Abstract:
Työn tavoitteena oli parantaa tuotannonohjausta ja ohjattavuutta, sekä vähentää tuottamattoman työn määrää kokoonpanossa. Suurimpana ongelmana tuotannonohjauksen ja ohjattavuuden kehittämisessä yrityksen tapauksessa voidaan pitää hajautettuun tietojärjestelmään liitettäviä ongelmia. Tarvittava tieto on hankalasti löydettävissä ja sen takia myös hankalasti hyödynnettävissä. Yrityksellä on käytössään monia irrallisia tietojärjestelmiä, joiden välillä tieto ei kulje järjestelmästä toiseen, kuten ideaalitilanteessa olisi tarkoitus. Uusien keskitettyä tiedonhallintaa hyödyntävien tietojärjestelmien käyttöönottaminen on monivuotuinen prosessi. Tämän vuoksi diplomityössä keskityttiin tekemään konkreettisia parannuksia, joita voidaan tehdä työn puitteissa lyhyelläkin aikavälillä. Työn tutkimusstrategiaksi valittiin kirjallisuustutkimus, joka kattoi rajallisen tutkimusotteen. Ratkaisua yrityksen tuotannonohjattavuusongelmaan lähdettiin hakemaan teoriaosuudessa käsiteltyjen vaihtoehtojen kriittisellä vertailulla yrityksen nykytilanteeseen. Yrityksen nykytilannekartoituksen ja pitkän tähtäimen kehityssuunnitelmat huomioon ottaen lähdettiin työssä hakemaan lyhyellä aikavälillä tehtäviä mahdollisia toimenpiteitä, jotka tukevat yrityksen pitkäntähtäimen suunnitelmaa mahdollisimman hyvin. Konkreettisena toimenpiteenä tuotannonohjauksen parantamiseksi uudistetaan tuotannon FLOW-järjestelmä. FLOW-järjestelmään tuodaan mahdollisuuksien mukaan kaupallisille MES-järjestelmille (Manufacturing Execution System) tyypillisiä ominaisuuksia. Tuotannonohjattavuuden parantamiseksi suunniteltiin yrityksen ETO (Engineering to Order) eli tilausohjautuvaan suunnitteluun perustuvaan kokoonpanotuotantoon räätälöidyt tuotannon mittarit. Tuotannon mittareiden toteutus vaatii datan keräystavan ja käyttöliittymän muuttamista mittareille sopiviksi. Tuotannon käyttöliittymä suunniteltiin uudestaan. Käyttöliittymään suunniteltiin tarvittavia leimauskohtia eri työvaiheisiin liittyen, ja käyttöliittymään suunniteltiin parannuksia, joiden avulla onnistutaan vähentämään tuottamattoman työn määrää. Käytännössä parannuksilla tarkoitetaan työskentelyyn tarvittavan tiedon löytymiseen tehtäviä parannuksia, sekä yleisiä muita parannuksia, joilla vähennetään työntekijän aikaisemmin monimutkaisina pidettyjen toimenpiteiden tekemistä. Käyttöliittymään linkitetään muun muassa suora pääsy kommunikointikeskuksen aloitussivulle, josta työntekijä voi helposti tehdä turvahavaintoja ja kehitysehdotuksia, sekä muita aiheeseen liittyviä toimenpiteitä. Tuotannon käyttöliittymän ja tuotannon mittareiden suunnittelussa onnistuttiin loistavasti. Yleisesti tehtyihin toimintatapamuutoksiin ollaan monesti vastahakoisia, mutta yrityksen tapauksessa myös tuotannon lattiatason työntekijät suhtautuivat hankkeeseen positiivisesti. Positiiviseen suhtautumiseen vaikutti todennäköisesti monen asian summa, mutta yhtenä tekijänä voidaan pitää tuotannon työntekijöiden mukaan ottamista kehityshankkeeseen. Suunnitellusta uudesta käyttöliittymästä saatiin siisti, havainnollinen ja moderninnäköinen. Lisäksi käyttöliittymään suunniteltuihin parannuksiin oltiin yleisesti tyytyväisiä ja niillä pystytään helpottamaan päivittäistä työskentelyä. Käyttöliittymällä saadaan kerättyä tarvittava tieto mittareiden muodostamiseksi. Turhat aiemmin tehdyt merkinnät, jotka eivät tuo mittaukseen lisäarvoa, poistetaan käyttöliittymästä. Tuotantoon tuotuihin tabletteihin ja näyttöihin ollaan tyytyväisiä, ja niiden koetaan helpottavan työskentelyä ja työohjeiden lukemista. Käyttöön otettavia mittareita voidaan tehtyjen toimenpiteiden myötä esittää näytöissä sähköisessä muodossa ja mittareiden avulla voidaan muodostaa yhteenveto jokaisen projektin läpiviennin onnistumisesta. FLOW-järjestelmään tehtävien muutoksien suunnittelussa onnistuttiin hyvin. Tehtävien kehitystoimien myötä FLOW-järjestelmä vastaa ominaisuuksiltaan yhä enemmässä määrin kaupallisia MES-järjestelmiä
The aim of this master’s thesis was to improve production management and controllability and also try to reduce nonproductive work in assembly. The biggest problems in trying to improve the company’s production management and control are related to the decentralized information management system. The needed data can be difficult to find and utilize. Presently the company is using many data systems that do not communicate with each other. Employing a new centralized information management system is a time consuming process. Therefore, the focus of this thesis is on practical improvement that can be realized in the short term. The research strategy was chosen the literature survey, which was taken into account a limited group of materials and surveys. Production management problems were tackled by comparing theoretical alternatives to current practices. As a result the production system in use, FLOW, was redesigned with an objective to integrate features of commercial manufacturing execution systems into it. Key performance indicators (KPI) were customized for engineered-to-order (ETO) assembly work with an aim to improve production control. Implementation of the new KPI’s required changes to the user interface, and to the data gathering. While planning the user interface improvements were made to include checklists for assembly and to reduce unprofitable work. In practice the improvements help in finding needed information and reduce the amount of tasks previously seen as complicated. In addition, a direct link from the user interface to the communication center webpage enables the user to give feedback and report safety observations or continuous improvement ideas. Planning the new user interface and the KPI’s was a success. Often reforms and improvements of working practices are received reluctantly. However, in this case also the assembly workers had a positive attitude towards the project. Involving the workforce in developing their own work was likely one of the reasons for this. The look of the newly designed user interface is clear, illustrative and modern. Users were generally satisfied with the improvements which were seen to facilitate daily work. All the data required by the KPI’s can now be gathered while previous unnecessarily fed entries have been removed. Workers on the shop floor are satisfied with the tablet computers and displays as they assist in performing work tasks and reading instructions. The new KPI’s will be easy to display on the shop floor in real time. It will also be possible to summarize how well projects have succeeded, and highlight problems. We make the grade with the development of FLOW. Its features increasingly resemble those of commercial MES-systems

