Abstract:

Past research has shown that entrainment facilitates social bonding in intimates and strangers. The current study investigated if synchronicity in romantic couples is disrupted by relationship discord. Rocking chair movements were used as an objective measure of synchronicity. Couples rocked together for 3 minutes to assess their baseline synchronicity, and again for 3 minutes after inducing a threat to the relationship in one member of each couple. It was hypothesized that satisfied couples would be more entrained than dissatisfied couples at baseline, as well as after inducing a relationship threat. Results indicated no significant difference in rocking between satisfied and dissatisfied couples at baseline. However, results supported that synchronicity was significantly disrupted in dissatisfied couples, but not satisfied couples, after the threat was induced. These results suggest that relationship satisfaction acts as a buffer to relationship threats and/or that satisfied couples are more likely to remain entrained even in the face of hardship.

Abstract:
This research explores the question of what role medical family therapists play on a health care team when working with serious pediatric illness. Seven participants from three different health care settings were interviewed. Results were organized within the ecological systems framework (Bronfenbrenner, 1979) according to the various ways participants became a part of the familyâ s illness experience. Participants identified roles directly with the family, with the health care team, and within the larger health care system. Clinical implications are identified concerning the preparation of the health care system for the continued growth and evolution of the field of medical family therapy.

Abstract:
The purpose of this study was to explore the effects of perceived family competence on late adolescent problem-solving abilities, family relationships, and affective experience. Specific areas of interest were perceived confidence in problem-solving and approach rather than avoidance of problems; intergenerational intimacy, intergenerational individuation, and personal authority in the family system as the adolescent relates to parents; and level of depression. Subjects were 256 late adolescents whose parents were still married and living together. Results indicated that perceived family competence had an effect on the dependent variables in the expected directions. Specifically, individuals who scored high on perceived family competence were high on perceived problem-solving confidence, approached problem-solving, were high on intergenerational intimacy, intergenerational individuation, and personal authority in relation to parents, and had less depression than individuals low on perceived family competence. Several sex differences were noted. Females had significantly higher approach to problem-solving than did males. Women reported significantly higher intergenerational intimacy with parents than did men. There was a significant interaction on personal authority such that for the high perceived family competence group, women scored higher than men. However, there were no significant differences between males and females in the low perceived family competence group.

Our presentation was aimed at providing a thorough overview of concepts that interfere with an adolescent’s ability to stay in school and graduate. Additionally, the presentation addressed what other factors of poverty, such as risk-taking behaviors (e.g., marijuana use, binge drinking, or sexual activity), might carry over into college life and affect a student’s academic career. Variables connected to family community, family responsibilities, and adolescents’ self-esteem were described as well. Finally, the presentation discussed factors that play into a student’s willingness to seek out college campus resources for support. With this paper, we hope to outline variables that lead to academic drawbacks within our youth and to raise awareness about the multi-faceted phenomenon of retention of students.

Abstract:
According to the American Veterinary Medical Association, there are currently more than 60 million pet dogs in the United States. This is an increase of nearly eighteen percent since 1991, coinciding with a growing area of research on human’s relationships with companion animals and companion animals’ place in society. For years dogs have been thought of as “man’s best friend” because of their loyalty and faithfulness. The increasing popularity of activities such as canine daycare and puppy school suggests that dogs have become more than a best friend to some and even an integral part of the American family unit. The bond and emotional connection between humans and canines is a unique relationship, yet the depth of that relationship is not fully understood academically. In order to contribute to our understanding of this special bond, I conducted seven in-depth interviews with canine companions. My research allowed me to explore how contemporary Americans understand their relationship with their companion dogs. Not only was I able to shed more light on how people think about and treat their canine companions, but I also investigated what benefits are reaped from relationships with dogs. Based on my informants’ reflections and stories, it became clear that their canines were more than just pets. The people in my study described dogs as their best friends, babies and even sons. My interviewees described canine companions who are active participants in their families and in human social life in general. Those who hope to understand this life cannot afford to ignore the canine companion’s changing and important contributions to society.

