Abstract:
As the nineteenth century drew to a close, people living in coastal Louisiana noticed that local rodents called muskrats were rapidly increasing and quickly becoming pests by digging up crops and into levees. Property owners soon demanded their elimination, but to the ire of many, Louisiana officials chose to develop a market for muskrat fur and protect its supply through management laws. The state sought the cooperation of trappers in order to maintain global demand, but when nutria were released alongside the muskrat, the ecological balance of the marsh was permanently altered. Muskrats shrank back into obscurity, and trappers struggled to embrace the nutria as a substitute. This thesis will trace the Louisiana muskrat industry's development starting with its rise in the 1890s, continuing through its years as a leading furbearer, and ending with its replacement by the nutria in the 1960s.

The semiconductor industry is an exciting and challenging industry. Strong demand at the application end, plus the high capital intensity and rapid technological innovation in manufacturing, makes it difficult to manage supply chain planning and investment in technology transitions. Better understanding the essence of the industry dynamics will help firms win competitive advantages in this turbulent market. In this thesis, we will study semiconductor industry dynamics from three different angles: quantitative modeling, industry dynamics simulation, and strategic analysis. First, we develop a stochastic linear optimization model to address the supplier's "order fulfillment dilemma" suggested by previous empirical studies. The model provides optimal equipment production decisions that minimize the total cost under stochastic demand. To solve the large scale problem, we introduce the Bender's Decomposition, which is proven to outperform the pure Simplex method. Furthermore, we extend the basic model to multiple periods, allowing equipment inventory planning over a period of time. Second, we build a macro-level industry dynamic model using the methodology of System Dynamics. The model includes components of electronics demand projection, fabrication capacity allocation, fabrication cost structure, technology roadmapping as well as equipment production and R&D. The model generates projections of demand , industry productivity, schedule of building new fabrication, adoption of the latest process technology, etc., which are validated by actual industry data. In addition, we devise a control panel in the software that enables the users to implement flexible scenario and sensitivity analysis. Third, we propose a strategic framework for companies to pinpoint the root causes of the supply-demand mismatch problem.
(cont.) This framework considers long lead times, fast clockspeeds, Moore's Law, and risky product and technology, which transitions contribute to the pronounced volatility amplification occurring in the semiconductor industry. This framework, along with several industry successful practices, will assist companies to mitigate the demand volatility and improve their supply chain performance.

Abstract:
At a time when environmental mitigation is firmly at the centre of the agenda for sustainable development, there is no shortage of research in the field of green logistics. However, little has been done in an attempt to provide integrated solutions for industry, based on a practical assessment of the interrelationships between specific measures. This research investigates the current status of the British logistics industry in terms of its environmental sustainability, by examining 14 sustainable measures that feature strongly in contemporary logistics practice and policies, and determines the interrelationships among them. The primary data was collected using a combined approach involving a postal questionnaire survey and in-depth company interviews. The survey covered both logistics services providers (LSPs) and logistics service users (LSUs), together with the targeted inclusion of more specific actors within the logistics industry. Through analysis of the sustainable practice and implementation process as seen in the experiences and judgments of key actors in the logistics industry, it is shown that the role of the actors as either logistics providers or users has a certain impact on their perceptions of, and behaviours in, sustainable logistics. Particular characteristics of the company and the sector it belongs to also exert influence, to various extents, on its response to sustainability. The findings also highlight cost-effectiveness as another critical factor determining companies’ sustainability policies. On the basis of the assessment of the measures, in particular their effectiveness and cost efficiency, the thesis concludes with proposals for sustainable packages constructed from different perspectives, along with suggestions for their implementation. This evidence-based research thus informs policy-makers of appropriate and viable sustainable strategies with the right incentives in various circumstances, and the potential to bring about tangible improvements in environmental performance.

