Abstract:
The structural specificity and hydrological sensitivity of loess have a strong impact on its long-term stability and safety. This topic is being actively researched and focuses on the macromechanical behavior of the shear strength of loess disturbed and its micromechanisms from the perspective of the dry–wet cycle (especially involving soluble salt erosion). In this paper, the correlation between micro-structural shear functional units and macroscopic degradation behavior was established by combining the changes in physicochemical properties of mass loss, surface cracking, strength deterioration, and structural disturbance of the loess with scanning electron microscopy (SEM) microscopic images in different dry–wet cycles and different salt contents. Results revealed that with the increase in dry–wet cycles and salt content, the mass loss of soil deteriorated and the surface crack rate increased. The cohesion of soil showed an overall decreasing trend, which decreased more obviously in the early stage of the dry–wet cycle, followed by a slow decrease, and tended to be constant after nine dry–wet cycles. However, the internal friction angle increased and then decreased during the whole cycle, and its value generally changed little. According to the deterioration and decay of shear strength, it can be concluded that the structural disturbance of loess increased with the increase in dry–wet cycles and salt content. At the same time, further linear quantization fitting of the structural disturbance parameters showed that the structural parameters had a positive correlation with salt content and a power function with dry–wet cycles, where dry–wet cycles seemed to play a dominant role in the loess structural deterioration rather than salt content. The microscopic study demonstrates that the dry–wet cycles and salt content do not directly affect the cohesion and internal friction angle of soil but change the basic shear structural unit of aggregate and then cause an essential impact on c and φ, which in turn have an essential impact on soil strength attenuation. This paper not only helps to elucidate the essence of water–soil–salt structural interactions but also provides theoretical references for sustainable development research in environmental engineering, geological engineering, and other related fields.

Abstract:
Protein p53 is one of the most studied proteins. This attention is primarily due to its key role in the cellular mechanisms associated with carcinogenesis. Protein p53 is a transcription factor involved in a wide variety of processes: cell cycle regulation and apoptosis, signaling inside the cell, DNA repair, coordination of metabolic processes, regulation of cell interactions, etc. This multifunctionality is apparently determined by the fact that p53 is a vivid example of how the same protein can be represented by numerous proteoforms bearing completely different functional loads. By alternative splicing, using different promoters and translation initiation sites, the TP53 gene gives rise to at least 12 isoforms, which can additionally undergo numerous (>200) post-translational modifications. Proteoforms generated due to numerous point mutations in the TP53 gene are adding more complexity to this picture. The proteoforms produced are involved in various processes, such as the regulation of p53 transcriptional activity in response to various factors. This review is devoted to the description of the currently known p53 proteoforms, as well as their possible functionality.

Background: In neuroscience research about functional magnetic resonance imaging (fMRI), accurate inter-subject image registration is the basis for effective statistical analysis. Traditional fMRI registration methods are usually based on high-resolution structural MRI with clear anatomical structure features. However, this registration method based on structural information cannot achieve accurate functional consistency between subjects since the functional regions do not necessarily correspond to anatomical structures. In recent years, fMRI registration methods based on functional information have emerged, which usually ignore the importance of structural MRI information. Methods: In this study, we proposed a non-rigid cycle consistent bidirectional network with Transformer for unsupervised deformable functional MRI registration. The work achieves fMRI registration through structural MRI registration, and functional information is introduced to improve registration performance. Specifically, we employ a bidirectional registration network that implements forward and reverse registration between image pairs and apply Transformer in the registration network to establish remote spatial mapping between image voxels. Functional and structural information are integrated by introducing the local functional connectivity pattern, the local functional connectivity features of the whole brain are extracted as functional information. The proposed registration method was experimented on real fMRI datasets, and qualitative and quantitative evaluations of the quality of the registration method were implemented on the test dataset using relevant evaluation metrics. We implemented group ICA analysis in brain functional networks after registration. Functional consistency was evaluated on the resulting t-maps. Results: Compared with non-learning-based methods (Affine, Syn) and learning-based methods (Transmorph-tiny, Cyclemorph, VoxelMorph x2), our method improves the peak t-value of t-maps on DMN, VN, CEN, and SMN to 18.7, 16.5, 16.6, and 17.3 and the mean number of suprathreshold voxels (p < 0.05, t > 5.01) on the four networks to 2596.25, and there is an average improvement in peak t-value of 23.79%, 12.74%, 12.27%, 7.32%, and 5.43%. Conclusions: The experimental results show that the registration method of this study improves the structural and functional consistency between fMRI with superior registration performance.

