Abstract:
RésuméLa neurotransmission de sérotonine (5-HT) est régulée, via un rêtro-contrôle négatif, par des réceptcurs situés an niveau du raphé nucleus et des terminaisons neuronales. Ces autorécepteurs sérotoninergiques ont été étudiés aux niveaux comportemental, électrophysiologique et biochimique. Les réponses comportementales et électropliysiologiques sont atténuées par l'administration chronique des antidépresseurs. De plus, in vitro, les inhibiteurs de capture de 5-HT atténuent l'aulorégulation de la libération de [3H]5-HT. La présente étude concerne l'effet, sur cette libération induite par stimulation électrique de l'hypothalamus; de rat et sa modulation, de l'administration chronique de 2 antidépresseurs, le citalopram, inhibiteur spécifique de la capture de 5-HT, et le milnacipran (anciennement midalcipran, F 2207), qui bloque la capture de 5-HT et de noradrénaline avec la même puissance. Le eitalopram était administré à 50 et à 10 mg/kg/j pendant 21 jours et 24 h après, les rats étaient décapités, des coupes d'hypoihalamus incubées avec la [3H]5-HT, superfusées avec une solution de Krebs et stimulées électriquement. La quantité de [3H]5-HT libélée par stimulation électrique étail augmented et l'effet inhibiteur de l'agoniste, LSD, diminué chez les rats traités. Ces effets étaient présents aux 2 doses mais plus marqués à la forte dose. Donc un traitement chronique avec le citalopram provoque une «down-regulation» de l'autorécep-teur sérotonergique, permettant ainsi une augmentation de la neurotransmission de 5-HT. Cet effet peut expliquer, an moins en partie, l’action antidépressive de ce produit et it esl possible qu'il représente un mécanisme fondamental à beaucoup d'antidépresseurs qui inhibent la capture de 5-HT. Cependant, après 21 jours de traitement avec le milnacipran, à 50 mg/kg/j, aucun des paramétres étudiés n'était modifié. Cela suggère que l’hypothèse de la «down-regulation» de l’autorècepteur n'est pas l'hypothèse universelle qui explique l’action de tous les antidépresseurs agissant sur la capture de 5-HT.

AbstractBackground:Essential tremor (ET) is the most common tremor disorder in adults. In addition to upper limbs, the tremor in ET may also involve head, jaw, voice, tongue, and trunk. Though head tremor (HT) is commonly present in patients with ET, large comparative studies of ET patients with HT (HT+) and without HT (HT−) are few.Methods:To determine whether ET with HT is a distinct clinical subtype by comparing ET patients with and without HT, a chart review of 234 consecutive patients with ET attending the neurology clinics of the National Institute of Mental Health and Neurosciences, India, was done. A movement disorder specialist confirmed the diagnosis of ET in all patients using the National Institutes of Health collaborative genetic criteria.Results:HT was present in 44.4% of the patients. Comparison between HT+ and HT− showed that the HT+ group patients: (1) were older, (2) had later onset of tremor, (3) had unimodal distribution of age at onset with a single peak in the fifth decade, (4) had more frequent voice tremor, and (5) were more likely to have mild cervical dystonia. HT was part of presenting symptoms in nearly two thirds of the ET patients and in the rest it was detected during clinical examination.Conclusions:Several demographic and clinical variables suggest that ET patients with HT have a distinct clinical phenotype.

Abstract:
Interactions between 5-hydroxytryptamine (5-HT) and endothelin-1 (ET-1) relative to contraction of rat aortic rings were examined in this study. Pretreatment of rings with threshold concentration of 5-HT potentiated the subsequent contractile response to ET-1. However, pretreatment with threshold concentration of ET-1 did not potentiate the contractile response to 5-HT. The 5-HT receptor antagonist LY 53857 blocked the synergistic contractile effects of 5-HT and ET-1 on rat aortic rings. Indomethacin and the thromboxane A2/endoperoxide receptor antagonist SQ 29548 also attenuated (P less than 0.05) the synergistic contractile effects of 5-HT and ET-1, suggesting release of thromboxane A2 or expression of thromboxane A2 receptors during this interaction. The calcium channel blocker verapamil also decreased the synergistic contractile effects of 5-HT and ET-1. Contraction of aortic rings by 5-HT alone was abolished by LY 53857 and attenuated by verapamil, diltiazem, and SQ 29548. Decrease in the force of contraction by verapamil as well as diltiazem indicates activation of voltage-dependent calcium channels during 5-HT-mediated contraction and perhaps during amplification of the vasoconstrictor activity of ET-1 by 5-HT.

