Abstract:
ABSTRACTBordetella pertussisfimbriae (Fim2 and Fim3) are components of a five-component acellular pertussis vaccine (diphtheria–tetanus–acellular pertussis vaccine [DTaP5]), and antibody responses to fimbriae have been associated with protection. We analyzed the IgG responses to individual Fim2 and Fim3 in sera remaining from a Swedish placebo-controlled efficacy trial that compared a whole-cell vaccine (diphtheria-tetanus-whole-cell pertussis vaccine [DTwP]), a two-component acellular pertussis vaccine (DTaP2), and DTaP5. One month following three doses of the Fim-containing vaccines (DTwP or DTaP5), anti-Fim2 geometric mean IgG concentrations were higher than those for anti-Fim3, with a greater anti-Fim2/anti-Fim3 IgG ratio elicited by DTaP5. We also determined the responses in vaccinated children following an episode of pertussis. Those who received DTaP5 showed a large rise in anti-Fim2 IgG, reflecting the predominant Fim2 serotype at the time. In contrast, those who received DTwP showed an equal rise in anti-Fim2 and anti-Fim3 IgG concentrations, indicating that DTwP may provide a more efficient priming effect for a Fim3 response following contact withB. pertussis. Anti-Fim2 and anti-Fim3 IgG concentrations were also determined in samples from two seroprevalence studies conducted in Sweden in 1997, when no pertussis vaccine was used and Fim2 isolates predominated, and in 2007, when either DTaP2 or DTaP3 without fimbriae was used and Fim3 isolates predominated. Very similar distributions of anti-Fim2 and anti-Fim3 IgG concentrations were obtained in 1997 and 2007, except that anti-Fim3 concentrations in 1997 were lower. This observation, together with the numbers of individuals with both anti-Fim2 and anti-Fim3 IgG concentrations, strongly suggests thatB. pertussisexpresses both Fim2 and Fim3 during infection.

ABSTRACTBordetella pertussisexpresses two serologically distinct fimbriae (Fim2 and Fim3) which are included in the Sanofi Pasteur 5-component acellular pertussis vaccine, and antibody responses to these antigens have been shown to be associated with protection. Studies to date have assessed the IgG response to this vaccine using a copurified mixture of Fim2 and Fim3, and the response to the individual antigens has not been characterized. We have purified separate Fim2 and Fim3 from strains that express either Fim2 or Fim3 and have used these antigens in an enzyme-linked immunosorbent assay (ELISA) to quantify IgG responses following immunization with 5-component acellular pertussis vaccine in 15-month-old, 4- to 6-year-old, and 11- to 18-year-old subjects. All individuals showed increases in Fim2 and Fim3 IgG concentrations following immunization, with 3-fold-greater Fim2 than Fim3 IgG concentrations seen in the younger two age groups. Fim2 IgG concentrations were 1.5-fold greater than Fim3 IgG concentrations in the 11- to 18-year-olds. We have also compared Fim2 and Fim3 IgG concentrations in individuals with prolonged cough who were diagnosed as having recent pertussis using a pertussis toxin (Ptx) IgG ELISA with individuals with prolonged cough but without elevated Ptx IgG concentrations. Individuals with evidence of recent pertussis had greater Fim3 IgG concentrations, consistent with the predominant serotype of isolates obtained in the United Kingdom. However, a surprising number of individuals had moderate Fim2 IgG concentrations despite very few isolates of that serotype obtained in the sampling period.

Abstract:
ABSTRACT Type I fimbriae in Salmonella enterica serovar Typhimurium are surface appendages that facilitate binding to eukaryotic cells. Expression of the fim gene cluster is known to be regulated by three proteins—FimW, FimY, and FimZ—and a tRNA encoded by fimU. In this work, we investigated how these proteins and tRNA coordinately regulate fim gene expression. Our results indicate that FimY and FimZ independently activate the P fimA promoter which controls the expression of the fim structural genes. FimY and FimZ were also found to strongly activate each other's expression and weakly activate their own expression. FimW was found to negatively regulate fim gene expression by repressing transcription from the P fimY promoter, independent of FimY or FimZ. Moreover, FimW and FimY interact within a negative feedback loop, as FimY was found to activate the P fimW promoter. In the case of fimU, the expression of this gene was not found to be regulated by FimW, FimY, or FimZ. We also explored the effect of fim gene expression on Salmonella pathogenicity island 1 (SPI1). Our results indicate that FimZ alone is able to enhance the expression of hilE, a known repressor of SPI1 gene expression. Based on our results, we were able to propose an integrated model for the fim gene circuit. As this model involves a combination of positive and negative feedback, we hypothesized that the response of this circuit may be bistable and thus a possible mechanism for phase variation. However, we found that the response was continuous and not bistable.

