Abstract:
Abstract Factor J (FJ) is a cationic glycoprotein that is able to inhibit in vitro both the classical and alternative pathways of complement. FJ was purified to homogeneity from human urine by sequential chromatographic steps. To examine the expression of FJ in human cells we obtained mAbs against urine-purified FJ. Preliminary studies by immunocytochemistry revealed that one of the anti-FJ mAbs recognized cell surface components of certain cell lines, such as K562 and U937 cells, so we have focused subsequently on the detection of these homologue membrane-bound FJ Ags (FJ-h Ags) in cell lines of lymphoid (Ramos and Jurkat) and mieloyd (U937 and K562) origin, as well as in peripheral blood cells. The flow cytometry analysis of the examined cell lines revealed partial staining ranging from 10% (U937) to 29% (K562) positive cells. Flow cytometry of peripheral blood cells showed a positive staining in a small but consistent population of lymphocytes (mean = 11%, n = 17) but none at all on monocytes, granulocytes, erythrocytes, or platelets. Double Ab immunostaining of lymphocytes showed that the FJ-h positive population included mainly B lymphocytes (a mean of 63% CD19+ were FJ-h positive). When we analyzed peripheral blood lymphocytes from a patient with chronic lymphocytic leukemia B (95% CD19+/CD5+), the majority of these (55%) bore FJ-h on their surface. Acid strip of these cells did not abrogate the surface staining, which supports the finding that the Ag is tightly bound to the membrane. Immunoprecipitation from U937 cell lysates showed a single 65 kDa band under reducing conditions. FJ-h Ags purified from K562 and U937 cells displayed inhibitory activity in the functional EAC14 assay for the classical complement pathway, as did urine FJ, and they were recognized immunochemically by five different (one polyclonal and four monoclonal) anti-FJ Abs. In conclusion, FJ-homologues are present in the membranes of several human cell lines that show functional and antigenic characteristics similar to soluble urine FJ. They are also found in a small subset of peripheral blood lymphocytes, mainly B cells. The structural relationship between both soluble urine FJ and these membrane-bound FJ-h remains to be established.

Abstract:
The purpose of this study was to investigate hospital variation in the placement, surgical techniques, and safety of feeding jejunostomies (FJ) during minimally invasive esophagectomy (MIE) in the Netherlands. This nationwide cohort study analyzed patients registered in the Dutch Upper Gastrointestinal Cancer Audit (DUCA) that underwent MIE for cancer. Hospital variation in FJ placement rates were investigated using case-mix corrected funnel plots. Short-term outcomes were compared between patients with and without FJ using multilevel multivariable logistic regression analysis. The incidence of FJ-related complications was described and compared between hospitals performing routine and non-routine placement (≥90%–<90% of patients). Between 2018–2020, an FJ was placed in 1481/1811 (81.8%) patients. Rates ranged from 11–100% among hospitals. More patients were discharged within 10 days (median hospital stay) without FJ compared to patients with FJ (64.5% vs. 50.4%; OR: 0.62, 95% CI: 0.42–0.90). FJ-related complications occurred in 45 (3%) patients, of whom 23 (1.6%) experienced severe complications (≥Clavien–Dindo IIIa). The FJ-related complication rate was 13.7% in hospitals not routinely placing FJs vs. 1.7% in hospitals performing routine FJ placement (p < 0.001). Significant hospital variation in the use of FJs after MIE exists in the Netherlands. No effect of FJs on complications was observed. FJs can be placed safely, with lower FJ-related complication rates, in centers performing routine placement.