Abstract:
Elevated levels of serotonin (5-HT) and endothelin-1 (ET-1) may be involved in cardiovascular complications of diabetes mellitus. Data suggest supraphysiological concentrations of 5-HT (10−6 M) potentiate the ability of ET-1 to stimulate DNA synthesis and vascular smooth muscle cell (VSMC) proliferation in vitro via activation of mitogen-activated protein kinase (p42/44 MAPK) and Janus kinase 2 (JAK2) pathways. Additionally, 5-HT enhances agonist-induced contractions via p42/44 MAPK and an unknown tyrosine kinase. However, the exact mechanisms of the 5-HT/ET-1 interactions and whether these effects occur at physiological levels (10−9 M) are unknown. Therefore, we hypothesized that interactions between 5-HT and ET-1 at physiological concentrations in VSMC enhanced activation of both p42/44 MAPK and JAK2 pathways contributing to vascular growth and contractile responses. With the use of rat VSMC and Western blot analysis, our data suggest no effect of acute (30 min) preincubation with 5-HT (10−9 M) and/or ET-1 (10−9 M) on the activation of either pathway in normal or high glucose conditions. To determine if there was altered vascular reactivity in intact vessels we tested the effects of 5-HT and ET-1 interaction using myographs to measure isometric contractions of rat thoracic aortic rings. 5-HT (10−9 M) and ET-1 (10−12 M) stimulate enhanced contractile responses to each other that were inhibited by JAK2 and p42/44 MAPK antagonists. Our findings demonstrate that both 5-HT and ET-1 at physiological concentrations could interact with each other and activate p42/44 MAPK and JAK2 signaling pathways to cause an increase in smooth muscle contraction that could lead to altered vascular function.

A high adherence to a Mediterranean diet has been related to numerous beneficial effects in human health, including a lower incidence and mortality of prostate cancer (PCa). Olive oil is an important source of phenolic bioactive compounds, mainly hydroxytyrosol (HT), of this diet. Because of the growing interest of this compound and its derivatives as a cancer chemopreventive agent, we aimed to compare the in vitro effect of HT isolated from olive mill wastewaters and five semisynthetic alkyl ether, ester, and nitro-derivatives against prostate cancer (PCa) cell lines. The effect in cell proliferation was determined in RWPE-1, LNCaP, 22Rv1, and PC-3 cells by resazurin assay, the effect in cell migration by wound healing assay, and tumorsphere and colony formation were evaluated. The changes in key signaling pathways involved in carcinogenesis were assessed by using a phosphorylation pathway profiling array and by Western blotting. Antiproliferative effects of HT and two lipophilic derivatives [hydroxytyrosyl acetate (HT-Ac)/ethyl hydroxytyrosyl ether (HT-Et)] were significantly higher in cancerous PC-3 and 22Rv1 cells than in non-malignant RWPE-1 cells. HT/HT-Ac/HT-Et significantly reduced migration capacity in RWPE-1 and PC-3 and prostatosphere size and colony formation in 22Rv1, whereas only HT-Ac and HT-Et reduced these functional parameters in PC-3. The cytotoxic effect in 22Rv1 cells was correlated with modifications in the phosphorylation pattern of key proteins, including ERK1/2 and AKT. Consistently, HT-Ac and HT-Et decreased p-AKT levels in PC-3. In sum, our results suggest that HT and its lipophilic derivatives could be considered as potential therapeutic tools in PCa.

Les hormones thyroïdiennes (HT) sont essentielles pour le bon fonctionnement du cerveau tout au long de la vie des vertébrés, dès les stades précoces du neuro-développement. Des études épidémiologiques ont montré l’importance des HT de la mère pendant les premiers mois du développement fœtal : une déficience précoce en HT maternelles entraîne à long terme des altérations du développement cognitif et du comportement social de l’enfant. L’apport des modèles animaux, non seulement les modèles mammifères mais également les modèles alternatifs (poisson zèbre, xénope, poulet), a permis de décrypter les mécanismes cellulaires et moléculaires gouvernés par les HT lors du développement cérébral. En particulier le modèle rongeur a contribué à montrer que les HT ont également un rôle crucial chez l’adulte, principalement au sein de deux niches neurogéniques majeures, la zone sous-ventriculaire et la zone sous-granulaire de l’hippocampe où elles régulent finement le destin des cellules souches neurales (CSN). Une question essentielle en biologie des cellules souches est de comprendre, comment les HT gouvernent le devenir des CSN vers un destin neural ou glial et ce, afin de contribuer au développement du cerveau et de maintenir ses fonctions tout au long de la vie adulte dans des conditions physiologiques et lors d’un dommage cérébral (maladies neurodégénératives, maladies démyélinisantes ou accident vasculaire cérébral). Notre revue fait le point sur les connaissances actuelles sur le rôle d’un signal endocrinien clé, les HT, lors du développement du cerveau et de la neurogenèse adulte, et principalement chez les mammifères, notamment l’Homme.