AbstractWe determined the molecular epidemiology ofBordetella pertussisisolates to evaluate its potential impact on pertussis reemergence in a population of Mexico. Symptomatic and asymptomatic cases were included. Pertussis infection was confirmed by culture and real-time polymerase chain reaction (PCR). SelectedB. pertussisisolates were further analysed; i.e. clonality was analysed by pulsed-field gel electrophoresis (PFGE) andptxP-ptxA, prn,fim2 andfim3 typing was performed by PCR and sequencing. Out of 11 864 analysed samples, 687 (5.8%) were positive for pertussis, with 244 (36%) confirmed by both culture and PCR whereas 115 (17%) were positive only by culture and 328 (48%) were positive only by PCR. One predominant clone (clone A,n= 62/113; 55%) and three major subtypes (A1, A2 and A3) were identified by PFGE. All 113 selected isolates had the allelic combinationptxP3-ptxA1.The predominant clone A and the three major subtypes (A1, A2 and A3) corresponded to the emerging genotypesptxP3-ptxA1-prn2-fim2-1-fim3-2 andptxP3-ptxA1-prn2-fim2-1-fim3-1. In conclusion, the presence of an endemic clone and three predominant subtypes belonging to the genotypesptxP3-ptxA1-prn2-fim2-1-fim3-2andptxP3-ptxA1-prn2-fim2-1-fim3-1were detected. This finding supports the global spread/expansion reported for these outbreaks associated genotypes.

ABSTRACT Two types of adhesive fimbriae are expressed by Actinomyces; however, the architecture and the mechanism of assembly of these structures remain poorly understood. In this study we characterized two fimbrial gene clusters present in the genome of Actinomyces naeslundii strain MG-1. By using immunoelectron microscopy and biochemical analysis, we showed that the fimQ-fimP-srtC1-fimR gene cluster encodes a fimbrial structure (designated type 1) that contains a major subunit, FimP, forming the shaft and a minor subunit, FimQ, located primarily at the tip. Similarly, the fimB-fimA-srtC2 gene cluster encodes a distinct fimbrial structure (designated type 2) composed of a shaft protein, FimA, and a tip protein, FimB. By using allelic exchange, we constructed an in-frame deletion mutant that lacks the SrtC2 sortase. This mutant produces abundant type 1 fimbriae and expresses the monomeric FimA and FimB proteins, but it does not assemble type 2 fimbriae. Thus, SrtC2 is a fimbria-specific sortase that is essential for assembly of the type 2 fimbriae. Together, our experiments pave the way for several lines of molecular investigation that are necessary to elucidate the fimbrial assembly pathways in Actinomyces and their function in the pathogenesis of different biofilm-related oral diseases.

Abstract:
ABSTRACT Bordetella pertussis fimbriae bind to sulfated sugars such as heparin through the major subunit Fim2. The Fim2 subunit contains two regions, designated H1 and H2, which show sequence similarity with heparin binding regions of fibronectin, and the role of these regions in heparin binding was investigated with maltose binding protein (MBP)-Fim2 fusion proteins. Deletion derivatives of MBP-Fim2 showed that both regions are important for binding to heparin. The role of H2 in heparin binding was confirmed by site-directed mutagenesis in which basic amino acids were replaced by alanine. These studies revealed that Lys-186 and Lys-187 are important for heparin binding of MBP-Fim2, whereas Arg-179 is not required. Peptides derived from H1 and H2 (pepH1 and pepH2) also showed heparin binding activity. Using a series of peptides, in each of which a different basic amino acid was substituted for alanine, we demonstrated that the structural requirements for heparin binding differ significantly among pepH1 and pepH2 peptides. A Pepscan analysis of Fim2 revealed regions outside H1 and H2 which bind heparin and showed that not only basic amino acids but also tyrosines may be important for binding to sulfated sugars. A comparison of the heparin binding regions of Fim2 with homologous regions of Fim3 and FimX, two closely related but antigenically distinct fimbrial subunits, showed that basic amino acids and tyrosines are generally conserved. The major heparin binding regions identified in Fim2 are part of epitopes recognized by human antibodies, suggesting that the heparin binding regions are exposed at the fimbrial surface and are immunodominant. Since B. pertussis fimbriae show weak serological cross-reactivity, the differences in primary structure in the heparin binding regions of Fim2, Fim3, and FimX may affect antibody binding but not heparin binding, allowing the bacteria to evade antibody-mediated immunity by switching the fimbrial gene expressed.

