Abstract:
This Master thesis deals with the valuation of Leo Express, a. s. to the date January 1, 2016. As a valuation method was used the earnings valuation method based on discounted free cash flow to the firm and equity (FCFF). The thesis is split into two parts The methodical part defines the terminplogy and used indicators. The practical part is a detailed analysis of the market for transport of passenger rail services in the Czech Republic, Slovak Republic and Poland by used PEST analysis. The part of Master thesis is also financial analysis of the company. Process of valuation was implemented on the base of well-prepared financial plan and other value drivers. The evaluation itself has been accomplished using three-phase DCF method,which is consist of a prediction phase,second faze and a residual value phase.

Abstract:
The Atomic Force Microscope (AFM) is an invention that has enabled a significant number of studies and discoveries in the field of nanotechnology. It is well-known that the resolution of AFM-based applications is critically dependent on the tip bluntness of the probe utilised. Numerous researchers have proposed different approaches to assess the condition of AFM probe tips. In spite of these efforts, further advances are still needed for the express analysis of the bluntness of such tips. In this context, the overall aim of the research work presented in this Thesis was to investigate a novel in-situ technique for assessing the apex condition of AFM tips. In particular, this technique relies on the analysis of depth-sensing data obtained from the nanoindentation of the probe tip into a soft elastic sample. Nanoindentation is a process that is readily implemented on AFM devices. For this reason, the proposed technique could be a fast an efficient approach for deciding when AFM probes should be replaced. The theoretical argument on which the technique is based is that the current shape of the tip apex in its working position within an AFM device can be approximated as a power-law function and that the exponent of this function can be used as a quantitative measure of the tip bluntness. Based on this approximation and the use of the self-similar (scaling) approach to depth-sensing indentation, it is possible to extract this bluntness parameter, herein also referred to as the degree of tip bluntness, from AFM nanoindentation data. The practical implementation of this technique was realised using a commercial AFM device and commercial probes. The actual geometry of the apex of these probe was also studied in details using additional experimental methods via the use of Scanning Electron Microscopy and also via the so-called “reverse imaging” method to obtain two- and three-dimensional data about the tip apex of these probes. Among the different iv contributions made from the work carried out in this research, the most important conclusion is that a good agreement was found between values of the bluntness parameter evaluated by the proposed technique and the effective bluntness obtained from analysing the actual three dimensional geometry of the AFM tips. Thus, it can be argued that the technique put forward in this work for the express analysis of the bluntness of AFM probe tips using depth-sensing nanoindentation can be considered as a valid method when assessing the condition of AFM probes.

The aim of this thesis is to determine the market value of DHL Express (Czech Republic) s.r.o. based on publicly available information as at 31.12.2011. The first section details the methodological framework for strategic and financial analysis, building a financial plan and determining the market value of a company. The beginning of the practical part contains the evaluated company information and declares the purpose of determining the market value of a company. The strategic analysis and evaluation of a company's financial health is an essential part of the evaluation process. In addition, the individual company value generators are prognosed on the basis of which the financial plan for the next period is built. On the assumption of company's continued existence, the discounted cash flow method DCF was selected.

Low temperature is one of the environmental factors that cause substantial crop losses in the world. Recent advances in the study of plants native from temperate regions, have established that cold acclimation is regulated at the gene expression leveL To study the function ofaXerophyta viscosa cold responsive protein that accumulates in plant cells under various environmental stresses, we have cloned the Xerophyta viscosa cold-responsive cDNA gene designed Xvcor, into a less complex host, Saccharomyces cerevisiae with the purpose of carrying out in vivo functional analysis. Sequence analysis showed that Xvcor encodes a 264 amino acid residue protein (Garwe et al 2002). The Hydropathy plot indicated that the XVCOR protein is highly hydrophobic and contains 6 transmembrane domains (Garwe et al 2002). In order to achieve high-level expression of Xvcor, the gene was placed under the control of a strong promoter (Phosphoglycerate kinase). Reverse-transcription PCR amplification revealed that the Xvcor transcript accumulated in yeast cells. However, SDS-PAGE analysis could not detect the predicted 29.6 kDa recombinant protein, suggesting that the translation might be hampered. Tests designed to measure the enhancement of stress tolerance between the recombinant and the control showed similar growth performance, confirming the absence of recombinant protein. A computer search for codon usage showed that the codon usage bias in Xvcor was low compared with that of highly expressed genes of S. cerevisiae. Together, these results suggest that the codon usage in Xvcor could influence its expression in yeast.

Abstract:
Instagram, as one of today’s largest social media platforms, plays a significant part in the maintaining and reproducing of existing stereotypes and role expectations for women. The purpose of the thesis is to study how Instagram interprets violations against its guidelines, and whether decisions to remove certain pictures from the platform are in line with terms of use, or part of human subjectivity. The noticeable pattern among the removed pictures is that they are often norm-breaking. The thesis discusses communication within Instagram to reveal how and why some pictures are removed while others are not, which limits women’s possibilities to express themselves in non-conventional settings. The study applies semiotics to analyse 12 pictures that were banned from the platform without directly violating its guidelines. Role theory and norms are used to supplement semiotics and shed light on the underlying societal structures of female disadvantage. Two major conclusions are presented: 1) Instagram has unclearly communicated its guidelines to women’s disadvantage, and 2) Instagram has therefore been subjective in the decisions to have the pictures removed from the platform, also to women’s disadvantage. Further, the discussion focuses on how Instagram handles issues related to (1) female sexuality, (2) women stereotyping, and (3) female self-representation.