Abstract The recent discovery of various mutations of the CALR gene that are mutually exclusive with JAK2 and MPL mutations has allowed a correct diagnosis in about 90% of adult cases of essential thrombocythemia (ET). Moreover, the mutation status of JAK2 and CARL defines subtypes of ET in adults with a substantially different clinical course and outcome. Based on our experience, we suggested that primary thrombocythemia (PT) in children is characterized by subtypes that differ from those found in adult ET. The present study was carried out in children and adolescents with PT in order to (a) characterize the various subtypes of the disease and (b) analyze their clinical and biologic features, treatment approach and outcome. PT patients aged <20 years (yrs) at diagnosis (dx) were evaluated for mutations of JAK2, thrombopoietin (TPO) and its receptor (MPL) and CALR genes, and for clonal hematopoiesis (females). The presence of MPLS505A (confirmed on DNA from buccal swabs) defined a hereditary thrombocytosis (HT). ET was diagnosed according to WHO 2008 criteria. For wild type patients, an additional inclusion criteria was a follow-up >24 months. Among 58 PT patients (males: 23; females: 35; median age at dx: 14.4 yrs), 21 (36%) had HT due to MPLS505A, 14 were JAK2V617F-mutated (24%), 9 (16%) harbored CALR mutations and 14 (24%) were wild type for JAK2, CALR and MPL (Fig 1). JAK2- and CALR-mutated were older than those with wild type ET or with HT (median age, 17.6 and 16.1 vs 10.4 and 13.7 yrs, p .028). As to the hematologic findings, HT patients showed both hematocrit values (median, 36.3%) and leukocytes counts (median, 9.53 x109/L) significantly lower than ET patients, whatever the subtypes (median, 41.2% and 11.2 x109/L, p .006 and p .029, respectively). No differences were found with regard to platelets both between HT and ET and among the different ET subtypes. JAK2-mutated patients exhibited more frequently symptoms (69%) compared to CALR-mutated (22%), wild-type ET (14%) and HT (14%) patients (p. 0057). Splenomegaly at diagnosis was recorded more frequently in JAK2-mutated than in CALR-mutated or wild type-ET or HT (50%, 33% 21% and 14% , respectively, p .122). Antiplatelet agents, mostly acetylsalicylic acid (ASA), were started less frequently in HT than in ET patients, irrespective of the subtypes (57% vs 81%, p .05). The use of ASA progressively decreased over the time; at the last follow-up, 2 patients with HT, 2 CALR-mutated and 1 JAK2-mutated patients were still receiving ASA, while no wild type ET patient was on treatment. Cytoreductive agents, hydroxyurea and/or interferon and/or anagrelide, were used in a minority of HT patients (19%) in comparison with ET patients (65%), p .001, mainly with those wild-type (78%, p <.001). At the last observation, one HT patient was still receiving cytoreductive agents compared to 30% of ET patients whatever the subtypes (p .024). After a median follow-up of 196 months (similar in the different subtypes), all patients are alive. On the whole, 5 thrombotic events were recorded in 3 patients with HT and in 2 ET patients (1 JAK2-mutated and 1 JAK2 and CALR wild-type), without any significant thrombophilic abnormalities during treatment with ASA and/or cytoreductive agents. A progressive splenomegaly was recorded in 9 (15%) patients (2 HT, 4 JAK2-mutated, 3 CALR-mutated) and it was combined with grade ≥2 medullar fibrosis in 2/4 JAK2-mutated and in 2/3 CALR-mutated patients. None of the JAK2 and CALR wild-type patients had spleen enlargement or reticulin fibrosis (p .022). Two untreated patients (1 HT and JAK2 and CALR wild-type) developed malignancies. On the whole, these data emphasize that in young patients with PT, hereditary forms can be frequently observed. Thrombotic events, recorded mainly in HT patients despite treatment with ASA, were probably due to a MRP4 protein overexpression that was found in our MPLS505A HT. Moreover, our observations highlight that, in contrast to adult ET, more than one third of young ET patients have no JAK2 or CALR mutations. Figure 1 Figure 1. Cette etude avait pour objet la caracterisation biochimique et moleculaire de deux recepteurs de la serotonine (5-hydroxytryptamine, 5-ht), 5-ht#3 et 5-ht#1#a. Le recepteur 5-ht#1#a a deja ete purifie, clone et sequence, en revanche, l'on possede tres peu d'information sur le recepteur 5-ht#3. En vue de la purification du recepteur 5-ht#3 et en utilisant le derive tritie d'un antagoniste selectif, le zacopride, nous avons demontre la presence, dans les membranes des cellules de l'hybridome ng108-15, d'un recepteur 5-ht#3 identique au recepteur 5-ht#3 cerebral. A partir des membranes de cellules ng108-15, nous avons obtenu une forme soluble du recepteur 5-ht#3. C'est un polymere, glycosyle, de poids moleculaire apparent eleve et de coefficient de svedberg de 12 s. Sa sous-unite de liaison a un poids moleculaire apparent de 35 kda. Un (ou plusieurs) residu(s) tryptophane est (sont) implique(s) dans la liaison des agonistes et des antagonistes au recepteur 5-ht#3. Les recepteurs 5-ht#1#a sont apparemment heterogenes en regard de leur interaction avec les agonistes puisqu'il a ete etabli que la liaison a haute affinite de ces composes a une partie des recepteurs 5-ht#1#a centraux est insensible au gtp. Nous avons demontre (dans l'hippocampe de rat) que cette insensibilite au gtp etait la consequence d'une oxydation du recepteur 5-ht#1#a. En vue de l'etude des mecanismes de regulation des recepteurs 5-ht#1#a aux niveaux transcriptionnel et traductionnel, nous avons determine la cartographie de l'arn messager du recepteur 5-ht#1#a par hybridation in situ d'une sonde arnc. De plus, nous avons compare les distributions respectives de l'arnm, de la proteine receptrice (par immunohistochimie) et des sites de liaison (par marquage autoradiographique) du recepteur 5-ht#1#a dans le cerveau de rat. Elles sont superposables et concentrees dans le systeme limbique