Abstract:
Customization is an important way for suppliers to offer value to customers and to be competitive. There is a variety of methods suppliers can adopt to offer customization. What they have in common is that some form of interaction between customer and supplier is required as customization is based on involving individual customers in specifying a solution to be produced. This can be achieved, for example, by allowing the customer to choose from components or values that have been defined in advance, later to be assembled or put together according to the customer's wishes. As the possibilities for customization are clearly defined in advance, the supplier can rationalize their procedure to capture customer needs and propose an appropriate solution. This differs from cases where the customer is already involved in the development, design, or engineering stage ± so-called engineer-to-order (ETO) scenarios. Here, the customer is not bound to predefined possibilities for customization, which means that the interaction required to define the solution can extend beyond that required when customization possibilities are predefined and thus limited. In this thesis, customization-based interaction in ETO is investigated, both with the intention of improving the understanding of interaction in this context as such but also to offer ways of explaining how some approaches to customization have implications for interaction that differ from others. The result of the research consists of an account of how interaction is manifested in ETO and how different approaches to customization can be understood to clarify their implications for how solutions are defined.
Kundanpassning är ett viktigt sätt för leverantörer att erbjuda värde till sina kunder och på så vis vara konkurrenskraftiga. Det finns en rad olika metoder som leverantörer kan anta för att erbjuda kundanpassning. Gemensamt är att någon form av interaktion mellan kund och leverantör förutsätts, då kundanpassning baseras på att involvera den enskilda kunden i att specificera den lösning som ska produceras. Det kan exempelvis göras genom att kunden tillåts välja mellan olika komponenter eller värden som definierats i förväg, som därefter monteras eller sätts samman enligt kundens önskemål. Eftersom möjligheterna till kundanpassning är tydligt definierade i förväg kan leverantören därför rationalisera deras procedur för att fånga upp kundkrav och föreslå en lämplig lösning. Denna metod skiljer sig från fall då kunden involveras redan i utvecklings, design, eller konstruktionsfasen, så kallade konstruktion-mot-order (engineer-to-order, ETO) scenarion. Kunden är i dessa fall inte bunden till fördefinierade möjligheter för kundanpassning, vilket gör att interaktionen som krävs för att definiera lösningen inte kan rationaliseras i samma utsträckning. I denna uppsats undersöks kundanpassningsbaserad interaktion i ETO, både med avsikt att förbättra förståelsen för interaktion i denna kontext som sådan men även för att erbjuda sätt att förklara hur vissa förhållningssätt till kundanpassning har implikationer för interaktion som skiljer sig från andra. Resultatet av forskningen utgörs av en redogörelse för hur interaktion tar sitt uttryck i ETO samt hur olika förhållningssätt till kundanpassning kan förstås för att tydliggöra deras innebörd för hur lösningar definieras.

Today’s manufacturing industry is characterized by being highly competitive and drives companies to strive for more efficient product and production system development. One way of achieving this, is by continuous improvement of processes. One important part of lowering waste is by an efficient  production planning. A competitive production system can improve product quality, decrease costs, and improve the general efficiency. Engineering-to-order (ETO) systems face a challenge due to the difficulty to attain an efficient production planning. The difficulty strives from the complex character of the manufacturing processes and the uniqueness of product properties, which makes a good standardization work a challenge. ETO companies often have high reliance on manual-labor with manual data acquisition, which makes it hard to perform effective production planning with high accuracy. One common technique for executing production planning in the ETO-industry represent a multivariate regression analysis     The thesis includes a case study that examines how regression analysis used for production planning within a production of an ETO company can be improved and what effects it has on processes and departments within the company. The problem was addressed by a mixed method approach, qualitative data were collected from semi-structured interviews, observations, and a thorough literature review and quantitative data were mainly collected through document studies. The study shows that regression analysis is suitable for production planning in the ETO industry while emphasizing the importance of high-quality data, partly through implementation of standardizations. The updated model improves main processes in production which in turn gives rise to improvements in support processes performed by other departments.

The American military experience in the European Theater of Operations during the Second World War is one of the most heavily documented topics in modern historiography. However, within this plethora of scholarship, very little has been written on the contributions of the American corps cavalry to the operational success of the Allied forces. The 13 mechanized cavalry groups deployed by the U.S. Army served in a variety of roles, conducting screens, counter-reconnaissance, as well as a number of other associated security missions for their parent corps and armies. Although unheralded, these groups made substantial and war-altering impacts for the U.S. Army.

The competition in the manufacturing industry has never been higher. Owing to the technological changes and advancements in the market, readily available data is no longer a thing of the past. Numerous studies have discussed the impact of industry 4.0, digital transformation as well as better production planning methods in the manufacturing industry.  The Mass-Manufacturing industry, in specific, has gained efficiency levels in production that were previously unimaginable. Industry 4.0 has been discussed as the ‘next big thing’ in the manufacturing context. In fact, it is seen as a necessity for manufacturing companies to stay competitive. However, efficient production planning methodologies are a preliminary requirement in order to successfully adopt the new manufacturing paradigms. The Engineering-to-order (ETO) industry is still widely unexplored by the academia ETO industries, barely have any production planning methodologies to rely on owing to their complex production processes and high reliance on manual-labour. Regression techniques have repeatedly been used in the production planning context. Considering its statistical prowess, it is no surprise that even the newer machine-learning techniques are based on regression. Considering its success in the mass-manufacturing industry for production planning, is it possible that its usage in the ETO industry might lead to the same results? This thesis involves a case study that was performed at an electrical transformer manufacturing plant in Sweden. After understanding the several operations that are performed in the production process, regression techniques are employed to estimate man-hours. The results from the study reconfirm the statistical prowess of regression and show the possibility of using regression in order to estimate man-hours in the ETO industry. In addition, several factors that can affect successful adoption of this tool in the production planning context are discussed. It is hoped that this study will lay the foundation for better production planning methodologies for the ETO industries in the future which might subsequently result in more data-driven decision making rather than instincts.

Abstract:
ERP systems are useful for MTS and ATO manufacturing strategies. When evaluating the ERP systems in an ETO environment there is not much research available on the matter, but the general understanding is that ERP system lacks specific business processes that are special to companies in the sector. In addition, the few studies that are available on the subject conclude that there is need for more empirical studies. By conducting a thorough literature study on the topics ERP, ETO, and project life-cycle processes, information was gathered and used for creating a survey and interview questions. All the questions were based on how companies handled the business processes that were considered critical in an ERP solution for ETO companies. The companies that participated in the case study were in the maritime and offshore ETO segment and they were currently using the ERP software Microsoft Dynamics AX. The results from the surveys are a contribution to the empirical study this specific area of research needs. During the evaluation of the current solutions there were areas where AX was not sufficient at all, areas where AX needed extra functionality, and areas where AX was sufficient. Where AX was not performing up to standards, new software had been implemented and most often integrated with AX. Industry specific requirements had also been included as new, customizable functionality in AX. This extra functionality was very important because of the focus on a specific industry. In conclusion, the findings from the case studies were used to generate requirements that represented real needs to be fulfilled by an eventual solution. The general solution was therefore based on a combination of “best-practice” solutions from all three cases. Benefits of the solution were also identified.

Sharing Knowledge is considered an important part of managing new product development (NPD) research on the process of NPD and Knowledge Management methods have influenced industry in various ways. For example the management of the NPD process, the use of tools, techniques and the organisation of teams, and the integration of the marketing and manufacturing have resulted in considerable progress within NPD process. Prior studies on the NPD problems have delivered various models of the NPD process and a variety of supporting methods, tools and techniques in a generic context. A more realistic scenario however, is to consider the needs of firms that develop products on a Make-to-Order (MTO) or Engineer-to-Order (ETO) basis. The research methodology adopted was based on extracting a preliminary ETO model supported by variety of Knowledge Management methods, tools and techniques from the review of literature. To examine the applicability of these models and methods and also the influential factors on the NPD process a survey by questionnaire and structured interviews in UK industrial companies was carried out. Findings were bound together to provide a generic model of the ETO process and a framework for the knowledge sharing on the specific needs of ETO manufacturing companies. IDEFO technique was used to develop the preliminary and the generic models. The objective of this research is to construct a structured and practical framework for supporting the opportunity for knowledge sharing within one-off projects. The knowledge sharing framework referred to as 'Sharing-ETO-Knowledge' (SETOK) was translated into a computer program using the "MS Visio' enterprise modelling systems. It was examined by applying the system program to the data of the two cases that had been obtained at the case study stage. The framework has been fruitful in the provision of a guideline for the implementation of the knowledge sharing in various NPD-ETO projects. The SETOK framework may be viewed as a practical, robust generic tool to assess the process performance of ETO manufacturing projects. The outcome of this study would help ETO manufacturing companies in their knowledge sharing and decision making processes with regards to NPD-ETO manufacturing projects.