The 70 kDa heat shock protein (Hsp70) chaperone system is ubiquitous, highly conserved, and involved in a myriad of diverse cellular processes. Its function relies on nucleotide-dependent interactions with client proteins, yet the structural features of folding-competent substrates in their Hsp70-bound state remain poorly understood. Here we use NMR spectroscopy to study the human telomere repeat binding factor 1 (hTRF1) in complex with Escherichia coli Hsp70 (DnaK). In the complex, hTRF1 is globally unfolded with up to 40% helical secondary structure in regions distal to the binding site. Very similar conformational ensembles are observed for hTRF1 bound to ATP-, ADP- and nucleotide-free DnaK. The patterns in substrate helicity mirror those found in the unfolded state in the absence of denaturants except near the site of chaperone binding, demonstrating that DnaK-bound hTRF1 retains its intrinsic structural preferences. To our knowledge, our study presents the first atomic resolution structural characterization of a client protein bound to each of the three nucleotide states of DnaK and establishes that the large structural changes in DnaK and the associated energy that accompanies ATP binding and hydrolysis do not affect the overall conformation of the bound substrate protein.

Given the unique problem of the extremely high potential of the oxidant P + 680 that is required to oxidize water to oxygen, the photoinactivation of photosystem II in vivo is inevitable, despite many photoprotective strategies. There is, however, a robustness of photosystem II, which depends partly on the highly dynamic compositional and structural heterogeneity of the cycle between functional and non–functional photosystem II complexes in response to light level. This coordinated regulation involves photon usage (energy utilization in photochemistry) and excess energy dissipation as heat, photoprotection by many molecular strategies, photoinactivation followed by photon damage and ultimately the D1 protein dynamics involved in the photosystem II repair cycle. Compelling, though indirect evidence suggests that the radical pair P + 680 Pheo – in functional PSII should be protected from oxygen. By analogy to the tentative oxygen channel of cytochrome c oxidase, oxygen may be liberated from the two water molecules bound to the catalytic site of the Mn cluster, via a specific pathway to the membrane surface. The function of the proposed oxygen pathway is to prevent O 2 from having direct access to P + 680 Pheo – and prevent the generation of singlet oxygen via the triplet–P 680 state in functional photosytem IIs. Only when the, as yet unidentified, potential trigger with a fateful first oxidative step destroys oxygen evolution, will the ensuing cascade of structural perturbations of photosystem II destroy the proposed oxygen, water and proton pathways. Then oxygen has direct access to P + 680 Pheo – , singlet oxygen will be produced and may successively oxidize specific amino acids of the phosphorylated D1 protein of photosystem II dimers that are confined to appressed granal domains, thereby targeting D1 protein for eventual degradation and replacement in non–appressed thylakoid domains.

In addition to the protein coding information, viral RNA genomes code functional information in structurally conserved units termed functional RNA domains. These RNA domains play essential roles in the viral cycle (e.g., replication and translation). Understanding the molecular mechanisms behind their function is essential to understanding the viral infective cycle. Further, interfering with the function of the genomic RNA domains offers a potential means of developing antiviral strategies. Aptamers are good candidates for targeting structural RNA domains. Besides its potential as therapeutics, aptamers also provide an excellent tool for investigating the functionality of RNA domains in viral genomes. This review briefly summarizes the work carried out in our laboratory aimed at the structural and functional characterization of the hepatitis C virus (HCV) genomic RNA domains. It also describes the efforts we carried out for the development of antiviral aptamers targeting specific genomic domains of the HCV and the human immunodeficiency virus type-1 (HIV-1).