L'objectif de notre etude consiste a apporter des elements d'information sur la topographie du site recepteur serotoninergique 5-ht#3, notamment dans la region interagissant avec la partie aromatique des ligands antagonistes specifiques. Dans ce but, nous avons concu et synthetise des composes derives des benzamides serotoninergiques presentant un cycle intramoleculaire destine a maintenir les principaux elements pharmacophores, noyau aromatique et groupe carbonyle amidique, dans une position coplanaire. Nous avons tente d'apporter des elements de comparaison des proprietes biologiques observees pour des molecules originales dans la serie des quinazoline-2,4-diones, par rapport aux ligands de reference comme le zacopride. La preparation des quinazoline-2,4-diones derivees de la quinuclidine a ete optimisee par la mise au point de methodes de cyclisation intramoleculaire a l'aide de reactifs carbonyles et de substituts du phosgene. Dans une deuxieme approche, nous avons synthetise et evalue les proprietes d'esters de divers acides aromatiques capables de reveler l'activite antagoniste des recepteurs 5-ht#3, par rapport aux esters serotoninergiques classiques. Les affinites des molecules synthetisees pour le recepteur 5-ht#3 ont ete evaluees par des essais de deplacement du radioligand #3h brl 43694 et une approche theorique a ete tentee par des etudes de modelisation moleculaire a l'aide du logiciel sybyl 6. 0. Nous constatons de nombreuses variations des proprietes biologiques de nos composes a structure cyclique dicarbonylee, par rapport aux benzamides serotoninergiques classiques

Abstract:
L'objectif de notre etude est d'apporter des elements d'information sur la topographie du site recepteur serotoninergique 5-ht#3 et d'etudier les contraintes steriques au niveau du site anionique. Pour cela, nous avons concu et synthetise des composes possedant de nouveaux systemes azacycliques ou azapolycycliques tels que des piperidines trisubstituees, des derives du tropane et de la quinuclidine et ainsi que des molecules analogues de la coccinelline. La cyclisation de dieckmann, la cyclisation de robinson-schopf ont ete utilisees pour preparer ces azapolycycles cetoniques. Ces dernieres permettent d'acceder de facon stereoselective aux amines ou aux alcools correspondants qui ont ete condenses avec des acides aromatiques capables de reveler l'activite antagoniste pour les recepteurs 5-ht#3. Dans une deuxieme approche, nous avons synthetise une serie d'analogues du metoclopramide, dans lesquels un cycle de trois a six chainons est introduit en position beta de la fonction amide. Les molecules synthetisees ont ete evaluees par des essais de deplacement du radioligand #3h brl 43694 et pour certaines d'entre elles au moyen du test de bezold-jarisch. Nous constatons que la plupart des substitutions sur la piperidine, excepte celles localisees en position anti par rapport au doublet libre de l'azote, conduisent a une importante diminution de l'affinite pour le recepteur 5-ht#3. Des etudes de modelisation moleculaire, associees aux resultats biologiques, nous ont permis de dire que le site anionique du recepteur 5-ht#3 se trouve localise dans une cavite etroite et hydrophobe. L'introduction d'un cycle dans le metoclopramide augmente nettement l'affinite pour les recepteurs 5-ht#3. Elle depend de la dimension du cycle et nous remarquons que la presence d'un heteroatome comme le soufre ou l'oxygene est defavorable. D'autre part, la specificite de ces molecules est considerablement accrue

Dès la découverte des récepteurs 5-HT₇ en 1993, des études intenses de relation structure-activité ont été poursuivies par plusieurs groupes de recherches, dont notre laboratoire. En conséquence, un nombre impressionnant de ligands potentiels 5-HT₇ sont référencés dans la littérature mais peu sont des agonistes sélectifs. Dans ce contexte, ce travail de thèse a pour but principal, la conception de quatre classes distinctes de ligands et leur évaluation en tant que ligand agoniste des 5-HT₇R. Dans un premier temps, nous nous sommes intéressés à la préparation de ligands de type 2,4-diaminopyrido [2,3-d]pyrimidinique et 2,4-diaminopyrimidinique. Par la suite, nous avons élaboré une troisième famille de composés bâtis sur une charpente pyridinique, à partir d’un des plus intéressants agonistes sélectifs 5-HT₇ actuels et ce, via une stratégie multi-étape efficace. La dernière partie de ce mémoire a été consacrée au développement d’une méthode originale et rapide de synthèse des tétrahydro-1,6-naphtyridines polysubstituées au départ de 3-vinyl-1,2,4-triazines judicieusement fonctionnalisées via une séquence domino d’addition d’aza-Michael et une cyclisation intramoléculaire de Diels-Alder à demande électronique inverse
Serotonin-activated cell-surface receptor 5-HT₇ is the most recently discovered 5-HT receptor. Intensive studies of structure-activity relationships have been pursued by several research groups, including our laboratory, and have resulted in the publication in the literature of an impressive number of potential ligands 5-HT₇. In this context, the main purpose of this thesis is to design four distinct classes of ligands. First, we were interested in the preparation of 2,4-diaminopyrido[2,3-d]pyrimidinique and 2,4-diaminopyrimidinique ligands. Subsequently, we developed a third family of compounds built on a pyridine scaffold, from one of the most interesting current 5-HT₇ selective agonists, via an effective multistep strategy. Nous avons étudié 2 protéines impliquées dans l'absorption iléale de la vitamine B12 ou cobalamine : le récepteur du complexe facteur intrinsèque-cobalamine et la transcobalamine. Le récepteur du complexe facteur intrinsèque-cobalamine est exprimé dès la dixième semaine du développement fœtal chez l'homme. Le récepteur iléal porcin a été purifié par chromatographie d'affinité sur un support sépharose-facteur intrinsèque. Il se compose de 4 polypeptides dont trois sont glycosyles. La liaison entre le récepteur et son ligand peut être inhibé en présence d'agglutinine de germe de blé. L'inhibition est due à la fixation de cette lectine à la sous-unité alpha du récepteur qui contient le site de fixation du complexe facteur intrinsèque-cobalamine. Les résidus galactosyls de la copule glucidique du facteur sont importants dans la formation de la liaison entre le récepteur et son ligand. Enfin, nous avons montré l'existence de la synthèse et de la sécrétion de transcobalamine par les cellules HT 29. Ces cellules ont été stabilisées à partir d'un carcinome colique et représentent un modèle entérocytaire in vitro

