Abstract:
The quality of eggs is tightly associated with freshness. New possibilities for the determination of egg freshness were studied. The volatile compounds of eggs and their changes during storage were followed. Three methods for extraction of volatiles were compared: dynamic headspace (Purge and Trap), static headspace (Solid Phase of Microextraction – SPME) and extraction according to Likens-Nickerson by simultaneous distillation-extraction (SDE) with diethyl ether as organic solvent. The extracts were analysed by GC/FID. The volatiles in an extract obtained by SDE method were identified by GC-MS. The extract includes aldehydes, alcohols, acids and esters. The volatiles in an extract obtained by SPME and Purge and Trap have not been identified until now. The changes in volatiles during storage of eggs using the above mentioned methods were studied.

Abstract:
The purpose of this research was to produce the enriched eggs, but low in cholesterol content. Forty eight layer chickens aged 72 weeks (strain Dekalb Warren) were distributed into six treatment groups. One group of laying hens was fed diets without Sauropus androgynusleaf extract (SALE) as the control, and another five groups were fed diet supplemented to 9 g SALE, 18 g SALE, 27 g SALE, 36 g SALE, and 45 g SALE/kg diet, respectively. Completely randomized design was used in the present study. The experimental results showed that SALE supplementation had no effect on egg production, feed intake, feed conversion ratio, egg weight, yolk weight, albumen weight, shell egg, egg index, eggshell thickness, and yolk index (P>0.05), but significantly affected Haugh Unit, albumen index, yolk color, egg odor, egg taste, and yolk color (P<0.05). The protein, fat, calsium, phosphor, kaliumand iron contents of eggs did not change (P>0.05), but the cholesterol content of egg was significantly reduced (P<0.05). In conclusion, the supplementation of SALE did not enhance performance, but it improve egg quality and its chemical composition.

Abstract:
The assembly of microtubules during mitosis requires many identified components, such as γ-tubulin ring complex (γ-TuRC), components of the Ran pathway (e.g., TPX2, HuRP, and Rae1), and XMAP215/chTOG. However, it is far from clear how these factors function together or whether more factors exist. In this study, we used biochemistry to attempt to identify active microtubule nucleation protein complexes from Xenopus meiotic egg extracts. Unexpectedly, we found both microtubule assembly and bipolar spindle assembly required glycogen, which acted both as a crowding agent and as metabolic source glucose. By also reconstituting microtubule assembly in clarified extracts, we showed microtubule assembly does not require ribosomes, mitochondria, or membranes. Our clarified extracts will provide a powerful tool for activity-based biochemical fractionations for microtubule assembly.

Abstract:
A low level of vigilance is one of the main reasons for traffic and industrial accidents. We conducted experiments to evoke the low level of vigilance and record physiological data through single-channel electroencephalogram (EEG) and electrocardiogram (ECG) measurements. In this study, a deep Q-network (DQN) algorithm was designed, using conventional feature engineering and deep convolutional neural network (CNN) methods, to extract the optimal features. The DQN yielded the optimal features: two CNN features from ECG and two conventional features from EEG. The ECG features were more significant for tracking the transitions within the alertness continuum with the DQN. The classification was performed with a small number of features, and the results were similar to those from using all of the features. This suggests that the DQN could be applied to investigating biomarkers for physiological responses and optimizing the classification system to reduce the input resources.

Abstract:
As the first step in determining whether or not pre-replication complexes are assembled at specific sites along mammalian chromosomes, nuclei from G(1)-phase hamster cells were incubated briefly in Xenopus egg extract in order to initiate DNA replication. Most of the nascent DNA consisted of RNA-primed DNA chains 0.5 to 2 kb in length, and its origins in the DHFR gene region were mapped using both the early labeled fragment assay and the nascent strand abundance assay. The results revealed three important features of mammalian replication origins. First, Xenopus egg extract can selectively activate the same origins of bi-directional replication (e.g. ori-beta) and (beta') that are used by hamster cells in vivo. Previous reports of a broad peak of nascent DNA centered at ori-(beta/(beta)' appeared to result from the use of aphidicolin to synchronize nuclei and from prolonged exposure of nuclei to egg extracts. Second, these sites were not present until late G(1)-phase of the cell division cycle, and their appearance did not depend on the presence of Xenopus Orc proteins. Therefore, hamster pre-replication complexes appear to be assembled at specific chromosomal sites during G(1)-phase. Third, selective activation of ori-(beta) in late G(1)-nuclei depended on the ratio of Xenopus egg extract to nuclei, revealing that epigenetic parameters such as the ratio of initiation factors to DNA substrate could determine the number of origins activated.