Abstract:
Advancements in the power electronics systems have been directly related to the availability of improved power semiconductor devices. The device performance greatly determines the efficiency, reliability, volume, and cost of the power electronics system. This dissertation is dedicated to develop an advanced high power semiconductor device, the emitter turn-off (ETO) thyristor, which is targeted to improve the limitations of the present high power devices. Major improvements in electrical and mechanical designs of the ETO for high power and high frequency operation are proposed which result in improved snubberless turn-off capability, low conduction loss, and low gate drive power consumption of the new generation ETO. A revolutionary self-power generation method of the ETO is proposed. Different from the conventional high power devices which require the external power input for their gate drivers, ETO achieves complete optically controlled turn-on and turn-off and all the internal power required is self-generated. This advancement will have a major impact to high power converter designs. A novel integrated method to eliminate the dead-time requirement is proposed for ETO. This method not only improves the output waveform quality but also increases the reliability and reduces the cost of the high power PWM voltage source converters. With this unique function, the upper and the lower ETO's within a converter phase leg can receive the ideal complementary (without dead-time) PWM signals and solve shoot-through problems. Method to measure the ETO current and transfer the current information to a PWM signal is proposed. Based on the ETO's built-in current sensor, the over-current protection function of the ETO is designed as well. The experimental results show that the built-in current sensor has a very high precision, and the over-current protection function can effectively protect the ETO during the short circuit faults. In order to improve ETO's turn-off capability, a comprehensive investigation of the turn-off failure mechanism of the ETO was performed. A series of simulations and experiments are carried out to study the ETO turn-off operation. The detail turn-off failure mechanisms are presented. The conditions to cause the ETO failure are addressed. The approaches to improve the ETO's turn-off capability are discussed.

Abstract:
Lean management is considered a key to lead time reduction while being efficient, in order to survive in the actual market. Despite this, there is relatively little research done about implementation of lean management in ETO processes. Knowing that ETO processes are a reality in the industry, because there is a demand of customized products in the market. Nonetheless, the early phases of the ETO processes are critical. The purpose of this study was therefore to investigate how lean management can be used to understand and early phases of ETO processes, in order to evaluate potential lead time reduction. Analysing issues that arise when implementing lean in this kind of processes, such as variety, variation and standardization. The empirical data was collected from two different companies with ETO processes on different stages of lean implementation. A theoretical background in the area is supported by a comparison of the historical development and current state of two companies. The access to these two companies provides a view of the state before lean implementation and during implementation, and also a brief view of the effects. This helps to understand how lean management could be implemented in ETO processes. As a result, this research describes what should be considered when implementing lean management in early phases of ETO processes and what the effect of this implementation could be.

Abstract:
The AM L1/ETO fusion protein in t(8;21) acute myeloid leukaemia is not sufficient for leukaemogenesis, but requires the acquisition of additional co-operating mutations. The "mutator phenotype" model of cancer suggests that the normal mutation rate in human somatic cells is not sufficient to mediate transformation, and that initiating mutations that increase the mutation rate are an essential "driver." As such, the major hypothesis of this thesis is that AML1/ETO confers a mutator phenotype and predisposes cells to the acquisition of co-operating mutations. Using cell lines transduced with the AML1/ETO fusion gene, investigations described within this thesis show that AML1/ETO confers a mutator phenotype, predisposing cells to acquisition of mutations at the TK and PIGA gene loci. This increase in mutation frequency in AML1/ETO-transduced cells is observed both spontaneously and after treatment with genotoxic agents, such as ionising radiation or the chemotherapy agent doxorubicin. Disruption of carefully controlled DNA repair mechanisms is well described to increase propensity to the acquisition of mutations. The effect of AML1/ETO on DNA repair in ectopic AML1/ETO-positive cell line models was investigated revealing dysregulated expression of several DNA repair-associated genes. The DNA base-excision repair pathway was notably affected, including downregulation of the OGGl DNA glycosylase, which is primarily responsible for the repair of 7,8-dihydro-8-oxoguanine lesions in DNA. The role of AML1/ETO in modulating angiogenic regulatory factors, was also investigated in ectopic AML1/ETO-transduced cell lines and t(8;21)-positive patient-derived cell lines. AML1/ETO was found to substantially upregulate the angiogenic regulatory factor ANGPTl. The study of dysregulated angiogenesis in leukaemia is a rapidly expanding field of research, and the investigations presented on AML1/ETO and ANGPT1, suggest that ANGPTl may be a possible therapeutic target in t(8;21) AML. The work presented in this thesis has provided new information as to how the AML1/ETO fusion protein promotes the acquisition of co-operating mutations leading to leukaemic transformation, and identified a 'possible role for AML1/ETO as a regulator of angiogenesis. This information could provide the basis for development of new methods of therapeutic intervention in t(8;21) AML.

The mutation of transcription factor genes is a main cause for acute myeloid leukaemia. RUNX1/ETO, the product of the t(8;21) chromosomal translocation, subverts normal blood cell development by impairing myeloid differentiation. RUNX1/ETO knockdown alleviates this block, with a global reprogramming of transcription factor binding and initiation of myeloid differentiation. Co-depletion of the myeloid transcription factor C/EBPα with RUNX1/ETO suppressed this differentiation response. Furthermore, C/EBPα overexpression largely phenocopied the effect of RUNX1/ETO knockdown. Our data show that low levels of C/EBPα are critical to the maintenance of t(8;21) AML and that C/EBPα drives the response to RUNX1/ETO depletion. To examine how changes in transcription factor binding impact on the activity of cisregulatory elements we mapped genome wide promoter-distal-element interactions in a t(8;21) AML cell line, via Capture HiC, and found that hundreds of interactions were altered by RUNX1/ETO knockdown. Differentially interacting elements exhibited changes in C/EBPα binding and were enriched for the CTCF motif. Our results demonstrate that the presence or absence of RUNX1/ETO has a profound impact on the intra-nuclear organisation of t(8;21) AML cells, and indicate which transcription factors are driving these changes. This work provides a novel mechanism for the RUNX1/ETO mediated differentiation block in t(8;21) AML.

Abstract:

This dissertation aims in part to redress the shortage of close readings of Vladimir Mayakovsky and Aleksandr Rodchenko's joint project, the book Pro Eto. It explores the relationship between the book's visual and verbal aspects, treating the book and its images as objects that repay attentive looking and careful analysis. By these means this dissertation finds that the images do not simply illustrate the text, but have an intertextual relationship with it: sometimes the images suggest their own, alternative narrative, offering scenes that do not exist in the poem; sometimes they act as literary criticism, suggesting interpretations, supplying biographical information, and highlighting with their own form aspects of the poem's.

This analysis reveals Pro Eto's strong links with distant forms of art and literature. The poem's intricate ties to the book of Genesis and Victor Shklovsky's novel Zoo, written while the former literary critic was in exile in Berlin, evince an ambivalence about the manifestations of socialism in early-1920s Russia that is missing from much of Mayakovsky's work. At the same time Rodchenko's images, with their repeated references to Byzantine icons and Dadaist photomontage, expand the poem's scope and its concerns far beyond NEP-era Moscow. Thus my analysis finds that although Pro Eto is considered to be an emblematic Constructivist work, many of the received ideas about Russian Constructivism—the unswerving zeal of its practitioners, the utility of its production, and in particular the ideology-driven, sui-generis nature of the movement itself—are not supported by the book. Pro Eto's deep connections with art and literature outside of Bolshevik Russia contradict the idea—first set out by the Constructivists themselves and widely accepted by subsequent scholars—of Constructivism as an autochthonous movement, born of theory, and indebted neither to historical art movements nor to contemporary western ones. My analysis suggests that reading Pro Eto through the lens of Constructivist theory denies the work the richness, ambivalence and humor it gains when that theory is understood as being in conversation with artistic practice, rather than defining it.