Genomic information for lepidopteran insects, particularly agricultural pest species, is very limited but urgently needed due to their economic importance and biodiversity. The huge economic losses ($ 1-2 billons / year) caused by the European corn borer (Ostrinia nubilalis, Hübner, ECB) makes this insect species one of the major pests of corn in the United States and western world. Management of ECB by conventional methods is limited but has had a great success by transgenic Bt (Bacillus thuringiensis) corn, which targets insect gut. However, the widespread use of Bt corn may lead to the development of Bt resistance in ECB. Knowledge of genes expressed in the insect gut is considered crucial for understanding basic physiology of food digestion, their interactions with Bt toxins and pathogens, and for discovering new targets for pest management. A large database of 15,000 expressed sequence tags (ESTs) was established from the ECB larval gut. To our knowledge, this database represents the largest gut-specific EST database from a lepidopteran pest. Analysis of 10 aminopeptidase-like genes between Cry1Ab–resistant and –susceptible ECB larvae revealed that aminopeptidase P-like (OnAPP) gene is a strong candidate for its role in Bt toxicity and resistance. The RNA interference mediated reduction in the transcript level of OnAPP gene in ECB larvae resulted in their reduced susceptibily to Cry1Ab. Analysis of the chitinase-like gene (OnCht) revealed its essential role in regulating chitin content of peritrophic membrane (PM). Our results suggest that OnCht may influence food digestion, nutrient absorption or movement of digestive enzymes through the PM and can be an important target for insect management. We also identified and characterized six genes involved in the innate immune defense response in ECB and showed that the expression of these genes were induced when challenged with bacteria. In addition to these results, this research generated significant genomic information for the development of microarray from the larval gut of ECB. The establishment of the feeding-based RNA interference technique could potentially help in delivering dsRNA orally to ECB for high throughput screening of effective genes to be targeted for insect pest management.

Abstract:
This thesis presents a statistical analysis of weaving in a managed lane system which is evolving from a High-Occupancy Vehicle (HOV) system to a High-Occupancy Toll system (HOT). Weaving was, assessed along the I-85 corridor in Atlanta, during three different phases in the conversion from HOV to HOT: 1) the existing HOV managed lane system prior to conversion to HOT lanes, 2) after restriping of some weaving zones but prior to conversion of the HOV lanes to HOT lanes and, 3) after the HOT managed lane system opened. Each phase was analyzed to see how weaving behavior into and out of the managed lane system was affected by changes in the system. To accomplish the analysis, video was collected using Georgia Department of Transportation cameras along the corridor. The videos were transferred to an Android Tablet, in which an App developed by the research team was used to record data from the videos. Using the processed weaving data, a comparison of weaving activity during each phase was performed. Data were also analyzed across time of day, speed differentials, and whether the weaves in question were performed legally (within established weaving zones) or illegally (across double-solid striped lane markings). After a comparison of weaving behavior along different variables, a regression tree analysis was completed. The analysis showed that weaving intensity increased as the system was converted from HOV to HOT. However, illegal weaving decreased significantly once the HOT system was in place, perhaps due to stricter enforcement or perhaps due to driver response to illegally entering and leaving tolled lanes. The regression tree analyses indicated that weaving intensity was highly dependent upon whether it was legal or illegal to weave and upon the phase of conversion during which the weave occurred.

This work regards to the structural conception as a formal element of design in wood architecture. On this approach, the group of projects studied is formed by some realized works of two important Brazilian architects: Severiano Porto and Marcos Acayaba. The time interval comprises the period from 1971 to 1997, which correspond respectively to the years of the creation of first and the last of the analyzed constructions. The research perspective concerns to the relationship between the parts, the whole and the building techniques adopted in each project. Moreover, the analysis focuses on the structure as the link among the different projectual ideas. Thus, the research method firstly includes a survey of specific bibliographies and documents which refer to the structural conception in wood architecture. Secondly,the buildings are analyzed according to the methods of architectural composition discussed through this work.
Este trabalho aborda a concep??o estrutural como elemento formal expressivo na arquitetura em madeira. O universo analisado ? constitu?do de obras constru?das de dois arquiteto nacionais considerados importantes pela cr?tica arquitet?nica: Severiano Porto e Marcos Acayaba. Abstract:
This thesis examines the Grand Paris Express project currently underway in the Île-de-France. The basis of this project is a series of new and renovated railway lines to connect and span across the entirety of the region. They are being planned with the hopes to improve urbanization for the outlying suburbs. The Île-de-France is the wealthiest region in France, but it has high economic inequality between its departments. One hard hit area is Seine-St-Denis. This is the area I will be focusing primarily on. Department number 93, its urban landscape holds histories of rioting, unemployment, and large immigrant populations. In its underdeveloped growth, residents are calling for socio-spatial justice. Professor of Urban and Regional planning Edward Soja (1980) coins this as a “social-spatial dialectic” (208), where any given space is inherently political. Currently, residents in Department 93 are engaged in this “dialectic”, taking action by means of redefining the places where they live. In addition to demands for spatial equality is a more complex and hidden protest: the demand from marginalized citizens for an adjusted French ideology with respect to national identity. In their spatial marginalization, groups are contesting the egalitarian claims of the state based on where they live. I explore the historical significance of the Seine-St-Denis department in the Île-de-France’s urbanizing efforts. This thesis argues that planning for the Grand Paris Express represents the effects of a new French identity in formation. But while Paris is being recreated to accommodate those living in the outer communes, an anti-immigrant state narrative is also resulting in reformed urban planning in an existing discriminatory French ideology. However, as I plan to prove later in my research, the Grand Paris Express will provide more social access for France’s marginalized citizens.