Abstract:
Microtubule dynamics underlie spindle assembly, yet we do not know how the spindle environment affects these dynamics. We developed methods for measuring two key parameters of microtubule plus-end dynamic instability in Xenopus egg extract spindles. To measure plus-end polymerization rates and localize growing plus ends, we used fluorescence confocal imaging of EB1. This revealed plus-end polymerization throughout the spindle at ∼11 μm/min, similar to astral microtubules, suggesting polymerization velocity is not regionally regulated by the spindle. The ratio of EB1 to microtubule fluorescence revealed an enrichment of polymerizing ends near the spindle middle, indicating enhanced nucleation or rescue there. We measured depolymerization rates by creating a front of synchronized depolymerization in spindles severed with microneedles. This front could be tracked by polarization and fluorescence microscopy as it advanced from each cut edge toward the associated pole. Both imaging modalities revealed rapid depolymerization (∼30 μm/min) superimposed on a subset of microtubules stable to depolymerization. Larger spindle fragments contained a higher percentage of stable microtubules, which we believe were oriented with their minus ends facing the cut. Depolymerization was blocked by the potent microtubule stabilizing agent hexylene glycol, but was unaffected by α-MCAK antibody and AMPPNP, which block catastrophe and kinesin motility, respectively. These measurements move us closer to understanding the complete life history of a spindle microtubule.

Abstract:
It is a significant issue to accurately and quickly extract brain evoked potentials under strong noise in the research of brain-computer interface technology. Considering the non-stationary and nonlinearity of the electroencephalogram (EEG) signal, the method of wavelet transform is adopted to extract P300 feature from visual, auditory and visual-auditory evoked EEG signal. Firstly, the imperative pretreatment to EEG acquisition signals was performed. Secondly, respectivly obtained approximate and detail coefficients of each layer, by decomposing the pretreated signals for five layers using wavelet transform. Finally, the approximate coefficients of the fifth layer were reconstructed to extract P300 feature. The results have shown that the method can effectively extract the P300 feature under the different visual-auditory stimulation modes and lay a foundation for processing visual-auditory evoked EEG signals under the different mental tasks.

Abstract:
ABSTRACT An extract from activated Xenopus eggs joins both matching and nonmatching ends of exogenous linear DNA substrates with high efficiency and fidelity (P. Pfeiffer and W. Vielmetter, Nucleic Acids Res. 16:907–924, 1988). In mammalian cells, such nonhomologous end joining (NHEJ) is known to require the Ku heterodimer, a component of DNA-dependent protein kinase. Here I investigated whether Ku is also required for the in vitro reaction in the egg extract. Immunological assays indicate that Ku is very abundant in the extract. I found that all NHEJ was inhibited by autoantibodies against Ku and that NHEJ between certain combinations of DNA ends was also decreased after immunodepletion of Ku from the extract. The formation of a joint between a DNA end with a 5′-protruding single strand (PSS) and an end with a 3′-PSS, between two ends with 3′-PSS, and between two blunt ends was most Ku dependent. On the other hand, NHEJ between two DNA ends bearing 5′-PSS was Ku independent. These results show that theXenopus cell-free system will be useful to biochemically dissect the role of Ku in eukaryotic NHEJ.

