Abstract:
An effort has been made to integrate insights on charge-based interactions in immune complex glomerulonephritis with nuclear antigen involvement in lupus nephritis. Attention was focussed on the histones, a group of highly cationic nuclear constituents, which could be expected to bind to fixed anionic sites present in the glomerular basement membrane (GBM). We demonstrated that all histone subfractions, prepared according to Johns (4), have a high affinity for GBM and the basement membrane of peritubular capillaries. Tissue uptake of 125I-labeled histones was measured by injecting 200 micrograms of each fraction into the left kidney via the aorta and measuring organ uptake after 15 min. In glomeruli isolated from the left kidneys, the following quantities of histones were found: f1, 13 micrograms; f2a (f2al + f2a2), 17 micrograms; f2b, 17 micrograms; and f3, 32 micrograms. Kinetic studies of glomerular binding showed that f1 disappeared much more rapidly than f2a. The high affinity of histones (pI between 10.5 and 11.0; mol wt 10,000-22,000) for the GBM correlates well with their ability to form aggregates (mol wt greater than 100,000) for comparison lysozyme (pI 11, mol wt 14,000), which does not aggregate spontaneously bound poorly (0.4 micrograms in isolated glomeruli). The quantity of histones and lysozyme found in the isolated glomeruli paralleled their in vitro affinity for a Heparin-Sepharose column (gradient elution studies). This gel matrix contains the sulfated, highly anionic polysaccharide heparin, which is similar to the negatively charged heparan sulfate present in the GBM. Lysozyme eluted with 0.15 M NaCl, f1 with 1 M NaCl, and f2a, f2b, and f3 could not be fully desorbed even with 2 M NaCl; 6 M guanidine-HCl was necessary. Two further findings of great relevance for the concept of induction of immune complex glomerulonephritis by histones were: (a) glomerular-bound histone was accessible for specific antibody given intravenously; and (b) prior binding of histones promoted glomerular deposition of anionic antigens, as could be shown with ssDNA fragments. These data justify the proposal that glomerular deposition of histones can induce immune complex formation, start an inflammatory process, and produce tissue damage.

Abstract:
The gravity map of Ouaddai in the eastern region of Chad exhibits important anomalies that are often delimitated by the high gravity gradients, resulting from the density contrasts between various anomaly sources. This presumes an important tectonic activity in zone. Because of its arid and desert nature, the region is water-poor or even lacking water resources. The main source of water in the region is made of very deep aquifers. The gravity analysis from this study helps to better understand the network of faults in the area. Many complementary approaches of the gravity data processing have been applied, namely the horizontal gradient coupled to the upward and downward continuation and the Euler method. Results from the data filtring have allowed highlighting faults network in various directions (SSW-NNE S-N E-W, NE-SW) and the main direction. In total, 37 major faults were detected with various lengths including 11 (F6, F9, F10, F12, F17, F29, F31, F34, F35, F37) for a cumulative total length of 353 km oriented towards Bongor basin and 15 others (F1, F4, F7, F8, F11, F13, F18, F20, F23, F25, F26, F27, F28, F30, F33), for a cumulative total length of 621 km, oriented towards Doba basin. These faults are of paramount importance with great potential impacts on the regions hydrocarbon reservoirs. Thise results in one hand confirmed some known faults from the previous investigations. In the other hand, the study helps to identify other new and unknown tectonic signatures.