Industry 4.0 is at the moment one of the most talked-about topics amongst industry companies. Management teams of discuss the area as something they want to implement, but since it is an ongoing industrial revolution it cannot be implemented straight away. This study is looking into the subject of Industry 4.0 and the identification of nine different technology drivers that a manufacturing company can use as tools in a production environment. However, what is less known is how the tools can be applied to a global company and its different cultures. Because even though Industry 4.0 is about decentralization and giving responsibility to every co-worker, they need to work with a common framework, thus standardization. A company that has started to look into Industry 4.0 is Lantmännen Unibake, who is a global bakery group. Therefore, they serve as the case company in this exploratory study where the goal is to conduct a pre-study that gives them, and the food industry, information and guidelines on how Industry 4.0 applies to their industry. Hence, what improvements it can lead to, what tools that will be suitable, and what the prerequisites for a successful Industry 4.0- transformation will be. The method to answer the research question starts with conducting a comprehensive literature study, followed by several interviews around Europe, and ending in an analysis of the current state with help from the literature framework. The analysis has ended in a conclusion that there is no single or easy answer to the questions since food industries might vary a lot from one to another. Although, in this case, the answer would be that six out of nine tools would be applicable in a global food-supply-chain, some needing more preparatory work than others. By implementing these tools, companies will see more engaged employees that are prone to take autonomous decisions, more accurate production planning, proactive rather than reactive working ways, fewer defects, and higher customer satisfaction. Regarding the prerequisites for transformation, the answer depends on what tools that are used. But what all cases have in common is that there is a need for aware and engaged employees. Everyone will have to be in on the change, understanding why and how, and what benefits that will come out of it. For future studies, it is suggested to look into how maturity models can be applied to these kinds of issues smoothly. The existing maturity models for Industry 4.0 are very complicated and difficult to apply; thus, they would need some development. Another area would be a cost model of the different tools, as well as a comparison of the tools and food regulations.

Abstract:
Researchers often regard remanufacturing as an environmentally beneficial end-of-life option. There have been, however, few environmental measurements performed in the area. The aim of this paper is to identify general environmental pros and cons with remanufacturing. This is done through the analysis of practical examples in remanufacturing industries. Life Cycle Assessment methodology has been used for the environmental validations. The first conclusion, based on the industrial cases and the literature review, is that remanufacturing is preferable from a material resource perspective when compared with manufacturing of new products. The second conclusion is that remanufacturing is preferable from a more overarching perspective for some of the investigated cases, but it is not possible to draw any general conclusions since the companies studied are few and benefits from remanufacturing are highly context-related.

There are tremendous potential applications for blockchain technology, an innovative distributed ledger database system, within the real estate industry. This paper will explore one aspect-recording property titles- by comparing the benefits and limitations of a blockchain with those of the current record keeping system. This paper will begin with a brief overview of the current state of the title recording system in the U.S. followed by a deeper look into the procedures of one rapidly growing American city, Nashville (Davidson County), Tennessee. The goal is to understand current real estate title systems and technologies in order to identify their benefits and limitations. Next, this paper will introduce the concept of blockchain technology, starting with a high level technical overview of how the technology works, as well as its benefits and limitations. It will also examine Bitcoin, which operates on the largest blockchain, as a potential model, whose practical applications may be adapted in creating a more efficient and safer title registry system. Recommendations will then be made for possible methods of implementing a blockchain-based registry and how its use might change the way real estate title transactions are handled in Davidson County, TN in order to determine if the collective benefits outweigh the costs. The research to date leads to the following conclusions: A blockchain title recording system is the future of title record keeping and would provide immediate benefits over the current title recording system, with additional benefits accruing in the future as blockchain technology grows in acceptance. However, at the moment, these benefits do not yet outweigh the costs and challenges associated with implementing a prototype blockchain title registry system in Davidson County, or elsewhere in the country. That being said, steps can, and should be taken now to lay the foundation for a blockchain system.