This research was carried out to determine the effect of Jerusalem artichoke (JA) with or without 5, 10% vetch (V) supplementation on performance, egg quality characteristics and egg cholesterol content. In the study, seventy-five 25 weeks-old commercial white laying hens were randomly divided into one control and 4 treatment groups each containing 15 hens. Control group was fed basal diet without JA or V. Treatment group 1, 2, 3 and 4 were fed diets containing 5% V, 5% JA, 5% JA + 5% V and 10% JA + 10% V, respectively. The feeding period lasted 16 weeks. In the study, it was determined that dried-ground JA contained dry matter, crude protein, ether extract, crude fibre, crude ash, inulin and metabolizable energy (ME) at the level of 93.30, 10.02, 0.36, 5.64, 9.05, 15.80% and 3 060 kcal/kg, respectively. At the end of the study, live weight was lower by 4.36–10.09% in the treatment group 10% JA + 10% V compared with the other groups, but feed efficiency was improved in this group. There were statistically significant differences between the groups in egg quality characteristics (P < 0.05, P < 0.001). Egg production was not affected by supplementation of 5, 10% JA with or without 5, 10% V supplementation. Abstract:
Nuclei assembled in Xenopus egg extract from purified DNA or chromatin resemble their natural counterparts in a number of structural and functional features. However, the most obvious structural element of normal interphase nuclei, the nucleolus, is absent from the in vitro reconstituted nuclei. By EM, cytological silver staining, and immunofluorescence microscopy employing antibodies directed against various nucleolar components we show that nuclei assembled in vitro contain numerous distinct aggregates that resemble prenucleolar bodies (PNBs) by several criteria. Formation of these PNB-like structures requires pore complex-mediated nuclear transport of proteins but is independent of the genetic content of the in vitro nuclei as well as transcriptional and translational events. Our data indicate that nuclei assembled in vitro are capable of initiating early steps of nucleologenesis but that the resulting PNBs are unable to fuse with each other, probably due to the absence of a functional nucleolus organizer. With appropriate modifications, this experimental system should be useful to define and analyze conditions promoting the site-specific assembly of PNBs into a coherent nucleolar body.

Abstract:
When demembranated sperm nuclei are placed in a Xenopus egg extract, they become surrounded by a nuclear envelope and then swell to form morphologically typical pronuclei. Granules ranging from < 1.0 to approximately 3.0 microns in diameter appear within such nuclei. Bell et al. identified four nucleolar proteins in these "prenucleolar bodies" by immunofluorescent staining (fibrillarin, nucleolin, B23/NO38, 180-kDa nucleolar protein). By in situ hybridization we show that these bodies also contain U3 and U8 small nuclear RNAs (snRNAs), known to be involved in pre-rRNA processing. Moreover, they contain all the snRNAs involved in pre-mRNA splicing (U1, U2, U4, U5, and U6), as well as U7, which is required for histone pre-mRNA 3' end formation. In addition to the nucleolar antigens previously identified, we demonstrated staining with antibodies against the Sm epitope, trimethylguanosine, and coilin. Because the composition of these prenucleolar bodies is closer to that of coiled bodies than to nucleoli, we propose that they be referred to as coiled bodies. The existence of large coiled bodies in transcriptionally inactive pronuclei suggests that they may play a role in the import, assembly, and storage of RNA processing components but are not themselves sites of processing. In transcriptionally active nuclei coiled bodies could serve as sites for initial preassembly and distribution of snRNP complexes for the three major RNA processing pathways: pre-mRNA splicing, pre-rRNA processing, and histone pre-mRNA 3' end formation.

Abstract:
Xenopus embryos initiate replication at random closely spaced sites until a certain concentration of nuclei is achieved within the embryo, after which fewer, more specific chromosomal sites are utilized as origins. We have examined the relationship between nucleo-cytosolic ratio and origin specification when Chinese hamster ovary (CHO) cell nuclei are introduced into Xenopus egg extracts. At concentrations of intact late-G1-phase nuclei that approximate early Xenopus embryos, the entire genome was duplicated nearly 4 times faster than in culture, accompanied by a de-localization of initiation sites at the dihydrofolate reductase (DHFR) locus. As the concentration of nuclei was increased, the number of initiation sites per nucleus decreased and initiation at the DHFR locus became localized to the physiologically utilized DHFR origin. Origin specification was optimal at nuclear concentrations that approximate the Xenopus mid-blastula transition (MBT). Higher concentrations resulted in an overall inhibition of DNA synthesis. By contrast, with intact early G1-phase nuclei, replication initiated at apparently random sites at all concentrations, despite an identical relationship between nucleo-cytosolic ratio and replicon size. Furthermore, permeabilization of late-G1-phase nuclei, using newly defined conditions that preserve the overall rate of replication, eliminated site-specificity, even at nuclear concentrations optimal for DHFR origin recognition. These data show that both nucleo-cytosolic ratio and nuclear structure play important but independent roles in the regulation of replication origin usage. Nucleo-cytosolic ratio clearly influences the number of replication origins selected. However, titration of cytosolic factors is not sufficient to focus initiation to specific sites. An independent mechanism, effecting changes within G1-phase nuclei, dictates which of many potential initiation sites will function as an origin.