Emulgel is one of the topical dosage which dermatologically has several beneficial properties, namely thixotropic, not oily, easy to spread, easy to clean, soft, easy to wash, long lasting, transparent and comfortable when used. The purpose of this research was to formulate moringa (Moringa oleifera Lam) leaves extract into emulgel dosage forms and determine the antioxidant activity of the dosage using DPPH method.The research began with extraction of moringa leaves and optimization of the base by varying the concentration of carbopol 940 as gelling consisting of F1 0.5%, F2 1%, F3 1.5% and F4 2%. The base that met the requirements of good physical stability was F2. The F2 base was then made into emulgel dosage with 3 concentration variations of the extract, namely F2a 4%, F2b 5% and F2c 6%. The physical stability test result of each formula met the organoleptic test,the pH test, the dispersion test, the adhesion test, the viscosity test, and the freeze-thaw test. The One way ANOVA statistical test result showed that the p value was greater than 0.05, which meant that the emulgel dosage had good physical stability. The IC50values of each antioxidant activity result were F2a (t0 = 120.464 g/mL; t28 = 144.887 g/mL), F2b (t0 = 113.642 g/mL; t28 = 128.407g/mL), F2c (t0 = 74.745 g/mL; t28 = 90.618 g/mL). The statistical results of the t-test showed thep value = 0,027, (0.05), This indicated that there were significant difference results of the antioxidant activity test between the three formulas on the first day (t0) and on the 28th day (t28).

Analysis of the respiratory syncytial virus (RSV) fusion (F) protein in RSV-infected Vero cells showed the presence of a single F1 subunit and at least two different forms of the F2 subunit, designated F2a (21 kDa) and F2b (16 kDa), which were collectively referred to as [F2]a/b. Enzymatic deglycosylation of [F2]a/b produced a single 10 kDa product suggesting that [F2]a/b arises from differences in the glycosylation pattern of F2a and F2b. The detection of [F2]a/b was dependent upon the post-translational cleavage of the F protein by furin, since its appearance was prevented in RSV-infected Vero cells treated with the furin inhibitor dec-RVKR-cmk. Analysis by protein cross-linking revealed that the F1 subunit interacted with [F2]a/b, via disulphide bonding, to produce equivalent F protein trimers, which were expressed on the surface of infected cells. Collectively, these data show that multiple F protein species are expressed in RSV-infected cells.

Many biomedical applications absolutely require, or are substantially enhanced by, coexpression of multiple proteins from a single vector. Foot-and-mouth disease virus 2A (F2A) and “2A-like” sequences (e.g.,Thosea asignavirus 2A; T2A) are used widely for this purpose since multiple proteins can be coexpressed by linking open reading frames (ORFs) to form a single cistron. The activity of F2A “cleavage” may, however, be compromised by both the use of shorter versions of F2A and the sequences (derived from multiple-purpose cloning sites) used to link F2A to the upstream protein. To characterise these effects, different lengths of F2A and T2A were inserted between green and cherry fluorescent proteins. Mutations were introduced in the linker region immediately upstream of both F2A- and T2A-based constructs and activities determined using both cell-free translation systems and transfected cells. In shorter versions of F2A, activity may be affected by both the C-terminal sequence of the protein upstream and, equally strikingly, the residues immediately upstream introduced during cloning. Mutations significantly improved activity for shorter versions of F2A but could decrease activity in the case of T2A. These data will aid the design of cloning strategies for the co-expression of multiple proteins in biomedical/biotechnological applications.

ObjectiveThis study sought to explore the views and experiences of a group of people with severe mental disorders (SMDs) who performed volunteer services.DesignA qualitative phenomenological study.SettingCommunity public mental health services of the Community of Madrid and the province of Barcelona (Spain).ParticipantsPurposive sampling techniques were used between September 2016 and April 2017. The inclusion criteria were: individuals aged 18–65 years who participated in volunteer activities during the performance of this study, based on the regulations of volunteer services in Spain and the community of Madrid; a diagnosis of non-organic psychotic disorder (F20.x, F21, F22, F24, F25, F28, F29, F31.x, F32.3 and F33) according to the International Classification of Diseases, 10th Revision; an evolution of ≥2 years; and a moderate to severe dysfunction of global functioning with scores ≤70 in the Global Assessment of Functioning Scale. Ultimately, 23 people with SMD participated in the study with a mean age of 47 years (SD 8.23).MethodsData were collected through in-depth interviews and researcher field notes. A thematic analysis was performed following appropriate guidelines for qualitative research.ResultsTwo main themes emerged to describe the experience of participating in volunteer activities: (1) rebuilding self-identity, based on the participant’s experience of volunteering, of acquiring a new role and a new perceived identity that made them feel valued and respected; and (2) being a so-called normal person with a ‘normal’ life, based on recovering a sense of normality, unmarked by the illness, thanks to daily responsibilities and occupations.ConclusionsQualitative research offers insight into the way people with SMD experience volunteering and may help to improve understanding of the underlying motivations that drive these individuals. These findings may be applied to improve guidance during their process of recovery and subsequent inclusion into society.