Abstract:
This thesis explores how Dual Income No Kids (DINK) couples within the United States approach family planning. The study is based on ethnographic work I carried out over the course of 2011, including a nationwide survey and in-depth interviews I conducted in Atlanta, Georgia, Fort Lauderdale, Florida, and Denver Colorado. Specifically, I was interested in investigating why these couples were “delaying” having children based on the national average. While current literature points to changes in education, healthcare, and societal values as being the catalyst for the DINK movement, I wanted to understand Americans’ childbearing decisions on a more personal level. Through this project I looked at how both the social goals (parent and peer role models) and personal pressures (prioritization of education, career and marital partnership) influence an individual’s decision about whether and when to have children. As such, I also explore themes of identity, life narrative, and choice in regards to family planning. Whereas the popular stereotype of DINK suggests that these couples are uninterested in family or “family values”, my research shows that many couples actually choose to be DINK for a time because they are actively pursuing and preparing for parenthood.

Abstract:
Purpose – The purpose of the research is to explain how and why the internal and external influences faced by family-run wineries affect their CSR practices and communication through institutional theory and stakeholder theory. Methodology – Ten wineries were analyzed in winemaking regions in Hungary and the US, of which nine semi-structured interviews were undertaken with family members and employees.  The research available implies that family-run businesses within the wine industry are regulated by institutionalism (Brundin & Wigren-Kristoferson, 2013). Therefore, institutional theory is applied to the analysis of the data. The lens of institutional theory uses three pillars, regulatory, normative, and cultural cognitive to identify the external and internal influences on these businesses.  Thematic analysis was utilized to code and analyze the data. In addition, the content of each interviewed winery is examined using thematic analysis. Thematic analysis is also applied to content provided on each winery’s website to back up the data. Findings –In Hungary, the use of sustainable production methods in wineries is part of the winemakers’ personal value system. In the US, wineries have a fierce commitment to the land. In recent years they have worked hard to communicate their sustainability practices. Winegrowers have incorporated modern and old-school practices addressing sustainability issues to meet the expectations of all stakeholders. Through institutional theory, it has been demonstrated that numerous drivers are influencing family-run wineries to implement environmental and social sustainability practices. However, there is a significant difference between these CSR practices and the need to communicate them between Hungary and the US. There is a significant difference between the communication methods of Hungarian and US wineries. In Hungary, there is little communication of sustainability practices, although there is movement towards more communication about sustainability practices since the introduction of the web and social media. The experience, in general, was that instead of communication, the emphasis is put more on living and working according to the principles of sustainability – whether it is environmental, social, or economic. In the US, the communication of CSR is used as a marketing tool to engage and educate their stakeholders, mainly through social media. Research implications –The implications of our research contribute to the knowledge of corporate social responsibility and how institutional influences affect social and environmental responsibility within family-run wineries by bringing the wine industry, family businesses, and CSR together under the analysis of institutional theory. The findings in this paper should be looked upon by family wineries as a resource related to the current communication of social responsibility in the wine industry. Research limitations – A limitation of this research is the lack of personal meetings with interviewees. Due to the COVID-19 lockdowns, the possibility of conducting the interviews face-to-face was not an option. The researchers were challenged to obtain more personal information about the familial, organizational structure without conducting multiple interviews, building trust and rapport.

Researchers have long argued the importance of shared book reading and its potential impact on future reading growth. With the increasing popularity of e-books and the introduction of interactive iPad books, more and more parents are sharing books with their children in digital format. This may have a direct impact on the nature of the interactions that occur during young children's early book sharing experiences. This multiple-case study examined the nature of the interactions between six mother-child dyads as they read a story on an iPad compared with the interactions that happened during a traditional book reading. A coding system was developed to categorize behaviors during the reading sessions. The categories included (a) meaning talk, (b) text and print talk, (c) technology talk, and (d) the nature of the affective climate.The results of this study indicate that mother-child traditional read-alouds and digital iPad read-alouds are different experiences. During digital readings there was an increase in the number of vocabulary-related (a subcategory of meaning) interactions but a decrease in the number of interactions related to text and print. The results also showed that as the dyad shared a digital storybook, they engaged in many interactions about the technological elements of the iPad texts, which may be important to the development of digital literacies in young children. The findings also indicate that a child is more engaged with digital texts than with traditional texts, which is important because engagement is an essential component in literacy development. However, sometimes the child's increased engagement with the digital texts resulted in both members of the dyad being less sensitive to the other. Digital storybook reading is a relatively new experience for many parents and thus, they may not know how to interact with their children in ways that promote the development of traditional and digital literacies.

