String theory, with its richness of dynamical scenarios, represents a microscopic tool of utmost relevance to jointly describe all the fundamental forces of nature and simultaneously it involves a wide and fascinating spectrum of geometrical implications. Because of its nature, in many cases research in string theory cannot be carried through on a purely locally-based analysis, but it should often be supplemented with intrinsically global investigations and consistency checks. The mathematical apparatus needed for such a study, however, turns out to be usually much more sophisticated than the one which is su cient for eld theory-like computations, and, unfortunately, it is not always really familiar to the physics community. The arguments discussed in this thesis constitute manifest examples of this situation...

This thesis explores new analytical advances using compact disk biosensing technologies, and comprises six scientific publications distributed along four chapters. Special attention is herein payed to Thermochromic Etching Disks (TED) technology (Chapter 1), rational design of disk-based biorecognition assays (Chapter 2), and label-free detection systems for point-of-need analysis (Chapters 3 and 4).
First, insights into a novel light-mediated signal developing system for biorecognition assays (based on TED disks and drives) are provided together with an overview of the state-of-the art and future trends in photo- and thermochromic biosensing. This signal developing approach exploits photo- and thermochromism for biosensing in an original manner and represents a potential strategy to simplify signaling processes in bioanalytical systems. Then, how to transform TED technology into lab-on-a-disk systems is addressed. TED has proven to be a very versatile tool to perform sensitive analysis of biorecognition assays, using platforms and scanners easily obtained from regular disks and drives, respectively. Biologically relevant assays of different nature (microarray, cell culture, immunofiltration, turbidimetry, etc.) have been arrayed in a single disk and sensitively analyzed by imaging.
Regarding rational design, a theoretical-experimental method (INSEL) based on kinetics and mass-transport modelling for optimizing biorecognition assays and exploring their behavior is presented. INSEL has been implemented as an in silico tool that enables to characterize biointeractions with minimal experimentation, to perform optimizations directed towards custom objectives defined by the user, and to easily compute the effect of critical variables without further experiments.
In another study included in this thesis, polycarbonate grooved structures obtained from standard recordable disks (CD-R and DVD-R) were coated with silver and tailored to become SERS-active. This strategy represents a cost-effective and industrially scalable alternative to the SERS substrates typically used for bioanalysis. These disk-based materials have presented tunable plasmonic responses, significant Raman enhancement, and have allowed complex biological targets (such as proteins and exosomes) to be analyzed by SERS without using labeled reagents as tracers. In addition to introduce inexpensive and large-scale SERS substrates for biosensing, this study also suggests the development of prospective Raman scanners based on disk drives.
Another approach herein presented addresses the implementation of diffraction-based sensing (DBS) in TED technology in order to conceive disk-based label-free biosensors based on standard disks and drives. At first, a comprehensive experimental assessment of the analytical possibilities offered by DBS is presented. Then, the fabrication of arrays of diffractive protein networks on TED disks is investigated, with which sensitive analysis of antibodies in label-free conditions has been demonstrated, using adapted drives as scanners. This investigation provides important insights into cost-effective and industrially scalable functional materials and detection setups that exploit consumer electronics for label-free biosensing.Esta tesis explora nuevos avances en química analítica usando tecnologías de biosensado basadas en sistemas de disco compacto y comprende seis publicaciones científicas distribuidas a lo largo de cuatro capítulos. Los estudios se han centrado en la tecnología Thermochromic Etching Disks (TED) (Capítulo 1), el diseño racional de ensayos de bioreconocimiento en discos compactos (Capítulo 2), y la detección sin marcaje para realizar análisis in situ (Capítulos 3 y 4). Primero, enmarcado en una discusión del estado del arte y futuras tendencias en biosensado foto y termocrómico, se presenta un nuevo sistema (basado en discos y lectores TED) mediado por luz para el desarrollo de señales en ensayos de bioreconocimiento. Ésta constituye una estrategia novedosa para aprovechar el foto y termocromismo en biosensado, y presenta un gran potencial para simplificar los procesos de desarrollo de señal en sistemas bioanalíticos. A continuación, se aborda cómo transformar la tecnología TED en sistemas analíticos integrados basados en discos compactos. TED ha demostrado ser una herramienta muy versátil para analizar, de forma sensible, ensayos de bioreconocimiento usando plataformas y escáneres fácilmente obtenidos a partir de discos y lectores convencionales, respectivamente. Un único disco ha mostrado poder albergar varios ensayos biológicos importantes y de distinta naturaleza (micromatriz, cultivos celulares, inmunofiltración, turbidimetría, etc.), para ser analizados de forma sensible a través de imágenes En cuanto al diseño racional, se presenta un método teórico-experimental (INSEL), basado en modelos cinéticos y de transporte de masa, para optimizar ensayos de bioreconocimiento y explorar su comportamiento. INSEL se ha implementado como una herramienta in silico que permite caracterizar biointeracciones mediante mínima experimentación, realizar optimizaciones dirigidas a objetivos particulares definidos por el usuario, y computar el efecto de variables críticas de forma sencilla y sin experimentos adicionales. En otro estudio incluido en esta tesis, nanoestructuras en forma de surco obtenidas a partir de discos regrabables convencionales (CD-R y DVD-R) fueron recubiertas con plata y adaptadas para ser activas en SERS. Esta estrategia supone una alternativa, económicamente efectiva e industrialmente escalable, a los sustratos SERS típicamente usados en bioanálisis. Estos materiales han mostrado respuestas plasmónicas sintonizables, una amplificación Raman significativa, y han permitido analizar muestras biológicas complejas (como proteínas y exosomas) mediante SERS sin usar marcadores. Además de introducir sustratos SERS grandes y baratos, este trabajo también sugiere el desarrollo de escáneres Raman basados en lectores de disco. Otra aproximación presentada en esta tesis aborda la implementación de DBS (diffraction-based sensing) en tecnologías TED, con el fin de desarrollar biosensores para detección sin marcaje basados en discos y lectores convencionales. Primero, se presenta una amplia evaluación experimental de las posibilidades analíticas ofrecidas por DBS. A continuación, se investiga la fabricación de multitud de redes difractivas de proteínas sobre discos TED, con las que se ha demostrado la determinación sensible y sin marcaje de anticuerpos, usando lectores adaptados como escáneres analíticos. Esta investigación introduce avances importantes que apuntan al desarrollo de materiales funcionales y sistemas de detección, baratos e industrialmente escalables, que aprovechen las tecnologías de consumo para realizar biosensado sin marcaje.