ABSTRACT Type 1 fimbriae of Salmonella enterica serovar Typhimurium are surface appendages that carry adhesins specific for mannosylated host glycoconjugates. Regulation of the major fimbrial subunit is thought to be controlled by a number of ancillaryfim genes, including fimZ, fimY,fimW, and fimU. Previous studies using a FimZ mutant have indicated that this protein is necessary forfimA expression, and in vitro DNA binding assays determined that FimZ is a transcriptional activator that binds directly to thefimA promoter. To determine the role of FimY as a potential regulator of fimbrial expression, a fimY mutant of serovar Typhimurium was generated by allelic exchange. This mutant was found to be phenotypically nonfimbriate. No transcription from thefimA promoter was detected in a fimY mutant containing a fimA-lacZ reporter construct located on the chromosome. In addition, transcription from the cloned fimYpromoter was not detected in Escherichia coli unless both FimZ and FimY were present, indicating that these proteins also act as coactivators of fimY expression. Consistent with these results, there is no transcription from a fimY-lacZreporter construct within a serovar Typhimurium fimY orfimZ mutant. Studies using the fimY-lacZconstruct reveal that expression of this gene varies with environmental conditions in a manner similar to fimA expression. Extensive in vitro DNA binding assays using extracts from E. coli that overexpress FimY, as well as partially purified FimY, were unable to identify a specific interaction between FimY and thefimA or fimY promoter. The results indicate that FimY is a positive regulator of fimbrial expression and that this protein acts in cooperation with FimZ to regulate the expression ofSalmonella type 1 fimbrial appendages.

In many countries of the world despite the extensively vaccination against pertussis has increased the incidence of the whooping cough in all age group of the population. The MLST, MLVA and CGH investigations revealed the differences in genotypes between the vaccine strains B.pertussis and the circulating isolates B.pertussis in consequence of adaptation of the bacterium B.pertussis to the immunized hosts. It is lead to waning immynity and outbreak of incidence of pertussis. The mutations in the genes encoding the major virulence factors, the allelic polimorfism and decreasing the genome size of B.pertussis strains are the basis of the B.pertussis adaptation to the immunized hosts and dependent on the type of the vaccine used for immunization Programme. In countries that use acellular pertussis vaccine for vaccination programme the dominant isolates genotypes are: ptxА1-ptxС2- ptxР3-prn2- tcfA2-1-fim3-2 и ptxА1- ptxС2- ptxР3-prnА2- tcfA2- fim2-1- fim3-1, and that use the cellular pertussis vaccine the dominant isolates genotypes are ptxА1-ptxС1- ptxР1-prn1- tcfA2- fim2-2 fim3-1 и ptxА1- ptxС1- ptxР1- prn2- tcfA2- fim2-1- fim3-1. The constant monitoring of the genotypes of isolates B.pertussis is necessary to reveal the dominant genotypes and include them in the national immunization programme in combination with vaccine strains B.pertussis.

ABSTRACTThe production of type 1 fimbriae inSalmonella entericaserovar Typhimurium is controlled, in part, by three proteins, FimZ, FimY, and FimW. Amino acid sequence analysis indicates that FimZ belongs to the family of bacterial response regulators of two-component systems. In these studies, we have demonstrated that introducing a mutation mimicking phosphorylation of FimZ is necessary for activation of its target gene,fimA. In addition, the interaction of FimZ with FimW, a repressor offimAexpression, occurs only when FimZ is phosphorylated. Consequently, the negative regulatory effect of FimW is most likely due to downmodulation of the active FimZ protein. FimY does not appear to function as a response regulator, and its activity can be lost by mimicking the phosphorylation of FimY. Overproduction of FimY cannot alleviate the nonfimbriate phenotype in a FimZ mutant, whereas high levels of FimZ can overcome the nonfimbriate phenotype of a FimY mutant. It appears that FimY acts upstream of FimZ to activatefimAexpression.

Abstract:
The fimbriae of Bordetella pertussis are required for colonization of the human respiratory tract. Two serologically distinct fimbrial subunits, Fim2 and Fim3, considered important vaccine components for many years, are included in the Sanofi Pasteur 5-component acellular pertussis vaccine, and the World Health Organization recommends the inclusion of strains expressing both fimbrial serotypes in whole-cell pertussis vaccines. Each of the fimbrial major subunit genes, fim2, fim3, and fimX, has a promoter poly(C) tract upstream of its −10 box. Such monotonic DNA elements are susceptible to changes in length via slipped-strand mispairing in vitro and in vivo, which potentially causes on/off switching of genes at every cell division. Here, we have described intra-culture variability in poly(C) tract lengths and the resulting fimbrial phenotypes in 22 recent UK B. pertussis isolates. Owing to the highly plastic nature of fimbrial promoters, we used the same cultures for both genome sequencing and flow cytometry. Individual cultures of B. pertussis contained multiple fimbrial serotypes and multiple different fimbrial promoter poly(C) tract lengths, which supports earlier serological evidence that B. pertussis expresses both serotypes during infection.