Remote work has become a popular topic within organizations and the popular press. However, academic research has been inconclusive as to whether remote work is related to benefits of increased job performance and lessened work-family conflict. This study examined remote work resources to gain an in-depth understanding of how remote work relates to job performance and work-family conflict. One hundred fifty-one salespeople participated in two time-lagged surveys regarding remote work resources (autonomy, feedback, access to information and interaction with one’s supervisor), outcomes (subjective and objective job performance and work family-conflict) and demographics. Remote work resources were not significantly related to job performance. Contrary to hypotheses, more control over work schedule and control over work process were related to more FIW. In support of hypotheses, more interaction with one’s supervisor was related to less FIW and more access to information was related to less WIF. There was no support for hypothesized mediation or moderation but exploratory analyses revealed that proactive personality moderated the relationship between interaction with one’s supervisor and objective job performance such that the relationship was stronger for less proactive employees than for more proactive employees. Overall, findings support the value of fine-grained analysis of remote work’s resources to provide a nuanced look into their relationships with outcomes.

Abstract:
Understanding why persons repeatedly place themselves at risk for Human Immunodeficiency Virus (HIV), amidst the wealth of prevention information available is of profound importance. Presently, scientific research of this phenomenon has been dominated by the cognitive models of health behaviour, but these were criticised for ignoring emotional, social and cultural influences on sexual behaviour. This thesis explored and investigated some of these non-cognitive factors within the specific cultural context of St. Vincent and the Grenadines, with sole reference to women, to understand why women put themselves at risk and also help inform the country’s efforts to tackle the problem. This research comprised of four studies, each targeting women ages (18-40 yrs) and sexually involved in relationships. Study one was a qualitative study (N= 10), which explored women’s perceptions of the socio-cultural influences which contributes to their decision to engage in risky sex. Study two was a quantitative study in which (N=75), HIV+ women were surveyed, on whether they contracted HIV from within their long term relationships. Study three was a qualitative Interpretative Phenomenological Analysis (IPA) study (N=9); in-depth interviews investigated the intricacies of long-term relationships that made them more likely to influence unsafe sexual practices. Study four was a quantitative study (N=60) women; used questionnaires to investigate the validity that tolerance to infidelity and non use of condoms in long term relationships, which contributes to HIV transmission amongst Vincentian women. This research confirmed the existing limitations of the Cognitive models on health when applied to sexual behavior and produced evidence that Vincentian women more at vulnerable to contracting HIV within their long term relationships.

Denna uppsats är en litteraturstudie av Roots: The Saga of an American Family. Studien syftar till att undersöka argumentet att rasism har en långvarig effekt på de människor som utsätts för rasism. I detta fall, karaktärerna i romanen som utsätts för rasismens olika hemskheter så som slaveri. Genom användningen av Robert Jensen, WEB Du Bois och Edward Saids teorier (dvs. Privileged/unprivileged, The Veil, Double Consciousness och Others) har romanen Roots analyserats. Slutsatsen visar på hur slaveriet påverkade huvudpersonerna och hur effekten av slaveri lyckats beröva de svarta människorna i romanen deras ursprungliga identitet. Slaveriet ersattes istället av en kollektiv identitet av mindre värde än vita människors identitet vilket lämnat långvariga psykiska ärr för karaktärerna i romanen. Det pedagogiska fokus som används i uppsatsen är den så kallad cooperative learning metoden.

Abstract:
The primary goal of this study was to evaluate the effectiveness of a program being run by the Bedford Police Department’s Repeat Victimization Unit on domestic violence and mental health and mental retardation. The study sought to determine whether the program was effective in reducing instances of repeat victimization in domestic violence and MHMR victims. Additionally the program investigated whether or not the program was effective at reducing victimization severity, and which demographic could be identified as the most victimized. Participants consisted of 157 domestic violence and MHMR victims in the city of Bedford, Tx between November 11, 2012 to July 30, 2013. Findings indicate that levels of repeat victimization for domestic violence and MHMR are relatively low regardless of whether the victim received services through the repeat victimization program or not. Additionally the severity of these repeat victimizations remains relatively constant regardless of whether services were received through the program or not. Implications and findings are discussed.

На основі аналізу ознак інноваційного тренду family look було розроблено систему основних принципів об’єднання виробів в даному стилі і визначені базові показники художньо-композиційних рішень моделей промислової колекції. Встановлено перелік основних вимог до виробів в порядку їх вагомості. Сформовано технічне завдання на розробку промислової колекції У другому розділі виконане ескізне проектування моделей-ідей та моделей-пропозицій. Виконана специфікація деталей і визначена оцінка ступеня уніфікації деталей. За методикою ЦОТШЛ побудовано кресленики базової конструкції і виконано конструктивне моделювання моделей-пропозицій. Розроблені схеми градації основних лекал суконь та «Технічний опис» на базову модель. У третьому розділі обрано раціональні методи оброки основних вузлів та обґрунтовано технологію їх виготовлення. Результативність економічної ефективності проєктних рішень підтверджено розрахунками техніко-економічних показників якості майбутнього виробу. Для виготовлення суконь промислової колекції підібрано пакет матеріалів.