Aquesta tesi explora nous avanços en la química analítica usant tecnologies de biosensat basades en sistemes de disc compacte, i comprèn sis publicacions científiques distribuïdes en quatre capítols. Els estudis s'han centrat en la tecnologia Thermochromic Etching Disks (TED) (Capítol 1), el disseny racional d'assajos de bioreconeixement en discos compactes (Capítol 2), i la detecció sense marcatge per realitzar anàlisi in situ (Capítols 3 i 4). Primer, dins del marc d'una discussió de l'estat de l'art i tendències futures en biosensat foto i termocròmic, es presenta un nou sistema (basat en discos i lectors TED) per al desenvolupament de senyals mitjançant llum, en assajos de bioreconeixement. Aquesta constitueix una nova estratègia per aprofitar el foto i termocromisme en biosensat, mentre que també presenta una gran potencial per simplificar els processos de desenvolupament de senyal en sistemes bioanalítics. Tot seguit, s'aborda com transformar la tecnologia TED en sistemes analítics integrats basats en discos compactes. TED ha demostrat ser una eina molt versàtil per analitzar, de forma sensible, assajos de bioreconeixement usant plataformes i escàners fàcilment obtinguts a partir de discos i lectors convencionals, respectivament. A més d'introduir substrats SERS grans i barats, aquest treball també suggereix el desenvolupament d'escàners Raman basats en lectors de disc. Una altra aproximació presentada en aquesta tesi aborda la implementació de DBS (diffraction-based sensing) en tecnologies TED, per tal de desenvolupar biosensors basats en discos i lectors convencionals que permeten detecció sense marcatge. Magistrinio darbo problematikos formulavimas vadovaujasi prielaida, jog dalis moderniosios filosofijos projektų formavosi kaip reakcija į Vakarų Europos mąstymo tradiciją, kurios formuojantį pagrindą sudarė filosofija. Dėl šios priežasties neatsiejamu šių projektų egzistavimo požymiu laikytina nuosekli filosofijos istorijos kritika, tuo pačiu pateikianti alternatyvius filosofijos supratimo orientyrus. Tokiu radikaliu filosofijos sąjūdžiu, XX a. laikomas egzistencializmas. Magistriniam darbui pasirinktos dvi filosofų figūros: Rosenzweigas ir Heideggeris. Abu filosofai kritiškai reflektavo filosofijos istoriją bei abu pateikė alternatyvius mąstymo projektus. Pagrindiniu Vakarų tradicijos kritikos bei naujo mąstymo suformulavimo svarbiausiu aspektu laikome fundamentalų „pirminių mąstymo prielaidų” perklausimą. Atlikęs tokį perklausimą bei taip atsiribojęs nuo „mąstymo metafizikos”, Rosenzweigas susikuria naujas prieigas prie tikėjimo problematikos. Panaudodamas Johano Wolfgango Geothes profenomeno sampratos bei savo mokytojo – Hermanno Coheno diferencialinės matematikos teorijos idėjas, jis pateikia trijų (metafizinio Dievo, metaloginio pasaulio ir metaetinio žmogaus) profenomenų aprašymą. Metaetinio žmogaus sąmonės struktūros gretinimas su „naujojo mąstymo” (tokį pavadinimą Rosenzweigas pats prisiskiria savo filosofijai) pirminėmis prielaidomis, atveda prie dialoginės (Zwiesprache) apreiškimo (Offenbarung) sąmonės struktūros aprašymo. Tik apreiškimo sąmonei atsiveria pirmapradė... [toliau žr. visą tekstą]The formulation of theme in this paper is followed the assumption that a part of projects of modern philosophy were generated as a reaction to the tradition of Western Europe thinking, which was influenced by philosophy. That is the reason why coherent criticism of history of philosophy, in the same time giving the perception guides of philosophy, is considered as a concurrent attribute of that project existence. Existentialism of the 20th century is considered as such radical movement of philosophy. Two philosophical figures were chosen from this general context: Rosenzweig and Heidegger. Both thinkers made critical reflections on history of philosophy and both give the alternative projects of thinking. The fundamental reasking of „prime thought assumptions” is considered as an important aspect of main criticism of Western tradition and the formulation of new thinking. After this performed reasking and disassociation from “metaphysics of thought”, Rosenzweig creates new approaches to the faith theme. Using the prophenomenon perception of Johan Wolfgang Geothe and his teachers, Herman Cohen’s ideas about diferencial mathematics theory, he presents description of three prophenomenon (metaphysical God, metalogical world and metaethical man). Esta peregrinação teve o seu início no final de 1552 e terminou em Setembro de 1558. O seu autor fez uma descrição pormenorizada de tudo o que viu e ouviu, desde a saída de Lisboa até chegar à Casa Santa de Jerusalém e do seu regresso até à Universidade de Salamanca, onde dá por terminada a peregrinação. Esta edição contém 503 páginas, em formato A4. É composta por uma introdução e notas, seguida de oito livros.: Livro primeiro do monge peregrino aa Casa Sancta. Hypothèse: l'enfant aux prises avec sa névrose infantile agit selon sa perception de son Autre, personne ou lieu d'adresse de sa parole. Chez les premiers psychanalystes d'enfants, nous vérifions si oui ou non, un écueil de l'acte est que l'analyste en vienne à se conformer à ce que l'enfant induit de sa perception de son Autre. Le fantasme devient réalité. Les défenses sont renforcées. La méprise: l'analyste saurait le bien de l'enfant, sa nature et agirait en conséquences: comme père, mère, enfant, analyste. . . Idéal. S. Ferenczi et Freud se disputent leur savoir sur le Père (de la psychanalyse), Bon analyste. M. Klein redouble l'appel à la bonne Mère de certains transferts. A. Freud selon le fantasme de l'enfant d'âge œdipien construira des lois de son développement psychosocial. D. W. Winnicott en jouant, déjoue les agirs infantiles, et dessine la présence symboliques de l'analyste dans la cité (exigence de la parole). F. Dolto se fait l'Autre de la Vérité, donne à l'enfant le savoir sur son fantasme. Pour tous la castration symbolique est le but de la cure: l'analyste la donne en comblant les besoins "psy ". R et R. Lefort ne vérifient pas ce collage. Le sujet s'ouvre à son désir, non d'un savoir sur lui, mais de son accès à sa parole. Le rapport du sujet aux signifiants, au défaut phallique de la chose pulsionnelle est mis en fonction. L'hypothèse est vérifiée. L'analyste d'enfant n'est pas les figures imaginaires ou réelles de l'Autre. L'histoire des concepts vérifie que le savoir inconscient est côté enfant et non, côté analyste. La fin de la cure: l'enfant se fait sujet du discours de la science, dont il est l'objet du malaise de la civilisation. La définition ne se fait plus par opposition au social, mais selon la visée des demandes qui la fonde sur l'appel du sujet. Freud builds, according to the fantasy of the child in his/her oedipian age, laws to his/her psychosocial development. D. W. Winnicott, playing, frustrates