Abstract:
A Ética Nicomaqueia tem como tema central a felicidade (eudaimonia). No entanto, há um problema, pelo menos aparente, em relação a como a eudaimonia é caracterizada principalmente no livro I e no livro X da EN. A partir do que é desenvolvido no primeiro livro e nos livros centrais da EN, podemos entender que a felicidade contém boas ações, isto é, ações de acordo com as virtudes de caráter. No entanto, no livro X, a felicidade é identificada com a contemplação, ou seja, o exercício da virtude teórica, e as ações moralmente virtuosas acabam ficando em uma posição inferior. Surge, então, o problema de determinar se a felicidade é um ou mais bens e como combiná-los. Esse problema foi apontado por W. F. R. Hardie no artigo “O bem final na ética de Aristóteles” e, a partir de sua publicação, iniciou-se uma discussão a respeito de a eudaimonia ser um fim inclusivo ou um fim dominante. O objetivo deste trabalho é investigar a noção de eudaimonia na EN a partir da discussão mencionada acima. Para isso, analiso as passagens e os conceitos importantes para avaliar os argumentos apresentados e as interpretações sustentadas pelos comentadores inseridos nessa discussão. Ao longo do trabalho, mostro que a identificação da eudaimonia com a atividade teórica virtuosa não é incompatível com o exercício das virtudes morais e que ambos fazem parte da eudaimonia.
The central theme of the Nicomachean Ethics is happiness (eudaimonia). However there is a problem, at least apparent, regarding the characterization of eudaimonia, particulary in books I and X of the NE. Based on what is developed in the first and central books of NE we can understand that happiness contains good actions, that is, actions in accordance with character virtues. In book X, however, happiness is identified with contemplation, that is, the exercise of theorethical virtue, and morally virtuous actions ends up occupying a lower position. What follows is the emergence of the problem of determining whether happiness is one or more goods and how to combine them. This problem was pointed out by W. F. R. Hardie in the paper “The final good in Aristotle's Ethics”, and since its publication, it started a discussion about eudaimonia being an inclusive or a dominant end. The aim of this work is to study the idea of eudaimonia in the NE based on the discussion mentioned above. To achieve that, I analyze important passages and concepts to assess the arguments presented and the interpretations sustained by critics involved in the discussion. In this study, I show that the identification of eudaimonia with theorethical activity is not inconsistent with the exercise of moral virtues, and that both are part of eudaimonia.