Abstract:
This thesis describes the characterisation of the PQ-loop repeat family, a novel family of membrane proteins from Arabidopsis thaliana. The family consists of six members (AtPQL1-6) all predicted to contain seven transmembrane segments and two copies of the PQ-loop domain. The repeated PQ-loop domain is a well conserved feature present in several eukaryotic proteins. However, the functional significance of the domain remains to be determined. Although most PQ-loop repeat proteins are of unknown function some have been characterised from non-plant species. Those studied include CTNS - a lysosomal cystine transporter from humans, STM1 - a putative G-protein coupled receptor from Schizosaccharomyces pombe, ERS1 - a Saccharomyces cerevisiae protein isolated as a suppressor of a trafficking mutant and LEC35/MPDU1 - an endoplasmic reticulum protein from mammals involved in the utilisation of dolichol monosaccharides during glycosylation. The AtPQL family can be divided into three groups based on similarity with their non-plant homologues. Detailed sequence analysis was carried out to investigate conserved motifs, topology, domain organisation and genome distribution of the six genes. The implications of these findings for putative functions are discussed. AtPQL gene expression was investigated using promoter::GUS fusions and quantitative PCR. Whereas AtPQL4 and AtPQL6 appear to be expressed ubiquitously, AtPQL1, AtPQL2 and AtPQL5 are all preferentially expressed in floral tissues and AtPQL3 is expressed primarily in roots. Several family members showed rhythmic expression over the course of a day and induction by both viral infection and wounding. Localisation at the sub-cellular level was investigated using green fluorescent protein fusions and confocal microscopy. At least four AtPQL proteins are targeted to intracellular compartments. AtPQL1 and AtPQL5-GFP fusions both highlight the tonoplast whereas AtPQL4 and AtPQL6-GFP appear to highlight the ER. A number of knockout and over-expresser mutants were isolated for each gene. Although no physiological phenotype could be found, mutants with altered expression of AtPQL4 and AtPQL6, the two closest A. thaliana homologues of MPDU1/LEC35, showed a clear molecular phenotype. These mutants displayed differential regulation of a number of genes encoding N-glycoproteins as well as defence, cell-surface, lipid biosynthesis and ER stress associated proteins. Abstract:
In this thesis, we propose a family of concurrency control protocols for high data contention database environments. The first one is called the Prudent-Precedence Concurrency Control (PPCC) protocol. It is prudently more aggressive in permitting more serializable schedules than two-phase locking and maintains a restricted precedence among conflicting transactions and commits the transactions according to the serialization order established in the executions. The second one is a family of hierarchical concurrency control protocols called the Hierarchical Precedence Concurrency Control (HPCC) protocols. It maintains cycle-free precedence hierarchies for conflicting transactions. Conflicting operations are allowed to proceed only if the hierarchical orderings of precedence is not violated. Transactions also commit based on the serialization order established during the executions. Detailed simulation models have been implemented for all these protocols and extensive experiments have been conducted to evaluate the performance of the proposed approaches. The results demonstrate that the proposed algorithms outperform the two-phase locking and optimistic concurrency control over a wide range of system workloads.

Other than mobile hydraulics and high voltage switchgears, Bucher Hydraulics is also involved in the less-known area of hydraulic lifts. In fact, Bucher Hydraulics did invent the electronically controlled lift valve in the 1970s. Since then, Bucher Hydraulics developed a wide line of products for hydraulic elevators, such as valves and power units. In 2012, this valve family included various sizes, pressure ranges, systems with constant motor speeds, inverter-driven motors, energy-efficient solutions with hydraulic counterweight, as well as customized solutions. As the common principle, all these solutions apply an electronic closed-loop control that uses a volumetric flow sensor and a proportional actuator. Since 2012, Bucher Hydraulics is substituting this valve family with a new generation, the iValve. Every iValve uses several self-teaching algorithms to adapt to its environment. Their on-board and cabinet electronics control solenoid currents and measure flow, pressure, and temperature. These features enable the iValve to self-monitor, to adapt to operating parameters, and to analyze and log information about itself and the attached system. This report on a highly specialized product is meant to provide inspiring insights.