Abstract:
Trois magmas (haplotonalite synthétique, basalte et dacite de Santorin) ont été juxtaposés et déformés à l'aide d'une presse de type Paterson pour contraindre les conditions de mélange de magmas à haute pression (300 MPa). Ces trois magmas ont été utilisés en conditions sèches ou hydratées et dans une gamme de température comprise entre 600 et 1200°C, permettant d'obtenir plusieurs fractions cristallines et contrastes de viscosité. Les textures de mélange produites lors des expériences dépendent de la fraction cristalline et sont similaires aux textures rencontrées dans la nature. Les textures de mélange mécanique (mingling) produites sont la ségrégation de cristaux depuis leur magma source, la formation d'enclaves par détachement et de filaments par étirement de parcelles de magma. Le mélange chimique (mixing) est illustré par des zones d'interactions comportant une large variété de liquides intermédiaires et la cristallisation de nouvelles phases. Le mélange est produit en l'absence d'une charpente cristalline dans l'un ou l'autre des magmas, si le contraste de viscosité est faible (< 0,3 unité log) et en dessous d'un seuil de viscosité absolue, déterminé entre 107 et 108 Pa.s. Par ailleurs, l'eau joue un rôle important sur les propriétés rhéologiques des magmas, et donc sur leurs capacités de mélanges : elle abaisse le seuil de mélange de près de 200°C entre des magmas saturés en eau, et son exsolution (présence de bulles) entraîne une réduction significative des viscosités promouvant le mélange à des fractions cristallines plus faibles. Les conditions rhéologiques favorables aux mélanges entre le magma d'un réservoir et un magma plus chaud qui le recharge ont été déterminées selon la fraction de magma injecté. Cette fraction est de 0,5 minimum pour la majorité des réservoirs typiques du contexte d'arc. La comparaison avec des systèmes plutoniques et volcaniques exposant des figures de mélange montre que les conditions favorables aux mélanges sont atteintes avec des fractions inférieures. Ceci suggère que le fonctionnement d'un réservoir magmatique est influencé par ses styles et taux de recharge, ainsi que la quantité de volatils.

Cette these est consacree a l'etude de deux recepteurs de la serotonine, un neuromediateur implique dans le controle de nombreuses fonctions du systeme nerveux central. Ces recepteurs caracterisent les deux grandes familles de recepteurs de neuromediateurs puisque le recepteur 5-ht#1#a est couple a une proteine g, tandis que le recepteur 5-ht#3 est un canal ionique. Des anticorps polyclonaux anti-recepteur 5-ht#1#a de rat ont ete produits chez le lapin en utilisant comme antigene, un peptide synthetique correspondant a un domaine tres specifique de la troisieme boucle intracytoplasmique de ce recepteur. Ces anticorps n'immunoprecipitent que les sites 5-ht#1#a dans un solubilisat de membranes d'hippocampes et ne reconnaissent en immunoblot qu'une seule proteine d'un poids moleculaire apparent de 63 kda, identique a celui du recepteur 5-ht#1#a determine par d'autres techniques. Enfin, la distribution du marquage immunohistochimique par ces anticorps dans le systeme nerveux central du rat se superpose exactement a celle des sites 5-ht#1#a identifies par la liaison d'un radioligand selectif, le #3h8-oh-dpat. Dans des cultures primaires de neurones provenant du septum de rats au 17#e#m#e jour d'age ftal, les anticorps anti-recepteur 5-ht#1#a revelent la presence de ce recepteur sur des neurones cholinergiques identifies par des anticorps anti-choline acetyltransferase. Cette observation est a la base de la seconde partie de ce travail de these qui a conduit a la mise en evidence d'une influence trophique de l'activation des recepteurs 5-ht#1#a sur les neurones cholinergiques en culture. Ainsi, les agonistes des recepteurs 5-ht#1#a (8-oh-dpat, gepirone, buspirone et surtout ipsapirone) stimulent la croissance de l'arborisation neuritique de ces neurones, entrainant une augmentation significative de l'activite choline acetyltransferase dans les cultures. Le mecanisme cellulaire a l'origine de cet effet reste a preciser ; quoi qu'il en soit, il n'implique ni le ngf, ni la proteine s-100. En mesurant le flux entrant du cation #1#4cguanidinium dans les cellules hybrides ng108-15 en culture, nous avons montre que la composante serotoninergique de la stimulation obtenue en presence de 5-ht (1 m) et de sp (10 m) est liee a l'activation des recepteurs 5-ht#3 a la surface de ces cellules. A partir de cette observation, une methode simple et rapide a ete developpee pour la determination des proprietes agonistes ou antagonistes de nouveaux ligands des recepteurs 5-ht#3, sans le recours a des investigations electrophysiologiques souvent lourdes et delicates. L'etude des mecanismes a l'origine de l'augmentation d'influx de #1#4cguanidinium dans les cellules ng108-15 exposees a un agoniste des recepteurs 5-ht#3, montre que cette reponse (qui traduit l'ouverture d'un canal cationique) requiert le ca#2#+ intracellulaire, au travers du complexe ca#2#+-calmoduline