The aim of this study was to uncover and illuminate aspects of spirituality, which may be present in the work of primary school teachers. Four themes emerged out the analysis and were coded using the methodology consistent with an Interpretative Phenomenological Analysis (IPA). The themes were: ● Spirituality as an aspect of identity formation and understanding. ● Relationships as central to understanding and formation. ● Teaching and learning as a shared encounter through mentoring. ● Spirituality as a contextual resource. IPA was chosen as a methodology as it places the participant as central to and expert in their lived experience whilst acknowledging the ways in which the researcher impacts on the interpretative process. Drawing on phenomenology whilst adopting a systematic process of analysis, the material uncovered ways in which spirituality can be used a resource in the professional encounter. The implications for this study points towards a new definition of spirituality that encompasses ‘moments of profundity and connection with other that leads to change’. This is particularly important in relation to the teachers in this study and could be of value to others in the education profession. Seeing one’s spirituality as a source of wisdom and as a contextual resource has allowed the participants to make connections with their colleagues and pupils that draw heavily on their spirituality in order to make sense of and bring change to situations and relationships. These instances are profound in nature for each individual but has brought about change in the situation, relationship or way of teaching and relating to one’s environment. As such the definition of spirituality in this study both encompasses established understanding of what it means to be spiritual but places this in the educational environment and profession. It is hoped that the findings of this study lead to an awareness of the way in which one’s spirituality can be drawn upon as a positive agential resource will be acknowledged in teacher education programmes and school-based teaching.

Reflective practice is deemed as one of the essential learning tools in professional education, particularly in the field of Medicine. Educators have recognised the role of reflective writing in focusing students’ reflective learning (Moon, 2004). Research in this writing genre has highlighted the centrality of successful management of affective meanings to achieve critical reflection (Boud et al., 1985). However, the volume of studies that explored the structure and linguistic features of authorial attitudes is limited, indicating a lack of knowledge. This study investigates the macrostructure of reflective writing and the strategies used by writers to position their stance towards entities and propositions across the sections of the reports. To achieve this aim, a corpus of 47 reflective reports written by four-year medical students at the Birmingham Medical School was compiled and interviews with supervisors were conducted. Drawing on the appraisal theory (Martin and White, 2005) and the parameter-based approach (Bednarek, 2006), a framework of evaluation was developed, featuring the analysis of the targets of the evaluation. The findings demonstrated the role of the situational context in selecting those targets, and the way writers position their stance towards them reflects their awareness of the genre conventions and their professional identity. They also revealed the influence the discourse community in structuring the reports. This thesis concludes with an evaluation of the framework and suggestions for pedagogical implications.

In this context, the purpose of this study is to apply the Analytic Hierarchy Process (AHP) in the evaluation process and selection of a logistics services providers (LSP) for the courier delivery services company, analyzing qualitative and quantitative criteria that express the real needs of the parties, whose aim is to improve the indicators in terms of quality and customer satisfaction. The results indicated that the information technology, responsiveness and quality of management are the most relevant criteria for the contractor because of the need of preserving the enterprise reliability front of your customers and to remain competitive in the courier delivery services area. The results also showed that the criteria and method used could contribute for classification of LSP suitable for the service in terms of deadline, demand and traceability required for operational success. A sensitivity analysis was also carried out to assess the robustness of the decision. Finally, the proposed model proved to be a good alternative to support the decision on the selection of logistics providers for express charge, since the analysis of qualitative and quantitative criteria adhering to organizational practice, can lead to choosing companies that can meet the quality requirements of the sector.
A terceirização das atividades logísticas no Brasil vem crescendo, permitindo as empresas a direcionar os esforços no sentido de suas atividades principais e obter ganhos estratégicos devido ao know-how de empresas especializadas e a possibilidade de reduzir os custos com os ativos da empresa contratada. Desta forma, ferramentas que auxiliem na seleção adequada de fornecedores, pode ser importante para que a empresa atinja seus objetivos. Nesse contexto, o objetivo deste estudo é investigar o uso do método de decisão multicritério Analytic Hierarchy Process (AHP) para a avaliação e seleção de provedores de serviços logísticos (PSLs) para empresas do segmento de cargas expressas analisando critérios qualitativos e quantitativos que expressam as reais necessidades das partes envolvidas, cujo intuito é melhorar os indicadores em termo de qualidade, prazo e satisfação dos clientes. Os resultados demonstraram que a tecnologia da informação, responsividade e a gestão da qualidade são os critérios mais relevantes para a contratante em razão da necessidade da preservação da confiabilidade da empresa frente a seus clientes e para manter-se competitiva no segmento de cargas expressas. Constatou-se que os critérios utilizados e o método contribuíram para a classificação de PSLs adequados para o atendimento em termos de prazo, demanda e rastreabilidade exigidos para o sucesso operacional. Uma análise de sensibilidade também foi desenvolvida para avaliar a robustez da decisão. O modelo proposto mostrou ser uma boa alternativa para apoio a decisão na seleção de fornecedores logísticos para carga expressa, uma vez que a análise de critérios qualitativos e quantitativos aderentes à prática organizacional, pode levar a escolha de empresas que possam atender as exigências de qualidade do setor.