Le transport de composés à travers les membranes biologiques est crucial pour la physiologie des cellules eucaryotes. Cependant la fonction de nombreux transporteurs putatifs reste inconnue. C’est notamment le cas de nombreux transporteurs intracellulaires exportant les catabolites du lysosome. Le transporteur lysosomal de cystine, baptisé cystinosine, se caractérise par la présence d’un motif dupliqué appelé " boucle PQ ". Sa dysfonction entraîne une maladie lysosomale, la cystinose, caractérisée par l'accumulation de cystine dans les lysosomes. Les protéines possédant un motif PQ sont retrouvées plus souvent dans les cellules eucaryotes et, à l'exception de la cystinosine, leur fonction reste inconnue. Dans cette thèse, nous démontrons qu'une autre protéine à motif PQ, PQLC2 est le transporteur responsable de l'efflux lysosomal des acides aminés cationiques et qu'il est impliqué dans le traitement de la cystinose.L'hypothèse de départ était basée, d'une part, par sur des prédictions par analyse protéomique de la localisation lysosomale de PQLC2 et, d'autre part, sur des résultats chez S.cerevisiae impliquant les orthologues putatifs de PQLC2, situés à la membrane de la vacuole, dans l'homéostasie des acides aminés cationiques. En utilisant une approche consistant à délocaliser PQLC2 à la membrane plasmique et à acidifier le pH extracellulaire pour mimer la lumière acide du lysosome, nous avons pu, par mesure d'accumulation intracellulaire de composés radiomarqués et par mesure électrophysiologique sur cellule entière, faire la preuve du transport sélectif, actif à bas pH et de faible affinité des acides aminés cationiques par PQLC2. Dans une seconde partie, nous avons mis en évidence l'implication de ce transporteur dans l'efflux lysosomal du produit de réaction entre la cystine accumulée dans les lysosomes de cellules de patients cystinotiques et le principe actif (cystéamine) du traitement pharmacologique de la cystinose.Enfin, dans une dernière partie, nous avons effectué une analyse bioinformatique préliminaire des protéines à motif PQ qui exploitait la pseudo-symétrie de ces protéines pour identifier des résidus potentiellement impliqués dans l'activité de transport
Transport of solutes across biological membranes is crucial to eukaryotic cell physiology. However, the function of many putative transporters remains unknown, such as the proteins responsible for lysosomal export of metabolites. Cystinosin, the lysosomal cystine exporter defective in cystinosis, is characterized by a duplicated motif termed the PQ loop. PQ-loop proteins are more frequent in eukaryotes than in prokaryotes, and, except for cystinosin, their molecular function remains unknown. Here we show that another PQ-loop protein, PQLC2, is a lysosomal transporter for cationic amino acids and that it is required for the treatment of cystinosis. The hypothesis that PQLC2 is a lysosomal metabolite transporter was based on a proteomic study predicting that PQLC2 is located at the lysosomal membrane and on a genetic study that linked putative yeast orthologues with cationic amino acid homeostasis. Using an approach that consisted in misrouting PQLC2 to the plasma membrane of frog oocytes and in acidifying the extracellular medium to mimic the acidic lysosomal lumen, we showed an accumulation of radiolabelled cationic amino acids into mRNA-injected oocytes and an electrogenic, inward current due to a selective, pH-dependent, low-affinity transport of cationic amino acids by PQLC2. Moreoever, we showed that PQLC2 exports a key chemical intermediate (cysteamine-cysteine mixed disulfide) from cystinotic lysosomes treated with the aminothiol drug cysteamine, thus explaining the mechanism underlying the current drug therapy of cystinosis. Finally, in a last chapter, we performed a preliminary bioinformatic study of the family of PQ-loop proteins that took advantage of the pseudo-symmetric structure of these proteins to identify residues potentially important for the transport activity

