Abstract:
Human oral squamous cell carcinomas (OSCCs) have high cancer mortality and a 5-year survival rate lower than that of most other carcinomas. New therapeutic strategies are required for the treatment and prevention against OSCCs. An approach to cancer therapy using plant-derived natural compounds has been actively in progress as a trend. Falcarindiol (FALC), or its isolated form Ostericum koreanum Kitagawa (O. koreanum), is present in many food and dietary plants, especially in carrots, and this compound has a variety of beneficial effects. However, biological activity of FALC has not been reported in OSCCs yet. This study aimed to demonstrate the antitumor effects of FALC against OSCCs, YD-10B cells. In this study, FALC was selected as a result of screening for compounds isolated from various natural products in YD-10B cells. FALC suppressed cell growth, and FALC-induced apoptotic cell death was mainly accompanied by the dephosphorylation of PI3K, AKT, mTOR, and p70S6K. The apoptotic cell death was also associated with autophagy as evidenced by the expression of Beclin-1, the conversion of LC3-II, and the formation of autophagosome. FALC-induced autophagy was accompanied by MAPKs including ERK1/2 and p38. Furthermore, FALC caused the antimetastatic effects by inhibiting the migration and invasion of YD-10B cells. Taken together, the findings suggest the potential value of FALC as a novel candidate for therapeutic strategy against OSCCs.

Abstract:
Fiber reinforced aerated lightweight concrete (FALC) was developed to reduce concrete's density and to improve its fire resistance, thermal conductivity, and energy absorption. Compression tests were performed to determine basic properties of FALC. The primary independent variables were the types and volume fraction of fibers, and the amount of air in the concrete. Polypropylene and carbon fibers were investigated at 0, 1, 2, 3, and 4% volume ratios. The lightweight aggregate used was made of expanded clay. A self-compaction agent was used to reduce the water-cement ratio and keep good workability. A surfactant was also added to introduce air into the concrete. This study provides basic information regarding the mechanical properties of FALC and compares FALC with fiber reinforced lightweight concrete. The properties investigated include the unit weight, uniaxial compressive strength, modulus of elasticity, and toughness index. Based on the properties, a stress-strain prediction model was proposed. It was demonstrated that the proposed model accurately predicts the stress-strain behavior of FALC.

Abstract:
Abstract Innate immune responses are important in combating various microbes during the early phases of infection. NK cells are innate lymphocytes that, unlike T and B lymphocytes, do not express antigen receptors but rapidly exhibit cytotoxic activities against virus-infected cells and produce various cytokines. Here we report a new type of innate lymphocyte present in a novel lymphoid structure associated with adipose tissues in the peritoneal cavity. These cells do not express lineage (Lin) markers but do express c-Kit, Sca-1, IL7R and IL33R. Similar lymphoid clusters were found in both human and mouse mesentery and we term this tissue ‘FALC’ (fat-associated lymphoid cluster). FALC Lin-c-Kit+Sca-1+ cells are distinct from lymphoid progenitors and lymphoid tissue inducer cells. These cells proliferate in response to IL2 and produce large amounts of Th2 cytokines such as IL5, IL6 and IL13. IL5 and IL6 regulate B-cell antibody production and self-renewal of B1 cells. Indeed, FALC Lin-c-Kit+Sca-1+ cells support the self-renewal of B1 cells and enhance IgA production. After helminth infection and in response to IL33, FALC Lin-c-Kit+Sca-1+ cells produce large amounts of IL13, which leads to goblet cell hyperplasia—a critical step for helminth expulsion. In mice devoid of FALC Lin-c-Kit+Sca-1+ cells, such goblet cell hyperplasia was not induced. Thus, FALC Lin-c-Kit+Sca-1+ cells are Th2-type innate lymphocytes, and we propose that these cells be called ‘natural helper cells’.

