Background: Fluralaner is a novel isoxazoline compound and the only systemically distributed ectoparasiticide approved in the United States for redosing at up to 12-week intervals for flea and tick control in cats. Other feline ectoparasiticides, including other systemic isoxazolines, are approved for redosing at monthly intervals. A survey developed in 2016 to assess the satisfaction, preference, and adherence of dog owners prescribed fluralaner as an ectoparasiticide with the treatment and veterinary flea and tick protection recommendations was adapted for completionby cat owners in the USA. Aim: The study objective was to use cat-owner survey data obtained at US veterinary practices to assess client satisfaction and utilization practices for fluralaner, and to evaluate owner adherence to current flea and tick control recommendations. Methods: US veterinary practices (n = 26) were asked to obtain completed surveys for up to 25 active clients who were currently treating their cats with a topical preparation of fluralaner for flea and tick control. Clients who had previously used flea and tick products for cats other than fluralaner were enrolled in the study. Participating cat owners completed an 11-question survey on their satisfaction with, preference for, and adherence to treatment recommendations for topically applied fluralaner as a feline flea and tick control medication. Results: The average cat in this study had a mean (± SD) body weight of 5.1 (± 0.9) kg and was 7.1 (±1.4) years old. Most cats lived in a home versus an apartment and more than half spent some time outside. Satisfaction was assessed with a 5-point Likert scale, with nearly all cat owners (97%) indicating that they were satisfied or very satisfied with fluralaner. Most of them (66%) had previously used other monthly flea and tick products for cats. Owners were not excluded if they had previously used a canine flea and tick product. The extended dosing interval up to 12 weeks was the most frequently selected benefit of fluralaner. Nearly 9 out of 10 respondents indicated they readministered fluralaner mostly on time or delayed by a few days, and most said they were more likely to give a repeat dose of fluralaner at the recommended redosing interval compared to monthly products. 87% of the responding cat owners preferred topical fluralaner over the monthly flea and tick products they had used. Conclusion: The extended dosing interval of up to 12 weeks was the leading preference factor and the key driver of user satisfaction with fluralaner leading to improved adherence to redosing recommendations. Cat owners said they were more likely to administer fluralaner at the recommended redosing interval compared to monthly products, indicating that less frequent redosing contributes to improved adherence.

Fluralaner is a new molecular entity from the isoxazoline class that exhibits acaricidal and insecticidal activity and it is recommended as an anti-ticks drugs. The aim of this study is to analyze the effect of fluralaner against dog infested by tick. A total of 10 dogs weighing about 20-40 kg which consists of various races, were divided into control groups with-out medication (n=2), and treatment group (n=8) received fluralaner tablets once by orally, at a dose of 25 mg / kg body weight. The results showed that based on observation of day of drug application at 24th hour was the time where the drug reaches the standard of efficacy with decrease number of the tick by 97.3%. In 28 and 56 days post treatment were the highest levels of efficacy with a decrease number of tick reaching 100%. The efficacy of the fluralaner was still very high until 84 days post treatment reached up to 94.1%. This study also proved that 12 weeks of drug work believed to overcome reinfestation of the tick. The rapid tick-killing effect and the long duration of efficacy showed the potency fluralaner to aid in the prevention of tick borne diseases.

RESUMO O objetivo do presente trabalho foi relatar a eficácia do fluralaner no tratamento da demodicidose juvenil generalizada canina. Dois caninos, apresentando dois e três meses de idade, com diagnóstico clínico e parasitológico de demodicidose generalizada, foram tratados com administração única de fluralaner, na dose recomendada em bula pelo fabricante. Além disso, foi realizada a terapia adjuvante à base de xampu de peróxido de benzoíla e domperidona. Observou-se uma resposta terapêutica satisfatória, com repilação completa ao 30º dia após a administração do fármaco. Para cada paciente, foi realizado exame parasitológico do raspado cutâneo aos 75 e 90 dias seguintes ao início do tratamento, e todos foram negativos. Tendo em vista a elevada eficácia do tratamento proposto, sugere-se a inclusão do fluralaner nos protocolos terapêuticos destinados à demodicidose canina, particularmente nos pacientes pediátricos e com a forma generalizada da doença.