Various shadowing aspects of nuclear parton distributions are discussed in a parton-model framework. We explain existing x dependent data of F2A /F2D, then the model is applied to a flavour asymmetry – d in a nucleus, to shadowing in valence-quark distributions, and to nuclear dependence of Q2 evolution. First, we find that a finite nuclear ū – đ distribution could be possible due to nuclear interactions even in the flavour symmetric case in the nucleon ( ū – đ = 0). Second, it is indicated that valence-quark shadowing could be used for discriminating among various shadowing models. Third, we find that nuclear dependence of Q2 evolution ∂ [F2Sn/F2C]/∂[ln Q2], which was found by the New Muon Collaboration (NMC), is essentially understood by modification of parton distributions in nuclei. However, higher-twist effects in nuclear interactions could be tested by studying the details of the nuclear Q2 evolution.

Abstract. Material flow analysis and environmental contamination analysis are merged into a flux-flow analysis (F2A) as illustrated for the metal circulation in the Seine River catchment. F2A combines about 30 metal flows in the anthroposphere (14 million people) and/or metal fluxes in the environment (atmosphere, soils, and aquatic system) originating from two dozens of sources. The nature and quality of data is very heterogeneous going from downscaled national economic statistics to upscaled daily environmental surveys. A triple integration is performed: space integration over the catchment (65000 km2), time integration for the 1950–2000 trend analysed at 5 year resolution, and a conceptual integration resulting in two F2A indicators. Despite the various data sources an average metal circulation is established for the 1994–2003 period and illustrated for zinc: (i) metal circulation in the anthroposphere is now two orders of magnitude higher than river outputs, (ii) long term metal storage, and their potential leaks, in soils, wastedumps and structures is also orders of magnitude higher than present river fluxes. Trend analysis is made through two F2A indicators, the per capita excess load at the river outlet and the leakage ratio (excess fluxes/metal demand). From 1950 to 2000, they both show a ten fold improvement of metal recycling while the metal demand has increased by 2.5 to 5 for Cd, Cu, Cr, Pb and Zn, and the population by 50%.

Abstract:
A single dose of monocrotaline, a pyrrolizidine alkaloid, was injected into rats in order to produce 25 (Group I) and 45 (Group II) days later a progressive and so called delayed liver injury. The present study investigated the prostanoid production of Kupffer cells and endothelial cells separated from Monocrotaline and saline (Group III) injected rat livers. Kupffer cells: formation of 6 keto Prostaglandin F1a, the major prostacyclin metabolite, gradually decreased in Groups I vs II (P50.01) and in both Groups I and II vs Controls (P50.01). In addition Prostaglandin F2a showed a significant increase in Groups I and II when compared to Group III, (P50.001), and Thromboxane B2 was present in both Groups of Monocrotaline treated animals, while it was not detectable in the control Group III. Endothelial cells:6ketoProstaglandin F1a decreased in Groups I vs II. This differences was significant when compared, and compared to controls (Group III, P50.001). Prostaglandin E2 was detected only in Groups I and II. Prostaglandin F2a and Thromboxane B2 could not be detected in any Group. Ultramicroscopy showed morphological cell damage in nonparenchymal cells in Monocrotaline intoxication in Group II, rats sacrified 45 days after the injection, while it shows normal features in those treated animals sacrified 25 days after the injection, as well as in control group. Conclusion: Asingle Monocrotaline injection produces, 25 and 45 days later, severe and progressive alterations in the prostanoid production in Kupffer and Endothelial cells, while ultramicroscopic alterations was only observed 45 days after the injection of Monocrotaline. A decreased production of vasodilators and the presence of vasoconstrictor prostanoids that can participate in the production of the circulatory derangements enhancing liver injury and portal hypertension were also observed.