La question principale que dispose cette thèse est relative à la restructuration du secteur et des entreprises chimiques à cause des nouvelles frontières stratégiques, notamment concurrentielles, technologiques, financières et institutionnelles. L’objectif principal c’est de comprendre et caractériser les comportements hétérogènes des firmes chimiques, les en classent en groupes distincts. La première prémisse de cette thèse est que l'apprentissage cumulatif scientifique ou les DPI (droits de propriété intellectuelle) ne sont plus les principaux obstacles aux nouveaux entrants dans le secteur; elles ont été additionnées à la suprématie des finances, certifications HQES (santé, qualité, environnement, sécurité ) et les valeurs intangibles (la marque, la chaîne d'approvisionnement, réseau, les clusters, la compréhension des clients, le marketing, la responsabilité sociale, la gestion et de main-d'œuvre qualifiée). Le second postulat c’est que l’environnement concurrentiel et technologique a recrée des nouveaux limites stratégiques en partagent les firmes en sous-secteurs comme les sciences hybrides, sciences de la vie, substances de base ou les produits chimiques spécialisés. Cette nouvelle configuration induit les opérations de l'acteur dans leur portefeuille de produits, leurs actifs, production, R&D, F&A, réseau, JV, alliances, licences, connaissances spécifiques et le marketing. The principal matter that disposes this thesis is concerning the changes in the chemical firms for “new strategic boundaries”. The main objective is to understand and characterize the heterogeneous chemical companies’ strategies and to classify in distinct groups. The first premise of this thesis is that the accumulative scientific learning or IPR (Intellectual property rights) is not anymore the main barrier to new entrants into the segment; it was replaced by finance supremacy, HESQ (health, environmental, safety and quality) certifications and intangible values (brand, supply chain, network, clusters, customer understanding, marketing, social responsibility, management and skilled labor). Several new players appeared supported by national governments and using licensees or technologies from specialized engineering firms (SEF), raw material suppliers & equipment fabricators to produce locally and replace imported products, furthermore those new players through foreign direct investment( FDI), merger & acquisition (M&A) or joint ventures (JV) expand their operations abroad in direct concurrence with the traditional groups. Second premise is that the latest competitive environment are recreating new strategic firms’ boundaries separated in sub sectors like hybrid science, basic or specialty chemical inducing the main player’s actions in product portfolio , fixed assets, process, R&D , M&A, network, JV, alliances, licenses , knowledge capital and marketing. The thesis concludes that the changes in the chemical sector evolve inside a new framework typify at 4 main axes: value creation, globalization, restructuring and regulation

While the pace of technical innovation in the semiconductor industry continues to accelerate, business processes and supply chain techniques have not kept up. Microprocessor performance improvement continues to follow Moore's Law, but increased variability has complicated efforts to accurately forecast demand and set inventory targets. Products are becoming more complex, often containing assemblies of multiple parts. Lifecycles are becoming shorter; made possible by technology breakthroughs and efficient manufacturing ramp-ups. Demand and supply are ever more stochastic and non-stationary. Inventory is one of the few ways that a firm can buffer themselves from the inherent and increasing variability, while still meeting required service levels. We explore the sources of the variability in the semiconductor supply chain. On the supply side, we evaluate variability in throughput time, yield and other factors not explicitly considered in standard models. Here, we primarily focus on the natural stochasticity of the manufacturing process and disregard the variability arising from forecasting of these supply parameters. For demand, the natural stochastic process is not well understood, so we evaluate the forecast error and use it as a proxy for demand variability. We then apply these data to the base-stock model - constrained by its associated assumptions - to calculate inventory targets required to meet a certain level of service. Using a two-node base-stock model in conjunction with the actual variability data, we develop inventory estimates across the network and evaluate tradeoffs between different inventory strategies. We then determine what each variability parameter contributes to inventory. The combination of a simple yet representative
(cont.) model of the semiconductor supply chain with actual data from the variability characterization provides the tools to make powerful recommendations to reduce variability and decrease inventories throughout the supply network.