Abstract:
The plant Saussurea costus (Falc) Lipsch has many biological activities and a strong curative and preventive power against a variety of diseases including cancer, diabetes, and hemorrhoids. In the current study, phytochemical screening was carried out as well as an investigation of the antilithiatic and antioxidant activities of aqueous and ethanolic extracts of this plant. The results showed that aqueous and ethanolic extracts were effective in reducing cystine stone mass and that the aqueous extract of Saussurea costus (Falc) Lipsch had the highest percentage of dissolution (6.756 ± 1.024) (p < 0.05). A turbidimetric method and a crystallization test were used to evaluate the antilithiatic activity of an aqueous and ethanolic extract of this plant on calcium oxalate crystallization. The results of these methods revealed that the ethanolic extract of this plant has a significant inhibitory effect on calcium oxalate crystallization, with a percentage inhibition of (91.017 ± 0.299) (p < 0.05) for a concentration of 2 mg mL−1. The DPPH method revealed that the ethanolic extract of Saussurea costus (Falc) Lipsch with a concentration of (IC50 = 0.12325 mg mL−1) had the highest IC50, whereas the FRAP method revealed that the aqueous extract of Saussurea costus (Falc) Lipsch with a concentration of 300 µg mL−1 has the most significant reducing power with (OD = 0.56 ± 0.05). These findings indicate that aqueous and ethanolic extracts of Saussurea costus (Falc) Lipsch had a significant effect on whewellite and weddellite and a greater free radical scavenging effect but had no effect on cystine dissolution.

Abstract:
Abstract Fibroblastic reticular cells (FRCs), a subpopulation of stromal cells in lymphoid organs and fat-associated lymphoid clusters (FALCs) in peritoneal fat, play immune-regulatory roles in the host defense. However, the mechanisms underlying the regulation of FRC pathobiology, especially in response to Toll-like receptor (TLR) signaling is unclear. Therefore, we aimed to understand the roles of TLR signaling in FRC pathobiology during sepsis. FALC cells were isolated from mesenteric adipose tissue in wild-type mice before and after cecal ligation and puncture (CLP). The transcriptomic profile of FALC cells were assessed using single cell RNA sequencing. Notably, FRCs were a major population of FALC cells (50 +/− 10.2 % in control, 30 +/− 8.6% in CLP). One population of FRCs were identified in control mice, and two populations of FRCs were identified in CLP mice. According to gene set enrichment analysis, FRCs after CLP are enriched in gene expression associated with inflammatory response and chemotaxis. Interestingly, activation of TLR4 (LPS) and TLR3 (Poly I:C) signaling in mesenteric adipose tissue derived FRCs significantly increased the expression of chemokines known to attract neutrophils (Cxcl2 and Cxcl5), monocytes (Cxcl3), and lymphocytes (Cxcl13 and Ccl20), while activation of TLR9 (ODN1518) significantly decreased these chemokine gene expressions. Importantly, we found similar regulatory effects of TLR3, 4, and 9 on chemokine production in human adipose tissue-derived FRCs. Together, our results indicate that TLRs plays critical roles in regulating chemokine production by FRCs. These data form a knowledge basis upon which to design new therapeutic strategies of FRC-based treatments for sepsis.

Abstract:
Abstract Fibroblastic reticular cells (FRC) are highly heterogeneous. Distinct FRC subsets play unique roles in the formation of secondary lymphoid organs and immune responses. We have previously shown that TLR9 signaling plays a critical role in regulating peritoneal immunity via suppressing chemokine production in FRC in the fat associated lymphoid clusters (FALC). However, the subset-specific roles of TLR9 in FRC of FALCs remain unknown. Using single-cell RNA-sequencing, we identified three distinct subsets of FRC (CD55hi, CD9hi and CD55loCD9lo) in mouse mesenteric FALCs at baseline. Based on the gene set enrichment analysis, the Cd55hi subset was enriched in gene expression related to cell differentiation. The CD9hi subset was enriched in gene expression related to immune response. The CD55loCD9lo subset was enriched in the gene expression related to extracellular matrix formation. Furthermore, we found that CD9hi FRC from Tlr9−/− mice increased gene expression associated with inflammation. Using flow cytometry and bulk-RNA seq, we successfully isolated and validated these three subsets in mouse FALCs. Interestingly, activation of TLR9 signaling using ODN1585 significantly decreased CD9 expression in FRC in vivo and in vitro. Furthermore, treatment of ODN1585 suppressed the proliferation of CD9hi FRC, evidenced by decreased expression of Ki67. These results indicate that CD9hi FRC in FALC are immunoregulatory. TLR9 signaling may modulate peritoneal immunity via regulation of the proliferation of CD9hi FRC. Understanding the mechanism of how TLR9 regulates CD9hi FRC may lead to discovering new therapies for inflammatory diseases.