Abstract:
The clinical effectiveness of the implimintation of the Prostaglandin's E2 and F2a for labor preparation, for induction and regulation of the childbirth process was studied. Prepidil gel was applied for cervix preparation for labor by various testimony in 142 patients. The maturing of the cervix was note in 50,3% patients, and the effect of stimulation of labor was reach in 46,5% patients. Prostin E2 and F2a were used in case of PPROM and for treatment of the dystocia in 53 patients. The clinical analysis of the labors was performed. It was discovered, that Prostaglandin's have high effectiveness for cervix preparation in labor, for stimulation of childbirth process and labor induction, for the prevention of long pregnancy.

Abstract:
Objective. The main objective of this study was to select lactic acid bacteria strains with antimicrobial activity and to identify and characterize the antimicrobial compounds. Methods. In this study we tested the antimicrobial activity of 153 lactic bacteria strains by disk diffusion method against 6 microbial pathogenic strains isolated from patients with urinary and vaginal infections. Results. Antimicrobial test results revealed that most of lactic acid bacteria strains exhibited high antimicrobial activity against pathogenic microorganisms. For most of lactic bacteria strains antimicrobial activity has been correlated with the production of organic acids and only for two strains with the biosynthesis of bacteriocins. Bacteriocin produced by Lactococcus (Lc.) lactis F2a strain presented a broad spectrum of activity and high activity (51,200 AU/ml) compared with bacteriocins isolated from Lactobacillus (Lb.) paracasei ssp. paracasei JR strain (400 AU/ml). The stability tests of bacteriocin revealed that the bacteriocin produced by Lc. lactis F2a strain, it is stable at acid pH while exposure for long time to 600C causes a drastic decrease in bacteriocin activity. Conclusions. Lactic bacteria strains showed a high antimicrobial activity against both prokaryotic and eukaryotic pathogen strains. Two bacterial strains have bacteriocins. Bacteriocins isolated from Lc. lactis F2a strain showed a high activity and a broad spectrum of action.

Abstract:
Plasmodium falciparum, mainly distributed in tropical and subtropical regions of the world, has received widespread attention owing to its severity. As a novel protein, P. falciparum surface-related antigen (PfSRA) has the structural and functional characteristics to be considered as a malaria vaccine candidate; however, limited information is available on its immunogenicity. Here, we expressed three fragments of recombinant PfSRA in an Escherichia coli system and further analyzed its immunogenicity. The results showed that rPfSRA-immunized mice produced specific antibodies with high endpoint titers (1:10,000 to 1:5,120,000) and affinity antibodies (i.e., rPfSRA-F1a (97.70%), rPfSRA-F2a (69.62%), and rPfSRA-F3a (91.87%)). In addition, the sera of immunized mice recognized both the native PfSRA and recombinant PfSRA, the rPfSRA antibodies inhibited the invasion of P. falciparum into the erythrocytes, and they were dose-dependent in vitro. This study confirmed PfSRA could be immunogenic, especially the F1a at the conserved region N-terminal and provided further support for it as a vaccine candidate against P.falciparum.

The purpose of this study was to estimate combining ability of 58 sorghum lines previously selected for Aluminum (Al) tolerance. One hundred sixty-five hybrids were evaluated at three levels of Al saturation (0%, 20% and 50%) at the same season. General Combining Ability (GCA) effects were significant for female lines for all three traits. GCA effects for male lines were significant only for plant height. Specific Combining Ability (SCA) effects were significant only for flowering time. The ratio GCA to SCA was greater than the unity, indicating the prevalence of additive effects for the control of Al tolerance. F7, F14, F17, F20, F21, F24, F29, F31, F41, F42, F48, F51, F54 and F55 lines contributed to increase yield, while F29, F48 and F51 also contributed to reduce flowering time. M2 was the best male line since it contributed to increase yield and plant height, and to reduce flowering time.