Over the last decade, several large-scale randomized trials have reported results that disagreed substantially with the motivating observational studies on the value of various chronic disease–prevention strategies. One high-profile example of these discrepancies was related to postmenopausal hormone therapy (HT) use and its effects on cardiovascular disease and cancer. The Women’s Health Initiative (WHI), a National Heart, Lung, and Blood Institute–sponsored program, was designed to test three interventions for the prevention of chronic diseases in postmenopausal women, each of which was motivated by a decade or more of analytic epidemiology. Specifically, the trials were testing the potential for HT to prevent coronary heart disease (CHD), a low-fat eating pattern to reduce breast and colorectal cancer incidence, and calcium and vitamin D supplements to prevent hip fractures. Over 68,000 postmenopausal women were randomized to one, two, or all three randomized clinical trial (CT) components between 1993 and 1998 at 40 U.S. clinical centers (Anderson et al., 2003a). The HT component consisted of two parallel trials testing the effects of conjugated equine estrogens alone (E-alone) among women with prior hysterectomy and the effect of combined estrogen plus progestin therapy (E+P), in this case conjugated equine estrogens plus medroxyprogesterone acetate, among women with an intact uterus, on the incidence of CHD and overall health. In 2002, the randomized trial of E+P was stopped early, based on an assessment of risks exceeding benefits for chronic disease prevention, raising concerns among millions of menopausal women and their care providers about their use of these medicines. The trial confirmed the benefit of HT for fracture-risk reduction but the expected benefit for CHD, the primary study end point, was not observed. Rather, the trial results documented increased risks of CHD, stroke, venous thromboembolism (VTE), and breast cancer with combined hormones (Writing Group for the Women’s Health Initiative Investigators, 2002). Approximately 18 months later, the E-alone trial was also stopped, based on the finding of an adverse effect on stroke rates and the likelihood that the study would not confirm the CHD-prevention hypothesis.

ABSTRACT: Reservoir thermal energy storage (RTES) is a promising technology to balance the mismatch between energy supply and demand. In particular, high temperature (HT) RTES can stabilize the grid with increasing penetration of renewable energy generation. This paper presents the investigation of the mechanical deformation and chemical reaction influences on the performance of HT-ATES for the Lower Tuscaloosa site. Thermo-hydraulic (TH), thermo-hydro-mechanical (THM), and thermo-hydro-chemical (THC) coupled simulations were performed with different operational modes and injection rates for a fixed five-spot well configuration and a seasonal cycle. The results show that (1) geomechanical-induced porosity change is mainly contributed by effective stress change, and the porosity change is distributed through the whole system; (2) geochemistry-induced porosity change is located near the hot well, and its change is one order of magnitude higher than the geomechanical effect; (3) both the operation mode and the injection rate have a huge influence on the RTES performance and lower injection rate with push-pull operation mode has the best performance with recovery factor around 70% for this RTES system. These results shed light on the deployment of HT-RTES in the US and around the world. 1 INTRODUCTION The concept of reservoir thermal energy storage (RTES), also known as geological thermal energy storage (GeoTES) or aquifer thermal energy storage (ATES), to mitigate the mismatch between energy supply and demand has been applied around the world since the 1960s with mixed success. Given its nearly unlimited storage capacity and easy accessibility, RTES has the potential to become an indispensable component to achieve the goal of carbon-neutral energy. Most successful deployments of RTES are operated at low temperatures (LT) (&lt; 25°C), mainly to heat buildings by storing excess thermal energy during the low-use periods (summer) and recovering it during peak energy demand periods (winter). As reviewed by Fleuchaus et al. (2018), there are currently more than 2800 RTES systems worldwide, and 99% are LT-RTES. However, only high-temperature (HT) RTES has the capacity to serve as an earth battery for stabilizing the grid as indicated in McLing et al. (2019). The research and development of HT-RTES have mainly focused on site suitability studies and performance optimization by only considering fluid flow and heat transfer. For example, Schout et al. (2014) extended the widely adopted Rayleigh number - recovery factor relationship for identifying site suitability of LT-RTES systems (Gutierrez-Neri et al., 2011) to HT-RTES systems. Sheldon et al. (2021) further improved the Rayleigh number relationship to consider daily cycles for HT-RTES systems. In addition to recovery factor, the performance metrics of HT-RTES include storage capacity, operational duration, etc. Jin et al. (2021, 2022) performed stochastic thermo-hydraulic simulations and used a machine learning algorithm to directly correlate formation parameters and operational conditions with multiple HT-RTES performance metrics using the simulated big data. All these investigations can facilitate the deployment of HT-RTES. However, geomechanical response and geochemical reactions involved during the operation of a HT-RTES system can potentially induce risks as identified by Fleuchaus et al. (2020), and their effects on HT-RTES performance have not been systematically reported.

