Abstract:
Fusaricidins and related LI-F compounds are effective bactericides and fungicides. Recently, we have found that they are highly toxic to mammalian cells. Here, we studied the effect of fusaricidin-type compounds (FTCs) on the membranes of mammalian cells. Ethanol extracts from Paenibacillus polymyxa strains, RS10 and I/Sim, were fractionated and analyzed by HPLC and mass spectrometry. The effects of FTCs on mitochondrial functions and integrity were studied by standard methods: measurements of swelling, membrane potential (ΔΨm), respiration rate, cytochrome c release, and pore sizes. Superoxide flashes were registered by 3,7-dihydro-2-methyl-6-(4-methoxyphenyl)imidazol[1,2-a]pyrazine-3-one (MCLA). Plasma membrane permeability was assessed by propidium iodide (PI) staining and ATP release. FTCs caused the permeabilization of the inner mitochondria membrane (IMM) to ions and low-molecular-weight (~750 Da) solutes. The permeabilization did not depend on the permeability transition pore (mPTP) but was strongly dependent on ΔΨm. Fusaricidins A plus B, LI-F05a, and LI-F05b–LI-F07b permeabilized IMM with comparable efficiency. They created pores and affected mitochondrial functions and integrity similarly to mPTP opening. They permeabilized the sperm cell plasma membrane to ATP and PI. Thus, the formation of pores in polarized membranes underlays the toxicity of FTCs to mammals. Besides, FTCs appeared to be superior reference compounds for mPTP studies.

Abstract:
Objective Influenza season occurs every year in China, but its presentation was unusual in the period from December 2017 to early 2018. During this period, influenza activity was increasing across the country and was much greater than during the same period in previous years, with great harm to people’s health. Methods In this study, we isolated two human influenza virus strains—A/Hebei/F076/2018(H1N1) and B/Hebei/16275B/2018—from patients with severe influenza in Hebei, China, during the flu season in January 2018, and explored their genetic characteristics, pathogenicity, and transmissibility. Results A/Hebei/F076/2018(H1N1) belongs to the human-like H1N1 influenza virus lineage, whereas B/Hebei/16275B/2018 belongs to the Victoria lineage and is closely related to the World Health Organization reference strain B/Brisbane/60/2008. Pathogenicity tests revealed that A/Hebei/F076/2018(H1N1) replicated much more strongly in mice, with mice exhibiting 40% mortality, whereas B/Hebei/16275B/2018 was not lethal. Both viruses could be transmitted through direct contact and by the aerosol route between guinea pigs, but the H1N1 strain exhibited higher airborne transmissibility. Conclusions These results may contribute to the monitoring of influenza mutation and the prevention of an influenza outbreak.

Las plantas asintomáticas son reservorios de patógenos, ya que pueden permanecer infectadas la mayor parte de su ciclo de desarrollo, convirtiéndose en fuente de contaminación para el resto del cultivo. El objetivo de este estudio fue evaluar un método de detección y discriminación de dos cepas de Fusarium oxysporum en tomate usando espectroscopia. La enfermedad en las plantas de tomate inoculadas con la cepa aislada de gulupa (F05) fue mayor a la observada en la cepa aislada de tomate (F07), presentando valores de 60,0% (11 días) y 81,8% (22 días); la cepa F07 presentó incidencias de 30,0 y 64,3% en ambas mediciones. La planta infectada con la cepa F05 fue mejor discriminada en el periodo de incubación de la enfermedad en ambos periodos de tiempo en los Análisis de Componentes Principales (PCA) y Análisis Discriminantes Lineales (LDA) realizados con los controles en comparación con la cepa F07. Estos resultados sugieren que la espectroscopia de reflectancia VIS es un método sensible y confiable que puede ser adecuado para el diagnóstico temprano de enfermedades en plantas.

The cyclic peptide core of the antifungal and antibiotic cyclic depsipeptide LI-F04a was synthesised by using a modified Yamaguchi macrolactonization approach. Alternative methods of macrolactonization (e.g., Corey–Nicolaou) resulted in significant epimerization of the C-terminal amino acid during the cyclization reaction. The D-stereochemistry of the alanine residue in the naturally occurring cyclic peptide may be required for the antifungal activity of this natural product.