Abstract:
Membrane pyrophosphatases (M-PPases) couple pyrophosphate hydrolysis to the translocation of sodium ions/protons, using the resulting ion gradients to drive abiotic stress resistance and in the infectivity of protozoan parasites. I have solved two M-PPase structures in different catalytic states, combining these with previous structures to update the model of the catalytic cycle of M-PPases. These new structures confirm previous findings that substrate binding breaks interactions between K12.50 and D6.43 due to motion of helix 12, leading to a rearrangement of helix 6 and priming the enzyme for hydrolysis. Previously this information was only visible between the structures of two-distinct M-PPases, a H+-PPase and Na+-PPase. The current structures allow for comparisons to be made between structures of the same type of M-PPase. Electrometric data was acquired using the Nanion SURFE2R technique, which showed a proton-pumping signal was generated by the non-hydrolysable inhibitor, imidodiphosphate. This provided sufficient information to update the model of the complete catalytic cycle, favouring the hypothesised Binding change mechanism, in which substrate binding induces a series of conformational changes during which ion pumping occurs first, followed by substrate hydrolysis. Additionally, crystal optimisation techniques improved the resolution of the Pyrobaculum aerophilum M-PPase structure to 3.8, providing an overview of the K+-independent M-PPase. The hydrolytic centre and ion gate regions showed similar coordination to previous structures, with differences seen in the conformation of several outer ring helices, potentially linked to K+-independence. I also carried out mutational studies investigating K12.46 and T12.49, both involved in K+-independence and found that both mutations were required to generate a K+-dependent variant of PaPPase. Overall, this information has improved our understanding of the structure and function of the membrane pyrophosphatases, providing a basis for drug-design programmes targeting protozoan parasites, to which the membrane pyrophosphatases are a vital part of growth and infectivity.

Abstract:
In the recent years, a rapidly increasing amount of experimental data has been generated by high-throughput technologies. Despite of these large quantities of protein-related data and the development of computational prediction methods, the function of many proteins is still unknown. In the human proteome, at least 20% of the annotated proteins are not characterized. Thus, the question, how to predict protein function from its amino acid sequence, remains to be answered for many proteins. Classical bioinformatics approaches for function prediction are based on inferring function from well-characterized homologs, which are identified based on sequence similarity. However, these methods fail to identify distant homologs with low sequence similarity. As protein structure is more conserved than sequence in protein families, structure-based methods (e.g. fold recognition) may recognize possible structural similarities even at low sequence similarity and therefore provide information for function inference. These fold recognition methods have already been proven to be successful for individual proteins, but their automation for high-throughput application is difficult due to intrinsic challenges of these techniques, mainly caused by a high false positive rate. Automated identification of remote homologs based on fold recognition methods would allow a signi cant improvement in functional annotation of proteins. My approach was to combine structure-based computational prediction methods with experimental data from genome-wide RNAi screens to support the establishment of functional hypotheses by improving the analysis of protein structure prediction results. In the first part of my thesis, I characterized proteins from the Ska complex by computational methods. I showed the benefit of including experimental information to identify remote homologs: Integration of functional data helped to reduce the number of false positives in fold recognition results and made it possible to establish interesting functional hypotheses based on high con dence structural predictions. Based on the structural hypothesis of a GLEBS motif in c13orf3 (Ska3), I could derive a potential molecular mechanism that could explain the observed phenotype. In the second part of my thesis, my goal was to develop computational tools and automated analysis techniques to be able to perform structure-based functional annotation in a high-throughput way. I designed and implemented key tools that were successfully integrated into a computational platform, called StrAnno, which I set up together with my colleagues. These novel computational modules include a domain prediction algorithm and a graphical overview that facilitates and accelerates the analysis of results. StrAnno can be seen as a first step towards automatic functional annotation of proteins by structure-based methods. First, the analysis of long hit lists to identify promising candidates for further analysis is substantially facilitated by integration and combination of various sequence-based computational tools and data from functional databases. Second, the developed post-processing tools accelerate the evaluation of structural and functional hypotheses. False positives from the threading result lists are removed by various filters, and analysis of the possible true positives is greatly enhanced by the graphical overview. With these two essential benefits, fold recognition techniques are applicable to large-scale approaches. By applying this developed methodology to hits from a genome-wide cell cycle RNAi screen and evaluating structural hypotheses by molecular modeling techniques, I aimed to associate biological functions to human proteins and link the RNAi phenotype to a molecular function. For two selected human proteins, c20orf43 and HJURP, I could establish interesting structural and functional hypotheses. These predictions were based on templates with low sequence identity (10-20%). The uncharacterized human protein c20orf43 might be a E3 SUMO-ligase that could be involved either in DNA repair or rRNA regulatory processes. Based on the structural hypotheses of two domains of HJURP, I predicted a potential link to ubiquitylation processes and direct DNA binding. In addition, I substantiated the cell cycle arrest phenotype of these two genes upon RNAi knockdown. Fold recognition methods are a promising alternative for functional annotation of proteins that escape sequence-based annotation due to their low sequence identity to well-characterized protein families. The structural and functional hypotheses I established in my thesis open the door to investigate the molecular mechanisms of previously uncharacterized proteins, which may provide new insights into cellular mechanisms.