Abstract:

______________________________________________________________________________ ABSTRACT
Consciously that racism and sexism operates dehumanizing black lives and establishes social relations for a society that has slavery, phallocentric practices and misogyny in its history, the purpose of the present study is to present an analysis of black women feminist theories. This theoretical apparatus provides us with experience on the application of theories on black feminism, interseccionality, womanization and transgressive education as a requirement to social empowerment focusing on gender and race. Education, as a powerful social field is also an area in which several types of violence are inflicted towards minorities and also sets on the exclusion process; In order to face those conflicts from a different perspective, as those proposed on this paper it is indispensable to suggest that the theoretical scope can be an aid, to avoid universalistic social policies, capable of substantiating a solid work towards Black women in their empowerment process through education.

SUBBURAMAN, RAJESH. "Fall Prediction and Controlled Fall for Humanoid Robots." Doctoral thesis, Università degli studi di Genova, 2019. http://hdl.handle.net/11567/939791.

Full text
Abstract:
Humanoids which resemble humans in their body structure and degrees of freedom are anticipated to work like them within infrastructures and environments constructed for humans. In such scenarios, even humans who have exceptional manipulation, balancing, and locomotion skills are vulnerable to fall, humanoids being their approximate imitators are no exception to this. Furthermore, their high center of gravity position in relation to their small support polygon makes them more prone to fall, unlike other robots such as quadrupeds. The consequences of these falls are so devastating that it can instantly annihilate both the robot and its surroundings. This has become one of the major stumbling blocks which humanoids have to overcome to operate in real environments. As a result, in this thesis, we have strived to address the imminent fall over of humanoids by developing different control techniques. The fall over problem as such can be divided into three subissues: fall prediction, controlled fall, and its recovery. In the presented work, the first two issues have been addressed, and they are presented in three parts. First, we define what is fall over for humanoids, different sources for it to happen, the effect fall over has both on the robot and to its surroundings, and how to deal with them. Following which, we give a brief introduction to the overall system which includes both the hardware and software components which have been used throughout the work for varied purposes. Second, the first sub-issue is addressed by proposing a generic method to predict the falling over of humanoid robots in a reliable, robust, and agile manner across various terrains, and also amidst arbitrary disturbances. The aforementioned characteristics are strived to attain by proposing a prediction principle inspired by the human balance sensory systems. Accordingly, the fusion of multiple sensors such as inertial measurement unit and gyroscope (IMU), foot pressure sensor (FPS), joint encoders, and stereo vision sensor, which are equivalent to the human’s vestibular, proprioception, and vision systems are considered. We first define a set of feature-based fall indicator variables (FIVs) from the different sensors, and the thresholds for those FIVs are extracted analytically for four major disturbance scenarios. Further, an online threshold interpolation technique and an impulse adaptive counter limit are proposed to manage more generic disturbances. For the generalized prediction process, both the instantaneous and cumulative sum of each FIVs are normalized, and a suitable value is set as the critical limit to predict the fall over. To determine the best combination and the usefulness of multiple sensors, the prediction performance is evaluated on four different types of terrains, in three unique combinations: first, each feature individually with their respective FIVs; second, an intuitive performance based (PF); and finally, Kalman filter based (KF) techniques, which involve the usage of multiple features. For PF and KF techniques, prediction performance evaluations are carried out with and without adding noise. Overall, it is reported that KF performs better than PF and individual sensor features under different conditions. Also, the method’s ability to predict fall overs during the robot’s simple dynamic motion is also tested and verified through simulations. Experimental verification of the proposed prediction method on flat and uneven terrains was carried out with the WALK-MAN humanoid robot. Finally, in reference to the second sub-issue, i.e., the controlled fall, we propose two novel fall control techniques based on energy concepts, which can be applied online to mitigate the impact forces incurred during the falling over of humanoids. Both the techniques are inspired by the break-fall motions, in particular, Ukemi motion practiced by martial arts people. The first technique reduces the total energy using a nonlinear control tool, called energy shaping (ES) and further distributes the reduced energy over multiple contacts by means of energy distribution polygons (EDP). We also include an effective orientation control to safeguard the end-effectors in the event of ground impacts. The performance of the proposed method is numerically evaluated by dynamic simulations under the sudden falling over scenario of the humanoid robot for both lateral and sagittal falls. The effectiveness of the proposed ES and EDP concepts are verified by diverse comparative simulations regarding total energy, distribution, and impact forces. Following the first technique, we proposed another controller to generate an online rolling over motion based on the hypothesis that multi-contact motions can reduce the impact forces even further. To generate efficient rolling motion, critical parameters are defined by the insights drawn from a study on rolling, which are contact positions and attack angles. In addition, energy-injection velocity is proposed as an auxiliary rolling parameter to ensure sequential multiple contacts in rolling. An online rolling controller is synthesized to compute the optimal values of the rolling parameters. The first two parameters are to construct a polyhedron, by selecting suitable contacts around the humanoid’s body. This polyhedron distributes the energy gradually across multiple contacts, thus called energy distribution polyhedron. The last parameter is to inject some additional energy into the system during the fall, to overcome energy drought and tip over successive contacts. The proposed controller, incorporated with energy injection, minimization, and distribution techniques result in a rolling like motion and significantly reduces the impact forces, and it is verified in numerical experiments with a segmented planar robot and a full humanoid model.