Abstract:
Black garlic is a type of garlic that is processed for a specific period, with precisely controlled degrees of moisture and heat until the color of raw garlic turns black. Black garlic plays a strong role in preventing diseases such as hyperlipidemia. This study aims to compare the effect of Egyptian black garlic extract (EBG) and Egyptian raw garlic extract (ERG) at different concentrations on serum biochemical parameters in hyperlipidemia rats’ model. Fifty albino rats were divided randomly into 10 groups with five animals. Rats were fed a normal diet (control) G1, high-fat diet (HFD) 40% lard w/w were (G2- G10) for four weeks to promoting hypercholesterolemia for all rats except G1 (control). After that, HFD groups were treated with 1ml oral administration three times a week of 100% (G3), 75% (G4), 50% (G5), and 25% (G6) EBG extract for 6 weeks. G2 (HFD) was a high-fat diet alone without any extract. ERG extract feed group were treated in the same manner described above, and G7 (HFD+100% ERG extract), G8 (HFD+75% ERG extract), G9 (HFD+50% ERG extract), and G10 (HFD+25% ERG extract) as written here. EBG extract-treated group showed significantly increased high-density lipoprotein cholesterol (HDL-C) in serum (p < 0.05), decreased both level of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG) in serum. EBG extract with high concentration (G3) causes down regulation of lipid and cholesterol (p < 0.05) comparing with ERG extract indicating usefulness for treating hypercholesterolemia. Body weights on rats decrease by feeding of EBG extract, along with liver enzymes decrease in EBG fed rats. This indicates the efficiency of black garlic extract in reducing the level of hypercholesterolemia and avoiding heart disease and atherosclerosis. Abstract:
Novel trends in affective computing are based on reliable sources of physiological signals such as Electroencephalogram (EEG), Electrocardiogram (ECG), and Galvanic Skin Response (GSR). The use of these signals provides challenges of performance improvement within a broader set of emotion classes in a less constrained real-world environment. To overcome these challenges, we propose a computational framework of 2D Convolutional Neural Network (CNN) architecture for the arrangement of 14 channels of EEG, and a combination of Long Short-Term Memory (LSTM) and 1D-CNN architecture for ECG and GSR. Our approach is subject-independent and incorporates two publicly available datasets of DREAMER and AMIGOS with low-cost, wearable sensors to extract physiological signals suitable for real-world environments. The results outperform state-of-the-art approaches for classification into four classes, namely High Valence—High Arousal, High Valence—Low Arousal, Low Valence—High Arousal, and Low Valence—Low Arousal. Emotion elicitation average accuracy of 98.73% is achieved with ECG right-channel modality, 76.65% with EEG modality, and 63.67% with GSR modality for AMIGOS. The overall highest accuracy of 99.0% for the AMIGOS dataset and 90.8% for the DREAMER dataset is achieved with multi-modal fusion. A strong correlation between spectral- and hidden-layer feature analysis with classification performance suggests the efficacy of the proposed method for significant feature extraction and higher emotion elicitation performance to a broader context for less constrained environments.

Abstract:
The basic problem of literary translation lies in its very possibilities, in the limits and aims of all reflection on it, and in the capacity to shift this reflection to practice. In this context, the methodological starting point is normally the target text, from which premises, methods and procedures are extracted. In this paper, however, our aim is to take translation itself, as applied to didactics, as our starting point in order to extract from the supervised translating process possible conclusions regarding the essence of the process and its generalization for practice. The final aim is to extract theoretical principles based on practical principles, reflections on recurring difficulties, information about possible textual markers pointing to problem-solving formats (source text quality, loyalty concept, tone, etc., which are always understood from the point of view of what can be verified or pointed out specifically in texts). This paper aims, through examples, to draw attention to the peculiarity of literary translation didactics as a possible research method into literary translation.