Authentic approaches to design education are typically defined as experiences centered on industry involvement. This industry connection is commonly either in the form of projects provided by industry partners or practicing engineers that serve as mentors to students. After exploring the goals and current practices of design education, this dissertation proposes an expanded definition of authentic design education: any design project with impact beyond the classroom environment that encourages the development of a student's self-identity as an engineer. To investigate the potential benefits afforded by an expanded definition of authentic design, a new design class was developed, taught, and evaluated across four years. The class, entitled Design for the Wilderness, was developed with a focus on projects that have impact beyond the classroom environment. Students were required to design and build products that they relied on while traveling in remote wilderness environments.
These impactful projects required students to experience the results of their design decisions. Building on our experiences implementing Design for the Wilderness, a curricular approach of Design for Use is introduced that requires students to use products developed by their peers. Design for Use helps increase students' understanding of human-centered design principles by encouraging students to confront the interplay between their intentions when designing a product and their experiences when failing to understand the intentions behind products designed by their peers. This dissertation also considers a mechanical engineering capstone design class (MIT's 2.009). An interesting outcome of this class is that some teams continue to work on commercializing their products after the semester ends. Team characteristics most strongly correlated with persisting on product development beyond the end of the class are related to healthy team dynamics and a positive social environment.
Teams that persisted spent more of their time working together, had fewer teammates that worked significantly more or less than the team average, and spent more of their time simply "hanging out" in lab. Drawing on our findings from investigating multiple approaches to authentic design education, recommendations are made for the future development of effective engineering design classes.

The objective of this thesis is to develop a conceptual design of a computerized environment for detailed design of construction activities associated with projects characterized by significant repetition. High-rise building construction is used as the example of repetitive construction projects. The construction cycle design of a typical floor structure is studied to gain an understanding of the difficulty and complexity involved in the activity design process. Modeling techniques currently used in construction planning, modeling techniques developed in the field of operations research, and assembly line balancing techniques used in industrial engineering are reviewed to determine their applicability for detailed construction cycle design. Using the concept of decision support systems developed in the fields of management science and knowledge engineering for solving ill-structured and ill-defined problems, a conceptual design of a decision support system for construction cycle design is developed.

The Electricity Industry Reforms (EIR) is a worldwidephenomenon that is inducing an intensive business orientedcontext in the organization, processes and functions of theElectricity Industry (EI). The electric power grid is nowtreated as the electricity market, the consumer as thecustomer, the power system activities are related to marketactors (i.e. generators, distributors and retailers) and theElectricity Industry Control Centers (EICC) provide theessential coordination and economic trade functions andtransactions.

The EICC are looking for solutions to introduce, amongstothers, emerging information processes in the business orientedcontext with all the Electricity Industry actors without losingthe technical reliability of the EI. The EIR is delineated as amajor change on the traditional EI relying heavily oninformation exchange amongst the market actors.

In the Central American countries, these EIR started around1996. The EICC in this region are facing the demanded changeswith information legacy systems, in operation long time beforethe EIR and considered already obsolete.

This thesis exploresand summarizes, as a researchcontribution, the main problems at the EICCs in CentralAmerica, in managing the incorporation of emerging informationproc-esses.

An Information System Migration Framework (ISMF) for theEICC is proposed as a re-search contribution and solution tothe problems identified. The ISMF here presented provides a setof steps and guidelines to follow for managing any emerginginformation processes in a systematic, feasible and reliableway. The ISMF is a continuous description of emerginginformation processes, risk assessments, requirementelicitation and specifica-tions with traceable and incrementalimplementations without completely replacing the system. TheISMF has as fundamental characteristic that provides a feasibleview of the current operation of the EICC, a continuous anditerative process of controlled changes and a systematicprocess to update the EICC operation model with a set of stepsthat are implementation independent, technology independent,process ori-ented and user centered.

The ISMF’s feasibility, replicability and useracceptance has partially tested at the EICC in El Salvador andNicaragua.