Abstract:
Background: Traumatic brain injury(TBI) is an important component of mortality rate(MR) among young adults. Hypothermia(HT) is a treatment with questionable efficacy, given existing discordant views. Objective: investigate the performance of HT in TBI. Methods: Randomized controlled trials(RCT) from PUBMED, over the last five years, were analysed through the descriptors “Traumatic brain injury”; “Therapeutic hypothermia” and its MeSH variations. RCTs in individuals over 19 were included. After criteria were applied, four were selected for this review. This systematic review was submitted for registration on the PROSPERO plataform(ID: 253817). Results: Eurotherm3235 segregated n=387 to a group subjected to HP and another to standard treatment, evaluating them through the Glasgow Scale. The RCT was interrupted due potential damage to the participants, and showed greater MR(p <0.05) in the experimental group (EG). The POST-HOC of the B-HYPO sorted n=129 into a control group on fever management, and another group subjected to HT, separating them according to the injury. The patients with minor severity from the EG showed higher MR(p <0.05). The RCT from Chunhai T et al.(n=60) subjected to decompressive hemicraniectomy, which were divided into those who received HT, and a control group, displaying an inferior MR in the EG (p<0.05). Feng JZ et al. with n=14 subjected the EG to HT, and analysed the participants through the magnetic resonance metabolomics, showing lesser MR and better recovery in the EG(p<0.05). Conclusion: The studies indicated disparate results. However, those with larger samples indicated worse prognosis between patients subjected to HT.

Abstract:
The occlusion time of a Butterfly 25 short cannula inserted into the cubital vein was assessed as a new bleeding time modification. The method excludes the influences of vessel wall and tissue factors since primary haemostasis takes place on the standard surface of the cannula. Avoiding scar formation, the technique can be repeated deliberately. It could be shown that HT correlated better than the Duke and Ivy/ template bleeding times with reduced platelet counts and reflected the effects of aspirin and unfractionated heparin in the same way (Janzarik et al., Blut 1986, 52:345).HT was applied simultaneously with the Duke and Ivy/ template or Simplate bleeding times in haemophilia and von Willebrand's disease (vWD). According to most of the other authors, the skin bleeding times were normal in patients with haemophilia. However, HT was prolonged in 9/16 patients irrespective of the factor VIII:C/IX:C values. The duration of HT correlated significantly to the onset of bleeding symptoms and to the degree of spontaneous bleeding (e.g., haemarthro-ses). In 31 patients with clinically mild or moderate vWD, the Duke bleeding time was prolonged in 3/31 patients, Simplate II in 13/31 patients and HT in 14/31 patients. With the two latter methods, corresponding results were observed in 20 cases, different results in 11 cases. HT and both the other methods correlated significantly to f. VIII:RCo and VIII:C but not to VIII: vWFWe conclude that the repeatable HT method is an interesting diagnostic parameter in haemophilia and vWD. With the Simplate and HT methods simultaneously performed, the influence of vessel wall factors on primary haemostasis may be differentiated.

Large Eddy Simulation (LES) has become an increasingly attractive option for turbulence modeling due to the rise in computing power and the improvement in sub-grid scale (SGS) parameterizations. This study tests the improvements in simulations of wall-bounded flows over heterogeneous surfaces attained by the implementation of three improvements in the eddy-viscosity SGS closure: the dynamic model by Germano et al. [1], the Lagrangian model by Meneveau et al. [2], and the scale-dependent approach by Porte´-Agel et al. [3]. The dynamic model consists of using the resolved scales to ‘measure’ the model coefficient during the simulation; therefore, no a-priori knowledge of the coefficient or the flow physics is needed. The traditional dynamic approach averages the coefficient over statistically homogeneous directions to numerically stabilize the simulations. The Lagrangian model relaxes the need for homogeneous directions by averaging the coefficient over pathlines, hence allowing local determination of the coefficient and facilitating applications to complex-geometry flows. The scale-dependent approach uses the dynamic formulation but does not assume that the SGS coefficients are scale-invariant, as is the case in traditional dynamic formulations. The deficiencies of the traditional Smagorinsky model are confirmed. Implementation of a dynamic model treats some of these deficiencies but is found to be under-dissipative close to the wall in high Reynolds number LES that does not resolve the viscous layer. The sensitivity of the model coefficient to the wall roughness is demonstrated thus confirming the need for a local SGS model such as the Lagrangian model used here. Finally, when the Lagrangian-dynamic model is implemented with the scale-dependent formulation, the results improve significantly.