Dans les vasières intertidales dominées par les diatomées, la production primaire est particulièrement forte à marée basse. Ce microphytobenthos peut être limité par les nutriments azotés en lien avec les communautés de procaryotes impliquées dans le cycle de l’azote. Ainsi, ce travail de thèse cherche, via une approche écologique, à décrire le rôle des communautés de procaryotes benthiques notamment liées au cycle de l’azote et ce, suivant différentes échelles temporelles liées aux cycles du microphytobenthos. Des échantillons de sédiment ont été prélevés dans la baie de Marennes-Oléron (Côte Atlantique, France) entre 0 et 10 cm de profondeur suivant 5 couches (0-0,5 cm, 0,5-1 cm, 1-2 cm, 2-5 cm, 5-10 cm). Différents facteurs biotiques et abiotiques ont été mesurés et mis en relation avec la production bactérienne, les activités enzymatiques et les gènes fonctionnels liés au cycle de l’azote (impliqués dans la nitrification, la dénitrification et l’anammox). De plus, la diversité bactérienne et archéenne a été évaluée par pyroséquençage 454 afin de caractériser les communautés et leurs dynamiques en lien avec les facteurs forçants biotiques et abiotiques. Dans le but d’évaluer l’influence des paramètres abiotiques et de la production du microphytobenthos, des mesures in situ ont été couplées avec des mesures en conditions semi-contrôlées
In diatoms-dominated intertidal mudflats, at low tide the primary production is particularly high and microphytobenthos that can be limited by nitrogen-related nutrients is linked with N-related prokaryotic communities. Thus, this PhD thesis aim at describing by ecological approach, the role of benthic prokaryotic communities especially N-related ones, at various temporal scales linked to microphytobenthos life cycles. Sediment samples from Marennes-Oleron mudflat (Atlantic coast, France) were collected according to 5 layers : 0-0.5 cm, 0.5-1 cm, 1-2 cm, 2-5 cm and 5 to 10 cm below sediment surface (bsf). Various biotic (i.e. chlorophyll a) and abiotic parameters (i.e. nutrients, exopolymeric substances, water content, salinity, pH, temperature…) were recorded and linked with benthic bacterial production, enzymatic activities and N-related functional genes (i.e. implied in nitrification, denitrification, and anammox). Furthermore, the bacterial and archaeal diversity was assessed by 454 pyrosequencing in order to characterize the communities and shift in link with biotic and abiotic drivers. Aiming at evaluating the influence of abiotic parameters and microphytobenthic activities on the prokaryotic communities, in situ measurements were coupled to a semi-controlled approach