the child's actings and outlines the symbolical presence of the analyst in the city (the demand of speech). F. Dolto becomes the Other of the Truth, gives the child the knowledge on his/her fantasy. Symbolical castration is, for everyone, the goal of the cure : the analyse delivers it while fulfilling the so called "psy" needs. R et R. Lefort do not check this collage. The subject opens to his/her desire, not of a knowledge of himself/herself, but of his/her access to his/her speech. The relationship of the subject with the signifiers, with the phallic deficiency of the drive, is put into function. The hypothesis is confirmed. The analyst for children is not the imaginary or real images of the Other. The history of concepts confirms that unconscious knowledge is on the child's side- and not on the analyst's. The end of the cure: the child becomes a subject to the science discourse of which he/she is the object of the discontent of civilisation. The definition is no longer given in opposition with the social but according to the aim of the questions which founds it on the invocation of the subject. In this work a study of social networks based on analysis of family names is presented. A basic approach to the mathematical formalism of graphs is developed and then main theoretical models for complex networks are presented aiming to support the analysis of surnames networks models. These, in turn, are worked so as to be drawn leading quantities, such as aggregation coefficient, minimum average path length and connectivity distribution. Based on these quantities, it can be stated that surnames networks are an example of complex network, showing important features such as preferential attachment and small-world character
Neste trabalho ? apresentado um estudo das redes sociais baseado na an?lise dos nomes de fam?lias. Faz-se uma abordagem b?sica do formalismo matem?tico dos grafos e em seguida apresenta-se os principais modelos te?ricos para as Redes Complexas com o objetivo de fundamentar a an?lise das redes dos sobrenomes. Estas, por sua vez, s?o trabalhadas de modo a serem extra?das as principais grandezas, tais como coe ciente de agrega??o, menor caminho m?dio e distribui??o de conectividades. Com base nestas grandezas, pode-se a rmar que as redes de sobrenomes s?o um exemplo de rede complexa, exibindo caracter?sticas importantes como liga??o preferencial e o car?ter de mundo pequeno.

This dissertation considers the mystery of the relationship between human free choice and God by focusing on the Controversy de Auxiliis (1582-1607) and the thought of Domingo Báñez, O.P. (1528-1604) in particular. The dissertation comprises four chapters and a conclusion preceded by a preface and brief historical introduction. The preface introduces the issue to be explored and the motivations for exploring it before providing a general synopsis of the dissertation that is more detailed than the present abstract. The historical summary that follows introduces a theological debate that has become widely unfamiliar to contemporary theology, even while conceptually, that debate remains perennial. The four-chapter body that follows may be divided into two general parts: Broadly, chapters One and Two exposit Báñez’s thought, while chapters Three and Four critique it. Chapter One explores Báñez’s positive account of physical premotion, human freedom and sin. Chapter Two examines Báñez’s critique of Luis de Molina S.J.’s alternative proposal, in conjunction with some contemporary sources from both sides of the debate (Molina was Báñez’s principal adversary in the Controversy de Auxiliis). Báñez’s line of critique in Chapter Two is found to be cogent. Chapter Three investigates Molina’s critique of Báñez and finds it too to be cogent, even though Molina’s positive account was found to be problematic in Chapter Two. Chapter Four begins by exploring Bernard Lonergan S.J.’s work on divine causality and human free choice. Lonergan attempts to provide a fresh historical reading of Aquinas that is unencumbered by the presuppositions of the Controversy de Auxiliis. The first part of Chapter Four explains Lonergan’s critique of Báñez and finds it convincing, while the second part of the chapter finds Lonergan’s interpretation of Aquinas problematic from a theoretical standpoint. Chapter Four then offers a constructive critique of Lonergan’s interpretation before advancing an alternative way to think about God’s causation of human free choices. In closing, this dissertation argues that God creates human free choices, but that in creating a human free choice, God, or God’s creative will, is not an antecedent condition that determines choice. Rather, God creates the entire reality of a human free choice—both what it is and that it is—and in so doing, part of the reality God creates just is that choice’s being up to its human agent.

This essay examines the role of repetition in the two novels Älskaren (The Lover) by Marguerite Duras and Den amerikanska flickan (The American Girl) by Monika Fagerholm, through a thematic and comparative reading. The study mainly uses Bruce F. Kawin’s analysis of repetition in literature and language as well as it refers to Cathy Jellenik and her analysis of rewriting in the works of Duras. The study also takes into Julia Kristeva’s idea of Duras as a writer lacking of catharsis. Furthermore, the essay analyses both novels by applying Sigmund Freud’s term “repetition compulsion”. The essay shows how Fagerholm and Duras both use permissive writing, which means that the authors boldly write clichéd replications in their texts. Both writers practice what Kristeva calls “an aesthetics of awkwardness”. Aswell, the authors both question whether a story at all is able to be told: they work with what Jellenik calls “an aesthetics of doubt”, but in different ways. Moreover, by using the work of Kawin, the essay shows how the authors have different ways of handling time. The study shows indications of repetition compulsion in the characters and also exemplifies instances of what Kristeva calls “reduplication”. Through this a fragmentary and Nietzschean perspective of the self is displayed in the novels. The study review how Duras can be said to be a writer lacking of catharsis, while Fagerholm on the other hand liberates one of her protagonists from repetition compulsion by allowing the character to undergo a catharsis. Finally, the essay conclusion points out that Duras and Fagerholm are postmodern writers who know how to challenge the concept of truth. Even if they are different writers in many aspects, they both use repetition as a method to express something unspeakable. They can be said to write also by an aesthetics of search for the truth; a search for an origin tale which they both argue as biased and is therefore impossible to capture.