Tese apresentada à Universidade Fernando Pessoa como parte dos requisitos para obtenção do grau de Doutor em Ciências Sociais, especialidade em Sociologia
Nos anos terminais da Guerra do Ultramar, a Academia Militar deixou de cumprir, por falta de candidatos, a sua missão capital: formar as elites militares intermédias de combate. Os Capitães do Quadro Permanente soçobraram. Ficou-se em presença de um Exército quase miliciano, num clima de forte contestação à guerra e de fraca motivação para lhe dar continuidade. No entanto, a defesa do Império, a todo o custo, era a política vigente, apesar do visível cansaço da Nação. Para ajudar a obviar este desiderato, e de forma surpreendente, foi determinada por despacho de 20 de Julho de 1970 do Ministro do Exército, Horácio José de Sá Viana Rebelo, a formação acelerada de Capitães milicianos na Escola Prática de Infantaria. Formaram-se à volta de cento e sessenta por ano. Em cerca de catorze meses fazia-se de um estudante universitário, quase sempre, em estádio avançado de licenciatura ou já licenciado, um Capitão combatente para actuar nos teatros de guerra mais exigentes de Angola, da Guiné e de Moçambique. O modo de selecção destas novas elites pelejadoras, e a formação apressada a que foram sujeitas, deram ensejo a que alguns as apelidassem, desdenhosamente, de “Capitães de proveta” ou “de “aviário”. Aplica-se, neste contexto, em alternativa, a expressão Capitães do Fim. Como foram seleccionados, formados e que desempenhos e protagonismos tiveram estes Capitães, nos teatros de Angola, da Guiné e de Moçambique, no comando de Companhias em quadrícula ou independentes de intervenção? A resposta tem enquadramento na sociologia e na história, ainda que de especificidade militar, em coerente continuum de procedimentos metodológicos qualitativos e quantitativos. Com recurso a autobiografias e a histórias de vida de guerra de muitos dos actores-Capitães ainda vivos, valorando-se dessa forma o tecido da participação-acção, entrecruzando os fios de vida das pessoas com aquilo que foi o seu terreno, o irrepetível e o individual, e contribuindo para a construção de conhecimento, procura trazer-se luz a um assunto que, no tempo, vai certamente, com reinterpretações, ter continuidade de investigação.
In the final years of the Ultramarine War, due to candidates´ scarcity, the Military Academy stopped complying to its core mission - to train the middle ranked fighting military elites. The commissioned captains foundered. All left was a quasi-militia army amongst strong opposition to war and lack of motivation to pursue it. However, the stated policy was the protection of the Empire, at all costs, despite the evident strain of the Nation. To support this aspiration, and surprisingly, it was determined, by the promulgation of the Army Minister, Horácio José de Sá Viana Rebelo, decree of 20th July 1970, to speedily train a militia of captains at the Infantry Practical School. Around one hundred and sixty captains were graduated per year. Within about fourteen months, a university student, often in later stages of his five-year full-time first degree (licenciatura), or already graduated, was converted into a fighting captain to act in the most demanding war circumstances in Angola, in Guinea and in Mozambique. The selection methods of these new belligerent elites and their rushed training, led to their, pejorative, appellation of the "beaker” or “avian” captains. Alternatively, henceforth, in this context, they will be referred to as Captains of the End. How were these captains selected, trained and which performance and role had in the war environments in Angola, in Guinea and in Mozambique, in charge of both territorial companies or independent platoons? The answer is framed in sociology and history, even though with military specificity, in coherent continuum of qualitative and quantitative methodological procedures. Resorting to autobiographies and histories of war episodes of many still alive actors-captains, and hence valuing the fabric of the participation-action, and crisscrossing life yarns of people with what was their own grounds, the unrepeatable and the individual, and hence contributing to building knowledge, to shine light on one subject that, in the future, will certainly, be subject to reinterpretations and further research.
Au cours des années des terminaux guerre outre-mer, l'Académie militaire n'a pas respecté, faute de candidats, leur capitale mission: former des élites militaires provisoires de lutte. Le cadre permanent de capitaines a sombré. Est resté en présence d'une armée presque milicien dans un climat de forte opposition à la guerre et de la faible motivation de donner une continuité. Toutefois, la défense de l'Empire était à tout prix la politique en vigueur, malgré la fatigue visible dans la nation. Pour remédier à ce souhait et étonnamment, il a été déterminé, par arrêté du ministre de l’armée, 20 juillet 1970, Horácio José de Sá Viana Rebelo, accéléré la formation des capitaines de milice à l’école d'infanterie. Formé d'environ cent soixante par an. À environ quatorze mois était un étudiant, presque toujours, à un stade avancé de degré, ou déjà une licence, un combattant de capitaine à effectuer dans les théâtres les plus exigeants de la guerre en Angola, en Guinée et au Mozambique. La sélection de ces nouvelles élites combattants et la formation hâtive dont ils sont victimes, a donné lieu à certains de l'appellent, avec mépris, comme «in vitro» ou capitaines de «Volière». S’applique, dans ce contexte, comme alternative, les Capitaines de la Fin. Comment ont été sélectionnés, formés et ce en direct et l'envie d'avoir ces capitaines dans les théâtres de l'Angola, en Guinée et au Mozambique, en charge de compagnies de la grille ou indépendante de l'intervention? La réponse a cadre en sociologie et en histoire, quoique spécificité militaire cohérente continuum des procédures méthodologiques qualitatives et quantitatives. En utilisant les autobiographies et des récits de vie de beaucoup de joueurs guerre-capitaines encore en vie, en prenant soin lui-même, de cette manière, le tissu de participation-action, traversant les fils de la vie des personnes atteintes de ce qu’était sa terre, l’inimitable et l’individu et qui contribuent à la construction des connaissances, cherche à apporter la lumière sur une question qui, dans le temps, va certainement, avec des réinterprétations, avoir une continuité de la recherche.