Abstract:
We investigated the effects of histone H1s on DNA replication using Xenopus egg extract. Mouse variants H1c and H10 were assembled onto Xenopus sperm chromatin by the extract during the remodeling that accompanies nuclear decondensation. The association of H1 with chromatin was rapid and concentration dependent. H1-associated chromatin displayed a typical nucleosomal repeat pattern indicating that linker histones are properly positioned along the DNA. The presence of H1 on sperm chromatin reduced both the rate and extent of DNA replication in egg extract. This reduction in rate is due, in part, to a delay in initiation of replication within individual nuclei. Initiation in extract is dependent upon nuclear assembly. Analysis of the assembly process revealed that H1 does not inhibit nuclear membrane formation or the import of nuclear protein, however, it does slow the rate of nuclear lamina formation. This H1-induced delay in lamina assembly is responsible for the delay in initiation as pre-assembled H1-containing nuclei initiate replication at the same time as control nuclei. However, H1 inhibits replication even when lamina assembly is complete suggesting that H1 also affects replication directly. These data indicate that H1 modulates DNA replication through multiple pathways in egg extract.

Hen egg is easy to get and affordable. Besides having a complete nutritional content, chicken eggs are also easily damaged due to microbial activity during storage so that it can reduce egg quality. Therefore, a preservation process is needed to maintain egg quality. This study aims to determine the effect of soursop leaf extract concentrations on decreasing the quality of eggs. This research used a completely randomized design with two factors, the first factor was the concentration of soursop leaf extract (20, 30, 40%) and the second was the shelf life (0, 7, 14, 21 and 28 days). Observation included egg white index, egg yolk index, haugh unit, and weights of eggs. The results indicate that eggs with soursop leaf extract immersion treatment can maintain egg quality. The concentration of soursop leaf extract as much as 40% gives better results in maintaining the quality of eggs.

Abstract:
The main purpose of this study was to evaluate the characteristics of eggs produced by Japanese quails fed with guava extract. A total of 400 eggs collected from the quails fed with experimental diets, were distributed in a completely randomized design in factorial arrangement 4 x 4 with main effects including four dietary levels of guava extract (0, 3, 6 and 9 g kg-1) and four storage periods (0, 9, 18 and 27 days) totalizing 16 treatments with five replicates of five eggs. The eggs were produced by Japanese quails, fed with experimental diets. The diets were isonutritive formulated by corn-soybean basis. The eggs were stored for different periods, at the same conditions, to constitute the treatments. The parameters evaluated were egg weight loss (g and %), yolk color, Haugh unit, specific gravity, eggshell thickness and pH of the yolk and albumen. Data were submitted to ANOVA, and means were compared using Scott-Knott test, using α = 0.05. The specific gravity of fresh eggs was better with the use of 6 g kg-1 of guava extract in the diet. The use of 3 g kg-1 of guava extract resulted in smaller eggshell thickness of fresh eggs. The Haugh unit was affected just by the storage periods. The dietary guava extract (3, 6 or 9 g kg-1) increased the pH of albumen in eggs stored for 27 days. The use of 6 g kg-1 of guava extract in quails diet increase the specific gravity in fresh eggs and increase the yolk color when the eggs are stored.

Abstract:
The purpose of this study was to study the effect of immersion on dayak onion extract and storage time and the interaction of these two factors on decreasing of egg weight, haugh unit value and albumen pH of duck egg. The study used a completely randomized design of factorial pattern 4 x 4 with 4 replications. The treatment is as follows: The first factor is the concentration of onion dayak extract (A), consisting of: A0 = Eggs storage placed at room temperature; A1 = 10% concentrated of dayak onion extract; A2 = 20% concentrated of dayak onion extract; A3 = 30% concentrated of dayak onion extract; The second factor is the length of storage (B) consists of: B0 = 7 days. B1 = 14 days. B2 = 21 days. And B3 = 28 days. Research data were analyzed using analysis of variance and continued by Tukey Test. The results showed that the concentration of dayak onion extract did not affect the reduction in egg weight, but did affect the haugh unit and albumen pH. Egg weight reduction ranged from 0.92-1.02. Haugh unit and pH of albumen at 10% and 30% concentration of dayak onion extract did not differ, while the 0% concentration differed from all other treatments. Interaction effects between concentration of dayak onion extract and length storage were also significant for egg weight loss, Haugh unit, and pH of albumen. It is recommended that for preserving duck eggs, it is better to use a concentration of 30% onion extract.
