Full textKinetic monitoring of protein-protein interactions offers fundamental insights of their cellular functions and is a vital key for the improvement of diagnostic tests as well as the discovery of novel therapeutic drugs. Surface plasmon resonance (SPR) is an established biosensor technology routinely used for kinetic studies of biomolecular interactions. While SPR offers the benefits of real-time and label-free detection, it requires expensive and sophisticated optical apparatus and highly trained personnel, thus limiting the accessibility of standard laboratories. In this PhD project, we have developed an alternative and cost-effective biosensor platform exploiting biofunctionalized nanofluidic slits, or nanoslits, combined with a bench-top fluorescence microscope. Our approach enables the visualization of protein interactions in real-time with the possibility to determine associated kinetic parameters along with optimized response times and enhanced binding efficiency. We have demonstrated the effectiveness of our devices through kinetic studies of two representative protein-receptor pairs with different binding affinities: streptavidin-biotin and mouse IgG/anti-mouse IgG interactions. Good agreement of extracted kinetic parameters between our device, SPR measurements and literature values indicated that this approach could be readily applicable to study kinetics of protein interactions with sensitivity down to 1 pM on a large scale of dissociation constants. In addition, we have incorporated a microfluidic gradient generator to our validated nanoslit device, which has allowed one-shot parallel kinetic measurements to be realized in a single-experiment. This integrated system provides advantages of diminished material consumption and analysis time over the conventional kinetic assays. We believe that this innovative technology will drive future advancements not only in the discipline of biomedical and personalized medicine, but also in basic chemical/biological research

Full textLe but de la thèse est de développer une ou plusieurs techniques d'imagerie quantitatives et instantanées de température et de concentration. Deux axes d'études complémentaires ont été abordés au cours de ce travail. Il s'agit de la fluorescence induite par laser sur le radical OH pour l'étude des milieux en combustion et de la fluorescence induite par laser d'un traceur moléculaire (acétone) pour l'étude des écoulements non réactifs. Ces deux axes d'étude ont été menés en parallèle avec la même approche : après compilation des données bibliographiques, des modèles de fluorescence induite par laser de ces deux espèces ont été développés. Les résultats de ces modélisations ont servi à sélectionner les meilleures stratégies expérimentales.
Des expériences pilotes ont été mises en place en vue développer ces techniques d'imagerie. Elles consistent en l'utilisation des modèles proposés pour les deux traceurs étudiés afin d'obtenir des images instantanées et couplées de température et de concentration dans différents types d'écoulements (combustion hydrogène–air, mélange de jets, ...).
D'autres applications ont été réalisées sur des bancs d'essais à caractère semi-industriel afin de caractériser l'utilisation de ces techniques dans des conditions de mesures sévères.
Les résultats obtenus au cours de ces différentes expériences montrent que l'outil d'imagerie quantitative et instantanée développé est opérationnel, répondant ainsi à un besoin d'informations quantitatives et spatialement résolues et complétant les possibilités offertes par les autres diagnostics optiques.

Full textThe discovery of cell free foetal DNA (cffDNA) in 1997 allows for the combination of accuracy as well as non-invasiveness for prenatal diagnosis. This non-invasive genetic test requires only a maternal blood sample from which the cffDNA can be isolated and analysed. In this work cffDNA was isolated from a maternal blood sample using a micro-fluidic device which was fabricated using hot embossing and laser ablation techniques. The DNA sample was first pre-concentration by electrokinetic trapping (EKT) and then isotachophoresis (ITP). The concentrated sample was then separated by size using capillary electrophoresis (CE), all in a single device. All parameters and processes concerned with the micro-fluidic device were optimised sequentially. These parameters include both the chemical components as well as the physical processes which occur. The DNA used for the optimisation protocol was analysed using fluorescence spectroscopy, agarose gel electrophoresis as well as an Agilent Bioanalyser. The optimised protocol included a 9% acrylamide/pDMA matrix, 3 M N,N-dimethylurea as a denaturing agent, with tris based buffers for pre-concentration steps and 1X TBE (tris/borate/EDTA) buffer for capillary electrophoresis. The applied voltage of ITP was 300 V and CE was carried out at 180 V. The timing at which DNA was extracted from the device was kept at time = 60 s intervals. The optimised protocol was then used for real sample analysis and these samples were obtained from mothers pregnant with male foetuses. The DNA extracted from the micro-fluidic device was then analysed using real time PCR (RT-PCR) in order to distinguish which was maternal and which was foetal. This was carried out by amplification of male and general (present in male and female) genes respectively. RT-PCR results confirmed that only the male specific gene was amplified in initial samples exiting the device and it was thus successful in isolating cffDNA from a maternal plasma sample.

Full textThe concurrent spreading of a boundary layer type diffusion flame is studied. The impossibility of obtaining a low velocity laminar flow without any perturbation induced by buoyancy has lead to the development of an experimental apparatus for use in micro-gravity facilities. Based on previous experimental observations, an original numerical approach has been developed showing, first the dominating role of the radiative heat transfer on the structure of the flame and second the major role of the soot on the extinction phenomenon at the flame trailing edge. The influence of the forced flow velocity, the fuel injection velocity and oxygen concentration on the geometry of the flame has been examined by imaging of CH* and OH* radicals spontaneous emission. Laser-Induced Incandescence (LII) is used to determine the soot field concentration in the flame. The soot formation has been studied by Laser Induced Fluorescence (LIF) of Polycyclic Aromatic Hydrocarbons (PAHs). The interaction between the reaction zone and the field of soot formation/oxidation is taken into account to analyze the flame length. These results can be used as the experimental input data for a future complete validation of numerical model simulating the soot formation and oxidation in this kind of flame