Resumo: Tem a presente pesquisa o intuito de identificar e mapear na obra ainda em formação do filósofo italiano Giorgio Agamben o percurso que, do ponto de vista da filosofia política, leva de um composto de reflexões em torno da crítica da cultura às incursões decisivas do que aqui chamei de crítica do jurídico, priorizando num primeiro momento as leituras de Infanzia e storia, La comunità che viene e Il tempo che resta, e num segundo momento os livros que se inserem no projeto Homo sacer, especialmente Homo sacer: il potere sovrano e la nuda vita, Stato di eccezione e Quel che resta di Auschwitz. Se o nosso século é aquele em que a sociedade tornada espetacular culmina na erosão de toda experiência possível, ele é também aquele em que os conceitos jurídicos perdem sempre mais sua materialidade: em nome da defesa do direito chegamos contraditoriamente a uma realidade jurídica rarefeita. Explorando os diálogos estabelecidos pelo filósofo com outros filósofos, tais como Walter Benjamin e Michel Foucault, e outras áreas do saber constituído, tais como a história, a literatura ou a teoria social, o texto que se segue buscará permitir visualizar um complexo diagnóstico. Através do uso de conceitos políticos basilares e do auxílio de determinadas figuras paradigmáticas, veremos como o liame entre soberania, exceção e vida nua contamina todo o espaço político contemporâneo. O fruto do trabalho que agora se apresenta não quer todavia somente decifrar ou diagramar um cenário decomposto. Pode ser que, em última instância, ele queira também contribuir, ainda que modestamente, para desobstruir - como escreve o filósofo na introdução do primeiro livro da série Homo sacer - o campo em direção à nova política que ainda resta inventar
Abstract: The present research's goal is to map and identify a path in Giorgio Agamben's on-going work that, from the perspective of political philosophy, leads from a cluster of reflections around cultural critics to decisive incursions of what here I called juridical critics, prioritizing in the first moment the readings of Infanzia e storia, La comunità che viene and Il tempo che resta, and in a second moment the books which belong to the project Homo sacer, specially Homo sacer: il potere sovrano e la nuda vita, Stato di eccezione and Quel che resta di Auschwitz. If our century is the one in which society becoming spectacular culminates in the erosion of every possible experience, it is also the one where the juridical concepts loose more and more their materiality: in behalf of right's defense we arrive contradictorily at a thin juridical reality. Exploring the dialogs established by Agamben with others philosophers, such as Walter Benjamin and Michel Foucault, as well with others areas of constitutive knowledge, like history, literature or social theory, the text that follows will search to allow the visualization of a complex diagnosis. Through the use of basic political concepts and with the assistance of some paradigmatic figures, we shall see how the bond between sovereignty, exception and bare life contaminates the entire contemporaneous political space. However, the fruit of the work which now comes to presentation does not only intend to decipher or to diagrammatize a decomposed scenario. It might be that, in the last instance, it also wishes to contribute, even if modestly, to open the field up - or as the philosopher writes in the introduction of the first book of his Homo sacer series - to clean the way towards the new politics which remains largely to be invented

Diversas aplicações de redes de sensores sem fio necessitam de serviços de segurança, como confidencialidade, integridade e autenticação de origem de dados. Contudo, dadas as limitações de processamento, memória e suprimento de energia dos dispositivos, os mecanismos de segurança tradicionais podem causar efeitos indesejáveis na rede, como atraso na comunicação e aumento no consumo de energia, impondo obstáculos para seu uso na tecnologia em questão. Muitas propostas de esquemas de segurança baseados em criptografia simétrica projetados especificamente para redes de sensores sem fio são encontradas na literatura. Contudo, essas soluções são focadas na segurança salto-a-salto. Tal abordagem é adequada para garantir a segurança dos enlaces deste tipo de rede, mas não garante a segurança na comunicação fim-a-fim. Neste trabalho são apresentados cenários e desafios de implementação de segurança neste tipo de rede, e a concepção, o projeto e a implementação de uma arquitetura de segurança para redes de sensores sem fio, que tem como objetivos: prover segurança na comunicação fim-a-fim; permitir a interoperabilidade entre diferentes sistemas; e possibilitar uma maior flexibilidade em relação à utilização de chaves criptográficas em diferentes cenários e topologias. Adicionalmente, a solução proposta suporta ativação e desativação de seus serviços em tempo de execução. O projeto da referida arquitetura, atuante na camada de aplicação da pilha de protocolos de rede, foi construído com base na análise das características de arquiteturas encontradas na literatura, bem como de estratégias adotadas por estas. Para a construção da implementação foram selecionados mecanismos e algoritmos criptográficos a partir da avaliação de desempenho que considerou assimétricas de uso de memória, tempo de execução e consumo de energia. Como resultados são apresentados a especificação da arquitetura, a avaliação qualitativa da mesma e a avaliação de desempenho da implementação desenvolvida como prova de conceito. Além disso, é apresentada uma análise do impacto de diferentes topologias e características de disposição na tarefa de distribuição de chaves criptográficas em redes de sensores sem fio.
Many wireless sensor networks applications need security services, such as confidentiality, data integrity and data source authentication. On the other hand, because of device limitations, security mechanisms may affect the network energy consumption and communication delay, which impose a great challenge for practical implementation of security mechanisms in such scenario. Many solutions based on symmetric cryptography were proposed for the specific challenges of wireless sensor networks. Nevertheless, they are focused on hop-by-hop security. Such approach is suited to provide link-layer security, but it cannot guarantee end-to-end security. This work presents scenarios and challenges to implement security in wireless sensor networks, and the conception, design and implementation of a security architecture, which aims to provide: security in end-to-end communication; interoperability between different systems, and enable greater flexibility in cryptographic keys distribution in different scenarios and topologies. Additionally, the proposed solution supports on-the-y adjustment of its security services. The architecture design, which targets the application layer of the network protocol stack, was based on the main properties of the architectures found in literature as well as adopted strategies. For the implementation, mechanisms and cryptographic algorithms were selected through the performance evaluation that considers memory usage, execution time and power consumption as metrics. The results were the architecture specification and its qualitative analysis, and the performance evaluation of the implementation developed as proof of concept. Furthermore, we present an analysis of topology and deployment impact on key distribution task.