Abstract Background Clostridioides difficile is an anaerobic spore-forming, toxin-producing Gram-positive bacillus listed by the CDC as an “urgent threat” pathogen. Epidemiologic studies using whole-genome sequencing (WGS) have found that genetically distinct lineages infections occur in hospitalized patients, in addition to the fact that C. difficile spores persist on hospital surfaces after disinfection. The purposes of this study were to isolate and characterize C. difficile from the environment of a large Texas hospital. Methods We collected 330 swab samples of hospital environmental surfaces using sterile cotton gauze. The samples were then anaerobically enriched in brain heart infusion broth for 48–72 hours and plated onto cycloserine–cefoxitin fructose agar (CCFA). Suspected colonies were then genetically characterized using PCR (for tcdA, tcdB, cdtA, cdtB and tpi genes) and genotyped using fluorescent PCR ribotyping techniques. Results A total of 90/330 (27.3%) environmental samples were culture positive for C. difficile, of which 75/90 (82.1%) tested were toxigenic C. difficile by the presence of tcdA, tcdB, cdtA or cdtB. A total of 16 distinct ribotypes were identified from 41 C. difficile isolates tested using a fluorescent-ribotyping method. The predominant ribotypes isolated were F078–126 (8/41), F002 (5/41), F106 (4/41), F255 (4/41), and F014-020 (3/41). Conclusion We found a diversity of C. difficile strain types in various hospital high-touch surface environment in addition to ribotype F027 and F078, suggesting the hospital environment a reservoir and significant source C. difficile infections. Latar Belakang. Laserasi perineum adalah salah satu akibat dari proses persalinan denganberbagai penyebab seperti kelahiran kepala secara tiba-tiba dan cepat, berat janin yang berlebihan, danjaringan kulit ibu yang mudah robek. Komplikasi dari laserasi perineum adalah penyembuhan lukayang tertunda, infeksi, nyeri, takut bergerak, dan terhambatnya pengeluaran lochea. Hambatan aliranlochea pada infeksi postpartum dapat menyebabkan kematian ibu. Binahong (Anredera Cordifolia(ten) Steenis) adalah zat alami yang membantu penyembuhan luka. Kandungan asam askorbat dalambinahong penting untuk mengaktifkan enzim hidroksilasi prolil yang mendukung tahap hidroksilasidalam pembentukan kolagen, yang mempercepat proses penyembuhan luka. Tujuan. Penelitian inibertujuan untuk mengetahui sifat fisik, homogenitas, uji pH, dan organoleptik ekstrak daun binahongsetelah diformulasikan ke dalam sabun cair dengan variasi konsentrasi 100%. Metode. Penelitianini menggunakan metode eksperimental, yang dilakukan di laboratorium farmasi. Ekstraksi daunbinahong dilakukan dengan merebus daun selama 30 menit dengan suhu 90°C menggunakanpelarut air. Sabun cair dibuat dengan dasar sabun PEG, carbopol (f0a), esaflor (f0b), HPMC (f0c),asam sitrat dan sodium hidrogen fosfat. Hasil f0c Formula dengan HPMC memiliki evaluasi terbaikdibandingkan dengan f0a dan f0b yang terlihat memiliki pH netral, organoleptik dan sifat fisikterbaik di antara formula lainnya. Kesimpulan. Sabun cair Binahong (Formula f0c) layak sebagaibahan sabun cair anti-septik dari hasil evaluasi stabilitas formula dan persyaratannya. Saran. Hasilpenelitian ini dapat digunakan sebagai referensi untuk tahap selanjutnya, yaitu optimalisasi rumusformulasi sabun cair sebagai sabun antiseptic

Abstract:
Мета роботи – встановлення клінічних особливостей пограничних нервово-психічних розладів у хворих на ВІЛ-інфекцію у I клінічній стадії недуги. Пацієнти і методи. Здійснили порівняльне дослідження пограничних нервово-психічних розладів (ПНПР) при I клінічній стадії ВІЛ-інфекції (основна група – 86 хворих) та в осіб з несправжньопозитивним скринінговим тестом на ВІЛ (група порівняння – 43 пацієнти). Результати досліджень. При клініко-психопатологічному обстеженні встановлено, що усі 100,0 % пацієнтів основної групи страждають клінічно вираженими ПНПР. У групі порівняння зазначена симптоматика проявлялася у (67,4±7,1) % осіб (Р<0,001) і характеризувалася відносно легшою клінікою. Найчастіше у пацієнтів з ВІЛ-інфекцією реєструються: органічні психічні розлади (F06-F07) – (32,6±5,1) %; розлади зрілої особистості та поведінкові розлади (F60-F61) – (27,9±4,8) %; поєднані психічні порушення (F60-F61; F06-F07; F40-F48; F30-F39) – (20,9±4,4) %; невротичні розлади (F40-F48) – (12,7±3,6) %; афектні розлади (F30-F39) – (5,8±2,5) %. Виявлена психічна патологія залежить від преморбідного фону і впливу ВІЛ-інфекції. Зокрема у преморбідних психічно гармонійних осіб після отримання інформації про інфікування ВІЛ формуються невротичні та афектні розлади. Переважання органічних психічних розладів у пацієнтів в досліджених групах пояснюється тим, що ВІЛ-інфекція нашаровується на попередній преморбідний фон, змінений вживанням психоактивних речовин, черепномозковими травмами і хронічними соматичними захворюваннями (хронічні гепатити В і/або С, венеричні хвороби). Частота розладів зрілої особистості обумовлюється несприятливими мікросоціальними факторами в дитячо-підлітково-юнацькому віці та патологічним розвитком особистості після інфікування ВІЛом. Вплив ВІЛ-інфекції на психічне здоров’я пацієнтів проявляється в достовірно частішому «нашаруванні» органічних, невротичних, афектних порушень на наявні розлади особистості.