Background: Over the past years, companies have competed in a globalized and dynamic environment which has forced them to implement various incentives in order to gain cost advantages. Companies have tried to implement Sales and Operations Planning (S&OP) but many have not managed to get the expected results. Coordinating sales and operations remains a crucial challenge because of the multiple differences and conflicts in objectives and responsibilities, referred to as having a silo culture. As many companies have encountered this barrier in conjunction with an S&OP implementation, investigating the S&OP process in a company with functional silos and a complex planning process remains.  Objectives: The objective with this study is to provide a deeper understanding of how to manage the silo culture challenge, in order to achieve a higher S&OP maturity level in a manufacturing with a complex planning process.  Methods: The objective with this study was fulfilled through an in-depth empirical case study research in an Engineering-to-order (ETO) company. Data was collected through semi-structured interviews with employees working in the departments involved in the S&OP process, along with a survey that was used to identify the organizational culture. Moreover, a framework synthesis was carried out in order to identify the most suitable S&OP maturity model for assessing the maturity level.  Results: The results revealed that no written rules or policies govern the organizational activities, but an organization chart and some job descriptions exists. The total work is divided, resulting in a functional organizational design. The results showed that the departments facing supply and demand, have different objectives due to their functional specialization and that no common goals exist between the departments. The results also showed that the prevailing organizational culture appeared as different among the departments. The S&OP diagnosis reveled that the S&OP maturity level was low on most of the dimensions except from the IT dimension. Moreover, it revealed the lack of a shared understanding of the S&OP process among the different departments.  Conclusions: The main findings for managing the silo culture in order to reach a higher S&OP maturity level was to: (1) possess a organizational design that facilitates cross-functional alignments, (2) focus on people by ensuring that the S&OP process is fully understood, (3) define clear job roles and job descriptions, (4) state goals for each department and incorporate common goals along with metrics for all departments, (5) incorporate a strong organizational culture with shared values and beliefs and (6) have a clear decision making process along with moving towards a decentralization in some areas.
Bakgrund: Under de senaste åren har företag konkurrerat i en globaliserad och dynamisk miljö, vilket skapat incitament för att erhålla kostnadsfördelar. Flertalet företag har försökt implementera Sälj- och Verksamhetsplanering (SVP) men få har lyckats erhålla de förväntade resultaten. Koordineringen av försäljning och verksamhet återstår vara en avgörande utmaning på grund av flertalet skillnader och konflikter i mål och ansvarsområden, även kallat för silo kultur. Eftersom många företag stött på detta hinder i samband med en SVP implementering, återstår det att undersöka SVP-processen i ett företag med funktionella områden (avdelningar) och en komplex planeringsprocess.  Syfte: Syftet med denna studie är att ge en djupare förståelse för hur man hanterar silo kultur utmaningen, för att uppnå en högre SVP-mognadsnivå i en tillverkande industri med en komplex planeringsprocess.  Metod: För att uppnå syftet med studien genomförde författarna en djup empirisk fallstudie i ett kundorderstyrt företag. Data samlades in genom semi-strukturerade intervjuer med anställda som tillhör de avdelningar som är involverade i SVP-processen, och genom en enkät som användes för att identifiera organisationskulturen. Dessutom genomfördes en syntes av befintliga SVP-ramverk för att identifiera den mest lämpliga SVP-mognadsmodellen för att bedöma mognadsnivån.  Resultat: Resultaten visade på att inga skriftliga regler eller riktlinjer reglerar organisationsaktiviteterna, men att ett organisationsschema och ett fåtal arbetsbeskrivningar existerar. Det totala arbetet är uppdelat mellan olika avdelningar, vilket resulterar i en funktionell organisationsdesign. Resultaten avslöjade även att de avdelningar som står inför utbud och efterfrågan har olika mål på grund av sin funktionella specialisering och att det inte finns några gemensamma mål mellan avdelningarna. Dessutom visade resultaten att den nuvarande organisationskulturen uppfattas som annorlunda mellan avdelningarna. SVP-diagnosen avslöjade att SVP-mognadsnivån var låg på de flesta dimensioner förutom IT-dimensionen. Vidare avslöjar resultatet bristen på en gemensam förståelse för SVP-processen mellan de olika avdelningarna.  Slutsatser: Huvudresultaten för att hantera silo kulturen för att uppnå en högre SVP-mognadsnivå var att: (1) ha en organisationsdesign som främjar tvärfunktionella anpassningar, (2) fokusera på människorna genom att säkerställa att SVP-processen är fullt förstådd, (3) definiera tydliga arbetsroller och arbetsbeskrivningar, (4) ange mål för varje avdelning och inför gemensamma mål tillsammans med gemensamma mätvärden för alla avdelningar, (5) inför en stark organisationskultur som präglas av gemensamma värderingar och övertygelser och (6) erhåll en tydlig beslutsprocess tillsammans med att gå mot en decentralisering inom vissa områden.

This study focuses on the factors that cause project delays in an industrial hydraulic company. More specifically, the article also addresses the development of on-demand engineered products and the impact of the procurement process on delays. As part of the investigation, the authors assessed the historical data of the projects completed in 2017 before determining the critical path for each project in an attempt to identify which project stages may be improved. After further assessing the resulting data, the planning and procurement procedures were deemed as requiring improvement. Next, the article proposes a training plan and new procedures based on the Project Management Body of Knowledge guidelines for Engineer-to-Order projects. In conclusion, the results showed that delays were effectively reduced after implementing these new procedures in both Project Management stages.

Prostate cancer is the most prevalent malignant neoplasia affecting men over 65 years of age. While prostate carcinoma is considered mostly curable when confined to the prostatic capsule, more aggressive forms have a predilection to metastasise to the bone. Increased expression of RUNX2 in primary tumours has been proposed to promote bone metastasis due to its critical role in bone regulation. However a role in tumour development and progression remains to be elucidated for RUNX2, or its β-subunit, CBFb. This thesis examines a role for RUNX2 during prostate cancer progression and defines a novel role for CBFβ and the RUNX-associated transcriptional repressor, ETO in prostate development and tumorigenesis. ETO was differentially expressed during prostate development and tumorigenesis, in vivo. Additionally, in vitro, ETO protein expression was confirmed in all prostate cancer cell subtypes, with high expression associated with aggressive, metastatic prostate cancer cells. Manipulation of ETO in vitro demonstrated a requirement for ETO in the growth and invasion of human prostate cancer cells. Subsequent examination of these cells in subcutaneous xenograft models revealed a requirement for ETO in aggressive tumour growth in vivo. Examination of CBFβ in vivo, demonstrated its differential expression during prostate development and tumorigenesis. CBFβ knockdown in an aggressive prostate cancer cell line demonstrated that CBFβ expression was required for growth and invasion in vitro and delayed tumour growth following in vivo xenograft examination. RUNX2 was shown to promote increased cell growth and invasion in human prostate cancer cells in vitro. Subsequent in vivo examination of a non-aggressive prostate cancer cell line following forced RUNX2 expression demonstrated decreased primary tumour growth but was associated with increased metastasis. These data provide a new role for ETO and CBFβ in prostate development and tumorigenesis and a novel role for RUNX2 in the phenotypic shift associated with metastatic prostate cancer.