Galactans are polysaccharides sulfated present in the cell wall of red algae. Carrageenans are galactans well known in the food industry as gelling polysaccharides and for induce inflammatory process in rodents as animal model. The extraction of polysaccharides from A. multifida has been carried out by proteolysis and precipitation in different volumes of acetone, which produced three fractions (F1, F2, and FT). Chemical and physical analyses revealed that these fractions are sulfated galactan predominantly. Results of the antioxidant activity assays showed that all of these fractions have antioxidant activity and that was associated with sulfate content of the analysis of reducing power and total antioxidant capacity. However, these fractions were not effective against lipid peroxidation. The fraction FT presented higher activity on the APTT test at 200 μg (> 240 s). The assessment of the hemolytic activity showed that the FT fraction has the best activity, increasing lyses by the complement system to 42.3% (50 μg) (p< 0,001). The fraction FT showed the best yield, anticoagulant and hemolytic activity between the three fractions and therefore it was choose for the in vivo studies. The Inflammation assessment using the FT fraction (50 mg / kg MB) showed that the cellular migration and the IL-6 production increased 670.1% (p< 0,001) and 531.8% (p< 0,001), respectively. These results confirmed its use as an inflammation inducer in animal model. Cytotoxicity assay results showed that all fractions have toxic effects on 3T3 and HeLa cells after exposition of 48 hours, except when 100 μg for both F1 and FT were used. These results arise the discussion whether these polysaccharides it should be used as additive in foods, cosmetics and medicines.

An?lises f?sicas e qu?micas revelaram que essas fra??es s?o predominantemente galactanas sulfatados. Resultados dos ensaios de atividade antioxidante mostraram que todas essas fra??es apresentam atividade antioxidante e que esteve associada ao teor de sulfato na an?lise da redu??o da pot?ncia e capacidade antioxidante total. No entanto, estas fra??es n?o foram eficazes contra a peroxida??o lip?dica. A fra??o FT apresentou maior atividade no teste de APTT a 200 mg (> 240 s). A avalia??o da atividade hemol?tica mostrou que a fra??o FT tem a melhor atividade, aumentando a lise pelo sistema complemento para 42,3% (50 mg) (p< 0,001). A fra??o FT apresentou o melhor rendimento, atividade anticoagulante e hemol?tica entre as tr?s fra??es e por isso foi escolhido para os estudos in vivo. En quoi la psychanalyse de Freud et de Lacan auraient quelque chose à dire concernant l’inutile dans l’art ? C’est la question centrale dans cette thèse. Une condition inutile qui caractérise les arts autant de nos jours qu’à certains moments dans l’histoire. L’argumentation montre en quoi cette condition est partagée avec la qualité paradoxale du sujet de la psychanalyse. Cette qualité paradoxale l’est, selon notre hypothèse, par rapport au sujet de la science et de la philosophie, raison pour laquelle le fil rouge de cet analyse sera le texte « La science et la vérité. En effet, si l’on suive les efforts de Lacan pour distinguer le sujet de la psychanalyse de celui de la science et de la philosophie, nous trouvons des éléments très propres aux poétiques. Ceux-ci sont constatés en parcourant l’histoire de l’art, l’esthétique et les théories utilitaristes de Bentham et Mill. Ainsi, l’hypothèse principale ici proposée c’est l’inutile dans l’art comme un corrélat culturel du sujet de la psychanalyse. Sans chercher ni à établir une définition de l’inutile, ni à décider si les arts le sont ou ne le sont pas, cette thèse propose une théorie psychanalytique qui par structure s’occupe de l’inutile, comme la règle fondamentale l’atteste. En le proposant comme un reste conditionné par la propre structure de l’être parlant, l’inutile apparaît comme un réel qui revient toujours à sa place. Si nous proposons un inutile dans l’art comme corrélat culturel du sujet de la psychanalyse, l’art vient être la manifestation privilégié au sein du lien sociale de ce qu’il y a de radicalement asociale et inutile chez le sujetWhat can be said about the useless character of the arts? How does the condition of ‘uselessness’ which has permeated art across its history, resonate with the paradoxical quality of the Lacanian barred subject? This is the central question of this thesis. In ‘Science and Truth’, a key text for this work, Jacques Lacan makes a crucial distinction between the poetic dimension of the divided subject of psychoanalysis and the subject that philosophy and science presupposes. The poetic dimension that Lacan locates in this seminal text and the specific elements that distinguish its subject from the subject of philosophy and science can be traced in the history of art and aesthetics as well as in the utilitarianism of Bentham and Mills. It is precisely these resonances that lead us to think that the useless character of art is the cultural correlate of the subject of psychoanalysis. Such is the key hypothesis that orients this thesis.While purposely suspending the temptation to extract a closed definition of the term ‘useless’ and equally avoiding speculation around the question of how useless art is or isn't, this work proposes that Freud’s theory is forged and structured around the idea of the useless as suggested by the fundamental rule of free association. In disregard of the effort that the subject enlists to make life useful, they will unavoidably trip over a useless remainder. Freud’s observations imply that the structure of the speaking being produces an excess, a left over that Lacan called, jouissance - a useless real that always returns to the same place. If we accept the useless character of art, art manifests in the social bond as precisely this remainder: the privileged manifestation of a radical uselessness in the human subject

This study sets out to explore attitudes and intentions towards nicotine product goods and how it is affected by the goal framing effect. The presence of this effect has been shown in the advertising of both green products and everyday products. The aim of this research is to explore whether or not this also holds true when it comes to unhealthy products, more precisely tobacco free all white nicotine products. A quasi experimental study was conducted with the use of a fictitious brand of All-white tobacco. All 63 participants in the three different experimental groups of the study met the mandatory age requirement. One of the three groups received a positively framed advert, another received a negatively framed advert, whereas the last group acted as the control group and therefore received a neutral advertisement stimuli. All participants submitted their answers on a questionnaire created for this study, which was validated using Cronbach’s alpha and factor analysis. The results suggest that the use of goal framing is beneficial when advertising nicotine product goods. Both positive and negative goal-framing showed a greater effect on purchase intention and product attitude than the control group. The negative goal-framing advert proved to be statistically different than the control group when measuring product attitude. Furthermore, the positively framed advert showed a statistically significant difference in effect on both product attitude and purchase intention compared to the control group. The findings suggest that positively framed goal-framing has an effect on both attitudes and purchase intention, compared to the neutral stimuli. The negatively framed goal-framing had an effect on attitudes, compared to the neutral stimuli. The practical implication of this study could possibly be that when constructing advertisements for tobacco free snus products, it could be preferable to make use of the positive goal-framing effect in order to affect the consumers purchase intention and attitude towards the product. This study has confirmed to some extent that the framing effect is a factor prevalent in the advertising of tobacco free products. For further research it would be highly interesting to delve deeper in comparing positive and negative goal-framing in order to find further evidence of which one has greater effect on consumers.