Sugarcane is one of the most important products of the world and Brazil is responsible for 25 % of the world production. One problem of this culture at northeast of Brazil is the early flowering. In our laboratory, it has been made before four subtractive libraries using early and late flowering genotypes in order to identify messages related to the flowering process. In this work, two cDNAs were chosen to make in silico analysis and overexpression constructs. Another approach to understand the flowering process in sugarcane was to use proteomic tools. First, the protocol for protein extraction using apical meristem was set up. After that, these proteins were separated on two bidimensional gels. It was possible to observe some difference for some regions of these gels as well as some proteins that can be found in all conditions. The next step, spots will be isolated and sequence on MS spectrometry in order to understand this physiological process in sugarcane

Road transport is most commonly used out of other modes in terms of freight and passenger transportation on local and regional levels. For European citizens it is a primary mean of their access to services, social activities and employment. The scope of road infrastructure differs in some regions. The major disparity appears when the East of Europe is compared with the West. Actions to link periphery with the core of Europe are being continuously performed, what in this case represents itself in constructing Pan-European Transport Corridors, which will contribute to the connection of the whole territory of Europe. The Via Baltica, chosen as a case for this thesis, is a route to connect Helsinki, Tallinn, Riga, Kaunas, Budzisko and Warsaw. It is the common initiative of Finland, Estonia, Lithuania, Latvia and Poland. In addition, it is to be an express road and part of the I Pan-European Transport Corridor, which is going to connect Baltic countries with the West and South part of Europe. We focus on presenting how the Via Baltica is introduced in official documents and compare it with the process in practice. Our final goal is to assess the planning process of the Via Baltica route in Poland according to top down planning approaches and sustainability criteria. We are trying to find out what are the advantages and disadvantages of the ongoing process. After introducing results of the analysis made for the Via Baltica by Polish experts, we present how stakeholders and local authorities try to reinforce the process, regardless of environmental issues, to achieve economic and social profits. As the result we try find out how are the dimensions of sustainable development handled in the Via Baltica case in Poland. The conclusions are based on our findings concerning several conflicts which appeared during the planning process. They are also based on one-dimension as well as multi-criteria analyses.

The primary focus of this thesis is on representations of Germany and Germans in the sports pages of English newspapers from the mid-1950s to the mid-1990s, when EURO 96 generated press coverage that prompted much comment and criticism, both in England and in Germany. Studies focusing on media representations from the mid 1990s onwards, such as those by Maguire, Poulton and Possamai (1999), Garland and Rowe (1999) and Garland (2004) have been helpful in deconstructing the language used by football journalists and in identifying negative national stereotyping. More recently, however, Ramsden (2007) and Young (2007) have developed our understanding of Anglo-German cultural relations and how they have changed since 1945. In the light of these recent developments this thesis seeks, firstly, to analyse the discourses embedded within the ‘Two World Wars and One World Cup’ meta-narrative which has characterized press coverage of Anglo-German football since international fixtures between the two countries were resumed in 1954 and, secondly, to contextualize them in the broader history of Anglo-German cultural relations and how they developed over the forty years or so that followed. Though drawing on some insights from both cultural and media studies the methodology employed is essential historical. This does not mean, however, that press reports and comment are regarded as unproblematic primary sources. Recent methodological approaches the history of sport, notably by Booth (2005) and Hill (2006), have pointed to the importance of viewing such sources as texts which are thus open to deconstruction. A complementary emphasis on historical context is nevertheless justified, principally because it is important to explain variations that have occurred over time. Though there were some similarities in the way that Anglo-German football was covered in 1954 and 1996 – and at various points in between - there are also striking differences which it is argued here are primarily explained by conditions prevailing at the particular historical junctures at which representations were generated. The relationship which existed between Britain and the Federal Republic of Germany (West Germany) in the 1950s, 1960s and 1970s was significantly different to that which existed between Britain and re-unified Germany in the 1990s. This was an important contingent factor and helps to explain variations in the deployment of journalistic discourses over the years. Thus this thesis breaks new ground in that it emphasizes the historical contextualization of representations over a long period and seeks to counter any tendency to look backwards from the viewpoint of the mid 1990s. The discussion proceeds chronologically from the 1950s to the 1990s in order to demonstrate variations in the way that discourses were deployed over the years. Thus the representations generated provide a way of reading the state of underlying Anglo-German relations at any given point. One chapter is devoted to the 1966 World Cup Final on account of its significance in press discourses relating to Anglo-German football and in what is popularly referred to in England as the 'thirty/forty years of hurt' that followed. Whereas academic attention in relation to football-related representations has previously concentrated on the downmarket tabloid press, this study is equally concerned with quality and middlemarket titles. Thus The Times and the Daily Express are considered alongside the Daily Mirror and the Sun. Finally – and in contrast to previous accounts which have considered the English press in isolation – a chapter on German newspaper coverage (principally Bild, Die Welt and Frankfurter Allgemeine Zeitung) has been included to allow some comparisons to be made and to point to directions in which future research might be pursued.

The purpose of this study is to find and clarify the most limited set of metrics that reflect the main directions of assessing the success and condition of the enterprise and are suitable for conducting rapid analysis. The system of coefficients for express analysis of financial statements proposed in this study is presented. For each of the presented areas of analysis, there are a large number of coefficients and methods for calculating them, but the ideology of express analysis implies minimizing their number, ideally, to one indicator for each of the areas. Therefore, the coefficients are selected on the basis of the principle of the most general representation of the relevant direction of analysis.