There is a huge energy demand from offshore renewable energy resources. To maximize the use of various renewable energy sources, a combined floating energy system consisting of different types of energy devices is an ideal option to reduce the levelized cost of energy (LCOE) by sharing the infrastructure of the platform and enhancing the power production capacity. This study proposed a combined concept of energy systems by combing a heave-type wave energy converter (WEC) with a semisubmersible floating wind turbine. In order to investigate the power performance and dynamic response of the combined concept, coupled aero-hydro-servo-elastic analysis was carried out using the open-source code F2A, which is based on the coupling of the FAST and AQWA tools by integrating all the possible environmental loadings (e.g., aerodynamic, hydrodynamic). Numerical results obtained by AQWA are used to verify the accuracy of the coupled model in F2A in predicting dynamic responses of the combined system. The main hydrodynamic characteristics of the combined system under typical operational conditions were examined, and the calculated responses (motions, mooring line tension and produced wave power) are discussed. Additionally, the effect of aerodynamic damping on the dynamic response of the combined system was examined and presented. Moreover, a second fully coupled analysis model was developed, and its response predictions were compared with the predictions of the model developed with F2A in order for the differences of the calculated responses resulted by the different modeling techniques to be discussed and explained. Finally, the survivability of the combined concept has been examined for different possible proposed survival modes.

Nitric oxide (NO) has diverse physiological and pathophysiological effects. The roles of NO in the renal and cardiac dysfunction found in cirrhosis are reviewed. In the kidneys of experimental animals with cirrhosis, several lines of evidence speak in favour of an enhanced production of NO, through the activation of both endothelial constitutive and inducible isoforms of NO synthase. In contrast with the situation in normal animals, inhibition of NO synthesis in rats with cirrhosis improves sodium and water excretion via blood pressure-dependent and -independent mechanisms, which indicates that the renal sodium and water retention of cirrhosis is related to an excess of NO production. The deleterious effect of excessive NO on the kidney may be mediated by peroxynitrite, a potent oxidant that is readily formed whenever superoxide anions and the ·NO radical are produced together. The peroxidation of arachidonic acid by peroxynitrite leads to the formation of F2a-isoprostanes, which are powerful renal vasoconstrictors. F2a-isoprostane levels are correlated with the severity of liver injury during cirrhosis. However, whether peroxynitrite or F2a-isoprostanes are the elusive mediator of the NO-induced renal alterations in cirrhosis remains to be firmly established. NO is also involved in cardiac contractility, probably in the normal heart as well as in disease conditions such as non-cirrhotic and cirrhotic cardiomyopathy. In the latter state, evidence suggests that inducible NO synthase attenuates ventricular contractility, mediated by cGMP. Another gas that transduces its signal through cGMP, carbon monoxide, is also likely to play a role in cirrhotic cardiomyopathy, but the nature of the interaction between NO and carbon monoxide in this syndrome remains unclear.