Abstract:
The effect of concentration of cobalt chromium in hemolysis test for dental implant. Dental implants are used to replace tooth/teeth loss and its function. Cobalt chromium has ideal characteristics to be made as dental implants material. It is required that the material to be implanted must be biocompatible with cells and tissues. One of biocompatibility characteristics is hemocompatibility. Hemocompatibility of materials can be observed with hemolysis test. Thus the purpose of this research is to know whether cobalt chromium as dental implants material affect the hemolysis of rabbit blood or not. This research was done with rabbit blood (Oryctolagus cuniculus) and devided into 3 groups (treatment, positive and negative control). The tested material was cobalt chromium Remanium® GM 800, a product from Dentaurum. The contact between blood and material was done with ASTM-F075 hemolysis test. Cobalt chromium was not hemolytic at 2,5%, 5%, and 10% of concentration, slightly hemolytic at 20% of concentration, and hemolytic at 40% and 80% of concentration. The conclusion of this research was variety of concentration of cobalt chromium affected hemolysis percentage signi cantly.ABSTRAKImplan gigi digunakan untuk mengganti gigi yang hilang untuk dan dapat mengembalikan fungsi gigi. Cobalt chromium memiliki sifat-sifat yang memenuhi persyaratan sebagai material implan. Material yang diimplankan dalam tubuh harus memiliki sifat biokompatibilitas. Salah satu sifat biokompatibilitas yang harus dimiliki material yang diimplankan dalam tubuh adalah sifat hemokompatibilitas. Sifat hemokompatibilitas dapat diketahui dengan uji hemolisis. Penelitian ini bertujuan untuk mengetahui ada/tidaknya pengaruh logam cobalt chromium sebagai material implan gigi terhadap hemolisis pada darah kelinci. Penelitian ini dilakukan menggunakan sampel darah yang didapat dari kelinci (Oryctolagus cuniculus) yang dibagi menjadi 3 kelompok, yaitu perlakuan, kontrol positif dan negatif. Bahan penelitian yang diuji adalah cobalt chromium Remanium GM 800 produksi Dentaurum. Kontak darah dengan bahan uji dilakukan menggunakan uji hemolisis ASTM-F075. Hasil penelitian menunjukkan material logam cobalt chromium tidak menimbulkan hemolisis pada konsentrasi 2,5%, 5%, dan 10%, menimbulkan hemolisis ringan pada konsentrasi 20%, dan hemolisis pada konsentrasi 40% dan 80%. Kesimpulan dari penelitian ini adalah perbedaan konsentrasi logam cobalt chromium berpengaruh secara signikan terhadap persentase hemolisis.

Abstract:
This study aimed to evaluate in vitro the plant growth promotion capacity of rhizospheric fungi from Opuntia cochenillifera. The isolates were submitted to phosphate solubilization, acetic acid-3-indole (AIA) production, antagonism against phytopathogens, and growth under water activity. All test results were subjected to statistical analysis. The studied isolates were able to solubilize phosphate, being F04 (Aspergillus sp.) and F05 (Coprinellus radians) the ones that presented the highest solubilization indices. As to produce AIA, F02 (Penicillium sp.) and F07 (Aspergillus sp.) were the most promising. All isolates could inhibit the growth of Colletotrichum sp. and Fusarium sp. Water restriction provided mycelial growth for all isolates. Therefore, these characteristics confer characteristics as growth promoters in plants.