Resumo: Pelo que sabemos, este é o primeiro estudo do canto da população brasileira de Baleia-jubarte Megaptera novaeangliae em sua área de reprodução. Realizamos gravações em equipamento digital (DAT) na região do banco dos Abrolhos, Bahia, de julho a novembro de 2000. Analisamos auditivamente e espectralmente 20 ciclos de canto, perfazendo um total de 5 horas de canto gravado em 10 ocasiões diferentes. O fim de uma seção de canto foi gravado em uma ocasião. Os tipos de notas foram classificados com base em seus parâmetros acústicos: duração, duração do intervalo entre cada nota, freqüências mínimas e máximas. Medimos também o ritmo (número de notas por segundo) de emissão das notas do canto. Identificamos 24 tipos de notas organizadas em 5 temas (cada tema contém um ou mais tipos de frases). Todos os temas são compartilhados por todos os indivíduos da nossa amostra. Diferentemente de todos os outros estudos que conhecemos, em nenhuma ocasião os cantores da nossa amostra omitiram algum tema ou reverteram a ordem de emissão dos temas no canto. Em um dos temas encontramos tipos diferentes de frases cujas terminações são iguais, estando a diferença, portanto, na parte inicial da frase. Estudos anteriores sugeriram que se trata de um fenômeno mnemônico, análogo às rimas que usamos em nossa linguagem, cujo principal propósito seria facilitar o aprendizado da alteração do canto ao longo do tempo. Às vezes a passagem de um tema para o seguinte ocorre de forma gradual. Isto se dá devido à ocorrência de frases de transição; frases formadas por notas de dois temas adjacentes que são emitidas logo antes da passagem de um tema para o outro. Estas ocorrem na passagem entre dois temas dos cantos da nossa amostra. Uma característica marcante do canto da Baleia-jubarte é sua alteração irregular ao longo do tempo, alteração esta que sempre ocorre durante a temporada reprodutiva. Em nosso estudo, observamos o aparecimento de um tipo de nota e o desaparecimento de um tipo de 'terminação de frase', estando ambos aparentemente relacionados ao progresso da temporada. Uma outra característica conhecida é que todos os indivíduos de uma mesma população cantam igual. Em contraste, observamos uma variação individual na emissão de determinadas notas, todas concentradas em um mesmo tema. Acreditamos que esta variação esteja relacionada à capacidade que cada cantor tem de compor e/ou de aprender as modificações no canto ao longo do tempo. A partir da observação de que existe uma variação individual no canto desta espécie, fenômeno este até o presente momento não relatado em nenhum outro estudo sobre o qual temos conhecimento, apresentamos a idéia de que a capacidade de um cantor compor e/ou aprender as alterações do canto ao longo do tempo influencie, em última instância, sua aptidão
Abstract: To our knowledge, this is the first study of the song of the Brazilian population of Humpback Whale Megaptera novaeangliae in its breeding and calving ground. Digital recordings (DAT) were made at the Abrolhos Bank, Bahia, from July to November 2000. Aural and spectral analyses were carried out for approximately 20 song cycles, in a total of 5 hours of songs from 10 different recording events. The end of a song session was recorded once. Note types were classified using acoustical parameters, namely: duration, length of the intervals between them, minimum and maximum frequencies. The rhythm of note utterance was also measured. We identified twenty-four note types, organized in 5 themes (each theme has one or more types of phrases). All themes are shared by all singers. In contrast to all other studies that we are aware of, in no occasion the singers in our sample omitted a particular theme or reversed the order in which they were sung. In one of the themes we found different types of phrases that contain the same kind of ending. Previous studies have suggested that this is a mnemonic device, analogous to the rhymes we use in our language, being its main purpose to facilitate the learning of the song alteration through time. Sometimes the passage from one theme to another occurs gradually. This is due to the occurrence of transitional phrases, in which notes of the two adjacent themes are emitted in the same phrases prior to the change of theme. These transitional phrases occur in the passage between two themes. One of the well established characteristics of the song is its irregular alteration through time, which seems always to occur during the breeding season. In the songs we have analyzed, we observed the appearance of a particular note type and the dissapearance of a particular phrase ending, and both seem to be related to the progression of the breeding season. Another characteristic is the fact that all individuals in a population tend to sing the same current version of the song. In contrast, we observed an individual variation in the performance of certain note types, which are all concentrated in the same theme. We believe that this variation is related to each singer's ability to compose and/or to learn the song alterations through time. From the observation that an individual variation in the song of the species exists, something to our knowledge not reported in any other study so far, we present the idea that a singer' s ability to compose and/ or learn the song alterations through time influences his fitness

The Gate Turn-Off (GTO) Thyristor has the best voltage blocking and current conducting capabilities among all known high power semiconductor devices. To improve its dynamic performances to meet the increased demand in high-performance high-power applications, a special driving technique, namely unity gain turn-off, is studied. Several innovative approaches, which realize this driving requirement, are proposed, analyzed and experimentally demonstrated in this dissertation. The Emitter Turn-Off Thyristor (ETO) is a new family of high power semiconductor devices that is suitable for megawatt power electronics application. ETOs with voltage and current ratings of 4.0~6.0 kV and 1.0~4.0 kA, have been developed and demonstrated. These power levels are the highest in silicon power devices and are comparable to those of the GTO. Compared to the conventional GTO, the ETO has a much shorter storage time, voltage controlled turn-off capability, and a much larger reverse biased safe operation area (RBSOA). These combined advantages make the ETO based power system simpler in terms of dv/dt snubber, di/dt snubber and over current protection, resulting in significant savings at the system level. Experimental and numerical simulation results that demonstrate the advantages of the ETO are presented. A new family of snubberless turn-off GTO, the Resonant Gate Commutated Thyristor (RGCT) is proposed and investigated. By using a transient high commutation voltage, the RGCT can achieve unity turn-off gain and snubberless turn-off capability even with a relatively high gate loop stray inductance. Therefore conventional GTOs with flexible gate lead can be used to achieve the state-of-the-art performance similar to that of the Integrated Gate Commutated Turn-Off thyristor (IGCT). Detailed current commutation analysis and experimental results are presented. A novel equivalent circuit model for the GTO under the unity gain turn-off is proposed. This model is composed of a step current source, which represents the open-base PNP turn-off behavior, in series with a diode that represents the GTO's gate-cathode junction. This equivalent circuit can be used to analyze the turn-off transient behavior of a system employing this GTO. A new mechanism that dominates the failure of the GTO under the unity gain turn-off condition is identified and analyzed. Innovative hybrid GTO-based devices all have significant gate lead stray inductance. During the turn-off transition, this stray inductor will interact with the turn-off voltage source, the junction capacitance of the GTO's gate-cathode, causing effective current injection into the GTO's emitter junction when the voltage on the device is already high. Design guidelines and solutions for different types of GTO-based hybrid devices are provided.