This paper presents a standalone coupled aeroelastic framework for highly flexible flapping wings in hover for Micro Aerial Vehicle (MAVs) application. A realistic hover-capable flapping-wing MAV utilizes highly flexible wings operating at high frequencies/amplitudes, which causes extreme wing deflections and highly unsteady, vortical flows. The strong fluid-structure coupling and the complicated flow physics make the performance of such a system extremely difficult to predict. In the present model, in order to accurately capture the large nonlinear wing deflections, a geometrically exact shell theory has been adopted. A potential-flow-based unsteady aerodynamic model is then coupled with the fully nonlinear structural model to generate high fidelity coupled aeroelastic framework. Both the structural and aerodynamic models are validated independently before coupling. The coupling process required spatial and temporal interpolation functions for the two models to communicate with each other. An innovative flapping wing experimental setup was designed and built to measure the time-history of lift forces acting on a flexible wing during a flap cycle. The instantaneous lift force predictions from the coupled aeroelastic model is compared with the results from the in-house flapping wing experiments at moderately high frequencies.

This paper investigates the feasibility of using machine learning to predict whirl flutter bifurcation diagrams. The machine learning techniques selected for the study are XGBoost and the long short-term memory neural network. These techniques are selected for their suitability for sequential and nonlinear data. The techniques are investigated for a propeller-nacelle test case with polynomial structural nonlinearities resulting in supercritical or subcritical whirl limit-cycle oscillations. The techniques are trained to learn the bifurcation diagram for the amplitude variation of pitch angle limit-cycle oscillations of the propeller-nacelle system as a function of the forward speed for various levels of cubic structural nonlinearity. Bifurcation diagram learning and testing data are generated using the bifurcation forecasting method. XGBoost is computationally faster to train but less accurate for low amounts of learning data, especially for the most weakly and strongly nonlinear cases. The long short-term memory neural network is more computationally expensive to train but shows a less scattered error pattern for sparse learning data. However, it is sensitive to the amplitude resolution of the bifurcation diagrams. The approach to sample the cubic nonlinearity range does not significantly impact the results once the techniques have a sufficient amount of data to learn from. The data requirements observed in the study suggest that, for these techniques, direct learning of bifurcation diagrams may not scale beyond a handful of input parameters.

The approach uses free decay output data for a quantity of interest at various forward speeds to estimate the system's recovery rate to equilibrium while capturing its variation with amplitude. The recovery rate is then extrapolated to predict the bifurcation diagram, which gives the limit-cycle oscillation amplitude for the quantity of interest as a function of the forward speed. The approach is demonstrated using output data from transient simulations of a notional tiltrotor model with polynomial structural nonlinearities. The approach accurately predicts the tiltrotor whirl flutter speed and limitcycle oscillation amplitudes while only requiring two free decays. This approach can facilitate whirl flutter bifurcation analyses of tiltrotor systems exhibiting nonlinear dynamics.

The approach applies the matrix pencil method to a short time window that slides along the free decay of a quantity of interest, quantifying the variation in the system's recovery rate to equilibrium with amplitude. The recovery rates at each amplitude and various forward speeds are extrapolated to predict the critical forward speed of zero recovery rate at those amplitudes. This process yields a set of limit-cycle oscillation solutions that can be visualized as a bifurcation diagram. The approach is demonstrated using output data from transient simulations of a propeller-nacelle test case with hardening structural nonlinearities. The impact of each parameter in the sliding-window matrix pencil method is first characterized via sensitivity analyses. Next, the bifurcation diagram is predicted using the recovery rates for the optimal parameter values. The results are compared with direct time marching and with the extrapolation of recovery rates estimated from envelope functions. The proposed method accurately captures the bifurcation diagram using two pre-flutter transient simulations with no need for envelope functions. This approach shows promise for output-based bifurcation analysis of nonlinear aeroelastic systems exhibiting limit-cycle oscillations associated with whirl flutter.