The 22q11.2 deletion syndrome (del22q11.2) is a genetic condition caused by a microdeletion in the chromosome 22q11.2 region. The performance of individuals with this condition at the receptive level of spoken language when compared to that at the expressive level is still controversial. Some studies show superiority in language production with regard to reception, while others suggest that both levels are compromised. A study on speech fluency in narrative tasks could contribute to a better characterization of the expressive level of language in these individuals. The purpose of this study was, therefore, to investigate the speech fluency profile in del22q11.2 syndrome individuals through the characterization, identification, localization and comparison of their disfluencies and speech rate with those of individuals with typical language development. In this study, 15 subjects, male and female, between 7 and 17 years of age, and diagnosed with del22q11.2 syndrome (Experimental Group – EG) were compared with 15 age and gender-matched control subjects with typical language development (Control Group – CG). In order to characterize the subjects, the Wechsler Intelligence Scale, the Peabody Picture Vocabulary Test (PPVT), and Teste de Desempenho Escolar (TDE) were used. The speech fluency profile was investigated using an oral narrative sample elicited by the book Frog, Where Are You? Each oral narrative was transcribed and analyzed according to the Teste de Linguagem Infantil ABFW, which included the type and frequency of normal and stuttered disfluencies and the speech rate (words and syllables per minute). The total number of normal, stuttered and overall disfluencies was obtained based on the frequency of occurrence of disfluencies in the narrative. These data were used to calculate the percentage to match the data and compare the groups... (Complete abstract click electronic access below)

Abstract:
In November 2016, Colombia’s five decades of armed conflict, which had claimed hundreds of thousands of victims, ended in a peace agreement between the government of President Juan Manuel Santos and the FARC guerrilla movement. This analysis examines the progress made in the five main points of the accord: comprehensive rural development reform, political transition, the process of demobilization and social inclusion, the eradication of illicit crop production, and the reform of the justice system. Although violence has decreased overall, and tens of thousands of families have been provided with legal ways of making a living as well as access to land and basic services, the results fall short of the stated goals, in which the changes in government and public policy that have taken place over the years have played a significant role.