A six-year-old spayed female silver fox (Vulpes vulpes) presented with bilateral ocular discharge, lethargy, facial alopecia and dermatitis of six months’ duration. The fox was immobilised with ketamine (4 mg/kg), dexmedetomidine (0.01 mg/kg) and butorphanol (0.05 mg/kg) intramuscularly for skin cytology, skin scrapings, dermatophyte culture, bloodwork, faecal floatation and skin biopsies. Diagnostics revealed numerous Demodex species mites. Histopathology showed folliculitis, perifolliculitis and furunculosis with mites, identified as Demodex canis using RT-PCR. The fox received meloxicam (0.2 mg/kg by mouth, three days), cefovecin (8.6 mg/kg subcutaneously, single injection), praziquantel (56.8 mg, subcutaneously, single injection), and fluralaner (250 mg, by mouth, once every 12 weeks, Bravecto). Complete resolution was noted one month after initiating treatment. Thirteen months later the fox remains mite-free and is receiving fluralaner by mouth every 12 weeks. This case report describes successful management of adult-onset generalised Demodex species dermatitis in a silver fox using oral fluralaner.

The toxicity of three new insecticides indoxacarb, chlorfenapyr and fluralaner for house flies of the susceptible laboratory strain S-NIID has been studied. A comparison is made of their insecticidal properties in the contact and intestinal routes of entry into the body. Sugar baits with this three insecticides are highly toxic to houseflies. Insecticidal baits containing 0.1–0.6 % a.i. of indoxacarb are highly effective against houseflies. Apparently, indoxacarb, chlorfenapyr, and fluralaner can be recommended for the introduction of insecticides into rotation schemes to combat winged flies, both in the form of concentrates used by spraying or insecticidal baits.

Isoxazolines are oral insecticidal drugs currently licensed for ectoparasite control in companion animals. Here we propose their use in humans for the reduction of vector-borne disease incidence. Fluralaner and afoxolaner rapidly killed Anopheles, Aedes, and Culex mosquitoes and Phlebotomus sand flies after feeding on a drug-supplemented blood meal, with IC50 values ranging from 33 to 575 nM, and were fully active against strains with preexisting resistance to common insecticides. Based on allometric scaling of preclinical pharmacokinetics data, we predict that a single human median dose of 260 mg (IQR, 177–407 mg) for afoxolaner, or 410 mg (IQR, 278–648 mg) for fluralaner, could provide an insecticidal effect lasting 50–90 days against mosquitoes and Phlebotomus sand flies. Computational modeling showed that seasonal mass drug administration of such a single dose to a fraction of a regional population would dramatically reduce clinical cases of Zika and malaria in endemic settings. Isoxazolines therefore represent a promising new component of drug-based vector control.

ZusammenfassungIm Jahr 2016 kamen für Kleintiere zwei neue Wirkstoffe auf den deutschen Tierarzneimittelmarkt: das Mineralokortikoid Desoxycorticosteron (Zycortal®) und Sarolaner (Simparica®), ein Ektoparasitikum der Isoxazolingruppe. Ein Wirkstoff erhielt eine Tierartenerweiterung. So wurde Fluralaner (Bravecto®), ebenfalls ein Ektoparasitikum der Isoxazolingruppe, auch für Katzen zugelassen. Außerdem kamen ein Präparat mit einer neuen Wirkstoffkombination, ein Wirkstoff in Monozubereitung und zwei Wirkstoffe in einer neuen Darreichungsform für Kleintiere auf den Markt.