O uso da tecnologia de voz sobre IP (VoIP) vem se tornando cada vez mais comum no mundo todo. No entanto, a implantação dessa tecnologia associada à necessidade de garantia de sigilo do canal compatível a sua precursora, a telefonia comutada, ainda é um desafio. Apesar de haver certo consenso em como prover o transporte seguro do dado multimídia transportado em uma conferência, ainda existe discussões de como efetuar a negociação de forma protegida das chaves que permitirão a criação do canal criptografado. Quando se deseja criar esse canal de forma independente de qualquer infra-estrutura de segurança existente, tais como nos cenários fim-a-fim, onde apenas os terminais envolvidos devem ser os responsáveis por essa proteção, as alternativas se resumem a apenas três: o uso dos protocolos S/MIME, MIKEY ou ZRTP. Apesar desses frameworks resolverem satisfatoriamente o problema, eles possuem limitações que os impedem de atender todos os cenários de implementação existentes. O presente artigo apresenta um novo modo de transporte para o MIKEY, o MIKEY-DHHMAC-SAS, que mantém as características principais do protocolo, mas adiciona algumas existentes no ZRTP, solucionando assim as limitações dos protocolos existentes, podendo ser usado em qualquer conferência segura fim-a-fim.

Este trabalho visa a implementação e análise de um ambiente formado por redes WiMAX e MetroEthernet. A rede WiMAX oferece acesso de banda larga sem fio com capacidade ampla de cobertura de transmissão para áreas dispersas. Essa rede, no entanto irá permitir a interconexão com redes MetroEthernet, com aplicabilidade principalmente para órgão públicos e de pesquisa, que proporcionam uma infra-estrutura óptica moderna e de alta velocidade com conexão de até 10Gbps. O mapeamento aqui apresentado trata de uma implementação de Qualidade de Serviço (QoS) baseada na crescente demanda por banda para trafegar aplicativos como dados, vídeo e voz no cenário apresentado anteriormente. O ambiente de teste implementado, condizente com a arquitetura de uma rede MetroEthernet e WiMAX, nos possibilitou obter métricas de configuração e mapeamento de Qualidade de Serviço (QoS) para as futuras redes da INFOVIA. Os resultados obtidos foram amplamente positivos baseados nas métricas de Qualidade de Serviço. Até a elaboração deste trabalho não existe um estudo teórico e prático das características de tráfego de tempo real em redes WiMAX inter-conectando MetroEthernet no Brasil. _________________________________________________________________________________________ ABSTRACT
This work aims the implementation and analysis of an environment formed by WiMAX and MetroEthernet networks. WiMAX offers unwired broadband access with high capacity of data transmission for dispersed areas. This network, however will allow the interconnection of MetroEthernet, with public agencies and research centers, providing a modern optical infrastructure and high speed connection up to 10Gbps. The mapping herein presented deals with an implementation of Quality of Service (QoS) based in the increasing demand for band to allow data, video and voice traffic previously presented in the above scenario. The testbed deals with the architecture of a MetroEthernet and WiMAX network, which made possible obtaining new metrics for configuration and mapping of Quality of Service (QoS) for future INFOVIA networks. The results are widely positive and are based on QoS metric ones. Until the elaboration of this work a theoretical and practical study of the characteristics of real time traffic in WiMAX interconnecting MetroEthernet networks in Brazil does not exist.