Due to industrial development and technological innovation, many problems arise from the increasing number of electronics devices that need to be managed at end of life. The two main market failures related to waste electrical and electronic equipment (WEEE) are economic loss and environmental externalities. Incomplete recovery of materials in waste mobile phones results in a great amount of economic loss. Many studies aim to improve the situation by characterizing metals within printed circuit boards (PCBs). My work focuses on the evolving composition and the flow of materials located outside of PCBs. I have discovered significant economic potential of non-PCB metals, and provide suggestions for optimization of different preprocessing steps. I recommend that preprocessors pay special attention to precious metals in fine shredding. And I have also provided strategies for plastics recycling by forecasting the demand and supply of it in Portugal. Besides opportunities for addressing environmental impact in the end-of-life stage of electronics products, there are also opportunities in other life cycle stages such as GHG savings by the enablement of semiconductors in the use stage. I identify six areas of GHG savings and provide insights for more informed policy to better evaluate the influence of technologies in the electronics industry.

The body of this research seeks to create an Industry Technology Roadmap for one segment of the U.S. textile and apparel industry, specifically, the flushable pre-moistened wipes industry. Industry Technology Roadmapping is a new type of strategic planning method. Previous technology forecasting methods allowed planners to identify several alternate future states or scenarios and provided uncertainty in either product needs or technological developments. In contrast, Industry Technology Roadmapping allows a company or an industry to proactively plan and prepare for its future by offering future goals, critical requirements and, finally, solutions to achieve the future goals. In addition, Roadmapping works as an effective organizational learning and knowledge creation tool and induces collaboration and new partnerships among companies as well. Currently, flushable pre-moistened nonwoven wipes emerge as new textile products and attempt to expand their future markets. During this research, industry expert interviews and surveys, consumer surveys, heavy user surveys, creation of an Internet blog, and literature review including a patent analysis were conducted for data gathering. The resulting Roadmap provides the nonwoven wipes industry valuable information about the potential future markets, required properties, the current environment of flushable wipes, including challenges, strength, and opportunities, and the solutions. These can help the U.S. nonwoven wipes industry to do informed investment into research, development and manufacturing facilities.

Our understanding of the effects that human production and consumption has on our planet and its resources has challenged us to think differently when developing new products. In response to these problems, Eco Design has been developed over the last few decades. Eco Design is a process integrated into product and engineering design that aims to lower the environmental impact of products across their life cycle, whilst not hindering design brief criteria such as function, price, performance, and quality. Research in Eco Design has focused mainly on the development of new tools and ways to implement Eco Design in industry. However, there is still little empirical knowledge today regarding the state of Eco Design implementation and practices in industry; in addition to the prerequisite needs and factors to successfully implement Eco Design. The aim of this research has been to review the level and type of Eco Design in the British Product Design industry and to identify recurrent themes helping or hindering implementation. This was achieved through the use of a pilot study followed by a two stage case study design, involving 20 cases and 57 participants across 65 interviews. The investigation and its analysis produced 12 confirmed themes, each generating their own drivers and barriers to Eco Design implementation. This research into Eco Design implementation provides a unique contribution and a timely insight into the Eco Design practices of the British Product Design industry today. The research also provides the novel contribution of identifying the drivers and barriers to implementing and sustaining Eco Design, as well as an understanding of the strengths and shortfalls of the current Eco Design processes and tools. These contributions to knowledge in the field of Eco Design will help future research formulate better solutions to implement Eco Design processes in the Product Design industry.