This study focuses on preparing surface enhanced fluorescence (SEF) substrates for use in the enhancement of the emission signal of rhodamine B and fluorescein dyes.
Fluorescence spectroscopy has been widely utilized owing to its high sensitivity. SEF is a process where the interactions of fluorophores with the localized surface plasmons of metal nanoparticles results in fluorescence enhancement, increased photostability and rates of system radiative decay which leads to a decreased lifetime. One of the most important factors of SEF studies is to provide a uniform distance between fluorophore and metal nanoparticle in a controlled mannerotherwise, Fö
rster resonance energy transfer takes place from fluorophore to metal nanoparticle and emission intensity of fluorophore is quenched. The spherical gold and silver nanoparticles were prepared using the well known and straightforward chemical reduction method, in which sodium citrate acted both as a reducing agent and a stabilizer around the formed nanoparticles. Silver and gold were chosen because of their high plasmon field enhancement. Since plasmon field strongly depends on the shape and size of the nanoparticles, the prepared nanoparticles were characterized using absorption spectroscopy and field emission scanning electron microscopy (FE-SEM). Prior to deposition of silver or gold nanoparticles on glass slides, the slides were derivatized by immersing them into an aqueous solution of 3-Aminopropylethoxysilane (APTES). Following derivatization, silver or gold nanoparticles were deposited by immersing the slides into the colloid mixture. Metal nanoparticle coated slides were characterized using absorption spectroscopy and field emission scanning electron microscopy (FE-SEM). Surface enhanced Raman scattering (SERS) measurements were carried out to observe the plasmon efficiency of the deposited nanoparticles. The SERS measurements were repeated for the duration of two weeks in order to check the stability of the plasmon efficiency. In this study, different types of materials (silica, zinc oxide, gold, stearic acid.) were employed as spacers to observe their effects on fluorescence enhancement. Physical vapor deposition (PVD) and Langmuir-Blodgett (LB) film deposition techniques were used for the formation of the spacer within the substrate. Fluorescence enhancement of rhodamine B and fluorescein was observed on the prepared SEF substrates. Obtained enhancement factors indicate that SEF substrates have the potential for sensitivity improvements of fluorescence sensing in many fields.

A MATTER OF NATIONAL CONCERN examines the Kennedy Administration’s contribution to the restoration of public education in Prince Edward County, Virginia, and determines if those actions support the dominant narrative of Kennedy’s overall civil rights record – a historical assessment generally generated from a few acute crises. For five consecutive years (1959-1964), in defiance of federal court orders, the county board of supervisors refused to levy taxes to operate public schools, marking Prince Edward County as the only locale in the nation without free public education. The county leadership organized a segregated private school system for the 1,400 white children, but afforded no formal education for the 1,700 African American students. The Kennedy Administration inherited the Prince Edward County school situation – a crisis that threatened to cripple a generation, and, if replicated, destroy public education. In the Prince Edward County school dilemma, the Kennedy Administration took proactive measures, proved sympathetic to the plight of African Americans, challenged Virginia’s congressional delegation, and appointed federal judges that supported President Kennedy’s civil rights agenda. The Prince Edward County story generally, and the federal government’s actions specifically, have been virtually overlooked by historians. A MATTER OF NATIONAL CONCERN challenges scholars to re-evaluate the Kennedy Administration’s civil rights record by including all of the civil rights events of the Kennedy years, thus developing a thorough, comprehensive assessment. A MATTER OF NATIONAL CONCERN is the product of the study of unpublished archival documents, oral histories, interviews, newspaper reports, and secondary sources. The horse has a natural predisposition for the sport, however, its use in competitions can result in stress related problems that impair its sporting performance and especially its health. In this way it is fundamental not only to understand how the different risk and resilience factors to different stressors influence the response to stress, but also to develop strategies that can prevent or minimize the deleterious effects of stress. In this sense, acupuncture is an ancient technique of Traditional Chinese Medicine that has been used in the treatment and prevention of stress-related diseases. The present study proposed the use of two models of stress: one physical (physical exercise) and another psychological (startle model) to verify the reactivity to the stress of athletes horses. In addition, it was also evaluated if horses of different sporting modalities present different psychological stress responses and if acupuncture can alter the responses to physical stress. In the experiment 1, 16 Thoroughbred race horses were submitted to a exercise in the field of high intensity and short duration (12 m / s, 4 min). The RR intervals for analysis of the Heart Rate Variability were acquired through the Polar Equine ? heart rate monitor and blood samples were collected before and immediately after 2h, 4h, 6h, and 24h after exercise. The exercise promoted autonomic alterations in the sympatho-vagal balance since it significantly increased the low frequency component (LF), the heart rate and the LF / HF ratio, and decreased the high frequency component (HF) (p <0.01). There was an increase in hematocrit, plasma proteins, glucose and plasma lactate immediately after exercise (p <0.001). There was an increase (p <0.01) in serum cortisol values after 30 minutes, returning to baseline values after 60 min. However, no significant difference was observed in plasma cytokines IL-1? and IL-6 between moments after exercise and baseline. In experiment 2, horses of the experimental group 1 after exercise were randomly divided into two groups: CTL (C2): Control (without manipulation) and ACUP (C2)): animals submitted to weekly sessions of acupuncture at points VG1, C7, VG20 and B52 for 10 weeks. After the treatment period the animals repeated the same exercise and the same parameters were analyzed. Acupuncture reduced the LF / HF ratio, promoting a faster recovery of the animals, showing no influence on the other parameters analyzed. In the experiment 3, 24 equines were used, from three equestrian modes: Polo (PSI) (n = 9), Dressage (Brazilian Horse Riding) (n = 6) and Endurance (n=6) were subjected to the experimental model of startling through the abrupt opening of an umbrella. The results showed that endurance horses presented a significantly less intense startle-induced autonomic response than Polo and Dressage horses (lower LF / HF ratio at the time of the jump), paradoxically Enduro horses have cortisol levels in response in response to the startle than Polo horses. However, there was no difference between the modalities in the behavioral response after the startle, and Polo horses had significantly higher baseline levels of cortisol than the other modalities and did not change their cortisol levels in response to stress. Thus, we can conclude that 1) the exercise in the field of high intensity and short duration promoted adaptive changes characteristic of stress, being able to be used in studies of reactivity to stress in race horses; 2) acupuncture has a modulating effect on the stress-induced autonomic response in athletic horses, and 3) the equestrian modalities of Dressage, Polo and Endurance present different stress reactivity
