatures were 25.6°F (-3.6°C) and 78.1°F (25.6°C), respectively, at the Mica Mountain station, and 37.7°F (3.2°C) and 102.3°F (39.1°C), respectively, at the Desert Research Learning Center station. Overall temperatures in WY2019 were cooler than the mean for the entire record. The reconnaissance drought index for the Mica Mountain station indicated wetter conditions than average in WY2019. Both of the park’s NOAA COOP stations (one in each district) had large data gaps, partially due to the 35-day federal government shutdown in December and January. For this reason, climate conditions for the Tucson Mountain District are not reported. The mean groundwater level at well WSW-1 in WY2019 was higher than the mean for WY2018. The water level has generally been increasing since 2005, reflecting the continued aquifer recovery since the Central Avra Valley Storage and Recovery Project came online, recharging Central Arizona Project water. Water levels at the Red Hills well generally de-clined starting in fall WY2019, continuing through spring. Monsoon storms led to rapid water level increases. Peak water level occurred on September 18. The Madrona Pack Base well water level in WY2019 remained above 10 feet (3.05 m) below measuring point (bmp) in the fall and winter, followed by a steep decline starting in May and continuing until the end of September, when the water level rebounded following a three-day rain event. The high-est water level was recorded on February 15. Median water levels in the wells in the middle reach of Rincon Creek in WY2019 were higher than the medians for WY2018 (+0.18–0.68 ft/0.05–0.21 m), but still generally lower than 6.6 feet (2 m) bgs, the mean depth-to-water required to sustain juvenile cottonwood and willow trees. RC-7 was dry in June–September, and RC-4 was dry in only September. RC-5, RC-6 and Well 633106 did not go dry, and varied approximately 3–4 feet (1 m). Eleven springs were monitored in the Rincon Mountain District in WY2019. Most springs had relatively few indications of anthropogenic or natural disturbance. Anthropogenic disturbance included spring boxes or other modifications to flow. Examples of natural disturbance included game trails and scat. In addition, several sites exhibited slight disturbance from fires (e.g., burned woody debris and adjacent fire-scarred trees) and evidence of high-flow events. Crews observed 1–7 taxa of facultative/obligate wetland plants and 0–3 invasive non-native species at each spring. Across the springs, crews observed four non-native plant species: rose natal grass (Melinis repens), Kentucky bluegrass (Poa pratensis), crimson fountaingrass (Cenchrus setaceus), and red brome (Bromus rubens). Baseline data on water quality and chemistry were collected at all springs. It is likely that that all springs had surface water for at least some part of WY2019. However, temperature sensors to estimate surface water persistence failed...