On September 6, 2008, at the start of what would amount to the greatest financial crisis since the Great Depression, the U.S. Government took two publicly traded Government Sponsored Enterprises (GSEs), Fannie Mae and Freddie Mac, and placed them into conservatorship. Operating losses by these entities had created strong market concerns about their ability to function, threatening to adversely affect the US housing finance market. Over the last two decades, the GSEs have played a major role in the multifamily industry, routinely accounting for approximately 30% of annual multifamily financing; at the height of the financial crisis in 2009, GSEs accounted for 86% of all new multifamily loans. Although GSEs account for a substantial portion of multifamily financing, very little research has been conducted in order to examine the future of the multifamily industry post-GSE conservatorship. In part, the reason for minimal research and a lack of media attention on this issue is that GSEs play a far greater role in terms of total dollars in the single-family residential market. This thesis helps to fill this void by closely examining the GSEs role in the multifamily market and surmising the impact to the multifamily sector post-GSE conservatorship. In order to examine this issue, this thesis focuses on the history and role of GSEs in the multifamily market; examines the guidelines, structure and securitization process of GSE multifamily loans; examines current multifamily market conditions and trends; provides a performance comparison of GSE securitized loans to other multifamily loans; and examines proposed GSE legislation. This thesis then synthesizes and prognosticates the current and potential future multifamily market conditions utilizing the 4-Quadrant model and the role the government should play in the multifamily finance market post-GSE conservatorship. This thesis concludes and surmises that reduced government involvement in the multifamily finance market will affect the multifamily industry, causing increased borrowing costs, decreased property values, and increased property value volatility. The extent to which the multifamily industry is affected depends upon the outcome of the GSE conservatorship, although it appears that increased multifamily demand due to favorable demographic trends may help to negate the impact of decreased GSE involvement in the multifamily industry for the foreseeable future.

This thesis provides insight into methods for data analysis of testing procedures to optimize the overall testing times within the electronics manufacturing industry. By analyzing each test regime within the manufacturing sequence individually, with the goal of overall test time reduction, better test system optimization may occur. Specifically, within Burn In testing it was found that failure rates were heavily dependent upon the device on/off cycle. Once discovered new test cycles were proposed to reduce overall test times by 50%. Once implemented such new test cycles increased early failure capture as expected. In addition, industry benchmarking studies showed new forms of testing such as Highly Accelerated Stress Testing (HAST) are pushing the product testing earlier into the product life cycle where in-process tests such as Burn In may be reduced. In the case of HAST testing, the tests are being conducted in the design phase reducing more costly Burn In testing in the production phase.

Zara is the world's leader in the fast-fashion industry and introduces over 10,000 unique designs to their stores each year. Zara's parent company, Inditex, reported profits of $2.2 billion in 2012, an increase of 27% for the year. They opened 360 new stores in over 50 markets across all of their brands last year. Zara contributes approximately 60% of these profits and has around 1,720 stores in over 80 countries worldwide. Zara is committed to meeting the needs of their customer through continuous improvement of their processes and systems. Though they continually improve their already advanced forecasting and distribution methods, there is significant variability in demand that remains challenging to predict. Due to this uncertainty in demand and the short life cycle of trendy clothing articles, it is imperative that Zara is able to quickly respond to changing demand patterns. After initial distribution, inventory can be redistributed among stores in order to satisfy their customers' demand and maximize sales. This critical step in the distribution process is known as inventory transfers. The purpose of this project was to develop a demand forecast model, optimization model and operational process to optimize and standardize these inventory transfers among the complex network of thousands of Zara stores. The key performance indicator was an increase in profit of at least three percent. The research process was first to identify the key decision-making criteria and variables affecting transfer decisions; second to use that criteria to build an optimization model to propose optimal redistribution of articles among stores; and third to prepare the roll-out and integration of the new approach in the existing operational process and IT system. This project required integration with Zara stakeholders across many functions including product management, buying, distribution and information technology. Crucial to the success of the project was remaining focused on these stakeholder needs to ensure the model would be easily adopted and fully implemented while also considering demand, costs, logistics, feasibility and many other factors. The new model provides a profit increase of 21 percent for those articles transferred and is the first model of this kind applied in retail supply chain management.

The most time consuming and capital intensive portion in the assembly of power electronic devices is the test system. A comprehensive test system including functional and stress screening technologies can significantly increase assembly times and more than double the capital investment required in a new assembly line. The primary purpose of the test system is to screen components for early life failures and to verify proper assembly. Determination of key performance characteristics and the resultant test system are developed during the product design phase and are seldom revised after the product has been released to manufacturing. This thesis explores best practices in testing methods and develops new methods to analyze test system performance. Both efforts were conducted in an effort to optimize existing test regimes. Upon completion of the above analyses the existing test sequence was reduced by 50%. This was primarily due to a discovery in the Burn In test cycle which indicated that failures correlated strongly with the on/off cycles inherent in the test sequence. A new test cycle was proposed to accommodate this finding and test results verified the initial hypothesis. Additionally, the summary of best practices identified new forms of product testing including Highly Accelerated Stress Testing (HAST), moving additional product testing into the development phase consequently reducing testing requirements during assembly.