Participation in military operations causes high incidence of psychogenic disorders, which can be the reason for psychosis. Assessment of the structure, features of formation, manifestation, symptoms of psychotic disorders in combatants will contribute to the development of appropriate measures for their psychoprophylaxis and treatment.Purpose. To study the clinical symptoms of psychotic disorders of combatants.Material and methods. 64 medical histories of combatants, who suffered from psychosis and were treated in psychiatric hospitals, were analyzed. According to the medical records, the following was studied: the structure of psychotic disorders, the symptoms during hospitalization. The obtained data were processed according to the criterion χ2.Results. The analysis of 64 medical cases showed that the majority of combatants, who were treated at psychiatric hospitals, had the following diseases: schizophrenia (F20) – 28 persons, schizotypical disorders (F21) – 4 persons, acute and transient psychotic disorders (F23) – 25 persons, schizoaffective disorders (F25) – 7 persons.In 38 (59.4%) patients, there was the situation when psychiatric disorders began acutely after some trauma, while in the rest of the persons, their mental disorders gradually increased, which excluded their further military service. Thinking disorders, delusional ideas, and inadequate affect predominated in the structure of psychopathological symptoms. The manifestations of delusional disorders and affective reactions were associated with military operations, injuries and fatalities of some combatants (47 combatants (73.4%)). Certain symptoms comorbid to psychotic disorders and advanced post-traumatic stress disorder (PTSD) before the beginning of treatment took place in 52 patients (81.3%).Conclusion. Analysis of the prevalence and structure of psychotic disorders in combatants using the medical records was carried out. The following was studied: nosological spectrum, manifestation of psychosis, detailed comparative analysis of clinical symptoms in the most common psychoses (schizophrenia (F20) and acute and transient psychotic disorders (F23)). Участие в военных операциях вызывает высокую частоту психогенных нарушений, которые в свою очередь могут быть пусковым фактором возникновения психозов. Оценка структуры, особенностей формирования, манифестации, симптоматики и течения психотических расстройств у комбатантов будет способствовать разработке соответствующих мер их психопрофилактики и лечения.Цель работы: исследовать клиническую симптоматику при психотических расстройствах у комбатантов.Материал и методы. Проведен анализ 64 историй болезней комбатантов, перенесших психозы и лечившихся в психиатрических стационарах. По данным медицинской документации были исследованы: структура психотических расстройств, симптоматика при госпитализации. Обработку полученных данных проводили по критерию χ2.Результаты и обсуждение. Результаты анализа 64 историй болезней показали, что у большинства пациентов-комбатантов, которые лечились стационарно в психиатрических учреждениях, были выставлены следующие основные диагнозы: шизофрения (F20) – у 28 пациентов, шизотипические расстройства (F21) – у 4 человек, острые и преходящие психотические расстройства (F23) – у 25, шизоаффективные расстройства (F25) – у 7 пациентов.У 38 (59,4%) комбатантов психотические расстройства начинались остро, после перенесенной психотравмы, тогда как у остальных лиц постепенно усиливались расстройства психики и поведения, исключающие их дальнейшую военную службу. Расстройства мышления, бредовые идеи и неадекватный аффект преобладали в структуре психопатологической симптоматики. В проявлениях бредовых расстройств и аффективных реакций наблюдались переживания, которые были связаны с военными операциями, травмами и смертельными случаями однополчан у 47 комбатантов (73,4%). Коморбидные к психотическим расстройствам отдельные симптомы и развернутая симптоматика ПТСР до начала лечения имели место у 52 пациентов (81,3%).Вывод. Осуществлен анализ распространенности и структуры психотических расстройств у комбатантов с помощью анализа медицинской документации. Исследованы нозологический спектр, манифестация психозов, проведен подробный сравнительный анализ клинической симптоматики при наиболее часто встречающихся психозах – шизофрении (F20), острых и транзиторных психотических расстройствах (F23).

The sequence polymorphism of mitochondrial DNA (mtDNA) hypervariable Segment 1 (HV1) and hypervariable Segment 2 (HV2) are studied and applied to genetic diversity and human evolution assessment, forensic genetics, consanguinity determination, and mitochondrial disease diagnosis. Nucleotide sequence variations in HV1 and HV2, two hypervariable segments of the noncoding control region of human mitochondrial DNA (mtDNA), in selected ethnics of the Vietnamese population were elucidated through sequencing. In this study, we define the variations of HV1 and HV2 of 517 unrelated Vietnamese individuals in Kinh, Muong, Cham, and Khmer ethnic. We found 50 haplogroups: F1a haplogroup frequency is the highest at 15.7%; B5a haplogroup frequency is 10.8%, M haplogroup frequency is 8.9%, M7b1 haplogroup frequency is 7.7%; B6, D4e, D5a, E, F1c, F2a, F3a, G2a, M9b, N, N21 and U5a haplogroup frequencies are the lowest (1%). The frequency of SNP A263G are 100%; A73G is 99.6%, 315insC is 96%; 309insC is 56%; C16223T is 41%, and T16189C is 39%. We have assessed the genetic polymorphism of mtDNA HV1 and HV2 of 517 Kinh, Muong, Cham, Khmer ethnic samples.