The main objective of this thesis is to present proposals to improve the integration between Product Development (PD) and Production Planning and Control (PPC) in production systems that operate in Engineering-to-Order (ETO) environment. The specific objectives are: to develop a theoretical framework which identifies concepts and management tools applicable in business process studied that helps to explore integration opportunities; to map the studied scenario in order to understand it comprehensively in what it refers to interactions between PD and PPC; to identify the information technology tools used on studied companies, as well as ways to maximize their usage and to evaluate the proposals with the assistance of professionals who work at PD and PPC on the studied companies. The integration is critical for the production system operations performance, looking to reduce the time and costs for product placement in the market and to increase the quality of the products and services offered. To reach that goal, a literature review on the interest topics and case studies was performed to support the development of the proposals to improve the integration between PD and PPC. The case studies were carried out in large metal-mechanical companies that develop and manufacture capital goods on demand, located in the state of São Paulo. Among the proposals, models which attempt to describe, prescribe and represent aspects that can contribute to improving the integration between PD and PPC are presented, as well as a set of steps to be followed to achieve the process of improving the functional integration. It is expected that the results will contribute to a better understanding of the factors that enable a better integration between PD and PPC in this type of company as well as creating knowledge to the researched theories lines related to the topic and contribute to the management of the studied companies.
O objetivo geral desta tese é apresentar propostas para melhorar a integração entre Desenvolvimento de Produto (DP) e Planejamento e Controle da Produção (PCP) em sistemas de produção que atuam em ambiente Engineering-to-Order (ETO). Como objetivos específicos, tem-se: construir um referencial teórico que permita identificar os conceitos e ferramentas de gestão aplicáveis ao processo de negócio estudado de modo a explorar as oportunidades de integração; mapear o cenário estudado a fim de entendê-lo de forma abrangente no que se refere às interações entre DP e PCP; identificar as ferramentas de tecnologia de informação utilizadas nas empresas estudadas, assim como formas de potencializar sua utilização; avaliar as propostas apresentadas com auxílio de profissionais que atuam no DP e PCP das empresas. A integração é fundamental para o desempenho das operações de um Sistema de Produção, objetivando a redução do tempo e custos para colocação de produtos no mercado e aumento da qualidade dos produtos e serviços oferecidos. Para atingir o objetivo proposto, foram realizados uma revisão bibliográfica sobre os temas de interesse e estudos de casos para subsidiar a elaboração das propostas para melhoria da integração entre DP e PCP. Os estudos de casos foram realizados em empresas metal-mecânicas de grande porte que desenvolvem e fabricam bens de capital sob encomenda, localizadas no estado de São Paulo. Dentre as propostas são apresentados modelos que buscam descrever, representar e prescrever aspectos que possam contribuir para a melhoria da integração entre DP e PCP, assim como um conjunto de passos a serem seguidos para consecução do processo de melhoria da integração interfuncional. L'hématopoïèse est un processus complexe et dynamique qui conduit à la formation ainsi qu'au remplacement continu et régulé des cellules sanguines. Au cours de ce processus, une série de facteurs de transcription contrôle l'apparition, l'engagement et la différentiation des cellules souches dans un lignage déterminé. En particulier, chez les vertébrés, le facteur RUNX1/AML1 (pour Acute Myeloid Leukemia 1) est requis pour l'émergence des cellules souches hématopoïétiques ainsi que pour la différentiation des lignées myéloïdes et lymphoïdes. Chez l'homme, différentes altérations génétiques affectant AML1 sont liées au développement de pathologies du système hématopoïétique. Notamment, la translocation chromosomique t(8;21), codant la protéine de fusion AML1-ETO, est associée à 10-15 % des cas de leucémies myéloïde aiguës. Il a été décrit que AML1-ETO agit essentiellement en interférant avec la fonction de AML1 au cours de la différentiation hématopoïétique. Cependant, le mode d'action de cet oncogène reste mal compris et peu de facteurs modulant son activité sont connus. Récemment, de nombreux travaux ont mis en évidence que divers aspects du développement des cellules hématopoïétiques sont conservés de la Drosophile aux vertébrés. Notamment, les facteurs de transcription des familles RUNX et GATA, acteurs clefs de l'hématopoïèse chez les vertébrés, contrôlent aussi le développement des cellules sanguines chez la Drosophile. Tirant profit d'une part de la conservation phylogénétique des circuits géniques régulant l'hématopoïèse chez l'homme et la Drosophile et d'autre part des puissants outils d'analyses génétiques disponibles chez la Drosophile, nous avons cherché à utiliser la Drosophile comme système modèle pour étudier le mécanisme d'action de l'oncogène humain AML1-ETO et identifier des modulateurs de son activité. Nos résultats montrent que AML1-ETO exerce dans des cellules sanguines de la Drosophile dont la différentiation dépend de l'expression d'un facteur RUNX (Lozenge, LZ), des effets similaires à ceux observés dans des cellules leucémiques humaines portant la translocation t(8;21), à savoir : un blocage de la différenciation des cellules hématopoïétiques exprimant LZ et une augmentation de leur nombre. Grâce à la réalisation d'un crible génétique in vivo par interférence à l'ARN, nous avons identifié calpainB comme requis pour que AML1-ETO induise ces effets. De plus, nous avons pu montrer que l'inhibition des calpaines provoque la dégradation de AML1-ETO et diminue le potentiel clonogénique de cellules leucémiques humaines exprimant cette chimère. Hematopoiesis is a complex and dynamic process that leads to the formation and continuous blood cells replenishment. During this process, a series of transcription factors controls the appearance, commitment and differentiation of stem cells into specific lineages. In particular, in vertebrates, the factor RUNX1/AML1 (for Acute Myeloid Leukemia 1) is required for the emergence of hematopoietic stem cells and for the differentiation of both myeloid and lymphoid lineages. In humans, several genetic alterations affecting AML1 are linked to the development of different hemopathies. Notably, the chromosomal translocation t (8; 21), encoding the fusion protein AML1-ETO, is associated with 10-15% of cases of acute myeloid leukemia. It has been described that AML1-ETO acts essentially by interfering with AML1 function during hematopoietic differentiation. However, the mode of action of this oncogene remains poorly understood and few factors modulating its activity are known. Recently, numerous studies have shown that several aspects of hematopoietic development are conserved from Drosophila to vertebrates. Notably, transcription factors of RUNX and GATA families, which are key players in vertebrate's hematopoiesis, also control the development of blood cells in Drosophila. Taking advantage of the phylogenetic conservation of genetic circuitry regulating hematopoiesis between humans and Drosophila and of the powerful genetic tools available in Drosophila, we assessed whether Drosophila can provide a suitable model system to study the mechanism of action of the human oncogene AML1-ETO and to identify modulators of its activity. Our results show that AML1-ETO exerts in Drosophila blood cells expressing a RUNX factor (Lozenge, LZ) similar effects to those observed in human leukemic cells carrying the t(8;21), namely a differentiation blockage and an increased proliferation. In addition, by performing a large scale in vivo screen based on RNA interference, we identified calpainB as required for AML1-ETO-induced blood cell disorders in Drosophila. In addition, we showed that calpains inhibition resulted in AML1-ETO degradation and reduced the clonogenic potential of human leukemic cells expressing intrinsically this chimera. Our results establish that Drosophila can be used as an alternative model to better understand AML1-ETO function and to identify new factors regulating its activity

In order to gain competitive advantages in markets, companies have provided a variety of customized products to satisfy customer-specific requirements, leading to not only a large amount of product data but also high cost, long lead-time and complexity of quality control. Efficient product data management throughout the product lifecycle has become increasingly crucial, of which product structure management is regarded as the most important constituent.  The study took ABB Mine Hoist system as a case to investigate how to construct a generic product structure model fit for engineer-to-order system offerings with the consideration of their sales-delivery product lifecycle. The aim of the model is to facilitate the product-related information sharing and reuse across a company, and the integration of different business operations throughout the entire product lifecycle as well. Based on the current situation analysis of product data management on ABB Mine Hoist, three major issues were identified which need to be addressed in the formulation of a generic structure model: namely the integration of requirements of multiple disciplines; the consistency of product information throughout the product lifecycle; and the constant update of product repository. Through illustrating the formulation of ABB Mine Hoist generic structure model, the method of how to construct a generic product structure model for engineer-to-order system product was presented. The model was achieved by applying the framework of the step-based product model and was regarded as a result of integrating domain-specific requirements. The adaptive generic product structure model was then employed to display the role of this generic model in the different phases of a sales-delivery lifecycle. The model could serve as a “master concept” to transfer common product information in the product lifecycle. It’s expected to benefit the business of engineer-to-order system product through improving the integration of different disciplines, enhancing information exchange and reuse. It could also provide an abstract and conceptual basis for potential product repository to reinforce data consistency and completeness.