Einleitung Dermanyssus gallinae ist ein weit verbreiteter Ektoparasit bei Hühnern in Europa. Vor allem in den Sommermonaten entwickeln sich in Hühnerställen sehr große Milbenpopulationen. Die Milbenstiche und der damit einhergehende Blutverlust gehen zulasten des Tierwohls und verursachen hohe wirtschaftliche Einbußen. Die Milben befinden sich nur nachts für den Saugakt auf den Tieren und verstecken sich tagsüber in trockener Einstreu oder in dunklen Ritzen und Spalten. Dies macht die Bekämpfung schwierig und aufwendig. Seit September 2017 ist ein Arzneimittel mit dem Wirkstoff Fluralaner auf dem deutschen Markt verfügbar. Dieses soll die Bekämpfung der Roten Vogelmilbe einfacher machen. Ziel der Untersuchungen Ziel dieser Dissertation ist die Bewertung der Milbenreduktion bei Behandlung mit Fluralaner in der Dosierung 0,5 mg/kg Körpergewicht bei zweimaliger Applikation im Abstand von sieben Tagen in natürlich infizierten Geflügelställen. Verglichen wurde jeweils ein behandelter Versuchsstall mit einem unbehandelten Kontrollstall unter Beachtung der Good Clinical Practice (GCP). Tiere, Material und Methoden In der vorliegenden Arbeit wurden die Ergebnisse von zwei Junghennenbetrieben, drei Legehennenbetrieben und einem Elterntierbetrieb betrachtet. Auf jedem Betrieb waren zwei möglichst ähnliche Ställe mit vergleichbaren Tieren vorhanden. Die Ställe durften vor der Studie länger als drei Monate nicht mit Antiparasitika oder Silikatstaub behandelt worden sein. Die Höhe des Milbenbefalls in den Ställen wurde mit Fallen der Firma AviVet B.V. (Niederlande) ermittelt. Diese wurden in regelmäßigen Abständen vor und nach der Behandlung an gleichmäßig im Stall verteilten, festen Plätzen für 24 Stunden im Stall ausgebracht. Anschließend wurden sie tiefgefroren und bei Avivet B.V. in Lunteren, Niederlande ausgezählt. Die Hühner im Behandlungsstall wurden zwei Mal im Abstand von sieben Tagen mit einer Fluralaner-Lösung (10 mg/ml) behandelt. Die Dosierung betrug 0,5 mg/kg Körpergewicht. Die Studie galt nach einer Versuchsdauer von sechs Monaten nach der ersten Applikation, nach Beendigung eines Produktionszyklus, nach einem starken Absinken der Milbenreduktion oder wenn der Kontrollstall aufgrund des Tierwohls gegen D. gallinae behandelt werden musste, als beendet. Außerdem wurden Mortalität, klinische Auffälligkeiten und bei Legehennen die Legeleistung beobachtet. Die Milbenreduktion der Fluralaner-Lösung wurde mit der Henderson-Tilton Formel berechnet. Tests auf signifikante Unterschiede zwischen der behandelten und unbehandelten Gruppe wurden mit dem U-Test nach Mann-Whitney durchgeführt. Ergebnisse Durch die Behandlung mit Fluralaner konnte bei allen Betrieben eine signifikante (P < 0,05) Milbenreduktion erreicht werden. Bei drei Betrieben wurde eine Milbenreduktion von über 99 % erreicht, in den anderen drei Betrieben lag die Milbenreduktion zwischenzeitlich bei 100 %. Die Dauer der Milbenreduktion unterschied sich jedoch von Betrieb zu Betrieb zum Teil stark. In den Aufzuchtbetrieben wurde am Ende des Produktionszyklus (42 Tage nach erster Fluralaner Applikation) noch eine Wirksamkeit von 96 % beziehungsweise 99,6 % und damit ein signifikanter Unterschied (P < 0,05) festgestellt. In zwei Legehennenbetrieben sank die Milbenreduktion 70 Tage nach der ersten Fluralaner Applikation unter 90 % (P > 0,05). In einem Legehennenstall lag die Wirksamkeit 119 Tage nach der ersten Fluralaner Applikation (Produktionsende) noch bei 98,95 % (P < 0,05). Bei den Elterntieren sank die Milbenreduktion 126 Tage nach der ersten Fluralaner Applikation unter 90 % (P > 0,05). Bei der Mortalität konnten keine signifikanten (P > 0,05) Unterschiede zwischen Versuchs- und Behandlungsstall festgestellt werden. Auch klinisch gab es keine Auffälligkeiten. Bei zwei von drei Betrieben war die Legeleistung im Behandlungsstall gegenüber dem Kontrollstall erhöht. In einem dieser Betriebe könnte diese Differenz aber durch ein unterschiedliches Tieralter verursacht worden sein. Schlussfolgerungen Auf allen untersuchten Betrieben konnte eine Milbenreduktion von über 90 % erreicht werden. Eine zweimalige Applikation im Abstand von sieben Tagen mit Fluralaner in der Dosierung 0,5 mg/kg Körpergewicht ist als wirksam gegen D. gallinae einzustufen. Aufgrund der einfachen Applikation über das Trinkwasser bietet die Behandlung mit Fluralaner viele Vorteile gegenüber im Stall auszubringenden Akariziden und Silikatstäuben.
