The MK 92 MOD 2 Fire Control System is a complex weapons system based on 1970's technology. It is a maintenance intensive system, requiring extensive technical trouble-shooting and, occasionally, supplemental shore based support. Development of an expert maintenance system for the MK 92 MOD 2 Fire Control System offers a viable solution to the labor intensive efforts of the technicians, reduces the number of Visits by shore based support staff, and provides relief to an already overburdened maintenance budget. It will also significantly reduce the depot repair 'no fault evident' rate which is the result of good parts replaced because of defective trouble shooting. This thesis addresses the first iteration of prototype development of the performance channels of the MK 92 MOD 2 Maintenance Advisor Expert System. Specific issues covered include the scope of the project, hardware selection, system shell selection, knowledge acquisition, knowledge representation, knowledge implementation, and lessons learned in the process.

Abstract:
The conserved N-linked glycosylation site on the Fc domain of IgG1 antibodies is essential for maintaining a functionally active conformation of the antibody. Different glycoforms of the Fc exhibit widely different effector functions. Similarly, therapeutic antibodies, with engineered glycosylation, exhibit altered binding to cellular Fc receptors (FcRs). Here, X-ray crystallographic structures were obtained for biosynthetic intermediate glycoforms of human IgG1 Fc bearing: unprocessed oligomannose-type, intermediate hybrid-type, and mature complex-type glycans. The fully processed Fc protein crystallised in an “open” conformation with glycans forming canonical stabilising interactions on the protein surface. Analysis of the biosynthetic intermediates revealed that these stabilising hydrophobic protein-glycan interactions are formed only after processing by Golgi -mannosidase II. Mutagenesis of hydrophobic residues on Fc disrupted crucial protein-glycan interactions resulting in the selective destabilization of the 3-arm of the glycan chain with the 6-arm closely matching that seen for the native structure. However, carbohydrate analysis of released glycans shows increased processing on both arms indicating a more accessible and flexible glycan in the mutant structure suggesting that the crystallographic structure of these antibody glycans represents a minor low-energy conformation. The importance of Fc glycosylation is highlighted by endoglycosidases which eliminate Fc effector function. The crystallographic structure of enzymatically deglycosylated IgG Fc revealed a significant collapse of the of Cγ2 domains resulting in a ‘closed’ quaternary conformation, incompatible with Fc receptor binding. This provides a structural explanation for immune deactivating properties of endoglycosidases including those under preclinical development for the treatment of antibody-mediated immune pathology. One such bacterial endoglycosidase, Endo S, was studied further and revealed a specificity for complex-type glycans of the type found on IgG but no hydrolytic activity towards an engineered IgG Fc with oligomannose-type glycans. Introduction of both the engineered monoclonal IgG and endoglycosidase in serum led to a dramatic increase in FcR binding as the competitive binding of serum IgG for FcRs was selectively eliminated. This approach is a general technique for boosting the effector signal of therapeutic antibodies.

<p>TQL Stadium is the first dedicated home for FC Cincinnati, a major league soccer team. Buro Happold provided structural engineering services for the stadium. The stadium design meets all Major League Soccer criteria as well as being able to host FIFA sponsored events. All 26,000 seats in the newly designed stadium have excellent sightlines. A complete, 360-degree canopy roof covers every single seat in the stadium. Particular special characteristics of the structural design included the many varieties of seating and hospitality arrangement, the fast schedule and the structural façade interaction. This paper undertakes four exercises in order to get a better understanding of these vulnerabilities. First, probit/logit estimations are used to identify the firm-level and macroeconomic determinants of FC borrowing by non-financial corporations. Second, the implications of the balance sheet vulnerability for real activity are investigated. Evidence is found of an FC balance sheet effect that transmits exchange rate fluctuations to firm-level investment, and show that that this effect is asymmetric, much greater for depreciations than for appreciations. Third, using logit/probit estimations, it is shown that not all firms use forward exchange derivatives solely to hedge their FC liabilities. This might be a consequence of exchange rate intervention by the monetary authority, protecting against extreme exchange rate misalignments. Finally, results are reported of a survey-based qualitative analysis on the hedging policies and activities of 12 large non-financial firms.

Abstract:
This study evaluates the visual performance of four intersection lighting designs and five midblock crosswalk lighting designs along with two pedestrian safety countermeasures (rectangular rapid flashing beacons and flashing signs) at three light levels. The study involved a pedestrian detection task, which was completed at night on a realistic roadway intersection and a midblock crosswalk. The results from the study showed that driver nighttime visual performance at intersection and midblock crosswalks was influenced by the lighting design and light level. Intersections should be illuminated to an average horizontal illuminance of 14 lux (1.3 fc). This light level ensures optimal visibility of pedestrians regardless of the lighting design (or luminaire layout) of the intersection. The average horizontal illuminance of 14 lux (1.3 fc) also increases the visibility of pedestrians when glare from oncoming vehicles is present. The 14 lux (1.3 fc) average horizontal illuminance is valid for all lighting designs evaluated except the lighting design that illuminated the exits of the intersection. When the exits of the intersection are illuminated, an average horizontal illuminance of 24 lux (2.2 fc) is needed to offset the disability glare from opposing vehicles. Midblock crosswalks should be illuminated to an average vertical illuminance of 10 lux (0.9 fc) to ensure optimal pedestrian visibility. Where overhead lighting is available, midblock crosswalk lighting designs that render the pedestrian in positive contrast are recommended. Where overhead lighting is not available, crosswalk illuminators can be used to illuminate midblock crosswalks. At night, pedestrian crossing treatments such as rectangular rapid flashing beacons and flashing signs should not be used for pedestrian visibility at midblock crosswalks. Pedestrians crossing treatments should be used in conjunction with overhead lighting or crosswalk illuminators at the established vertical illuminance to ensure optimal pedestrian visibility at midblock crosswalks.