Serotonin (5-HT) stimulates prostacyclin (PGI2) production by bovine aortic smooth muscle cells in culture via 5-HT2 receptors (1). These cells express a synthetic phenotype (2), whereas the majority of the smooth muscle cells in the media from adult arteries are in a contractile state. We have now shown that 5-HT (1-10 μM) also stimulates PGI2 production by a preparation of contractile smooth muscle cells : explants from bovine aortic media cultured for short periods. This effect is independent from 5-HT2 receptors : it is only partially inhibited (±30%) by ketan-serin (a selective and potent 5-HT2 antagonist) and is perfectly mimicked by a 5-HT1 agonist, 5-carboxamidotryptamine. 5-HT2 receptors seem to be linked to a phospholipase C (3), with subsequent accumulation of inositol tr isphosphate , Ins(1,4,5)P3, and diacylglycerol, an activator of protein kinase C. We have observed a stimulatory effect of phorbol 12-myristate, 13-acetate (a selective activator of kinase C) on PGI2 production by the bovine aortic smooth muscle cells (synthetic state), whereas it was totally ineffective on media explants preparation (contractile state). Furthermore, in the smooth muscle cells in culture, the 5-HT effect can be inhibited by (ethyl-isopropyl)amiloride, a potent and selective inhibitor of the Na+/H+ antiporter. In conclusion it appears that the regulation mechanisms of PGI2 production in arterial smooth muscle cells are strongly dependent an the phenotypic state of these cells. Abstract:
The paper presents a new approach to predict the two-phase performance of jet-pumps under interference conditions. We limit our study mainly to diffuser and transport regions of the jet pump. The five essential pre-requisites which form the backbone of our approach are a fairly generalized and accurate approach to (i) solid-fluid interaction, (ii) particle diffusion under generalized flow field, (iii) friction factor-Reynolds number equation, (iv) solid-fluid flow through ducts and (v) mixing of primary and secondary jets using the approach of Wang et al. [1] based on boundary layer concept. The extensive experimental data of several research workers along with fresh data generated on specially designed test-rig support the new approach.

Thermionic emission in vacuum could be a highly efficient cooler or power generator if the work function, the minimum work for electrons to go into vacuum, is around 0.3–0.4 eV for heat source at a temperature below 500C [Mahan, 1994]. Unfortunately, the work function of existing materials is currently above 1 eV. Theoretical and experimental studies have shown that the work function can be reduced to 0.3–0.4 eV if the distance between the two electrodes (cathode and anode) of the thermionic emission cooler/power generator is below 10 nm [Hishinuma, et al, 2001, 2003]. At this nanometer scale, electron transport between the two electrodes takes two paths: electron tunneling and thermionic emission. The combined physical processes result in a desired work function. However, maintaining a nanometer gap for two parallel plates within an area larger than 1 cm2 is a daunting task, if not impossible, especially if the power generator is mounted on a moving or vibrating device. Even a slight vibration or thermal expansion of the two plates (electrodes) could cause direct contact between the two plates (electrodes), and thus shorten the circuits. Thus vacuum thermionic power generator based on difficult to make and to operate [Tavkhelidze, et al., 2002]. In this study, we propose to use a solid insulating spacer for preventing the shortening and for feasibility of manufacturing. The spacer is less than 5nm, and electron transport as thermionic emission and tunneling concurrently. In this study, we first investigate electron and phonon transport in single-layer (spacer) double heterostructures by including the tunneling effects. It is found that single-layer generator can have a high efficiency, but small power intensity due to the small temperature difference between the two electrodes. We then investigate the efficiency of multilayer-layer power generator. Calculations show that the solid power generator operating at a temperature below 500°C, can have an efficiency of larger than 40% of the Carnot efficiency.

Abstract:
As advocated by Denton [1], three-dimensional CFD calculations are more and more used in the process of designing centrifugal compressors. Therefore, numerical accuracy is an issue if valuable design decisions are to be based on CFD results. This paper aims at correlating mesh-parameters and calculated flow quantities. The design-of-experiment method (DOE) is used to identify which mesh-parameters influence any given flow quantity. The emphasis is put on global performances (mass-flow rate, pressure-ratio and efficiency), and on local features (friction co-efficients and local velocity-profiles). The outcome of the study is the connection between mesh-parameters and specific flow-properties. This can be further used to derive numerical bench-marks, such as reported by Dufour et al. [2] for the tip-clearance flow.

Abstract:
The v2–f models of Durbin and the new v2–f model of Jones et al. were used to compute the blade heat transfer for the transonic cascade of Giel et al.. Three different experimental cases were considered. Calculations were performed using the Glenn-HT code modified so that it solves these turbulence model equations using an implicit scheme. An iterative matrix solver, namely GMRES was used for this purpose. Comparisons were made with previously published results using the SST model and a simple algebraic model modified for the stagnation heat transfer effects and transition effects. The results, albeit limited to the present case, show that the v2–f model of Durbin provides good predictions on the pressure surface. But there is some discrepancy between the suction surface heat transfer predictions and experimental measurements. The newer model of Jones et al. requires further modification before it can be routinely used for blade heat transfer.