With the rise in digital information and technologies, businesses are aiming at measuring their processes and thereby getting better control over their operations. Industries are striving to increase efficiency and productivity to survive in the high global competitive market by adopting a digital tool. Simulation has been one of the most common tools of Industry 4.0 that allows a virtual representation of the real system. The use of simulation has tremendously increased in manufacturing industries due to its advantages where changes can be made and checked virtually before implementing them to the physical system. Data acquisition has been an important aspect that remains unfocussed especially in Engineer to Order (ETO) and Make to Order (MTO) environments where the production processes are complex, nonstandardised and depend heavily on manual work leading to a prolonged barrier towards digitalisation. So, can data create new values for companies? Are firms realising the importance of data? As the economy is moving towards a more data-driven state, it is essential for companies to realise the importance of data and find efficient ways of data collection. The thesis is based on a case study at an electrical transformer manufacturing firm taking its first step towards digital technology to have better control over its manufacturing processes. However, this transformation is primarily hindered by the limited availability of data and poor data quality due to manual data acquisition methods. Analysis of organisational documents and Value Stream Mapping (VSM) have been used to analyse missing primary and secondary production data. The results of this study conclude that modelling of the system can give additional data gaps, and the simulation model can be served as a powerful decision support tool for the firm. This switch in approach from intuition-based decision making to more fact-based decision making can result in better planning and control over the production. This thesis will lay the foundation for studies related to data acquisition and simulation in an ETO environment as it is not widely discussed in the existing literature

La sterilizzazione è di primaria importanza, specialmente per quanto riguarda il campo dei dispositivi medici (MDs), nel quale praticamente tutti i prodotti devono essere sterilizzati prima di essere immessi nel mercato, per prevenire l’esposizione a malattie e infezioni del paziente. Il tirocinio in LivaNova mi ha permesso di analizzare il processo di sterilizzazione ad ossido di etilene (ETO) attualmente in uso nel plant di Mirandola. Ho avuto modo di familiarizzare con il processo, con le regolamentazioni di riferimento e le procedure interne. In particolare, il mio studio si è concentrato sul processo e specialmente sul ciclo, composto di numerose variabili e parametri fisici. L’elaborato riporta, dopo un’introduzione all’azienda, una descrizione dettagliata del generico processo di sterilizzazione ETO e dello specifico processo utilizzato in LivaNova, fornendo una panoramica di tutti i parametri di processo. Inoltre, vengono fornite tutte le informazioni relative al processo di validazione del ciclo e i relativi metodi utilizzati. Il nucleo centrale del lavoro, invece, riguarda l’analisi dei principali parametri di processo del ciclo (umidità, pressione, temperatura) e lo studio causa-effetto di una criticità emersa negli scorsi mesi: nello specifico, tale criticità impatta sulla parte più esterna del packaging di prodotto, che a valle della sterilizzazione viene ritrovata deformata e quindi non conforme qualitativamente. Dopo la definizione dello stato AS-IS del processo, tramite l’utilizzo di analisi di correlazione prettamente qualitative, performate su dati appartenenti a cicli in cui si era presentato il problema, ho individuato informazioni e relazioni significative che sono state utilizzate per progettare dei test sperimentali, ovvero dei cicli di sterilizzazione con caratteristiche appositamente scelte per testare i vari parametri. Infine, con i risultati ottenuti, ho delineato un ipotetico stato TO-BE con relativa analisi tecnico economica.

This research discusses the use of Delta in regulating the investment behaviour of the Trustees of Self-Managed Superannuation Funds (SMSFs) who use Exchange Traded Options (ETOs) in their investment strategies. An ETO represents a contract between two parties, giving the taker (the buyer) the right, but not the obligation, to buy or sell a parcel of shares at a predetermined price, on or before a predetermined date, to or from the writer (the seller). It is acceptable for SMSF Trustees to use ETO investments as part of their overall investment strategy, providing that leverage or mere speculation are not the reasons behind that investment. It is important to note that neither the Regulator, the Australian Taxation Office (ATO), nor its predecessor, the Australian Prudential Regulatory Authority (APRA), actually state what constitutes 'speculation', or what the allowable uses for derivatives are. There are no practical guidelines. This is a key issue for this research, which aims, as practically as possible, to fill these crucial gaps. A Trustee must abide by their superannuation fund's overriding covenants and investment strategy, and inform its members, through Risk Management Statements, of the trust's derivative strategy. While ETOs can be used to manage risk, they also carry a level of risk themselves. Delta measures an ETO's value movement in correlation with a movement in the option's underlying share price. An ETO carrying a low Delta generally means a cheaper price (premium) per contract than an option carrying a higher Delta. The lower the Delta, however, the lower the chance there is of a positive result for the buyer. This research shows that an ETO Delta of less than 0.2 gives results in favour of buyers in only 11 out of 100 occurrences. This figure rises to 42 out of 100 when Delta is greater than 0.8. From the sampled data, there is an overall financial loss to the buyer of -1.91%, with the financial return results being mixed at all levels of Delta. The overall return results have been compiled without preference to market direction, and clearly highlight the natural premium bias (which the buyer pays) to the seller. What this data does is reenforce the need for Trustees to have a solid view of market directions, or a set strategy in place, as buyers of ETOs. The conclusions drawn from the findings show that the chance of loss (when buying), or gain (when selling) ETOs with a Delta of; - less than 0.20 is 89%; - less than 0.40 is 74%; - less than 0.60 is 66%; - less than 0.80 is 57%; - greater than 0.80 is 58%; For example, a Trustee buying an ETO with a Delta of less than 0.20, faces an 89% chance of loss; a Trustee selling an ETO with a Delta of less than 0.20, faces an 89% chance of gain. The findings on overall financial returns (profit or loss) offer additional support to this critical review of Delta as a risk measurement tool. Whist it is impossible to know the motives or actual positions of portfolio managers of SMSF at any time, the aim of the thesis is to provide a measurement tool that can be used to assist the trustee at any given time by measuring the option risk element alone. When interpreting the findings, the reader must remember that ETO strategies are numerous, and a high-risk profile for one strategy may represent a low risk for another. Further to this, an ETO strategy's risk profile may change with the overlaying of another ETO. For example, where a Call option is bought, the risk involved in that purchase is represented by the premium paid. However, another Call option can then be sold against that position, with a later (or earlier) date to expiry, and with a higher strike price. This 'overlay' reduces the initial risk, but impacts on the maximum gain. It is vital that Trustees have a solid understanding of the basics of ETO strategies before considering using Delta as a measure of risk. Delta may not be suitable, however, for measuring multiple layers of combined ETO positions, a type of derivative strategy not suited to or usual in the context of measuring risk within a SMSF. (table inserted) There is a major difference between simple and simplistic solutions offering practical answers in an environment of increasing complexity. Often, simple solutions offer far more value to the less experienced, when compared to complex ones, especially given the growing number of SMSFs, and the increasing lack of expertise in the areas of superannuation and risk management that this growth implies.