Shape Memory Polymer Composites (SMPCs) are smart materials showing the structural properties of long-fiber polymer-matrix together with the functional behavior of shape memory polymers. In this study, SM carbon fiber reinforced (CFR) composites have been produced by using a SM interlayer between two CFR prepregs. Their SM properties have been evaluated in comparison with traditional structural CFR composites without the SM interlayer by using an especially designed test. Active and frozen forces are measured during a thermo-mechanical cycle in the three-point bending configuration. Experimental results show that SMPCs are able to fix a temporary deformed shape by freezing high stresses.

Abstract Structural batteries are multifunctional systems that can store electrochemical energy and provide load-bearing capability. An important aspect is the mechanical testing of such multi-functional systems to establish mechanical properties while maintaining electrical functionality. This article proposes a tensile testing protocol while simultaneously cycling the structural pouch batteries electrochemically. Since most batteries require encapsulation and cannot be used in exposed environments, damage to the protective encapsulation (pouch) needs to be avoided to ensure that the battery function is not affected. The protocol ensures that the critical encapsulation layer remains intact. The fabrication and testing of structural batteries in practical operating conditions are demonstrated in this work. The method is demonstrated with a structural battery (in pouch cell format) fabricated with commercial carbon fibre nylon (CFN) unidirectional composite (which acts as a mechanical substrate onto which the anode and cathode is coated). The fabricated cells are subjected to mechanical tension load and electrochemical testing simultaneously. The load’s effect on the electrochemical cycling is studied and the cells are observed to cycle electrochemically (i.e. remain functional) even after the conclusion of the mechanical tensile test. The potential of the presented method for structural battery evaluation and development will be discussed.

Abstract This paper describes the integration of Failure Modes and Effects Analysis (FMEA) into a computer-aided tool for Serviceability Design. FMEA is an important yet often neglected design practice that is critical in ensuring the product’s ownership quality. A computer aid that integrates FMEA into other life-cycle design evaluation tool should encourage engineers to use this important methodology more readily. The proposed method, service modes and effects analysis (SMEA) combines the functional block diagram (FBD) and structural graph (LINKER) as design inputs. The user defines functional discrepancies or component failures as service mode inputs, and the tool computes the failure criticality for each mode and life-cycle service costs. The key technology lies in representing the relationship between functions and structure and maintaining the consistency between function and component-based service modes. SMEA builds on our prototype life-cycle design analysis tool, LASeR, which evaluates not only the life-cycle ownership quality but also assemblability and recyclability.

Low cycle fatigue (LCF) induces damage accumulation in structural components used in various applications. LCF typically describes conditions for which plastic strains are larger than elastic strains. In order to certify and qualify a structural component, manufactured from a given material, that requires high reliability for operation and safety, fundamental material properties should be experimentally investigated and validated. The traditional strain–life approach serves as the underlying experimental method for most LCF investigations. Building upon that background, the purpose of this paper is to investigate the statistical variability and appropriately model that variability for life in LCF. Specifically, the variability associated with the median behavior in a strain–life graph for data is examined. The ensuing analyses are based on data for a cold-rolled, low carbon, extra deep drawing steel; ASTM A969 which is appropriate for applications where extremely severe drawing or forming is envisioned. It is frequently used in the automotive industry for components such as inner door components and side body components. For substantiation of the proposed modeling techniques, data for 9Cr-1Mo steel is also investigated. Such steel is frequently used in the construction of power plants and other structures that experience operating temperatures in excess of 500°C. The commonly used universal slopes approach for fatigue life modeling for which the strain–life computation employs the standard Coffin–Manson relationship is compared to a statistical methodology using a distribution function frequently used in structural reliability. The proposed distribution function for characterizing the fatigue life is a generalized Weibull distribution function that empirically incorporates load history and damage accumulation.