Resumo: O Brasil tem a maior produção de café no mundo e é o segundo mercado consumidor, atrás somente dos EUA. O consumo de café está crescendo constantemente, graças a seus efeitos fisiológicos e ao seu sabor e aroma agradáveis. Associado a este consumo, a maior preocupação com a saúde estimulou a área de pesquisa e desenvolvimento de alimentos de baixa caloria e adoçantes não calóricos. Os edulcorantes podem produzir uma variedade de características sensoriais, dependendo do alimento ou bebida ao qual foi adicionado, sendo necessário estudá-los individualmente. O objetivo deste estudo foi avaliar sensorialmente os cafés expressos adoçados com sacarose e diferentes edulcorantes (sucralose, aspartame, neotame, mistura de ciclamato/sacarina e estévia). A avaliação sensorial foi realizada através da determinação da Doçura Ideal, Teste de Equivalência em Doçura, Análise Descritiva Quantitativa (ADQ®), Análise de Aceitação e Análise Tempo-Intensidade para o estímulo gosto doce. A ADQ, Análise Tempo-Intensidade e Teste de aceitação foram avaliados por análise de variância (ANOVA), teste de Tukey e Análise de Componentes Principais (ACP). A doçura ideal de sacarose no café expresso foi de 12,5%. O teste de equivalência de doçura revelou que a amostra com neotame teve a maior potência edulcorante entre os cinco edulcorantes testados, enquanto que a estévia apresentou o menor poder de dulçor. Os termos descritivos do café expresso com sacarose e adoçantes diferentes foram: cor marrom escuro (bebida café), cor marrom claro (espuma), presença de espuma, viscosidade aparente, aroma de café, aroma doce, aroma de baunilha, aroma torrado, sabor de caramelo, gosto doce, gosto doce residual, gosto amargo, gosto amargo residual, sabor de café, adstringência, sabor torrado e corpo. A ACP sugeriu que a amostra com sacarose foi caracterizada principalmente pelos atributos corpo e viscosidade, a amostra com sucralose pelo aroma doce, a amostra com o aspartame por doçura, a amostra com o neotame por adstringência, a amostra com a mistura de ciclamato/sacarina por amargor e gosto residual amargo e, por fim, a amostra com estévia por gosto doce residual. Os resultados não mostraram uma grande diferença no perfil sensorial das amostras de café expresso, mas a amostra com estévia foi a que apresentou maior variação. As amostras de café expresso foram muito semelhantes em relação à aparência, textura e aroma. O teste de aceitação verificou que a amostra comaspartame apresentou maior aceitação entre os consumidores. Não houve diferença estatística significativa (p> 0,05) em relação aos atributos de aparência, aroma e textura, entre as seis amostras. Abstract: Brazil has the biggest coffee beans production among all countries and is the second consumer market, following USA. The consumption of coffee beverages is growing steadily thanks to their physiological effects and pleasant flavor and aroma. In addition concern about health has given inputs to research and development on low calorie foods and non-caloric sweeteners. Sweeteners can produce a variety of sensory characteristics depending on the food or beverage in which they have been added, therefore it is necessary to study them individually. The objective of this study was to sensorially evaluate espresso coffee sweetened with various sweeteners (sucralose, aspartame, neotame, mixture of cyclamate/saccharin 2:1 and stevioside) and sucrose. The sensory evaluation was done through ideal and equivalent sweetness determinations, Quantitative Descriptive Analysis (QDA), Acceptance Analysis, and Time-Intensity Analysis for stimuli sweet taste. The QDA, time intensity analysis and acceptance test data were evaluated by the analysis of variance (ANOVA), Tukey's Test and the principal component analysis. The statistical analysis was carried out using the SAS program. The ideal sweetness of sucrose in espresso coffee was 12,5%. The equivalent sweetness determinations showed that neotame had the greatest potency amongst the five intensity sweeteners tested, while stevioside exhibited the smallest. The descriptive terms of the coffee espresso with sucrose and different sweeteners were: dark brown color, light brown color, foam, viscosity, coffee aroma, sweet aroma, vanilla aroma, roasted aroma, caramel flavor, sweetness, sweet aftertaste, bitterness, bitter aftertaste, coffee flavor, astringency, roasted flavor and body. The Principal Component Analysis (PCA) suggested that the sample with sucrose was mainly characterized by the body and viscosity, the sample with sucralose by the sweet aroma, the sample with aspartame by the sweetness, the sample with neotame by astringency, the sample with cyclamates/saccharine (2:1) blend by bitterness and bitter aftertaste, and, finally, the sample with stevioside by sweet aftertaste. The results did not show a great difference in the sensory profile of espresso coffee samples, however the sample with stevioside showed more variation. The samples were very similar in relation to appearance, aroma, and texture. The acceptance test showed a greater acceptance for the sample with aspartame. No statistically significant differences (p > 0.05) were observed for appearance, aroma and texture, between all the samples. The Time-Intensity Analysis showed that the sample with stevioside presented highest Imax (maximum intensity), Timax (time in which the maximum intensity was recorded); Area (area of the curve time×intensity) and Ttot (total duration time of the stimulus) of the all samples

Este trabalho estuda a obra de Itamar Assumpção, investigando os principais procedimentos de composição musical utilizados, bem como a relação que possuem com suas vivências pessoais de aprendizagem musical e teatral. Sua obra promove algumas sínteses de materiais musicais e poéticos da tradição popular brasileira e internacional, referencias que são reelaboradas no processo de criação de suas canções. São demonstradas e analisadas a confluência e a atrelagem que existe entre a ação da performance e oito matrizes composicionais identificadas ao longo das diferentes fases de sua obra.
This paper studies the music of Itamar Assumpção, exploring not only the compositional procedures employed, but also their relation to the artist\'s musical and theatrical life experiences. Assumpção\'s work harmonizes musical and poetic material from the Brazilian and international popular tradition, a synthesis that is reconstructed in the process of songwriting. This paper demonstrates and analyzes the confluence and interconnection between the act of performance and eight compositional structures identified throughout different phases of his work.