Biopharmaceutical companies are focusing on operational efficiency more than ever before due to cost pressures, generic competition, complex pricing, regulations, and globalization. Due to the low probabilities of success of drug development and achieving market penetration, it is critical for a company to optimize the sizing of a manufacturing facility to achieve maximum capacity utilization while avoiding or minimizing non-productive, idle capacity. In order to achieve high manufacturing efficiency and minimize capital risks, many firms elect to outsource various phases of the drug development value chain. This thesis investigates the major drivers for vertical integration and outsourcing in the biopharmaceutical industry. This work examines outsourcing and vertical integration decisions including their financial, organizational, and strategic effects on the organization. It presents a methodology and framework for strategic sourcing decisions and includes a set of quantitative tools for assessing outsourcing options and guidelines for selecting, implementing, and managing relationships with contract manufacturers. These frameworks are applied to case studies conducted at the Genzyme Corporation. The strategic sourcing decisions tools developed in this work are highly customizable and can be adopted by other companies in the biopharmaceutical industry.

Historically the pharmaceutical industry has been highly profitable. However, the increasing regulatory requirements, bargaining power of buyers, and drug failures together with the threat of biosimilars and decreasing R&D productivity are creating challenges for research driven pharmaceutical companies. With future revenue growth uncertain, pharmaceutical companies must focus on cost reduction to sustain the profit margins needed to support research and development of new medicines. The lean methodology first developed by Toyota is recommended as a way to achieve operational success. A deep analysis of the current state of the pharmaceutical industry and the operational inefficiencies inherent in regulated drug production is provided. The renewed importance of operations within the pharmaceutical business model is explored through a case study of the biotechnology segment's leader, Amgen. Specifically, the design and initial rollout of the Amgen Process Excellence (APEX) initiative is studied. The APEX methodology is a six step process based on lean and six-sigma principles to guide operational improvement activities at Amgen. During the author's internship at the Rhode Island site the rollout of the APEX movement included a current state analysis of the site's financial and operational performance. As a result of this analysis, a prioritized list of improvement ideas was generated and incorporated into a future state vision for the site. Implementation of these improvement ideas is estimated to result in a reduction in cycle time by 55%, lower inventory levels, and the elimination of millions of dollars in waste. The following major conclusions were developed as a result of this work.
(cont.) First, substantial improvement opportunities exist within current pharmaceutical manufacturing. Second, pharmaceutical companies must build operational efficiencies into manufacturing process design. Lastly, operational excellence cannot simply be attained through the implementation of an improvement toolkit.

Both color keyed and color specific liquid primers have been used successfully in automotive paint application, reducing the use of costly topcoat materials. Generally, color keyed primer is close in color to the topcoat and is applied to the exterior surfaces of the vehicle body. Color specific primer identically matches the topcoat material color, and can be used as a replacement for topcoat on interior surfaces. Application of color powder paint primer would provide for additional cost savings, more environment-friendly manufacturing, and higher customer satisfaction. Recent technology breakthroughs have led to the technical feasibility of applying multicolor powder paint with full reclaim and therefore 100% material utilization. The field feasibility of applying multicolor powder paint in an automotive manufacturing facility is being assessed and validated by GM. More specifically, GM has developed the GM Canister Powder Paint Delivery System to meet the challenges of color powder paint application. In anticipation of multicolor powder paint application, GM is completing its Manufacturing Systems Qualification (MSQ) Process for the Canister System. Due to the newness of the enabling technology, GM is investigating all aspects of materials, process designs, facilities, operations, and people, for the implementation of color keyed/color specific powder paint primer into new GM paint shops. Similar to GM's MSQ Process, qualification requirements in the pharmaceutical and medical device industry attest to the importance of validation in the implementation of new manufacturing technology. Beyond its technical purposes, validation can serve as a bridge between development and operation. During development, formal and relational contracts with
(cont.) suppliers should be established to provide incentives for the supplier to perform throughout validation and commissioning. Involving the plant in validation along with suppliers is one means of education and thus empowerment, a key phase of organizational change as described by Professor Shoji Shiba. Leadership can use several change management techniques to help prepare the plant organization for operation of the new manufacturing technology.