Abstract:
Background: Concomitant placement of Feeding Jejunostomy (FJ) tubes is common after pancreaticoduodenectomy for optimizing postoperative nutrition. The aim of this study is to determine the role FJ tube placement for postoperative nutrition following pancreaticoduodenectomy. Methods: All patients undergoing concomitant FJ following pancreaticoduodenectomy (Whipples procedure) from July 2014 to July 2016 were reviewed retrospectively. FJ feeds were routinely started on POD 2. Jejunostomy feeds were discontinued once patient is able to take adequate oral feeds. Data were represented by frequency and mean. Results: A total of 28 patients underwent pancreaticoduodenectomy in the study period and concomitant FJ was performed in all those patients. A total of 18 (64.3%) were men and mean age was 47.1 years. Majority (25, 89.3%) of these patients had malignancy as an indication for surgery. Only 6 out of 28 (21.4%) patients required nutrition supplementation through FJ on POD 7. Only 2 out of 28 (7.1%) patients required FJ feeds on POD 30. None of these patients had tube related complications in 30-day postoperative period. There was no FJ related mortality in 30-day postoperative period. The patients who required prolonged FJ feeds had grade C pancreatic leak, gastrojejunal anastamotic leak and GJ anastamotic stricture. Conclusions: Concomitant FJ can be used as a routine in patients undergoing pancreaticoduodenectomy. It is especially beneficial among patients requiring prolonged postoperative nutritional supplementation due to grade c pancreatic leak and gastrojejunal anastamotic leak. Literature review suggested that one third of the nasojejunostomy tubes dislodge and TPN doubles the risk of infection, hence FJ is considered safe and effective adjunct for patients undergoing pancreaticoduodenectomy.

Abstract:
Dietary guidelines often deal with 100% fruit juice (FJ) inconsistently because it represents a source of free sugars. However, FJ also provides bioavailable micronutrients and plant bioactives at levels similar to those found in whole fruits. The present review weighs up the evidence from high-quality studies investigating a potential health harm for FJ against evidence from studies which indicate a potential health benefit. The findings reveal that FJ consumption, at moderate intakes consistent with the dietary guidelines for the US and some European countries (75–224 mL daily), does not increase the risk of obesity, type 2 diabetes, cardiovascular disease or poor glycaemic control. In contrast, regular consumption of FJ—even up to 500 mL per day in short-to-medium-term studies—appears to confer a health benefit in terms of vascular function and reduced blood pressure. Emerging evidence for cognitive health benefits requires further investigation in human trials. Observational studies report associations between FJ and nutrient adequacy and suggest that FJ consumption is associated with reduced risk of stroke. In conclusion, FJ appears to offer more benefit than risk and there appears to be no justification for discouraging FJ within a balanced diet for children and adults.

Abstract:
ABSTRACT In 2001 and 2003, we isolated two H5N1 viruses, A/swine/Fujian/1/01 (SW/FJ/01) and A/swine/Fujian/1/03 (SW/FJ/03), from pigs in Fujian Province, southern China. Genetically, these two viruses are similar, although the NS gene of the SW/FJ/03 virus has a 15-nucleotide deletion at coding positions 612 to 626. The SW/FJ/01 virus is highly lethal for chickens, whereas the SW/FJ/03 virus is nonpathogenic for chickens when administrated intravenously or intranasally. To understand the molecular basis for the difference in virulence, we used reverse genetics to create a series of single-gene recombinants of both viruses. We found that a recombinant virus containing the mutated NS gene from the SW/FJ/03 virus in the SW/FJ/01 virus background was completely attenuated in chickens. We also found that viruses expressing the mutant NS1 protein of SW/FJ/03 did not antagonize the induction of interferon (IFN) protein. Conversely, only the recombinant virus containing the wild-type SW/FJ/01 NS gene in the SW/FJ/03 background was lethal in chickens and antagonized IFN protein levels. Further, we proved that the NS1 genes of the two viruses differ in their stabilities in the host cells and in their abilities to interact with the chicken cleavage and polyadenylation specificity factor. These results indicate that the deletion of amino acids 191 to 195 of the NS1 protein is critical for the attenuation of the SW/FJ/03 virus in chickens and that this deletion affects the ability of the virus to antagonize IFN induction in host cells.

Abstract:
We retrospectively analyzed CT scans of 620 individuals, who presented to our traumatology department between 2008 and 2010. Facet joint (FJ) arthritis was present in 308 (49.7%) individuals with a mean grade of 1. It was seen in 27% of individuals ≤40 years and in 75% of individuals ≥41 years () as well as in 52% of females and 49% of males (). Mean FJ orientation was 30.4° at L2/3, 38.7° at L3/4, 47° at L4/5, and 47.3° at L5/S1. FJ arthritis was significantly associated with more coronal (increased degree) FJ orientation at L2/3 () with a cutoff point at ≥32°. FJs were more coronally oriented (48.8°) in individuals ≤40 years and more sagittally oriented (45.6°) in individuals ≥41 years at L5/S1 (). Mean FJ asymmetry was 4.89° at L2/3, 6.01° at L3/4, 6.67° at L4/5, and 7.27° at L5/S1, without a significant difference for FJ arthritis. FJ arthritis is common, increases with age, and affects both genders equally. More coronally oriented FJs (≥32°) in the upper lumbar spine may be an individual risk factor for development of FJ arthritis.