Abstract:
The formation of viable pollen relies upon a complex interaction of genes in time and space within the anther. One of the most important maternal tissues involved in the production of functional pollen is the tapetum, which is a highly active tissue that plays a major secretory role during pollen development. This project involved the molecular analysis of genes that are expressed in the anther tapetum and are critical for functional pollen development. A number of these are thought to be regulated by, or interact with MALESTERILITY1 (MS1), a transcriptional regulator of male gametogenesis (Yang et al., 2007) or ABORTED MICROSPORE (Xu et al., 2010). Work involved analysis of an ABC transporter (At3g13220), which has been shown to be critical for viable pollen formation and confirmed as directly regulated by AMS (Xu et al., 2010). Another protein, POB2, which appears to be involved in ubiquitin-based proteolytic breakdown, is thought to interact with the MS1 protein. POB2 was identified from a previous screen of a stamen specific yeast-2-hybrid library using the MS1 protein. This interaction has been subsequently confirmed in this work by further yeast two hybrid analyses and bifunctional fluorescent complementation. Further work involved verification of this interaction in vitro and in planta by pull-downs and transient expression of proteins in E. coli and Nicotiana benthamiana respectively. Other work focused on identifying factors that regulate MS1 expression; this identified novel male sterile mutants derived from screening fast neutron mutagenised seed carrying the MS1Prom:MS1-GFP functional fusion protein. Microscopic observation of the fluorescent reporter showed changes in the stage specific expression of MS1 in some of these mutants. Backcrossing of the male sterile mutants with the parental plants (carrying the MS1Prom:MS1-GFP fusion construct) and the ms1 mutant confirmed one as a new mutant and the other three as being allelic to the ms1 mutation. Gene mapping of this mutant was subsequently conducted and suggest that it may be located on chromosome 3. These results are providing insight into the regulatory network of MS1 and AMS during anther development.

Obesity is a complex and chronic disease that is associated with many complications including type 2 diabetes (T2D). However, the mechanism leading to these events is unclear. To determine the role played by obesity in the development of T2D, it was purposed to examine serum protein profiles of diet-induced obese Wistar rats. These protein(s) will be used as potential biomarkers for early detection, diagnosis, as target for drug discovery and hopefully treatment of obesity and obesity-induced disorders. An animal model of obesity-induced T2D was established by feeding male Wistar rats a diet with a high fat content for 44 weeks. Weight changes and food intake were monitored weekly during the diet phase. Fasting blood glucose levels were measured while blood was collected for serumpreparation every second week for the first eight weeks; using these parameters, a model for the identification of diet-induced obese rats was established. Serum protein profiles were then compared between the two groups using 2D-PAGE analysis coupled to MALDI-MS at termination of the study. Several proteins showed differences in their expression when compared between the low fat (LF) and high fat (HF) groups. However, only proteins that showed expression that was either two fold low or high between the two groups were considered to be differentially expressed. High fat fed rats weighed significantly heavier starting at the fourth week on diet and throughout the study, their glucose homeostasis and serum protein expression was altered. Three protein spots were identified as dysregulated in the HF group, where Apolipoprotein-AIV was found to have been up-regulated whereas C-reactive protein and Fetuin-A were down-regulated. These proteins might help in the understanding of the mechanism(s)that underlie the pathogenesis of obesity and its related disorders. However, further studies are required to determine the relationship between these proteins, obesity development and its comorbidities.

Abstract:
The avocado mesocarp contains up to 60-70% oil by dry weight where triacylglycerol (TAG) is the major constituent. There is significant human nutritional demand for vegetable oil, but its use in production of renewable biomaterials and fuels has intensified the need to further increase oil production. In plants, the final and committed step in TAG biosynthesis is catalyzed by diacylglycerol acyltransferases (DGAT) and/or a phospholipid: diacylglycerol acyltransferases (PDAT). Both DGAT and PDAT contribute to TAG biosynthesis in an independent or overlapping manner, depending on the species. However, preferred pathway for TAG biosynthesis is not well studied in nonseed tissues such as mesocarp. Based on the transcriptome data of Persea americana it is hypothesized that both DGAT and PDAT are likely to catalyze the conversion of diacylglycerol to TAG. In this study, putative DGAT1 was identified and comprehensive in silico analyses were conducted to determine the respective start codons, full-length coding sequences, transmembrane domains, predicted protein structures and phylogenetic relationships with other known DGAT1s. These data reveal that the putative DGAT1 of a basal angiosperm species retain features that are conserved not only among angiosperms but also other eukaryotes. For further functional analysis, the avocado DGAT1 was expressed in H1246, a TAG-deficient yeast strain and lipotoxicity rescue assays, TLC analysis, Nile Red staining were conducted. The complementation of this yeast strain confirmed enzyme activity and supported the possible role of avocado DGAT1 in TAG biosynthesis. Finally, substrate specificity of DGAT was determined by incubating microsomes with different radiolabeled substances and found that avocado DGAT1 has a preference toward oleic acid (18:1) compared to palmitic acid (16:0) while it is converting diacylglycerol (DAG) to triacylglycerol. In summary, we characterized functional DGAT1 in a basal angiosperm species, which may be metabolically engineered into crop species to produce TAG enriched in oleic acid.