Les α-galactosides de la famille du raffinose (RFO) sont, avec le saccharose, les principaux oligosaccharides des légumineuses. Cependant, aucune activité α(1,6)-galactosidase n'est retrouvée au niveau de l'épithélium intestinal humain, les RFO sont donc exclusivement fermentés par les enzymes microbiennes. Ces travaux introduisent une enzyme bifonctionnelle de Ruminococcus gnavus E1, un membre majoritaire du microbiote intestinal humain, présentant une activité α(1,6)-galactosidase/ saccharose kinase (AgaSK). L'analyse de la séquence peptidique montre qu'AgaSK présente deux domaines : un domaine homologue aux α-galactosidases GH36, et un autre contenant un motif de fixation des nucléotides (motif A de Walker). La caractérisation des paramètres biochimiques d'AgaSK met en évidence cette bifonctionnalité puisqu'elle est capable d'hydrolyser les α(1,6)-galactosides solubles, et parallèlement en présence d'ATP de phosphoryler le saccharose spécifiquement sur la position C6 du glucose. La production directe de saccharose-6-phosphate à partir de l'hydrolyse du raffinose constitue une voie métabolique jamais décrite chez les bactéries. L'analyse de chacun des domaines montre que les domaines isolés d'AgaSK sont actifs mais la comparaison de leurs paramètres cinétiques montre qu'il y a des différences entre la protéine entière et les domaines isolés. Α-galactosides are non digestible carbohydrates present in many leguminous plants. Soluble α-galactosides consist of galactose units α(1,6) linked to different carbohydrates. Among these, the raffinose family oligosaccharides (RFO) and sucrose, are the most abundant oligosaccharides found in legumes. However, no α(1,6)galactosidase activity exists in the human intestine mucosa and α-galactosides are exclusively fermented by microbial α(1,6)galactosidases (EC3.2.1.22). Here we introduce a bifunctional enzyme, the α(1,6)galactosidase/sucrose kinase (AgaSK) whose gene is highly transcribed in vivo by Ruminococcus gnavus E1, a major member of human dominant intestinal microbiota. Sequence analysis showed that AgaSK is composed of two domains: one closely related to α-galactosidases from glycoside hydrolase family GH36 and the other containing a nucleotide binding motif (Walker A motif). Its biochemical characterization showed that AgaSK is able to hydrolyze efficiently soluble α-galacosides. Furthermore, AgaSK it is able to bind nucleotide to phosphorylate specifically on the C6 position of glucose sucrose. The production of sucrose-6-P directly from raffinose brings out a glycolytic pathway in bacteria, not described so far. In addition, AgaSK isolated domains are active but the biochemical characterization has shown that there are differences in the activities between the whole protein and isolated domains. The crystal structures of the galactosidase domain in complex with the product shed light onto the reaction and substrate recognition mechanisms and highlight an oligomeric state necessary for efficient substrate binding

Dynamic of settlement and space changes are in the heart of this research. Thus, the studied population, listed from 1860 to 1950 in a town center street of an inner suburb of Lille, revealed distinctive links between family ties and location of residence. For some residents this street is quite a tight space. It is true "centre of the world" for those who are integrated in a wide family using their land inheritance for the exercice of crafts or proximity. At the same time, this street constitutes for others a transitory stopping place, in a slightly polarized migratory space, Lille being increasingly present as soon as the family links become looser. Mon travail de thèse s'articule autour de l'étude d'une protéine-kinase bactérienne d'un nouveau genre, l'HPr Kinase/Phosphorylase (HPrK/P). Cette enzyme bifonctionnelle régule chez les bactéries à Gram positif la répression cataabolique du carbone, qui permet aux bactéries de s'adapter rapidement à la disponibilité en source de carbone. HPrK/P ne présente aucune similarité structurale avec les protéine-kinases eucaryotes. Elle phosphoryle la serine 46 des protéines HPr et Crh, qui activent alors le régulateur transcriptionnel CcpA. La conformation phosphorylase de l'enzyme est connue depuis 2001. Lors de ma thèse, l'étude cristallographique du mutant V267F du domaine catalytique de l'HPrK/P de L. Casei a permis de résoudre sa conformation kinase. L'analyse structurale des interactions entre CcpA et les formes phosphorylées de HPr et Crh a également été réalisée. Enfin, j'ai étudié les protéines Fap7 et YFH7 de S. Cerevisiae dans le cadre d'un projet d'analyse systématique des protéines à P-loop de fonction inconnue chez la levure
My PhD work concerns the study of a new type of bacterial protein-kinase, the HPr Kinase/Phosphorylase (HPrK/P). This bifunctional enzyme is a key sensor of the Carbon catabolite regulation mechanism in low G+C Gram positive bacteria. HPrK/P exhibits no similiarity with eukaryotic protein kinases. HPrK/P phosphorylates residue serine 46 of the HPr and Crh proteins, allowing them to activate the transcriptional regulator CcpA, and to regulate the expression of carbon catabolism genes. This mechanism enables bacteria to adapt to their environment. The structure of the phosphorylase conformation of HPrK/P was known. My work on mutant V267F of L. Casei HPrK/P catalytic domain allowed me to gain insights into the kinase conformation. I also performed structural analysis of the interactions between CcpA and the phosphorylated forms of HPr and Crh. Finally, I analysed the proteins Fap7 and YFH7 of S. Cerevisiae in a project of systematic study of yeast P-loop containing proteins of unknown function