Les intégrines, principaux récepteurs de la matrice extracellulaire (MEC), entretiennent un lien étroit avec la MEC exerçant ainsi un contrôle majeur sur le comportement des cellules, en agissant sur la migration, la prolifération et la survie cellulaire. Les travaux actuels sur les voies de signalisation impliquant les intégrines commencent à mieux appréhender les effets spécifiques et complexes exercés par la MEC et les intégrines sur les cellules. Ainsi, afin d’identifier de nouveaux partenaires de la sous-unité d’intégrine 1, un crible double hybride chez la levure a été réalisé à partir d’une banque d’ADNc de cerveaux embryonnaires murins. Nous avons pu mettre en évidence une interaction entre le domaine cytoplasmique de l’intégrine 1A et les membres d’une famille de protéine appelée ETO. Nous avons démontré que cette interaction pouvait s’effectuer dans les cellules et que ETO était présente dans des structures particulières de la membrane appelés « ruffles ». The integrins are major receptors for the extracellular matrix (ECM). By their connexion with the ECM, they can modulate the cellular behaviour by regulating the cell adhesion, motility and survival. Now, the studies on the signalling pathways mediated by the integrins help us to better understand the specific and complex effects of ECM on the cells. For this reason, we set up a yeast two-hybrid assay where the screening of mouse embryonic brain cDNA library enables us to find new cytoplasmic partners for the 1A integrin subunit. By this approach, we found that the cytoplasmic domain of 1A integrin can associate with members of the ETO family. We have demonstrated that this interaction can occur in cells and ETO is present in membrane ruffles. Moreover, knock-down of ETO expression results in decreased adhesion and spreading, suggesting an additional and novel role for this family in integrin signalling pathways

A evapotranspiração é de suma importância ao balanço hídrico, originando demandas crescentes de informações quantitativas que proporcionem o planejamento hídrico de forma sustentável. Este trabalho determina a evapotranspiração de referência (ETo) na bacia experimental do riacho Gameleira PE, utilizando lisímetro de pesagem hidráulica e métodos indiretos. A bacia experimental do riacho Gameleira está inserida na bacia do rio Tapacurá, localizada na parte Nordeste da Mesoregião Geográfica do Agreste do Estado de Pernambuco, nas coordenadas UTM 245.186 E; 9.108,157 N; 248.694 E e 9.103,632 N no meridiano central 25, incluindo a estação climatológica da Escola Agrotécnica Federal de Vitória de Santo Antão PE. O lisímetro instalado é do tipo pesagem hidráulica com pistão de mercúrio, área superficial de 1,52 m2 e profundidade de 0,80 m. A análise estatística foi feita nas escalas diária e pentada. A resposta das células de carga hidráulica às massas-padrão aplicadas na calibração foi linear, com coeficiente de determinação na faixa de 0,9993 a 0,9999 e erro padrão da estimativa na faixa de 0,60 a 1,45 mm dia-1 na escala diária. A resolução do lisímetro é de 0,1543 mm de ETo. O erro médio posicional obtido foi de 2,31%. A relação área de borda e superfície é de 18%. O coeficiente de calibração (k) de 2,13513 mm kg-1 define que cada mm de ETo ocorrido no tanque corresponde a uma variação de leitura no manômetro de 3,24 mm. Foi feita avaliação de desempenho entre o lisímetro como padrão e os métodos indiretos tanque Classe A, Camargo, Makkink, Radiação Solar, Jensen-Haise, Hargreaves-Samani, Blaney- Criddle, Penman e Penman-Monteith. O método da Radiação Solar foi o que apresentou melhor índice de desempenho em relação ao lisímetro e o método padrão da parametrização da FAO Penman-Monteith apresentou desempenho bom nas escalas diária e pentada

Abstract:
The digital wave that has engulfed the construction industry has in many ways changed how consultants work. The new working methods have created new conditions and opportunities for how architectural planning in construction can be conducted. Traditional planning is a project-oriented ETO process that can be compared with manufacturing of complex one-of-a-kind products (Jensen, 2014). Many consultants' business models based on the process gets criticized for not encouraging the reuse of solutions or development of their ways of working (Lidelöw, Stehn, Lessing, & Engström, 2015). In the manufacturing industry, processes based on the concept of mass customization are used. The purpose of these processes is to improve the flexibility of the final product, at the same time as the standardization and economies of scale is preserved in the components (Jensen, 2014). Modularization and configuration is central within mass customization. These concepts along with automation formed the basis of the study. The concept intended to incorporate these influences into traditional A-planning, is called room objects. Room objects are parametrically driven objects that collect furnishings and equipment as modules in a BIM environment. In this study the room objects has been developed as products families with the intention and ability to meet the underlying needs (Jiao, Simpson, & Siddique, 2007). The needs consisted of room layouts gathered from reference projects and regulations and have been identified through case studies. The needs have then been revised in stages to ultimately result in the design parameters used in the construction of the room objects. In this thesis, the development of the room objects has been delimited against WC environments. The technical use of the objects has been studied within the concept room objects. The result of the study is presented as a methodology for the use and management inside Autodesk Revit. A big part of the use consists of various tools in the form of scripts developed in the software Dynamo. The tools allow some operations within the use, and thus within the planning, to be automated. They also enable the creation of configurators outside of Revit that facilitates the configuration of the room objects. The analysis and evaluation of the flexibility and usability showed of the room objects, shows that room objects as concept has the potential to work in the phase of A-planning. The flexibility and modularity of the room objects could within the context of the work meet 97 % of the designs given by reference projects and 100% of the designs given by the regulations. The limits of the adjustment possibilities are situated in complex room shapes that require an advanced parametric basis. The usability of the objects appeared to be limited by technical knowledge. The configuration of the room objects requires an insight of the parametric construction. The modification of the room objects requires the user to possess an understanding of parametric modelling at family level in Revit. The alteration of the scripts demands knowledge of visual programming. The potential is there to solve or manage these limitations through technical means and suitable project organization. How the concept can be implemented purely in terms of the process should be further investigated. In this study, there has been a discussion of how the project group can be organized in order to fully utilize the benefits room objects can bring in terms of time savings, experience feedback and quality assurance.

La mejora de procesos es una de las estrategias más utilizadas por las empresas para mejorar su posición competitiva. De sus disciplinas, el rediseño de procesos (BPR, por Business Process Redesign) es una de las más usadas y ha permitido a muchas empresas mejorar la eficiencia y eficacia de sus procesos de forma considerable, dejando de lado las pretensiones de cambios radicales que la literatura de BPR proponía inicialmente. Enercom S.A. es una empresa chilena especializada en el diseño y fabricación de equipos industriales en procesos térmicos, de secado y combustión. Estos son altamente complejos y diseñados a medida según las especificaciones del cliente, clasificando a la empresa como de diseño a la medida (ETO, por Engineered to Order). La gerencia de la empresa desea realizar una revisión de sus procesos, en particular del proceso de ingeniería de detalle que detectan como fuente de numerosos errores. El objetivo del proyecto se centrará en la reducción del número de Productos No Conformes (PNC) originados en el proceso de ingeniería de detalle, basándose en la metodología de Cameron y Braiden [2004]. Una evaluación de la organización permite comprender el funcionamiento y las características particulares del proceso de venta, diseño y fabricación de equipos, que concentra el 90% de sus ingresos anuales. Se detecta en esta etapa que el proceso de ingeniería de detalle concentra el 38,7% de los Productos No Conformes detectados, validando la decisión de rediseñar este proceso particular. El consecuente levantamiento del proceso de ingeniería de detalle, junto con un análisis de causa-efecto y de causa raíz de los PNC detectados, deja en evidencia que la falta de especificación de requerimientos y criterios de diseño y el alto nivel de incertidumbre del proceso son responsables de casi el 80% de las no conformidades detectadas. Durante el rediseño del proceso se pone especial énfasis en mitigar las principales causas de no conformidad detectadas, seleccionando de la literatura las mejores prácticas que reduzcan los costos y mejoren la calidad del proceso. También se rediseñan los sistemas que soportan el proceso: la estructura organizacional y los sistemas de información de la empresa. Finalmente, se incluye una propuesta de plan de implementación como un primer acercamiento hacia la implementación de este rediseño en la empresa. Este trabajo muestra que el proyecto tiene beneficios económicos importantes y que el costo de implementación es relativamente bajo, por lo que se recomienda continuar con las siguientes etapas del proyecto propuestas en la metodología. Se determina que BPR es aplicable a empresas del sector ETO y que permite obtener beneficios importantes, siempre que se limite a procesos unitarios y dentro una misma unidad de negocios.