In summary intensive studies carried out this year in both the US and Israel had established the methodology necessary for the achievement of the specific aims of the original proposal. Two complementary approaches to effectively neutralize the luteal ET- system were developed. In light of recent publications indicating that ET-2 might also have a physiological role in ovulation, the objectives of the original proposal have even more significant. Not only were the technologies to neutralize the luteal endothelin system developed in these studies, but additional important implications about the role of ET-1 were revealed. For example, direct early inhibitory effects of PGF2α were unmasked. It is possible that these early direct inhibitory effects could be related to functional aspects of luteal regression, while the effects observed after 12 hours of the PGF2α injection and that reversed by the ET receptor antagonist, could coincide with structural aspects of regression. Nevertheless, overall, the results clearly indicate that serum progesterone concentrations can effectively be elevated by the receptor antagonist which of great practical importance.

This paper develops a flexible, semi-structural framework to empirically quantify the non-linear transmission of income shocks to household portfolio choice decisions both at the extensive and intensive margins. I model stock market participation and portfolio allocation rules as age-dependent functions of persistent and transitory earnings components, wealth and unobserved taste shifters. I establish non-parametric identification and propose a tractable, simulation-based estimation algorithm, building on recent developments in the sample selection literature. Using recent waves of PSID data, I find heterogeneous income and wealth effects on both extensive and intensive margins, over the wealth and life-cycle dimensions. These results suggest that preferences are heterogeneous across the wealth distribution and over the life cycle. Moreover, in impulse response exercises, I find sizeable extensive margin responses to persistent income shocks. Finally, I find heterogeneity in participation costs across households in the wealth distribution.

Earthquakes, which can cause tremendous local stress and strain on infrastructure, can cause reinforced concrete (RC) bridges to collapse due to the concrete cracking and fracture of the steel reinforcement rebars. The fracture of longitudinal reinforcing steel due to low-cycle fatigue is one of the main causes of failure in RC structures under earthquake loading. The purpose of this research is to include the effects of low-cycle fatigue fracture of longitudinal reinforcing steel bars on the seismic performance of reinforced concrete bridge piers. To obtain a greater understanding of low-cycle fatigue failure of steel reinforcement of RC bridge piers subjected to seismic loadings, its properties were studied by considering the slenderness ratio to observe its effects on the behaviors of the steel material. The slenderness ratio are functions of unsupported length, diameter of the circular cross section of the longitudinal reinforcing bars, and the spacing of transverse reinforcing bars. The seismic performance of RC single-column pier-supported bridges with flexural failure under near-fault ground motion were assessed with the use of damage indices. The damage indices can numerically assess the damaged state of RC bridge piers and show the gradual accumulation of damage. Four numerical models are developed with fiber-based nonlinear beam-column elements to simulate the damage accumulation on RC bridge piers under seismic loadings, considering variables such as low-cycle fatigue, tensile strain damage, global buckling of longitudinal steel bars, the cracking and spalling of cover concrete, and the bond-slip between concrete and longitudinal steel bars. Bond-slip is related to the interaction between the longitudinal steel rebars and the concrete for load bearing and coordination deformation. The four numerical models were developed with different considerations of low-cycle fatigue and bond-slip: Model 1 (without bond-slip and without fatigue), Model 2 (without bond-slip and with fatigue), Model 3 (with bond-slip and without fatigue), and Model 4 (with bond-slip and with fatigue). The models underwent nonlinear time-history analyses. The results were compared with experimental testing results. All four numerical models are optimal to assess the seismic performance of RC single-column pier-supported bridges. The proposed damage indices can reasonably reflect the damage states in accordance with the experimental results. The proposed models can reasonably predict the damage states and seismic behavior of RC bridge columns and could be available to the structural engineering community for non-linear analysis and performance assessment of RC bridge structures.

The process of mastering, systematizing and automatizing systems language skills occupies a key place in the theory and practice of teaching foreign languages and cultures. Following the main trends of modern applied linguistics in the field of multilingual research, we hypothesize the advisability of using the lexical approach in mastering the entire complex of systems skills (grammar, vocabulary, phonology, functions, discourse) in students receiving multilingual education at higher educational institutions. In order to theoretically substantiate the hypothesis, the authors carry out structural, semantic, and phonological analysis of the main lexical units (collocations). After this, linguodidactic analysis of students’ hypothetical problems and, as a result, problems related to the teaching of relevant linguistic and axiological features is carried out. At the final stage of the paper, a list of possible outcomes from the indicated linguistic and methodological problematic situations is given. This article is the first in the cycle of linguodidactic studies of the features of learning and teaching systems language skills in a multilingual educational space.