Abstract:
O Bom Fim mudou muito nos últimos anos. A vida noturna, que era tida como sua característica principal nas décadas de 1980 e 1990, não está mais lá. O cinema e os bares também não fazem mais parte daquela paisagem. A intenção da presente dissertação é compreender este momento do bairro que acabou. Para isso, foram utilizados os conceitos de espaço e lugar de Michel de Certeau. Segundo este autor, o lugar é constituído a partir da ação disciplinadora que organiza cada coisa em uma localidade específica, uma separada da outra. O espaço é o lugar praticado. Nos grandes centros urbanos se impõe a disciplina, enquanto seus habitantes se apropriam dos limites impostos, criando brechas. O lugar é a ordem e busca a permanência no tempo, o espaço muda constantemente. Práticas criativas cotidianas transformaram a vida do Bom Fim desde o seu começo. Durante as duas últimas décadas do século XX, as práticas noturnas ocuparam o bairro, modificando ele e as representações dele. A porta de entrada para este novo Bom Fim foi um conjunto de bares próximo a UFRGS, conhecido como Esquina Maldita. Desde a década de 1970 tem ocorrido um intenso crescimento urbano e populacional na cidade de Porto Alegre. Durante esse processo, houve a abertura do bairro enquanto um novo terreno de ação, onde era possível praticar novas formas de comportamento, explorar novos caminhos e novas maneiras de produzir arte na cidade. A partir da Esquina ocorreu um movimento de expansão, exploração e ocupação das outras ruas, cinemas, bares, auditórios e do parque da Redenção. Muitos bares começaram a surgir na Avenida Osvaldo Aranha e nas transversais. Mais pessoas começaram a ir a esses bares e locais. A Esquina, filmes, músicas, shows, bares ajudaram a fixar o Bom Fim como um espaço aberto, jovem, de boêmia transgressora e produção artística intensas, peculiares e criativas. A partir do final da década de 1980 foi imposto um disciplinamento que acabou com estas características.
The Bom Fim has changed a lot on the last couple of years. The night live, a main characteristic trough out the 80's and 90's, it is not there anymore. The movie theaters and bars are not part of its landscape. The intention of the present study is to understand this neighborhood moment that has ended. To do that, Michel de Certeau concepts of place and space were used. According to the author, the place is constructed from the disciplinary action that organizes everything in a specific location, one apart from the other. The space is the place in practice. In large urban centers discipline is forced, while its habitants appropriates the imposed limits, creating gaps. The place is the order and seeks permanence on time, space is constantly changing. Creatively daily practices have transformed Bom Fim's life since it is beginning. During the last two decades of 20th century, night practices occupied neighborhood, modifying it and its representations. The gateway of this new Bom Fim was set of bars next to the UFRGS, in a corner known as the Esquina Maldita. Since the 1970's, the city of Porto Alegre has been experiencing an intense urban and populational growth. During this period, the neighborhood appears as a new field of action, where it was possible to practice new forms of behavior, explore new paths and new ways of producing art in the city. From the Esquina up there was a movement of expansion, exploration and occupation of the other streets, movie theaters, bars, auditoriums and Redenção Park. Many bars began to arise on Osvaldo Aranha Avenue and its cross streets. More people have started to attend to this establishments and bars. The Esquina, movies, songs, shows, bars helped to determined Bom Fim as an young, open space, of intense transgressive bohemia and intense artistic production, peculiars and creatives. A disciplinary process was imposed at the late 1980's that consummate those characteristics.

O fatiamento de rede tem se apresentado como uma tecnologia promissora para o gerenciamento de recursos em redes de computadores modernas como o 5G, criando diferentes redes virtuais sobre uma mesma infraestrutura física. Considerando escalabilidade, flexibilidade e aspectos financeiros, simuladores têm se tornado um possível ambiente para avaliar novas soluções desenvolvidas para esse cenário. Nesse contexto, o simulador MobFogSim foi desenvolvido para avaliar soluções em ambientes de Computação em Nuvem e Névoa, incluindo o suporte para migração de serviços, fatiamento de rede e gerenciamento da mobilidade dos usuários. Neste trabalho, apresentamos as novas funcionalidades do simulador em termos de escalabilidade e suporte ao fatiamento ponta-a-ponta, o que inclui recursos de armazenamento e processamento. Simulações considerando dados reais como parâmetros mostraram o impacto de diferentes critérios na alocação de recursos das redes virtuais, além do aumento da escalabilidade do simulador.

Abstract:
Full information maximum likelihood (FIML) is an important approach to compensating for nonresponse in data analysis. Unfortunately, only a few software packages implement FIML and even fewer have the capability to compensate for missing not at random (MNAR) nonresponse. One of these packages is Statistical Innovations’ Latent GOLD; however, the user documentation for Latent GOLD provides no mention of this capability. The purpose of this paper is to provide guidance for fitting MNAR FIML models for categorical data items using the Latent GOLD 5.0 software. By way of comparison, we also provide guidance on fitting FIML models for nonresponse missing at random (MAR) using the methods of Fuchs (1982) and Fay (1986), who incorporated item nonresponse indicators within a structural modeling framework. We compare both FIML for MAR and FIML for MNAR nonresponse models for independent and dependent variables. Also, we provide recommendations for future applications of FIML using Latent GOLD.