Els mètodes microbiològics utilitzats a les companyies farmacèutiques per analitzar l’ambient del procés de fabricació així com els productes finals estan basats en mètodes de cultiu tradicional. La tecnologia d’aquests mètodes manca de precisió comparada amb les tecnologies més modernes de detecció i identificació microbiològica. Cada vegada, els requeriments dels controls microbiològics de productes i processos són més estrictes, el que fa que es necessiten mètodes cada vegada més ràpids i precisos. Dintre d’aquest context s’han desenvolupat els mètodes de microbiologia ràpida (RMM) per a la seva implantació a la industria farmacèutica, avalada per les diferents agències reguladores. Les diferents tecnologies en les quals es basen els RMM es coneixen a l’àmbit acadèmic des de fa dècades, no obstant la seva implantació i validació a les indústries farmacèutiques és relativament recent. La implementació de noves metodologies a l’àmbit farmacèutic està regulada per la normativa de bones practiques de fabricació (BPF), el qual va associat a una adequada validació de les tècniques. Aquesta tesi descriu la implementació de RMM al control microbiològic del procés de fabricació i productes a l’empresa Laboratoris Reig Jofré (RJF). El principal objectiu ha estat dividit en 3 blocs individuals: implementació d’un programa d’identificació microbiana per als aïllats del procés de fabricació i productes; implementació d’un mètode de detecció de microorganismes a l’aire de sales classificades per fabricació asèptica basat en fluorescència induïda per làser i l’ avaluació d’un sistema basat en citometria en fase sòlida per la detecció de microorganismes en productes filtrables. En general, els RMM implementats han contribuit a obtenir resultats de manera més ràpida, el qual permet mitigar el risc de contaminació en el mateix moment que ha sigut detectat. Aquest projecte assenta les bases per futures aplicacions de RMM en el procés de fabricació farmacèutica.
Microbiological testing plays an essential role in the Pharmaceutical Industry as it is an indicator of safety in drug products. The microbiological methods used in the pharmaceutical companies for testing the environment of the manufacturing process as well as the final products are based on traditional culture methods. These methods, although being appropriate for their intended use, rely on century-old technology that lacks accuracy when compared to most up-to-date methodologies for microbial detection and identification. Every time the requirement of microbiological testing of products and processes increases, arising the need of faster and more accurate methods. In this paradigm, rapid microbiological methods (RMM) are developed for their implementation in the pharmaceutical industry encouraged by regulatory agencies. The different technologies in which rapid microbiological methods are based have been known in the academic field for decades, however their implementation and validation in the pharmaceutical industries is relatively recent. Implementation of new methodologies in a pharmaceutical environment ruled under Good Manufacturing Practice (GMP) guidelines needs proper validation of the techniques involved. This thesis describes the implementation of RMM in the microbial monitoring of the pharmaceutical manufacturing process and products in Reig Jofré Laboratories (RJF). The main objective has been divided into three different blocks: implementation of a microbial identification program for the isolates found in the production process and products; implementation of a laser-induced fluorescence system for the detection of airborne microorganisms in cleanrooms for aseptic processing and evaluation of a solid-phase cytometry system for the detection of microorganisms in filterable products. In general, the RMM implemented have contributed to obtain faster results which allows to mitigate contamination risk at the moment it is detected. This projects lays the ground for further applications of RMM in the pharmaceutical manufacturing process.