In this project, we studied the photosynthetic apparatus during dehydration and rehydration of the homoiochlorophyllous resurrection plant Craterostigmapumilum (retains most of the photosynthetic components during desiccation). Resurrection plants have the remarkable capability to withstand desiccation, being able to revive after prolonged severe water deficit in a few days upon rehydration. Homoiochlorophyllous resurrection plants are very efficient in protecting the photosynthetic machinery against damage by reactive oxygen production under drought. The main purpose of this BARD project was to unravel these largely unknown protection strategies for C. pumilum. In detail, the specific objectives were: (1) To determine the distribution and local organization of photosynthetic protein complexes and formation of inverted hexagonal phases within the thylakoid membranes at different dehydration/rehydration states. (2) To determine the 3D structure and characterize the geometry, topology, and mechanics of the thylakoid network at the different states. (3) Generation of molecular models for thylakoids at the different states and study the implications for diffusion within the thylakoid lumen. (4) Characterization of inter-system electron transport, quantum efficiencies, photosystem antenna sizes and distribution, NPQ, and photoinhibition at different hydration states. (5) Measuring the partition of photosynthetic reducing equivalents between the Calvin cycle, photorespiration, and the water-water cycle. At the beginning of the project, we decided to use C. pumilum instead of C. wilmsii because the former species was available from our collaborator Dr. Farrant. In addition to the original two dehydration states (40 relative water content=RWC and 5% RWC), we characterized a third state (15-20%) because some interesting changes occurs at this RWC. Furthermore, it was not possible to detect D1 protein levels by Western blot analysis because antibodies against other higher plants failed to detect D1 in C. pumilum. We developed growth conditions that allow reproducible generation of different dehydration and rehydration states for C. pumilum. Furthermore, advanced spectroscopy and microscopy for C. pumilum were established to obtain a detailed picture of structural and functional changes of the photosynthetic apparatus in different hydrated states. Main findings of our study are: 1. Anthocyan accumulation during desiccation alleviates the light pressure within the leaves (Fig. 1). 2. During desiccation, stomatal closure leads to drastic reductions in CO2 fixation and photorespiration. We could not identify alternative electron sinks as a solution to reduce ROS production. 3. On the supramolecular level, semicrystalline protein arrays were identified in thylakoid membranes in the desiccated state (see Fig. 3). On the electron transport level, a specific series of shut downs occur (summarized in Fig. 2). The main events include: Early shutdown of the ATPase activity, cessation of electron transport between cyt. bf complex and PSI (can reduce ROS formation at PSI); at higher dehydration levels uncoupling of LHCII from PSII and cessation of electron flow from PSII accompanied by crystal formation. The later could severe as a swift PSII reservoir during rehydration. The specific order of events in the course of dehydration and rehydration discovered in this project is indicative for regulated structural transitions specifically realized in resurrection plants. This detailed knowledge can serve as an interesting starting point for rationale genetic engineering of drought-tolerant crops.

Abstract:
We study the determinants of active debt management through issuance and refinancing decisions for firms around the world. We leverage instrument-level data to create a comprehensive picture of the maturity, currency, and security type composition of firms' debt for a large cross-section of countries. At the instrument level, we estimate a predictive model of prepayment as a function of interest costs savings and maturity lengthening motives. We document that there is substantial heterogeneity in prepayment across bonds and loans and across firms, depending on their reliance on bank lending. While debt prepayment is generally successful at extending average maturities and lowering interest rate costs at the firm level, these benefits appear smaller for issuers in emerging market economies. Tight global credit conditions reduce both the ability to prepay debt early and the effectiveness of debt refinancing in reducing interest costs and rollover risk. Put together, our results show that the impact of global credit conditions on firms' debt structure can be traced back to how instrument-level prepayment incentives change over the global credit cycle.