O cavalo tem uma predisposi??o natural para o esporte, no entanto, o seu uso em competi??es pode resultar em problemas relacionados ao estresse que prejudicam seu desempenho esportivo e principalmente a sua sa?de. Desta forma ? fundamental n?o apenas entender como os diferentes fatores de risco e de resili?ncia a diferentes estressores influenciam a resposta ao estresse, como tamb?m desenvolver estrat?gias que possam prevenir ou minimizar os efeitos delet?rios do estresse. Neste sentido a acupuntura ? uma t?cnica milenar da Medicina Tradicional Chinesa que tem sido utilizada no tratamento e preven??es de doen?as relacionadas ao estresse. O presente estudo prop?s o uso de dois modelos de estresse: um f?sico (exerc?cio f?sico) e outro psicol?gico (modelo de sobressalto) para verificar a reatividade ao estresse de cavalos atletas. Al?m disso, tamb?m foi avaliado se cavalos de diferentes modalidades esportivas apresentam respostas ao estresse de psicol?gico distintas e se acupuntura pode alterar as respostas ao estresse f?sico. No experimento 1, 16 equinos de corrida da ra?a Puro Sangue Ingl?s foram submetidos ao exerc?cio a campo de alta intensidade e curta dura??o (12 m/s, 4min). Os intervalos RR para an?lise da Variabilidade da Frequencia Card?aca foram adquiridos atrav?s do frequenc?metro card?aco Polar Equine? e as amostras de sangue foram coletadas antes e, imediatamente, 2h, 4h, 6h, e 24h ap?s o exerc?cio. O exerc?cio promoveu altera??es auton?micas no balan?o simpato-vagal uma vez que aumentou significativamente o componente de baixa frequ?ncia (LF), a frequ?ncia card?aca e a raz?o LF/HF e diminuiu o componente de alta frequ?ncia (HF) (p < 0.01). Houve aumento do hemat?crito, das prote?nas plasm?ticas, glicose e lactato plasm?tico imediatamente ap?s o exerc?cio (p < 0.001). Houve aumento (p<0.01) nos valores s?ricos de cortisol ap?s 30 minutos, retornando aos valores basais ap?s 60min. No entanto, n?o foi observado diferen?a significativa nas citocinas plasm?ticas IL-1? e IL-6 entre os momentos ap?s exerc?cio e o momento basal. No experimento 2: os equinos do experimento 1 ap?s o exerc?cio foram aleatoriamente divididos em dois grupos: CTL (C2): Controle (sem manipula??o) e ACUP (C2)ACUP (C2): animais submetidos a sess?es semanais de acupuntura nos pontos VG1, C7, VG20 e B52 durante 10 semanas. Ap?s o per?odo de tratamento os animais repetiram o mesmo exerc?cio e foram analisados os mesmos par?metros. A acupuntura reduziu a raz?o LF/HF, promovendo uma recupera??o mais r?pida dos animais n?o apresentando influ?ncia nos demais par?metros analisados. No experimento 3, foram utilizados 24 equinos, pertencentes a tr?s modalidades equetres: P?lo (ra?a PSI) (n=9), Adestramento (ra?a Brasileiro de Hipismo) (n=6) e Enduro (Puro Sangue ?rabe) (n=9) submetidos ao modelo experimental de sobressalto atrav?s da abertura abrupta de um guarda-chuva. Os resultados mostraram que cavalos de enduro apresentaram resposta auton?mica induzida pelo sobressalto significativamente menos intensa que cavalos de Polo e Adestramento (menor raz?o LF/HF no momento do sobressalto), paradoxalmente cavalos de Enduro possuem n?veis de cortisol em resposta ao sobressalto mais altos que cavalos de Polo. N?o houve diferen?a entre as modalidades na resposta comportamental ap?s o sobressalto, no entanto cavalos de P?lo apresentam n?veis basais de cortisol significativamente mais altos que as demais modalidades e n?o variaram seus n?veis de cortisol em resposta ao estresse. Previous issue date: 2009-07-30Introduction: The ability to walk is impaired in obese by anthropometric factors (BMI and height), musculoskeletal pain and level of inactivity. Little is known about the influence of body adiposity and the acute response of the cardiovascular system during whole the 6-minute walk test (6mWT). Objective: To evaluate the effect of anthropometric measures (BMI and WHR waist-to-hip ratio), the effort heart and inactivity in ability to walk the morbidly obese. Materials and Methods: a total 36 morbidly obese (36.23 + 11.82 years old, BMI 49.16 kg/m2) were recruited from outpatient department of treatment of obesity and bariatric surgery in University Hospital Onofre Lopes and anthropometric measurements of obesity (BMI and WHR), pulmonary function, pattern habitual physical activity (Baecke Questionnaire) and walking capacity (6mWT). The patient was checking to measure: heart rate (HR), breathing frequency (BF), peripheral oxygen saturation, level of perceived exertion, systemic arterial pressure and duplo-produto (DP), moreover the average speed development and total distance walking. The data were analysed between gender and pattern of body adiposity, measuring the behavior minute by minute of walking. The Pearson and Spearmam correlation coefficients were calculated, and stepwise multiple Regression examined the predictors of walking capacity. All analyses were performed en software Statistic 6.0. Results: 20 obese patients had abdominal adiposity (WHR = 1.01), waist circumference was 135.8 cm in women (25) and 139.8 cm in men (10). Walked to the end of 6mWT 412.43 m, with no differences between gender and adiposity. The total distance walked by obesity alone was explained by BMI (45%), HR in the sixth minute (43%), the Baecke (24%) and fatigue (-23%). 88.6% of obese (31) performed the test above 60% of maximal HR, while the peak HR achieved at 5-minute of 6mWT. Systemic arterial pressure and DP rised after walking, but with no differences between gender and adiposity. Conclusion: The walk of obese didn?t suffers influence of gender or the pattern of body adiposity. The final distance walked is attributed to excess body weight, stress heart, the feeling of effort required by physical activity and level of sedentary to obese. Pouco se sabe sobre a influ?ncia do perfil da adiposidade corporal, do g?nero e da resposta aguda do sistema cardiovascular sobre a capacidade de caminhada do obeso. Objetivo: Avaliar o efeito de medidas antropom?tricas (IMC e WHR, waist-to-hip ratio), esfor?o card?aco e sedentarismo sobre a capacidade de caminhada de portadores de obesidade m?rbida. Materiais e M?todos: Entre setembro de 2007 e setembro de 2008, 36 obesos m?rbidos (idade 36,23 + 11,82; IMC 49,16 kg/m2) foram recrutados no ambulat?rio de tratamento da obesidade e cirurgia bari?trica do Hospital Universit?rio Onofre Lopes e avaliados quanto marcadores antropom?tricos de obesidade, fun??o pulmonar, n?vel de atividade f?sica habitual (Question?rio de Baecke) e capacidade de caminhar (TC6M). As correla??es de Pearson e Spearmam foram analisadas. A Regress?o M?ltipla buscou preditores da capacidade de caminhada. Foi utilizado o software Statistic 6.0 para an?lise estat?stica. Resultados: 20 obesos tinham adiposidade abdominal (WHR = 1,01), circunfer?ncia da cintura de 135,8 cm nas mulheres (25) e de 139,8 cm nos homens (10). Durante TC6M, foi caminhada uma dist?ncia de 412,43 m, sem diferen?as entre g?nero ou adiposidade. Essa dist?ncia total percorrida foi explicada isoladamente pelo IMC (45%), FC no sexto minuto (43%), Baecke (24%) e fadiga (-23%). 