Objective: The aim of this study was to evaluate the antioxidant property of the isolated phytocompounds from TA (Terminalia arjuna) bark and in vivo study for nephro-protective and oxidative stress reducing activity in experimentally induced albino male rats.Methods: Fractions from methanol crude TA extract were collected by column chromatography and F27, F28, F29 fractions were selected on the basis of antioxidant property by 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging assay. The in vivo study performed by 30 albino male rats which were randomly divided into five groups: Group I (control)were taken normal food and water, Groups II (uremic) were injected acetaminophen intraperitoneally at the dose of 500 mg/kg/d for 10 d, Group III, IV and V(extract treatment) acetaminophen intraperitoneally at the dose of 500 mg/kg/d for 10 d with co-administered orally of methanol fraction F27, F28, F29 at the dose of 100 mg/kg/d for 15 d respectively.Results: After scarification of rats, the uremic marker plasma urea (80%), creatinine (85%) were elevated and antioxidant enzyme marker such as plasma SOD and catalase level were significantly increased (p<0.05)in Group IV compared to Group II. The total phenolic content of the F28 methanolic fraction was (815.48±8.11) mg gallic acid equivalent/g of extract. For isolation of available compound by 1H NMR study in F28 methanol fraction of TA bark was arjunoside IV which contained olefinic proton (a pair of carbon atom linked with double bond).Conclusion: Among the three methanolic fraction of TA bark, F28 was shown best antioxidative, nephron-protective and oxidative stress reducing property.

Estrus was synchronized in dairy cattle using the synthetic prostaglandin F2a (PG) analog Estnunate (Coopers Animal Health Ltd., Berkhamsed, England. UK; 250 ug/ml of cloprostenol). PG was administered by two routes at two different doses, 500 ug by im route (control) and 125 pg by intravulval lip (ivu) route (treatment). All animals were bred by artificial insemination (AI) to detected estrus using the am-pm breeding rule (BR). First service conception rate (FSCR) for all dairy cows and heifers was 70% for the control and 54% for the treatment groups (P>0.05). The FSCR was also not different (P>0.05) between Holstein (HOL) and Australian Milking Zebu (AMZ) breeds (HOL 52% and AMZ 78%). Moreover, all the animals in this study were confirmed pregnant by the end of the breeding season. In conclusion, dairy cows and heifers can be successfully synchronized using only 125 pg of cloprostenol without impairing fertility. This dose corresponds to only one fourth of the recommended dose of 500 pg of cloprostenol by the manufacturer.

All does were synchronized during the transitional period (during July prior to the breeding season) using fluorogestone acetate (FGA) impregnated intra-vaginal sponges, equine chorionic gonadotropin (eCG) and different prostaglandin F2a (PGF2α) analogues. Four doses of prostaglandin F2a (group 1: 4·5 mg luprostiol; group 2: 2·25 mg luprostiol; group 3: 0·2 mg cloprostenol; group 4: 0·1 mg cloprostenol) were administered i. m. 48 h prior to the end of progestagen treatment and 400 IU eCG was administered at sponge removal. All does were inseminated with fresh semen (300 ✕ 106 spermatozoa per insemination) 42 to 44 h after sponge removal. There were no significant differences (P > 0·05) in the number of goats showing ovarian activity (blood progesterone concentration ³ 1 ng/ml) prior to oestrous synchronization between PGF2α treatment groups. No significant differences (P > 0·05) were observed for the kidding rates between the treatment groups. Our results indicate that goats with ovarian activity prior to sponge application who received high doses of PGF2α showed lower conception rates than those detected in anoestrus (P 0·05). No differences (P 0·05) were observed in conception rates between groups that received lower doses of PGF2α. July is considered to be a transitional period suitable for oestrous synchronization for both Swiss and indigenous dairy goats. Natural mating during the two subsequent cycles after artificial insemination improved kidding rate, but without any significant difference either between treated groups or between Swiss and indigenous breeds.