Abstract:
ObjectivesThe size and shape of a joint cavity are the key determinates for the mobility of the joint. The anatomy and configuration of the facet joint (FJ) recesses at different levels of the spine remain unclear and controversial. The aim of this study was to identify the configuration of the FJ recesses in the cervical, thoracic and lumbar spine using a combination of micro-CT and sheet plastination techniques.MethodsOf 19 cadavers (9 males, 10 females, age range of 54–89 years), the FJ cavities of 3 spines were injected with contrast filling and scanned with micro-CT, and 16 plastinated spines were prepared as the series of sagittal (9 sets), transverse (5 sets) or coronal (2 sets) sections with a thickness of 2.5 mm and examined under a stereoscopic microscope.ResultsThis study characterised the FJ spaces and recesses of the spine and found that (1) the configuration and extent of the FJ recesses varied along the spine. The optimal needle approach to the FJ cavity was via an anterolateral or posterolateral recess at the cervical level, along the tip of the inferior articular process at the thoracic level and via the posteromedial recess at the lumbar level. (2) The FJ cavity did not communicate with the retrodural space.ConclusionThe anatomical features of the FJ recesses at different levels of the spine confirm no direct communication between the FJ cavity and retrodural space.

Abstract:
The fossil hominid site of Fejej FJ-1 (Ethiopia) is located near the lower Omo valley and the eastern shore of lake Turkana, close to the border of Kenya. A pre-Oldowayan lithic assemblage and a collection of mammal bones were excavated. The FJ-1 sites’ stratigraphy is divided into 5 members, composed river bed and flood plain sediment and a regional volcanic event. The sedimentological study reveals that sediment originated from the metamorphic Hamar Range and from the Oligo-Miocene basaltic High Plateau, both located to the northeast of Fejej FJ-1. The sediment was transported and deposited by a radial river tributary to the Paleo-Omo River. The tephrostratigraphical correlation between the Fejej FJ-1 tuff (member IV) and the G-29 tuff of the Shungura formation and the Borana tuff of the Koobi Fora formation correlated the FJ-1 stratigraphical sequence to the Upper G member of the Shungura and Upper Burgi member of Koobi Fora. This correlation is in agreement with chronological and biostratigraphical results. The latter results, as well as those from the magnetostratigraphy and the regional sedimentation rate allow us propose an age of 1,95 ± 0,05 Ma for the pre-oldowan level of Fejej FJ-1

Abstract:
Trabalho Final de Mestrado Integrado, Ciências Farmacêuticas, Universidade de Lisboa, Faculdade de Farmácia, 2014
FJ 194940.1 gene was shown to be a potential molecular marker of cancer development and progression. This gene is located in chromosome 1, consists of 5 exons and probably undergoes alternative splicing in malignant tissues. Despite promising results in cancer, in other diseases its expression remains unknown. This work consists in the investigation of the expression of this gene in depression, a prevalent, severe and life-threatening disease that lacks biomarkers that could predict therapeutic responses and help prevent side effects. This study has demonstrated the presence of FJ 194940.1 gene in some of the studied cases, however, its expression was only found to be related with two of the analyzed clinical features: age at the time of diagnosis and HDRS before treatment. No statistically significant correlation was found with other clinical features such as gender, HDRS before treatment and HDRS change after treatment. The presence of the protein product of FJ 194940.1 gene was also investigated in serum samples of patients with depression, and in this case, all of them were positive.