Full textThe objective of this work was to construct the recombinant phage D29::gfp and to use this phage as an indicator agent of cell viability in a minimal inhibitory concentration (MIC) assay for the mains drugs used for tuberculosis treatment. The recombinant phage contains the mycobacteria-specific hsp70 promoter controlling the green fluorescent protein gene (gfp) and was constructed by Spe I restriction in the intergenic region next to the right cohesive termini of the D29 phage genome. An hsp 70 promoter and gfp previously cloned in p YL GFP was amplified by PCR using primers with Spe I sites. The Spe I-restricted D29 phage DNA was ligated with the hsp 70-gfp fragment using T4 DNA ligase and ligated product was transformed using the packing protocol. Infection of M.smegmatis with this recombinant phage indicated the expression of green f1uorescent protein (GFP). To use the recombinant phage for assaying the activity of anti-TB drugs, 100 clinical isolates was tested for susceptibility to isoniazid (H), rifampicin (R), streptomycin (S), and ethambutol (E) using both the proportion method on Lowenstein-Jensen (L-J) medium, resazurin microtiter assay plate (REMA), as well as a microplate assay using D29::gfp. The REMA plate method showed that 30 clinical isolates were susceptible to H and 58 (66%) clinical isolates were resistant, where the MICs were 1 µg/mL or higher for 41 (71%). The R MICs for 49 (56%) resistant clinical isolates were 0,5 µg/mL for 17 (35%). The S MICs for 33 (37%) resistant clinical isolates were 2 µg/mL for 13 (40%) and E MICs for 34 (39%) resistant clinical isolates were 16 µg/mL or higher for 19 (56%). Molecular characterization by PCR IS6110 showed that 88 clinical isolates were M.tuberculosis and by PRA hsp65 were seven clinical isolates were M.kansasii and four was M.abscessus, and one M.zulgai. After using the recombinant phage as an indicator agent of cell viability for assaying the activitity of anti-TB drugs we can conclude that the expression of green fluorescent protein was non-specific and not reproducible, rendering it not useful for the determination of the MIC of the principal drugs used for the treatment of tb.

The control of their implementation requires comprehension of their behavior and constitutes one of the challenges of nowadays research. In this work of thesis, we were interested in the sedimentation process of concentrated suspensions of non-colloidal and inert particles whose size distribution is mono, bi or tri-disperse. Two types of sedimentation were studied: decantation and thickening. The objective is to do an experimental and in a lesser way numerical study to characterize sedimentation the most completly possible with a detailed attention to flow and deposit structure, concentration profiles and determination of sedimentation modes. Used experimental means are the usual techniques of two-dimensional measurement (laser velocimetry and direct visualization) which authorize the data acquisition for configurations where particles evolve in the plan. These laser techniques were improved to simultaneously follow various particles populations and access to various outpouts such as velocities field, local volumic fraction, and velocities of various sedimentation fronts. A simple numerical tool was also built using Stokesian Dynamics and seizing the main part of the physical phenomena driving sedimentation leading to the modeling of the hydrodynamic interactions

Full textLitterature review showed the conventional concentration probes are now limited both in size and frequency cutoff. Fluorescence spectroscopy seemed promising. Theoretical basis and applications study explain how it can provide spectral, spatial and temporal resolution indispensable to improve local concentration measurements. Difficulties when studying the elements of the experimental setup are described with their complete adjustments. Characteristic electrical and mechanical noises and corresponding remedies, when existing, are give. The absorption-emission stochastic process involved by the fluorescence of the tracer appears to depend on a Poisson statistic : direct proportionality no more exists between local concentration and measured signal. But relationships between their statistical properties have been established, with the corresponding signal processing only able to eliminate the superimposed random noise pollution. The mixing of two feed streams in a standard continuous stirred tank, one with the fluorescent tracer, the other without, results in a turbulent fluctuating concentration field, used to validate the sensor. Theoretical predictions for the signal processing appear to be relevant. Effects of position, stirring speed, mean residence time and viscosity were studied on local intensity of segregation, spectral density functions, power dissipation spectra and autocorrelations. Minimum values for stirring speed or mean residence time seem to exist, above which fluctuations are no more detectable. Dimensionless reduced spectra take a unique shape, as predicted by the turbulence theory. Viscosity, while increasing segregation, seems no to influence spectra because the Schmidt number was probably not modified. Perpectives of improvement are suggested

Full textThe maximum efficiency limit attainable with a single-junction PV cell is ~ 33% according to the detailed balance formalism (also known as Shockley-Queisser model), which remains far from the Carnot limit, predicting a solar to electricity efficiency upper value of 93%. The large gap between both limits is due to intrinsic loss mechanisms, including the inefficient conversion of the solar spectrum and the large discrepancy between the solid angles of absorption and emission. To overcome these losses and get closer to the Carnot limit, three different strategies are considered in this thesis: concentrated multi-junction solarcells, the combination of solar concentration and angular confinement, and hybrid PV/CSP systems. Each strategy is inherently limited by several loss mechanisms that degrade their performances. The objective of this thesis is, hence, to better understand the extent to which these strategies are likely to be penalized by these losses, and to tailor the cell properties toward maximizing their efficiencies. To address these questions, a detailed-balance model of PV cell accounting for the main loss mechanisms was developed. A genetic-algorithm optimization tool was also implemented, aiming at exploring the parameter space and identifying the optimal operation conditions. We demonstrate the uttermost importance of tailoring the electronic properties of the materials used with both multi-junction solar cells undergoing significant series resistance losses, and PV cells operating at temperature levels exceeding ambient temperature. We also investigate the extent to which series resistances losses and non-radiative recombination are likely to affect the ability of PV cells simultaneously submitted to concentrated sunlight and angular restriction of the light emitted by band-to-band recombination