Abstract:
The recent increase in availability of next generation sequencing methodologies has led to extensive analysis of the genome of Clostridium difficile. In contrast, protein expression analysis, crucial to the elucidation of mechanisms of disease, has severely lagged behind. In this study, in-depth proteomic analysis of three strains of varying virulence, demonstrated previously in an animal model, has been undertaken against a background of the sequenced genomes. Strain B-1 is a historic, virulent, ribotype 005 clone, strain A represents the emerging hypervirulent 027 ribotype, while strain Tra5/5, ribotype 001, is of low virulence. To undertake a comprehensive overview of the expressed proteome, both 1D and 2D gel electrophoresis were used to separate and display the protein content of each isolate. This was coupled to MALDI-TOF and LC-MS/MS mass spectrometry for protein identification. A total of 888 different proteins were characterised by comparative analysis of isolates grown in parallel for 64 hours on blood agar. Of these, only 38% were shared between all isolates. An additional 350, 243 and 398 proteins were detected from broth cultures, and the use of a hexapeptide bead library, designed to capture low abundance proteins, led to the detection of a further 148, 127, and 171 proteins in strains A, B-1 and Tra5/5 respectively. Relative differential expression was investigated using Differential In Gel Electrophoresis (DIGE), and five proteins were shown to have a statistically higher concentration in strain A, twelve in strain B-1 and eight in strain Tra5/5. A number of these were surface proteins, with selected S-layer proteins found to be up-regulated in each strain, and the flagellar protein, FliC, up-regulated in both A and B-1. Furthermore, differential post-translation modification events were seen in flagellar and S-layer proteins. In-vivo expression of these proteins was mapped using Western blotting. Immunodetection of the majority of these, including FliC and the high molecular weight S-layer protein, were conserved between the three strains, but a notable series of immunoreactive protein spots were present in strains A and Tra5/5 but not B-1, most likely corresponding to an additional S-layer protein present in the genomes these strains, but not that of B-1. Protein expression differences for a number of previously proposed virulence proteins were evident between strains, including toxin B, sporulation, flagella and the S-layer proteins, metabolic enzymes, stress response proteins and ABC transporters. This study strongly supports the view that the virulence of Clostridium difficile is multifactorial, and that a number of related factors, although not directly required for pathogenicity, may serve to modulate the virulence of individual strains.

Resumo: Arranjos de DNA são uma poderosa técnica de monitoramento da expressão gênica em larga escala. No entanto, a grande quantidade de dados gerados com esse tipo de experimento requer um tratamento estatístico adequado às suas características. Uma aplicação importante dos arranjos de DNA é a identificação de genes diferencialmente expressos em diferentes amostras de RNA. Essa seleção demanda testes estatísticos apurados, capazes de distinguir, entre o grande número de genes usualmente presentes nos arranjos, aqueles cuja expressão é significativamente diferenciada. Neste trabalho nós desenvolvemos algoritmos para a análise estatística dos dados provenientes de arranjos de DNA, eficientes em lidar com os problemas usuais nesse tipo de dados, como o número limitado de réplicas. Aplicados a dados simulados, os algoritmos desenvolvidos mostraram-se competitivos com outros métodos de análises descritos na literatura, superando-os em algumas situações. A aplicação desses algoritmos foi também demonstrada em um experimento voltado à identificação de genes de cana-deaçúcar diferencialmente expressos em resposta a deficiência de fosfato. O fósforo é um macronutriente essencial, captado pelas plantas principalmente na forma de fosfato inorgânico (Pi). A deficiência de fosfato é freqüente na natureza, especialmente nos solos ácidos das regiões tropicais e subtropicais. Devido a grande importância econômica da cana-de-açúcar, a identificação de genes diferencialmente expressos em resposta à deficiência de fosfato nesta espécie é de grande interesse científico e agronômico. Algoritmos de agrupamento foram também aplicados aos dados de expressão obtidos no experimento, identificando padrões de expressão gênica nos diferentes estágios da resposta da cana a esse estresse e proporcionando assim uma caracterização adicional da mesma. Futuramente, os resultados desse trabalho podem conduzir ao desenvolvimento de linhagens de cana-de-açúcar com melhor desempenho em solos pobres de fosfato, o que seria de extremo interesse agronômico
Abstract: DNA arrays are a powerful technique for monitoring gene expression in large scale. However, the great amount of data generated by this kind of experiment requires a statistical treatment adequate to its characteristics. An important application of DNA arrays is the identification of differentially expressed genes in different RNA samples. This selection demands refined statistical tests, able of distinguish among the great number of genes usually present in the arrays those which expression is significantly different. In this work, we have developed algorithms for the analysis of DNA array data, efficient in handling the usual problems in this kind of data, as the limited number of replicates. When applied to data simulations the developed algorithms showed to be competitive with other methods of analysis described in the literature and widely used, overperforming them in some situations. The application of these algorithms was also demonstrated in an experiment devoted to the identification of sugarcane genes differentially expressed in response to phosphate deficiency. Phosphorous is an essential macronutrient, absorbed by plants mostly in the form of inorganic phosphate (Pi). The phosphate deficiency is frequent in the nature, especially in the acid soils of tropical and subtropical areas. Because of the great economical importance of sugarcane, the identification of genes that are differentially expressed in response to phosphate deficiency in this species is of great scientific and agronomic interest. Clustering algorithms were also applied to the expression data obtained in the experiment, identifying patterns of gene expression in the different stages of the sugarcane response to the stress, thus providing an additional characterization of it. In the future, the results of this work can lead to the development of sugarcane cultivars that have better performance in phosphate deficient soils, what would be of great agronomic interest