Abstract:
A rich supply of nerve fibers containing neuropeptide Y-like (NPY-LI) and tyrosine hydroxylase-like immunoreactivity was seen in human cerebral arteries, arterioles and veins. Only a sparse supply of vasoactive intestinal polypeptide (VIP-LI), substance P (SP-LI), and calcitonin gene-related peptide (CGRP-LI) was demonstrated in the walls of human cerebral vessels. In isolated ring segments of human cerebral arteries, NPY and noradrenaline caused vasoconstriction but did not potentiate each other. VIP, peptide histidine methionine, SP, neurokinin A, and CGRP relaxed arteries precontracted by prostaglandin F2a. The degree of innervation and the vasomotor responses are discussed in relation to migraine pathophysiology.

Abstract:
This paper describes a new synthesis of monomeric 2-/(2-methyl-2-propenoyl)oxy/ethyl esters of prostaglandins (IIa-IId). The monomeric esters were prepared by reaction of an alkali metal or triethylammonium salt of a prostaglandin of the F2a or A2 series (Ia-Id) with 2-iodoethyl (IIIa) or 2-bromoethyl-2-methyl-2-propenoate (IIIb) in good to high yields. These monomers have been used for preparing reversible anchoring of prostaglandins on a modified three-dimensional biocompatible hydrogels prepared by copolymerization with 2-hydroxyethyl 2-methyl-2-propenoate. The application of functionalized polymers could be used in drug forms with controlled liberation of biologically active PG derivatives.

The vasoreactivity of the intracranial segment of the internal carotid artery to transmitters, present in the perivascular sympathetic, parasympathetic and sensory nerves, as well as to other vasoactive agents of relevance for headache, was tested in man and monkey. The total arterial segment from both species is equipped with contractile receptors for noradrenaline, serotonin, prostaglandin F2a, ergotamine and sumatriptan. Further, the total arterial segment dilated upon exposure to calcitonin gene-related peptide in both species. Other vascoactive transmitters, acetylcholine, substance P and neurokinin A, caused only weak dilatation, restricted to the proximal extracavernous segment in the monkey. The findings are discussed in relation to the pathogenesis and treatment of cluster headache.

Background: The use of continuing antipsychotic medication is an established evidence-based strategy for preventing relapse in people with schizophrenia, but medication adherence is known to be suboptimal. Covert non-adherence can be eliminated by the use of long-acting injectable (LAI) formulations. We sought to (1) raise awareness among clinicians of the potential benefits of LAI antipsychotic formulations, (2) increase use of these formulations for the treatment of schizophrenia in routine clinical practice and thereby (3) reduce the number of relapses requiring hospitalisation in patients with schizophrenia under our care. Method: Educational initiatives, promotion of reflective practice and patient-specific reminders were used to prompt increased use of LAI antipsychotic medication for patients with schizophrenia. Data relating to the use of these medications and the number of acute admissions for schizophrenia spectrum disorders (F20-29, ICD-10) over time were extracted from existing clinical information systems. Results: Over the 3-year time frame of our local initiative, the use of LAI antipsychotic preparations increased by 11%, the number of acute admissions for schizophrenia/schizoaffective disorder (F20 and F25) decreased by 26% and the number of acute bed days occupied by patients with these diagnoses decreased by 8%. The number of admissions for other psychosis diagnoses (F21-24 and F28-29) did not show the same pattern of improvement. Conclusion: In our health care organisation, raising clinicians’ awareness of the evidence base relating to the potentially favourable benefit–risk balance for LAI antipsychotic medication compared with oral formulations resulted in more use of the former. There were accompanying reductions in acute admissions and occupied bed days for patients with schizophrenia.