88,6% dos obesos (31) realizaram o teste acima de 60% da FCM?xima, sendo o pico de FC atingido aos 5 minutos de caminhada. PA e DP aumentaram significativamente com a caminhada, mas sem diferen?as entre g?nero ou adiposidade. La situation actuelle des institutions psychiatriques en Israël est partagée entre un discours d'orientation médicale mêlée à une orientation de réhabilitation. La question qui guide la recherche est de savoir si la psychanalyse, dans une orientation freudo-lacanienne, pourrait guider la pratique thérapeutique dans un service psychiatrique, pourrait s’écarter du réel de la pulsion, de l’objet a dans le contexte de la vie institutionnelle, pourrait donner des possibilités de rééducation dans la pratique médicale et dans toute l’activité existant dans le service d’un hôpital psychiatrique dans lequel on prendrait en compte le sujet au sens psychanalytique. Dans le cadre de cette thèse, nous essayerons de chercher un éclairage de certains aspects de la vérité de la pratique d'un service psychiatrique, au travers de l’orientation psychanalytique lacanienne, par l'intermédiaire d'une lecture de la pratique et d'entretiens qualitatifs avec les membres clés d’un service de psychiatrie en Israël, et par conséquent ajouter ou ouvrir de nouvelles possibilités permettant de meilleurs soins.Cette recherche nous permettra de démontrer à travers trois présentations de cas, l’usage du discours psychanalytique dans un service psychiatrique de l’hôpital Shalvata dont le lien avec la psychanalyse est étroit. Pourtant, la confusion entre l’approche humaniste et l’orientation psychanalytique y est grande. Quand l'humanisme prend les commandes de la rencontre thérapeutique et que la psychanalyse ou la psychothérapie devient un outil pour réaliser le but humaniste, elles perdent de leur puissance et ne servent plus que comme défense contre l'angoisse. L’approche humaniste est une condition nécessaire mais non suffisante, pour que la psychanalyse soit un discours utilisé pour guider la cure dans un service. Il s'agit de s’appuyer sur le discours analytique, faire du réel une question clinique commune. Notre analyse indique que la psychanalyse appliquée à la thérapie dans un service psychiatrique n’est pas une affaire de cadre, de setting, mais bien une affaire de discours. L’orientation pratique dans l’institution est telle qu’il s’agit de traiter le symptôme du sujet avec respect et de tenter de l’accompagner dans la recherche pragmatique d’une solution plus efficace, qui lui permettrait de mieux s’entendre avec l’autre, de modérer sa jouissance, de mieux s’entendre avec son corps, tout cela à partir des indications que nous donne le sujetThe current situation of psychiatric institutions in Israel is divided between a discourse of medical orientation mixed with a rehabilitation orientation. The question which guides the research is to know if psychoanalysis, in a Freudo-Lacanian orientation, could guide the therapeutic practice in a psychiatric service, could deviate from the real of the drive, of the object a in the context of the institutional life, could provide possibilities for re-education in medical practice and in all the activity existing in the service of a psychiatric hospital in which the subject is taken into account in the psychoanalytic sense.In the framework of this thesis, we will try to shed light on certain aspects of the truth of the practice of a psychiatric service, through the Lacanian psychoanalytic orientation, through a reading of the practice and of qualitative interviews with key members of a psychiatric service in Israel, and therefore add or open up new possibilities enabling better care. This research will allow us to demonstrate, through three case presentations, the use of psychoanalytic discourse in a psychiatric service of the Shalvata hospital, which has a close link with psychoanalysis. However, there is a great confusion between the humanist approach and the psychoanalytic orientation. When humanism takes control of the therapeutic encounter and psychoanalysis or psychotherapy becomes a tool to achieve the humanist goal, they lose their power and only serve as a defense against anxiety. The humanist approach is a necessary but not sufficient condition for psychoanalysis to be a discourse used to guide the treatment in a service. It is about using the analytical discourse, making of the real a common clinical question. Our analysis indicates that psychoanalysis applied to therapy in a psychiatric service is not a matter of frame, of setting, but rather of discourse. The practical orientation in the institution means treating the subject's symptom respectfully and of trying to accompany him in the pragmatic search for a more effective solution, which would allow him to better manage with the other, to moderate his jouissance, to get along better with his body, all this based on the indications given to us by the subject

Ethnographic collections in western museums such as the Royal Albert Memorial Museum (RAMM) carry many meanings, but by definition, they represent an intercultural encounter. This history of this encounter is often lost, overlooked, or obscured, and yet it has bearing on how the objects in the collection have been interpreted and understood. This thesis uncovers the hidden history of one particular collection in the RAMM and examines the multiple meanings that have been attributed to the objects in the collection over time. The Richard Dennett Collection was made in Africa in the years when European powers began to colonise the Congo basin. Richard Edward Dennett (1857-1921) worked as a trader in the Lower Congo between 1879 and 1902. The collection was accessioned by the RAMM in 1889. The research contextualises the collection by making a close analysis of primary source material which was produced by the collector and by his contemporaries, and includes publications, correspondence, photographs and illustrations which have been studied in museums and archives in Europe and North America. Dennett was personally involved with key events in the colonial history of this part of Africa but he also studied the indigenous BaKongo community, recording his observations about their political and material culture. As a result he became involved in the institutions of anthropology and folklore in Britain which were attempting to explain, classify and interpret such cultures. Through examining Dennett’s history this research has been able to explore the Congo context, the indigenous society, and those European institutions which collected and interpreted BaKongo collections. The research has added considerably to the museum’s knowledge about this collection and its collector, and the study responds to the practical imperative implicit in a Collaborative Doctoral Project, by proposing a small temporary exhibition in the RAMM to explore these histories and meanings. In making this proposal the research considers the current curatorial debate concerning responsible approaches to colonial collections, and assesses some of the strategies that are being employed in museums today.