Abstract:
This thesis provides an in-depth analyze of expressed sequence tags (EST) that represent pieces of eukaryotic mRNA by using unsupervised hidden Markov model (HMM). ESTs are short nucleotide sequences that are used primarily for rapid identificationof new genes with potential coding regions (CDS). ESTs are made by sequencing on double-stranded cDNA and the synthesizedESTs are stored in digital form, usually in FASTA format. Since sequencing is often randomized and that parts of mRNA contain non-coding regions, some ESTs will not represent CDS.It is desired to remove these unwanted ESTs if the purpose is to identifygenes associated with CDS. Application of stochastic HMM allow identification of region contents in a EST. Softwares like ESTScanuse HMM in which a training of the HMM is done by supervised learning with annotated data. However, because there are not always annotated data at hand this thesis focus on the ability to train an HMM with unsupervised learning on data containing ESTs, both with and without CDS. But the data used for training is not annotated, i.e. the regions that an EST consists of are unknown. In this thesis a new HMM is introduced where the parameters of the HMM are in focus so that they are reasonablyconsistent with biologically important regionsof an mRNA such as the Kozak sequence, poly(A)-signals and poly(A)-tails to guide the training and decoding correctly with ESTs to proper statesin the HMM. Transition probabilities in the HMMhas been adapted so that it represents the mean length and distribution of the different regions in mRNA. Testing of the HMM's specificity and sensitivityhave been performed via BLAST by blasting each EST and compare the BLAST results with the HMM prediction results.A regression analysis test shows that the length of ESTs used when training the HMM is significantly important, the longer the better. The final resultsshows that it is possible to train an HMM with unsupervised machine learning but to be comparable to supervised machine learning as ESTScan, further expansion of the HMM is necessary such as frame-shift correction of ESTs byimproving the HMM's ability to choose correctly positioned start codons or nucleotides. Usually the false positive results are because of incorrectly positioned start codons leadingto too short CDS lengths. Since no frame-shift correction is implemented, short predicted CDS lengths are not acceptable and is hence not counted as coding regionsduring prediction. However, when there is a lack of supervised models then unsupervised HMM is a potential replacement with stable performance and able to be adapted forany eukaryotic organism.

BACKGROUND: Elucidation of candidate colorectal cancer biomarkers often begins by comparing the expression profiles of cancerous and normal tissue by performing high throughput gene expression profiling. While many such studies have been performed, the resulting lists of differentially expressed genes tend to be inconsistent with each other, suggesting that there are some false positives and negatives. One logical solution to this problem is to determine the intersection of the lists of differentially expressed genes from independent studies. It is expected that genes that are biologically relevant to cancer tumorigenesis will be reported most often, while sporadically reported genes are due to the inherent biases and limitations of each of the profiling platforms used. However, the statistical significance of the observed intersection among many independent studies is usually not considered. PURPOSE: To address these issues, we developed a computational meta-analysis method that ranked differentially expressed genes based on the following criteria, which are presented in order of importance: the amount of intersection among studies, total tissue sample sizes, and average fold change in expression. We applied this meta-analysis method to 25 independent colorectal cancer profiling studies that compared cancer versus normal, adenoma versus normal, and cancer versus adenoma tissues. RESULTS: We observed that some genes were consistently reported as differentially expressed with a statistically significant frequency (P <.0001) in the cancer versus normal and adenoma versus normal comparisons, but not in the cancer versus adenoma comparison. We performed a review of some of the high ranking candidates and determined that some have previously been shown to have diagnostic and/or prognostic utility in colorectal cancer. More interestingly, the meta-analysis method also identified genes that had yet to be tested and validated as biomarkers. Thus, these candidates are currently being validated at the protein level on colorectal tissue microarrays. CONCLUSION: Our meta-analysis method identified genes that were consistently reported as differentially expressed. Besides identifying new biomarker candidates, our meta-analysis method also provides another filter to remove false positive genes from further consideration. In conclusion, the genes presented here will aid in the identification of highly sensitive and specific biomarkers in colorectal cancer.

Abstract:
Increasing evidence indicates a shift is occurring in the distribution of lung cancer subtypes, with relative and absolute increases in the number of adenocarcinomas. The exact reason for this remains unclear. Molecular and cytogenetic studies have identified several genetic changes occurring in the development of pulmonary adenocarcinoma but many more remain unknown. Until recently, identification of unknown genes relied heavily on the use of linkage-based studies, which are both laborious and time-consuming. Recently, a new technique has been developed to investigate gene expression with greater accuracy and experimental ease: differential display reverse transcription polymerase chain reaction (DDRT-PCR).;In this study DDRT-PCR has been applied to search for novel, differentially expressed genes in human pulmonary adenocarcinoma, using both human normal and adenocarcinoma cell lines and matched surgical tissues. Human cell lines were used in preliminary experiments to adapt and establish the experimental techniques. Results from these studies produced three differentially expressed bands, one of which was a true positive. This cDNA tag was found to show 100% sequence homology to human fibronectin mRNA. Fibronectin is known to show different patterns of expression in various tumours. Similar studies were set up using human surgical specimens collected from two major thoracic units. Results from one matched human tissue sample identified eight differentially expressed bands. Again only one of these demonstrated true differential expression with a northern blot.;Expression of the tissue-positive band, called H8, was diminished in pulmonary adenocarcinoma RNA samples and may represent a novel tumour suppressor gene. The sequence data that has been obtained from the H8 cDNA tag appears to correspond to the 3' untranslated region. Northern blotting studies revealed high levels of H8 in normal lung tissue matched to other types of lung tumour, normal liver, stomach and colon, with lower expression in the corresponding tumour samples.