The current study describes the directive behaviors of seven mothers with their toddlers ranging in age from 12 to 35 months throughout the day. This study explores the behaviors of a sample with lower socioeconomic status without the use of unnatural measures or artificial environments that may enhance the likelihood of observing atypical behaviors and perhaps perpetuate a deficit-based interpretation of the poverty context. Nine hours of observation for each dyad were collected as part of a larger study concerning the daily experiences of toddlers with the exception of one participant who dropped out of the study after three hours of observation. The current study analyzed maternal behaviors while the mother was present with her toddler and the toddler was awake. Observations used in the current analysis lasted between 90 to 450 minutes for each participant. The importance of the extended observational protocol used in the current study was specifically investigated by comparing parenting behaviors that occurred during the first 45 minutes of observation to those which occurred during subsequent observational segments. This study also explored a more complete conception of directiveness in a lower socioeconomic context by defining two separate variables for responsive and adultinitiated directiveness. The situational contexts that influence mothers’ directive behaviors were then examined. The results of the current study suggest that when mothers with lower socioeconomic status are observed for an extended amount of time they vary greatly in the amount of directiveness that they use with their children. These directive behaviors occurred at a much higher rate during the first segment of time mothers were observed. Directive behaviors did not cluster as either adult-initiated or responsive as expected. Rather, directive behaviors clustered according to the contexts of caregiving or play interactions. Only three toddlers engaged in any structured activities while in the care of their mothers. Results of the current study challenge the methodology used in previous research that has resulted in the wide spread stereotype of parents with lower socioeconomic status parenting in a harsh and deficient manner. Implications for family functioning assessment and intervention are also discussed.

While the study of volunteer tourism has been growing over the past decade, there is limited literature that takes a global approach, whereby one explores the perspective of a number of stakeholder groups in such experiences. The aim of this project was to fill this gap by looking at the volunteer tourism experience from four perspectives, namely the volunteer, the organization, the host family, and the community at large. The focus in this case was on the experiences and expectations of these groups. The research was qualitative; based primarily upon interviews. The research for this project took place in Ghana. It was found that the three local stakeholders (the organization members, the host family members and people from the broader community) generally viewed the experience similarly. Volunteers often expressed views that set them apart from the other groups. Main themes for the groups generally revolved around cultural differences, communication and progression through stages of the experience. The stages of the experience showed three main stages, those referring to the time before the volunteer, during the volunteer???s stay and the time after the volunteer???s departure. Cultural differences included discussions of culture shock, stereotypes, and mis-communications in cross-cultural dialogue. Communication was a major issue, and was to be found lacking between all groups. In regards to communication there was also a suggestion for continuing contact and relationships between the locals and volunteers beyond the volunteer???s time in the community.

abstract: When my attention was brought to the overwhelming lack of family policy support in the United States, my curiosity led me to look into what other industrialized nations are doing to support growing families and find out what policies and programs have been put in place to better facilitate the work-home balance. I first provide a brief background context of family policy in the United States, leading up to the development and implementation of our nation's parental leave legislation, the Family and Medical Leave Act (FMLA). I present the crucial concerns of this provision, as well as the effects that policy has on children's well-being. The second major part of this analysis deals with child care programs and the myriad challenges so many families encounter in this realm. Specifically addressed are the topics of affordability, accessibility and quality of child care found in the U.S. After an in-depth look at U.S. policies, I transition to a comparative analysis of parental leave and child care provision in a range of other nations in the Organization for Economic Co-operation and Development (OECD), specifically Canada, Australia, the United Kingdom, France, Sweden and Norway. I carefully chose these countries to offer a broad spectrum of family policies to compare to our own. I then return to a discussion of limitations of U.S. family policy and the values and ideology it represents, as well as the importance of strengthening such policies.