Elle prend appui pour sa démonstration, dans une première partie sur une analyse des travaux théoriques traitant de la notion de moi idéal de Freud à Françoise Dolto en analysant notamment en détail les travaux de Viktor Tausk, Herman Nunberg. Daniel Lagache et Jacques Lacan ainsi que les élaborations anglo-saxonnes de Heinz Kohut, Otto Kernberg et Annie Reich. Cette partie commence par une analyse de la relation transférentielle Freud Jung afin de souligner le fait que la théorie du narcissisme de Freud est produite dans un contexte clinique transférentiel ou il est confronté à la pathologie narcissique de Jung. Aussi bien, la recherche théorique s'appuiera-t-elle des lors sur les développements de Lagache. Lacan et Dolto qui ont permis de dépasser les impasses de Freud. La théorie lacanienne de l'imaginaire, jusque dans ces dernières avancées, devient de ce fait le support de cette thèse. Analyzing attentively the works of Viktor Tausk. Hermann Numberg, Daniel Lagache and Jacques Lacan and some of Anglo-Saxons authors such as Heinz Kohut, Otto Kernberg and Annie Reich. A part of this first chapter shows how the transferential relationship between Freud and Jung was important in the context of the elaboration of the theory of narcissism because of Jung’s narcissics troubles. Then, this research is found upon Lacan's theory of "imaginaire" which enables a better understanding of the narcissism. The second part of the research is based upon seven clinical studies of adults and children that have been reported through their therapies. Their analyses put the light on the fact that each psychoanalyst confronted with narcissics troubles should accept to be involved narcissically himself in the cure. The last part deals with the theory of the ideal founder agency : it postulates a knot (cf. Lacan) including instincts with a "pre-imaginaire" surface (ideal ego) and with a pre-symbolical materiel (ego ideal). The description of the agency through two poles-ideal ego and ego idealenables a coordination of the dynamics and the economics in that structure and gives the psychoanalyst a new way of thinking the pre-oedipian building of the ego. The main aim of this thesis is the analysis, characterization and emphasis of the phenomenon of Free Spirit in Nietzsche's Human, All Too Human, volumes I. and II. The thesis attempts to portray the Free Spirit as a holistic concept which, in addition, converges Nietzsche's most prevalent subjects, the criticism of religion, metaphysics and culture, and the importance of education and development. The character of Free Spirit shall be used to illustrate Nietzsche's vision of the future and the possible roads, which lead to it shall be described. In the first part of the thesis, I intend to focus on the creation of the Free Spirit with emphasis on "the great liberation" and on the circumstances from which it emerges, especially in contrast to the fettered spirit. The central motive then shifts to the importance of culture, its development and escalation and its cohesion with the Free Spirit. The final part is dedicated to the effects of the Free Spirit, its tasks and possibilities, which it has for the free ranging spirit. Dissertação de Mestrado apresentada no ISPA - Instituto Universitário para obtenção do grau de Mestre na especialidade de Psicologia ClínicaO presente estudo visa contribuir para a compreensão da vivência subjetiva (intra e interpsíquica) do sujeito transexual e avaliar o impacto das alterações corporais inerentes ao Processo de Redesignação Sexual. Foram realizadas duas entrevistas, com o espaçamento de um ano entre elas, seguindo o Método de Narrativas de Associação Livre (Hollway & Jefferson, 2000), a um participante do género masculino em processo de redesignação para o género feminino. A análise das narrativas, orientada pelos postulados teóricos da metodologia proposta, revela que a transformação corporal consiste numa solução defensiva e radical face a um ataque e depreciação da masculinidade do sujeito, levando-o a constituir uma identidade de superfície feminina que recobre a identidade e permite a continuidade psíquica. A transformação corporal ganha sentido na fantasia de um “renascer”, com o propósito de varrer da consciência um passado doloroso e remeter as suas experiências ao esquecimento. A nova “etapa”, retratada e marcada no corpo, inspirada na fuga e no desespero, constitui a derradeira tentativa de proteger um território psíquico na iminência de desaparecer. O corpo torna-se palco de transformações sucessivas que parecem intensificar a estranheza da experiência corporal e gerar uma maior e perigosa desorganização psíquica.
ABSTRACT: The present study aims to contribute to the understanding of the subjective experience (intra and interpsychic) of the transsexual subject and to evaluate the impact of the corporal changes inherent in the Sexual Reassignment Process. Two interviews were carried out, with the spacing of one year between them, following the Free Association Narratives Method (Hollway & Jefferson, 2000), to a participant of the masculine gender in the process of redesignation to the feminine gender. The analysis of the narratives, guided by the theoretical postulates of the proposed methodology, reveals that the corporal transformation consists in a defensive and radical solution to an attack and depreciation of the masculinity of the subject, leading him to constitute a feminine surface identity that encases the identity and allows for psychic continuity. Corporal transformation makes sense in the fantasy of a "rebirth", with the purpose of wiping from consciousness a painful past and remitting its experiences to oblivion. The new "stage," portrayed and marked in the body, inspired by escape and despair, is the ultimate attempt to protect a psychic territory in the imminence of disappearing. The body becomes the stage of successive transformations that seem to intensify the strangeness of bodily experience and generate a greater and dangerous psychic disorganization.