Abstract:
In a very traditional village game, popular over the period of Lent (usually on the pigñata day, the first Sunday of Lent), a young player, suitably blindfolded and armed with a long cudgel, tries to hit a pot (the pigñata) located some distance away, in order to win the sweetmeats contained inside. To break the pot they take random steps, and at each step they try to hit the pot with the cudgel. Is it possible to guess the distance of the player from their starting position after n random steps? Is it possible to guess the direction of the vectorial resultant of the n steps? A very simple analysis of the problem suggests that the distance after n steps may be estimated but the direction of the resultant step cannot, because a preferred privileged orientation does not exist. The situation is very similar to structure factor statistics. Each of the N atoms in the unit cell provides the vectorial contribution . . . fj = fj exp(2πih · rj) = fj exp(i θj). . . to the structure factor; this is equivalent to a vectorial step of the pigñata player. The modulus of the atomic contribution, like the amplitude of the step in the pigñata game, is known (because the chemical composition of the molecules in the unit cell is supposed to be known), but the phase θj (corresponding to the direction of the step) remains unknown; indeed we do not know the position rj of the j th atom. The analogy with the pigñata game suggests that some information on the moduli of the structure factors can be obtained via a suitable statistical approach, while no phase information can be obtained using this approach. This chapter deals just with this statistical approach and owing to the relevant contributions of A. J. C. Wilson, we call this chapter Wilson statistics.

Abstract:
Ladner-Fischer [1980] was the first one to demonstrate size vs. depth trade-off in parallel prefix circuits. In this Chapter, a lower bound from Snir [1986] on (size + depth) for prefix circuits is first derived. Several designs of parallel prefix circuits with optimal (size + depth) trade-offs are described. In this section, we derive a lower bound from Snir [1986] on the sum of depth and size of a class of prefix circuits. Let x = { x1, x2, • • • , XN }, be a set of N variables. Let D be the domain over which elements of x take their values, and let FN = { f1, f 2, . . . , fN } be a set of functions satisfying the following conditions: (SR1) fi, depends only on the variables x1, x2 , • • • xi, and (SR2) For each variable xi, i = 1, 2, • • • , N, there exist values ai, in D, such that, the set of functions fj |xi = ai = fj(x1,x2, . . . , xi-1, xi = ai, xi+1, . . . , xj), j=1,2, . . . , N, contains a family of (N - 1) functions satisfying these conditions. The family FN satisfying these two conditions is called a self reducible family of functions. Clearly, fi (x1, x2, . . . , xi) = x1 o x2 o . . . o xi, 1 ≤ i ≤ N, satisfies these conditions. Following is an example of a self-reducible family of functions. An associative binary operation θ is said to be non-trivial if the following conditions are satisfied: (i) Let z = x θ y. Then z depends on both x and y, that is, θ is not a projection, nor a constant operation, and (ii) There exists a (right) unit element e of θ, such that, x = x θ e.

Abstract:
Section 6.5.2 of N283.3-10 of the CSA Standard [1] describes analytical methods for seismic qualification of nuclear components. Clause 6.5.2.2 of this Section instructs how to prepare seismic input for the time history method, specifically it states: “Time-histories of support point motion (displacement, velocity, or acceleration) may be used as dynamic inputs to components. To take into account the effects of possible frequency variations of component and structure, the analysis shall be carried out using three different time-history excitations. These time-histories shall be obtained by varying time scale of the original support point time-history by (a) 1.0; (b) 1 - Δfj/fj; and (c) 1 + Δfj/fj, where fj = the dominant structural frequency; Δfj = a parameter defining the frequency variation due to uncertainties in structural soil properties. The most severe effects obtained from these three time history analyses shall be considered in the design of the components. Notes: (1) A value of 15% for Δfj/fj may be used for the time-history analysis specified in this Clause; (2) For structures directly on bedrock, a value of Δfj/fj = 0% may be used”. This paper identifies some ambiguities in the approach described above. It shows, by theory and examples, that significantly different responses are obtained depending on which form of excitation is used (acceleration, velocity or displacement). In a typical acceleration excitation approach, the response may be over or under estimated. To remove this ambiguity, the paper proposes a simple modification of the broadening procedure described in [1]. The problem discussed in this paper may also be meaningful for the broadening time history described in Appendix N of ASME Code [2].

Abstract:
Abstract We consider the question of recovering coefficients such as q or k from the equation −∇.(k∇uj) + q(x)uj = fj(x) in a domain Ω ⊂ Rn where uj(x) is subject to Dirichlet boundary conditions. The non-homogeneous source terms {fj(x)}j=1∞ form a basis for L2(Ω). We will prove that a unique determination is possible from data measurements consisting of the average flux leaving a fixed subset of the boundary of the region for each input source. The particular aplication we have in mind is the recovery of interior parameters in heat conduction problems given that we can only make measurements on the boundary, but are able to generate a sequence of internal heat sources. An algorithm that allows efficient numerical reconstruction of the coefficients from finite data will be given.