This study focus on how the couples overcome the pressure from the long-team essentials in the process of raising their Asperger children. Moreover, it emphasizes not only how to maintain a good co-parenting relationship during the process, but also how the couples use the resources when they are under stress. In addition, the study talks about how these couples adapt themselves as husband and wife with appropriate and cognitive behaviors in the process of parenting. This study analyzes cases based on the interviews with three couples who have gone through 18 years of Asperger parenting stress while maintaining good marital relationships successfully. The results were summarized as follows: 1. Couples with Asperger children would face different pressures as their children grow up. The sequence of the pressures is in the following order: confusion in the beginning of the stage, the difficulty in parenting care, school acceptances, the misunderstanding from the society and the worry of their children’s future. 2. As a matter of fact, those three couples relied on the professional information, and support each other emotionally. It is worth mentioning that two of the couples used their personal inner resources to build a supporting system in order to help other families that were in a similar situation. In return, the system supported and helped the couples themselves as well. 3. In terms of resolving problems, these 3 couples tend to educate their Asperger children with attitudes that are consisted of educational philosophy, positive thoughts and rational perspective. For the education of Asperger children, instead of putting more pressure on the parents, which causes problem in their relationship, it actually leads the parents to opportunities on facing the issues in their relationship especially when the parents are going through marriage crisis. 4. Children are considered as the top priority for these couples in terms of co-parenting interaction. In general, the wives understand how to treat their husbands and children with the use of homeopathic-roundabout strategies and patience. On the other hand, husbands usually treat their wives with respect, trust, tolerance and acceptance. In addition, these couples support each other financially and spiritually without falling into the traditional roles and rules of genders in their relationships.

This thesis examines the discourses on the promotion of the Lippes Loop and the experiences of the women users in Taiwan’s Family Planning Program in the period between 1960s and 1980s. The materials include oral history interviews with 10 Lippes Loop users, 4 other IUD users, 1 gynecologist, 1 midwife, 1 family planning worker, and other historical documents. By analyzing several relevant groups for Lippes Loop, including the government, the medical community, and women users, I found that the government used goal-oriented strategy to promote the use of Lippes Loop. In addition, the medical community, with its professional and cultural authority, played an important role in promoting IUDs in public, and individual physicians were instrumental in installing IUDs in their medical practice. However, by examining various medical journals it appears that members of the medical community did not share the same opinions regarding Lippes Loop. Finally and most importantly, the choices that the users made were affected by the historical circumstances. It is only through revealing the experiences of women users, we may see the other side of the Taiwan Family Planning Program. Taiwan’s Family Planning Program has been much acclaimed; it is often described as a miracle. Yet, little attention has been paid to the gender politics involved in the implantation of the so-called miraculous program. Women were treated much like experimental subjects, and their reported reactions to the device were often ignored as over-sensitiveness; the physicians often condescended on women’s ways of dealing with the Loop. Only when one looks into these historical circumstances, including individual women’s condition, their notions of the body and their tacit knowledge, does one begin to understand their lived body experiences.

Abstract:
The resurgence of Texas' snook (Family Centropomidae) recreational fishery is coupled with an uncertainty as to what species occur in State waters, a limited understanding of life history, and habitat needs of its constituents. This study described species composition and early life history aspects of juvenile (< 100 mm SL) centropomids taken in bag seine collections in estuarine and freshwater habitats along the upper, middle, and lower coast of Texas during 2006 to 2010. Centropomus specimens (n = 548) captured from 41 locations across the Texas coast as well as congeners from Mexico (n = 24), Florida (n = 7), and Costa Rica (n = 3) were used in a genetics- and meristic-based determination of species composition, growth rates, range of hatching dates, geographic distribution, and habitat association. Genetic analyses of the mitochondrial DNA 16s ribosomal RNA gene and the mitochondrial control region (D - loop) validated the presence of smallscale fat snook (C. parallelus Poey, 1860, n = 333), common snook (C. undecimalis Bloch 1792, n = 212) and Mexican snook (C. poeyi Chavez, 1961, n = 3) in Texas, with the last of these validations representing the first known record of this species in Texas. AMOVA of 16s and D - loop sequences failed to detect genetic differentiation within Texas for C. parallelus and C. undecimalis. However, AMOVA for 16s and D - loop C. undecimalis sequences did yield significant genetic differences between Texas and Mexico against those from Florida and Costa Rica. Juvenile centropomids (< 100 mm SL) in Texas occupied backwater habitats with dissipated currents similar to those of Florida congeners (tidal sloughs, freshwater habitats, and structured shorelines). Coastal ranges of these species differed with C. parallelus taken from the Rio Grande to West Galveston Bay, whereas C. undecimalis was captured from the Rio Grande northward near Palacios. Three C. poeyi were captured at only two locations (Laguna Vista and Port Aransas). Daily growth rates varied between species and capture years, with these ranging from 0.22 to 0.97 mm d^-1. Analyses of hatch-date distribution suggest centropomids in Texas begin spawning in August and continue it through late September into mid-November.
